Neuroinflammatory TNFα Impairs Memory via Astrocyte Signaling.

The occurrence of cognitive disturbances upon CNS inflammation or infection has been correlated with increased levels of the cytokine tumor necrosis factor-α (TNFα). To date, however, no specific mechanism via which this cytokine could alter cognitive circuits has been demonstrated. Here, we show that local increase of TNFα in the hippocampal dentate gyrus activates astrocyte TNF receptor type 1 (TNFR1), which in turn triggers an astrocyte-neuron signaling cascade that results in persistent functional modification of hippocampal excitatory synapses. Astrocytic TNFR1 signaling is necessary for the hippocampal synaptic alteration and contextual learning-memory impairment observed in experimental autoimmune encephalitis (EAE), an animal model of multiple sclerosis (MS). This process may contribute to the pathogenesis of cognitive disturbances in MS, as well as in other CNS conditions accompanied by inflammatory states or infections.

Mediastinite fibrosante: relato de caso

Neste trabalho é relatado o caso de uma mulher de 51 anos de idade, com dor torácica, dispneia e engurgitamento de vasos do tórax superior, sem evidências clínicas de doença granulomatosa. O estudo por imagem do tórax revelou massa mediastinal com calcificações, abordada por mediastinoscopia com biópsia. A análise histopatológica mostrou padrão inflamatório crônico sem etiologia definida, compatível com mediastinite fibrosante.

Aspectos tomográficos e anatomopatológicos da sarcoidose pulmonar

OBJETIVO: Analisar os aspectos radiológicos observados nas tomografias computadorizadas de alta resolução de pacientes com sarcoidose e fazer a correlação com os achados anatomopatológicos. MATERIAIS E MÉTODOS: Foram revistos os aspectos radiológicos observados nas tomografias computadorizadas de alta resolução de dez pacientes com sarcoidose e feita correlação com material obtido de biópsias cirúrgicas ou necrópsias de quatro desses pacientes. RESULTADOS: O aspecto mais frequentemente observado foi o de nódulos, com distribuição perilinfática, predominando ao longo das bainhas broncovasculares e da superfície pleural, com nódulos subpleurais e cissuras nodulares. Outros achados menos comuns foram as opacidades em vidro fosco e o espessamento de septos interlobulares. CONCLUSÃO: Em geral, todos esses achados corresponderam, anatomopatologicamente, ao acúmulo de granulomas nessas regiões.

Achados da tomografia computadorizada de alta resolução na histiocitose de células de Langerhans pulmonar

OBJETIVO: Caracterizar, por meio da tomografia computadorizada de alta resolução, as principais alterações pulmonares da histiocitose de células de Langerhans. MATERIAIS E MÉTODOS: Foram avaliadas, retrospectivamente, as tomografias computadorizadas de alta resolução de oito pacientes com diagnóstico comprovado da doença a partir de biópsia pulmonar a céu aberto, biópsia transbrônquica, estudos de imuno-histoquímica e/ou lesões extrapulmonares associadas. RESULTADOS: Pequenas lesões císticas, arredondadas e de paredes finas foram observadas em todos os pacientes. Nódulos, com distribuição predominantemente periférica no parênquima pulmonar, estavam presentes em 75% dos exames estudados. As lesões apresentaram distribuição difusa, com predomínio nos terços superior e médio dos pulmões em todos os casos, mas acometimento dos recessos costofrênicos foi observado em 25% dos pacientes. CONCLUSÃO: A comparação das tomografias computadorizadas de alta resolução com radiografias de tórax mostrou que cistos de paredes finas e pequenos nódulos não podem ser avaliados satisfatoriamente por radiografias convencionais. A tomografia computadorizada de alta resolução, por sua capacidade de detectar e caracterizar cistos e nódulos pulmonares, permite o diagnóstico de histiocitose de células de Langerhans pulmonar com alta probabilidade.

Cytokine-induced sleep: Neurons respond to TNF with production of chemokines and increased expression of Homer1a in vitro.

Interactions of neurons with microglia may play a dominant role in sleep regulation. TNF may exert its somnogeneic effects by promoting attraction of microglia and their processes to the vicinity of dendrites and synapses. We found TNF to stimulate neurons (i) to produce CCL2, CCL7 and CXCL10, chemokines acting on mononuclear phagocytes and (ii) to stimulate the expression of the macrophage colony stimulating factor (M-CSF/Csf1), which leads to elongation of microglia processes. TNF may also act on neurons by affecting the expression of genes essential in sleep-wake behavior. The neuronal expression of Homer1a mRNA, increases during spontaneous and enforced periods of wakefulness. Mice with a deletion of Homer1a show a reduced wakefulness with increased non-rapid eye movement (NREM) sleep during the dark period. Recently the TNF-dependent increase of NREM sleep in the dark period of mice with CD40-induced immune activation was found to be associated with decreased expression of Homer1a. In the present study we investigated the effects of TNF and IL-1β on gene expression in cultures of the neuronal cell line HT22 and cortical neurons. TNF slightly increased the expression of Homer1a and IL-1β profoundly enhanced the expression of Early growth response 2 (Egr2). The data presented here indicate that the decreased expression of Homer1a, which was found in the dark period of mice with CD40-induced increase of NREM sleep is not due to inhibitory effects of TNF and IL-1β on the expression of Homer1a in neurons.

Pulmonary tuberculosis in a patient with rheumatoid arthritis undergoig immunosuppressive treatment: case report

Rheumatoid arthritis is a disease which characteristically affects the joints. Because it is an autoimmune disease, immunosuppressive drugs are widely used in its treatment. The present case report illustrates the association of immunosuppressive treatment with the development of opportunistic infections in a 64-year-old patient.

D’altra banda la novamanera caravaggesca es contraria a la proposada per Annibale Carracci al programa dela Galeria Farnese i, alhora, per la defensada per tota la tractadística italiana del segleXVII, des de Giovanni Battista Agucchi, en el seu Tractat de la pintura(1607-1615),fins a Giovan Pietro Bellori en el seu discurs L’Idea del pittore, dello scultore edell’architetto, scelta dalle bellezze naturali superiore alla natura que seguint els vellsprincipis de la Poètica defensen que l’obra d’art ha de tenir versemblança, ha d’imitar els millors i pot corregir la realitat. No es estrany, doncs, que Agucchi posi comexemple a seguir l’art d’Annibale Carracci i que compari a Caravaggio amb l’escultorgrec Demetri, que va seguir tant al peu de la lletra la semblança que no va tenirconsideració per la bellesa.

Variação anual de metilxantinas totais em amostras de Ilex paraguariensis A. St. - Hil. (erva-mate) em Ijuí e Santa Maria, Estado do Rio Grande do Sul

Ilex paraguariensis is a native plant in Southern Brazil, whose leaves are utilized in beverages such as chimarrão and tererê. This study investigated variations of total methylxanthines in leaves from two Ilex populations collected at one-month intervals over the course of one year. The levels varied from 1.92 to 10.37 mg/g (Ijuí) and 1.77 to 9.17 mg/g (Santa Maria). Statistical analysis indicated a significative variation in the contents of all samples analysed, confirming seasonal fluctuations. These results show that the period of harvest might decisively influence the organoleptic characteristics of raw material used in industry and therefore in commercial products.

Development and physicochemical characterization of dexamethasone-loaded polymeric nanocapsule suspensions

The influence of drug concentration, oil phase, and surfactants on the characteristics of dexamethasone-loaded nanocapsules was investigated. The best formulations were obtained at dexamethasone concentrations of 0.25 and 0.50 mg.mL-1 (encapsulation efficiency: 80-90%; mean size: 189-253 nm). The type of oil phase influenced only the stability of dexamethasone-loaded nanocapsules. The association of polysorbate 80 and sorbitan monooleate provided a more stable formulation. Sunflower oil and sorbitan sesquioleate used for the first time as oil phase and surfactant for nanocapsules, respectively, have allowed obtaining suspensions with low mean size and narrow size distribution.

Atividade antiproliferativa de Sloanea garckeana K. Schum.

The crude methanol extract and hexane, ethyl acetate, chloroform and methanol fractions from leaves of S. garckeana were examined in vitro for their antiproliferative activity on MCF-7, NCI-ADR, NCI-460, UACC-62, 786-0, OVCAR-03, PCO-3 and HT-29 human cancer cell lines. Among the assayed fractions, the ethyl acetate fraction showed to be cytotoxic against 786-0, UACC-62, OVCAR-03 and NCI-ADR cell lines with IC50 values of 12 µg/mL, 42 µg/mL, 53 µg/mL and 51 µg/mL, respectively. Through fractionation and isolation procedures compound (1) was obtained from the EtOAc fraction and its structure was elucidated by spectral techniques.

Obtenção e avaliação de concentrados proteicos de corvina (Micropogonias furnieri) processados por extração química

The objective of this study was to obtain and evaluate physicochemical and functional properties of protein concentrates from Micropogonias furnieri produced by pH shifting process, using alkaline and acid solubilization followed by isoelectric precipitation of muscle proteins. The concentrates showed high protein content and the maxim solubility for the minced (96.5%), for the alkaline (97.5%) and acid (93.7%) extraction was obtained at pH 11.0. The water holding capacity of the alkaline concentrate resulted in a value same or superior to water holding capacity of the acid concentrate in all investigated values of pH. The oil holding capacity of alkaline and acid concentrates showed no significant difference between the studied processes.
