Microsatellites or simple sequence repeats (SSRs) are ubiquitous in eukaryotic genomes. Single-locus SSR markers have been developed for a number of species, although there is a major bottleneck in developing SSR markers whereby flanking sequences must be known to design 5'-anchors for polymerase chain reaction (PCR) primers. Inter SSR (ISSR) fingerprinting was developed such that no sequence knowledge was required. Primers based on a repeat sequence, such as (CA)(n), can be made with a degenerate 3'-anchor, such as (CA)(8)RG or (AGC)(6)TY. The resultant PCR reaction amplifies the sequence between two SSRs, yielding a multilocus marker system useful for fingerprinting, diversity analysis and genome mapping. PCR products are radiolabelled with P-32 or P-33 via end-labelling or PCR incorporation, and separated on a polyacrylamide sequencing gel prior to autoradiographic visualisation. A typical reaction yields 20-100 bands per lane depending on the species and primer. We have used ISSR fingerprinting in a number of plant species, and report here some results on two important tropical species, sorghum and banana. Previous investigators have demonstrated that ISSR analysis usually detects a higher level of polymorphism than that detected with restriction fragment length polymorphism (RFLP) or random amplified polymorphic DNA (RAPD) analyses. Our data indicate that this is not a result of greater polymorphism genetically, but rather technical reasons related to the detection methodology used for ISSR analysis.

Background: Several studies have already reported the utilization of fibrin glue in microvascular anastomoses to minimize the number of sutures and to decrease the operative time. Despite the good results obtained in most of these experiments, its clinical application has not launched. The aim of this study was to clarify the controversies around the safeness of fibrin glue application in microvascular anastomoses, and also to demonstrate the potential benefits of fibrin glue application in a realistic free flap model. Methods: Twenty-seven rabbits were used in this study The experimental model consisted of a free groin flap transfer to the anterior cervical region. The flap`s circulation was restored by means of an end-to-side anastomosis between the femoral and carotid arteries, and an end-to-end anastomosis between the femoral and external jugular veins. The animals were divided into two groups (n = 10) according to the anastomosis technique: Group I (conventional suture) and group 11 (fibrin glue). Results: The number of sutures required to complete the arterial and venous anastomoses was reduced in 39 and 37% in group 11, respectively. Despite this reduction, the anastomoses maintained adequate patency rates and mechanical strength. Both arterial and venous anastomoses benefited from fibrin glue application, which made them easier and faster to perform. The flaps` ischemic time and the total operative time were also significantly shortened. Conclusions: In this study, the application of fibrin glue in microvascular anastomoses was safe and reliable. The risk-benefit ratio of fibrin glue application in microvascular anastomoses is favorable for its use. (c) 2008 Wiley-Liss, Inc.

Water deficit and ABA application on leaf gas exchange and flavonoid content in marigold (Calendula officinalis L.).The goal of this study was to evaluate the effects of water deficit and abscisic acid (ABA) application on physiological parameters and flavonoid production in marigold plant. The experiment was performed under nursery conditions with potted plants. It was tested water deficit by withholding water (control - diary irrigation, 3, 6 and 9 days without irrigation) followed by 3 ABA concentrations (0, 10 e 100 mu M) applied in the beginning of blooming. It was evaluated the relative water content and the leaf gas exchange using a portable infrared gas analyzer (A: net photosynthesis, gs: stomatal conductance, E: transpiration, Ci: CO(2) intercellular concentration and EUA: water use efficiency). At the end of 9 days of water deficit there were significant decreases in all the characteristics evaluated, independent of ABA application. This suggests that the main effect of ABA was to cause a reduction on gs which was accompanied of a reduction in A, only when the plants were submitted to the water deficit. There was no significant difference among the levels of water deficit tested in relation to the total flavonoid content in inflorescences. However, ABA restricted the flavonoids biosynthesis both in control plant and stressed plants.

Incremental parsing has long been recognized as a technique of great utility in the construction of language-based editors, and correspondingly, the area currently enjoys a mature theory. Unfortunately, many practical considerations have been largely overlooked in previously published algorithms. Many user requirements for an editing system necessarily impact on the design of its incremental parser, but most approaches focus only on one: response time. This paper details an incremental parser based on LR parsing techniques and designed for use in a modeless syntax recognition editor. The nature of this editor places significant demands on the structure and quality of the document representation it uses, and hence, on the parser. The strategy presented here is novel in that both the parser and the representation it constructs are tolerant of the inevitable and frequent syntax errors that arise during editing. This is achieved by a method that differs from conventional error repair techniques, and that is more appropriate for use in an interactive context. Furthermore, the parser aims to minimize disturbance to this representation, not only to ensure other system components can operate incrementally, but also to avoid unfortunate consequences for certain user-oriented services. The algorithm is augmented with a limited form of predictive tree-building, and a technique is presented for the determination of valid symbols for menu-based insertion. Design of liquid retaining structures involves many decisions to be made by the designer based on rules of thumb, heuristics, judgment, code of practice and previous experience. Various design parameters to be chosen include configuration, material, loading, etc. A novice engineer may face many difficulties in the design process. Recent developments in artificial intelligence and emerging field of knowledge-based system (KBS) have made widespread applications in different fields. However, no attempt has been made to apply this intelligent system to the design of liquid retaining structures. The objective of this study is, thus, to develop a KBS that has the ability to assist engineers in the preliminary design of liquid retaining structures. Moreover, it can provide expert advice to the user in selection of design criteria, design parameters and optimum configuration based on minimum cost. The development of a prototype KBS for the design of liquid retaining structures (LIQUID), using blackboard architecture with hybrid knowledge representation techniques including production rule system and object-oriented approach, is presented in this paper. Trials conducted in Queensland, Australia between 1997 and 2002 demonstrated that fungicides belonging to the triazole group were the most effective in minimising the severity of infection of sorghum by Claviceps africana, the causal agent of sorghum ergot. Triadimenol ( as Bayfidan 250EC) at 0.125 kg a. i./ha was the most effective fungicide. A combination of the systemic activated resistance compound acibenzolar-S-methyl ( as Bion 50WG) at 0.05 kg a. i./ha and mancozeb ( as Penncozeb 750DF) at 1.5 kg a. i./ha has the potential to provide protection against the pathogen, should triazole-resistant isolates be detected. Timing and method of fungicide application are important. Our results suggest that the triazole fungicides have no systemic activity in sorghum panicles, necessitating the need for multiple applications from first anthesis to the end of flowering, whereas acibenzolar-S-methyl is most effective when applied 4 days before flowering. The flat fan nozzles tested in the trials provided higher levels of protection against C. africana and greater droplet deposition on panicles than the tested hollow cone nozzles. Application of triadimenol by a fixed wing aircraft was as efficacious as application through a tractor-mounted boom spray.

In microarray studies, the application of clustering techniques is often used to derive meaningful insights into the data. In the past, hierarchical methods have been the primary clustering tool employed to perform this task. The hierarchical algorithms have been mainly applied heuristically to these cluster analysis problems. Further, a major limitation of these methods is their inability to determine the number of clusters. Thus there is a need for a model-based approach to these. clustering problems. To this end, McLachlan et al. [7] developed a mixture model-based algorithm (EMMIX-GENE) for the clustering of tissue samples. To further investigate the EMMIX-GENE procedure as a model-based -approach, we present a case study involving the application of EMMIX-GENE to the breast cancer data as studied recently in van 't Veer et al. [10]. Our analysis considers the problem of clustering the tissue samples on the basis of the genes which is a non-standard problem because the number of genes greatly exceed the number of tissue samples. We demonstrate how EMMIX-GENE can be useful in reducing the initial set of genes down to a more computationally manageable size. The results from this analysis also emphasise the difficulty associated with the task of separating two tissue groups on the basis of a particular subset of genes. These results also shed light on why supervised methods have such a high misallocation error rate for the breast cancer data.

Neste trabalho, analisamos aspectos relacionados a como a tecnologia computacional é utilizada no Atendimento Educacional Especializado (AEE) e como se deu a formação de professores para utilizar esses recursos. Para tanto, delimitamos os seguintes objetivos específicos: investigar a utilização da tecnologia assistiva (TA) computacional no âmbito das salas de recursos multifuncionais (SRM); problematizar as tensões, dificuldades e possibilidades relacionadas à TA com ênfase na tecnologia computacional para o AEE; analisar a formação do professor de educação especial para o AEE tendo como recurso a TA com ênfase na tecnologia computacional, visando à mediação dos processos de aprendizagem. O aporte teórico deste trabalho foi a abordagem histórico-cultural, tomando por referência os estudos de Vigotski e seus colaboradores. Trata-se de uma pesquisa qualitativa que fez uso de diferentes instrumentos metodológicos como, os grupos focais, o questionário online e a entrevista semiestruturada. Para desenvolvê-lo, realizamos a coleta de dados em diferentes contextos, começando pelos grupos focais da pesquisa inaugural do Oneesp, que serviram como dispositivo para esta pesquisa, seguida da aplicação de um questionário aos professores participantes da pesquisa-formação desenvolvida como um desdobramento no estado do Espírito Santo da pesquisa inaugural do Oneesp pelos integrantes do Oeeesp e da aplicação in loco de entrevistas semiestruturadas com professores de educação especial, de uma SRM do Tipo II. Foram oitenta e nove professores participantes na pesquisa do Oneesp, trinta professores na pesquisa-formação do Oeeesp e dois professores para aplicação da entrevista semiestruturada in loco, respectivamente. Esses dois professores participaram tanto da pesquisa do Oneesp como da pesquisa do Oeeesp. Analisamos esses três momentos, dos quais emergiram os apontamentos que nos proporcionaram pensar, com base nas narrativas orais e escritas dos professores: sua formação para uso da TA computacional; seu entendimento sobre sua formação para este fim; seus anseios por uma formação mais direcionada; a forma como utilizam a tecnologia na sala de recursos; seus entendimentos sobre as dificuldades e possibilidades relacionadas a TA com ênfase na tecnologia computacional para o AEE. Após essas análises, concluímos que poucos professores que atuam nas SRM tiveram uma formação que possibilitasse a aplicação das tecnologias computacionais em sua mediação pedagógica, aliando teoria e prática, com momentos de formação que privilegiassem os momentos presenciais e em laboratórios, onde possam interagir com os computadores e suas ferramentas simbólicas. Sem essa familiaridade com os recursos computacionais, os professores acabam sentindo-se inseguros para utilizá-los, deixando de potencializar, pela via desses recursos, os processos de ensinoaprendizagem do aluno com deficiência. Faz-se necessário um investimento nesse tipo de formação e, mais do que isso, que se viabilize para os professores que atuam ou que pretendem atuar nas SRM. A partir de uma formação apropriada é possível fazer com que os professores utilizem os recursos computacionais como mediadores dos processos de ensino-aprendizagem de seus alunos.

Graphical user interfaces (GUIs) are critical components of todays software. Given their increased relevance, correctness and usability of GUIs are becoming essential. This paper describes the latest results in the development of our tool to reverse engineer the GUI layer of interactive computing systems. We use static analysis techniques to generate models of the user interface behaviour from source code. Models help in graphical user interface inspection by allowing designers to concentrate on its more important aspects. One particularly type of model that the tool is able to generate is state machines. The paper shows how graph theory can be useful when applied to these models. A number of metrics and algorithms are used in the analysis of aspects of the user interface's quality. The ultimate goal of the tool is to enable analysis of interactive system through GUIs source code inspection.

In the context of an e ort to develop methodologies to support the evaluation of interactive system, this paper investigates an approach to detect graphical user interface bad smells. Our approach consists in detecting user interface bad smells through model-based reverse engineering from source code. Models are used to de ne which widgets are present in the interface, when can particular graphical user interface (GUI) events occur, under which conditions, which system actions are executed, and which GUI state is generated next.

A sociedade atual é marcada por uma crescente competição entre as organizações, as quais para garantirem a sua sobrevivência têm que evoluir e utilizar a tecnologia de informação mais adequada. Num período em que as PMES portuguesas necessitam de reduzir custos e concentrarem-se mais no seu processo de negócio surge um novo paradigma tecnológico, a Computação na Nuvem, esta ainda é um paradigma bastante recente, onde os recursos de TI que apresentam características de elasticidade e dinâmicas são oferecidos ao cliente como um serviço, no qual este paga apenas o que consome (pagamento por utilização). A Cloud Computing com o seu novo paradigma permite às organizações, principalmente às PMES reduzirem custos, concentrarem-se no seu processo de negócio sem se preocuparem com a parte informática (cópias de segurança, assistência e perda de dados) e obter um retorno mais rápido do seu investimento. Este trabalho consiste no Estudo da Adoção Individual da Cloud Computing no contexto das PMEs Portuguesas, o qual apresenta como principais objetivos obter uma melhor compreensão do conceito Cloud Computing, entender a sua importância para os indivíduos das PMES, identificar os seus fatores de sucesso e insucesso e identificar as limitações sentidas de uma forma generalizada e de uma forma específica para as PMES Portuguesas. Para cumprir os objetivos mencionados anteriormente, efetuou-se uma extensa revisão de literatura, propôs-se um modelo conceptual de investigação com adaptações à Teoria Unificada de Aceitação e Uso de Tecnologia (UTAUT) e utilizou-se como instrumento de recolha de dados um questionário que foi enviado às PMES Portuguesas.

The authors are developing a pilot project for a Municipality in the North of Portugal, envisaging the definition and implementation of an e-marketplace for healthcare and social services, in order to facilitate the interaction between healthcare and social services professionals and people with special needs (or their relatives). Based on the results of a survey on user needs analysis and expectations conducted in 2011, the paper discusses the relevance and interest of such platforms and the main drivers and motivations of the population for using such services, as well as which services would motivate citizens to use the platform. The results of the study will be used to select the products and services perceived to be the most desired by the potential users. The paper thus makes three main contributions: (1) the results of the study confirm the interest and the perceived potential of such a service, from the end-users perspective; (2) the findings support the advantage of expanding this pilot project to a full scale implementation; and (3) the performed analysis improves our understanding of the relations between the characteristics of the inquired population and the perceived interest in such platforms.
