En esta Tesis Doctoral se emplean y desarrollan Métodos Bayesianos para su aplicación en análisis geotécnicos habituales, con un énfasis particular en (i) la valoración y selección de modelos geotécnicos basados en correlaciones empíricas; en (ii) el desarrollo de predicciones acerca de los resultados esperados en modelos geotécnicos complejos. Se llevan a cabo diferentes aplicaciones a problemas geotécnicos, como es el caso de: (1) En el caso de rocas intactas, se presenta un método Bayesiano para la evaluación de modelos que permiten estimar el módulo de Young a partir de la resistencia a compresión simple (UCS). La metodología desarrollada suministra estimaciones de las incertidumbres de los parámetros y predicciones y es capaz de diferenciar entre las diferentes fuentes de error. Se desarrollan modelos "específicos de roca" para los tipos de roca más comunes y se muestra cómo se pueden "actualizar" esos modelos "iniciales" para incorporar, cuando se encuentra disponible, la nueva información específica del proyecto, reduciendo las incertidumbres del modelo y mejorando sus capacidades predictivas. (2) Para macizos rocosos, se presenta una metodología, fundamentada en un criterio de selección de modelos, que permite determinar el modelo más apropiado, entre un conjunto de candidatos, para estimar el módulo de deformación de un macizo rocoso a partir de un conjunto de datos observados. Una vez que se ha seleccionado el modelo más apropiado, se emplea un método Bayesiano para obtener distribuciones predictivas de los módulos de deformación de macizos rocosos y para actualizarlos con la nueva información específica del proyecto. Este método Bayesiano de actualización puede reducir significativamente la incertidumbre asociada a la predicción, y por lo tanto, afectar las estimaciones que se hagan de la probabilidad de fallo, lo cual es de un interés significativo para los diseños de mecánica de rocas basados en fiabilidad. (3) En las primeras etapas de los diseños de mecánica de rocas, la información acerca de los parámetros geomecánicos y geométricos, las tensiones in-situ o los parámetros de sostenimiento, es, a menudo, escasa o incompleta. Esto plantea dificultades para aplicar las correlaciones empíricas tradicionales que no pueden trabajar con información incompleta para realizar predicciones. Por lo tanto, se propone la utilización de una Red Bayesiana para trabajar con información incompleta y, en particular, se desarrolla un clasificador Naïve Bayes para predecir la probabilidad de ocurrencia de grandes deformaciones (squeezing) en un túnel a partir de cinco parámetros de entrada habitualmente disponibles, al menos parcialmente, en la etapa de diseño. This dissertation employs and develops Bayesian methods to be used in typical geotechnical analyses, with a particular emphasis on (i) the assessment and selection of geotechnical models based on empirical correlations; on (ii) the development of probabilistic predictions of outcomes expected for complex geotechnical models. Examples of application to geotechnical problems are developed, as follows: (1) For intact rocks, we present a Bayesian framework for model assessment to estimate the Young’s moduli based on their UCS. Our approach provides uncertainty estimates of parameters and predictions, and can differentiate among the sources of error. We develop ‘rock-specific’ models for common rock types, and illustrate that such ‘initial’ models can be ‘updated’ to incorporate new project-specific information as it becomes available, reducing model uncertainties and improving their predictive capabilities. (2) For rock masses, we present an approach, based on model selection criteria to select the most appropriate model, among a set of candidate models, to estimate the deformation modulus of a rock mass, given a set of observed data. Once the most appropriate model is selected, a Bayesian framework is employed to develop predictive distributions of the deformation moduli of rock masses, and to update them with new project-specific data. Such Bayesian updating approach can significantly reduce the associated predictive uncertainty, and therefore, affect our computed estimates of probability of failure, which is of significant interest to reliability-based rock engineering design. (3) In the preliminary design stage of rock engineering, the information about geomechanical and geometrical parameters, in situ stress or support parameters is often scarce or incomplete. This poses difficulties in applying traditional empirical correlations that cannot deal with incomplete data to make predictions. Therefore, we propose the use of Bayesian Networks to deal with incomplete data and, in particular, a Naïve Bayes classifier is developed to predict the probability of occurrence of tunnel squeezing based on five input parameters that are commonly available, at least partially, at design stages.

Cytochrome c oxidase is a membrane-bound enzyme that catalyzes the four-electron reduction of oxygen to water. This highly exergonic reaction drives proton pumping across the membrane. One of the key questions associated with the function of cytochrome c oxidase is how the transfer of electrons and protons is coupled and how proton transfer is controlled by the enzyme. In this study we focus on the function of one of the proton transfer pathways of the R. sphaeroides enzyme, the so-called K-proton transfer pathway (containing a highly conserved Lys(I-362) residue), leading from the protein surface to the catalytic site. We have investigated the kinetics of the reaction of the reduced enzyme with oxygen in mutants of the enzyme in which a residue [Ser(I-299)] near the entry point of the pathway was modified with the use of site-directed mutagenesis. The results show that during the initial steps of oxygen reduction, electron transfer to the catalytic site (to form the “peroxy” state, Pr) requires charge compensation through the proton pathway, but no proton uptake from the bulk solution. The charge compensation is proposed to involve a movement of the K(I-362) side chain toward the binuclear center. Thus, in contrast to what has been assumed previously, the results indicate that the K-pathway is used during oxygen reduction and that K(I-362) is charged at pH ≈ 7.5. The movement of the Lys is proposed to regulate proton transfer by “shutting off” the protonic connectivity through the K-pathway after initiation of the O2 reduction chemistry. This “shutoff” prevents a short-circuit of the proton-pumping machinery of the enzyme during the subsequent reaction steps.

The reduction of 12-oxophytodienoic acid (OPDA) to 3-oxo-2(2′[Z]-pentenyl)-cyclopentane-1-octanoic acid is catalyzed by 12-oxophytodienoate-10,11-reductase (OPR). Analysis of the isomer preference of OPR has indicated that the activity is composed of two isoenzymes exhibiting different stereoselectivities. The two isoforms of OPR have been separated, using protein extracts of Rock Harlequin (Corydalis sempervirens) as the starting material. OPRI, the enzyme reported earlier from the same species and corresponding to the cloned OPR from Arabidopsis, utilized 9R,13R-OPDA >> 9S,13R-OPDA but not the 13S-configured isomers, whereas the new activity, OPRII, effectively reduced all four OPDA isomers, including the natural 9S,13S-OPDA (cis-[+]-OPDA). OPRII activity is characterized in detail. The enzyme's enzymatic, biochemical, and immunological properties prove that it is a close relative of OPRI. The roles of OPRI and OPRII in octadecanoid biology are discussed.

A incorporação de elementos da cultura primeira do estudante no processo de ensino-aprendizagem foi defendida pelo pedagogo francês Georges Snyders (1988) em sua obra \"A Alegria na Escola\". Esta pesquisa contribuiu com essa interface, identificando, no discurso de canções do rock n\' roll, elementos textuais que possibilitem reflexões no âmbito conceitual, epistemológico e sociopolítico sobre a exploração do espaço. O objeto de estudo neste trabalho são canções do período entre as décadas de 1960 e 1970 que possuem representações sobre a astronomia e as missões espaciais. O uso do rock justificou-se pelo fato de temas sobre exploração espacial aparecerem no trabalho de diversos artistas desse gênero musical, permitindo reflexões em nível conceitual, epistemológico e sociopolítico sobre a ciência, a tecnologia e suas relações com a sociedade e o ambiente. Além disso, identificamos que tanto o rock quanto as missões espaciais foram fenômenos culturais que dependeram em sua gênese dos avanços da tecnologia e da ciência e tiveram sua repercussão na sociedade através de processos midiáticos. Essas canções foram selecionadas entre os diversos gêneros de rock, e analisadas a partir de referenciais semiodiscursivos. As atividades foram aplicadas em situações formais de ensino - ensino médio e ensino superior -, em formação continuada de professores e projetos de ensino não formal na escola. No processo de ensinoaprendizagem, foram desenvolvidas atividades que envolviam leitura-comentada da canção, identificando na letra, melodia e harmonia, aspectos que evidenciavam um discurso crítico sobre a ciência e sua relação com a sociedade e o ambiente. Essas atividades envolveram três instâncias: Elaboração, Aplicação e Análise. Como referencial norteador dessas etapas, nos valemos das teorias socioculturais de Vigotski (2001), Snyders (1988) e Freire (2013).

Rock mass characterization requires a deep geometric understanding of the discontinuity sets affecting rock exposures. Recent advances in Light Detection and Ranging (LiDAR) instrumentation currently allow quick and accurate 3D data acquisition, yielding on the development of new methodologies for the automatic characterization of rock mass discontinuities. This paper presents a methodology for the identification and analysis of flat surfaces outcropping in a rocky slope using the 3D data obtained with LiDAR. This method identifies and defines the algebraic equations of the different planes of the rock slope surface by applying an analysis based on a neighbouring points coplanarity test, finding principal orientations by Kernel Density Estimation and identifying clusters by the Density-Based Scan Algorithm with Noise. Different sources of information —synthetic and 3D scanned data— were employed, performing a complete sensitivity analysis of the parameters in order to identify the optimal value of the variables of the proposed method. In addition, raw source files and obtained results are freely provided in order to allow to a more straightforward method comparison aiming to a more reproducible research.

The San Julián’s stone is the main material used to build the most important historical buildings in Alicante city (Spain). This paper describes the analysis developed to obtain the relationship between the static and the dynamic modulus of this sedimentary rock heated at different temperatures. The rock specimens have been subjected to heating processes at different temperatures to produce different levels of weathering on 24 specimens. The static and dynamic modulus has been measured for every specimen by means of the ISRM standard and ultrasonic tests, respectively. Finally, two analytic formulas are proposed for the relationship between the static and the dynamic modulus for this stone. The results have been compared with some relationships proposed by different researchers for other types of rock. The expressions presented in this paper can be useful for the analysis, using non-destructive techniques, of the integrity level of historical constructions built with San Julián’s stone affected by fires.

The complete characterization of rock masses implies the acquisition of information of both, the materials which compose the rock mass and the discontinuities which divide the outcrop. Recent advances in the use of remote sensing techniques – such as Light Detection and Ranging (LiDAR) – allow the accurate and dense acquisition of 3D information that can be used for the characterization of discontinuities. This work presents a novel methodology which allows the calculation of the normal spacing of persistent and non-persistent discontinuity sets using 3D point cloud datasets considering the three dimensional relationships between clusters. This approach requires that the 3D dataset has been previously classified. This implies that discontinuity sets are previously extracted, every single point is labeled with its corresponding discontinuity set and every exposed planar surface is analytically calculated. Then, for each discontinuity set the method calculates the normal spacing between an exposed plane and its nearest one considering 3D space relationship. This link between planes is obtained calculating for every point its nearest point member of the same discontinuity set, which provides its nearest plane. This allows calculating the normal spacing for every plane. Finally, the normal spacing is calculated as the mean value of all the normal spacings for each discontinuity set. The methodology is validated through three cases of study using synthetic data and 3D laser scanning datasets. The first case illustrates the fundamentals and the performance of the proposed methodology. The second and the third cases of study correspond to two rock slopes for which datasets were acquired using a 3D laser scanner. The second case study has shown that results obtained from the traditional and the proposed approaches are reasonably similar. Nevertheless, a discrepancy between both approaches has been found when the exposed planes members of a discontinuity set were hard to identify and when the planes pairing was difficult to establish during the fieldwork campaign. The third case study also has evidenced that when the number of identified exposed planes is high, the calculated normal spacing using the proposed approach is minor than those using the traditional approach.

Although Portugal does not have a significant radical right presence in its party system, in the last decades the country did witness the development of a neo-Nazi skinhead movement that expresses its white nationalist nature and goals through the musical genres of Rock Against Communism (RAC) and the related Oi!. Utilizing various historical sources and theoretical analysis, this study contextualizes the development of nationalist music in Portugal, both before and especially during the democratic period (1974-2015). It focuses on its protagonists, domestic and international networks, as well as on the few attempts to establish a common cause with radical right-wing political parties at the turn of the century and in present times.

Soil degradation threatens agricultural production and food security in Sub-Saharan Africa. In the coming decades, soil degradation, in particular soil erosion, will become worse through the expansion of agriculture into savannah and forest and changes in climate. This study aims to improve the understanding of how land use and climate change affect the hydrological cycle and soil erosion rates at the catchment scale. We used the semi-distributed, time-continuous erosion model SWAT (Soil Water Assessment Tool) to quantify runoff processes and sheet and rill erosion in the Upper Ouémé River catchment (14500 km**2, Central Benin) for the period 1998-2005. We could then evaluate a range of land use and climate change scenarios with the SWAT model for the period 2001-2050 using spatial data from the land use model CLUE-S and the regional climate model REMO. Field investigations were performed to parameterise a soil map, to measure suspended sediment concentrations for model calibration and validation and to characterise erosion forms, degraded agricultural fields and soil conservation practices. Modelling results reveal current "hotspots" of soil erosion in the north-western, eastern and north-eastern parts of the Upper Ouémé catchment. As a consequence of rapid expansion of agricultural areas triggered by high population growth (partially caused by migration) and resulting increases in surface runoff and topsoil erosion, the mean sediment yield in the Upper Ouémé River outlet is expected to increase by 42 to 95% by 2025, depending on the land use scenario. In contrast, changes in climate variables led to decreases in sediment yield of 5 to 14% in 2001-2025 and 17 to 24% in 2026-2050. Combined scenarios showed the dominance of land use change leading to changes in mean sediment yield of -2 to +31% in 2001-2025. Scenario results vary considerably within the catchment. Current "hotspots" of soil erosion will aggravate, and a new "hotspot" will appear in the southern part of the catchment. Although only small parts of the Upper Ouémé catchment belong to the most degraded zones in the country, sustainable soil and plant management practices should be promoted in the entire catchment. The results of this study can support planning of soil conservation activities in Benin.
