A capillary electrochromatography (CEC) monolithic column with zwitterionic stationary phases was prepared by in situ polymerization of butyl methacrylate, ethylene dimethacrylate, methacrylic acid, and 2-(dimethyl amino) ethyl methacrylate in the presence of porogens. The stationary phases have zwitterionic functional groups, that is, both tertiary amine and acrylic acid groups, so the ionization of those groups on the zwitterionic stationary phase was affected by the pH values of the mobile phase, and further affects the strength and direction of the electroosmotic flow (EOF). Separations of alkylbenzenes and polycylic aromatic hydrocarbons based on the hydrophobic mechanism were obtained. Separation of various types of polar compounds, including phenols, anilines, and peptides, on the prepared column were performed under CEC mode with anodic and cathodic EOF, and different separation selectivities of those polar analytes were observed on the monolithic capillary column by using mobile phases with different pH values.

A novel ion-bonded discotic complex was prepared from 2,3,6,7,10,11-hexakis(N,N-dimethylaminopropylaminocarbonylmethoxy)triphenylene (HDTP) and 4'-dodecyloxybiphenyl-4-carboxylic acid (DBC) by ionic self-assembly (ISA) route and characterized by Fourier transform infrared (FTIR) spectrum. We found that the complex can self-assemble into stable gels in aromatic hydrocarbons. Nanofibers with diameters of 50-130 nm were observed in the gels by transmission electron micrograph (TEM).

The influences of additive, diluents, temperature, acidity of the aqueous phase on the interfacial behavior of primary amine N1923 in sulfate media have been investigated using the Du Nouy ring method. In addition, the effect of concentration of thorium(IV) loaded in the organic phase on the interfacial tension has also been studied. The interfacial tension isotherms are processed by matching different adsorption equations such as the Gibbs and the Szyszkowski. The surface excess at the saturated interface (Gamma (max)) and the minimum bulk concentration of the extractant necessary to saturate the interface (C-min) under different conditions are calculated according to two adsorption equations to be presented in comprehensive tables and figures. It appears that primary amine N1923 has strong interfacial activity and behaves very differently in various diluents systems. The surface excess at saturated interface increase with the type of diluerits in the following order: chloroform < aromatic hydrocarbons < aliphatic hydrocarbons. The relationship between the interfacial activity and kinetics of thorium extraction by primary amine N1923 has been discussed by considering different factors. However, the interfacial activity of primary amine N1923 is only a qualitative parameter suggesting the interfacial mechanism for thorium extraction, it cannot give strong evidence quantitatively supporting this mechanism.

Two kinds of macrocyclic arylene ketone oligomers have been synthesized in high yield from phthaloyl dichloride and various bridge-linking electron-rich aromatic hydrocarbons via the modified Friedel-Crafts acylation reaction. The presence of a Lewis base in this reaction is demonstrated to be advantageous for forming macrocycle oligomers. These resultant oligomers can undergo melt ring-opening polymerization to give polymers with high T. and excellent thermal stability.

In this paper, the extraction of Ce(IV) from nitric acid solutions is investigated using di-(2-ethylhexyl) 2-ethylhexyl phosphonate (DEHEHP, B) in heptane as extractant. Ce(IV) can be extracted effectively from nitric acid solution, whereas it is poorly extracted from sulfuric acid solution. Compared with some other organophosphorus esters, DEHEHP has moderate extractablity for Ce(IV). The extraction efficiency varies with diluent in the order: aliphatic hydrocarbons > nitrobenzene > aromatic hydrocarbons > carbon tetrachloride > chloroform. Regeneration and loading capacities of DEHEHP have also been examined. Ce(IV) extraction in HNO3 solutions as well as extraction of HNO3 and H2O have been systematically studied. The Ce(IV) extraction increases with an increasing of HNO3 concentration and exhibits the maximum distribution ratio at 1-1.5 mol/L HNO3. Nitric acid, as a source of nitrate ion, enhances the extraction of metal ion. But it also competes with metal ions for extractant molecules by its own extraction under high acidities. Heat shock protein 70 (HSP70) is an important member of the heat shock protein superfamily, and it plays a key role in the process of protecting cells, facilitating the folding of nascent peptides and responding to stress. The cDNA of bay scallop Argopecten irradians HSP70 (designated AIHSP70) was cloned by the techniques of homological cloning and rapid amplification of cDNA end (RACE). The full length of AIHSP70 cDNA was 2651 bp in length, having a 5' untranslated region (UTR) of 96 bp, a 3' UTR of 575 bp, and an open reading frame (ORF) of 1980 bp encoding a polypeptide of 659 amino acids with an estimated molecular mass of 71.80 kDa and an estimated isoelectric point of 5.26. BLAST analysis revealed that the AIHSP70 gene shared high identity with other known HSP70 genes. Three classical HSP signature motifs were detected in AIHSP70 by InterPro, analysis. 3-D structural prediction of AIHSP70 showed that its N terminal ATPase activity domain and,C terminal substrate-binding domain shared high similarity with that in human heat shock protein 70. The results indicated that the AIHSP70 was a member of the heat shock protein 70 family. A semi-quantitive RT-PCR method was used to analyse the expression of AIHSP70 gene after the treatment of naphthalin which is one kind of polycyclic aromatic hydrocarbon (PAH) and the challenge of bacteria. mRNA expression of AIHSP70 in scallop was up-regulated significantly after the stimulation of naphthalin and increased with increasing naphthalin concentration. A clearly time-dependent expression pattern of AIHSP70 was observed after the scallops were infected by Vibrio anguillarum, and the mRNA expression reached a maximum level at 8 h and lasted to 16 h, and then dropped progressively. The results indicated that AIHSP70 could play an important role in mediating the environmental stress and immune response in scallop. (c) 2006 Elsevier Ltd. All rights reserved.

Individuals of Mytilus edulis L., collected from the Erme estuary (S.W. England) in 1978, were exposed to low concentrations (7 to 68 μg l-1) of the water-accommodated fraction (WAF) of North Sea crude oil. The pattern of accumulation of petroleum hydrocarbons in the body tissues was affected by the presence of algal food cells, the period of exposure, the hydrocarbon concentration in seawater, the type of body tissue and the nature of the hydrocarbon. Many physiological responses (e.g. rates of oxygen consumption, feeding, excretion, and scope for growth), cellular responses (e.g. lysosomal latency and digestive cell size) and biochemical responses (e.g. specific activities of several enzymes) were significantly altered by short-term (4 wk) and/or long-term (5 mo) exposure to WAF. Stress indices such as scope for growth and lysosomal latency were negatively correlated with tissue aromatic hydrocarbons.

The marine gastropod Littorina littorea from four sites in the vicinity of the Sullom Voe Oil Terminal was found to display reduced cytochemically determined latency of lysosomal arylsulphatase, β-glucuronidase and acid phosphatase in comparison with snails from a nearby ‘clean’ site. This is interpreted as indicating lysosomal destabilization by environmental factors. Elevated total activities of particular lysosomal hydrolases were recorded at three of the sites in Sullom Voe. Animals from a fourth site (Swarta Taing) showed significant depression of arylsulphatase and β-glucuronidase. Cytochemically determined activity of blood cell NADPH-neotetrazolium reductase, which is a component of microsomal detoxication systems, was stimulated in these same sites in comparison with the ‘clean’ reference site. This stimulation or induction is interpreted as a response to the presence of oil-derived polynuclear aromatic hydrocarbons. These results are discussed in the light of previous work on the effects of hydrocarbons on lysosomes and in terms of the possible physiological consequences for the animals.

Structural changes were observed in the digestive tubule epithelial cells of Mytilus edulis following long-term exposure to the water accommodated fraction (WAF) of North Sea crude oil (30 μg · l−1 total oil derived aromatic hydrocarbons). The changes observed involved a reduction in the height of the digestive cells beyond that demonstrated in a normal feeding cycle. In addition there was a loss of the normal synchrony of the digestive cells to a point where nearly all the tubules exhibited an appearance similar to that which is usually termed ‘reconstituting’. These alterations were quantified using an image analysis technique and the mean height of the digestive cells used as an index of digestive function or state. Long-term exposure also induced a radical alteration of the structure of secondary lysosomes within the digestive cells, resulting in the formation of large lysosomes, believed to be autolysosomes. Stereological analyses showed that these lysosomes are reduced in numbers and greatly increased in volume in comparison with controls. There is a concomitant increase in surface area of lysosomes per unit volume of digestive cell compared with control conditions. These alterations are indicative of fundamental changes in secondary lysosomal function involving an autophagic response to oil derived hydrocarbons. which would contribute to the reduction of digestive cell cytoplasm. These cellular alterations are discussed in terms of their use as indices of cell injury, in response to oil.

Components of a xenobiotic detoxication/toxication system involving mixed function oxygenases are present inMytilus edulis. Our paper critically reviews the recent literature on this topic which reported the apparent absence of such a system in bivalve molluscs and attempts to reconcile this viewpoint with our own findings on NADPH neotetrazolium reductase, glucose-6-phosphate dehydrogenase, aldrin epoxidation and other reports of the presence of mixed function oxygenases. New experimental data are presented which indicate that some elements of the detoxication/toxication system inM. edulis can be induced by aromatic hydrocarbons derived from crude oil. This includes a brief review of the results of long-term experiments in which mussels were exposed to low concentrations of the water accommodated fraction of North Sea crude oil (7.7–68 µg 1−1) in which general stress responses such as reduced physiological scope for growth, cytotoxic damage to lysosomal integrity and cellular damage are considered as characteristics of the general stress syndrome induced by the toxic action of the xenobiotics. In addition, induction in the blood cells of microsomal NADPH neotetrazolium reductase (associated with mixed function oxygenases) and the NADPH generating enzyme glucose-6-phosphate dehydrogenase are considered to be specific biological responses to the presence of aromatic hydrocarbons. The consequences of this detoxication/toxication system forMytilus edulis are discussed in terms of the formation of toxic electrophilic intermediate metabolites which are highly reactive and can combine with DNA, RNA and proteins with subsequent damage to these cellular constituents. Implications for neoplasms associated with the blood cells are also discussed. Finally, in view of the increased use of mussel species in pollutant monitoring programmes, the induction phenomenon which is associated with microsomal enzymes in the blood cells is considered as a possible tool for the detection of the biological effects of environmental contamination by low concentrations of certain groups of organic xenobiotics.

Calanoid copepod eggs have been extracted from intertidal sediments and hatched in the laboratory. Although most of the eggs which hatched did so within < 7 days, the hatching of some continued over a more prolonged period (> 20 days). This indicates that there were a significant number of diapausing or delayed hatching eggs. The species of calanoids present include some of which are known to produce diapausing eggs. Hatching of nauplii from incubated sediment samples was slower than from the extracted eggs indicating dormancy induced by the effects of burial in the sediment. Viability of the eggs has been related to chronic industrial or urban pollution as indicated by polycyclic aromatic hydrocarbon levels. These hatchings were conducted simultaneously with those for cleaner locations. The viability of eggs was significantly depressed in the more heavily polluted sites. An oil spill arising from the grounding of the "Sea Empress" at Milford Haven, UK, in February 1996 has provided a comparison of the impact of an acute situation with these chronic effects. An immediate drastic reduction in viability was followed by a recovery in the year following the spill. The data have provided no evidence for differences in the response to pollution between diapausing and subitaneous eggs.

Freestanding polyparaphenylene films were obtained on polymerization of benzene at potential of 1.2 V versus Al wire on substrates like platinum/transparent conducting glass as an anode. The electrolyte used was chloroaluminate room-temperature melt, which was prepared by intimate mixing of a 1:2 ratio of cetyl pyridinium chloride and anhydrous aluminum chloride to yield a viscous liquid. This liquid was miscible in all proportions with benzene and other aromatic hydrocarbons in all proportions at room temperature. The polyparaphenylene films deposited on platinum anode exhibited a prominent cyclic voltammetric peak at 0.7 V versus Al wire as reference electrode in chloroaluminate medium. The impedance spectra gave low charge transfer resistance. The diffused reflectance electronic spectra of the film gave the peaks at 386 nm and 886 nm. The PPP films showed electronic conductivity around 3–4 × 104 S/cm by four probe method under nitrogen atmosphere. The polymer was also characterized by IR spectra, thermal studies, and SEM studies.

This paper is concerned with the chemical evolution of large molecules in interstellar clouds. We consider the chemistry and ionisation balance of large polycyclic aromatic hydrocarbon (PAH) type molecules in diffuse clouds and show that certain PAH molecules can be doubly ionised by the interstellar ultraviolet radiation field. If recombination of the dications so produced with electrons is dissociative rather than radiative, then PAHs are rapidly destroyed. PAHs which can only be singly ionised have much smaller recombination energies and can be long lasting in these regions. This type of property may be very important in selecting the PAH species which can populate the general interstellar medium and account for certain of the diffuse bands observed in optical spectra. Destruction of PAH molecules via formation of dications may be responsible for the weakening of the diffuse bands observed in regions of high UV flux.

No presente trabalho, foi estudado um largo espectro de efeitos genotóxicos e bioquímicos na tainha-garrento (Liza aurata). Nos Capítulos II e III são descritos os efeitos de exposição de curta duração ao fenantreno, um hidrocarboneto aromático policíclico (HAP). A exposição de curta duração (16 horas) demonstrou a capacidade deste composto induzir a actividade da enzima de fase I da biotransformação, etoxiresorufina O-desetilase (EROD), provocar decréscimos de integridade no ADN hepático e aumento de anomalias nucleares eritrocíticas (ANE). Em termos de respostas de stresse, os níveis plasmáticos de cortisol e glucose aumentaram face à exposição a este HAP. A exposição ao fenantreno induziu o decréscimo da glutationa peroxidase (GPx) nas guelras, enquanto que no fígado a actividade da GPx aumentou. No rim, a actividade da glutationa S-transferase (GST) foi inibida. Nas guelras, verificou-se um aumento da catalase. O fenantreno demonstrou igualmente a capacidade de induzir um aumento dos níveis de glutationa nas guelras e fígado. Estas respostas demonstraram a sensibilidade de L. aurata, a este HAP, realçando a especificidade das respostas em termos de órgãos. Apesar dos aumentos das defesas antioxidantes, o potencial tóxico deste composto foi demonstrado pelo aumento da peroxidação lipídica nos três órgãos. Nos capítulos seguintes, são descritas as respostas de L. aurata capturada na Ria de Aveiro, em locais com diferentes perfis de contaminação, inicialmente no Outono de 2005 (Capítulos III a IX) e posteriormente analisando respostas sazonais (Capítulos X e XI). A análise de respostas de stresse (cortisol, glucose e lactato) revelou que L. aurata capturada em Vagos (local contaminado por HAPs) apresentava níveis baixos de cortisol, enquanto que no Laranjo (local contaminado por mercúrio) apresentavam elevados níveis de glucose e lactato. Relativamente às hormonas do eixo hipotálamo – hipófise – tiróide (HHT), foram observados elevados níveis plasmáticos da hormona estimuladora da tiróide (TSH) nos organismos capturados no Laranjo, baixos níveis de tiroxina (T4) nos organismos da Barra (local sujeito a tráfego naval) e baixos níveis de triiodotironina (T3) no Rio Novo do Príncipe (próximo de um antigo efluente de pasta de papel), Laranjo e Vagos. A avaliação das defesas antioxidantes, dano oxidativo e genotóxico nas guelras, rim e fígado revelou diferenças significativas nas respostas dos órgãos. L. aurata capturada na Barra apresentou dano oxidativo nas guelras (Capítulo V). No rim foi detectada uma diminuição da integridade do ADN no Rio Novo do Príncipe e Vagos (Capítulo VI), enquanto que no fígado foi observado dano lipídico na Gafanha e Vagos (Capítulo VIII). O dano não esteve sempre associado a um decréscimo das defesas. As análises da água e do sedimento da Ria de Aveiro (Outono de 2005) revelaram elevadas concentrações de metais (Cd, Hg, Cu e Zn),principalmente, no Laranjo e Rio Novo do Príncipe. L. aurata capturada nestes locais apresentou os níveis mais elevados de metalotioninas hepáticas (Capítulo VII) que parecem responsáveis pela inexistência de danos no fígado (Capítulo VIII). O dano oxidativo no ADN, avaliado através da quantificação dos níveis plasmáticos de 8-hidroxi-2-desoxiguanosina (8-OHdG) e o dano clastogénico/aneugénico, avaliado através da quantificação da frequência de ANE, foram estudados, no Outono de 2005, em duas espécies de peixes (L. aurata e Dicentrarchus labrax - robalo) (Capítulo IX). Os resultados revelaram grande sensibilidade de D. labrax em termos de dano oxidativo no ADN na Gafanha, Rio Novo do Príncipe e Vagos, enquanto que L. aurata apresentou dano oxidativo apenas no Laranjo. O aumento da frequência de ANE apenas foi detectado em L. aurata, em Vagos, não se tendo detectado correlação entre estes dois parâmetros. O estudo sazonal (Maio de 2006 a Março de 2007) do dano oxidativo no ADN e frequência de ANE em L. aurata (Capítulo X) demonstrou a variação destes parâmetros com a estação do ano, apesar de não se ter verificado correlação com os parâmetros hidrológicos determinados. No entanto, no local de referência não se verificaram diferenças sazonais, o que sugere que estes biomarcadores reflectem variações de biodisponibilidade de contaminantes. A análise global dos resultados das diferentes estações do ano revelou que L. aurata capturada no Rio Novo do Príncipe e em Vagos apresentou maior susceptibilidade a dano oxidativo no ADN. No entanto, apenas L. aurata capturada em Vagos apresentou frequência de ANE superior à do local de referência. Os dados do estudo sazonal revelaram uma correlação entre dano oxidativo e ANE, sugerindo o stresse oxidativo como um possível mecanismo envolvido na formação de anomalias. A integridade do ADN das guelras, rim, fígado e sangue de L. aurata foi igualmente estudada ao longo de um ano (Capítulo XI), tendo-se verificado uma grande variabilidade ao longo deste período. Não foi demonstrada sensibilidade a um perfil de contaminação específico, tendo-se verificando variabilidade sazonal no local de referência. Globalmente, os resultados demonstraram a importância da utilização de uma bateria de biomarcadores na monitorização ambiental e a especificidade da resposta dos diferentes órgãos de L. aurata.

O objectivo geral deste trabalho consistiu no desenvolvimento de novos sensores químicos de fibra óptica (OF) para análise de compostos orgânicos voláteis (VOCs) em ambientes industriais. A componente de detecção dos sensores desenvolvidos é constituída por uma pequena secção de fibra óptica revestida com um filme de polímero. A morfologia dos filmes poliméricos foi analisada e caracterizada por microscopia electrónica de varrimento (SEM), sendo a espessura dos filmes determinada por espectroscopia de retrodispersão de Rutherford (RBS, acrónimo do inglês Rutherford backscattering spectrometry). O desempenho analítico dos sensores de OF foi avaliado relativamente a diferentes parâmetros operacionais, tais como, concentração da solução de revestimento, técnica de deposição do filme polimérico, temperatura da célula de injecção, temperatura de cura do material polimérico, caudal do gás de arraste, comprimento de onda e frequência de funcionamento do laser, configurações estruturais da célula de injecção e do tubo analítico. Foram desenvolvidos dois sensores de OF a operar na região do infravermelho para determinação de diferentes classes de VOCs, nomeadamente hidrocarbonetos aromáticos, clorados e alifáticos, além de álcoois. Os sensores de OF desenvolvidos apresentaram adequadas características analíticas em termos de sensibilidade, linearidade, repetitibilidade e reprodutibilidade do sinal analítico, sendo o tempo de resposta de aproximadamente 30 segundos. Foi também desenvolvido um sensor de OF para especiação de benzeno, tolueno e o-xileno a operar na região do visível (635 - 650 nm), tendo sido aplicado à análise de amostras reais de ar de uma indústria de solventes. Relativamente à monitorização de VOCs em ambientes industriais, foi desenvolvido um sensor de OF para monitorização in situ e de forma remota (até uma distância máxima de 60 metros do local de amostragem) de benzeno, tolueno, etilbenzeno, p-xileno, m-xileno e o-xileno (BTEX), utilizando um díodo laser a 1550 nm. O desempenho analítico do sensor desenvolvido foi comparado, para a determinação de BTEX, com a cromatografia gasosa acoplada à detecção com ionização de chama (GC-FID). Foram ainda desenvolvidos dois detectores de fibra óptica acoplados a um cromatógrafo de gás para especiação de álcoois e hidrocarbonetos aromáticos. A metodologia desenvolvida baseada em cromatografia gasosa acoplada a um detector de fibra óptica (GC-OF) foi aplicada à análise de amostras reais de ar de uma indústria de solventes, comparando os respectivos resultados com os obtidos por GC-FID. Por fim foi efectuado um estudo visando a obtenção de um modelo geral para a resposta analítica dos sensores de fibra óptica desenvolvidos.
