Estimates of dolphin school sizes made by observers and crew members aboard tuna seiners or by observers on ship or aerial surveys are important components of population estimates of dolphins which are involved in the yellowfin tuna fishery in the eastern Pacific. Differences in past estimates made from tuna seiners and research ships and aircraft have been noted by Brazier (1978). To compare various methods of estimating dolphin school sizes a research cruise was undertaken with the following major objectives: 1) compare estimates made by observers aboard a tuna seiner and in the ship's helicopter, from aerial photographs, and from counts made at the backdown channel, 2) compare estimates of observers who are told the count of the school size after making their estimate to the observer who is not aware of the count to determine if observers can learn to estimate more accurately, and 3) obtain movie and still photographs of dolphin schools of known size at various stages of chase, capture and release to be used for observer training. The secondary objectives of the cruise were to: 1) obtain life history specimens and data from any dolphins that were killed incidental to purse seining. These specimens and data were to be analyzed by the U.S. National Marine Fisheries Service ( NMFS ) , 2) record evasion tactics of dolphin schools by observing them from the helicopter while the seiner approached the school, 3) examine alternative methods for estimating the distance and bearing of schools where they were first sighted, 4) collect the Commission's standard cetacean sighting, set log and daily activity data and expendable bathythermograph data. Historical definitions of what determines whether one lives in a coastal area or not have varied over time. According to Culliton (1998), a “coastal county” is defined as a county with at least 15% of its total land area located within a nation’s coastal watershed. This emphasizes the land areas within which water flows into the ocean or Great Lakes, but may be better suited for ecosystems or water quality research (Crowell et al. 2007). Some Federal Emergency Management Agency (FEMA) documents suggest that “coastal” includes shoreline-adjacent coastal counties, and perhaps even counties impacted by flooding from coastal storms. An accurate definition of “coastal” is critical in this regard since FEMA uses such definitions to revise and modernize their Flood Insurance Rate Maps (Crowell et al. 2007). A recent map published by the National Oceanic and Atmospheric Administration’s (NOAA) Coastal Services Center for the Coastal Change Analysis Program shows that the “coastal” boundary covers the entire state of New York and Michigan, while nearly all of South Carolina is considered “coastal.” The definition of “coastal” one chooses can have major implications, including a simple count of coastal population and the influence of local or state coastal policies. There is, however, one aspect of defining what is “coastal” that has often been overlooked; using atmospheric long-term climate variables to define the inland extent of the coastal zone. This definition, which incorporates temperature, precipitation, wind speed, and relative humidity, is furthermore scalable and globally applicable - even in the face of shifting shorelines. A robust definition using common climate variables should condense the large broad definition often associated with “coastal” such that completely landlocked locations would no longer be considered “coastal.” Moreover, the resulting definition, “coastal climate” or “climatology of the coast”, will help coastal resource managers make better-informed decisions on a wide range of climatologically-influenced issues. Investigation were carried out on the effect of some locally available species in the enhancement of the organoleptic quality and the storage periods of smoked Heterotis niloticus using Pprosopis africana as common smoke sources. Samples of fresh H. niloticus were bought, cut into chunks while extract juice from pepper, ginger rhizomes, garlic, onion bulb were used as sources of spices. Samples of fish were divided randomly into five (5) batches dipped into spice extract juices for 10 minutes drained and smoked with common firewood. Treatment without spice extract juice served as control. Each batch of fish was smoked for 7 hours on a drum-made smoking kiln products were individually packaged in polythene bag stored at room temperature and used for sensory evaluation and microbial analysis. Results of the sensory evaluation indicated that there was significant difference (P<0.005) for taste, appearance, colour and overall acceptance for the treatments. Ginger juice extract had the best overall acceptance. Similarly there was significant difference (P>0.05) in the microbial analysis. The garlic juice extract had the longest storage period with minimum total plate and mould count after 8 weeks

Survey of Fungal infestation of some species of fish in Tagwai Dam Minna was carried out from March to June 2002. Fungi were isolated from the scale/skin, gills and fins. Twenty-one fungi species were identified from 18 species of fish microbial growth was measured by direct cell count using Stuart colony counter. Most of the fungi encountered were of the mould group and infestation occurred among all the species sampled. The infestation was predominantly by Aspergillus species and the scale/skin was most widely affected. The study showed the identified fungi in order of frequency to be as follows. Aspergillius niger. Rrhizopus spp. Mmucor spp Aspergilus flavum. Aspergillus parasitous. Aspergillus fumigatus, Microsporum canis. Penicillum virridicalumand Fusarium spp respectively. Aspergillus niger occurred on all the species of fish sampled. Barrilius spa and Chrysichrhy auratus longifilis had significantly higher (P<0.05) mean fungal load on their fins and gills. There was no significant difference (P>0.05) in the mean fungal load in different parts of the body of other fish species sampled

Studies were carried out using 96hr static toxicity bioassay to determine the effect of lethal concentrations of extracts from two local plants Tephrosia vogelii and Parkia clappertoniana which are known fish poison, on a species of mud fish. Clarias gariepinus Phytochemical analysis of the plant extracts was done and the extract from T. vogelii was found to contain alkaloids, tannins and flavonoids, while the extract from P. clappertoniana was formed to contain alkaloids tannins and saponins. Experimental fish were exposed to test water separately polluted by varying concentrations of extraction of both plant species ranging from 0.50mgl super(-1), 1.50mgl super(-1), 2.50mgl super(-1), 3.0mgl super(-1), 5.00mgl super(-1), 10.00mgl super(-1) in the case of T. vogelii and 5.00mgl super(-1), 7.50mgl super(-1), 10.00mgl super(-1), 15.00mgl super(-1), 20.00mgl super(-1) and 30.00mgl super(-1) in the case of P. clappertaniana. Behavioural hispathological and heamatological examinations were made. Both plant extracts were found to have lethal effects at the higher concentrations, affecting the gills and the central nervous system as well as having a depressive effect on the total count and increasing platelet and white blood cell count. Symptoms of toxicosis observed include, initial inactivation agitated swimming, tumbling movement air gulping, increased opercular beat and period of quiescence/knockdown before death. Marked differences were also observed in the hematological and histopathological analysis of poisoned fish. Lower concentrations of the extracts had sub lethal effects on the fish, which manifested as zigzag movement air gulping increased opercular movement etc. None of these effects were observed in the control experiment

The contributions of hematological factors to the distribution and estimations of Eustrongylides africanus larvae densities in Clarias gariepinus and C. anguillaris of Bida floodplain of Nigeria were documented for the first time. The hematological factors making the most important contributions to the distributions of E. africanus larvae infections in clarias species are mean corpuscular haemoglobin concentration (MCHC), mean corpuscular haemoglobin (MCH), mean corpuscular volume (MCV) and neutrophils count, in descending order of magnitude; having the manifestations for the months of January, March, September, and December of the year being closely related. Five haematological factors (neutrophils, lymphocytes and eosinophils counts; MCH and MCV) having positive or negative correlation coefficient (r) between 0.50 and 0.85 contributed to the estimated of E.africanus larvae densities in the wild population of Clarias species

Hematological effects of feeding varying dietary crude proteins levels to one hundred and fifty (150) H.longifilis fingerlings was examined on biweekly basis. The fingerlings of mean weights 1.26g plus or minus 0.24g were stocked in eight hapa nets (1mx1m) at 15 fingerlings per hapa. Four experimental diets with crude protein; 35%, 40%, 45% and 50% coded diet 1-4 respectively were fed to the fish for 8 weeks. The blood sample was taken and examined for packed cell volume (PCV) total protein (TP) Hemoglobin (Hb), Serum album, Erythrocyte count (RBC), while blood cell (WBC) mean corpuscle volume (MCV) and mean corpuscle hemoglobin, concentration (MCHC). There was an increase in the values of the hematological indices studied with increase in protein inclusion levels. A higher positive correlation with no significant difference (P greater than or equal to 0.05) exists between the treatments RBC, WBC, Hb and TP. The best RBC (2.10x10 super(6) count/l). WBC (7.65x10 super(4) count/l), PCV (35.4%) and Hb (5.79mg/l) were presented in fingerlings fed 40% crude protein followed by 45% crude protein. The dietary crude protein of 40% is recommended for H. longifilis for sound and healthy condition

En la última década, la diversidad social se ha visto reflejada en nuestras escuelas, y ha llevado a nuestro sistema educativo a desarrollar políticas y planes de actuación que favorezcan el desarrollo de una escuela de calidad para todos. En el presente trabajo planteamos analizar las prácticas educativas que llevan a cabo en el centro Hijas de la Caridad de Nuestra Señora de Begoña, y realizar propuestas de mejora que puedan contribuir al progreso hacia un centro más inclusivo. Para el desarrollo del trabajo hemos utilizado la técnica de grupos focales contando con la participación de miembros activos de la comunidad educativa. (Español)

This work seeks to understand past and present surface conditions on the Moon using two different but complementary approaches: topographic analysis using high-resolution elevation data from recent spacecraft missions and forward modeling of the dominant agent of lunar surface modification, impact cratering. The first investigation focuses on global surface roughness of the Moon, using a variety of statistical parameters to explore slopes at different scales and their relation to competing geological processes. We find that highlands topography behaves as a nearly self-similar fractal system on scales of order 100 meters, and there is a distinct change in this behavior above and below approximately 1 km. Chapter 2 focuses this analysis on two localized regions: the lunar south pole, including Shackleton crater, and the large mare-filled basins on the nearside of the Moon. In particular, we find that differential slope, a statistical measure of roughness related to the curvature of a topographic profile, is extremely useful in distinguishing between geologic units. Chapter 3 introduces a numerical model that simulates a cratered terrain by emplacing features of characteristic shape geometrically, allowing for tracking of both the topography and surviving rim fragments over time. The power spectral density of cratered terrains is estimated numerically from model results and benchmarked against a 1-dimensional analytic model. The power spectral slope is observed to vary predictably with the size-frequency distribution of craters, as well as the crater shape. The final chapter employs the rim-tracking feature of the cratered terrain model to analyze the evolving size-frequency distribution of craters under different criteria for identifying "visible" craters from surviving rim fragments. A geometric bias exists that systematically over counts large or small craters, depending on the rim fraction required to count a given feature as either visible or erased.

The ritterazine and cephalostatin natural products have biological activities and structures that are interesting to synthetic organic chemists. These products have been found to exhibit significant cytotoxicity against P388 murine leukemia cells, and therefore have the potential to be used as anticancer drugs. The ritterazines and cephalostatins are steroidal dimers joined by a central pyrazine ring. Given that the steroid halves are unsymmetrical and highly oxygenated, there are several challenges in synthesizing these compounds in an organic laboratory.

Ritterazine B is the most potent derivative in the ritterazine family. Its biological activity is comparable to drugs that are being used to treat cancer today. For this reason, and the fact that there are no reported syntheses of ritterazine B to date, our lab set out to synthesize this natural product.

Herein, efforts toward the synthesis of the western fragment of ritterazine B are described. Two different routes are explored to access a common intermediate. An alkyne conjugate addition reaction was initially investigated due to the success of this key reaction in the synthesis of the eastern fragment. However, it has been found that a propargylation reaction has greater reactivity and yields, and has the potential to reduce the step count of the synthesis of the western fragment of ritterazine B.

The genomes of many positive stranded RNA viruses and of all retroviruses are translated as large polyproteins which are proteolytically processed by cellular and viral proteases. Viral proteases are structurally related to two families of cellular proteases, the pepsin-like and trypsin-like proteases. This thesis describes the proteolytic processing of several nonstructural proteins of dengue 2 virus, a representative member of the Flaviviridae, and describes methods for transcribing full-length genomic RNA of dengue 2 virus. Chapter 1 describes the in vitro processing of the nonstructural proteins NS2A, NS2B and NS3. Chapter 2 describes a system that allows identification of residues within the protease that are directly or indirectly involved with substrate recognition. Chapter 3 describes methods to produce genome length dengue 2 RNA from cDNA templates.

The nonstructural protein NS3 is structurally related to viral trypsinlike proteases from the alpha-, picorna-, poty-, and pestiviruses. The hypothesis that the flavivirus nonstructural protein NS3 is a viral proteinase that generates the termini of several nonstructural proteins was tested using an efficient in vitro expression system and antisera specific for the nonstructural proteins NS2B and NS3. A series of cDNA constructs was transcribed using T7 RNA polymerase and the RNA translated in reticulocyte lysates. Proteolytic processing occurred in vitro to generate NS2B and NS3. The amino termini of NS2B and NS3 produced in vitro were found to be the same as the termini of NS2B and NS3 isolated from infected cells. Deletion analysis of cDNA constructs localized the protease domain necessary and sufficient for correct cleavage to the first 184 amino acids of NS3. Kinetic analysis of processing events in vitro and experiments to examine the sensitivity of processing to dilution suggested that an intramolecular cleavage between NS2A and NS2B preceded an intramolecular cleavage between NS2B and NS3. The data from these expression experiments confirm that NS3 is the viral proteinase responsible for cleavage events generating the amino termini of NS2B and NS3 and presumably for cleavages generating the termini of NS4A and NS5 as well.

Biochemical and genetic experiments using viral proteinases have defined the sequence requirements for cleavage site recognition, but have not identified residues within proteinases that interact with substrates. A biochemical assay was developed that could identify residues which were important for substrate recognition. Chimeric proteases between yellow fever and dengue 2 were constructed that allowed mapping of regions involved in substrate recognition, and site directed mutagenesis was used to modulate processing efficiency.

Expression in vitro revealed that the dengue protease domain efficiently processes the yellow fever polyprotein between NS2A and NS2B and between NS2B and NS3, but that the reciprocal construct is inactive. The dengue protease processes yellow fever cleavage sites more efficiently than dengue cleavage sites, suggesting that suboptimal cleavage efficiency may be used to increase levels of processing intermediates in vivo. By mutagenizing the putative substrate binding pocket it was possible to change the substrate specificity of the yellow fever protease; changing a minimum of three amino acids in the yellow fever protease enabled it to recognize dengue cleavage sites. This system allows identification of residues which are directly or indirectly involved with enzyme-substrate interaction, does not require a crystal structure, and can define the substrate preferences of individual members of a viral proteinase family.

Full-length cDNA clones, from which infectious RNA can be transcribed, have been developed for a number of positive strand RNA viruses, including the flavivirus type virus, yellow fever. The technology necessary to transcribe genomic RNA of dengue 2 virus was developed in order to better understand the molecular biology of the dengue subgroup. A 5' structural region clone was engineered to transcribe authentic dengue RNA that contains an additional 1 or 2 residues at the 5' end. A 3' nonstructural region clone was engineered to allow production of run off transcripts, and to allow directional ligation with the 5' structural region clone. In vitro ligation and transcription produces full-length genomic RNA which is noninfectious when transfected into mammalian tissue culture cells. Alternative methods for constructing cDNA clones and recovering live dengue virus are discussed.

O objetivo deste estudo foi avaliar o efeito da microestrutura dentinária na adesão de cimentos endodônticos modernos através: do desenvolvimento de uma metodologia para caracterizar microestruturalmente a dentina; da avaliação da resistência de união (através de ensaios de tração) dos cimentos endodônticos Epiphany SE, iRoot SP e AH Plus; da correlação dos dados obtidos da caracterização da microestrutura dentinária e dos ensaios de tração. Trinta terceiros molares inclusos, recém extraídos, foram embutidos em resina epóxi e seccionados 0,5mm abaixo da junção esmalte-dentina. Doze amostras foram eliminadas durante à preparação metalográfica devido à exposição da câmara pulpar. De cada dente, uma área de análise (AA) com 3,25 mm de diâmetro foi mapeada utilizando técnicas de microscopia ótica digital para: captura de ~400 imagens formando um mosaico; análise digital de imagens, obtendo os resultados de quantidade de túbulos (QT) e de fração de área de túbulos (FAT) do mosaico; conversão da imagem do mosaico em um mapa colorido em que as cores estão diretamente relacionadas à densidade de área tubular. As dezoito amostras restantes foram divididas em 3 grupos (N=6), de acordo com o cimento utilizado, para confecção dos corpos de prova para os ensaios de tração. Os valores de da resistência de união (RU) obtidos foram analisados estatisticamente com teste não-pareado t com correção de Welch e pelo teste F para comparar variâncias Os dados de FAT e de QT foram submetidos ao teste de D'Agostino & Pearson revelando-se não normais (P>0,05), o que indica grande variabilidade da amostragem. O cimento Epiphany SE apresentou uma resistência de união significativamente menor que o cimento AH Plus (P <0,05). Os corpos de prova do iRoot SP falharam prematuramente e não foram analisados. A aplicação do teste r de Spearman não demonstrou correlação significativa entre FAT e RU (P>0,05). O MEV de pressão variável foi utilizado para avaliar qualitativamente a superfície de fratura após os ensaios de tração. A análise revelou um padrão de fratura mista para o AH Plus e o Epiphany SE, além de diferenças no tamanho e na forma das partículas desses cimentos, o que pode influenciar no comportamento mecânico. Dentro da amostragem realizada, não se encontrou correlação significativa entre microestrutura dentinária e a resistência de união.

Um Atlas Digital é um atlas que foi concebido através de técnicas computacionais e que, consequentemente, pode ser acessado através de um computador. Estruturado em um ambiente gráfico, além dos mapas, pode-se contar também com textos, fotografias, dados estatísticos, gráficos e tabelas. Por estar em meio digital existe a possibilidade de utilização de uma expressiva gama de temas, formatos e escalas. Nesta dissertação apresenta-se um protótipo de Atlas Digital como uma colaboração ao Sistema de Informação Municipal SIM, para o município de São João de Meriti, RJ. O referido SIM, que tem como meta os serviços municipais, visa atender ao próprio município, ao cidadão e a outros interessados na cidade, sendo as suas informações fundamentais para a melhoria da gestão das prefeituras. A pesquisa foi direcionada para o tema da habitabilidade, que consiste num conjunto de condições voltadas para a construção de habitat saudável, abrangendo temas físicos, psicológicos, sociais, culturais e ambientais. Dentro do tema habitabilidade, foram trabalhados os subtemas relativos a infraestrutura de abastecimento de água, esgoto, coleta de lixo, saúde e educação, esses subtemas foram confrontados entre si para uma comparação entre os bairros do município. O SIM e a habitabilidade são contemplados no plano diretor da cidade e representa uma grande parte da sustentação teórica da dissertação. A modelagem e implementação do protótipo do Atlas Digital foram feitas com auxílio de softwares gratuitos, sendo possível acessar mapas temáticos e outras informações sobre São João de Meriti

Pesquisas recentes têm demonstrado que a periodontite pode modificar a concentração sanguínea de uma série de tipos celulares e substâncias bioquímicas, que são considerados fatores de risco para doenças cardiovasculares. Este trabalho tem como objetivo avaliar a associação entre a periodontite crônica e marcadores de risco para doença cardiovascular. No Estudo I foram examinados 100 pacientes aparentemente saudáveis sistemicamente, sendo 66 portadores de periodontite crônica e 34 pacientes controle, sem doença periodontal. Exames periodontais e exames sanguíneos foram realizados, e obtidas as espessuras das camadas íntima-média (IMT) da artéria carótida. No Estudo II, 66 pacientes participantes do Estudo I, diagnosticados com periodontite crônica, foram aleatoriamente submetidos a tratamento periodontal imediato (Grupo Teste, n=33) ou tratamento periodontal retardado (Grupo Controle, n=33). Os dados colhidos no Estudo I foram registrados como pré-tratamento (T0). Novos exames clínicos periodontais e laboratoriais foram realizados no período de 2 meses (T2) e 6 meses (T6) após os exames iniciais (Grupo Controle) ou conclusão do tratamento periodontal (Grupo Teste). Os dados colhidos foram analisados através de testes estatísticos. Os resultados mostraram que pacientes com periodontite crônica quando comparados ao grupo controle, apresentaram valores médios significativamente mais elevados na contagem total de hemácias (p<0,001), hemoglobina (p<0,001), hematócrito (p<0,001), contagem de plaquetas (p=0,019), velocidade de hemossedimentação (p<0,001), proteína C-reativa (p<0,001). Os níveis de HDL-colesterol foram significativamente mais baixos nos pacientes com periodontite crônica quando comparados ao grupo controle (p<0,001). As camadas íntima-média da parede da artéria carótida esquerda foram significativamente mais espessas nos pacientes com periodontite crônica quando comparados ao grupo controle (p=0,049). Os indíviduos com periodontite crônica também apresentaram 3,26 vezes mais chances de possuir Síndrome Metabólica do que aqueles indivíduos que não possuem doença peridontal (IC 95%: 1,8-5,9). No Estudo II, quando comparados os valores médios dos dados hematológicos após tratamento, no grupo teste, foi possível observar melhora estatisticamente significativa, entre T0/T2, dos valores de VHS e triglicerídeos (p=0,002; p=0,004; respectivamente). Redução nos valores médios da contagem total de leucócitos, VHS, CRP, transaminase glutâmico pirúvica, colesterol total e triglicerídeos, entre T0/T6, foi verificada no grupo teste pós-tratamento (p=0,028; p<0,001; p<0,001; p=0,010; p<0,001; p=0,015, respectivamente). Os resultados indicaram que a periodontite crônica severa está associada com níveis elevados de marcadores da inflamação e trombogênese, além de alterações no perfil lipídico em indivíduos sistemicamente saudáveis, podendo atuar como possível fator de risco para as doenças cardiovasculares. O tratamento periodontal não-cirúrgico mostrou-se eficaz na redução dos níveis dos marcadores sistêmicos da inflamação e na melhora do perfil lipídico em indivíduos com doença periodontal severa, consequentemente, reduzindo o risco de doenças cardiovasculares.

Wide field-of-view (FOV) microscopy is of high importance to biological research and clinical diagnosis where a high-throughput screening of samples is needed. This thesis presents the development of several novel wide FOV imaging technologies and demonstrates their capabilities in longitudinal imaging of living organisms, on the scale of viral plaques to live cells and tissues.

The ePetri Dish is a wide FOV on-chip bright-field microscope. Here we applied an ePetri platform for plaque analysis of murine norovirus 1 (MNV-1). The ePetri offers the ability to dynamically track plaques at the individual cell death event level over a wide FOV of 6 mm × 4 mm at 30 min intervals. A density-based clustering algorithm is used to analyze the spatial-temporal distribution of cell death events to identify plaques at their earliest stages. We also demonstrate the capabilities of the ePetri in viral titer count and dynamically monitoring plaque formation, growth, and the influence of antiviral drugs.

We developed another wide FOV imaging technique, the Talbot microscope, for the fluorescence imaging of live cells. The Talbot microscope takes advantage of the Talbot effect and can generate a focal spot array to scan the fluorescence samples directly on-chip. It has a resolution of 1.2 μm and a FOV of ~13 mm2. We further upgraded the Talbot microscope for the long-term time-lapse fluorescence imaging of live cell cultures, and analyzed the cells’ dynamic response to an anticancer drug.

We present two wide FOV endoscopes for tissue imaging, named the AnCam and the PanCam. The AnCam is based on the contact image sensor (CIS) technology, and can scan the whole anal canal within 10 seconds with a resolution of 89 μm, a maximum FOV of 100 mm × 120 mm, and a depth-of-field (DOF) of 0.65 mm. We also demonstrate the performance of the AnCam in whole anal canal imaging in both animal models and real patients. In addition to this, the PanCam is based on a smartphone platform integrated with a panoramic annular lens (PAL), and can capture a FOV of 18 mm × 120 mm in a single shot with a resolution of 100─140 μm. In this work we demonstrate the PanCam’s performance in imaging a stained tissue sample.
