The performance of the Weather Research and Forecast (WRF) model in wind simulation was evaluated under different numerical and physical options for an area of Portugal, located in complex terrain and characterized by its significant wind energy resource. The grid nudging and integration time of the simulations were the tested numerical options. Since the goal is to simulate the near-surface wind, the physical parameterization schemes regarding the boundary layer were the ones under evaluation. Also, the influences of the local terrain complexity and simulation domain resolution on the model results were also studied. Data from three wind measuring stations located within the chosen area were compared with the model results, in terms of Root Mean Square Error, Standard Deviation Error and Bias. Wind speed histograms, occurrences and energy wind roses were also used for model evaluation. Globally, the model accurately reproduced the local wind regime, despite a significant underestimation of the wind speed. The wind direction is reasonably simulated by the model especially in wind regimes where there is a clear dominant sector, but in the presence of low wind speeds the characterization of the wind direction (observed and simulated) is very subjective and led to higher deviations between simulations and observations. Within the tested options, results show that the use of grid nudging in simulations that should not exceed an integration time of 2 days is the best numerical configuration, and the parameterization set composed by the physical schemes MM5–Yonsei University–Noah are the most suitable for this site. Results were poorer in sites with higher terrain complexity, mainly due to limitations of the terrain data supplied to the model. The increase of the simulation domain resolution alone is not enough to significantly improve the model performance. Results suggest that error minimization in the wind simulation can be achieved by testing and choosing a suitable numerical and physical configuration for the region of interest together with the use of high resolution terrain data, if available.

Portugal, as well as the Mediterranean basin, is favorable to the occurrence of forest fires. In this work a statistical analysis was carried out based on the official information, considering the forest fires occurrences and the corresponding burned area for each of the districts of the mainland Portugal, between 1996 and 2010. Concerning to the forest fires occurrence it was possible to identify three main regions in mainland Portugal, while the burned area can be characterized in two main regions. Associations between districts and years are different in the two approaches. The results obtained provide a synthetic analysis of the phenomenon of forest fires in continental Portugal, based on all the official information available to date.

Due to their detrimental effects on human health, the scientific interest in ultrafine particles (UFP) has been increasing, but available information is far from comprehensive. Compared to the remaining population, the elderly are potentially highly susceptible to the effects of outdoor air pollution. Thus, this study aimed to (1) determine the levels of outdoor pollutants in an urban area with emphasis on UFP concentrations and (2) estimate the respective dose rates of exposure for elderly populations. UFP were continuously measured over 3 weeks at 3 sites in north Portugal: 2 urban (U1 and U2) and 1 rural used as reference (R1). Meteorological parameters and outdoor pollutants including particulate matter (PM10), ozone (O3), nitric oxide (NO), and nitrogen dioxide (NO2) were also measured. The dose rates of inhalation exposure to UFP were estimated for three different elderly age categories: 64–70, 71–80, and >81 years. Over the sampling period levels of PM10, O3 and NO2 were in compliance with European legislation. Mean UFP were 1.7 × 104 and 1.2 × 104 particles/cm3 at U1 and U2, respectively, whereas at rural site levels were 20–70% lower (mean of 1 ×104 particles/cm3). Vehicular traffic and local emissions were the predominant identified sources of UFP at urban sites. In addition, results of correlation analysis showed that UFP were meteorologically dependent. Exposure dose rates were 1.2- to 1.4-fold higher at urban than reference sites with the highest levels noted for adults at 71–80 yr, attributed mainly to higher inhalation rates.

RESUMO - Na radiologia de intervenção, e concretamente em ortopedia, os raios X são intensamente utilizados permitindo a visualização de diversas técnicas de intervenção cirúrgica. Do ponto de vista médico as vantagens dessa prática são enormes, contudo, os profissionais de saúde envolvidos são susceptíveis de estarem expostos a valores de dose de radiação que significam a sua classificação como trabalhadores expostos. O presente estudo realizou-se num hospital e teve como objectivo obter uma estimativa das doses envolvidas em ortopedia de intervenção, utilizando várias metodologias experimentais para caracterização do campo de radiação primário e secundário. Observaram-se distintos níveis de dose de acordo com a zona anatómica exposta à radiação X: (i) gónadas — 0,02 a 3 mGy/h; (ii) cristalino — 0,06 a 1 mGy/h e (iii) mãos — 0,6 mGy/h. Tais resultados evidenciam uma clara necessidade de utilização de equipamentos de protecção e de vigilância dosimétrica pelos profissionais de saúde envolvidos no acto cirúrgico.--------------------------ABSTRACT - In intervention radiology, and more specifically in orthopaedics, X-rays are intensely used allowing the visualization of many acts of clinical intervention. From a clinical perspective, the advantages of that practice are significant; however, involved health care professionals are susceptible of being exposed to radiation dose values that mean their classification as exposed workers. The present study, performed in a hospital, aimed to obtain an estimation of the doses involved in intervention orthopaedics through several experimental methodologies in order to characterise the primary and the secondary radiation fields. Different levels of dose were observed according to the anatomic area exposed to X radiation: (i) gonads — 0.02 a 3 mGy/h; (ii) crystalline lens — 0.06 a 1 mGy/h e (iii) hands – 0.6 mGy/h. The excessive use of pesticides and fertilisers in agriculture has generated a decrease in groundwater and surface water quality in many regions of the EU, constituting a hazard for human health and the environment. Besides, on-site sewage disposal is an important source of groundwater contamination in urban and peri-urban areas. The assessment of groundwater vulnerability to contamination is an important tool to fulfil the demands of EU Directives. The purpose of this study is to assess the groundwater vulnerability to contamination related mainly to agricultural activities in a peri-urban area (Vila do Conde, NW Portugal). The hydrogeological framework is characterised mainly by fissured granitic basement and sedimentary cover. Water samples were collected and analysed for temperature, pH, electrical conductivity, chloride, phosphate, nitrate and nitrite. An evaluation of groundwater vulnerability to contamination was applied (GOD-S, Pesticide DRASTIC-Fm, SINTACS and SI) and the potential nitrate contamination risk was assessed, both on a hydrogeological GIS-based mapping. A principal component analysis was performed to characterised patterns of relationship among groundwater contamination, vulnerability, and the hydrogeological setting assessed. Levels of nitrate above legislation limits were detected in 75 % of the samples analysed. Alluvia units showed the highest nitrate concentrations and also the highest vulnerability and risk. Nitrate contamination is a serious problem affecting groundwater, particularly shallow aquifers, especially due to agriculture activities, livestock and cesspools. GIS-based cartography provided an accurate way to improve knowledge on water circulation models and global functioning of local aquifer systems. Finally, this study highlights the adequacy of an integrated approach, combining hydrogeochemical data, vulnerability assessments and multivariate analysis, to understand groundwater processes in peri-urban areas.

The McArthur Township area in the Archean Abitibi Belt of northeast Ontario contains northwesterly trending volcanic rocks which are located on a limb of a large syncline. The axial trace of the syncline passes through the adjacent Douglas Township. The Archean volcanic rocks and associated sedimentary rocks are intruded and deformed by two large plutons and a few smaller hypabyssal intrusions. A petrographic and geochemical study of the Precambrian rocks exposed 1n the study area was undertaken in order to investigate the metamorphic grade and geochemical characteristics of the rocks. All the samples were studied with the microscope and analysis of 20 major and trace elements were determined on a selection of the less altered specimens by x-ray fluorescence. Three different periods of igneous activity have occurred in the study area. The first two periods were dominated by volcanic extrusive rocks accompanied by gabbroic sills. The third cycle is the diapiric intrusion of the granitic plutons and subsequent metamorphism of the older rocks to the low to medium grade. Two periods of sedimentation are also recognized in the study area which occurred after the first and second cycle of volcanism. Chemically, the lavas are subdivided into three main associations: (1) The komatiitic association is characterized by high MgO, high Ni, low Ti02 and a low FeO*/(FeO* + MgO) ratio. They occupy the base of each volcanic cycle and some of the flows exhibit spinifex textures. (2) The tholeiitic association displays distinct iron and titanium enrichment trends in the intermediate membersor -i r (3) The calc-alkaline association contains low FeO* and TI02 and high Ni contents relative to modern calc-alkaline types. They are formed at the end of each cycle of volcanism and overlie the tholeiitic flows. All three associations of the first volcanic cycle are exposed in the study area, while the second cycle is represented by a komatiltic sequence. The volcanic rocks were possibly formed by multiple partial melting of the Archean mantle to produce parental magmas under various P - T conditions.

The proce-ss ofoxygenic photosynthesis is vital to life on Earth. the central event in photosynthesis is light induced electron transfer that converts light into energy for growth. Ofparticular significance is the membrane bound multisubunit protein known as Photosystem I (PSI). PSI is a reaction centre that is responsible for the transfer of electrons across the membrane to reduce NADP+ to NADPH. The recent publication ofa high resolution X-ray structure of PSI has shown new information about the structure, in particular the electron transfer cofactors, which allows us to study it in more detail. In PSI, the secondary acceptor is crucial for forward electron transfer. In this thesis, the effect of removing the native acceptor phylloquinone and replacing it with a series of structurally related quinones was investigated via transient electron paramagnetic resonance (EPR) experiments. The orientation of non native quinones in the binding site and their ability to function in the electron transfer process was determined. It was found that PSI will readily accept alkyl naphthoquinones and anthraquinone. Q band EPR experiments revealed that the non-native quinones are incorporated into the binding site with the same orientation of the headgroup as in the native system. X band EPR spectra and deuteration experiments indicate that monosubstituted naphthoquinones are bound to the Al site with their side group in the position occupied by the methyl group in native PSI (meta to the hydrogen bonded carbonyl oxygen). X band EPR experiments show that 2, 3- disubstituted methyl naphthoquinones are also incorporated into the Al site in the same orientation as phylloquinone, even with the presence of a halogen- or sulfur-containing side chain in the position normally occupied by the phytyl tail ofphylloquinone. The exception to this is 2-bromo-3-methyl --.- _. -. - -- - - 4 _._ _ _ - _ _ naphthoquinone which has a poorly resolved spectrum, making determination of the orientation difficuh. All of the non-native quinones studied act as efficient electron acceptors. However, forward electron transfer past the quinone could only be demonstrated for anthraquinone, which has a more negative midpoint potential than phylloquinone. In the case of anthraquinone, an increased rate of forward electron transfer compared to native PSI was found. From these results we can conclude that the rate ofelectron transfer from Al to Fx in native PSI lies in the normal region ofthe Marcus Curve.

Photosynthesis is a process in which electromagnetic radiation is converted into chemical energy. Photosystems capture photons with chromophores and transfer their energy to reaction centers using chromophores as a medium. In the reaction center, the excitation energy is used to perform chemical reactions. Knowledge of chromophore site energies is crucial to the understanding of excitation energy transfer pathways in photosystems and the ability to compute the site energies in a fast and accurate manner is mandatory for investigating how protein dynamics ef-fect the site energies and ultimately energy pathways with time. In this work we developed two software frameworks designed to optimize the calculations of chro-mophore site energies within a protein environment. The first is for performing quantum mechanical energy optimizations on molecules and the second is for com-puting site energies of chromophores in a fast and accurate manner using the polar-izability embedding method. The two frameworks allow for the fast and accurate calculation of chromophore site energies within proteins, ultimately allowing for the effect of protein dynamics on energy pathways to be studied. We use these frame-works to compute the site energies of the eight chromophores in the reaction center of photosystem II (PSII) using a 1.9 Å resolution x-ray structure of photosystem II. We compare our results to conflicting experimental data obtained from both isolat-ed intact PSII core preparations and the minimal reaction center preparation of PSII, and find our work more supportive of the former.

La dihydrofolate réductase humaine (DHFRh) est une enzyme essentielle à la prolifération cellulaire. Elle réduit le dihydrofolate en tétrahydrofolate, un co-facteur impliqué dans la biosynthèse des purines et du thymidylate. La DHFRh est une cible de choix pour des agents de chimiothérapie comme le méthotrexate (MTX), inhibant spécifiquement l’enzyme ce qui mène à un arrêt de la prolifération et ultimement à la mort cellulaire. Le MTX est utilisé pour le traitement de plusieurs maladies prolifératives, incluant le cancer. La grande utilisation du MTX dans le milieu clinique a mené au développement de mécanismes de résistance, qui réduisent l’efficacité de traitement. La présente étude se penche sur l’un des mécanismes de résistance, soit des mutations dans la DHFRh qui réduisent son affinité pour le MTX, dans le but de mieux comprendre les éléments moléculaires requis pour la reconnaissance de l’inhibiteur au site actif de l’enzyme. En parallèle, nous visons à identifier des variantes plus résistantes au MTX pour leur utilisation en tant que marqueurs de sélection en culture cellulaire pour des systèmes particuliers, tel que la culture de cellules hématopoïétiques souches (CHS), qui offrent des possibilités intéressantes dans le domaine de la thérapie cellulaire. Pour étudier le rôle des différentes régions du site actif, et pour vérifier la présence d’une corrélation entre des mutations à ces régions et une augmentation de la résistance au MTX, une stratégie combinatoire a été dévelopée pour la création de plusieurs banques de variantes à des résidus du site actif à proximité du MTX lié. Les banques ont été sélectionnées in vivo dans un système bactérien en utilisant des milieux de croissance contenant des hautes concentrations de MTX. La banque DHFRh 31/34/35 généra un nombre considérable de variantes combinatoires de la DHFRh hautement résistantes au MTX. Les variantes les plus intéressantes ont été testées pour leur potentiel en tant que marqueur de sélection dans plusieurs lignées cellulaires, dont les cellules hématopoïétiques transduites. Une protection complète contre les effets cytotoxiques du MTX a été observée chez ces cellules suite à leur infection avec les variantes combinatoires. Pour mieux comprendre les causes moléculaires reliées à la résistance au MTX, des études de structure tridimensionnelle de variantes liées au MTX ont été entreprises. La résolution de la structure de la double variante F31R/Q35E lié au MTX a révélé que le phénotype de résistance était attribuable à d’importantes différences entre le site actif de la double variante et de l’enzyme native, possiblement dû à un phénomème dynamique. Une compréhension plus générale de la reconnaissance et la résistance aux antifolates a été réalisée en comparant des séquences et des structures de variantes de la DHFR résistants aux antifolates et provenant de différentes espèces. En somme, ces travaux apportent de nouveaux éléments pour la comprehension des intéractions importantes entre une enzyme et un ligand, pouvant aider au développement de nouveaux antifolates plus efficaces pour le traitement de diverses maladies. De plus, ces travaux ont généré de nouveaux gènes de résistance pouvant être utilisés en tant que marqueurs de sélection en biologie cellulaire.

Kabazi II est un site de plein air, situé sur la deuxième rangée des Monts de Crimée. Après sa découverte en 1986, les investigations archéologiques effectuées entre 1987 et le milieu des années 90 ont établi que Kabazi II avait auparavant servi de lieu de chasse et d’abattage pour les groupes néanderthaliens de la région. Les études archéozoologiques antérieures (Patou-Mathis 2003, 1999, 2005, 2006a, 2006b) ont déterminé que les stratégies de subsistance des Néanderthaliens du Kabazi II étaient très spécialisées et principalement axées sur la chasse des petits groupes de Equus hydruntinus mais aussi, à l’occasion, sur la chasse d’autres espèces. Ces comportements ont persisté malgré les changements climatiques et technologiques à travers l’histoire d’occupation du site. Cette étude présente l’analyse des assemblages fauniques encore inédits des niveaux II/1,II/2-1, II/2, II/3, II/4, II/5, II/7, II/8, II/9, II/13, II/13A de Kabazi II. Nos résultats sont en accord avec ceux obtenus parles d’études antérieures ; cependant, des différences par rapport à la fonction du site ont été constatées et un lien possible avec Kabazi V, un abri sur roche tout près de Kabazi II, a été établi. On croit que la persistance des activités de subsistance des Néanderthaliens de Kabazi II pendant presque 100 000 ans de présence est due à la polyvalence des ânes asiatiques tels que Equus hydruntinus, au contexte géographique et géologique de la région ainsi qu’aux caractéristiques du site elles-mêmes.

Ce mémoire tente d’établir la position sociale et culturelle de la communauté villageoise du XVᵉ siècle du site Droulers dans l’espace iroquoien du Saint-Laurent. À partir d’une analyse morpho-stylistique de la poterie, et particulièrement les vases décorés au dentelé, ce mémoire examine la variabilité culturelle du site et sa participation au sein de la sphère d’interactions des Iroquoiens du Saint-Laurent. La comparaison des tendances stylistiques de Droulers et des communautés voisines contribue ainsi à cerner la position chronologique ainsi que l’apparentement culturel du site au sein de sa région immédiate et des régions occidentale et centrale. Les caractères stylistiques à la fois conservateurs et progressistes relevés sur le site Droulers lui sont propres et expriment à la fois l’homogénéité du site et une certaine indépendance stylistique au sein de sa région. Sur cette base, nous avons déterminé que l’usage du dentelé n’a pas une valeur chronologique fiable à des fins de sériation dans le cas spécifique de Droulers, mais qu’il peut toutefois servir comme marqueur régional distinctif. Cet attribut ainsi que d’autres tendances régionales significatives nous ont ainsi servi à mieux cerner les similarités stylistiques entre les sites et à déterminer que Droulers s’apparente plus particulièrement aux sites Mandeville et Lanoraie, et dans une moindre mesure au site McIvor. De plus, nous avons pu établir que le site Droulers s’intègre dans un réseau d’interactions complexe, le rapprochant de communautés situées autant à l’Est qu’à l’Ouest le long de la vallée du Saint-Laurent. Finalement, l’ensemble des tendances morpho-stylistiques confirme la position chronologique du site, soit à la fin du XVᵉ siècle, et ce malgré une proportion importante du décor au dentelé, traditionnellement considéré comme une caractéristique des sites plus anciens.

Mon mémoire de maîtrise a été réalisé dans le cadre du projet Génorem (www.genorem.ca), un projet multidisciplinaire qui réunit différents chercheurs de l'Université de Montréal et de l'Université McGill dans le but d'améliorer les techniques utilisées en bioremédiation. Dans le cadre de l'étude, des saules à croissance rapide (Salix sp.) ont été utilisés comme plantes modèles dans l'étude. Ainsi, 11 cultivars de saule ont été suivis afin de déterminer leur potentiel à produire un bon rendement de biomasse, à tolérer des conditions de stress sévère causé par la présence de HAPs (hydrocarbures aromatiques polycycliques) , BPCs (biphényles polychlorés) et d'hydrocarbures pétroliers C10-C50. L'expérimentation consistait en une plantation de saule à forte densité qui a été mise en place en 2011 sur le site d'une ancienne industrie de pétrochimie à Varennes, dans le sud du Québec. Les boutures des génotypes sélectionnés ont été plantées sur une superficie d'environ 5000 m2. Les plantes ont été suivies pendant les deux saisons de croissance suivant le recépage et une série de paramètres de croissance et de mesures physiologiques ont été récoltés (surface foliaire, taux de chlorophylle, conductance stomatique et statut nutritionnel) dans le but d'évaluer et de comparer les performances de chaque génotype sur un sol pollué. Les analyses statistiques ont démontré que le cultivar S. miyabeana (SX61) était le meilleur producteur de biomasse sur le site contaminé, tandis que S. nigra (S05) et S. acutifolia (S54) présentaient la meilleure capacité photosynthétique. S. dasyclados (SV1), S. purpurea (‘Fish Creek’) et S. caprea (S365) ont semblé particulièrement affectés par la présence de contaminants. La capacité d'établissement et la croissance de S. nigra (S05), S. eriocephala (S25) and S. purpurea x S. miyabeana (‘Millbrook’) indiquent une tolérance globale supérieure à la pollution . Cette analyse comparative des différentes réponses physiologiques des saules cultivés sur un sol contaminé pourra guider le processus de sélection de plantes et les techniques de bioremédiation dans les futurs projets de phytoremédiation.
