As características da digestão das forragens podem ser mais bem compreendidas quando complementadas com estudos de anatomia dos tecidos de plantas forrageiras. Com o objetivo de agregar novas informações ao tema, determinou-se a área dos diversos tecidos presentes na folha e no colmo de Brachiaria brizantha e B. humidicola, em três diferentes níveis de inserção no perfilho. O experimento foi conduzido em casa de vegetação, com plantas mantidas em vasos plásticos contendo areia esterilizada e recebendo solução nutritiva de macro e micronutrientes. Após 70 dias do corte de uniformização das parcelas (vasos), coletou-se o segmento do colmo e da folha (limbo e bainha) situado na porção mediana de cada um dos três diferentes níveis de inserção do perfilho estudado. As amostras foram emblocadas em GMA, seccionadas em micrótomo e o laminário fotomicrografado. As fotografias foram digitalizadas e as áreas de tecidos, mensuradas com software Image Tool. Observou-se que o limbo das folhas basilares apresentou menor área de tecido vascular lignificado. Maior área de tecido parenquimático foi observada na bainha e no colmo da B. brizantha. em relação à B. humidicola, a B. brizantha apresentou maior área de tecido vascular lignificado no limbo, bainha e colmo. Maior distância entre os feixes vasculares foi observada no limbo e na bainha, sendo o número de feixes maior no limbo e no colmo.

Microhabitat distribution was investigated in five populations of Characeae (two of Chara guairensis, two of Nitella subglomerata and one of Nitella sp.) to determine the distributional patterns, the morphometric and reproductive adaptations to varying environmental conditions and niche width on a scale of few centimeters. Variations in physical variables revealed some general trends of microhabitat distribution for the Characeae populations studied, with occurrence under the following conditions: slow and narrow current velocities; substrata predominantly composed of small particle size (sand-clay); variable and generally low depths. In terms of morphological adaptations, we found some general patterns: plants with longer whorl branchlets also had longer internodes in all populations studied, whereas longer plants had also thicker axis. The former were generally associated with higher biomass (percent cover). Few correlations of morphological characters were observed with environmental variables (e.g. plant length with irradiance: negative in two populations and positive in one population). Despite the general patterns of occurrence mentioned above, our results also indicated that each population differed in its responses to environmental variables and had particular morphological and reproductive adaptations. The Characean populations occurred under a narrower range of microhabitat conditions than other macroalgae from lotic habitats, particularly lower current velocity (6.7-9.8 cm s(-1)) and a more specific substratum type (sand-clay). Niche width values (0.60-0.99) of the Characeae populations studied indicate a high degree of habitat specialization and are among the highest yet found in lotic macroalgae. The relatively narrow variations in microhabitat conditions and high niche widths here reported for Characean populations, suggest a lower tolerance to variations in current velocity, depth, irradiance and substratum type. These characteristics probably explain the relatively restricted distribution of Characeae species in streams of S (a) over tildeo Paulo State with low frequency of records in most regions.

An improved on-site characterization of humic-rich hydrocolloids and their metal species in aquatic environments was the goal of the present approach. Both ligand exchange with extreme chelators ( diethylenetetraaminepentaacetic acid ( DTPA), ethylendiaminetetraacetic acid ( EDTA)) and metal exchange with strongly competitive cations (Cu(II)) were used on-site to characterize the conditional stability and availability of colloidal metal species in a humic-rich German bogwater lake ( Venner Moor, Munsterland). A mobile time-controlled tangential-flow ultrafiltration technique (cut-off: 1 kDa) was applied to differentiate operationally between colloidal metal species and free metal ions, respectively. DOC ( dissolved organic carbon) and metal determinations were carried out off-site using a home-built carbon analyzer and conventional ICP-OES ( inductively-coupled plasma-optical emission spectrometry), respectively. From the metal exchange equilibria obtained on-site the kinetic and thermodynamic stability of the original metal species ( Fe, Mn, Zn) could be characterized. Conditional exchange constants K ex obtained from aquatic metal species and competitive Cu(II) ions follow the order Mn > Zn >> Fe. Obviously, Mn and Zn bound to humic-rich hydrocolloids are very strongly competed by Cu( II) ions, in contrast to Fe which is scarcely exchangeable. The exchange of aquatic metal species (e.g. Fe) by DTPA/EDTA exhibited relatively slow kinetics but rather high metal availabilities, in contrast to their Cu(II) exchange.

Mosquito larvae are believed to be capable of digesting chitin, an insoluble polysaccharide of N-acetylglucosamine, for their nutritional benefit. Studies based on physiological and biochemical assays were conducted in order to detect the presence of chitinase activities in the gut of the detritus-feeding Aedes aegypti larvae. Larvae placed for 24 h in suspensions of chitin azure were able to digest the ingested chitin. Semi-denaturing PAGE using glycol chitin and two fluorogenic substrate analogues showed the presence of two distinct chitinase activities: an endochitinase that catalyzed the hydrolysis of chitin and an endochitinase that cleaved the short substrates [4MU(GlcNAc)(3)] and [4MU(GlcNAc)(2)] that hydrolyzed the chitobioside [4MU(GlcNAc)(2)]. The endochitinase had an extremely broad pH-activity against glycol chitin and chitin azure, pH ranging from 4.0 to 10.0. When the substrate [4MU(GlcNAc)(3)] was used, two activities were observed at pH ranges 4.0-6.0 and 8.0-10.0. Chitinase activity against [4MU(GlcNAc)(3)] was detected throughout the gut with the highest specific activity in the hindgut. The pH of the gut contents was determined by observing color changes in gut after feeding the larvae with color indicator dyes. It was observed a correlation between the pH observed in the gut of feeding larvae (pH 10-6.0) and the optimum pH for gut chitinase activities. In this work, we report that gut chitinases may be involved in the digestion of chitin-containing structures and also in the partial degradation of the chitinous peritrophic matrix in the hindgut. OBJECTIVE: Protein malnutrition is characterized by a number of morphologic and physiologic alterations, including intestinal mucosal atrophy and impaired nutrient absorption. Impaired absorption accentuates nutritional deficiency and accelerates body weight loss and changes in body chemistry. Because leucine is a ketogenic and oxidative amino acid and stimulates the protein synthesis, we examined the ability of young rats to recover from protein malnutrition by feeding them a control balanced or a leucine-rich diet for 60 d.METHODS: At the end of the 60-d period, body, liver, and muscle weights; glucose, methionine, and leucine intestinal absorption; and carcass chemical composition were evaluated.RESULTS: Body weight gain was higher in the control balanced and leucine-rich groups than in control rats, indicating that adequate refeeding allows body weight to recover in these groups. Methionine and glucose absorptions were impaired in malnourished rats but were restored after nutritional recovery. The leucine-rich diet resulted in an increase in carcass collagen nitrogen but maintained the carcass structural nitrogen.CONCLUSIONS: These results indicated that leucine supplementation during nutritional recovery from protein malnutrition improves protein carcass restoration. However, the precise mechanism of the leucine effects involved in this response remains to be elucidated.

Three procedures (line transect, point intercept and quadrat) for estimating abundance of macroscopic components of periphyton (macroalgae and bryophytes) were compared in two bedrock streams from southeastern Brazil on the basis of frequency and percent cover. In addition, two quadrat sizes (25 and 50 cm) were tested for best size. Samplings were all made within a 10 m length stream reach. Differences were tested by means of analysis of variance, ANOVA - one way, Student's t test and Chi-square test, whereas associations among them were evaluated by Pearson's r correlation coefficient. Values of frequency and percent cover varied among the different techniques and periphyton species in both sites but ANOVA, t and Chi-square tests revealed no significant difference (p<0.05) for percent cover in the two sites. No consistent pattern was observed for the different procedures between sites. Values for quadrat (25 and 50 cm) percent cover were positively correlated among distinct periphyton species. Significant differences for frequency were found in only one site. Quadrats of 50 cm produced the highest frequency values, whereas point intercept the lowest in both sites. In terms of spatial variation within the stream segment, significant differences were generally observed among all procedures, except between quadrats. As a rule, highly significant correlations for percent cover were found among the different techniques along the stream reach. The strongest relationships were found for quadrats with the other procedures, whereas the weakest between Line and point. Pros and cons of the three techniques are fully discussed.

OBJECTIVE: the aim of this study was to determine the effects of diets rich in saturated and polyunsaturated fatty acids on metabolic pathways and the relation of metabolic shifting to oxidative stress in cardiac tissue.METHODS: Male Wistar rats (age, 60 d; n = 10) were fed with a control low-fat diet, a diet rich in saturated fatty acids (SFAs), or a diet rich in polyunsaturated fatty acids (PUFAs). After 5 wk of treatment, sera were used for protein and lipid determinations. Protein, glycogen, triacylglycerol, lactate dehydrogenase, citrate synthase, beta-hydroxyacyl coenzyme-A dehydrogenase, catalase, glutathione peroxidase, superoxide dismutase, lipoperoxide, and lipid hydroperoxide were measured in cardiac tissue.RESULTS: the SFA group had higher triacylglycerol, cholesterol, low-density lipoprotein cholesterol, and atherogenic index (ratio of cholesterol to high-density lipoprotein) than did the PUFA and control groups. The PUFA group had low serum cholesterol, triacylglycerol, and low-density lipoprotein cholesterol as compared with the SFA group. SFA increased myocardial lipid hydroperoxide and diminished glutathione peroxidase. Despite the beneficial effects on serum lipids, the PUFA diet led to the highest levels of myocardial lipoperoxide and lipid hydroperoxide and diminished superoxide dismutase and catalase activities. The PUFA effects were related to increased feed efficiency, increased susceptibility to lipoperoxidation, and metabolic shifting in cardiac tissue. PUFA elevated triacylglycerol levels and decreased myocardial glycogen concentrations. The ratios of lactate dehydrogenase to citrate synthase and beta-hydroxyacyl coenzyme-A dehydrogenase to citrate synthase were increased, indicating myocardial reduction of tricarboxylic acid cycle.CONCLUSIONS: PUFAs have been recommended as a therapeutic measure in preventive medicine to lower serum cholesterol, but PUFAs increased oxidative stress in the heart by providing cardiac susceptibility to lipoperoxidation and shifting the metabolic pathway for energy production. The control diet, which was much lower in calories and fat, produced better overall clinical outcomes, better fat profiles, and less oxidative stress than did the diets rich in fatty acids.

Diffusive gradients in thin films (DGT) and tangential-flow ultrafiltration (TF-UF) were combined for fractionation of Al and Cu in river water containing high content of dissolved organic carbon. A procedure based on ultrafiltration data is proposed to determine diffusion coefficients of the analytes in water samples and model solutions containing both free metal (M) and complex (metal - humic substance). Aiming to evaluate the accuracy of the proposed approach, the DGT results were compared with those from a protocol for determination of labile Al and Cu based on solid phase extraction (SPE). Good agreement between data from DGT and SPE were attained for model solutions. For analysis of real organic-rich water samples, differences between DGT and SPE measurements were consistent with the time-scales of the techniques. The concentration of labile Al determined by DGT were lower than the total dissolved concentrations (determined by inductively coupled plasma mass spectrometry) and exceeded the ultrafiltered concentration, indicating that inorganic Al species (species small enough to pass through 1 kDa membrane) were minor species as compared with Al organic complexes. Microhabitat distribution was investigated in three populations of C. coeruleus to determine the distributional patterns and their controlling factors, as well as morphometric adaptations to varying conditions on a scale of a few centimetres. Morphometric variations and their relations with physical variables (current velocity, irradiance, depth and type of substratum) revealed some particular characteristics for each population and indicate particular adaptations. However, some trends were clear: 1) larger plants (length and/or diameter) produced a higher quantity of monosporangia in the three populations; 2) plant length and diameter were positively correlated in two populations; 3) plant diameter was positively correlated with current velocity in two populations; 4) higher percent cover was associated with substrata composed of macrophytes in two populations. C. coeruleus occurred under relatively wide microhabitat conditions and had high niche width values, suggesting a tolerance to considerable variation in physical variables. These characteristics contribute to the species' wide distribution in Brazilian streams, both spatial (at distinct scales) and seasonal. (C) ADAC / Elsevier, Paris.

The evolution of As excess in As-rich Ga1-xAsx films is analyzed for distinct As concentrations and different annealing temperatures. Initially the samples are amorphous and crystallize partially after thermal annealing. The formation of both amorphous and crystalline As clusters is examined by micro-Raman and X-ray diffraction analysis. When highly and moderately unbalanced materials are compared, differences are clearly observed concerning the crystallization temperature and the migration kinetics of the As excess. These differences are explained by the fort-nation of As precipitates around the GaAs crystallites in the moderately unbalanced material, contrasting with the migration of the As excess to the film surface in the highly unbalanced material.

1. This study aimed to link basic ethnobiological research on local ecological knowledge (LEK) to the conservation of Brazilian streams, based on two case studies: original data on LEK of fishermen about freshwater fish in the Negro River, Amazon, and previously published data about LEK of farmers on the ecological relationship between forest and streams in the Macabuzinho catchment, Atlantic Forest.2. Information was obtained from fishermen through interviews using standard questionnaires containing open-ended questions. Informants for interview were selected either following some defined criteria or applying the 'snowball' method.3. Fishermen's LEK about the diets and habitats of 14 fish species in the Negro River provided new biological information on plant species that are eaten by fish, in addition to confirming some ecological patterns from the biological literature, such as dependence of fish on forests as food sources.4. In the Atlantic Forest, a comparison between farmers' LEK and a rapid stream assessment in the farmers' properties indicated that farmers tended to overestimate the ecological integrity of their streams. Farmers recognized at least 11 forest attributes that correspond to the scientific concept of ecosystem services. Such information may be useful to promote or enhance dialogue among farmers, scientists and managers.5. These results may contribute to the devising of ecosystem management measures in the Negro River, aimed to conserve both rivers and their associated floodplain forests, involving local fishermen. In the Atlantic Forest, we proposed some initiatives, such as to allow direct economic use of their forests to conciliate conflicting perceptions of farmers about ecological benefits versus economic losses from reforestation. Despite their cultural, environmental and geographical differences, the two study cases are complementary and cost-effective and promising approaches to including LEK in the design of ecological research. Background and objective: The purpose of this study was to analyze histologically the influence of autologous platelet-rich plasma on bone healing in surgically created critical-size defects in rat calvaria.Material adn Methods: Thirty-two rats were divided into two groups: the control group (group C) and the platelet-rich plasma group. An 8-mm-diameter critical-size defect was created in the calvarium of each animal. In group C the defect was filled by a blood clot only. In the platelet-rich plasma group, 0.35 mL of platelet-rich plasma was placed in the defect and covered by 0.35 mL of platelet-poor plasma. Both groups were divided into subgroups (n = 8) and killed at either 4 or 12 wk postoperatively. Histometric (using image-analysis software) and histologic analyses were performed. The amount of new bone formed was calculated as a percentage of the total area of the original defect. Percentage data were transformed into arccosine for statistical analysis (analysis of variance, Tukey, p < 0.05).Results: No defect completely regenerated with bone. The platelet-rich plasma group had a statistically greater amount of bone formation than group C at both 4 wk (17.68% vs. 7.20%, respectively) and 12 wk (24.69% vs. 11.65%, respectively) postoperatively.Conclusion: Within the limits of this study, it can be concluded that platelet-rich plasma placed in the defects and covered by platelet-poor plasma significantly enhanced bone healing in critical-size defects in rat calvaria.
