A aplicação superficial de calcário pode ser uma alternativa para a correção da acidez do solo sob plantio direto. Este trabalho avaliou as modificações em alguns atributos químicos provocadas pela aplicação de calcário superficial e incorporado ao solo a partir de pastagem natural. O experimento foi feito na área Experimental do Departamento de Solos da Universidade Federal de Santa Maria (RS), num Argissolo Acinzentado distrófico plíntico. Os tratamentos, com 0,0, 2,0, 8,5 e 17,0 t ha-1 de calcário incorporado na camada de 0-20 cm e em superfície, foram distribuídos em blocos ao acaso, com quatro repetições. Aos 24 meses, reaplicou-se em superfície a dose no tratamento de 2,0 t ha-1. Aos 6, 18, 36 e 48 meses da aplicação do calcário, coletaram-se amostras de solo nas profundidades de 0,0-2,5; 2,5-5,0; 5,0-10,0 e 10,0-15,0 cm. Avaliaram-se os atributos químicos relacionados com a acidez do solo. A correção da acidez do solo foi proporcional à dose de calcário aplicada, independentemente do modo de aplicação. A aplicação de calcário superficial criou uma frente de correção da acidez em profundidade proporcional à dose e ao tempo. Foram necessários 36 meses para que a saturação com alumínio atingisse nível próximo de zero na camada de 0,0-2,5 cm com a aplicação de 2,0 + 2,0 t ha-1; 36 meses para a camada 2,5-5,0 cm com 8,5 t ha-1; e 48 meses para a camada 5,0-10,0 cm com 17,0 t ha-1.

Injection drug use before and after liver transplantation: a retrospective multicenter analysis on incidence and outcome. Clin Transplant 2009 DOI: 10.1111/j.1399-0012.2009.01121.x.Background and aims: Injecting drug use (IDU) before and after liver transplantation (LT) is poorly described. The aim of this study was to quantify relapse and survival in this population and to describe the causes of mortality after LT. Methods: Past injection drug users were identified from the LT listing protocols from four centers in Switzerland and France. Data on survival and relapse were collected and used for uni- and multivariate analysis. Results: Between 1988 and 2006, we identified 59 patients with a past history of IDU. The mean age at transplantation was 42.4 yr and the majority of patients were men (84.7%). The indication for LT was for the vast majority viral cirrhosis accounting for 91.5% of cases, while alcoholic cirrhosis was 5.1%. There were 16.9% of patients who had a substitution therapy before and 6.8% who continued after LT. Two patients (3.4%) relapsed into IDU after LT and died at 18 and 41 months. The mean follow-up was 51 months. Overall survival was 84%, 66%, and 61% at 1, 5, and 10 yr after transplantation. Conclusions: Documented IDU was rare in liver transplanted patients. Past IDU was not associated with poorer survival after LT, and relapse after LT occurred in 3.4%.

This study was conducted to assess the pharmacologic properties of the new orally active angiotensin II subtype I (AT1) antagonist UR-7247, a product with a half-life >100 h in humans. The experiment was designed as an open-label, single-dose administration study with four parallel groups of four healthy men receiving increasing single oral doses (2.5, 5, and 10 mg) of UR-7247 or losartan, 100 mg. Angiotensin II receptor blockade was investigated < or =96 h after drug intake, with three independent methods [i.e., the inhibition of blood pressure (BP) response to exogenous Ang II, an in vitro Ang II-receptor assay (RRA), and the reactive increase in plasma angiotensin II. Plasma drug levels also were measured. The degree of blockade observed in vivo was statistically significant < or = 96 h with all UR-7247 doses for diastolic BP (p < 0.05) and < or =48 h for systolic BP. The maximal inhibition achieved with 10 mg UR-7247 was measured 6-24 h after drug intake and reached 54 +/- 17% and 48 +/- 20% for diastolic and systolic responses, respectively. Losartan, 100 mg, induced a greater short-term AT1-receptor blockade than 2.5- and 5.0-mg doses of UR-7247 (p < 0.001 for diastolic BP), but the UR-7247 effect was longer lasting. In vivo, no significant difference was observed between 10 mg UR-7247 and 100 mg losartan 4 h after drug intake, but in vitro, the blockade achieved with 100 mg losartan was higher than that seen with UR-7247. Finally, the results confirm that UR-7247 has a very long plasma elimination half-life, which may be due to a high but also tight binding to protein binding sites. In conclusion, UR-7247 is a long-lasting, well-tolerated AT1 receptor in healthy subjects.

In conducting genome-wide association studies (GWAS), analytical approaches leveraging biological information may further understanding of the pathophysiology of clinical traits. To discover novel associations with estimated glomerular filtration rate (eGFR), a measure of kidney function, we developed a strategy for integrating prior biological knowledge into the existing GWAS data for eGFR from the CKDGen Consortium. Our strategy focuses on single nucleotide polymorphism (SNPs) in genes that are connected by functional evidence, determined by literature mining and gene ontology (GO) hierarchies, to genes near previously validated eGFR associations. It then requires association thresholds consistent with multiple testing, and finally evaluates novel candidates by independent replication. Among the samples of European ancestry, we identified a genome-wide significant SNP in FBXL20 (P = 5.6 × 10(-9)) in meta-analysis of all available data, and additional SNPs at the INHBC, LRP2, PLEKHA1, SLC3A2 and SLC7A6 genes meeting multiple-testing corrected significance for replication and overall P-values of 4.5 × 10(-4)-2.2 × 10(-7). Neither the novel PLEKHA1 nor FBXL20 associations, both further supported by association with eGFR among African Americans and with transcript abundance, would have been implicated by eGFR candidate gene approaches. LRP2, encoding the megalin receptor, was identified through connection with the previously known eGFR gene DAB2 and extends understanding of the megalin system in kidney function. These findings highlight integration of existing genome-wide association data with independent biological knowledge to uncover novel candidate eGFR associations, including candidates lacking known connections to kidney-specific pathways. The strategy may also be applicable to other clinical phenotypes, although more testing will be needed to assess its potential for discovery in general.

Um dos desafios atuais da pesquisa é encontrar plantas e microssimbiontes tolerantes e que possibilitem a revegetação de áreas degradadas por excesso de metais pesados. Este experimento foi realizado no período de agosto a dezembro de 1998, em casa de vegetação do Departamento de Ciência do Solo da UFLA, Lavras (MG), com o objetivo de avaliar a tolerância a metais pesados e a capacidade de estabelecimento de simbiose de rizóbio de diferentes origens com Enterolobium contortisiliquum (tamboril), Acacia mangium (acácia) e Sesbania virgata (sesbânia), em misturas de solos, que continham proporções de solo contaminado (PSC): (0, 15, 30, 45 e 60% v/v) com Zn, Cd, Pb e Cu (18.600, 135, 600 e 596 mg dm-3, extraídos por aqua regia, respectivamente), diluído em Latossolo Vermelho distrófico. Estirpes recomendadas (E) e isolados de solo contaminado (ISC) e de solo não contaminado (ISNC), cuja tolerância a Cu, Cd e Zn foi determinada previamente "in vitro", foram inoculados. O aumento da PSC nas misturas inibiu o crescimento vegetativo, a produção de matéria seca e a nodulação das três espécies. A simbiose tamboril-BR4406 foi a mais tolerante e acácia-BR3617 a mais sensível à contaminação do solo. Os ISC que foram mais tolerantes "in vitro" formaram nódulos eficientes em solo sem contaminação, mas foram ineficientes em solos contaminados. Na PSC 15% (Zn = 750; Cd = 22,1; Pb = 65,1 e Cu = 111 mg dm-3 extraídos por DTPA) a atividade específica da nitrogenase aumentou 5 e 10 vezes em relação ao solo sem contaminação para as simbioses sesbânia-BR5401 e tamboril-BR4406, respectivamente. A tolerância de rizóbio a metais "in vitro" não correspondeu à tolerância da simbiose em solo contaminado.

Para estudar o efeito do Al no crescimento inicial de duas espécies florestais, foi instalado um experimento em novembro de 1998, em solução nutritiva, em casa de vegetação do Departamento de Ciência do Solo da Universidade Federal de Lavras. Após duas semanas em solução nutritiva diluída a 1/3 e sem Al, duas plantas de Moringa (Moringa oleifera) e duas de Angico (Anandenantha peregrina) foram transplantadas para vasos de 1,5 L e crescidas em solução nutritiva. Os teores de Al utilizados foram de 0,0; 2,5; 5,0; 7,5; e 10,0 mg L-1, com pH 4,0 ± 0,2, ajustado diariamente durante 70 dias. Após este período, foi determinada a altura das plantas, as quais, posteriormente, foram divididas em parte aérea e raiz, para determinação da matéria seca e dos teores de N e P. Os teores crescentes de Al reduziram progressivamente a altura das plantas e a produção de matéria seca das raízes nas duas espécies e da parte aérea no Angico. O teor de nitrogênio na Moringa diminuiu com o incremento nos teores de Al. O teor de fósforo na parte aérea da Moringa e do Angico diminui a partir de 5 mg L-1 de Al na solução nutritiva.

Na coleta da solução de solos alagados, é importante evitar o contato do oxigênio com a solução para prevenir reações redox que podem modificar sua composição química. Neste trabalho, realizado em casa de vegetação, foram feitos estudos sobre o estado de oxirredução e sobre a composição química de um Planossolo, em diferentes profundidades e tempos de alagamento, visando propor um novo método para a coleta da solução de solos alagados. O estudo foi feito em vasos plásticos com 7 kg de material de solo proveniente da camada superficial (0-20 cm), num delineamento inteiramente casualizado, com três repetições. O dispositivo de coleta da solução do solo consistiu de um tubo de polietileno com 70 cm de comprimento e 0,6 cm de diâmetro interno, com duas fileiras de orifícios de 1 mm de diâmetro, recoberto com tela de nylon de 400 mesh, construído na forma de espiral e acomodado nas profundidades de 2, 5 e 10 cm. A este dispositivo foi acoplado um sifão de vidro para a coleta da solução. Após a instalação do sistema coletor, o solo foi mantido alagado por 109 dias, tendo sido feitas coletas semanais da solução nas três profundidades. Foram analisados os valores de Eh e pH e os teores de Mn2+, Fe2+, Ca2+, Mg2+ e K+ na solução. As condições de oxirredução e os teores de cátions na solução nas diferentes profundidades, durante o período em que o solo se manteve alagado, permitiram concluir que o método utilizado pode ser indicado para coleta da solução de solos alagados, embora haja necessidade de estudos mais aprofundados sobre o assunto.

14C dating models are limited when considering recent groundwater for which the carbon isotopic signature of the total dissolved inorganic carbon (TDIC) is mainly acquired in the unsaturated zone. Reducing the uncertainties of dating thus implies a better identification of the processes controlling the carbon isotopic composition of the TDIC during groundwater recharge. Geochemical interactions between gas, water and carbonates in the unsaturated zone were investigated for two aquifers (the carbonate-free Fontainebleau sands and carbonate-bearing Astian sands, France) in order to identify the respective roles of CO2 and carbonates on the carbon isotopic signatures of the TDIC; this analysis is usually approached using open or closed system terms. Under fully open system conditions, the seasonality of the 13C values in the soil CO2 can lead to important uncertainties regarding the so-called "initial 14C activity" used in 14C correction models. In a carbonate-bearing unsaturated zone such as in the Astian aquifer, we show that an approach based on fully open or closed system conditions is not appropriate. Although the chemical saturation between water and calcite occurs rapidly within the first metre of the unsaturated zone, the carbon isotopic contents (δ13C) of the CO2 and the TDIC evolve downward, impacted by the dissolution-precipitation of the carbonates. In this study, we propose a numerical approach to describe this evolution. The δ13C and the A 14C (radiocarbon activity) of the TDIC at the base of the carbonate-hearing unsaturated zone depends on (i) the δ13C and the A 14C of the TDIC in the soil determined by the soil CO2, (ii) the water's residence time in the unsaturated zone and (iii) the carbonate precipitation-dissolution fluxes. In this type of situation, the carbonate δ13C-A 14C evolutions indicate the presence of secondary calcite and permit the calculation of its accretion flux, equal to ~ 4.5 ± 0.5 x 10-9 mol grock-1 yr-1. More generally, for other sites under temperate climate and with similar properties to the Astian sands site, this approach allows for a reliable determination of the carbon isotopic composition at the base of the unsaturated zone as the indispensable "input function" data of the carbon cycle into the aquifer.

OBJECTIVE: To assess the thermogenic response of dopamine at three different infusion rates and to analyze its effects on various biochemical variables. DESIGN: Randomized sequential experimental treatment bracketed by control periods. PATIENTS: Eight young healthy male volunteers with normal body weight (51 to 89 kg). INTERVENTIONS: Three experimental periods during which dopamine was administered iv in a randomized order at rates of 2.5, 5, or 10 micrograms/kg.min with one preinfusion baseline and two recovery periods in between. MEASUREMENTS AND MAIN RESULTS: A significant (p less than .01) increase in resting energy expenditure was observed in response to the two highest dopamine infusion rates (5 and 10 micrograms/kg.min), corresponding to 6% and 15% median increases, respectively, as compared with preinfusion values. At the lowest dopamine infusion rate, no variation in resting energy expenditure was observed. Dopamine induced a significant (p less than .01) increase in hyperglycemia at all three infusion rates, and, at the highest infusion rate, dopamine induced a significant (p less than .05) increase of plasma free fatty acid concentrations. Insulin plasma concentrations were significantly (p less than .05 to p less than 0.1) increased at the three dopamine infusion rates. CONCLUSIONS: Dopamine infusion produces a dose-dependent thermogenic effect and induces various metabolic actions in man.

In Brazil, Bradyrhizobium inoculation has successfully replaced the use of N fertilizer on soybean [Glycine max (L) Merr.] crops. However, with the expansion of no-tillage cropping systems in the Cerrados region, the idea that it is necessary to use small N rates at the sowing to overcome problems related with N immobilization has become widespread, mainly when soybean is cultivated after a non-legume crop. In this study we examined soybean response to small rates of N fertilizer under no-tillage (NT) and conventional tillage (CT) systems. Four experiments (a completely randomized block with five replicates) were carried out in a red yellow oxisol, during the periods of 1998/1999 and 1999/ 2000, under NT and CT. The treatments consisted of four urea rates (0, 20, 30 and 40 kg ha-1 N). All treatments were inoculated with Bradyrhizobium japonicum strains SEMIA 5080 and SEMIA 5079, in the proportion 1 kg of peat inoculant (1,5 x 10(9) cells g-1) per 50 kg of seeds. In both experiments, soybean was cultivated after corn and the N fertilizer was band applied at sowing. In all experiments, N rates promoted reductions of up to 50 % in the nodule number at 15 days after the emergence. Regardless of the management system, these reductions disappeared at the flowering stage and there was no effect of N rates on either the number and dry weight of nodules or on soybean yields. Therefore, in the Brazilian Cerrados, when an efficient symbiosis is established, it is not necessary to apply starter N rates on soybean, even when cultivated under notillage systems.

Para avaliar um esquema de fracionamento de zinco em Argissolo arenoso e suas relações com a disponibilidade de Zn para Cynodon spp cv. Tifton-85, realizou-se um experimento em casa de vegetação, em esquema fatorial 5 x 2 x 2 (5 doses de Zn, 2 doses de calcário e 2 épocas de amostragem), em delineamento inteiramente casualizado, com três repetições. As doses de Zn foram de 0; 2,5; 5,0; 7,5 e 10 mg dm-3; metade dos vasos não recebeu calagem (V = 42 %) e metade recebeu a calagem com vistas em elevar o índice de saturação por bases a 70 %. As épocas de amostragem foram 30 e 150 dias após aplicação de Zn, respectivamente, antes do plantio e depois do 3º corte de Tifton-85. A aplicação de Zn resultou em aumento significativo do elemento nas frações: trocável, óxidos de Mn, matéria orgânica e óxidos de Fe. O Zn ligado aos óxidos de Mn aumentou significativamente com a calagem. Após 150 dias de experimentação, houve diminuição do Zn trocável, ligado aos óxidos de Mn, à matéria orgânica e aos óxidos de Fe, e aumento na fração residual. A distribuição de Zn nas frações do solo foi: residual > óxidos de Fe > óxidos de Mn > trocável > matéria orgânica. As relações entre as características estudadas mostraram que tanto o Zn-DTPA quanto o Zn trocável, ligado à M.O. e aos óxidos de Mn, foram eficientes para representar o Zn absorvido pela planta.

PURPOSE: Recently, a 76-gene prognostic signature able to predict distant metastases in lymph node-negative (N(-)) breast cancer patients was reported. The aims of this study conducted by TRANSBIG were to independently validate these results and to compare the outcome with clinical risk assessment. EXPERIMENTAL DESIGN: Gene expression profiling of frozen samples from 198 N(-) systemically untreated patients was done at the Bordet Institute, blinded to clinical data and independent of Veridex. Genomic risk was defined by Veridex, blinded to clinical data. Survival analyses, done by an independent statistician, were done with the genomic risk and adjusted for the clinical risk, defined by Adjuvant! Online. RESULTS: The actual 5- and 10-year time to distant metastasis were 98% (88-100%) and 94% (83-98%), respectively, for the good profile group and 76% (68-82%) and 73% (65-79%), respectively, for the poor profile group. The actual 5- and 10-year overall survival were 98% (88-100%) and 87% (73-94%), respectively, for the good profile group and 84% (77-89%) and 72% (63-78%), respectively, for the poor profile group. We observed a strong time dependence of this signature, leading to an adjusted hazard ratio of 13.58 (1.85-99.63) and 8.20 (1.10-60.90) at 5 years and 5.11 (1.57-16.67) and 2.55 (1.07-6.10) at 10 years for time to distant metastasis and overall survival, respectively. CONCLUSION: This independent validation confirmed the performance of the 76-gene signature and adds to the growing evidence that gene expression signatures are of clinical relevance, especially for identifying patients at high risk of early distant metastases.
