O biodiesel produzido para ser comercializado no Brasil deve estar de acordo aos padrões de qualidade estabelecidos pela resolução de 4 de fevereiro de 2010 da Agência Nacional do Petróleo, Gás Natural e do Biocombustível (ANP, 04/2010). Neste trabalho, foi estudada a aplicação dos métodos ASTM D 6584 e EN 14105 para o biodiesel de mamona e biodiesel oriundo de rota etílica. Ambos os métodos empregam GC (Cromatografia Gasosa, do inglês Gas Chromatography) com FID (Detecção por Ionização em Chama, do inglês Flame Ionization Detection) e reação de sililação com N-metil-N-(trimetilsilil)trifluoracetamida (MSTFA). Os compostos foram identificados para quantificação pelos tempos de retenção, para os diglicerídeos e triglicerídeos foram utilizadas bandas de tempo de retenção. Os parâmetros de validação considerados foram: curva analítica, linearidade, sensibilidade, robustez, precisão e exatidão. Os métodos ASTM D 6584 e EN 14105 apresentaram sensibilidade semelhante para todos os compostos. Foram escolhidas as condições cromatográficas estabelecidas pelo método ASTM D 6584 por ser mais rápido que o EN14105 e ter sensibilidade semelhante. O método apresentou boa linearidade com todas as curvas analíticas com r maiores que 0,999. A reação de sililação com MSTFA foi otimizada para o biodiesel etílico de mamona em virtude da sua composição química. Um volume de 500 μL de MSTFA foi escolhido para realização dos ensaios de exatidão e precisão. Os valores de exatidão ficaram entre 67 e 145,9% com valores de precisão menores que 11%. Foi avaliada a ocorrência de efeito matriz para biodiesel etílico de mamona, sendo que esse efeito foi considerado baixo para glicerol, monooleína e dioleína e médio para trioleína. Mesmo havendo efeito de matriz o preparo das curvas analíticas em solvente conforme sugerido pelos métodos de referência foi mantido. O método foi robusto frente às variações da composição química da matriz. Na aplicação do método, esse se mostrou adequado para amostras de biodiesel etílico de mamona, de girassol e da mistura de sebo e soja.

The period of the 18th and 19th centuries was one of great change in the Highlands and Islands of Scotland. Improvement, the Jacobite Rebellions, and the Clearances transformed its communities and landscapes. These events have rightly been a focus of research. However, archaeological approaches have often sought simply to illustrate these processes, rather than create new narratives about life in the past. The resulting picture of the period can over-emphasise economic change whilst failing to reflect the richness and variety of everyday life. This thesis aims to suggest a new approach to the place and period, one which addresses matters often ignored in previous work. Whilst it has an archaeological sensibility, it draws on ideas from outside archaeology, such as landscape theory and on Gaelic oral tradition, an underused resource, to create a novel and broad-based approach to the period. An important part of the method is a synchronic approach that seeks to reconstruct the experience of the landscape at very particular times, engaging fully with the everyday experience of landscape rather than grand historical narratives. Two Hebridean case studies are utilised: Hiort (St Kilda) and Loch Aoineart, South Uist. Thematic discussions drawn from these landscapes are intended as critical assessments of the efficacy of the approach, as well as new narratives about life in the past in themselves. The thesis concludes by comparing the two case studies, reflecting on the merits of the approach, discussing recurrent themes in the work, and considering its wider context and implications. It is concluded that taking a novel approach to the case study landscapes can create narratives that often contrast or expand upon those produced by previous scholars, allow for a more detailed consideration of everyday life in the period, and open up new areas for archaeological enquiry. The extensive and critical use of evidence from Gaelic oral tradition is highlighted as crucial in understanding life and society in the period. The thesis questions the utility of grand historical narratives as a framework for archaeological study of post-medieval Gaeldom and suggests that our understanding of the past is best served by approaching the evidence in ways which allows for many different voices and stories from the past to emerge.

Background: The nitration of tyrosine residues in proteins is associated with nitrosative stress, resulting in the formation of 3-nitrotyrosine (3-NT). 3-NT levels in biological samples have been associated with numerous physiological and pathological conditions. For this reason, several attempts have been made in order to develop methods that accurately quantify 3-NT in biological samples. Regarding chromatographic methods, they seem to be very accurate, showing very good sensibility and specificity. However, accurate quantification of this molecule, which is present at very low concentrations both at physiological and pathological states, is always a complex task and a target of intense research. Objectives: We aimed to develop a simple, rapid, low-cost and sensitive 3-NT quantification method for use in medical laboratories as an additional tool for diagnosis and/or treatment monitoring of a wide range of pathologies. We also aimed to evaluate the performance of the HPLC-based method developed here in a wide range of biological matrices. Material and methods: All experiments were performed on a Hitachi LaChrom Elite® HPLC system and separation was carried out using a Lichrocart® 250-4 Lichrospher 100 RP-18 (5μm) column. The method was further validated according to ICH guidelines. The biological matrices tested were serum, whole blood, urine, B16 F-10 melanoma cell line, growth medium conditioned with the same cell line, bacterial and yeast suspensions. Results: From all the protocols tested, the best results were obtained using 0.5% CH3COOH:MeOH:H2O (15:15:70) as the mobile phase, with detection at wavelengths 215, 276 and 356 nm, at 25ºC, and using a flow rate of 1 mL/min. By using this protocol, it was possible to obtain a linear calibration curve (correlation coefficient = 1), limits of detection and quantification in the order of ng/mL, and a short analysis time (<15 minutes per sample). Additionally, the developed protocol allowed the successful detection and quantification of 3-NT in all biological matrices tested, with detection at 356 nm. Conclusion: The method described in this study, which was successfully developed and validated for 3-NT quantification, is simple, cheap and fast, rendering it suitable for analysis in a wide range of biological matrices.

Short sea shipping has several advantages over other means of transportation, recognized by EU members. The maritime transportation could be dealt like a combination of two well-known problems: the container stowage problem and routing planning problem. The integration of these two well-known problems results in a new problem CSSRP (Container stowage and ship routing problem) that is also an hard combinatorial optimization problem. The aim of this work is to solve the CSSRP using a mixed integer programming model. It is proved that regardless the complexity of this problem, optimal solutions could be achieved in a reduced computational time. For testing the mathematical model some problems based on real data were generated and a sensibility analysis was performed.

A medição precisa da força é necessária para muitas aplicações, nomeadamente, para a determinação da resistência mecânica dos materiais, controlo de qualidade durante a produção, pesagem e segurança de pessoas. Dada a grande necessidade de medição de forças, têm-se desenvolvido, ao longo do tempo, várias técnicas e instrumentos para esse fim. Entre os vários instrumentos utilizados, destacam-se os sensores de força, também designadas por células de carga, pela sua simplicidade, precisão e versatilidade. O exemplo mais comum é baseado em extensómetros elétricos do tipo resistivo, que aliados a uma estrutura formam uma célula de carga. Este tipo de sensores possui sensibilidades baixas e em repouso, presença de offset diferente de zero, o que torna complexo o seu condicionamento de sinal. Este trabalho apresenta uma solução para o condicionamento e aquisição de dados para células de carga que, tanto quanto foi investigado, é inovador. Este dispositivo permite efetuar o condicionamento de sinal, digitalização e comunicação numa estrutura atómica. A ideia vai de encontro ao paradigma dos sensores inteligentes onde um único dispositivo eletrónico, associado a uma célula de carga, executa um conjunto de operações de processamento de sinal e transmissão de dados. Em particular permite a criação de uma rede ad-hoc utilizando o protocolo de comunicação IIC. O sistema é destinado a ser introduzido numa plataforma de carga, desenvolvida na Escola Superior de Tecnologia e Gestão de Bragança, local destinado à sua implementação. Devido à sua estratégia de conceção para a leitura de forças em três eixos, contém quatro células de carga, com duas saídas cada, totalizando oito saídas. O hardware para condicionamento de sinal já existente é analógico, e necessita de uma placa de dimensões consideráveis por cada saída. Do ponto de vista funcional, apresenta vários problemas, nomeadamente o ajuste de ganho e offset ser feito manualmente, tornando-se essencial um circuito com melhor desempenho no que respeita a lidar com um array de sensores deste tipo.

Background: At the end of 80s, cloning technologies with the increase of the antibodies’ sensibility made easier the development of technologies based on Fluorescence in situ Hibridation (FISH). Nowadays, It’s widely used in the field of basic investigation as much as clinic diagnostic. Method: FISH is a technique that combines molecular biology with histochemistry way to detect specific nucleotide sequences so that chromosome’s section or even whole chromosome can be marked on metaphases cells (cell in division) and on attached cellular nucleus. This detection is realized using DNA fluorescence probes (marked with fluorophores), that can be different according to the structures manage to detect: large single-locus probes, small unique-sequence probes, chromosome- or region-specific “paints” or repetitive sequence probes and genomic DNA probes. Some of the applications of this technique is that can be so useful in the detection of numerical and structural chromosomal alterations such as polyploidies or genomic rearrangement, to mapping metaphases cells and even to detect bacteria or another type of microorganism. In addition, FISH allows us to monitoring diseases (antitumor therapies, quantification of genomic altered cells…) and the precise location of chromosomic broken spots on tumor searching for new genes involved in cancer and detect and map interested known genes. Conclusion: FISH has many advantages ahead of conventional cytogenetic techniques (bands G karyotype) overall at the time of establish a clinic diagnostic to detect tumors and chromosomic aberration, presenting a higher sensibility and specificity as well as being a relative quick technique (24 hours).

O presente estudo enquadra-se no domínio das Ciências da Educação e na área específica da Análise da Acão Educativa. Não se afastando das preocupações inscritas nos objetivos de um mestrado de autor "A Criança em Diferentes Contextos Educativos”, também se compagina com as nossas determinações pessoais e profissionais, empreendidas na convicção da investigação como um projeto de vida. Para o efeito, arquitetámos o nosso universo de estudo a partir da observação naturalista em sete salas de creche e de jardim-de-infância do distrito de Évora. Da população estudada fizeram parte diferentes acores educativos, entre eles, as crianças dos 3 meses aos 6 anos de idade, as educadoras e as auxiliares de Acão educativa. Pretendemos, a partir dos sinais emitidos pelas crianças, retirar indicadores de necessidades educativas, para compreender as atitudes e comportamentos dos agentes educativos (educadores e auxiliares) na sua prática pedagógica, inferindo das suas próprias necessidades formativas. Procurámos saber como eram feitos os diagnósticos de necessidades (educativas das crianças e formativas dos educadores) e como organizavam a sua relação com outros acores educativos, no sentido de um desenvolvimento holístico da criança. Dada a natureza e o objeto de estudo, optámos por uma metodologia qualitativa de carácter interpretativo, configurada num estudo de caso. Utilizámos a linha de Investigação-Acão/Formação, preconizada por Luís Barbosa, integrada na sua "Escola Sensível e Transformacionista", que radica os seus fundamentos na ideia de que o jardim-de-infância, tal como outra qualquer organização educativa, deve ser um "Observatório de Caracterização de necessidades educativas das crianças e formativas dos sues agentes educativos”, gerido na perspetiva da mudança. Neste enquadramento de ideias, ao qual aderimos por convicção, utilizámos, para caracterizar o real, a Observação como técnica privilegiada, pese embora as limitações que lhe são subjacentes. Minimizámos este facto, triangulando os dados com entrevistas semiestruturadas e notas de campo. Suportámo-nos da Técnica do Espelhamento, da Pedagogia de Ajuda, da Renomeação de Experiências e da Extensibilidade de Si, às quais, pela sua importância demos ênfase no enquadramento teórico. O nosso estudo permitiu-nos inferir que, a relação diagnostica nos processos de ensino/aprendizagem, naqueles contextos, parece estar correlacionada com o facto de que os agentes educativos ainda necessitam de adquirir um leque variado de competências, relacionadas com o Saber-Ser e o Saber-Estar, que lhes permitam atitudes relacionais pró-activas e maior sensibilidade para captar as emoções e as sensações das crianças. Este facto vem corroborar o facto de termos verificado que os agentes educativos não usam o diagnóstico de necessidades como âncora de organização das suas práticas educativas o que não contribui de forma sustentável para a promoção do bem-estar e do desenvolvimento integral da criança. ABSTRACT: The present study lies within the field of Educational Sciences in the specific area of analysis of education. Not moving away from the concerns listed in the goals of an author master degree "The Child in Different Educational Contexts”, it also fits our personal and professional decisions, undertaken in the belief of research as a project of life. We chose the universe of our study from naturalistic observation in seven rooms of Nurseries and Infant Schools in the district of Évora. The study population consisted of children from three months to six years old, educators and assistants of education. From signals emitted by children, we intended to take indicators of educational needs to understand the attitudes and behavior of the educators and their assistants in their pedagogical practice, inferring their own educational needs. We tried to know how the diagnoses of needs were made (educational needs of the children and training needs of the educators) and how they organized their relationship with other educational agents towards a holistic development of children. Considering the nature and purpose of the study we opted for a qualitative methodology of interpretative nature, in a case study. We used the action research training line, advocated by Luís Barbosa in his work "Escola Sensível e Transformacionista" (Sensitive and Transformational School), whose reason lies in the idea that Infant Schools, like other educational organizations, should be centers for the characterization of children’s educational needs and educational agents' formative needs, managed in terms of change. ln this framework of ideas, that we adopted by conviction, we used the observation as the main technique, despite its limitations. To minimize them we triangulated the data with semi structured interviews and field notes. We used the techniques of Mirroring, Pedagogy of Help, renaming of Experiences and Extensibility of Yourself, and, because of their importance, we focused on them in the theoretical framework. This study allowed us to infer that the diagnostic relationship in the teachinglearning processes, in those contexts, seem to be correlated with the fact that the educational agents still need to acquire a wide range of skills related to the Know How To Be, to provide them with proactive relational attitudes and a bigger sensibility to get the children's emotions and sensations. We also found that the educational agents don't use a diagnostic of needs as anchor of the organization of their educational practices, what does not contribute in a sustainable way for the promotion of the well being and integral development of the child.

ABSTRACT: Asian soybean rust, caused by the fungus Phakopsora pachyrhizi, is the most severe disease of the crop and can cause yield losses of up to 90%. The disease was first reported in Brazil in 2001. Epidemics of the disease are common in the country, where the fungus can survive year?round. Regulatory measures to reduce the inoculum between seasons and avoid late-season soybean have been adopted to manage the disease. Disease control has relied mainly on chemical control, but a lower sensibility of the fungus to fungicides has been reported in Brazil. Major?resistance genes have been mapped and incorporated into the cultivars. With the reduced efficacy of the fungicides, the adoption of integrated measures to control the disease will be important for the sustainability of the crop. This review presents the main changes in the soybean crop system caused by the introduction of the fungus in Brazil, the current management strategies adopted to avoid losses, and the new trends that, together with biotechnological strategies, can improve management in the future. RESUMO: A ferrugem?asiática da soja, causada pelo fungo Phakopsora pachyrhizi, é a doença mais severa da cultura e pode causar perdas de produtividade de até 90%. A doença foi relatada pela primeira vez no Brasil em 2001. Epidemias da doença são comuns no País, onde o fungo pode sobreviver durante todo o ano. Medidas regulatórias para reduzir o inóculo entre safras e evitar a semeadura tardia de soja têm sido adotadas para manejar a doença. O controle da doença tem se baseado principalmente no controle químico, mas uma menor sensibilidade do fungo aos fungicidas tem sido relatada no Brasil. Genes de resistência têm sido mapeados e incorporados às cultivares. Por causa da redução da eficiência dos fungicidas, a adoção de medidas integradas para o controle da doença será importante para a sustentabilidade da cultura. Este artigo de revisão apresenta as principais mudanças no sistema de produção da soja causadas pela introdução do fungo no Brasil, as medidas de controle atualmente usadas para evitar perdas, e as novas tendências que, juntas com estratégias biotecnológicas, podem melhorar o manejo da doença no futuro.

Spectral identification of individual micro- and nano-sized particles by the sequential intervention of optical catapulting, optical trapping and laser-induced breakdown spectroscopy is presented [1]. The three techniques are used for different purposes. Optical catapulting (OC) serves to put the particulate material under inspection in aerosol form [2-4]. Optical trapping (OT) permits the isolation and manipulation of individual particles from the aerosol, which are subsequently analyzed by laser-induced breakdown spectroscopy (LIBS). Once catapulted, the dynamics of particle trapping depends on the laser beam characteristics (power and intensity gradient) and on the particle properties (size, mass and shape). Particles are stably trapped in air at atmospheric pressure and can be conveniently manipulated for a precise positioning for LIBS analysis. The spectra acquired from the individually trapped particles permit a straightforward identification of the inspected material. The current work focuses on the development of a procedure for simultaneously acquiring dual information about the particle under study via LIBS and time-resolved plasma images by taking advantage of the aforementioned features of the OC-OT-LIBS instrument to align the multiple lines in a simple yet highly accurate way. The plasma imaging does not only further reinforce the spectral data, but also allows a better comprehension of the chemical and physical processes involved during laser-particle interaction. Also, a thorough determination of the optimal excitation conditions generating the most information out of each laser event was run along the determination of parameters such as the width of the optical trap, its stability as a function of the laser power and the laser wavelength. The extreme sensibility of the presented OC-OT-LIBS technology allows a detection power of attograms for single/individual particle analysis.

Character education has been viewed by many educators as having significant historical, academic, and social value. Many stakeholders in education argue for character development as a curricular experience. While understanding the degree to which character education is of worth to stakeholders of institutions is important, understanding students, teachers, and administrators perspectives from their lived experiences is likewise significant. The purpose of this phenomenological study was to gain a deeper understanding of character education within a Biblical framework environment by examining the lived experiences of students, administrators, and teachers of a Seventh-day Adventist School. Phenomenology describes individuals’ daily experiences of phenomena, the manner in which these experiences are structured, and focuses analysis on the perspectives of the persons having the experience (Moustakas, 1994). ). This inquiry was undertaken to answer the question: What are the perceptions of students, teachers, and an administrator toward character education in a Seventh-day Adventist school setting? Ten participants (seven students and three adults) formed the homogeneous purposive sample, and the major data collection tool was semi-structured interviews (Patton, 1990; Seidman, 2006). Three 90-minute open-ended interviews were conducted with each of the participants. Data analysis included a three-phase process of description, reduction and interpretation. The findings from this study revealed that participants perceived that their involvement in the school’s character education program decreased the tendency to violence, improved their conduct and ethical sensibility, enhanced their ability to engage in decision-making concerning social relationships and their impact on others, brought to their attention the emerging global awareness of moral deficiency, and fostered incremental progress from practice and recognition of vices to their acquisition of virtues. The findings, therefore, provide a model for teaching character education from a Seventh-day Adventist perspective. The model is also relevant for non-Seventh day Adventists who aspire to teach character education as a means to improving social and moral conditions in schools.
