The purpose of this study was to investigate whether the critical force (CritF) and anaerobic impulse capacity (AIC) - estimated by tethered swimming - reflect the aerobic and anaerobic performance of swimmers. 12 swimmers performed incremental test in tethered swimming to determine lactate anaerobic threshold (AnTLAC), maximal oxygen uptake (̇VO2MAX) and force associated with the ̇VO2MAX (i ̇VO2MAX). The swimmers performed 4 exhaustive (tlim) exercise bouts (100, 110, 120 and 130% i ̇VO2MAX) to compute the CritF and AIC (F vs. 1/tlim model); a 30-s all-out tethered swimming bout to determine their anaerobic fitness (ANF); 100, 200, and 400-m time-trials to determine the swimming performance. CritF (57.09±11.77 N) did not differ from AnTLAC (53.96±11.52 N, (P>0.05) but was significantly lower than i ̇VO2MAX (71.02±8.36 N). In addition, CritF presented significant correlation with AnTLAC (r=0.76; P<0.05) and i ̇VO2MAX (r=0.74; P<0.05). On the other hand, AIC (286.19±54.91 N.s) and ANF (116.10±13.66 N) were significantly correlated (r=0.81, p<0.05). In addition, CritF and AIC presented significant correlations with all time-trials. In summary, this study demonstrates that CritF and AIC can be used to evaluate AnTLAC and ANF and to predict 100, 200, and 400-m free swimming. © Georg Thieme Verlag KG Stuttgart . New York.

LaNiO3 thin films were deposited on SrLaAlO4 (1 0 0) and SrLaAlO4 (0 0 1) single crystal substrates by a chemical solution deposition method and heat-treated in oxygen atmosphere at 700° C in tube oven. Structural, morphological, and electrical properties of the LaNiO 3 thin films were characterized by X-ray diffraction (XRD), atomic force microscopy (AFM), field emission scanning electron microscopy (FE-SEM), and electrical resistivity as temperature function (Hall measurements). The X-ray diffraction data indicated good crystallinity and a structural preferential orientation. The LaNiO3 thin films have a very flat surface and no droplet was found on their surfaces. Samples of LaNiO3 grown onto (1 0 0) and (0 0 1) oriented SrLaAlO4 single crystal substrates reveled average grain size by AFM approximately 15-30 nm and 20-35 nm, respectively. Transport characteristics observed were clearly dependent upon the substrate orientation which exhibited a metal-to-insulator transition. The underlying mechanism is a result of competition between the mobility edge and the Fermi energy through the occupation of electron states which in turn is controlled by the disorder level induced by different growth surfaces. © 2013 Elsevier Ltd and Techna Group S.r.l.

We investigated the participation of A1 or A2 receptors in the gonadotrope and their role in the regulation of LH and FSH secretion in adult rat hemipituitary preparations, using adenosine analogues. A dose-dependent inhibition of LH and FSH secretion was observed after the administration of graded doses of the R-isomer of phenylisopropyladenosine (R-PIA; 1 nM, 10 nM, 100 nM, 1 µM and 10 µM). The effect of R-PIA (10 nM) was blocked by the addition of 8-cyclopentyltheophylline (CPT), a selective A1 adenosine receptor antagonist, at the dose of 1 µM. The addition of an A2 receptor-specific agonist, 5-N-methylcarboxamidoadenosine (MECA), at the doses of 1 nM to 1 µM had no significant effect on LH or FSH secretion, suggesting the absence of this receptor subtype in the gonadotrope. However, a sharp inhibition of the basal secretion of these gonadotropins was observed after the administration of 10 µM MECA. This effect mimicked the inhibition induced by R-PIA, supporting the hypothesis of the presence of A1 receptors in the gonadotrope. R-PIA (1 nM to 1 µM) also inhibited the secretion of LH and FSH induced by phospholipase C (0.5 IU/ml) in a dose-dependent manner. These results suggest the presence of A1 receptors and the absence of A2 receptors in the gonadotrope. It is possible that the inhibition of LH and FSH secretion resulting from the activation of A1 receptors may have occurred independently of the increase in membrane phosphoinositide synthesis.

A zona subgranular (ZSG) do giro denteado (GD) de mamíferos adultos é conhecida por produzir constantemente novos neurônios. A busca por novas moléculas que possam modular a formação de novas células neurais são bastante atuais. Visto que a Amazônia é conhecida mundialmente pela sua biodiversidade, com um potencial pouco explorado de fármacos naturais derivados de plantas medicinais típicas da região. O trabalho buscou investigar o efeito neurogênico do extrato aquoso (EA) da Physalis angulata e da substância purificada Fisalina D sobre as células-tronco do GD do hipocampo de camundongos adultos. Os camundongos machos (BALB/c), 6 a 8 semanas de idade foram divididos em quatro grupos experimentais: controle e tratados com EA ou substância purificada. Os animais receberam diferentes doses do extrato (0,1; 1 e 5 mg/Kg) e/ou substância purificada (5mg/Kg) ou salina (grupo controle), 5 horas depois uma única dose de 5-Bromodeoxiuridina (BrdU) [50mg/kg]. Em seguida, os animais foram sacrificados 24 horas ou 7 dias após a administração do BrdU. Os cérebros foram coletados e cortes coronais do hipocampo (40 μm) foram realizados para contagem das células BrdU-positivas no GD hipocampal. Para avaliação estatística realizamos análise de variância (ANOVA) das médias amostrais seguida pelo pós-teste t de Student. O EA promoveu um aumento significativo do número de células BrdU positivas no GD dos grupos tratados em relação ao grupo controle [Controle, 92±24 (n=9); 0,1mg/Kg, 160±22 (n=4); 1mg/Kg, 310±5 (n=4); 5mg/Kg, 501±24 (n=3)] nos animais sacrificados 24 horas após administração do BrdU. Quando os animais foram sacrificados 7 dias após administração do BrdU, o número de células BrdU+ no GD também foi maior no grupo tratado em relação ao controle [Controle, 107±7 (n=4); 5mg/Kg, 145±23 (n=4)]. Usando a substância purificada, Fisalina D, também observamos um aumento do número de células BrdU+ no GD do grupo tratado com a droga em relação ao grupo controle [Controle, 92±24 (n=9); Fisalina D, 5mg/Kg, 316±37 (n=3)]. Este resultado sugere que o EA e a sustância purificada, na dose de 5 mg/Kg, estimulam a proliferação de células BrdU-positivas na ZSG do GD do hipocampo de camundongos adultos.

Sildenafil attenuates acute pulmonary embolism-induced pulmonary hypertension. However, the hemodynamic effects of sildenafil in combination with other vasodilators during acute pulmonary embolism have not been examined yet. In the present study, we examined the hemodynamic effects of combined sildenafil (0.25 mg/kg, i.v.) and L-arginine (100, 200, 500, and 1000 mg/kg/h, i.v.) in an anesthetized dog model of acute pulmonary embolism. Plasma nitrite/nitrate (NOx) and cGMP concentrations were determined using an ozone-based chemiluminescence assay and a commercial enzyme immunoassay, respectively. We found that L-arginine alone did not attenuate acute pulmonary embolism-induced pulmonary hypertension. However, significant decreases in mean pulmonary artery pressure were observed 30, 45, 60, and 75 min after the administration of sildenafil alone or after the combined administration of sildenafil and L-arginine (all P<0.05). No significant differences among groups were observed in the respiratory parameters. While L-arginine significantly increased NOx concentrations, cGMP concentrations increased only when sildenafil was administered (all P<0.05). These results suggest that while sildenafil attenuates acute pulmonary embolism-induced pulmonary hypertension, L-arginine does not enhance the beneficial hemodynamic effects of sildenafil. In addition, these findings suggest that stimulation of NO synthesis with L-arginine during acute pulmonary embolism does not produce beneficial effects. (c) 2005 Elsevier B.V. All rights reserved.

Aim The aim of the present study was to evaluate the effect of surface roughness (roughness average [Ra] μm) on the hydrophobicity of a denture-base acrylic resin and the initial adherence and biofilm formation of Candida albicans (C. albicans). Methods Disk-shaped specimens were divided into six groups: Ra 0.05, Ra 0.2, Ra 0.4, Ra 0.8, Ra 1.5, and Ra 3.0. Water contact angles (WCA) were measured, and the specimens incubated with C. albicans for 90 min (initial adherence, n = 108) or 48 h (biofilm formation, n = 108). Adhered and biofilm cells were evaluated by c.f.u./mL and 2,3-bis-(2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazolium-5-carboxanilide (XTT), and the correlation between the two methods was evaluated. The surface of the specimens and cells (adhered and biofilm) were also analyzed by scanning electron microscopy (SEM). Results Groups Ra 0.05 and 3.0 exhibited the lowest (~75°) and the highest (~100°) WCA mean values, respectively. For both initial adherence and biofilm formation, no statistically-significant differences were observed among all groups, as determined by c.f.u./mL and XTT. A positive correlation between these two methods was found. SEM analysis showed the presence of scratches and valleys on the acrylic specimens and densely-packed yeast cells covering the entire surface. Conclusions Roughness significantly increased hydrophobicity (WCA), but had no effect on the number and metabolic activity of adherent and biofilm cells of C. albicans.
