Background: Quantitative data from gene expression experiments are often normalized by transcription levels of reference or housekeeping genes. An inherent assumption for their use is that the expression of these genes is highly uniform in living organisms during various phases of development, in different cell types and under diverse environmental conditions. To date, the validation of reference genes in plants has received very little attention and suitable reference genes have not been defined for a great number of crop species including Coffea arabica. The aim of the research reported herein was to compare the relative expression of a set of potential reference genes across different types of tissue/organ samples of coffee. We also validated the expression profiles of the selected reference genes at various stages of development and under a specific biotic stress.Results: The expression levels of five frequently used housekeeping genes (reference genes), namely alcohol dehydrogenase (adh), 14-3-3, polyubiquitin (poly), beta-actin (actin) and glyceraldehyde-3-phosphate dehydrogenase (gapdh) was assessed by quantitative real-time RT-PCR over a set of five tissue/organ samples (root, stem, leaf, flower, and fruits) of Coffea arabica plants. In addition to these commonly used internal controls, three other genes encoding a cysteine proteinase (cys), a caffeine synthase (ccs) and the 60S ribosomal protein L7 (rpl7) were also tested. Their stability and suitability as reference genes were validated by geNorm, NormFinder and BestKeeper programs. The obtained results revealed significantly variable expression levels of all reference genes analyzed, with the exception of gapdh, which showed no significant changes in expression among the investigated experimental conditions.Conclusion: Our data suggests that the expression of housekeeping genes is not completely stable in coffee. Based on our results, gapdh, followed by 14-3-3 and rpl7 were found to be homogeneously expressed and are therefore adequate for normalization purposes, showing equivalent transcript levels in different tissue/ organ samples. Gapdh is therefore the recommended reference gene for measuring gene expression in Coffea arabica. Its use will enable more accurate and reliable normalization of tissue/organ-specific gene expression studies in this important cherry crop plant.

Previous tests with essential oils from ripe chiropterochoric fruits suggested they can be used to attract and capture fruit-eating bats inside forest remnants. Here we evaluated the efficiency of these oils to attract frugivorous bats to open areas. We performed field tests with artificial fruits impregnated with essential oils of the genera Piper or Ficus that were attached to two groups of mist-nets set 50 m outside the border of a forest remnant. One group of artificial fruits received the corresponding oil isolated through hydrodistillation and the other received water only. Fruits with oils attracted significantly more fruit-eating bats, especially Artibeus lituratus that regularly crosses open habitats to reach other forest remnants. The highly significant attraction of A. lituratus by the oil of Piper was unexpected, since this bat is a specialist on Ficus fruits. We hypothesize that in habitats with no fruit available it is possible to attract frugivorous bats with the odor of several ripe fruit species. Furthermore, we verified that almost half of the individuals captured defecated seeds, indicating that the oils also attract recently fed bats, even when their preferred food is available nearby. This technique potentially may increase seed rain at specific locations, being particularly promising to restoration projects.

We investigated effects of fruit colour (red, black or white), habitat (anthropogenic edges and forest interior) and fragment size on the removal of artificial fruits in semideciduous forests in south-east Brazil. Eight forest fragments ranging from 251 to 36,000 ha were used. We used artificial fruits, which were placed on shrubs between I and 2 m in height and checked after 48 and 96 h for peck marks in the fruits. All three variables affected the probability of consumption of our fruit models. Red and black fruits were statistically more pecked than the white fruits. The probability of fruit consumption was lower in the interior than at the edge and less in small than in large fragments. However, the decrease fruit consumption in small compared with large fragments was more accentuated for red and black fruits than for white fruits. Our results show that habitat reduction and edges affect the chances of a fruit being eaten by birds, which may ultimately affect plant fitness in forest fragments. (C) 2003 Elsevier B.V. Ltd. Three Latin American oilseeds obtained from native fruits: nopal (tuna) (Opuntia ficus-indica), cherimoya (chirimoya) (Annona cherimola), and papaya, Chilean variety (Carica pubescens or C. candamarcensis) were studied for their fatty acid composition and bioactive compounds, such as tocols and phytosterols, looking for new sources of special oilseeds for this region. The results indicated that each species represents an interesting possibility. Nopal oilseed is a good source of linoleic acid (62%), with a good balance between SFA and MUFA (1: 1.3). Cherimoya oilseed presents quite a different composition, with 24% SFA, 43% MUFA and 33% PUFA. Palmitic and stearic acids (15% and 7.6%, respectively) are the main SFA. A good balance between oleic acid (42.7%) and linoleic acid (31%) was observed. Papaya oilseed is a highly MUFA oil (72% with 71% oleic acid), with a very interesting composition, according to the new nutritional and technological recommendations.With respect to bioactive compounds, the main tocol in these three oilseeds was gamma-tocopherol, with 136, 300 and 317 mg/kg for cherimoya, papaya and nopal oilseeds, respectively. According to the total tocol content, papaya oilseed presented the highest value with 384 mg/kg. The total amount and distribution of phytosterols was different, with values of 3092, 3554 and 5474 mg/kg for nopal, cherimoya and papaya oilseeds, respectively, with beta-sitosterol forming 47.6%, 65.0 % and 78.7% of the total phytosterol fractions, respectively. From the results obtained, Chilean papaya oilseed can be considered as a very promising new source of special plant oil for different applications, followed by cherimoya and nopal oilseeds.

The present work reports the characterization of Fastuosain, a novel cysteine protease of 25kDa, purified from the unripe fruits of Bromelia fastuosa, a wild South American Bromeliaceae. Proteolytic activity, measured using casein and synthetic substrates, was dependent on the presence of thiol reagents, having maximum activity at pH 7.0. The present work reports cDNA cloning of Fastuosain; cDNA was amplified by PCR using specific primers. The product was 1096pb long. Mature fastuosain has 217 residues, and with the proregion has a total length of 324 residues. Its primary sequence showed high homology with ananain(74%), stem bromelain (66%) and papain (44%).

Before 1989 all braconid wasps were thought to be parasitoids, but in that year the first phytophagous species was reported. Subsequently, a few other examples of phytophagy have been discovered, most of which are species of Allorhogas in the subfamily Doryctinae. Until now, all demonstrated examples of phytophagy in this genus have been as gall inducers in the fruits of Fabaceae. Here we describe a new species from Costa Rica, Allorhogas conostegia Marsh and Shaw, and provide evidence that it forms galls in the fruits of Conostegia xalapensis (Melastomataceae). We also provide information on the phenology of the plant and of the galls and the effects of the galls on the host plant, and we discuss the potential species richness of Allorhogas in the Neotropics.

Testes em placas de CCDC reveladas com solução de beta-caroteno mostraram a presença de duas substâncias com propriedades antioxidantes no extrato hexânico. Elas foram isoladas e identificadas como 3-metil-sargacromenol (1) and sargacromenol (2). O extrato hexânico forneceu ainda o ácido 3-metil-sargaquinóico (3) e o ácido sargaquinóico (4). O comportamento eletroquímico destas substâncias foi investigado em CH2Cl2/Bu4NBF4 sobre eletrodo de carbono vítreo. A oxidação do grupo fenólico nos tocotrienóis 1 and 2 é responsável por um pico anódico em potenciais de +0.23V e +0.32V, os quais são correlacionados com sua atividade antioxidante. O mecanismo de oxidação é comparado com o comportamento eletroquímico do antioxidante alfa-tocopherol.

This study investigated the composition and antifungal activity against Cladosporium sphaerospermum and Cladosporium cladosporioides of essential oils of leaves of Piper cernuum, Piper diospyrifolium, Piper crassinervium, Piper solmsianum and Piper umbelata and fruits of P. cernuum and P. diospyrifolium. The essentials oils were analyzed by GC-MS and submitted of the antifungal activity tests. The essential oils of fruits from P. cernuum and leaves of P. crassinervium and P. solmsianum showed potential antifungal activity against C. sphaerospermum and C. cladosporioides. In addition, this is the first report of the composition of essential oils of fruits of P. cernuum and P. diospyrifolium.
