The incidence of the trichothecenes deoxynivalenol (DON), diacetoxyscirpenol (DAS), and T2 toxin (T2) in popcorn was investigated in 90 samples, belonging to 2 commercial and 28 experimental hybrids planted at experimental stations of the Agronomic Institute of Campinas at the locations of Campinas, Mococa, and Capão Bonito, and in 15 samples of popcorn, 9 branded and 6 unbranded, acquired from commercial outlets in the city of Campinas, SP. The samples were analyzed by gas chromatography with flame ionization detector combined with an alumina:carbon cleanup column. The detection limits were 30ng/g for DON, 50ng/g for DAS, and 40ng/g for T2. Five samples were contaminated with DON, four of them commercial and one from an experimental cultivar. The level of contamination in the commercial samples ranged from 30 to 40ng/g. The sample from the experimental cultivar contained 770ng/g DON. DAS and T2 were not detected in any of the popcorn samples analyzed.

Aim and design: To evaluate family-based health counseling for young children, and to study the significance of adding parental self-care or the training of professionals to the programs. The effectiveness and acceptability of the programs were evaluated by comparing two new programs with an earlier one. Subjects and methods: The study was carried out in Vantaa, which was divided into three study areas. The subjects consisted of children born in 2008, particularly fi rstborn children, while children born in 2006 formed the historical control. The fi rst of the new programs emphasized oral hygiene and use of fl uoride, and the second program focused on proper diet and use of xylitol. The main outcome measure was mutansstreptococci (MS) in the dental biofi lm of two-year-olds, and the opinions of parents and dental professionals were evaluated using questionnaires. Results: The programs found wide acceptance among dental professionals. There were no group-related differences found in the MS scores of the two-year-olds. However, all groups combined, father’s advanced level of education and child’s proper use of xylitol were associated with negative MS scores. In the opinion of parents, the oral healthcare guidance at least somewhat met their expectations. Conclusions: The present fi ndings suggest that providing training and support for professionals in health education is important. The addition of parental self-care to supplement programs aimed at young children does not improve the program, although it may improve parental readiness to change their own health habits. Counseling for families might be best carried out through a routine patient-centered program.

Antimicrobial activities of two commercial disinfectants, alone or combined with heat, against three Salmonella strains and three Listeria monocytogenes strains were studied. The efficacy of disinfectants against planktonic bacteria and bacteria attached to three food contact industrial surfaces (stainless steel, polytetraflourethylene, and rubber) was investigated. The tests were conducted using the sanitizer (quaternary ammonium compounds, and alquyldiethylenediamineglycine and di-alquyldiamineethylglycine) concentrations recommended by the manufacturers, and concentrations twice and four times higher than those values. The recommended concentrations were not effective to kill bacteria, especially when they were attached to surfaces. Concentrations of disinfectants twice and four times higher than those recommended were needed to fully eliminate planktonic bacteria. These same sanitizer concentrations were not sufficient to remove attached bacteria. To remove them from the surfaces, a treatment with recommended concentrations in combination with heat was needed. Our results indicate that these two pathogenic bacteria could survive common sanitation programs used in the food industry.

A utilização das popularmente chamadas, sementes crioulas de soja ou sementes salvas pelos produtores é uma constante no Centro-Oeste Brasileiro. Este trabalho teve o objetivo de avaliar a qualidade das sementes de soja produzidas, colhidas e armazenadas por uma empresa produtora de sementes, e das sementes utilizadas pelos produtores de grãos, produzidas sem seguir os procedimentos técnicos de produção de sementes. As parcelas conduzidas para produção de sementes comerciais, foram retiradas do campo logo que atingiram a maturação de campo com umidade de 16,3%, receberam quatro aplicações de inseticidas durante o seu ciclo, para o controle de percevejos e, foram colhidas com colhedora de fluxo axial com rotação de cilindro de 500 rpm, enquanto as outras parcelas foram retiradas do campo com umidades de 14,7 e 12,3%, receberam apenas três aplicações de inseticida e, foram colhidas com colhedora de cilindro côncavo com velocidade de rotação do cilindro de 700 rpm. Após a colheita foram utilizados diferentes processos de secagem, beneficiamento e armazenamento para as sementes comerciais e para as sementes salvas. No início e no final do armazenamento, todas as unidades experimentais foram analisadas com os seguintes testes: de germinação, de tetrazólio 1-8 , do pH do exsudato, de envelhecimento acelerado, de desenvolvimento de plântula, de tetrazólio 1-3 e de 1º contagem do teste de germinação. Os resultados permitem concluir que: sementes comerciais, produzidas de acordo com as recomendações técnicas de produção de sementes, apresentam qualidade fisiológica superior; a qualidade fisiológica de materiais conduzidos como grãos é reduzida com o armazenamento, impedindo sua utilização como sementes para formação de lavouras de grãos comerciais.

Atherosclerosis is a chronic and progressive disease of the vasculature. Increasing coronary atherosclerosis can lead to obstructive coronary artery disease (CAD) or myocardial infarction. Computed tomography angiography (CTA) allows noninvasive assessment of coronary anatomy and quantitation of atherosclerotic burden. Myocardial blood flow (MBF) can be accurately measured in absolute terms (mL/g/min) by positron emission tomography (PET) with [15O] H O as a radiotracer. We studied the coronary microvascular dysfunction as a risk factor for future coronary calcification in healthy young men by measuring the coronary flow reserve (CFR) which is the ratio between resting and hyperemic MBF. Impaired vasodilator function was not linked with accelerated atherosclerosis 11 years later. Currently, there is a global interest in quantitative PET perfusion imaging. We established optimal thresholds of [15O] H O PET perfusion for diagnosis of CAD (hyperemic MBF of 2.3 mL/g/min and CFR of 2.5) in the first multicenter study of this type (Turku, Amsterdam and Uppsala). In myocardial bridging a segment of the coronary artery travels inside the myocardium and can be seen as intramural course (CTA) or systolic compression (invasive coronary angiography). Myocardial bridging is frequently linked with proximal atherosclerotic plaques. We used quantitative [15O] H O PET perfusion to evaluate the hemodynamic effects of myocardial bridging. Myocardial bridging was not associated with decreased absolute MBF or increased atherosclerotic burden. Speckle tracking allows quantitative echocardiographic imaging of myocardial deformation. Speckle tracking during dobutamine stress echocardiography was feasible and comparable to subjective wall motion analysis in the diagnosis of CAD. In addition, it correctly risk stratified patients with multivessel disease and extensive ischemia.

The frequency and type of agonistic displays involved in male-male encounters should be significantly influenced by the presence of females. Discrete agonistic displays vary in energy expenditure and risk, and therefore should be dependent on available resources. The influence of live females and the scent of females, on the frequency of male agonistic displays was observed in a laboratory terrarium using the field cricket Gryllus bimaculatus. The effect of energy constraints on display frequency was also determined. Half the males were fed a diet high in protein and fet; the other males were fed a lower quality diet, for a 7-11 day period. The frequency of five individual displays and mating frequency were recorded using an Event Recorder and notebook. Each group of males was presented with three experimental conditions, over three days, involving the presence or absence of live females and female scent. The presence of females elicited an increase in all displays except antennation; female scent increased the frequency of antennations, mandible flares and grapples, but to a lesser extent than did live females. The frequency of grapples significantly increased for males fed the high quality diet; however diet did not influence the other displays. The combined influence of diet and condition was significant for mandible flare only. Mating frequency was not influenced by diet. However, the frequency ofthe displays were positively correlated with mating frequency for high quality fed males. Escalated displays involving high costs, such as grapple and mandible flare, increased in frequency when the benefits of winning contests were high in G.bimaculatus. Escalation to grapple behaviour was less evident for males fed the lower quality diet as this imposed energy constraints on high cost displays.

The first objective of this study was to identify appropriate sensory descriptors to assess the astringent sub-qualities of red wine. The influence of pH and ethanol on the sensation of astringency in red wine was evaluated, using a de-alcoholized red wine. A portion of the wine was adjusted to the pH values of 3.2, 3.4, 3.6 and 3.8, and another portion was adjusted to ethanol concentrations of 0%, 6%, 12%, and 15%. In addition, the pH 3.4 and 3.6 treatments were adjusted to an ethanol concentration of 12% and 15% all wines were then assessed sensorially and seventeen terms were identified, through panel discussion, to describe the mouth-feel and taste qualities: velvet, aggressive, silk/satin, dry, fleshy, unripe, pucker viscosity, abrasive, heat, chewy, acidity, grippy/adhesive, bitter, balance, overall astringency, and mouth-coat. Descriptive analysis profiling techniques were used to train the panel and measure the intensity of these attributes. It was found that decreasing pH values (averaged across all ethanol concentrations) showed an increase in the overall astringency of the wine. The combined treatments of ethanol and pH, real wine parameters (pH 3.4 and 3.6; 12% and 15% ethanol) did not have an effect on the perception of the astringent sub-qualities of the wine. A time intensity study was also included using the pH and ethanol adjusted wines, which showed that as the ethanol level of the wines increased so did the time to maximum intensity. The second objective was to identify appropriate sensory descriptors to evaluate the influence of grape maturity and maceration technique (grape skin contact) on the astringency sub-qualities of red vinifera wines from Niagara. The grapes were harvested across two dates, representing an early harvest and a late harvest. A portion of the Cabernet Sauvignon grapes wine was divided into three maceration treatments of oneweek maceration, standard two-week maceration, three-week maceration, and MCM. Another portion of both the early and late harvest Cabernet Sauvignon grapes were chaptalized to yield a final ethanol concentration of 14.5%. The wines were assessed sensorially and thirteen terms were identified, through panel discussion, to describe the mouth-feel and taste qualities: carbon dioxide, pucker, acidity, silk/chamois, dusty/chalky/powdery, sandpaper, numbing, grippy/adhesive, dry, mouthcoat, bitter, balance and, overall astringency. Descriptive analysis techniques were used to train the panel and measure the intensity of these attributes. The data revealed few significant differences in the mouth-feel of the wines with respect to maturity; which included differences in overall astringency and balance. There were varietal differences between Cabernet Sauvignon, Cabernet Franc, and Pinot Noir and differences for Cabernet Sauvignon wines due to the length and manner of maceration and as a result of chaptalization. Statistical analysis revealed a more complex mouth-feel for the Pinot Noir wines; and an increase in the intensity of the astringent sub-qualities as a result of the addition of sugar to the wines. These findings have implications for how processing decisions, such as optimum grape maturity and vinification methods may affect red wine quality.

New immigrants to Canada typically have a more favourable health profile than the non-immigrant population. This phenomenon, known as the 'healthy immigrant effect', has been attributed to both the socioeconomic advantage (ie. educational attainment, occupational opportunity) of non-refugee immigrants and existing screening protocols that admit only the healthiest of persons to Canada. It has been suggested that this health advantage diminishes as the time of residence in Canada increases, due in part to the adoption of health-risk behaviours such as alcohol and cigarette use, an increase in excess body weight, and declining rates of physical activity. However, the majority of health research concerning immigrants to Canada has been limited to cross-sectional studies (Dunn & Dyck, 2000; Newbold & Danforth, 2003), which may mask an immigrant-specific cohort effect. Furthermore, the practice of aggregating foreign-bom persons by geographical regions or treating all immigrants as a homogeneous group may also obfuscate intra-immigrant differences in health. Accordingly, this study uses the Canadian National Population Health Surveys (NPHS) and data from the United Nations Development Program (UNDP) to prospectively evaluate factors that predict health status among immigrants to Canada. Each immigrant in the NPHS was linked to the UNDP Human Development Index of their country of birth, which uses a combined measure of health, education, and per capita income of the populace. The six-year change in health function, psychological distress, and self-rated health were considered from a population health perspective (Evans, 1994), using generalized-estimating equations (GEE) to examine the compounding effect of past and recent predictors of health. Catalase is the enzyme which decomposes hydrogen peroxide to water and oxygen. Escherichia coli contains two catalases. Hydroperoxidase I (HPI) is a bifunctional catalase-peroxidase. Hydroperoxidase II (HPII) is only catalytically active toward H202. Expression of the genes encoding these proteins is controlled by different regimes. HPJI is thought to be a hexamer, having one heme d cis group per enzymatic subunit. HPII wild type protein and heme containing mutant proteins were obtained from the laboratory of P. Loewen (Univ. of Manitoba). Mutants constructed by oligonucleotidedirected mutagenesis were targeted for replacement of either the His128 residue or the Asn201 residue in the vicinity of the HPII heme crevice. His128 is the residue thought to be analogous to the His74 distal axial ligand of the heme in the bovine liver enzyme, and Asn201 is believed to be a residue critical to the function of the enzyme because of its role in orienting and interacting with the substrate molecule. Investigation of the nature of the hemes via absorption spectroscopy of the unmodified catalase proteins and their derived pyridine hemochromes showed that while the bovine and Saccharomyces cerevisiae catalase enzymes are protoheme-containing, the HPII wild type protein contains heme d, and the mutant proteins contain either solely protoheme, or heme d-protoheme mixtures. Cyanide binding studies supported this, as ligand binding was monophasic for the bovine, Saccharomyces cerevisiae, and wild type HPII enzymes, but biphasic for several of the HPII mutant proteins. Several mammalian catalases, and at least two prokaryotic catalases, are known to be NADPH binding. The function of this cofactor appears to be the prevention of inactivation of the enzyme, which occurs via formation of the inactive secondary catalase peroxide compound (compound II). No physiologically plausible scheme has yet been proposed for the NADPH mediation of catalase activity. This study has shown, via fluorescence and affinity chromatography techniques, that NADPH binds to the T (Typical) and A (Atypical) catalases of Saccharomyces cerevisiae, and that wild type HPII apparently does not bind NADPH. This study has also shown that NADPH is unlike any other hydrogen donor to catalase, and addresses its features as a unique donor by proposing a mechanism whereby NADPH is oxidized and catalase is protected from inactivation via the formation of protein radical species. Migration of this radical to a position close to the NADPH is also proposed as an adjunct hypothesis, based on similar electron migrations that are known to occur within metmyoglobin and cytochrome c peroxidase when reacted with H202. Validation of these hypotheses may be obtained in appropriate future experiments.

Although a substantial amount of research has been done on all aspects ofHeliconius biology and their ecological interactions with Passiflora, there has not hitherto been a phylogenetic examination of this association for coevolution. To test the HeliconiuslPassilfora association for coevolutionary congruence, phylogenies for each group were established and compared. The phylogeny for 14 species ofHeliconiinae from Costa Rica was based on combined sequence data from rRNA ITS 2 and partial EF-1a gene regions. For the Passifloraceae, 17 host plant species were utilized to establish a phylogeny based on tRNALeucine and ITS 1/5.8S1 ITS 2 sequence data. The phylogenies for both groups were largely in agreement with current classification (for Passifloraceae) and previously established phylogenies. Associations with the large subgenera Passiflora and Decaloba correspond with the two major Advanced Radiation groups in Heliconius. Although strict congruence above subgenus level was not observed, broad scale congruence was evident. One main host shift as well as other possible explanations for lack of strict congruence are suggested.

This thesis explored early literacy development in young vulnerable readers. More specifically, this thesis examined an emergent literacy program called Reading Rocks Junior offered by the Learning Disabilities Association of Niagara Region to children four- to six-years of age living in low socioeconomic status communities. Three methodologies were combined to create a rich and complete picture of an effective and accessible literacy program. First of all, a description of the Reading Rocks Junior program is outlined. Secondly, quantitative data that was collected pre- and post- program was analyzed to demonstrate achievement gains made as a result of participating in the program. Finally, qualitative interviews with the program coordinator, the convener of the agency that funded Reading Rocks Junior and three parents whose children participated in the program were analyzed to determine the contextual factors that make Reading Rocks Junior a success.

Ce triptyque d’essais présente le caractère versatile et évasif du concept moderne de capital social à plusieurs niveaux – global, national et régional, ainsi que dans le présent et dans le passé. Le premier article conteste l’hypothèse prédominante selon laquelle il y a une cohabitation entre l’engagement civique et la démocratie. Malgré sa validité au niveau général, la relation n’est pas confirmée si les catégories hétérogènes sont désagrégées. Pour les pays post-communistes de l'Europe, la relation entre le type de régime et la tendance de s'associer ressemble à celle des démocraties latines consolidées si la participation dans les associations volontaires est choisie comme mesure de la vitalité du capital social. Par conséquent, la vie civique moins intense ne prédit pas de difficultés pour la démocratie. Le deuxième article est une compilation originale de plus de 100 organisations classifiées selon les standards contemporains et une collection de présentations d'une douzaine d'organisations bulgares, les plus populaires depuis le XIXème siècle. Cette contribution importante à l’historiographie de la vie associative bulgare jusqu’à 1944 est le résultat d'un travail qui combine des entrevues avec des historiens et une recherche dans les archives. Le panoptique organisationnel sert de réfutation empirique de l’hypothèse qui attribue la faiblesse organisationnelle présente du poste-communisme à la pénurie de vie organisationnelle développée par le passé. ii Les mérites du troisième article sont doubles. Au niveau empirique on démontre que l’organisation culturelle la plus importante en Bulgarie a apparu comme une institution nationaliste imitant les organisations similaires des autres pays Européens. Elle s’est développée graduellement par une adaptation des expériences étrangères aux conditions locales. La collection des références bulgares est unique et représente le produit d’un travail méticuleux sur les documents et les entrevues. Au niveau abstrait, on confirme l’applicabilité de la théorie du transfert de la politique publique à un cas historique existant avant la théorie elle-même. Finalement, l’analyse détaillée des précurseurs du cabinet de lecture bulgare représente une contribution à la sociologie politique de l’histoire de la lecture. Puisque l’altération des habitats d’eau douce augmente, il devient critique d’identifier les composantes de l’habitat qui influencent les métriques de la productivité des pêcheries. Nous avons comparé la contribution relative de trois types de variables d’habitat à l’explication de la variance de métriques d’abondance, de biomasse et de richesse à l’aide de modèles d’habitat de poissons, et avons identifié les variables d’habitat les plus efficaces à expliquer ces variations. Au cours des étés 2012 et 2013, les communautés de poissons de 43 sites littoraux ont été échantillonnées dans le Lac du Bonnet, un réservoir dans le Sud-est du Manitoba (Canada). Sept scénarios d’échantillonnage, différant par l’engin de pêche, l’année et le moment de la journée, ont été utilisés pour estimer l’abondance, la biomasse et la richesse à chaque site, toutes espèces confondues. Trois types de variables d’habitat ont été évalués: des variables locales (à l’intérieur du site), des variables latérales (caractérisation de la berge) et des variables contextuelles (position relative à des attributs du paysage). Les variables d’habitat locales et contextuelles expliquaient en moyenne un total de 44 % (R2 ajusté) de la variation des métriques de la productivité des pêcheries, alors que les variables d’habitat latérales expliquaient seulement 2 % de la variation. Les variables les plus souvent significatives sont la couverture de macrophytes, la distance aux tributaires d’une largeur ≥ 50 m et la distance aux marais d’une superficie ≥ 100 000 m2, ce qui suggère que ces variables sont les plus efficaces à expliquer la variation des métriques de la productivité des pêcheries dans la zone littorale des réservoirs.

In the present study the preparation and characterisation of rubber ferrite composites (RFC) containing barium ferrite (BaF) and strontium ferrite (SrF) have been dealt with. The incorporation of the hard ferrites into natural and nitrile rubber was carried out according to a specific recipe for various loadings of magnetic fillers. For this, the ferrite materials namely barium ferrite and strontium ferrite having the general formula MO6Fe2O3 have been prepared by the conventional ceramic techniques. After characterisation they were incorporated into the natural and nitrile rubber matrix by mechanical method. Carbon black was also incorporated at different loading into the rubber ferrite composites to study its effect on various properties. The cure characteristics, mechanical, dielectric and magnetic properties of these composites were evaluated. The ac electrical conductivity of both the ceramic ferrites and rubber ferrite composites were also calculated using a simple relation. The investigations revealed that the rubber ferrite composites with the required dielectric and magnetic properties can be obtained by the incorporation of ferrite fillers into the rubber matrix, without compromising much on the processability and mechanical properties.

This doctoral thesis addresses the growing concern about the significant changes in the climatic and weather patterns due to the aerosol loading that have taken place in the Indo Gangetic Plain(IGP)which includes most of the Northern Indian region. The study region comprises of major industrial cities in India (New Delhi, Kanpur, Allahabad, Jamshedpur and Kolkata). Northern and central parts of India are one of the most thickly populated areas in the world and have the most intensely farmed areas. Rapid increase in population and urbanization has resulted in an abrupt increase in aerosol concentrations in recent years. The IGP has a major source of coal; therefore most of the industries including numerous thermal power plants that run on coal are located around this region. They inject copious amount of aerosols into the atmosphere. Moreover, the transport of dust aerosols from arid locations is prevalent during the dry months which increase the aerosol loading in theatmosphere. The topography of the place is also ideal for the congregation of aerosols. It is bounded by the Himalayas in the north, Thar Desert in the west, the Vindhyan range in the south and Brahmaputra ridge in the east. During the non‐monsoon months (October to May) the weather in the location is dry with very little rainfall. Surface winds are weak during most of the time in this dry season. The aerosols that reach the location by means of long distance transport and from regional sources get accumulated under these favourable conditions. The increase in aerosol concentration due to the complex combination of aerosol transport and anthropogenic factors mixed with the contribution from the natural sources alters the optical properties and the life time of clouds in the region. The associated perturbations in radiative balance have a significant impact on the meteorological parameters and this in turn determines the precipitation forming process. Therefore, any change in weather which disturbs the normal hydrological pattern is alarming in the socio‐economic point of view. Hence, the main focus of this work is to determine the variation in transport and distribution of aerosols in the region and to understand the interaction of these aerosols with meteorological parameters and cloud properties.
