La metáfora y otros mecanismos imaginativos subyacentes al pensa- miento y lenguaje humanos pueden ser utilizados en el discurso diario y especializado (Lakoff y Johnson 1980; Lakoff y Nuñez 2000). Asimismo pueden aparecer en la comunicación no verbal (Forceville y Urios-Aparisi 2009; Littlemore et al. Este volumen).Partiendo de estudios cognitivos y de la teoría de integración conceptual (Fauconnier 1997; Fauconnier y Turner 2002), este artículo examina la presencia de la metáfora en la ingeniería. Primeramente, se analiza un corpus lingüístico procedente de artículos de investigación de ingeniería civil. Los datos revelan el uso de la metáfora antropomórfica, sobre todo en expresiones relativas a la salud, como “diagnóstico”, “auscultación” o “proceso de curación”. Se exploran además ejemplos de ingeniería cuya fuente son proyecciones conceptuales corporales. Finalmente, abordamos la función de la metáfora visual bajo la teoría de integración conceptual mediante representaciones de ingeniería que evocan la anatomía humana o animal. Metaphor and other imaginative mechanisms that underlie human thought and language such as metonymy are used in everyday and specialised discourse (Lakoff and Johnson 1980; Lakoff and Nuñez 2000) They can also be involved in non- verbal forms of communication (Forceville and Urios-Aparisi 2009; Littlemore et al. this volume). Drawing on metaphor cognitive studies and on conceptual integration theory (Fauconnier 1997; Fauconnier and Turner 2002) this paper examines the occurrence of metaphor in engineering. First, we analyse results from a linguistic corpus formed by research papers from civil engineering journals. These data reveal the use of anthropomorphic metaphor, especially related to health or medical mappings such as “diagnosing”, “auscultation” or “curing”. Then, we explore how engineering notions are instantiated by bodily conceptual mappings according to conceptual integration theory. Finally, the function of visual metaphor is examined with conceptual integration theory by using engineering images evoking parts of human or animal anatomy.

La calidad del hormigón prefabricado se determina mediante ensayos de rotura a compresión en probetas transcurridos los 28 días de curado, según establece la EHE-08. Sin embargo, en la plantas de prefabricados es necesario además saber cuándo el hormigón está listo para ser procesado (destensado, cortado, trasladado), por lo que es necesario hacer ensayos de resistencia a la compresión entre las 48 y 72 horas, este tiempo se determina a partir de la experiencia previa adquirida y depende de las condiciones de cada planta. Si las probetas no han alcanzado el valor establecido, normalmente debido a un cambio en las condiciones climatológicas o en los materiales utilizados como el tipo de cemento o agregados, la solución adoptada suele ser dejar curar el material más horas en la pista para que alcance la resistencia necesaria para ser procesado. Si sigue sin alcanzarla, lo cual sucede muy ocasionalmente, se intenta analizar cuál ha sido el motivo, pudiéndose tirar toda la producción de ese día si se comprueba que ha sido un fallo en la fabricación de la línea, y no un fallo de la probeta. Por tanto, esta metodología de control de calidad, basada en técnicas destructivas, supone dos tipos de problemas, costes y representatividad. Los métodos no destructivos que más se han aplicado para caracterizar el proceso de curado del hormigón son los ultrasónicos y la medida de la temperatura como se recoge en la bibliografía consultada. Hay diferentes modelos que permiten establecer una relación entre la temperatura y el tiempo de curado para estimar la resistencia a compresión del material, y entre la velocidad de propagación ultrasónica y la resistencia. Aunque estas relaciones no son generales, se han obtenido muy buenos resultados, ejemplo de ello es el modelo basado en la temperatura, Maturity Method, que forma parte de la norma de la ASTM C 1074 y en el mercado hay disponibles equipos comerciales (maturity meters) para medir el curado del hormigón. Además, es posible diseñar sistemas de medida de estos dos parámetros económicos y robustos; por lo cual es viable la realización de una metodología para el control de calidad del curado que pueda ser implantado en las plantas de producción de prefabricado. En este trabajo se ha desarrollado una metodología que permite estimar la resistencia a la compresión del hormigón durante el curado, la cual consta de un procedimiento para el control de calidad del prefabricado y un sistema inalámbrico de sensores para la medida de la temperatura y la velocidad ultrasónica. El procedimiento para el control de calidad permite realizar una predicción de la resistencia a compresión a partir de un modelo basado en la temperatura de curado y otros dos basados en la velocidad, método de tiempo equivalente y método lineal. El sistema inalámbrico de sensores desarrollado, WilTempUS, integra en el mismo dispositivo sensores de temperatura, humedad relativa y ultrasonidos. La validación experimental se ha realizado mediante monitorizaciones en probetas y en las líneas de prefabricados. Los resultados obtenidos con los modelos de estimación y el sistema de medida desarrollado muestran que es posible predecir la resistencia en prefabricados de hormigón en planta con errores comparables a los aceptables por norma en los ensayos de resistencia a compresión en probetas. ABSTRACT Precast concrete quality is determined by compression tests breakage on specimens after 28 days of curing, as established EHE-08. However, in the precast plants is also necessary to know when the concrete is ready to be processed (slack, cut, moved), so it is necessary to test the compressive strength between 48 and 72 hours. This time is determined from prior experience and depends on the conditions of each plant. If the samples have not reached the set value, usually due to changes in the weather conditions or in the materials used as for example the type of cement or aggregates, the solution usually adopted is to cure the material on track during more time to reach the required strength for processing. If the material still does not reach this strength, which happens very occasionally, the reason of this behavior is analyzed , being able to throw the entire production of that day if there was a failure in the manufacturing line, not a failure of the specimen. Therefore, this method of quality control, using destructive techniques, involves two kinds of problems, costs and representativeness. The most used non-destructive methods to characterize the curing process of concrete are those based on ultrasonic and temperature measurement as stated in the literature. There are different models to establish a relationship between temperature and the curing time to estimate the compressive strength of the material, and between the ultrasonic propagation velocity and the compressive strength. Although these relationships are not general, they have been very successful, for example the Maturity Method is based on the temperature measurements. This method is part of the standards established in ASTM C 1074 and there are commercial equipments available (maturity meters) in the market to measure the concrete curing. Furthermore, it is possible to design inexpensive and robust systems to measure ultrasounds and temperature. Therefore is feasible to determine a method for quality control of curing to be implanted in the precast production plants. In this work, it has been developed a methodology which allows to estimate the compressive strength of concrete during its curing process. This methodology consists of a procedure for quality control of the precast concrete and a wireless sensor network to measure the temperature and ultrasonic velocity. The procedure for quality control allows to predict the compressive strength using a model based on the curing temperature and two other models based on ultrasonic velocity, the equivalent time method and the lineal one. The wireless sensor network, WilTempUS, integrates is the same device temperature, relative humidity and ultrasonic sensors. The experimental validation has been carried out in cubic specimens and in the production plants. The results obtained with the estimation models and the measurement system developed in this thesis show that it is possible to predict the strength in precast concrete plants with errors within the limits of the standards for testing compressive strength specimens.

In order to reduce costs and time while improving quality, durability and sustainability in structural concrete constructions, a widely used material nowadays, special care must be taken in some crucial phases of the project and execution, including the structure design and calculation, the dosage, dumping and curing of concrete: another important aspect is the proper design and execution of assembly plans and construction details. The framework, a name designating the whole reinforcement bars cage already assembled as shown in the drawings, can be made up of several components and implies higher or lower industrialization degree. The framework costs constitute about one third of the price per cubic meter placed in concrete works. The best solutions from all points of view are clearly those involving an easier processing to achieve the same goal, and consequently carrying a high degree of industrialization, meaning quality and safety in the work. This thesis aims to provide an indepth analysis of a relatively new type of anchoring by plate known as headed reinforcement bars, which can potentially replace standard or L-shaped hooks, improving the cleaning of construction details and enabling a faster, more flexible, and therefore a more economical assembly. A literature review on the topic and an overview of typical applications is provided, followed by some examples of specific applications in real projects. Since a strict theoretical formulation used to provide the design plate dimensions has not yet been put forward, an equation is proposed for the side-face blowout strength of the anchorage, based on the capacity of concrete to carry concentrated loads in cases in which no transverse reinforcement is provided. The correlation of the calculated ultimate load with experimental results available in the literature is given. Besides, the proposed formulation can be expanded to cases in which a certain development length is available: using a software for nonlinear finite element analysis oriented to the study of reinforced concrete, numerical tests on the bond-bearing interaction are performed. The thesis ends with a testing of eight corner joints subjected to a closing moment, held in the Structures Laboratory of the Polytechnic University of Madrid, aiming to check whether the design of such plates as stated is adequate for these elements and whether an element with plate-anchored reinforcement is equivalent to one with a traditional construction detail.

Esta tesis presenta los resultados de la investigación realizada sobre la inertización de cenizas volantes procedentes de residuos sólidos urbanos y su posterior encapsulación en distintas matrices de mortero. Durante el proceso de inertización, se ha logrado la inertización de éste residuo tóxico y peligroso (RTP) y también su valorización como subproducto. De esta forma se dispone de nueva “materia prima” a bajo coste y la eliminación de un residuo tóxico y peligroso con la consiguiente conservación de recursos naturales alternativos. La caracterización química de las cenizas analizadas refleja que éstas presentan altas concentraciones de cloruros, Zn y Pb. Durante la investigación se ha desarrollado un proceso de inertización de las cenizas volantes con bicarbonato sódico (NaHCO3) que reduce en un 99% el contenido en cloruros y mantiene el pH en valores óptimos para que la concentración de los metales pesados en el lixiviado sea mínima debido a su estabilización en forma de carbonatos insolubles. Se han elaborado morteros con cuatro tipos distintos de cementos (CEM-I, CEM-II, CAC y CSA) incorporando cenizas volantes inertizadas en una proporción igual a un 10% en peso del árido utilizado. Los morteros ensayados abarcan distintas dosificaciones tanto en la utilización de áridos con distintos diámetros (0/2 y 0/4), como en la relación cemento/árido (1/1 y 1/3). Se han obtenido las propiedades físicas y mecánicas de estos morteros mediante ensayos de Trabajabilidad, Estabilidad Dimensional, Carbonatación, Porosidad y Resistencias Mecánicas. De igual forma, se presentan resultados de ensayos de lixiviación de Zn, Pb, Cu y Cd, sobre probetas monolíticas de los morteros con los mejores comportamientos físico/mecánicos, donde se ha analizado el contenido en iones de dichos metales pesados lixiviados mediante determinación voltamperométrica de redisolución anódica Se concluye que todos los morteros ensayados son técnicamente aceptables, siendo los más favorables los elaborados con Cemento de Sulfoaluminato de Calcio (CSA) y con Cemento de Aluminato de Calcio (CAC). En este último caso, se mejoran las resistencias a compresión de los morteros de referencia en más de un 48%, y las resistencias a flexión en más de un 67%. De igual forma, los ensayos de lixiviado revelan la completa encapsulación de los iones de Zn y la mitigación en el lixiviado de los iones de Pb. Ambos morteros podrían ser perfectamente validos en actuaciones en las que se necesitase un producto de fraguado rápido, altas resistencias iniciales y compensación de las retracciones con una elevada estabilidad dimensional. En base a esto, el material podría ser utilizado como mortero de reparación en viales y pavimentos que requiriesen altas prestaciones, tales como: soleras industriales, pistas de aterrizaje, aparcamientos, etc. O bien, para la confección de elementos prefabricados sin armaduras estructurales, dada su elevada resistencia a flexión. ABSTRACT This dissertation presents the results of a research on inerting fly ash from urban solid waste and its subsequent encapsulation in mortar matrixes. The inerting of this hazardous toxic waste, as well as its valorization as a by-product has been achieved. In this way, a new "raw material" is available through a simple process and the toxic and hazardous waste is eliminated, and consequently, conservation of alternative natural resources is strengthened. Chemical analysis of the ashes analyzed shows high concentrations of soluble chlorides, Zn and Pb. An inerting process of fly ash with sodium bicarbonate (NaHCO3) has been developed which reduces 99% the content of chlorides and maintains pH at optimal values, so that the concentration of heavy metals in the leachate is minimum, due to its stabilization in the form of insoluble carbonates. Mortars with four different types of cements (CEM-I, CEM-II, CAC and CSA) have been developed by the addition of inertized fly ash in the form of carbonates, in the proportion of 10% in weight of the aggregates used. The samples tested include different proportions in the use of aggregates with different sizes (0/2 and 0/4), and in the cement/aggregate ratio (1/1 and 1/3). Physical/mechanical properties of these mortars have been studied through workability, dimensional stability, carbonation, porosity and mechanic strength tests. Leaching tests of Zn, Pb, Cu and Cd ions are also being performed on monolithic samples of the best behavioral mortars. The content in leachated heavy metal ions is being analyzed through stripping voltammetry determination. Conclusions drawn are that the tested CAC and CSA cement mortars present much better behavior than those of CEM-I and CEM-II cement. The results are especially remarkable for the CAC cement mortars, improving reference mortars compression strengths in more than 48%, and also bending strengths in more than 67%. Leaching tests confirm that the encapsulation of Zn and Pb is achieved and leachate of both ions is mitigated within the mortar matrixes. For the above stated reasons, it might be concluded that mortars made with calcium aluminate cements or calcium sulfoaluminate with the incorporation of treated fly ash, may be perfectly valid for uses in which a fast-curing product, with high initial strength and drying shrinkage compensation with a high dimensional stability is required. Based on this, the material could be used as repair mortar for structures, roads and industrial pavements requiring high performance, such as: industrial floorings, landing tracks, parking lots, etc. Alternatively, it could also be used in the manufacture of prefabricated elements without structural reinforcement, given its high bending strength.

El empleo de biomasa como combustible para la generación de bio-energía va en aumento en la actualidad, debido a su impacto medioambiental nulo en cuanto a las emisiones de CO2. Por lo tanto la generación de cenizas de biomasa, residuo de la producción de esta energía, constituye un problema medioambiental con un claro impacto social y económico. Este tipo de ceniza tiene contenidos en óxidos que la hacen atractiva para su empleo como sustituto parcial del cemento Portland, lo cual proporciona una salida eco-eficiente a este residuo, reduciendo al mismo tiempo la emisión de gases de efecto invernadero asociada a la fabricación del cemento. Esta investigación se centra en el desarrollo de nuevos e innovadores materiales base-cemento eco-eficientes que incorporan ceniza de biomasa para su aplicación integral en construcción. Para ello, se emplea una ceniza de biomasa (CB) procedente de un combustor de lecho fluidizado, cuya biomasa de combustión es principalmente restos de corteza de eucalipto, suministrada por el grupo ENCE-Navia (Asturias). El trabajo desarrollado en la presente tesis doctoral, tiene como primera fase la caracterización de esta ceniza y el análisis de viabilidad de su valorización en materiales base-cemento. Dentro de este análisis, se propone la activación de la ceniza CB mediante tratamiento hidrotermal (TH) en diferentes condiciones de medio activante, temperatura y tiempo de proceso, con el objetivo de favorecer la formación de fases hidratadas que potencien la valorización de la ceniza en el campo de los materiales de construcción. Como fase hidratada de interés se obtiene la fase tobermorita (Ca2.25(Si3O7.5(OH)1.5)(H2O)), precursora del gel C-S-H, responsable del desarrollo de resistencias mecánicas en los materiales base-cemento. El proceso de TH se optimiza para la síntesis más eficiente de esta fase. El estudio posterior de las propiedades mecánicas y micro-estructurales de pastas de cemento eco-eficientes que incorporan la ceniza CB y la ceniza tratada hidrotermalmente, CB-TH, confirma una mayor viabilidad de incorporación de la ceniza CB como sustituto parcial del cemento Portland. Como siguiente paso en el desarrollo de estos innovadores materiales base-cemento eco-eficientes se amplía el estudio multi-escalar de los materiales que incorporan CB mediante diferentes ensayos físico-mecánicos y de durabilidad. Los resultados indican que la presencia de la ceniza de biomasa no tiene efectos negativos sobre las propiedades físicas de los morteros eco-eficientes estudiados. Sin embargo, la adición de CB proporciona una mejor durabilidad del material al producir modificaciones de la microestructura que dificultan el transporte de agentes agresivos. Por otro lado, los morteros con un 10 y 20% de sustitución parcial de cemento por la ceniza de biomasa CB (CB-10 y CB-20) presentan una resistencia a compresión de 53.3 y 50.5 MPa a 28 días de curado, respectivamente. Estos morteros son comparables con un cemento Portland tradicional tipo CEM I de clase de resistencia 42.5 R. Por último, y con el fin de proporcionar la apertura de estos nuevos cementos eco-eficientes al mercado en el campo de los materiales de construcción, se estudian propiedades concretas relacionadas con diferentes tipos de aplicaciones. Concretamente se estudian en detalle las propiedades relativas a la aplicación en baldosas de mortero y los resultados indican unas prestaciones del material eco-eficiente con incorporación de CB similares o mejoradas con respecto al cemento Portland. Se analiza también la viabilidad de aplicación estructural de los cementos eco-eficientes desarrollados mediante el estudio de la adherencia al acero, que resulta similar a la del material de referencia. En cuanto a los resultados de extracción y caracterización de la fase acuosa de los poros, en todas las matrices eco-eficientes se obtiene un pH que garantiza la pasivación de la armadura. Sin embargo, el alto contenido en cloruros de dicha fase acuosa sugiere la conveniencia de realizar un análisis más detallado para la aplicación de los nuevos materiales eco-eficientes en hormigón armado. Se comprueba que todas las matrices que incorporan CB en porcentajes entre un 10 y un 90%, se pueden considerar adecuadas como nuevos materiales de construcción más eco-eficientes en aplicaciones con distintos niveles de exigencias mecánicas y sin problemas ambientales asociados con procesos de lixiviación. Con el presente trabajo de investigación se completan los objetivos iniciales de la tesis, con la obtención de nuevos e innovadores materiales base-cemento eco-eficientes que incorporan cenizas de biomasa (CB) con aplicación integral en el campo de la construcción. ABSTRACT The use of biomass as a fuel for the generation of bio-energy is increasing nowadays, due to its zero environmental impact in terms of CO2 emissions. Therefore the generation of biomass ash, a by-product of this energy, is an environmental problem with a clear social and economic impact. This type of ash contains oxides that make it attractive to be used as a partial replacement of Portland cement, providing an eco-efficient solution to this residue, while reducing the emission of greenhouse gases associated with the production of cement. The present research is focused on the development of new and innovative eco-efficient cement-based materials that incorporate biomass ash for their comprehensive application in construction. For this purpose a biomass ash (CB) is used from a fluidized bed forest combustor mainly fed with the bark of eucalyptus trees, provided by the ENCE-Navia (Asturias) group. The work includes in the first stage the characterization of the raw materials and the analysis of viability of their valorization in cement-based materials. Within this analysis, the activation of the ash is proposed by hydrothermal treatment (HT) in different conditions of activation medium, temperature and process duration, aiming an enhanced formation of hydrated phases to improve the ash valorization in the construction materials field. As an interesting hydrated phase, the tobermorite (Ca2.25(Si3O7.5(OH)1.5)(H2O)) is obtained from the process. This phase is considered as a precursor of the gel C-S-H, responsible for the development of mechanical strength in cement-based materials. HT process is optimized for the most efficient synthesis of tobermorite. The analysis of mechanical and microstructural properties of eco-efficient cement pastes incorporating CB ash and hydrothermally treated ash, CB-TH, confirms an improved viability of incorporation of CB ash as a partial replacement for Portland cement in the case. As a next step in the development of these innovative eco-efficient cement-based materials, a multiscale study of the materials that incorporate CB by different physical-mechanical and durability tests is carried out. The results indicate that the presence of biomass ash does not give rise to negative effects on the physical properties of the eco-efficient mortars analyzed. Nevertheless, the addition of CB produces a better durability performance due to microstructural modifications that hinder the transport of aggressive agents through the material. Moreover, mortars with a 10% and 20% of partial substitution of cement by the CB biomass ash (CB-10 and CB-20) show a compressive resistance of 53.3 and 50.5 MPa at 28 days of curing, respectively. These mortars are comparable to an ordinary Portland cement type CEM I with a resistance class of 42.5R. Finally, and in order to provide the opening of these new eco-efficient cement to the market in the field of construction materials, certain properties specifically related to different types of applications are studied. Among these, the properties concerning the application in mortar tiles are analyzed and the results indicate a similar, or even better performance of the eco-efficient mortar that incorporates CB, with respect to Portland cement. The viability of structural application of the developed eco-efficient cement is also performed considering the study of the adhesion to steel, with results similar to those of the reference material. Regarding the results of extraction and analysis of the aqueous phase of the pores, a pH value guaranteeing reinforcement passivation is obtained for all the eco-efficient matrices. However, high chloride content is obtained suggesting the suitability of a more detailed study to evaluate the application of these new eco-efficient materials in reinforced concrete. It is established that all the matrices incorporating CB in percentages between 10 and 90% may be considered adequate as new more eco-efficient construction materials in applications with different levels of mechanical demand and without environmental problems associated to leaching processes. In this research the initial objectives of the thesis are fulfilled by obtaining new and innovative eco-efficient cement-based materials that incorporate biomass ashes (CB) with comprehensive application in the construction field.

In adhesion, the wetting process depends on three fundamental factors: the surface topography of the adherend, the viscosity of the adhesive, and the surface energy of both. The aim of this paper is to study the influence of viscosity and surface roughness on the wetting and their effect on the bond strength. For this purpose, an acrylic adhesive with different viscosities was synthesized and some properties, such as viscosity and surface tension, were studied before adhesive curing took place. Furthermore, the contact angle and the lap-shear strength were analyzed using aluminum adherends with two different roughnesses. Scanning electron microscopy was used to determine the effect of the viscosity and the roughness on the joint interface. The results showed that the adhesive exhibits an optimal value of viscosity. Below this value, at low viscosities, the low neoprene content produces poor bond strength due to the reduced toughness of the adhesive. Additionally, it also produces a high shrinkage during curing, which leads to the apparition of residual stresses that weakens the interfacial strength. However, once the optimum value, an increase in the viscosity produces a negative effect on the joint strength as a result of an important decrease in the wettability.
Effect of nano-Si2O and nano-Al2O3 on cement mortars for use in agriculture and livestock production

The effect of nano-silica, nano-alumina and binary combinations on surface hardness, resistance to abrasion and freeze-thaw cycle resistance in cement mortars was investigated. The Vickers hardness, the Los Angeles coefficient (LA) and the loss of mass in each of the freeze–thaw cycles to which the samples were subjected were measured. Four cement mortars CEM I 52.5R were prepared, one as control, and the other three with the additions: 5% nano-Si, 5% nano-Al and mix 2.5% n-Si and 2.5% n-Al. Mortars were tested at 7, 28 and 90 d of curing to determine compression strength, total porosity and pore distribution by mercury intrusion porosimetry (MIP) and the relationship between the CSH gel and Portlandite total by thermal gravimetric analysis (TGA). The capillary suction coefficient and an analysis by a scanning electron microscope (SEM) was made. There was a large increase in Vickers surface hardness for 5% n-Si mortar and a slight increase in resistance to abrasion. No significant difference was found between the mortars with nano-particles, whose LA was about 10.8, classifying them as materials with good resistance to abrasion. The microstructure shows that the addition of n-Si in mortars refines their porous matrix, increases the amount of hydrated gels and generates significant changes in both Portlandite and Ettringite. This produced a significant improvement in freeze–thaw cycle resistance. The effect of n-Al on mortar was null or negative with respect to freeze–thaw cycle resistance.

The cytoplasmic heritable determinant [PSI+] of the yeast Saccharomyces cerevisiae reflects the prion-like properties of the chromosome-encoded protein Sup35p. This protein is known to be an essential eukaryote polypeptide release factor, namely eRF3. In a [PSI+] background, the prion conformer of Sup35p forms large oligomers, which results in the intracellular depletion of functional release factor and hence inefficient translation termination. We have investigated the process by which the [PSI+] determinant can be efficiently eliminated from strains, by growth in the presence of the protein denaturant guanidine hydrochloride (GuHCl). Strains are “cured” of [PSI+] by millimolar concentrations of GuHCl, well below that normally required for protein denaturation. Here we provide evidence indicating that the elimination of the [PSI+] determinant is not derived from the direct dissolution of self-replicating [PSI+] seeds by GuHCl. Although GuHCl does elicit a moderate stress response, the elimination of [PSI+] is not enhanced by stress, and furthermore, exhibits an absolute requirement for continued cell division. We propose that GuHCl inhibits a critical event in the propagation of the prion conformer and demonstrate that the kinetics of curing by GuHCl fit a random segregation model whereby the heritable [PSI+] element is diluted from a culture, after the total inhibition of prion replication by GuHCl.

Smokers have a significantly higher risk for developing coronary and cerebrovascular disease than nonsmokers. Advanced glycation end products (AGEs) are reactive, cross-linking moieties that form from the reaction of reducing sugars and the amino groups of proteins, lipids, and nucleic acids. AGEs circulate in high concentrations in the plasma of patients with diabetes or renal insufficiency and have been linked to the accelerated vasculopathy seen in patients with these diseases. Because the curing of tobacco takes place under conditions that could lead to the formation of glycation products, we examined whether tobacco and tobacco smoke could generate these reactive species that would increase AGE formation in vivo. Our findings show that reactive glycation products are present in aqueous extracts of tobacco and in tobacco smoke in a form that can rapidly react with proteins to form AGEs. This reaction can be inhibited by aminoguanidine, a known inhibitor of AGE formation. We have named these glycation products “glycotoxins.” Like other known reducing sugars and reactive glycation products, glycotoxins form smoke, react with protein, exhibit a specific fluorescence when cross-linked to proteins, and are mutagenic. Glycotoxins are transferred to the serum proteins of human smokers. AGE-apolipoprotein B and serum AGE levels in cigarette smokers were significantly higher than those in nonsmokers. These results suggest that increased glycotoxin exposure may contribute to the increased incidence of atherosclerosis and high prevalence of cancer in smokers.

Plasmid-encoded addiction genes augment the apparent stability of various low copy number bacterial plasmids by selectively killing plasmid-free (cured) segregants or their progeny. The addiction module of plasmid prophage P1 consists of a pair of genes called phd and doc. Phd serves to prevent host death when the prophage is retained and, should retention mechanisms fail, Doc causes death on curing. Doc acts as a cell toxin to which Phd is an antidote. In this study we show that host mutants with defects in either subunit of the ClpXP protease survive the loss of a plasmid that contains a P1 addiction module. The small antidote protein Phd is fully stable in these two mutant hosts, whereas it is labile in a wild-type host. We conclude that the role of ClpXP in the addiction mechanism of P1 is to degrade the Phd protein. This conclusion situates P1 among plasmids that elicit severe withdrawal symptoms and are able to do so because they encode both a cell toxin and an actively degraded macromolecule that blocks the synthesis or function of the toxin.

Toxoplasma gondii is a coccidian parasite with a global distribution. The definitive host is the cat (and other felids). All warm-blooded animals can act as intermediate hosts, including humans. Sexual reproduction (gametogony) takes place in the final host and oocysts are released in the environment, where they then sporulate to become infective. In intermediate hosts the cycle is extra-intestinal and results in the formation of tachyzoites and bradyzoites. Tachyzoites represent the invasive and proliferative stage and on entering a cell it multiplies asexually by endodyogeny. Bradyzoites within tissue cysts are the latent form. T. gondii is a food-borne parasite causing toxoplasmosis, which can occur in both animals and humans. Infection in humans is asymptomatic in more than 80% of cases in Europe and North-America. In the remaining cases patients present fever, cervical lymphadenopathy and other non-specific clinical signs. Nevertheless, toxoplasmosis is life threatening if it occurs in immunocompromised subjects. The main organs involved are brain (toxoplasmic encephalitis), heart (myocarditis), lungs (pulmonary toxoplasmosis), eyes, pancreas and parasite can be isolated from these tissues. Another aspect is congenital toxoplasmosis that may occur in pregnant women and the severity of the consequences depends on the stage of pregnancy when maternal infection occurs. Acute toxoplasmosis in developing foetuses may result in blindness, deformation, mental retardation or even death. The European Food Safety Authority (EFSA), in recent reports on zoonoses, highlighted that an increasing numbers of animals resulted infected with T. gondii in EU (reported by the European Member States for pigs, sheep, goats, hunted wild boar and hunted deer, in 2011 and 2012). In addition, high prevalence values have been detected in cats, cattle and dogs, as well as several other animal species, indicating the wide distribution of the parasite among different animal and wildlife species. The main route of transmission is consumption of food and water contaminated with sporulated oocysts. However, infection through the ingestion of meat contaminated with tissue cysts is frequent. Finally, although less frequent, other food products contaminated with tachyzoites such as milk, may also pose a risk. The importance of this parasite as a risk for human health was recently highlighted by EFSA’s opinion on modernization of meat inspection, where Toxoplasma gondii was identified as a relevant hazard to be addressed in revised meat inspection systems for pigs, sheep, goats, farmed wild boar and farmed deer (Call for proposals -GP/EFSA/BIOHAZ/2013/01). The risk of infection is more highly associated to animals reared outside, also in free-range or organic farms, where biohazard measure are less strict than in large scale, industrial farms. Here, animals are kept under strict biosecurity measures, including barriers, which inhibit access by cats, thus making soil contamination by oocysts nearly impossible. A growing demand by the consumer for organic products, coming from free-range livestock, in respect of animal-welfare, and the desire for the best quality of derived products, have all led to an increase in the farming of free-range animals. The risk of Toxoplasma gondii infection increases when animals have access to environment and the absence of data in Italy, together with need for in depth study of both the prevalence and genotypes of Toxoplasma gondii present in our country were the main reasons for the development of this thesis project. A total of 152 animals have been analyzed, including 21 free-range pigs (Suino Nero race), 24 transhumant Cornigliese sheep, 77 free-range chickens and 21 wild animals. Serology (on meat juice) and identification of T. gondii DNA through PCR was performed on all samples, except for wild animals (no serology). An in-vitro test was also applied with the aim to find an alternative and valid method to bioassay, actually the gold standard. Meat samples were digested and seeded onto Vero cells, checked every day and a RT-PCR protocol was used to determine an eventual increase in the amount of DNA, demonstrating the viability of the parasite. Several samples were alos genetically characterized using a PCR-RFLP protocol to define the major genotypes diffused in the geographical area studied. Within the context of a project promoted by Istituto Zooprofilattico of Pavia and Brescia (Italy), experimentally infected pigs were also analyzed. One of the aims was to verify if the production process of cured “Prosciutto di Parma” is able to kill the parasite. Our contribution included the digestion and seeding of homogenates on Vero cells and applying the Elisa test on meat juice. This thesis project has highlighted widespread diffusion of T. gondii in the geographical area taken into account. Pigs, sheep, chickens and wild animals showed high prevalence of infection. The data obtained with serology were 95.2%, 70.8%, 36.4%, respectively, indicating the spread of the parasite among numerous animal species. For wild animals, the average value of parasite infection determined through PCR was 44.8%. Meat juice serology appears to be a very useful, rapid and sensitive method for screening carcasses at slaughterhouse and for marketing “Toxo-free” meat. The results obtained on fresh pork meat (derived from experimentally infected pigs) before (on serum) and after (on meat juice) slaughter showed a good concordance. The free-range farming put in evidence a marked risk for meat-producing animals and as a consequence also for the consumer. Genotyping revealed the diffusion of Type-II and in a lower percentage of Type-III. In pigs is predominant the Type-II profile, while in wildlife is more diffused a Type-III and mixed profiles (mainly Type-II/III). The mixed genotypes (Type-II/III) could be explained by the presence of mixed infections. Free-range farming and the contact with wildlife could facilitate the spread of the parasite and the generation of new and atypical strains, with unknown consequences on human health. The curing process employed in this study appears to produce hams that do not pose a serious concern to human health and therefore could be marketed and consumed without significant health risk. Little is known about the diffusion and genotypes of T. gondii in wild animals; further studies on the way in which new and mixed genotypes may be introduced into the domestic cycle should be very interesting, also with the use of NGS techniques, more rapid and sensitive than PCR-RFLP. Furthermore wildlife can become a valuable indicator of environmental contamination with T. gondii oocysts. Other future perspectives regarding pigs include the expansion of the number of free-range animals and farms and for Cornigliese sheep the evaluation of other food products as raw milk and cheeses. It should be interesting to proceed with the validation of an ELISA test for infection in chickens, using both serum and meat juice on a larger number of animals and the same should be done also for wildlife (at the moment no ELISA tests are available and MAT is the reference method for them). Results related to Parma ham do not suggest a concerning risk for consumers. However, further studies are needed to complete the risk assessment and the analysis of other products cured using technological processes other than those investigated in the present study. For example, it could be interesting to analyze products such as salami, produced with pig meat all over the Italian country, with very different recipes, also in domestic and rural contexts, characterized by a very short period of curing (1 to 6 months). Toxoplasma gondii is one of the most diffuse food-borne parasites globally. Public health safety, improved animal production and protection of endangered livestock species are all important goals of research into reliable diagnostic tools for this infection. Future studies into the epidemiology, parasite survival and genotypes of T. gondii in meat producing animals should continue to be a research priority.

As metalo-β-lactamases (MBL) são capazes de hidrolisar os carbapenêmicos, a classe de antimicrobianos com maior potência para o tratamento de infecções graves e de maior uso clinico. Dentre as MBL, o grupo mais recentemente descrito e que apresentou rápida disseminação em todo o mundo é o da New-Delhi-Metalo- β-lactamases (NDM). Nas enterobactérias, os genes que codificam essas enzimas estão mais frequentemente localizados em plasmídeos. O estudo da estabilidade de plasmídeos que albergam o gene blaNDM-1 é importante para entender a predominância de espécies que carregam esses plasmídeos, desvendar mecanismos moleculares envolvidos na sua persistência e para desenvolver novas drogas que possam diminuir a sua persistência. Estudos recentes sobre estabilidade plasmidial evidenciaram que a maprotilina é capaz de induzir perda plasmidial de até 90% em E. coli K12. Neste trabalho, foi estudado o efeito da maprotilina na indução de cura de plasmídeos, que albergam o gene blaNDM-1, em diferentes espécies da família Enterobacteriaceae. Nove isolados pertencentes a diferentes espécies foram incluídas no estudo. Os plasmídeos foram caracterizados quanto ao seu tamanho por eletroforese e por sequenciamento de DNA no sistema Illumina. A persistência plasmidial foi determinada pelo método de contagem em placa em LB ágar com e sem tratamento com maprotilina em concentrações sub-inibitórias (50mg/L). O experimento foi conduzido por 10 dias, representando aproximadamente 100 gerações. Neste estudo evidenciou-se que o grupo das enterobactérias estão envolvidas na disseminação de plasmídeos com blaNDM-1, sendo que plasmídeos do grupo IncF estão mais relacionados a essa dispersão. A maprotilina teve efeito de cura plasmidial em todos os isolados exceto em E. hormaechei \"subsp. oharae\" e C. freundii. O isolado P. rettgeri apresentou maior taxa de perda plasmidial e a análise comparativa da sequência nucleotídica do plasmídeo indicou que a presença da IS5 pode estar relacionada com a diminuição da persistência plasmidial. Diferenças na persistência plasmidial, quando tratados com maprotilina, entre E. hormaechei \"subsp. steigerwaltii\" e E. hormaechei \"subsp. oharae\" sugerem que E. hormaechei \"subsp. oharae\" pode ser um possível disseminador de plasmídeos albergando blaNDM-1, devido a processos de adaptação co-evolutivos.
Painéis de partículas homogêneas cimento-bagaço de cana-de-açúcar curados por carbonatação acelerada

O presente estudo teve como objetivo produzir e avaliar o desempenho de painéis de partículas homogêneas de cimento-bagaço de cana-de-açúcar curados por carbonatação acelerada. Para atingir os resultados foram realizados ensaios de caracterizações morfológica e físico-química das partículas de bagaço de cana-de-açúcar, bem como ensaio de termometria para identificar a compatibilidade da matéria prima (bagaço) com o cimento. Os painéis de partículas cimento-bagaço produzidos foram submetidos a dois processos de cura distintos: 1- cura por 48 h em câmara climática, seguida por 24 h em ambiente com concentração de 15% ±0.6 de CO2, seguida por 24 dias em ambiente saturado ao ar; 2- cura em câmara climática por 48 h, seguida por 25 dias em ambiente saturado ao ar. Ao final dos 28 dias de cura e após ensaio de envelhecimento acelerado de imersão e secagem foram realizadas as caracterizações físico-mecânicas seguindo as recomendações das normativas DIN: 310; 322 e 323, bem como caracterização microestrutural e de condutividade térmica do painel de partículas cimento-bagaço. Os resultados obtidos indicaram que os painéis de partículas cimento-bagaço curados por carbonatação acelerada apresentaram melhor desempenho físico-mecânico quando comparados aos painéis não carbonatados, pois a carbonatação melhorou a interface entre as partículas e a matriz cimentícia, proporcionando maior adesividade entre as fases. E, além disso, reduziu o pH do meio alcalino em que as partículas de bagaço de cana-de-açúcar estão inseridas, minimizando o processo de degradação da lignina, celulose e hemicelulose.

A reciclagem profunda com espuma de asfalto tem sido uma alternativa de sucesso para a restauração de pavimentos degradados. Em relação às soluções tradicionais de reabilitação, como os recapeamentos, tem a vantagem de proporcionar a correção de defeitos em camadas inferiores, com a manutenção ou pequena elevação do greide da pista, além de ganhos ambientais, como um menor consumo de materiais virgens da natureza e redução do volume de material descartado. Entretanto, no Brasil não há método para dimensionamento estrutural para esta tecnologia, o que dificulta seu emprego. Para o desenvolvimento de um procedimento de dimensionamento que contemple este tipo de solução, foram estudados métodos presentes na bibliografia internacional: guia da AASHTO de 1993 e Caltrans, dos EUA, TRL386 e TRL611, da Inglaterra, as duas versões do guia sul-africano TG2 e os métodos oriundos do Austroads, tanto o procedimento interino de 2011 como adaptações de órgãos da Austrália e Nova Zelândia. Observou-se divergência de opiniões quanto ao comportamento do material reciclado com espuma de asfalto. Alguns órgãos e autores consideram o comportamento do mesmo mais próximo às misturas asfálticas, sendo o mecanismo de falha o trincamento, e outros o definem como semelhante a um material granular modificado com alta coesão e ruptura devido às deformações permanentes. Correlaciona-se tal associação ao teor de espuma usualmente utilizado nas obras rodoviárias. Outros aspectos que se destacam para este tipo de base são o ganho de resistência ao longo do tempo devido à cura, mesmo com início da operação da rodovia e a importância da infraestrutura remanescente no dimensionamento. Tais fatos foram corroborados pelos estudos de caso e resultados do trecho experimental construído na Rodovia Ayrton Senna - SP 070, monitorado por meio de ensaios deflectométricos com FWD durante um ano. Como resultado do trabalho, foi proposto um procedimento para o dimensionamento estrutural de pavimentos com camadas recicladas a frio com espuma de asfalto utilizando dados deflectométricos que atende o método do Manual de Pavimentação do Departamento Nacional de Infraestrutura de Transportes (DNIT) e incorpora diferentes aspectos na análise mecanicista. Outras conclusões são a viabilidade técnica a longo prazo da solução mencionada e a importância do controle tecnológico, com ênfase para o monitoramento deflectométrico nos primeiros meses de operação do pavimento para averiguar a evolução da cura do material.

La temperatura de curado, utilizada por la industria de fibrocemento como catalizador en la hidratación del cemento para lograr alta resistencia inicial en sus productos, genera la hipótesis de expansión causada por formación tardía de etringita. Para el planteamiento de dicha hipótesis, el siguiente estudio contempló dos grupos de muestras con el objetivo de estudiar aisladamente su comportamiento químico y físico: uno de los grupos conformado por pasta cementicia con 35% de sustitución de calcario, y el otro conformado por fibrocemento con incorporación de PVA y celulosa. Por calorimetría isotérmica se observó la cinética de reacción de dos pastas, una de ellas curada a 23 ºC y la otra a 85 ºC donde esta última experimentó un efecto catalizador en sus reacciones. Ensayos de DRX y DTG/TG mostraron la presencia de etringita a 23 °C durante todas las edades de hidratación exhibiendo una tendencia de aumento en la intensidad de los picos y en la cantidad, respectivamente, en función del tiempo; sin embargo, a 85 ºC la presencia de esta fase fue inconstante durante las primeras edades de hidratación, a partir de mes de hidratación mostró aumento en cada una de las siguientes medidas realizadas mensualmente; a 85 °C hubo una menor formación de portlandita en comparación con los resultados obtenidos a 23 °C. Utilizando el método Rietveld y análisis químico fueron determinadas las cantidades y las composiciones químicas, respectivamente, de las fases del cemento anhidro y del calcario para alimentar el programa GEMS y simular la hidratación de la pasta estudiada en laboratorio para un tiempo infinito bajo la influencia de la temperatura. Simulaciones de la hidratación de la pasta se aproximaron a los resultados reportados por la revisión bibliográfica permitiendo predecir la presencia de fases en función de la estabilidad termodinámica. Resultados de porosimetría por inyección de mercurio mostraron una mayor concentración de poros, asociados a defectos, en muestras curadas a 85°C con respecto a las de 23 °C. Finalmente, medidas de variación longitudinal para muestras de fibrocemento elaboradas en laboratorio y en fábrica presentaron dispersión en los resultados de expansión para cada uno de los escenarios sin poder correlacionar todos los datos con el aumento de masa exhibido por cada una de las muestras; no obstante, se resalta el riesgo de deterioro del fibrocemento por el aumento de defectos causado por la temperatura así como la creación de todas las condiciones necesarias que favorecen la formación tardía de etringita a partir del proceso de fabricación empleado para este producto.
