Catalytically inactive phospholipase A2 (PLA2) homologues play key roles in the pathogenesis induced by snake envenomation, causing extensive tissue damage via a mechanism still unknown. Although, the amino acid residues directly involved in catalysis are conserved, the substitution of Asp49 by Arg/Lys/Gln or Ser prevents the binding of the essential calcium ion and hence these proteins are incapable of hydrolyzing phospholipids. In this work, the crystal structure of a Lys49-PLA2 homologue from Bothrops brazili (MTX-II) was solved in two conformational states: (a) native, with Lys49 singly coordinated by the backbone oxygen atom of Val31 and (b) complexed with tetraethylene glycol (TTEG). Interestingly, the TTEG molecule was observed in two different coordination cages depending on the orientation of the nominal calcium-binding loop and of the residue Lys49. These structural observations indicate a direct role for the residue Lys49 in the functioning of a catalytically inactive PLA2 homologue suggesting a contribution of the active site-like region in the expression of pharmacological effects such as myotoxicity and edema formation. Despite the several crystal structures of Lys49-PLA2 homologues already determined, their biological assembly remains controversial with two possible conformations. The extended dimer with the hydrophobic channel exposed to the solvent and the compact dimer in which the active site-like region is occluded by the dimeric interface. In the MTX-II crystal packing analysis was found only the extended dimer as a possible stable quaternary arrangement. © 2012 Elsevier B.V.

For intricate automotive systems that enclose several components, such as gearboxes, an important aspect of the design is defining the correct assembly parameters. A proper assembly can ensure optimized operating conditions and therefore the components can achieve a longer life. In the case of the support bearings applied to front-axle lightweight differentials, the assembly preload is a major aspect for an adequate performance of the system. During the design phase it is imperative to define reference values to this preload, so the application would endure its requirements. However, with the assistance of computer simulations, it is possible to determine an optimum condition of operation, i.e. optimum pre-load, which would increase the system reliability. This paper presents a study on the influence of preload on the rating life of tapered roller bearings applied to light-weight front axle differentials, evaluating how preload affects several key parameters such as rating life and displacement of components, taking into account the flexibility of the surrounding differential housing. An exocellular β-(1→6)-d-glucan (lasiodiplodan) produced by a strain of Lasiodiplodia theobromae (MMLR) grown on sucrose was derivatized by sulfonation to promote anticoagulant activity. The structural features of the sulfonated β-(1→6)-d-glucan were investigated by UV-vis, FT-IR and 13C NMR spectroscopy, and the anticoagulant activity was investigated by the classical coagulation assays APTT, PT and TT using heparin as standard. The content of sulfur and degree of substitution of the sulfonated glucan was 11.73% and 0.95, respectively. UV spectroscopy showed a band at 261 nm due to the unsaturated bond formed in the sulfonation reaction. Results of FT-IR and 13C NMR indicated that sulfonyl groups were inserted on the polysaccharide. The sulfonated β-(1→6)-d-glucan presented anticoagulant activity as demonstrated by the increase in dose dependence of APTT and TT, and these actions most likely occurred because of the inserted sulfonate groups on the polysaccharide. The lasiodiplodan did not inhibit the coagulation tests. © 2012 Elsevier Ltd.

The meta-analysis was used to evaluate the performance of piglets in post-weaning period, without imposition of sanitary challenge and fed diets containing blood plasma, obtained by spray-dried process (SDBP). Piglets are faced with normal challenges in post-weaning period such as environmental stress and the substitution of the liquid diet to a solid one. References regarding sanitary challenges were disregarded in this study. Only data regarding normal and expected challenges were considered. Data were obtained from indexed journals with information extracted from the material, methods and results sections of pre-selected scientific articles. First, the database was analyzed graphically to observe the distribution of data and presence of outliers. Afterwards correlation analysis and variance-covariance analyses were carried out. The database contained a total of 23 articles. The average initial weight of the piglets was 8.02. kg (4.00-9.28. kg) and the average initial age was 27 days (14-32 days). The average duration of feeding diets containing spray-dried blood plasma (SDBP) was 9 days (6-28 days). SDBP increased the feed conversion by 20.2% (P<0.05) during the initial period. Feed conversion during the total period was 10.2% higher (P<0.05) for animals fed with SDBP. Average daily weight gain and daily feed intake were not affected (P>0.05) during the entire period, but average daily gain was higher (P<0.05) for animals fed with SDBP during the initial period. The initial age of supplementation influenced the average daily weight gain and average daily feed intake of animals fed with SDBP. Better results were obtained than those obtained for animals up to 35 days of age fed diets without added SDBP supplementation. In early post-weaning period for piglets weaned up to 35 days of age, the SDBP supplementation positively influenced the average daily weight gain and feed conversion. © 2013 Elsevier B.V.

We evaluated messenger RNA (mRNA) expression of the growth-hormone (GHR) and insulin-like growth factor (IGF-1) genes in 28-day-old Japanese meat quails fed diets containing 0, 8, or 12% dietary glycerol in substitution of corn. Total RNA was extracted from the breast muscle and the DNA was amplified with specific primers using real-time PCR. Feed conversion ratio and feed intake were evaluated. The birds fed 8 and 12% glycerol presented higher IGF-1 mRNA expression [0.059 and 0.049 arbitrary units (AU), respectively] relative to those not fed with glycerol (0.029 AU), while 12% glycerol reduced GHR mRNA expression (0.022 AU). Dietary inclusion of 8% glycerol promoted similar performance results (feed conversion) as the diet with no glycerol. We conclude that inclusion of glycerol in the diet affects GHR and IGF-1 gene expression in Japanese meat quails. However, considering the performance results and the expression of the GHR and IGF-1 genes, 8% glycerol may be safely included in the diet of meat quails. © FUNPEC-RP.

The global radiation incident on a tilted surfaces consists of components direct, diffuse and reflected from the ground. On a hourly database, the direct radiation can be calculated by geometric projections (ratio of the incidence angle to the solar zenith angle). The reflected radiation has a small effect on calculations and may be calculated with an isotropic model. Both components presents dependence of measures in incidence or horizontal surface. The great difficulty is to evaluate the diffuse radiation by variations of circumsolar, brightness horizontal, isotropic and anisotropic subcomponents. This study evaluated twenty models to estimate hourly diffuse radiation incident on tilted surfaces at 12.85° (latitude - 10°), 22.85° (latitude) and 32.85° (latitude + 10°) facing to North, under different cloudiness sky conditions, in Botucatu, São Paulo State, Brazil (22°53' S, 48°26' W and 786 m above the mean sea level). In contrast, models for estimating the diffuse component show major differences, which justify the validation for local calibrations. There is a decrease of the maximum total radiation scattered with increasing atmospheric transmissivity and inclination angle. The best results are obtained by anisotropic models: Ma and Iqbal, Hay, Reindl et al. and Willmott; isotropic: Badescu and Koronakis, and the Circumsolar model. The increase of the inclination angle allows for a reduction in the performance of statistical parametric models for estimating the hourly diffuse radiation.

Este trabalho busca avaliar e quantificar os impactos na disponibilidade hídrica local na região da Amazônia Central, decorrentes de possíveis mudanças climáticas e modificações na cobertura vegetal, por meio de um experimento de simulação numérica com o modelo de biosfera Common Land Model (CLM), no modo “off line”. Os resultados de 9 modelos climáticos acoplados Oceano-Atmosfera para três cenários de alterações climáticas do Painel Intergovernamental sobre Mudanças Climáticas (IPCC-AR4) foram utilizados para compor a base das forçantes climáticas do modelo de biosfera CLM. Em relação ao uso da terra, foram utilizados cenários de dinâmica de desmatamento para a região no caso “business as usual” previstos para cada ano do período de 2001 a 2050. A área de estudo compreende o domínio da drenagem da Bacia do Rio Cuieiras na Amazônia Central. A partir dos resultados dos modelos analisaram-se, para cada conjunto de simulações, as incertezas das projeções em relação à precipitação e a temperatura e o impacto no ciclo hidrológico terrestre, considerando a variabilidade entre os modelos e os cenários de emissões de CO2; bem como as alterações nas componentes do balanço de água e energia a superfície associada a variações progressivas na cobertura de floresta e sua substituição por pastagem. Os resultados indicam que diante de um cenário de mudanças climáticas que resultem em uma diminuição (aumento) persistente na precipitação média anual, tanto o escoamento quanto o armazenamento de água no solo serão diretamente afetados. Em relação às alterações na cobertura vegetal tanto as componentes dos balanços de água e energia foram significativamente afetados pela substituição da floresta por pastagem, apresentando redução na evapotranspiração e aumentos no armazenamento de água no solo e do escoamento total.

ABSTRACT: The objective of this study was to evaluate the effect of inclusion of lipid residue of biodiesel originated in the processing of palm oil (Elaeis guineensis) in the diet on the digestibility of feedlot lambs. Twenty-five crossbred male castrated lambs, weighing 20±1.61 kg, were distributed in randomized blocks with five treatments and five replications. The experimental period lasted 22 days; 15 for diet adaptation, 2 for the adaptation to the indicator LIPE (lignin from Eucalyptus grandis isolated, purified and enriched, UFMG, Minas Gerais) and 5 for fecal sampling. Diets were formulated with 64% concentrate based on corn and soybean meal, 31% Massai grass (Panicum maximum cv. Massai) hay and 5% lipid supplementation from increasing levels of substitution of 0, 25, 50, 75 and 100% of palm oil for biodiesel oil from palm residue. The lambs were offered two meals a day, at 7h00 and 16h00. There was linear effect of inclusion of the residue from palm oil biodiesel on dry matter intake. There was no change in digestibility of nutrients except for ether extract. The use of biodiesel from palm oil residue up to 100% replacement for the lipid supplementation of sheep positively influences the consumption without altering the digestibility of nutrients.

Os subprodutos farelo de coco (FC) e a torta de amêndoa de dendê (TAD) disponíveis na agroindústria da Amazônia Oriental podem ser utilizados como alimentos alternativos na substituição de concentrados convencionais, porém com poucas informações a respeito da sua composição bromatológica, e sua relação com as características produtivas e reprodutivas dos animais. Assim, este trabalho visou avaliar o consumo alimentar, desempenho ponderal, perímetro escrotal, qualidade seminal e níveis de testosterona de búfalos suplementados com rações experimentais à base de TAD ou FC. O trabalho foi desenvolvido na Unidade de Pesquisa Animal “Senador Álvaro Adolpho”, da Embrapa Amazônia Oriental, em Belém-PA. Foram utilizados como doadores de sêmen 15 machos bubalinos (Bubalus bubalis) adultos, divididos em três grupos experimentais: Controle, Base-FC (farelo de coco) e Base-TAD (torta de amêndoa de dendê). Os concentrados experimentais foram fornecidos individualmente na proporção de 1% do peso vivo (PV) de cada animal, diariamente. As colheitas seminais (n=173) foram realizadas semanalmente, e iniciaram 112 dias após o início da suplementação. Foram avaliadas as características físicas (aspecto, cor, volume, concentração, pH, motilidade e vigor espermáticos) e morfológicas (defeitos espermáticos e integridade de membrana plasmática) do sêmen. Os resultados foram submetidos à análise de variâncias, pelo comando PROG GLM do SAS, com comparações de médias feitas por Teste t (P<0,05). O consumo médio de matéria seca dos concentrados experimentais foi de 4,778 ± 1,233 kg, no Grupo Controle, 3,112 ± 0,693 kg, no Base-FC, e 4,558 ± 1,077 kg no Base-TAD (P>0,05). Os pesos médios dos animais foram 591, 4 ± 103,3 kg, 566,4 ± 94,1 kg e 578,3 ± 107,5 kg (P>0,05), e as medidas de perímetro escrotal de 34,6 ± 2,0 cm, 33,0 ± 1,9 cm e 32,7 ± 1,9 cm, spectivamente, nos grupos Controle, Base-FC e Base-TAD. Não houve diferença na concentração espermática (x106 sptz/mL) entre Controle (1326,3 ± 893,8), Base-FC (1.698,1 ± 1023,0) e Base-TAD (1.003,2 ± 569,0). A integridade da membrana plasmática (%) foi de 68,0 ± 19,5, 72,0 ± 22,6 e 82,1 ± 12,2, respectivamente para Controle, Base-FC e Base-TAD, juntamente com a motilidade espermática progressiva (%), foram maiores (P<0,05) no grupo Base-TAD (71,7 ± 15,1%), quando comparadas ao Controle (59,3 ± 20,5%) e Base-FC (56,7 ± 24,8%). Foram observadas correlações entre as variáveis de resposta. O consumo dos concentrados experimentais e ingredientes contidos neles, como PB, lipídeos, macro, micronutrientes e metais foram maiores no Grupo Base-TAD, com exceção de K, maior no Grupo Controle, e de Ti, maior no Grupo Base-FC. Em relação os parâmetros seminais não houve diferença significativa (P>0,05) no volume espermático, movimento de massa, vigor espermático, pH e patologias espermáticas, contudo, maiores concentrações espermáticas foram observadas no Grupo Base-FC e, maior motilidade espermática e integridade de membrana plasmática no Grupo Base-TAD. Deste modo, podemos concluir que as rações à base de subprodutos, como farelo de coco e torta de amêndoa de dendê podem ser utilizadas para touros reprodutores bubalinos, contudo, a melhor qualidade seminal foi observada nas rações à base de torta de amêndoa de dendê.
