In dieser Arbeit wurde zunächst ein humanisiertes Mausmodell entwickelt für die Analyse von humanen DCs in vivo. Darüber hinaus wurden erste Versuche mit Nanopartikelbeladenen DCs durchgeführt, mit der Intention, durch diese Kombination humane DCs zu untersuchen. Es wurden immunsupprimierte NOD/LtSz-scid IL2R (NSG) Mäuse verwendet und mit humanen CD34+ PBSCs transplantiert. Es wurden insgesamt 14 Modelle getestet, mit einer durchschnittlichen Humanisierungsrate von 76 %. In allen Modellen konnten ab Woche sechs nach Transplantation humane CD45+ Zellen sowie humane Bund NK-Zellen und CD14+ Monozyten gefunden werden. Darüber hinaus waren myeloide DC-Vorläuferzellen, konventionelle HLA DR CD11c DCs (cDCs) und plasmazytoide DCs (pDCs) vorhanden. Humane T-Zellen konnten nicht vor Woche 18 nach Transplantation beobachtet werden. Neben der Rekonstitution humaner DCs in peripheren Organen, wurde ebenfalls nach gewebsständigen DCs, insbesondere den Langerhans Zellen (LCs) der Epidermis geschaut. Waren humane LC vorhanden, konnten diese ab Woche zwölf nach Transplantation in der murinen Epidermis detektiert werden. Diese waren konstant bis in Woche 30 nach Transplantation nachweisbar. In Hinblick auf die Etablierung der DCs in diesem humanisierten Mausmodells wurden verschiedene Einflussgrößen getestet. IL-7 führte zu keiner veränderten Hämatopoese, wohingegen Flt3L zu einer Zunahme von CD14+ Monozyten und cDCs führte. Darüber hinaus konnte eine drastische Abnahmernhumaner B-Zellen beobachtet werden. Es zeigte sich, dass der Zeitpunkt der Flt3LrnApplikation einen entscheidenen Faktor für den Effekt von Flt3L auf die Rekonstitution humaner Zellen darstellt. Für die in dieser Arbeit durchgeführten funktionellen in vivo Studien, wurden humanisierten Mäusen alloreaktive CD8+ T-Zellen appliziert. Somit sollte die Funktionalität der rekonstituierten humanen APCs getestet werden. Es wurde deutlich, dass Monozyten und DCs ihre Funktionalität erst ab Woche 14 nach Transplantation zu entwickeln schienen,rnwohingegen B-Zellen bereits zu früheren Zeitpunkten als Zielzellen für die alloreaktiven T-Zellen dienten. Dies wurde durch den Rückgang der jeweiligen Zellen nach Applikation der T-Zellen sichtbar. Zu erwähnen ist, dass das Anwachsen einer humanen Hämatopoese stark spenderabhängig ist und somit keine allgemeingültigen Aussagen hinsichtlich der in vivo Funktion getroffen werden können. Um im Gewebe verbliebende APCs zu manipulieren gibt es verschiedene Möglichkeiten. Im Rahmen dieser Arbeit wurden auf Polystyren-basierende Nanopartikel getestet. Die verwendeten Partikel hatten eine Größe von 80 bis 160 nm und waren unfunktionalisiert oder mit Amino- bzw. Carboxy-Gruppen versehen. Zusätzlich wurden die Partikel mit BODIPY (Durchflusszytometrie und kLSM-Messungen), einem Infrarotnahem Farbstoff IR 780 (BFI-Messungen) und Platin (in vivo Messungen) beladen. Der Carboxy-funktionalisierte Partikel zeigte den geringsten Einfluss auf die Vitalität von humanen DCs, wohingegen der Amino-funktionalisierte Partikel bei steigender Konzentration toxisch wirkte. Bei unfunktionalisierten Partikeln stieg die Toxizität bei zunehmender Konzentration. Hinsichtlich der Expression diverser DC spezifischer Oberflächenmoleküle nach Beladung mit Nanopartikeln zeigte sich, dass allein der unfunktionalisierte, mit Lutensol AT50 hergestellte Partikel zu einer leichten Hochregulation von MHC-Klasse-II Molekülen führte. Die Expression von CD86 wurde im Gegenzug nur durch die Beladung mit den Amino-, bzw. Carboxy funktionalisierten Partikeln und dem unfunktionalisierten, mit SDS hergestellten Partikel leicht gesteigert. Trotz der teilweise leicht veränderten Expression von Oberflächenmarkern, konnte mit Hilfe von IFN-g ELISpots keine Beeinflussungrnder Funktion als APCs von Nanopartikel-beladenen DCs beobachtet werden. In den in vivo Untersuchungen zeigten alle vier Partikel eine konstante Zirkulation imrnOrganismus und konnten bis 96 h nach Applikation nachgewiesen werden. Alle Partikel konnten primär in der Leber detektiert werden, wobei der unfunktionalisierte, mit Lutensol AT50 hergestelle Partikel das weiteste Verbreitungsmuster zeigte. Erste Versuche im humanisierten Mausmodell zeigten keine Beeinflussung der Verteilung und Kinetik von Nanopartikeln durch die humane Hämatopoese. Mit dem in dieser Arbeit etablierten humanisierten Mausmodell ist es möglich, die Entwicklung, Differenzierung, Aktivierung und Funktionalität humaner DCs in vivo zu untersuchen. Darüber hinaus kann das gezielte Adressieren von DCs in vivo analysiert werden, was sowohl die Möglichkeit der Manipulation von DCs zur Vermeidung einer akuten GvHD bietet als auch Verwendung in anderen DC-vermittelten Therapien (z.B.Vakzinationsstudien) findet.

Recurrent prostate cancer presents a challenge to conventional treatment, particularly so to address micrometastatic and small-volume disease. Use of α-radionuclide therapy is considered as a highly effective treatment in such applications due to the shorter range and exquisite cytotoxicity of α-particles as compared with β-particles. (213)Bi is considered an α-emitter with high clinical potential, due to its short half-life (45.6 minutes) being well matched for use in peptide-receptor radionuclide α-therapy; however, there is limited knowledge available within this context of use. In this study, two novel (213)Bi-labeled peptides, DOTA-PEG(4)-bombesin (DOTA-PESIN) and DO3A-CH(2)CO-8-aminooctanoyl-Q-W-A-V-G-H-L-M-NH(2) (AMBA), were compared with (177)Lu (β-emitter)-labeled DOTA-PESIN in a human androgen-independent prostate carcinoma xenograft model (PC-3 tumor). Animals were injected with (177)Lu-DOTA-PESIN, (213)Bi-DOTA-PESIN, or (213)Bi-AMBA to determine the maximum tolerated dose (MTD), biodistribution, and dosimetry of each agent; controls were left untreated or were given nonradioactive (175)Lu-DOTA-PESIN. The MTD of (213)Bi-DOTA-PESIN and (213)Bi-AMBA was 25 MBq (0.68 mCi) whereas (177)Lu-DOTA-PESIN showed an MTD of 112 MBq (3 mCi). At these dose levels, (213)Bi-DOTA-PESIN and (213)Bi-AMBA were significantly more effective than (177)Lu-DOTA-PESIN. At the same time, (177)Lu-DOTA-PESIN showed minimal, (213)Bi-DOTA-PESIN slight, and (213)Bi-AMBA marked kidney damage 20 to 30 weeks posttreatment. These preclinical data indicate that α-therapy with (213)Bi-DOTA-PESIN or (213)Bi-AMBA is more efficacious than β-therapy. Furthermore, (213)Bi-DOTA-PESIN has a better safety profile than (213)Bi-AMBA, and represents a possible new approach for use in peptide-receptor radionuclide α-therapy treating recurrent prostate cancer.

Brucella suis biovar 2 is the most common aetiological agent of porcine brucellosis in Europe. B. suis biovar 2 is considered to have low zoonotic potential, but is a causative agent of reproductive losses in pigs, and it is thus economically important. The multilocus variable-number of tandem repeats genotyping analysis of 16 loci (MLVA-16) has proven to be highly discriminatory and is the most suitable assay for simultaneously identifying B. suis and tracking infections. The aim of this study was to investigate the relatedness between isolates of B. suis biovar 2 obtained during a brucellosis outbreak in domestic pigs and isolates from wild boars and hares collected from proximal or remote geographical areas by MLVA-16. A cluster analysis of the MLVA-16 data revealed that most of the isolates obtained from Switzerland clustered together, with the exception of one isolate. The outbreak isolates constituted a unique subcluster (with a genetic similarity >93.8%) distinct from that of the isolates obtained from wild animals, suggesting that direct transmission of the bacterium from wild boars to domestic pigs did not occur in this outbreak. To obtain a representative number of isolates for MLVA-16, alternative methods of Brucella spp. isolation from tissue samples were compared with conventional direct cultivation on a Brucella-selective agar. We observed an enhanced sensitivity when mechanical homogenisation was followed by host cell lysis prior to cultivation on the Brucella-selective agar. This work demonstrates that MLVA-16 is an excellent tool for both monitoring brucellosis and investigating outbreaks. Additionally, we present efficient alternatives for the isolation of Brucella spp.

Toll-like receptors are pattern recognition receptors with which hosts recognize pathogen-associated molecular patterns (PAMP). This recognition process is translated rapidly into a meaningful defense reaction. This form of innate host defense is preserved in the animal kingdom: invertebrates heavily depend on it; higher vertebrates also have an adaptive immune system. Both adaptive and innate immune systems are intertwined in that the former also depends on an intact innate recognition and response system. Members of the TLR system cover recognition of parasitic, bacterial or viral germs. Due to the constraints imposed by the necessity to recognize PAMP and to interact with downstream signaling molecules, the TLR system is relatively conserved in evolution. Nevertheless, subtle species differences have been reported for several mammalian TLR members. Examples of this will be given. In all mammalian species investigated, part of the coding sequence is available for the most important TLR members, thus allowing study of expression of these TLR members in various tissues by reverse-transcription polymerase chain reaction in its classical (RT-PCR) and quantitative real time RT-PCR (qRT-PCR) form. In some species, the whole coding sequences of the most important or even all TLR members are known. This allows construction of cDNA and transfection of common host cells, thus permitting functional studies. Extensive investigations were devoted to the study of non-synonymous single nucleotide polymorphisms. In a few cases, expression of a given amino acid in the extracellular (ligand-binding) portion of TLR members could be associated with infectious diseases. This will be discussed below.

Professional veterinarians are one of the most affected professions when it comes to killing animals. However, in some situations the opinion about the acceptance of killing of animals differs between people, which can cause a dilemma for the executing person. In a pilot study based on questionnaires, veterinarians from different working fields and students of different branches stated their acceptance of killing of animals in diverse concrete situations. The result clearly demonstrates a higher acceptance of killing of animals among veterinarians with longtime experience in contrast to other groups and the almost same acceptance among agricultural students. The acceptance increased with age, however, we could not find a gender specific difference except of within a narrow age interval. The variability of acceptance within the same profession group differs between the situations. Veterinarians should be aware of their different thinking about killing of animals in some situations compared to other people and should know the reason of such differences. This is important not least to protect themselves and their opinion and to contribute to their societal responsibility by their veterinarian activity.

The commercial use of animal cloning for breeding food producing animals has been limited so far by biological and technical constraints such as adverse effects on the health and welfare of animals, especially high perinatal and postnatal disease and mortality of clones. However, the improvement of the technique may overcome those problems in future and contribute to the spread of cloning in agricultural production, which raises concern not only on health and welfare aspects but also on food safety and ethics. This may cause conflict in international trade. The present article reviews these topics on the basis of up-to-date scientific opinions.

Killing of animals is an important task to be performed by veterinarians. Killing decisions and their implementation often raise ethical questions. As a result of an interdisciplinary workshop targeting the subject "killing of animals" with veterinarians and ethicists, a three-dimensional dimension scheme was developed. Whereas the first two dimensions are focused on the animal's past and future life and are discussed with regard to life quality and life accomplishment (the "telos"), the third dimension incorporates the reason to kill and may integrate the concept of dignity. This form of dignity and the weighing of interests are applied to example scenarios and the resulting responsibilities of veterinarians and society are discussed.

The progress in molecular genetics in animal breeding is moderately effective as compared to traditional animal breeding using quantitative genetic approaches. There is an extensive disparity between the number of reported quantitative trait loci (QTLs) and their linked genetic variations in cattle, pig, and chicken. The identification of causative mutations affecting quantitative traits is still very challenging and hampered by the cloudy relationship between genotype and phenotype. There are relatively few reports in which a successful identification of a causative mutation for an animal production trait was demonstrated. The examples that have attracted considerable attention from the animal breeding community are briefly summarized and presented in a table. In this mini-review, the recent progress in mapping quantitative trait nucleotides (QTNs) are reviewed, including the ABCG2 gene mutation that underlies a QTL for fat and protein content and the ovine MSTN gene mutation that causes muscular hypertrophy in Texel sheep. It is concluded that the progress in molecular genetics might facilitate the elucidation of the genetic architecture of QTLs, so that also the high-hanging fruits can be harvested in order to contribute to efficient and sustainable animal production.

The etiology of brain tumors in children and adolescents is largely unknown, and very few environmental risk factors have been identified. The aim of this study was to examine the relationship between pre- or postnatal animal contacts or farm exposures and the risk of childhood brain tumors (CBTs), since infectious agents may pose a risk factor and a proposed mechanism is transferral of infectious agents from animals to humans.

Free-living amoebae (FLA) belonging to Acanthamoeba spp., Naegleria fowleri, Balamuthia mandrillaris, and Sappinia pedata are known to cause infections in humans and animals leading to severe brain pathologies. Worldwide, warm aquatic environments have been found to be suitable habitats for pathogenic FLA. The present study reports on screening for potentially pathogenic FLA in four hot spring resorts in Switzerland. Water samples were taken from water filtration units and from the pools, respectively. Amoebae isolated from samples taken during, or before, the filtration process were demonstrated to be morphologically and phylogenetically related to Stenoamoeba sp., Hartmannella vermiformis, Echinamoeba exundans, and Acanthamoeba healyi. With regard to the swimming pools, FLA were isolated only in one resort, and the isolate was identified as non-pathogenic and as related to E. exundans. Further investigations showed that the isolates morphologically and phylogenetically related to A. healyi displayed a pronounced thermotolerance, and exhibited a marked in vitro cytotoxicity upon 5-day exposure to murine L929 fibroblasts. Experimental intranasal infection of Rag2-immunodeficient mice with these isolates led to severe brain pathologies, and viable trophozoites were isolated from the nasal mucosa, brain tissue, and lungs post mortem. In summary, isolates related to A. healyi were suggestive of being potentially pathogenic to immunocompromised persons. However, the presence of these isolates was limited to the filtration units, and an effective threat for health can therefore be excluded.

BACKGROUND: Dogs experimentally inoculated with Angiostrongylus vasorum develop severe pulmonary parenchymal lesions and arterial thrombosis at the time of patency. HYPOTHESIS: A. vasorum-induced thrombosis results in arterial hypoxemia, pulmonary hypertension (PH), and altered cardiac morphology and function. ANIMALS: Six healthy Beagles experimentally inoculated with A. vasorum. METHODS: Thoracic radiographs and arterial blood gas analyses were performed 8 and 13 weeks postinoculation (wpi) and 9 weeks posttherapy (wpt). Echocardiography was done before and 2, 5, 8, 13 wpi and 9 wpt. Invasive pulmonary artery pressure (PAP) measurements were obtained 8 wpi. Two untreated dogs were necropsied 13 wpi and 4 treated dogs 9 wpt. RESULTS: All dogs had patent infections at 7 wpi and clinical respiratory signs at 8 wpi. Moderate hypoxemia (median PaO2 of 73 and 74 mmHg) present at 8 and 13 wpi had resolved by 9 wpt. Echocardiographically, no evidence of PH and no abnormalities in cardiac size and function were discernible at any time point. PAP invasively measured at 8 wpi was not different from that of control dogs. Severe radiographic pulmonary parenchymal and suspected thrombotic lesions at 13 wpi were corroborated by necropsy. Most histopathologic changes had resolved at 9 wpt, but focal inflammatory, thrombotic, and fibrotic changes still were present in all dogs. CONCLUSION: In experimentally infected Beagles, pulmonary and vascular changes induced by A. vasorum are reflected by marked radiographic changes and arterial hypoxemia. These did not result in PH and echocardiographic changes in cardiac size and function.

The search for an effective treatment for septic arthritis is ongoing. Current therapies are expensive since they require repeated joint lavage and long term antibiotic treatment. Local application of antimicrobial drugs is advantageous because high concentrations can be attained at the infection site, although repeated injections increase the risk of superinfection of the joint. Thus, slow release formulations, which have the advantage of local treatment yet single application of the drug, are appealing. Antibiotics used in slow release formulations are selected for tissue compatibility, an appropriate antibacterial spectrum, and stability both during the mixing procedure and within the carrier during the release period. Ideally the carriers should be bioresorbable. Promising reports on the clinical use of poly(methyl methacrylate) (PMMA) mixed with several different antibiotics, and of collagen sponges impregnated with gentamicin, should encourage the search for formulations optimally adapted to veterinary medical requirements.
