O número de mulheres jovens infectadas pelo HIV-1 vem crescendo desde o início da epidemia da aids, principalmente nos países em desenvolvimento, onde a frequência de gravidez é elevada. O desenvolvimento de estratégias para evitar a transmissão vertical tem aumentado o número de recém-nascidos não infectados nascidos de gestantes portadora do vírus. O objetivo da presente tese foi avaliar, in vivo e in vitro, o perfil de citocinas de neonatos não infectados, porém nascidos de mulheres infectadas pelo HIV-1. Os resultados demonstraram elevados níveis de citocinas pró-inflamatórias relacionadas ao fenótipo Th17, associadas à baixa produção de IL-10, tanto nos plasmas quanto nos sobrenadantes das culturas de células T ativadas obtidas do sangue do cordão umbilical de neonatos nascidos de gestantes infectadas pelo HIV-1 com cargas virais plasmáticas(PVL) detectáveis. De modo interessante, um perfil similar pró-inflamatório foi observado em amostras de sangue periférico de suas respectivas mães. Por outro lado, níveis elevados de IL-10, associados à reduzida produção de citocinas inflamatórias, foram dosados no sangue e nos sobrenadantes das culturas de células T ativadas de gestantes que controlavam a PVL, assim como de seus neonatos. Com relação à caracterização fenotípica das células T maternas produtoras de IL-10, a análise por citometria de fluxo demonstrou que essa citocina é majoritariamente produzida por células T CD4+Foxp3-. Curiosamente, a replicação viral in vitro do HIV-1 foi inferior nas culturas das gestantes infectadas pelo HIV-1 e foi relacionada aos efeitos inibitórios da IL-10 sobre a produção de TNF-α. Por fim, os neonatos não infectados expostosin utero à terapia antirretroviral (ART) apresentaram um menor peso ao nascer, e este achado foi inversamente correlacionado com os níveis periféricos maternos de TNF-α. Em conclusão, nossos achados sugerem que gestantes que conseguem controlar a PVL, e o incremento na produção de IL-10 materna possam favorecer a expansão das células Tr-1, as quais podem auxiliar em reduzir o risco de transmissão vertical do HIV-1 por reduzir a taxa de replicação viral. Por outro lado, outros resultados também apresentados aqui, apesar de preliminares, revelaram efeitos adversos da replicação viral e da ART em gestantes infectadas pelo HIV-1 no desenvolvimento funcional das células T de neonatos não infectados. Esses dados podem ajudar a explicar por que algumas dessas crianças apresentam elevado risco à morbidade e mortalidade devido a um estado basal de hipersensibilidade patológica.

O estresse durante o desenvolvimento está associado com diversas desordens neurocomportamentais, que podem persistir ao longo da vida. A hiperatividade é um dos transtornos comportamentais que, com maior frequência, observa-se em humanos submetidos ao estresse precoce. Esse transtorno pode ser a manifestação clínica predominante, ou mesclar-se com déficit de atenção, impulsividade e retardo da aprendizagem, constituindo o Transtorno de Déficit de Atenção e Hiperatividade (TDAH), com número de casos diagnosticados em ascensão. Diversos protocolos experimentais utilizam a separação materna (SM) de roedores neonatos para mimetizar as consequências do estresse precoce em humanos. Esta predileção por roedores recém-nascidos se deve à sua equivalência aproximada com fetos humanos no terceiro trimestre da gestação em termos de neurodesenvolvimento, quando ocorre o maior crescimento do Sistema Nervoso Central fetal. Neste trabalho, camundongos suíços neonatos foram submetidos a sessões diárias de isolamento com separação materna, entre o 2 e o 10 dias de vida pós-natal (PN2 a PN10), variando-se a temperatura de isolamento dos filhotes, que permaneciam sem aquecimento (na temperatura do biotério, entre 22 e 25C) ou eram mantidos aquecidos a 37C durante essas sessões. Portanto, foram três grupos experimentais: isolamento aquecido com SM; isolamento não aquecido com SM; e controle. Os animais do grupo controle foram pesados em PN2 e PN10 e, prontamente, devolvidos às progenitoras. Todos os animais foram desmamados e sexados em PN21, não sendo perturbados até a realização dos testes neurocomportamentais, a partir de PN30, que incluíram os Testes de Campo Aberto e de Esquiva Inibitória. Num segundo estudo, foram realizadas dosagens séricas da corticosterona basal e dos hormônios tireoidianos nos três grupos experimentais, em PN6, PN10 e PN30. Finalmente, num terceiro estudo, camundongos do grupo controle e do grupo submetido ao isolamento não aquecido com SM foram tratados com vimpocetina (20g/kg), um neuroprotetor potencial, ou com veículo (Dimetilsulfóxido), a fim de avaliar os efeitos da vimpocetina na atividade locomotora. O 1 estudo demonstrou que a temperatura foi um fator crítico para a manifestação de hiperatividade locomotora no Teste do Campo Aberto, que ocorreu, somente, nos animais do grupo submetido ao isolamento não aquecido. Adicionalmente, não houve diferenças entre os grupos experimentais no Teste da Esquiva Inibitória, quanto à memória e à aprendizagem. O 2 estudo demonstrou que a temperatura do isolamento influenciou os níveis da corticosterona basal e dos hormônios tireoidianos em PN10 e em PN30. No 3 estudo, a vimpocetina reduziu a hiperatividade locomotora no grupo de animais submetidos ao isolamento não aquecido com SM. De todo o exposto, conclui-se que o isolamento com SM de camundongos suíços neonatos à temperatura de 22 a 25C aumentou a atividade locomotora nesses animais, e que a vimpocetina foi capaz de atenuar esse comportamento, sem aumento da mortalidade. Finalmente, considerando-se o maior risco de desenvolvimento do TDAH em crianças e adolescentes com históricos de prematuridade, levanta-se a hipótese de que o estresse térmico pelo frio seja um dos fatores envolvidos na fisiopatogenia da hiperatividade nesses casos.

Two large hydrologic issues face the Kings Basin, severe and chronic overdraft of about 0.16M ac-ft annually, and flood risks along the Kings River and the downstream San Joaquin River. Since 1983, these floods have caused over $1B in damage in today’s dollars. Capturing flood flows of sufficient volume could help address these two pressing issues which are relevant to many regions of the Central Valley and will only be exacerbated with climate change. However, the Kings River has high variability associated with flow magnitudes which suggests that standard engineering approaches and acquisition of sufficient acreage through purchase and easements to capture and recharge flood waters would not be cost effective. An alternative approach investigated in this study, termed On-Farm Flood Flow Capture, involved leveraging large areas of private farmland to capture flood flows for both direct and in lieu recharge. This study investigated the technical and logistical feasibility of best management practices (BMPs) associated with On-Farm Flood Flow Capture. The investigation was conducted near Helm, CA, about 20 miles west of Fresno, CA. The experimental design identified a coordinated plan to determine infiltration rates for different soil series and different crops; develop a water budget for water applied throughout the program and estimate direct and in lieu recharge; provide a preliminary assessment of potential water quality impacts; assess logistical issues associated with implementation; and provide an economic summary of the program. At check locations, we measured average infiltration rates of 4.2 in/d for all fields and noted that infiltration rates decreased asymptotically over time to about 2 – 2.5 in/d. Rates did not differ significantly between the different crops and soils tested, but were found to be about an order of magnitude higher in one field. At a 2.5 in/d infiltration rate, 100 acres are required to infiltrate 10 CFS of captured flood flows. Water quality of applied flood flows from the Kings River had concentrations of COC (constituents of concern; i.e. nitrate, electrical conductivity or EC, phosphate, ammonium, total dissolved solids or TDS) one order of magnitude or more lower than for pumped groundwater at Terranova Ranch and similarly for a broader survey of regional groundwater. Applied flood flows flushed the root zone and upper vadose zone of nitrate and salts, leading to much lower EC and nitrate concentrations to a depth of 8 feet when compared to fields in which more limited flood flows were applied or for which drip irrigation with groundwater was the sole water source. In demonstrating this technology on the farm, approximately 3,100 ac-ft was diverted, primarily from April through mid-July, with about 70% towards in lieu and 30% towards direct recharge. Substantial flood flow volumes were applied to alfalfa, wine grapes and pistachio fields. A subset of those fields, primarily wine grapes and pistachios, were used primarily to demonstrate direct recharge. For those fields about 50 – 75% of water applied was calculated going to direct recharge. Data from the check studies suggests more flood flows could have been applied and infiltrated, effectively driving up the amount of water towards direct recharge. Costs to capture flood flows for in lieu and direct recharge for this project were low compared to recharge costs for other nearby systems and in comparison to irrigating with groundwater. Moreover, the potentially high flood capture capacity of this project suggests significant flood avoidance costs savings to downstream communities along the Kings and San Joaquin Rivers. Our analyses for Terranova Ranch suggest that allocating 25% or more flood flow water towards in lieu recharge and the rest toward direct recharge will result in an economically sustainable recharge approach paid through savings from reduced groundwater pumping. Two important issues need further consideration. First, these practices are likely to leach legacy salts and nitrates from the unsaturated zone into groundwater. We develop a conceptual model of EC movement through the unsaturated zone and estimated through mass balance calculations that approximately 10 kilograms per square meter of salts will be flushed into the groundwater through displacing 12 cubic meters per square meter of unsaturated zone pore water. This flux would increase groundwater salinity but an equivalent amount of water added subsequently is predicted as needed to return to current groundwater salinity levels. All subsequent flood flow capture and recharge is expected to further decrease groundwater salinity levels. Second, the project identified important farm-scale logistical issues including irrigator training; developing cropping plans to integrate farming and recharge activities; upgrading conveyance; and quantifying results. Regional logistical issues also exist related to conveyance, integration with agricultural management, economics, required acreage and Operation and Maintenance (O&M).

O uso combinado de etanol e bebidas energéticas tem aumentado entre adolescentes. Além disso, estudos epidemiológicos indicam que o co-uso aumenta a probabilidade de consumo abusivo e dependência de etanol. Apesar disso, pouco se sabe sobre as consequências neurocomportamentais da co-exposição no cérebro adolescente. Este estudo tem como objetivo avaliar o curso temporal dos efeitos agudos da exposição à bebida energética e/ou etanol na atividade motora e ansiedade no teste de campo aberto, como também, os efeitos agudos ou prolongados sobre aprendizagem/memória e coordenação motora em camundongos adolescentes. Camundongos Suíços foram divididos em 4 grupos: bebidas energéticas e etanol, bebida energética, etanol e água. Três estudos separados foram conduzidos para avaliar cada um dos objetivos específicos deste trabalho. No primeiro estudo, realizado em PN40, os animais receberam a administração de bebida energética (8 ml/kg) e/ou etanol (4 g/kg) por gavagem e após 55 minutos foram submetidos ao teste do campo aberto (sessão 1). Outras duas sessões foram realizadas em sequência usando a metade da dose inicial (sessão 2 e 3). Nos estudos 2 e 3, estudamos os efeitos agudos (PN40) e crônicos (exposição de PN30-40) sobre o teste de esquiva passiva (0,3 mA, 3 s) e sobre o desempenho no teste do cilíndro giratório (sessão de treinamento e após 1 e 3 horas da gavagem das drogas). Em ambos os casos, a dose de bebida energética (8 ml/kg) e/ou etanol (4 g/kg) foi administrada. No teste da esquiva passiva, as sessões de treino e retenção foram realizadas 1 e 24 horas após a administração da droga, respectivamente. No teste do Rotarod, cada sessão foi constituída por 5 tentativas em modelo de aceleramento contínuo (4 a 40 rpm/min em uma tentativa de 2 min). Os nossos dados indicam que a exposição concomitante a bebida energética potencializa o efeito de hiperatividade induzido pelo etanol, como também, gera uma resposta ansiogênica no teste do campo aberto. A exposição aguda ao etanol induz déficit de memória/aprendizagem que não é revertida pela BE. A co-exposição aguda a bebida energética e etanol prolongou incoordenação motora induzida pelo etanol. No entanto, a bebda energética reverteu o comprometimento da coordenação motora gerada pela exposição crônica de etanol em camundongos fêmeas. O presente estudo fornece evidência experimental de que bebida energética e etanol interagem durante a adolescência, resultando em padrões de comportamento que poderiam aumentar o risco de desenvolvimento de abuso ou dependência de etanol. Além disso, os dados indicaram que a exposição aguda à bebida energética não atenuou as consequências negativas geradas pela etanol no desempenho do motor e cognitivo.

Measurement of the physiological effects of the red algal epiphytes Smithora naiadum (Anders.) Hollenberg and Melobesia mediocris (Fosl.) Setch. and Mason on Phyllospadix torreyi Wats. were made near Hopkins Marine Station, Pacific Grove, California. Field studies revealed a significant influence of these epiphytes on both the breakage incidence and length, with that of Melobesia being the most pronounced. Analysis of the photosynthetic rate of the seagrass shows a decrease in the photosynthetic maxima in both epiphytized samples at a light saturating intensity. Under light limiting conditions, an increase in photosynthetic efficiency and a change in chlorophyll a composition in both epiphytized sample types suggest an adaptive mechanism similiar to those found in terrestrial and aquatic shade plants.

A exposição gestacional ao etanol produz um amplo espectro de defeitos neurocomportamentais que podem persistir ao longo da vida. Dentre os distúrbios mais comumente observados estão o transtorno do déficit de atenção e hiperatividade (TDAH) e os déficits de aprendizado e memória. Apesar da grande quantidade de estudos, os mecanismos envolvidos com a manifestação destes transtornos permanecem pouco conhecidos. Estudos em roedores vêm demonstrando que o período equivalente ao terceiro trimestre de gestação é critico para o aparecimento destas alterações comportamentais. Durante este período, que é caracterizado por intensa sinaptogênese, a neurotoxicidade do etanol vem sendo atribuída ao bloqueio dos receptores glutamatérgicos do tipo N-metil-D-aspartato (NMDA) e hiperativação dos receptores do ácido gama-aminobutírico do subtipo A (GABAA). Tendo em vista que ao longo do desenvolvimento estes receptores diferem em relação a função e distribuição espaço-temporal, neste estudo avaliamos a contribuição relativa do bloqueio dos receptores NMDA e hiperativação dos receptores GABAA durante o período equivalente ao terceiro trimestre de gestação para a manifestação da hiperatividade locomotora e para os distúrbios de aprendizado e memória de camundongos pré-púberes. Para tanto, este estudo foi realizado em duas etapas. Na primeira, investigamos os efeitos da exposição isolada ao bloqueador NMDA MK801 (MK) e ao agonista GABAA muscimol (MU). Para tanto, em dias alternados do segundo dia pós-natal (PN2) a PN8, os animais receberam uma injeção intraperitoneal de Salina (SAL), MK nas doses de 0,1, 0,3 ou 0,5 mg/kg ou de MU nas doses de 0,1 0,3 ou 0,5 mg/kg. Na segunda etapa investigamos os efeitos da administração simultânea de MK (0,1mg/kg) e MU (doses 0,02, 0,1 ou 0,5 mg/kg). Em PN25, a atividade locomotora foi automaticamente avaliada por 15 min no teste de campo aberto. Em PN31 e PN32, o aprendizado e memória foi avaliado no teste da esquiva passiva inibitória. Em relação aos resultados da exposição isolada a cada uma das drogas, apenas o tratamento com MK promoveu um aumento dose dependente na atividade locomotora. No teste da esquiva passiva inibitória, os animais expostos as maiores doses de MK e MU apresentaram déficits de aprendizado e memória. Em relação aos resultados da exposição combinada de MK e MU, não foram observadas diferenças significativas entre os grupos na atividade locomotora. Na esquiva passiva inibitória, a administração simultânea de MK e MU, em doses que administradas isoladamente não tiveram efeito, promoveu prejuízos de aprendizado e memória. Nossos resultados sugerem que, enquanto a hiperatividade locomotora está associada apenas com o bloqueio dos receptores NMDA, os déficits de aprendizado e memória podem ser produto de uma ação sinergista do etanol nos dois receptores.

This report describes a surveillance strategy to detect deepwater invasive species in the Northwestern Hawaiian Islands. A need for this strategy was identified in the Papahānaumokuākea Marine National Monument Management Plan and the Monument’s Draft Natural Resources Science Plan. This strategy focuses on detecting two species of concern, the octocoral Carijoa riisei and the red alga Hypnea musciformis. Most research on invasive species in the Hawaiian archipelago has focused on shallow water habitats within the limits of conventional SCUBA (0-30 m). Deeper habitats such as mesophotic reefs are much more difficult to access and consequently little is known about the distribution of deepwater invasive species or their impacts. Recent deepwater (>30 m) sightings of H. musciformis and C. riisei, in and near NWHI, respectively, have prompted a call for further research and surveillance of invasive species in deepwater habitats. This report compiles the most up to date information about these two species of concern in deepwater habitats. A literature search and conversations with subject matter experts was used to identify their current distribution, preferred habitat types, optimal detection methods and ways to efficiently sample the vast extent of NWHI. The proposed sampling strategy prioritizes survey effort where C. riisei and H. musciformis are most likely to be found. At coarse spatial scales (tens to hundreds of kilometers), opportunistic observations and distance from the Main Hawaiian Islands, a principal propagule source, are used to identify high-risk islands and banks. At fine spatial scales (meters to tens of kilometers) a habitat suitability model was developed to identify high-risk habitats. The habitat suitability model focused on habitat preferences of C. riisei, since the species is well studied and adequate data exists to map habitats. There was insufficient information to identify suitable habitat for H. muscifomis. Habitat preferences for the algae are poorly understood and there is a lack of data at relevant spatial scales to map those preferences which are known. The principal habitats identified by the habitat suitability model were ledges and the edges of rugose coral reefs, where the shade loving octocoral would likely be found. Habitat suitability maps were developed for seven atolls and banks to aid in survey site selection. The protocol relied on technical divers to conduct visual surveys of benthic habitats. It was developed to increase the efficiency of surveys, maximize the probability of detection, identify important information relevant to future surveys and standardize results. The strategy, model and protocol were tested during a field mission in 2009 at several atolls and islands in NWHI. The field mission did not detect any invasive species among deepwater habitats and much was learned to improve future surveys. Data gaps and improvements are discussed.

The occurrence of hypoxia, or low dissolved oxygen, is increasing in coastal waters worldwide and represents a significant threat to the health and economy of our Nation’s coasts and Great Lakes. This trend is exemplified most dramatically off the coast of Louisiana and Texas, where the second largest eutrophication-related hypoxic zone in the world is associated with the nutrient pollutant load discharged by the Mississippi and Atchafalaya Rivers. Aquatic organisms require adequate dissolved oxygen to survive. The term “dead zone” is often used in reference to the absence of life (other than bacteria) from habitats that are devoid of oxygen. The inability to escape low oxygen areas makes immobile species, such as oysters and mussels, particularly vulnerable to hypoxia. These organisms can become stressed and may die due to hypoxia, resulting in significant impacts on marine food webs and the economy. Mobile organisms can flee the affected area when dissolved oxygen becomes too low. Nevertheless, fish kills can result from hypoxia, especially when the concentration of dissolved oxygen drops rapidly. New research is clarifying when hypoxia will cause fish kills as opposed to triggering avoidance behavior by fish. Further, new studies are better illustrating how habitat loss associated with hypoxia avoidance can impose ecological and economic costs, such as reduced growth in commercially harvested species and loss of biodiversity, habitat, and biomass. Transient or “diel-cycling” hypoxia, where conditions cycle from supersaturation of oxygen late in the afternoon to hypoxia or anoxia near dawn, most often occurs in shallow, eutrophic systems (e.g., nursery ground habitats) and may have pervasive impacts on living resources because of both its location and frequency of occurrence.

The fishery for spiny lobster Panulirus argus in the Florida Keys National Marine Sanctuary is well chronicled, but little information is available on the prevalence of lost or abandoned lobster traps. In 2007, towed-diver surveys were used to identify and count pieces of trap debris and any other marine debris encountered. Trap debris density (debris incidences/ha) in historic trap-use zones and in representative benthic habitats was estimated. Trap debris was not proportionally distributed with fishing effort. Coral habitats had the greatest density of trap debris despite trap fishers’ reported avoidance of coral reefs while fishing. The accumulation of trap debris on coral emphasizes the role of wind in redistributing traps and trap debris in the sanctuary. We estimated that 85,548 ± 23,387 (mean ± SD) ghost traps and 1,056,127 ± 124,919 nonfishing traps or remnants of traps were present in the study area. Given the large numbers of traps in the fishery and the lack of effective measures for managing and controlling the loss of gear, the generation of trap debris will likely continue in proportion to the number of traps deployed in the fishery. Focused removal of submerged trap debris from especially vulnerable habitats such as reefs and hardbottom, where trap debris density is high, would mitigate key habitat issues but would not address ghost fishing or the cost of lost gear.

本研究首次揭示了七种苏铁类植物叶绿体的超微结构。即苏铁科(Cycadaceae)的攀枝花苏铁(C. panzhihuaensis).苏铁(C. revoluta)、叉叶苏铁(C.micholitzii)、 刺叶苏铁(C.rumphii和多歧苏铁(C.multipinnata),蕨铁科(Stangeriaceae)的蕨铁(Stangeria eriopus)和泽米科(Zamiaceae)的米德尔堡大苏铁(Encephalortos middelburgensis).根据它们叶绿体内膜结构的不同将其大体分为两种类型:1、阴生型叶绿体:多歧苏铁和刺叶苏铁为此类型,它们的叶绿体类囊体垛叠程度高,基粒垛较宽,单个基粒中类囊体的数量很多,有的甚至上百;2、阳生型叶绿体:攀枝花苏铁、米德尔堡大苏铁、苏铁、蕨铁和刺叶苏铁以及外类群的凤尾蕨(Pteris vittata)的叶绿体均有阳生型叶绿体的特征:类囊体膜垛叠程度低,基粒较小。根据它们叶绿体的结构特征,又将其分为两组: ( 1)攀枝花苏铁,米德尔堡大苏铁和叉叶苏铁叶绿体中均有类囊体膜膨大的现象: (2)苏铁和蕨铁在苏铁类中是比较原始的种类,它们的叶绿体与在系统进化上较苏铁类低等的凤尾蕨的叶绿体中都有几个基粒聚集成簇的现象,说明苏铁类在进化的同时一些较原始的性状仍保留了下来。这些苏铁都在温室相同的条件下生长了两年多,但它们的叶绿体结构仍然能够反应原产地生境特点,说明在长期进化中,叶绿体对环境的适应方式已在基因水平上稳定下来,苏铁类植物不同叶绿体结构的形成有其遗传基础。 多歧苏铁,攀枝花苏铁,叉叶苏铁的叶绿体膜垛叠程度依次降低,与此相应,它们的chla/b和F730/F684依次升高,反应了结构与功能的一致性。 选用具有阳生型叶绿体的攀枝花苏铁和有阴生型叶绿体的多歧苏铁用不同的C02浓度(350 umol mol-l和700umol mol-l)处理后观察其叶绿体结构的变化,结果发现C02浓度倍增对它们的叶绿体影响甚微,而作为对照的无论是C3植物小麦(Triticum italica)还是C4植物谷子(Setaria italica)在C02倍增的条件下叶绿体内均有大量淀粉粒积累,并有膜结构改变。这说明苏铁的叶绿体结构有保守性,这有可能是苏铁类能历经亿万年的沧桑而生存下来的结构基础之一。

Four experiments each with three replications were conducted in 12 experimental ponds to control the euglenophytes bloom viz. treatment 1 (T1, covering of one third of the water surface by duckweed (Lemna minor); treatment 2 (T2), application of 123.5 kg lime/ha/month; treatment 3 (T3), use of both duckweed as in T1 and lime as in T2; treatment 4 (T4) was considered as control where neither duckweed nor lime was applied. Fishes comprising of rohu (Labeo rohita), catla (Catla catla), mrigal ( Cirrhinus cirrhosus), silver carp (Hypophthalmichthys molitrix) and silver barb (Barbonymus gonionotus) were stocked at the rate of 1080 fishes/ha with the species ratio of 8:4:6:9:13, respectively. The lowest cell density of euglenophytes was found in the ponds of T3 followed by T2, and T1. In the ponds of T3, euglenophytes bloom did not occur possibly due to alkaline pH, shade and nutrient absorption by duckweed. Thin bloom was observed in the ponds of T1 where pH was neutral or slightly alkaline. The grazing on euglenophytes by the silver carp and silver barb also had some contribution in controlling the bloom. Growth of fishes was comparatively higher in the ponds of T3 and T1, which might be due to better water quality and availability of adequate food while the lower fish growth as recorded from the ponds of T4 might be due to euglenophytes bloom. Thick bloom inhibited light penetration which hampered photosynthesis and growth of other phytoplankton that are the preferred food of planktivorous fishes. Mortality of fishes in ponds having euglenophytes bloom was possibly due to formation of anoxic situation in the early morning or due to the combined effect of anoxic situation and toxic metabolites secretion by the euglenophytes.

Based on the statistical analysis of 119 human and 92 E. coli proteins it was found that for both human and E. coli, the mRNA sequences consisting of tri-codon and tetra-codon with high translation speed preferably code for alpha helices more than for coils. For beta strand, the preference/ avoidance oscillates with the translation speed. Moreover, the non-homogeneous usages of tri-codon and tetra-codon with different translation speeds in a given secondary structure have also been found. These results cannot be simply explained by the effect of stochastic fluctuation.

Matrix anisotropy is important for long term in vivo functionality. However, it is not fully understood how to guide matrix anisotropy in vitro. Experiments suggest actin-mediated cell traction contributes. Although F-actin in 2D displays a stretch-avoidance response, 3D data are lacking. We questioned how cyclic stretch influences F-actin and collagen orientation in 3D. Small-scale cell-populated fibrous tissues were statically constrained and/or cyclically stretched with or without biochemical agents. A rectangular array of silicone posts attached to flexible membranes constrained a mixture of cells, collagen I and matrigel. F-actin orientation was quantified using fiber-tracking software, fitted using a bi-model distribution function. F-actin was biaxially distributed with static constraint. Surprisingly, uniaxial cyclic stretch, only induced a strong stretch-avoidance response (alignment perpendicular to stretching) at tissue surfaces and not in the core. Surface alignment was absent when a ROCK-inhibitor was added, but also when tissues were only statically constrained. Stretch-avoidance was also observed in the tissue core upon MMP1-induced matrix perturbation. Further, a strong stretch-avoidance response was obtained for F-actin and collagen, for immediate cyclic stretching, i.e. stretching before polymerization of the collagen. Results suggest that F-actin stress-fibers avoid cyclic stretch in 3D, unless collagen contact guidance dictates otherwise.

Metal-catalyst-free chemical vapor deposition (CVD) of large area uniform nanocrystalline graphene on oxidized silicon substrates is demonstrated. The material grows slowly, allowing for thickness control down to monolayer graphene. The as-grown thin films are continuous with no observable pinholes, and are smooth and uniform across whole wafers, as inspected by optical-, scanning electron-, and atomic force microscopy. The sp 2 hybridized carbon structure is confirmed by Raman spectroscopy. Room temperature electrical measurements show ohmic behavior (sheet resistance similar to exfoliated graphene) and up to 13 of electric-field effect. The Hall mobility is ∼40 cm 2/Vs, which is an order of magnitude higher than previously reported values for nanocrystalline graphene. Transmission electron microscopy, Raman spectroscopy, and transport measurements indicate a graphene crystalline domain size ∼10 nm. The absence of transfer to another substrate allows avoidance of wrinkles, holes, and etching residues which are usually detrimental to device performance. This work provides a broader perspective of graphene CVD and shows a viable route toward applications involving transparent electrodes. © 2012 American Institute of Physics.
