This work was focused on the analysis of transport, thermomechanical and electrochemical properties of a series of perovskite-like oxide materials and composites for potential applications as anodes of intermediate-temperature solid oxide fuel cells (SOFCs) with lanthanum gallate and silicate solid electrolytes. The primary attention was centered on A(Mn,Nb)O3-δ (A = Sr, Ca) and (La,Sr)(Mn,Ti)O3-based systems, lanthanum chromite substituted with acceptor-type and variable-valence cations, and various Ni-containing cermets. Emphasis was given to phase stability of the materials, their crystal structure, microstructure of porous electrode layers and dense ceramics, electronic conductivity, Seebeck coefficient, oxygen permeability, thermal and chemical induced expansion, and anodic overpotentials of the electrodes deposited onto (La,Sr)(Ga,Mg)O3- and La10(Si,Al)6O27- based electrolyte membranes. In selected cases, roles of oxygen diffusivity, states of the transition metal cations relevant for the electronic transport, catalytically active additives and doped ceria protective interlayers introduced in the model electrochemical cells were assessed. The correlations between transport properties of the electrode materials and electrochemical behavior of porous electrodes showed that the principal factors governing anode performance include, in particular, electronic conduction of the anode compositions and cation interdiffusion between the electrodes and solid electrolytes. The latter is critically important for the silicatebased electrolyte membranes, leading to substantially worse anode properties compared to the electrochemical cells with lanthanum gallate solid electrolyte. The results made it possible to select several anode compositions exhibiting lower area-specific electrode resistivity compared to known analogues, such as (La,Sr)(Cr,Mn)O3-δ.

Introdução: O padrão de recrutamento temporal inerente a uma sequência de ativação muscular (SAM), permite a organização multi-segmentar para a realização de uma tarefa motora. Este depende da conexão neural entre estruturas corticais e sub-corticais, incluindo os núcleos da base (NB), podendo, assim, estar comprometido em indivíduos com Doença de Parkinson (DP). As SAM poderão ser melhoradas através de uma intervenção baseada no conceito de Bobath (CB). Objetivo: Estudar o potencial da intervenção, baseada no CB, a longo prazo, nas SAM ao nível da tibio-társica (TT), durante as tarefas motoras sit-to-stand (SitTS) e o stand-to-sit (StandTS), em quatro indivíduos com DP. Metodologia: O estudo apresenta quatro casos de indivíduos com DP, que realizaram intervenção em fisioterapia baseada no CB, durante 12 semanas. Antes e após a intervenção, foram avaliadas as sequências de ativação do gastrocnémio medial (GM), do solear (SOL) e do tibial anterior (TA), durante as tarefas SitTS e StandTS, recorrendo à eletromiografia de superfície e à plataforma de forças, para a divisão cinética das diferentes fases das tarefas. Avaliou-se ainda o equilíbrio funcional, através da Escala de Berg, e a percepção subjetiva dos indivíduos acerca da sua capacidade para realizar atividades sem cair, através da Modified Falls Efficacy Scale. Resultados: Após a intervenção, os indivíduos em estudo apresentaram, na sua maioria, uma diminuição da co-ativação muscular, bem como um aumento do equilíbrio funcional e diminuição da probabilidade de risco de queda, refletindo uma melhoria do controlo postural (CP). As modificações na percepção subjetiva dos indivíduos acerca da sua capacidade para realizar atividades sem cair não foram homogéneas. Conclusão: A intervenção baseada no CB teve efeitos positivos do ponto de vista do CP nos quatro indivíduos com DP. Pensa-se que uma intervenção mais duradoura poderá intensificar as melhorias observadas.

Na região de Nisa, concelho de Portalegre, situa-se um dos maiores e mais relevantes jazigos de urânio alguma vez descoberto em território nacional. Trata-se de um jazigo ainda inexplorado que encerra um potencial estimado em cerca de 32 milhões de toneladas de minério tal qual. Este jazigo ocorre à superfície e encontra-se em contexto metassedimentar embutido em xistos mosqueados, pertencentes ao Grupo das Beiras, nas proximidades do contacto com o granito do Maciço de Nisa. Este trabalho tem como objetivo estudar de forma qualitativa e quantitativa a dispersão geoquímica provocada por processos naturais de erosão e de transporte hidrogravítico na envolvente à anomalia geoquímica natural localmente induzida por este jazigo. Para o efeito estabeleceu-se uma metodologia com as seguintes etapas principais: (1) georreferenciação em SIG de elementos de cartografia; (2) planeamento e elaboração do plano de amostragem; (3) recolha e tratamento de amostras de solos e sedimentos; (4) ensaios não destrutivos de medição de radiação gama por SPP2 e determinações analíticas por XRF; (5) análise exploratória e tratamento estatístico de dados e análise espacial; (6) análise de resultados; (7) definição de teores geoquímicos de fundo local. As amostras de sedimentos foram retiradas de uma ribeira que intersecta a área da anomalia e alimenta uma barragem local de enrocamento; as amostras de solos foram retiradas de linhas de amostragem perpendiculares à ribeira. As determinações analíticas registaram os teores em diversos metais, como o urânio, crómio, molibdénio, nióbio, vanádio e zinco e do semimetal arsénio. Com exceção do zinco, os resultados evidenciam que as concentrações naturais nestes metais no local onde se localiza uma importante jazida de minérios de urânio são muito elevados, e quando comparados com os valores standard da norma Canadiana mostram poder existir risco para a saúde se não forem limitados os usos do local.

Samples of volcanic rocks from Alboran Island, the Alboran Sea floor and from the Gourougou volcanic centre in northern Morocco have been analyzed for major and trace elements and Sr-Nd isotopes to test current theories on the tectonic geodynamic evolution of the Alboran Sea. The Alboran Island samples are low-K tholeiitic basaltic andesites whose depleted contents of HFS elements (similar to0.5xN-MORB), especially Nb (similar to0.2xN-MORB), show marked geochemical parallels with volcanics from immature intra-oceanic arcs and back-arc basins. Several of the submarine samples have similar compositions, one showing low-Ca boninite affinity. Nd-143/Nd-144 ratios fall in the same range as many island-arc and back-arc basin samples, whereas Sr-87/Sr-86 ratios (on leached samples) are somewhat more radiogenic. Our data point to active subduction taking place beneath the Alboran region in Miocene times, and imply the presence of an associated back-arc spreading centre. Our sea floor suite includes a few more evolved dacite and rhyolite samples with (Sr-87/Sr-86)(0) up to 0.717 that probably represent varying degrees of crustal melting. The shoshonite and high-K basaltic andesite lavas from Gourougou have comparable normalized incompatible-element enrichment diagrams and Ce/Y ratios to shoshonitic volcanics from oceanic island arcs, though they have less pronounced Nb deficits. They are much less LIL- and LREE-enriched than continental arc analogues and post-collisional shoshonites from Tibet. The magmas probably originated by melting in subcontinental lithospheric mantle that had experienced negligible subduction input. Sr-Nd isotope compositions point to significant crustal contamination which appears to account for the small Nb anomalies. The unmistakable supra-subduction zone (SSZ) signature shown by our Alboran basalts and basaltic andesite samples refutes geodynamic models that attribute all Neogene volcanism in the Alboran domain to decompression melting of upwelling asthenosphere arising from convective thinning of over-thickened lithosphere. Our data support recent models in which subsidence is caused by westward rollback of an eastward-dipping subduction zone beneath the westemmost Mediterranean. Moreover, severance of the lithosphere at the edges of the rolling-back slab provides opportunities for locally melting lithospheric mantle, providing a possible explanation for the shoshonitic volcanism seen in northern Morocco and more sporadically in SE Spain. The Mont Collon mafic complex is one of the best preserved examples of the Early Permian magmatism in the Central Alps, related to the intra-continental collapse of the Variscan belt. It mostly consists (> 95 vol.%) of ol+hy-nonnative plagioclase-wehrlites, olivine- and cpx-gabbros with cumulitic structures, crosscut by acid dikes. Pegmatitic gabbros, troctolites and anorthosites outcrop locally. A well-preserved cumulative, sequence is exposed in the Dents de Bertol area (center of intrusion). PT-calculations indicate that this layered magma chamber emplaced at mid-crustal levels at about 0.5 GPa and 1100 degrees C. The Mont Collon cumulitic rocks record little magmatic differentiation, as illustrated by the restricted range of clinopyroxene mg-number (Mg#(cpx)=83-89). Whole-rock incompatible trace-element contents (e.g. Nb, Zr, Ba) vary largely and without correlation with major-element composition. These features are characteristic of an in-situ crystallization process with variable amounts of interstitial liquid L trapped between the cumulus mineral phases. LA-ICPMS measurements show that trace-element distribution in the latter is homogeneous, pointing to subsolidus re-equilibration between crystals and interstitial melts. A quantitative modeling based on Langmuir's in-situ crystallization equation successfully duplicated the REE concentrations in cumulitic minerals of all rock facies of the intrusion. The calculated amounts of interstitial liquid L vary between 0 and 35% for degrees of differentiation F of 0 to 20%, relative to the least evolved facies of the intrusion. L values are well correlated with the modal proportions of interstitial amphibole and whole-rock incompatible trace-element concentrations (e.g. Zr, Nb) of the tested samples. However, the in-situ crystallization model reaches its limitations with rock containing high modal content of REE-bearing minerals (i.e. zircon), such as pegmatitic gabbros. Dikes of anorthositic composition, locally crosscutting the layered lithologies, evidence that the Mont Collon rocks evolved in open system with mixing of intercumulus liquids of different origins and possibly contrasting compositions. The proposed model is not able to resolve these complex open systems, but migrating liquids could be partly responsible for the observed dispersion of points in some correlation diagrams. Neuroblastoma (NB) is a neural crest-derived childhood tumor characterized by a remarkable phenotypic diversity, ranging from spontaneous regression to fatal metastatic disease. Although the cancer stem cell (CSC) model provides a trail to characterize the cells responsible for tumor onset, the NB tumor-initiating cell (TIC) has not been identified. In this study, the relevance of the CSC model in NB was investigated by taking advantage of typical functional stem cell characteristics. A predictive association was established between self-renewal, as assessed by serial sphere formation, and clinical aggressiveness in primary tumors. Moreover, cell subsets gradually selected during serial sphere culture harbored increased in vivo tumorigenicity, only highlighted in an orthotopic microenvironment. A microarray time course analysis of serial spheres passages from metastatic cells allowed us to specifically "profile" the NB stem cell-like phenotype and to identify CD133, ABC transporter, and WNT and NOTCH genes as spheres markers. On the basis of combined sphere markers expression, at least two distinct tumorigenic cell subpopulations were identified, also shown to preexist in primary NB. However, sphere markers-mediated cell sorting of parental tumor failed to recapitulate the TIC phenotype in the orthotopic model, highlighting the complexity of the CSC model. Our data support the NB stem-like cells as a dynamic and heterogeneous cell population strongly dependent on microenvironmental signals and add novel candidate genes as potential therapeutic targets in the control of high-risk NB.

In the southwestern part of the Aiguilles Rouges massif (pre-Alpine basement of the Helvetic realm, western Alps), a metavolcanic sequence, newly defined as the ``Greenstone Unit'',is exposed in two NS trending belts of several 100 metres in thickness. It consists of epidote amphibolites, partly epidote and/or calcic amphibole-bearing greenschists, and small amounts of alkali feldspar-bearing greenschists, which underwent low- to medium-grade metamorphism during Visean oblique collision. Metamorphic calcic amphiboles and epidotes show strong chemical zoning, whereas metamorphic plagioclase is exclusively albitic in composition (An 1-3). The SiO2 content of the subalkaline tholeiitic to calc-alkaline suite ranges continuously from 44 wt% to 73 wt%,but andesitic rocks predominate. The majority of samples have chemical compositions close to recent subduction-related lavas; some are even restricted to recent oceanic arcs (extremely low Ta and Nb contents, high La/Nb and Th/Ta ratios). But several basaltic to basalto-andesitic samples resemble continental tholeiites (low Th/Ta, La/Nb ratio). As it is very probable that both lava types are to some extent contemporaneous, it is proposed that the Greenstone Unit represents a former oceanic volcanic are which temporarily underwent extension during which emplacement of continental tholeiite-like rocks occurred. The cause of the extension remains ambiguous. Considering palaeotectonic significance and age of other metavolcanic units in the Aiguilles Rouges massif, the Greenstone Unit most likely formed in the Early Palaeozoic.

Norah and Fred Fisher welcomed John Fisher into the world on November 29, 1912, not knowing what an influential role he would play in shaping Canada's history. John Fisher grew up as the middle child of five brothers and sisters in Frosty Hollow, New Brunswick, close to today’s town of Sackville. Sackville’s main industry was the Enterprise Foundry which the Fisher family owned and operated; however, Fisher had no plans of going into the family business. He was more inspired by his maternal grandfather, Dr. Cecil Wiggins, who lived with the family after retiring from the Anglican ministry. Wiggins encouraged all his grandchildren to be well read and to take part in discussions on current events. There were often visitors in the Fisher household taking part in discussions about politics, religion, and daily life. Fisher forced himself to take part in these conversations to help overcome his shyness in social settings. These conversations did help with his shyness and also in forming many opinions and observations about Canada. It put Fisher on the road to becoming Mr. Canada and delivering the many eloquent speeches for which he was known. Fisher did not venture far from home to complete his first degree. In 1934 he graduated from Mount Allison University in Sackville, NB with an Arts degree. The same year Fisher enrolled in Dalhousie’s law school. During his time at Dalhousie, Fisher discovered radio through Hugh Mills. Mills or “Uncle Mel” was on CHNS, Halifax’s only radio station at the time. Fisher began by making appearences on the radio drama show. By 1941 he had begun writing and broadcasting his own works and joined the staff as an announcer and continuity writer. In 1936 the Canadian Broadcasting Corporation was formed, the first National radio station. Fisher joined the CBC shortly after it’s beginning and remained with them, as well as the Halifax Herald newspaper, even after his law school graduation in 1937. By 1943 Fisher’s talks became a part of the CBC’s programming for a group of maritime radio stations. Fisher once described his talks as follows “my talks weren’t meant to be objective. . . they were meant to be favourable. They were ‘pride builders’” He began his famed John Fisher Reports at CBC Toronto when he transfered there shortly after the war. This program brought emmence pride to the fellow Canadians he spoke about leading to approximately 3500 requests per year to speak at banquets and meeting throughout Canada and the United States. Fisher was a well travelled indivdual who would draw on personal experiences to connect with his audience. His stories were told in simple, straight forward language for anyone to enjoy. He became a smooth, dynamic and passionate speaker who sold Canada to Canadians. He became a renowned journalist, folk historian, writer and broadcaster. Fisher was able to reach a vast array of people through his radio work and build Canadian pride, but he did not stop there. Other ways Fisher has contributed to Canada and the Canadian people include: Honoured by five Canadian Universities. 1956, became the Director of the Canadian Tourist Association. 1961, was appointed Special Assistant to the Prime Minister of Canada. 1963, Commissioner of the Centennial Commission (the Federal Agency Responsible for Canada’s 100th birthday) 1968, received the Service Medal , a coveted Order of Canada. President of John Fisher Enterprises Ltd., private consultant work, specializing in Centennial planning, broadcasts, lectures and promotion. John Fisher continued recording radio broadcasts even after his diagnosis with cancer. He would record 3 or 4 at a time so he was free to travel across Canada, the U.S., Europe and Mexico in search of treatments. Fisher passed away from the disease on February 15, 1981 and he is buried at Mount Pleasant Cemetery in Toronto.

A generalization to the BTK theory is developed based on the fact that the quasiparticle lifetime is finite as a result of the damping caused by the interactions. For this purpose, appropriate self-energy expressions and wave functions are inserted into the strong coupling version of the Bogoliubov equations and subsequently, the coherence factors are computed. By applying the suitable boundary conditions to the case of a normal-superconducting interface, the probability current densities for the Andreev reflection, the normal reflection, the transmission without branch crossing and the transmission with branch crossing are determined. Accordingly the electric current and the differential conductance curves are calculated numerically for Nb, Pb, and Pb0.9Bi0.1 alloy. The generalization of the BTK theory by including the phenomenological damping parameter is critically examined. The observed differences between our approach and the phenomenological approach are investigated by the numerical analysis.

This study examined the bone mineral content (BMC) in young women with Adolescent Idiopathic Scoliosis (AIS), treated with a brace (27.9 ±21.6 months, for 18.0±5.4 h/d) during adolescence (AIS-B, n = 15, 25.6 ±5.8 yrs), versus women with AIS but no treatment (AIS-NB, n = 15, 24.0 ±4.0 yrs), and women without AIS (C, n = 19, 23.5 ±3.8 yrs). After controlling for lean body mass, calcium and vitamin D daily intake, and strenuous physical activity, femoral neck BMC was lower in the AIS-B compared with AIS-NB and C (all p’s < .05). In summary, women with AIS, braced during their growing years are characterized by low lower limb BMC. However, the lack of a relationship between brace treatment duration and BMC, suggests that bracing was not the likely mechanism.

Le neuroblastome (NB) représente 8% de tous les cancers pédiatriques et est caractérisé par sa grande hétérogénéité clinique. Afin d’évaluer son pronostic, plusieurs facteurs génétiques sont utilisés : amplification de MYCN, délétion 1p, gain 11q et gain 17q. Les buts de notre travail étaient d’abord de vérifier si l’hybridation in situ en fluorescence (FISH) permet une analyse complète de ces anomalies et ensuite, en utilisant une analyse globale du génome telle le polymorphisme nucléotidique simple (SNP), de vérifier la concordance avec les résultats de la FISH et le pronostic potentiel des anomalies du 14q, en particulier du gène AKT. Nous avons donc établi un panel de sondes pour la FISH qui a été appliqué sur 16 tumeurs non-fixées. Après isolation de l’ADN de 36 tumeurs, nous avons effectué une analyse génotypique par SNP utilisant les puces « Affymetrix Genome-Wide Human SNP Array 6.0 » contenant 945,826 sondes non polymorphiques et 906,000 sondes polymorphiques. Nos résultats ont démontré que la FISH permet l’évaluation complète des anomalies génétiques importantes du NB et que les anomalies déséquilibrées sont détectées très précisément par SNP. Les anomalies du 14q tendent à être associées avec des facteurs cliniques comme le grade et l’évolution, contrairement aux anomalies d’AKT. L’analyse du 14q a révélé trois gènes d’intérêt, MAX, BCL11B et GPHN, qui devraient être analysés sur un plus grand échantillon. Ainsi, l’étude par FISH semble adaptée pour détecter les anomalies génétiques classiques du NB, alors que celles retrouvées en 14q représentent de potentielles cibles thérapeutiques pour cette tumeur.

Le neuroblastome (NB) est la tumeur solide extracranienne la plus fréquente et mortelle chez les jeunes enfants. Il se caractérise par une résistance à la chimiothérapie possiblement en partie dû à la présence de cellules initiatrices de tumeurs (TICs). Des études ont mis en évidence le rôle de CD133 comme un marqueur des TICs dans divers types de cancers. Les buts de notre travail étaient d’abord de démontrer les vertus de TICs des cellules exprimant CD133 et ensuite, en utilisant une analyse globale du génome avec des polymorphismes nucléotidiques simples (SNPs), d’effectuer une analyse différentielle entre les TICs et les autres cellules du NB afin d’en identifier les anomalies génétiques spécifiques. Des lignées cellulaires de NB ont été triées par cytométrie de flux afin d’obtenir deux populations: une enrichie en CD133 (CD133high), l’autre faible en CD133 (CD133low). Afin de déterminer si ces populations cellulaires présentent des propriétés de TICs, des essais sur les neurosphères, les colonies en agar mou et les injections orthotopiques de 500 cellules sélectionnées dans 11 souris ont été réalisées. Après une isolation de l’ADN des populations sélectionnées, nous avons effectué une analyse génotypique par SNP utilisant les puces « Affymetrix Genome-Wide Human SNP Array 6.0 ». Pour vérifier l’expression des gènes identifiés, des Western Blots ont été réalisés. Nos résultats ont démontré que la population CD133 avait des propriétés de TICs in vitro et in vivo. L’analyse génotypique différentielle a permis d’identifier deux régions communes (16p13.3 and 19p13.3) dans la population CD133high ayant des gains et deux autres régions (16q12.1 and 21q21.3) dans la population CD133low possédant des pertes d’hétérozygoties (LOH). Aucune perte n’a été observée. Parmi les gènes étudiés, l’expression protéique d’éphrine-A2 était corrélée à celle de CD133 dans 6 tumeurs et 2 lignées cellulaires de NB. De plus, l’augmentation de la concentration d’anticorps anti-éphrine-A2 dans le milieu diminue la taille des neurosphères. Ainsi, la population CD133high, qui a des vertus de TICs, possède des caractéristiques génotypiques différentes par rapport à celle CD133low. La présence d’éphrine-A2 dans les cellules exprimant CD133 souligne son importance dans le développement des TICs. Ces résultats suggèrent la présence de potentielle cible pour de nouvelles thérapeutiques ciblant les TICs mise en évidence par l’étude génomique.

Le neuroblastome (NB) est la tumeur solide extracranienne la plus fréquente chez le jeune enfant. En dépit de plusieurs avancements thérapeutiques, seulement 60% survivront à long terme. Cette résistance aux traitements est possiblement due, en partie, à la présence des cellules souches cancéreuses (CSC). PARP-1 joue un rôle important dans la chimiorésistance de certaines tumeurs et son inhibition a montré une potentialisation des agents anticancéreux conventionnels. De plus, Bcl-2 est surexprimé dans le NB et son expression accrue contribuerait à la résistance à la chimiothérapie. Le but de notre travail était de déterminer les effets in vitro d’un PARP inhibiteur, AG-014699 (AG), et d’un inhibiteur de Bcl-2, Obatoclax (Obx), in vitro et in vivo, en monothérapie ou en combinaison avec de la Doxorubicine (Doxo) ou du Cisplatin (Cis), deux agents anticancéreux classiquement utilisés dans le traitement du NB. Afin de déterminer l’expression de PARP-1 dans les tumeurs de NB, nous avons analysé une cohorte de 132 tumeurs. Nous avons utilisé le test MTT afin d’évaluer la sensibilité de 6 lignées cellulaires de NB et des CSC à un traitement avec AG seul ou en combinaison avec de la Doxo ou du Cis. Nous avons déterminé l’étendue de la mort cellulaire par Annexin-V et caractérisé les dommages à l’ADN à l’aide d’un marquage γH2aX. De plus, les modulations des voies de signalisation intracellulaire ont été analysées par Western Blot. La sensibilité des cellules à l’Obx a été analysée par MTT sur 6 lignées cellulaires de NB et sa combinaison avec le Cis a également été déterminée dans 2 lignées cellulaires. Le marquage Annexin-V et des combinaisons avec ZVAD-FMK ont aussi été utilisés pour caractériser les effets d’Obx sur l’apoptose. Des expériences in vivo ont également été faites. Nos résultats démontrent que l’expression de PARP-1 est associée aux tumeurs moins agressives. AG n’a peu ou pas effet sur la croissance tumorale et ne potentialise pas significativement les effets de la Doxo ou de Cis. AG combiné à la Doxo semble sensibiliser les CSC dans une lignée cellulaire. L’Annexin-V et le marquage γH2aX ne révèlent pas d’effets synergiques de cette combinaison et les dommages à l’ADN et la mort cellulaire observés sont attribués à la Doxo. Cependant, on observe une augmentation d’apoptose et de bris d’ADN dans une lignée cellulaire (SK-N-FI) lorsqu’AG est utilisé en monothérapie. On observe une surexpression de pAKT et pERK suite à la combinaison Doxo et AG. Les cellules de NB sont sensibles à l’Obx à des concentrations à l’échelle nanomolaire. De plus, Obx active la mort cellulaire par apoptose. Aussi, Obx a un effet synergique avec le Cis in vitro. In vivo, l’Obx diminue significativement la taille tumorale. Nous concluons que l’Obx présente une avenue thérapeutique prometteuse dans le traitement du NB alors que l’utilisation d’AG ne semble pas être aussi encourageante.

Le neuroblastome (NB) est une tumeur fréquente et agressive du jeune enfant. Les tumeurs de haut grade de forme métastatique, généralement développées chez l'enfant de plus de 1 an, sont associées à une importante mortalité malgré un traitement lourd incluant une chimiothérapie à haute dose. La chimiorésistance est donc un problème majeur qui caractérise ces NB de haut grade. Une des hypothèses pour expliquer cette chimiorésistance est celle de l’activation de l’autophagie, un mécanisme auquel recourent les cellules afin de faire face aux situations de stress. D’ailleurs, plusieurs études ont démontré que l'autophagie était activée à la suite des thérapies anticancéreuses. Son inhibition pourrait donc représenter une stratégie thérapeutique très intéressante pour le traitement de cancers. Le but de ce projet de recherche a été de mettre en évidence l'importance de l'autophagie dans les cellules du NB ainsi que l'effet de son inhibition sur la sensibilité des cellules du NB à la chimiothérapie conventionnelle. La présence d'autophagie dans les cellules de NB et sa valeur pronostic ont été évaluées par une étude immunohistochimique et par western blot sur 184 tumeurs patient. Ensuite, dans le but de déterminer l'effet de la chimiothérapie conventionnelle sur le niveau d'autophagie, des études in vitro sur 6 lignées cellulaires ont été effectuées utilisant des tests de mesure d'autophagie (MDC, monodanylcadaverine), de viabilité cellulaire (MTT) et de western blot. Celles ci ont été suivies par des tests d'inhibition de l'autophagie par deux méthodes: l’inactivation du gène ATG5 par un lentivirus contenant un shRNA ciblant ATG5 ou de l'hydroxychloroquine (HCQ), un inhibiteur pharmacologique de l’autophagie. Cette inhibition a été testée seule ou en combinaison avec une chimiothérapie conventionnelle dans le but de déterminer le rôle de l'autophagie dans la sensibilisation des cellules de NB à la chimiothérapie. Ensuite, l’intérêt de l’inhibition de l’autophagie a été évalué sur des modèles murins. Enfin, le niveau d'autophagie a été testé dans des cellules souches de NB. Notre étude a démonté que l'autophagie est présente à un niveau basal dans une majorité des NB mais qu'elle ne représentait pas un facteur pronostic dans ce type de tumeur. Les différentes chimiothérapies testées induisent une augmentation de l'autophagie dans les cellules du NB. Les deux tests d'inhibition ont démontré in vitro que l'autophagie participe à la résistance des cellules aux traitements chimiothérapeutiques classiques du NB. Le blocage de l’autophagie in vivo augmente l’efficacité de la chimiothérapie, cependant certaines données associées au traitement par HCQ devront être complétées. Cette étude démontre que l'inhibition de l'autophagie en combinaison avec la chimiothérapie classique représente une approche thérapeutique prometteuse dans le traitement du NB.
