Here the mechanism of arsenite transport into paddy rice (Oryza sativa) roots, uptake of which is described by Michaelis-Menten kinetics, is reported. A recent study on yeast (Saccharomyces cerevisiae) showed that undissociated arsenite (its pKa is 9.2) was transported across the plasma membrane via a glycerol transporting channel. To investigate whether the same mechanism of transport was involved for rice, competitive studies with glycerol, which is transported into cells via aquaporins, were performed. Glycerol competed with arsenite for transport in a dose-dependent manner, indicating that arsenite and glycerol uptake mechanisms were the same. Arsenate transport was unaffected by glycerol, confirming that arsenate and arsenite are taken up into cells by different mechanisms. Antimonite, an arsenite analogue that is transported into S. cerevisiae cells by aquaporins, also competed with arsenite transport in a dose-dependent manner, providing further evidence that arsenite is transported into rice roots via glycerol transporting channels. Mercury (Hg2+) inhibited both arsenite and arsenate uptake, suggesting that inhibition of influx was due to general cellular stress rather than the specific action of Hg2+ on aquaporins. Arsenite uptake by pea (Pisum sativum) and wheat (Triticum aestivum) was also described by Michaelis-Menten kinetics.

A range of fern species (45) and their allies, Equisetum (5) and Selaginella (2) species and Psilotum nudum were screened for their ability to hyperaccumulate arsenic, to develop a phylogenetic understanding of this phenomenon. A number of varieties (5) of a known arsenic hyperaccumulator Pteris cretica were additionally included in this study. This study is the first to report members of the Pteris genus that do not hyperaccumulate arsenic, Pteris straminea and tremula. A phylogenetic basis for arsenic accumulation in ferns was investigated. Some orders can accumulate more arsenic than others. Although members of the Equisetales and Blechnales did not hyperaccumulate arsenic, they still accumulated relatively high levels in their fronds, approaching 100 mg kg-1 when grown on a soil dosed with 100 mg kg-1 arsenic. Arsenic hyperaccumulation was identified as a phenomenon at the extreme range of fern arsenic accumulation. Ferns that exhibit arsenic hyperaccumulation arrived relatively late in terms of fern evolution, as this character is not exhibited by primitive ferns or their allies.

Phytochelatins (PCs) are required for arsenic (As) detoxification in nontolerant plants. In addition, a role for PCs in arsenate tolerance has recently been proven, with tolerant plants able to accumulate significantly higher concentrations of As-PC complexes at equivalent levels of stress than nontolerant plants. The relationship between arsenate influx and PC production in tolerant and non-tolerant Holcus lanatus plants was determined in this study, along with an investigation of the effect of inhibition of PC synthesis by buthionine sulfoximine (BSO) on arsenate tolerance. A strong correlation between PC production and arsenate influx was demonstrated in arsenate-tolerant plants. In addition, inhibition of PC synthesis by BSO in tolerant plants increased arsenate sensitivity to that of the nontolerant clone. This dramatic reduction in tolerance proves that PC production is an essential component of the arsenate tolerance mechanism in H. lanatus. This study proposes that while there is a single major gene for arsenate tolerance, hypostatic modifiers are also in operation, affecting the expression of the tolerance character. © New Phytologist (2002).

Ericoid mycorrhizas are believed to improve N nutrition of many ericaceous plant species that typically occur in habitats with impoverished nutrient status, by releasing amino acids from organic N forms. Despite the ubiquity of mycorrhizal formation the mechanisms and regulation of nutrient transport in mycorrhizal associations are poorly understood. We used an electrophysiological approach to study how amino acid transport characteristics of Calluna vulgaris were affected by colonization with the ericoid mycorrhiza fungus Hymenoscyphus ericae. Both the Vmax and Km parameters of amino acid uptake were affected by fungal colonization in a manner consistent with an increased availability of amino acid to the plant. The ecophysiological significance of altered amino acid transport in colonized root cells of C. vulgaris is discussed. © New Phytologist (2002).

The role of arbuscular mycorrhizal fungi (AMF) in arsenate resistance in arbuscular mycorrhizal associations is investigated here for two Glomus spp. isolated from the arsenate-resistant grass Holcus lanatus. Glomus mosseae and Glomus caledonium were isolated from H. lanatus growing on an arsenic-contaminated mine-spoil soil. The arsenate resistance of spores was compared with nonmine isolates using a germination assay. Short-term arsenate influx into roots and long-term plant accumulation of arsenic by plants were also investigated in uninfected arsenate resistant and nonresistant plants and in plants infected with mine and nonmine AMF. Mine AMF isolates were arsenate resistant compared with nonmine isolates. Resistant and nonresistant G. mosseae both suppressed high-affinity arsenate/phosphate transport into the roots of both resistant and nonresistant H. lanatus. Resistant AMF colonization of resistant H. lanatus growing in contaminated mine spoil reduced arsenate uptake by the host. We conclude that AMF have evolved arsenate resistance, and conferred enhanced resistance on H. lanatus.

Elevation of arsenic levels in soils causes considerable concern with respect to plant uptake and subsequent entry into wildlife and human food chains, Arsenic speciation in the environment is complex, existing in both inorganic and organic forms, with interconversion between species regulated by biotic and abiotic processes. To understand and manage the risks posed by soil arsenic it is essential to know how arsenic is taken up by the roots and metabolized within plants. Some plant species exhibit phenotypic variation in response to arsenic species, which helps us to understand the toxicity of arsenic and the way in which plants have evolved arsenic resistances. This knowledge, for example, could be used produce plant cultivars that are more arsenic resistant or that have reduced arsenic uptake. This review synthesizes current knowledge on arsenic uptake, metabolism and toxicity for arsenic resistant and nonresistant plants, including the recently discovered phenomenon of arsenic hyperaccumulation in certain fern species. The reasons why plants accumulate and metabolize arsenic are considered in an evolutionary context. © New Phytologist.

• Differential resistance to arsenate (AsO4 3-) is demonstrated here among populations of the ericoid mycorrhizal fungus Hymenoscyphus ericae isolated from Calluna vulgaris in natural heathland soils and soils contaminated with AsO4 3-. • Isolates (c. 25) of the fungus from each of two As and Cu mine sites, and a natural heathland site, were screened for AsO4 3- and Cu2+ resistance by growing isolates in media containing a range of AsO4 3- and Cu2+ concentrations. • H. ericae populations from the mine sites demonstrated resistance to AsO4 3- compared with the heathland population; the mine-site populations producing significant growth at the highest AsO4 3- concentration (4.67 mol m-3), whereas growth of the heathland population was almost completely inhibited. EC50 values for mine-site isolates were estimated to be 5-41-times higher than the heathland population. All isolates produced identical responses to increasing Cu2+ concentrations, with no differences observed between mine-site and heathland isolates. • Populations of H. ericae on the contaminated mine sites have developed adaptive resistance to AsO4 3-. By contrast, Cu2+ resistance appears to be constitutive.

A total of 107 putative ericoid mycorrhizal endophytes were isolated from hair roots of Calluna vulgaris from two abandoned arsenic/copper mine sites and a natural heathland site in southwest England. The endophytes were initially grouped as 14 RFLP types, based on the results of ITS-RFLP analysis using the restriction endonucleases Hinf I, Rsa I and Hae III. ITS sequences were obtained for representative isolates from each RFLP type and compared phylogenetically with sequences for known ericoid mycorrhizal endophytes and selected ascomycetes. The majority of endophyte isolates (62-92%) from each site were identified as Hymenoscyphus ericae, but a number of other less common mycorrhizal RFLP types were also identified, all of which appear to have strong affinities with the order Leotiales. None of the less common RFLP types was isolated from C. vulgaris at more than one field site. Neighbour-joining analysis indicated similarities between the endophytes from C. vulgaris and mycorrhizal endophytes isolated from other Ericaceae and Epacridaceae hosts in North America and Australia.

The effects of Cd and Zn on cross-colonization by Paxillus involutus of Scots pine seedlings was examined by using pairs of ectomycorrhizal (ECM) and non-mycorrhizal (NM) seedlings grown in the same vessel. This was done to assess, first, the ability of P. involutus to colonize NM Scots pine seedlings by growth from colonized roots of other Scots pine seedlings in the presence of Cd or Zn, and, second whether ECM colonization of Scots pine by P. involutus provided a competitive advantage over NM seedlings. Ectomycorrhizal colonization of Scots pine was shown to be more sensitive than Scots pine itself to Cd and Zn, but prior colonization did provide a competitive advantage with respect to biomass production. This beneficial effect over NM seedlings was, however, equal in the control, Cd and Zn treatments, and was due simply to growth stimulation in the presence of ECM colonization. Cross-colonization from an ECM to a NM seedling was reduced but not prevented by Cd and Zn. Cd had a more negative effect on cross-colonization than on initial colonization of seedlings, whereas Zn had an equally inhibitory effect on both parameters. These results have important implications for plant establishment on metal-contaminated sites. If cross-colonization between plants is reduced by toxic metals, plant establishment on contaminated sites might be retarded.

A number of ectomycorrhizal (ECM) fungi, from sites uncontaminated by toxic metals, were investigated to determine their sensitivity to Cd2-, Pb2+, Zn2+ and Sb3-, measured as an inhibition of fungal biomass production. Isolates were grown in liquid media amended with the metals, individually (over a range of concentrations) and in combination (at single concentrations) to determine any significant interactions between the metals. Significant interspecific variation in sensitivity to Cd2+ and Zn2+ was recorded, while Pb2+ and Sb3- individually had little effect. The presence of Pb2+ and Sb3- in the media did however, ameliorate Cd2+ and Zn2+ toxicity in some circumstances. Interactions between Cd2+ and Zn2+ were investigated further over a range of concentrations. Zn2+ was found to significantly ameliorate the toxicity of Cd2+ to three of the four isolates tested. The influence of Zn2+ varied between ECM species and with the concentrations of metals tested.

The Abbey Theatre played a leading role in the politicisation of the revolutionary generation that won Irish freedom, but comparatively little is known about the men and women who formed the lifeblood of the institution: those whose radical politics drove them to fight in the 1916 Rising.

BACKGROUND: Preclinical studies have shown that statins, particularly simvastatin, can prevent growth in breast cancer cell lines and animal models. We investigated whether statins used after breast cancer diagnosis reduced the risk of breast cancer-specific, or all-cause, mortality in a large cohort of breast cancer patients.

METHODS: A cohort of 17,880 breast cancer patients, newly diagnosed between 1998 and 2009, was identified from English cancer registries (from the National Cancer Data Repository). This cohort was linked to the UK Clinical Practice Research Datalink, providing prescription records, and to the Office of National Statistics mortality data (up to 2013), identifying 3694 deaths, including 1469 deaths attributable to breast cancer. Unadjusted and adjusted hazard ratios (HRs) for breast cancer-specific, and all-cause, mortality in statin users after breast cancer diagnosis were calculated using time-dependent Cox regression models. Sensitivity analyses were conducted using multiple imputation methods, propensity score methods and a case-control approach.

RESULTS: There was some evidence that statin use after a diagnosis of breast cancer had reduced mortality due to breast cancer and all causes (fully adjusted HR = 0.84 [95% confidence interval = 0.68-1.04] and 0.84 [0.72-0.97], respectively). These associations were more marked for simvastatin 0.79 (0.63-1.00) and 0.81 (0.70-0.95), respectively.

CONCLUSIONS: In this large population-based breast cancer cohort, there was some evidence of reduced mortality in statin users after breast cancer diagnosis. However, these associations were weak in magnitude and were attenuated in some sensitivity analyses.

“CAPACITAR O DOENTE PARA A GESTÃO DA DOR” INTRODUÇÃO: Este Programa surge no âmbito do projeto de doutoramento em Enfermagem em que se pretende estudar o efeito de um Programa de Intervenção de Enfermagem na capacidade de gestão da dor por parte do doente oncológico com doença avançada e/ou do cuidador informal em domicilio. JUSTIFICAÇÃO: Um dos sintomas mais frequentes no doente oncológico com doença avançada é a dor. Segundo Palliative Care in European, a dor oncológica, tem uma importância especial porque o cancro é a segunda causa de morte em Portugal e por existir dor moderada a intensa em mais de 90% dos doentes em situação oncológica terminal. O desenvolvimento de um programa estruturado de intervenção de enfermagem que vá de encontro às necessidades do doente oncológico com doença avançada e/ou cuidador, relativamente à gestão da dor, poderá ter um impacto muito significativo no controlo da dor e de outros sintomas. Foi realizada pesquisa relativamente às duas teorias que suportam o estudo, a Teoria de Gestão de Sintomas e a Teoria dos Sintomas Desagradáveis. O Modelo de Gestão de sintomas, foi introduzido em 1994 na Escola de Enfermagem da Universidade de São Francisco na Califórnia, tendo sido revisto em 2001 por (Dodd; JANSON et al.). Foram então, desenvolvidos estudos científicos para a construção de um modelo baseado na evidência científica e também para comparar e contrastar os conceitos dos sintomas, com essa revisão, surge então a Teoria de Gestão de Sintomas. A Teoria de Gestão de Sintomas tem três conceitos essenciais: a experiência de sintomas, as estratégias de gestão de sintomas, e os resultados dos estados dos sintomas. A experiência de sintomas é conceptualizada enquanto algo que influencia e é influenciado tanto pelas estratégias de gestão de sintomas, como pelos resultados dos estados dos sintomas. A Teoria Dos Sintomas Desagradáveis, apresenta três conceitos principais: o (s) sintoma (s), fatores que influenciam e resultados de desempenho. Os sintomas podem ocorrer quer isoladamente, ou em combinação com outros sintomas. Em algumas situações, um sintoma pode preceder ou possivelmente dar origem a outro. Na Teoria Dos Sintomas Desagradáveis, os sintomas são conceptualizados como manifestando múltiplas dimensões e variáveis mensuráveis. Todos os sintomas variam em intensidade ou gravidade, grau de sofrimento associado, duração, e qualidade (Liehr & Smith, 2008). OBJETIVO Implementar um programa de Intervenção de Enfermagem para capacitar o doente na gestão da dor PALAVRAS CHAVES: Programa Educativo; Doente oncológico; dor oncológica MATERIAL A implementação do programa tem a duração prevista para 6 semanas. Compreende duas sessões educativas, três telefonemas e uma sessão de avaliação final. MÉTODOS 1.3 Trabalho de campo/empírico desenvolvido ao longo do semestre Durante dois meses, foi desenvolvido trabalho de campo no Hospital de dia, serviço de Oncologia, onde foi iniciada a investigação, relativamente à identificação de doentes oncológicos em situação de doença avançada. Para a identificação destes doentes assistiu a consultas de oncologia, com diferentes médicos, surgindo uma amostra intencional. O objetivo deste trabalho, foi certificar a existência de doentes oncológicos com doença avançada, prevendo uma média de doentes que se encontram a ser acompanhados naquele serviço. Como resultados e não esgotando a população, foram identificados cinquenta e nove doentes, a maioria do sexo feminino, com uma média de idade nos 65 anos, as neoplasias mais frequentes são a neoplasia do intestino e da mama. Todos os doentes apresentam metastização, sendo a localização mais frequente, o fígado, pulmão e osso. A maioria dos doentes apresenta capacidade de auto - cuidado e disponibilidade para a informação e ensino. A dor física não é referida como sintoma principal, mas como um mal-estar generalizado. O programa foi elaborado, após uma Revisão sistemática de Literatura, com base no programa “ THE PRO-SELF”, nas Orientações Genéricas preconizadas pelo Plano Nacional de Luta Contra a dor e na Intervenção de enfermagem “Controle da Dor” da Nursing Intervention Classification (NIC). RESULTADOS Foi feita uma aplicação em pré-teste do programa num serviço de oncologia, a três doentes oncológicos com doença avançada em domicílio. No início os doentes não apresentavam informação relativamente à gestão da dor, no final demonstraram capacidade para a monitorização da dor e de outros sintomas e para a gestão da terapêutica, realizando os registos num diário de bordo. A proximidade da morte e a alteração do estado de consciência interferem na autonomia e na tomada de decisão. CONCLUSÕES A aplicação deste programa deverá ter início numa fase mais precoce e não só na fase final de doença.
