Recent investigation of the intestine following ischemia and reperfusion (I/R) has revealed that nitric oxide synthase (NOS) neurons are more strongly affected than other neuron types. This implies that NO originating from NOS neurons contributes to neuronal damage. However, there is also evidence of the neuroprotective effects of NO. In this study, we compared the effects of I/R on the intestines of neuronal NOS knockout (nNOS(-/-)) mice and wild-type mice. I/R caused histological damage to the mucosa and muscle and infiltration of neutrophils into the external muscle layers. Damage to the mucosa and muscle was more severe and greater infiltration by neutrophils occurred in the first 24 h in nNOS(-/-) mice. Immunohistochemistry for the contractile protein, alpha-smooth muscle actin, was used to evaluate muscle damage. Smooth muscle actin occurred in the majority of smooth muscle cells in the external musculature of normal mice but was absent from most cells and was reduced in the cytoplasm of other cells following I/R. The loss was greater in nNOS(-/-) mice. Basal contractile activity of the longitudinal muscle and contractile responses to nerve stimulation or a muscarinic agonist were reduced in regions subjected to I/R and the effects were greater in nNOS(-/-) mice. Reductions in responsiveness also occurred in regions of operated mice not subjected to I/R. This is attributed to post-operative ileus that is not significantly affected by knockout of nNOS. The results indicate that deleterious effects are greater in regions subjected to I/R in mice lacking nNOS compared with normal mice, implying that NO produced by nNOS has protective effects that outweigh any damaging effect of this free radical produced by enteric neurons.

Nitroglycerin (GIN) has been clinically used to treat angina pectoris and acute heart episodes for over 100 years. The effects of GTN have long been recognized and active research has contributed to the unraveling of numerous metabolic routes capable of converting GIN to the potent vasoactive messenger nitric oxide. Recently, the mechanism by which minute doses of GIN elicit robust pharmacological responses was revisited and eNOS activation was implicated as an important route mediating vasodilation induced by low GTN doses (1-50 nM). Here, we demonstrate that at such concentrations the pharmacologic effects of nitroglycerin are largely dependent on the phosphatidylinositol 3-kinase, Akt/PKB, and phosphatase and tensin homolog deleted on chromosome 10 (PTEN) signal transduction axis. Furthermore, we demonstrate that nitroglycerin-dependent accumulation of 3,4,5-InsP(3), probably because of inhibition of PTEN, is important for eNOS activation, conferring a mechanistic basis for GIN pharmacological action at pharmacologically relevant doses. Increased neuronal oxidative stress (OxS) induces deleterious effects on signal transduction, structural plasticity and cellular resilience, mainly by inducing lipid peroxidation in membranes, proteins and genes. Major markers of OxS levels include the thiobarbituric acid reactive substances (TBARS) and the enzymes superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase. Lithium has been shown to prevent and/or reverse DNA damage, free-radical formation and lipid peroxidation in diverse models. This study evaluates OxS parameters in healthy volunteers prior to and following lithium treatment. Healthy volunteers were treated with lithium in therapeutic doses for 2-4 weeks. Treatment with lithium in healthy volunteers selectively altered SOD levels in all subjects. Furthermore, a significant decrease in the SOD/CAT ratio was observed following lithium treatment, wich was associated with decreased OxS by lowering hydrogen peroxide levels. This reduction in the SOD/CAT ratio may lead to lower OxS, indicated primarily by a decrease in the concentration of cell hydrogen peroxide. Overall, the present findings indicate a potential role for the antioxidant effects of lithium in healthy subjects, supporting its neuroprotective profile in bipolar disorder (BD) and, possibly, in neurodegenerative processes.

Tempol (4-hydroxy-2,2,6,6-tetramethylpiperidine-1-oxyl) and other cyclic nitroxides have been shown to inhibit the chlorinating activity of myeloperoxidase (MPO) in vitro and in cells. To examine whether nitroxides inhibit MPO activity in vivo we selected acute carrageenan-induced inflammation on the rat paw as a model. Tempol and three more hydrophobic 4-substituted derivatives (4-azido, 4-benzene-Sulfonyl, and 4-(4-phenyl-1H-1,2,3-triazol-1-yl)) were synthesized, and their ability to inhibit the in vitro chlorinating activity of MPO and carrageenan-induced inflammation in rat paws was evaluated. All of the tested nitroxides inhibited the chlorinating activity of MPO in vitro with similar IC50 values (between 1.5 and 1.8 mu M). In vivo, the attenuation of carrageenan-induced inflammation showed some correlation with the lipophilicity of the nitroxide at early time points but the differences in the effects were small (< 2-fold) compared with the differences in lipophilicity (> 200-fold). No inhibition of MPO activity in vivo was evident because the levels of MPO activity in rat paws correlated with the levels of MPO protein'. Likewise, paw edema, levels of nitrated and oxidized proteins, and levels of plasma exudation correlated with the levels of MPO protein in the paws of the animals that were untreated or treated with the nitroxides. The effects of the nitroxides in vivo were compared with those of 4-aminobenzoic hydrazide and of colchicine. Taken together, the results indicate that nitroxides attenuate carrageenan-induced inflammation mainly by reducing neutrophil migration and the resulting MPO-mediated damage. Accordingly, tempol was shown to inhibit rat neutrophil migration in vitro. (C) 2012 Elsevier Inc. All rights reserved.

This article studied the applicability of poly(acrylamide) and methylcellulose (PAAm-MC) hydrogels as potential delivery vehicle for the controlled-extended release of ammonium sulfate (NH(4))(2)SO(4) and potassium phosphate (KH(2)PO(4)) fertilizers. PAAm-MC hydrogels with different acrylamide (AAm) and MC concentrations were prepared by a free radical polymerization method. The adsorption and desorption kinetics of fertilizers were determined using conductivity measurements based on previously built analytical curve. The addition of MC in the PAAm chains increased the quantities of (NH(4))(2)SO(4) and KH(2)PO(4) loaded and extended the time and quantities of fertilizers released. Coherently, both loading and releasing processes were strongly influenced by hydrophilic properties of hydrogels (AAm/MC mass proportion). The best sorption (124.0 mg KH(2)PO(4)/g hydrogel and 58.0 mg (NH(4))(2)SO(4)/g hydrogel) and desorption (54.9 mg KH(2)PO(4)/g hydrogel and 49.5 mg (NH(4))(2)SO(4)/g hydrogel) properties were observed for 6.0% AAm-1.0% MC hydrogels (AAm/MC mass proportion equal 6), indicating that these hydrogels are potentially viable to be used in controlled-extended release of fertilizers systems. The new pathway nitrate-nitrite-nitric oxide (NO) has emerged as a physiological alternative to the classical enzymatic pathway for NO formation from L-arginine. Nitrate is converted to nitrite by commensal bacteria in the oral cavity and the nitrite formed is then swallowed and reduced to NO under the acidic conditions of the stomach. In this study, we tested the hypothesis that increases in gastric pH caused by omeprazole could decrease the hypotensive effect of oral sodium nitrite. We assessed the effects of omeprazole treatment on the acute hypotensive effects produced by sodium nitrite in normotensive and L-NAME-hypertensive free-moving rats. In addition, we assessed the changes in gastric pH and plasma levels of nitrite, NOx (nitrate+ nitrite), and S-nitrosothiols caused by treatments. We found that the increases in gastric pH induced by omeprazole significantly reduced the hypotensive effects of sodium nitrite in both normotensive and L-NAME-hypertensive rats. This effect of omeprazole was associated with no significant differences in plasma nitrite, NOx, or S-nitrosothiol levels. Our results suggest that part of the hypotensive effects of oral sodium nitrite may be due to its conversion to NO in the acidified environment of the stomach. The increase in gastric pH induced by treatment with omeprazole blunts part of the beneficial cardiovascular effects of dietary nitrate and nitrite. (c) 2012 Elsevier Inc. All rights reserved.

Melanins have been associated with the development of melanoma and its resistance to photodynamic therapy (PDT). Singlet molecular oxygen (102), which is produced by ultraviolet A solar radiation and the PDT system, is also involved. Here, we investigated the effects that these factors have on DNA damage and repair. Our results show that both types of melanin (eumelanin and pheomelanin) lead to DNA breakage in the absence of light irradiation and that eumelanin is more harmful than pheomelanin. Interestingly, melanins were found to bind to the minor grooves of DNA, guaranteeing close proximity to DNA and potentially causing the observed high levels of strand breaks. We also show that the interaction of melanins with DNA can impair the access of repair enzymes to lesions, contributing to the perpetuation of DNA damage. Moreover, we found that after melanins interact with 102, they exhibit a lower ability to induce DNA breakage; we propose that these effects are due to modifications of their structure. Together, our data highlight the different modes of action of the two types of melanin. Our results may have profound implications for cellular redox homeostasis, under conditions of induced melanin synthesis and irradiation with solar light. Protein disulfide isomerase (PDI) and its homologs are oxidoreductases facilitating protein folding in the ER. Endo-PDI (also termed ERp46) is highly expressed in endothelial cells. It belongs to the PDI family but its physiological function is largely unknown. We studied the role of Endo-PDI in endothelial angiogenic responses. Stimulation of human umbilical vein endothelial cells (with TNFα (10ng/ml) increased ERK1/2 phosphorylation. This effect was largely attenuated by Endo-PDI siRNA, whereas JNK and p38 MAP kinase phosphorylation was Endo-PDI independent. Similarly, TNFα-stimulated NF-κB signaling determined by IκBα degradation as well as TNFα-induced ICAM expression was unaffected by Endo-PDI siRNA. The action of Endo-PDI was not mediated by extracellular thiol exchange or cell surface PDI as demonstrated by nonpermeative inhibitors and PDI-neutralizing antibody. Moreover, exogenously added PDI failed to restore ERK1/2 activation after Endo-PDI knockdown. This suggests that Endo-PDI acts intracellularly potentially by maintaining the Ras/Raf/MEK/ERK pathway. Indeed, knockdown of Endo-PDI attenuated Ras activation measured by G-LISA and Raf phosphorylation. ERK activation influences gene expression by the transcriptional factor AP-1, which controls MMP-9 and cathepsin B, two proteases required for angiogenesis. TNFα-stimulated MMP-9 and cathepsin B induction was reduced by silencing of Endo-PDI. Accordingly, inhibition of cathepsin B or Endo-PDI siRNA blocked the TNFα-stimulated angiogenic response in the spheroid outgrowth assays. Moreover ex vivo tube formation and in vivo Matrigel angiogenesis in response to TNFα were attenuated by Endo-PDI siRNA. In conclusion, our study establishes Endo-PDI as a novel, important mediator of AP-1-driven gene expression and endothelial angiogenic function

[EN]Most marine fish larvae require high amounts of n-3 HUFA (highly unsaturated fatty acids) such as eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) (Watanabe, 1982; Izquierdo, 1996). Fish larvae tissue lipids are also very high in n-3 HUFA, what implies a higher risk of peroxidation (Sargent et al. 1999) and cellular damage (Kanazawa, 1991), requiring then antioxidants to protect them intra- and extra-cellularly from free radical compounds. Vitamin E (Vit E) functions as a chain breaking antioxidant, reacting with the lipid peroxide radical produced and preventing the further reaction with a new PUFA. Hence their requirements are related with the dietary and tissue PUFA contents. The objective of the present study was to determine the effect of dietary Vit E on gilthead sea bream and sea bass survival, growth and stress, at different n-3 HUFA levels.

[EN] In the regions of Canary Islands, it is prevailing all the year a high level of solar radiation. This force plants to develop defence mechanisms against excessive production of free radicals through the accumulation of antioxidant substances. Plants contain a diverse group of phenolic compounds with the structural requirements of free radical scavengers. The effects of different extracting solvents have been tested for the extraction of phenolic compounds from plant material. On the present study we compare the total phenolic contents (TPC) and antioxidant activities of several extracts derived from Aloe vera plants. The phenolic compounds present in the extracts were identified and quantified by RP­HPLC techniques.

[EN] Phytochemical research of two Tolpis species, T. proustii and T. lagopoda, led to the isolation of three new compounds: 30-chloro-3beta-acetoxy-22alfa-hydroxyl-20(21)- taraxastene (1), 3beta,22alfa-diacetoxy-30-ethoxy-20(21)-taraxastene (2) and 3beta,28-dihydroxy- 11alfa-hydroperoxy-12-ursene (3). The structures of the new compounds were elucidated by means of extensive IR, NMR, and MS data and by comparison of data reported in the literature. The in vitro antioxidant activities of the extracts were assessed by the DPPH and ABTS scavenging methods. The cytotoxicity of several known compounds and its derivatives was also assessed against human myeloid leukemia K-562 and K-562/ADR cell lines.

Oxidative stress is considered to be of major relevance for a variety of pathological processes. Thus, it is valuable to identify compounds, which might act as antioxidants, i.e. compounds that antagonize the deleterious action of reactive oxygen species (ROS) on biomolecules. The mode of action of these compounds could be either to scavenge ROS directly or to trigger protective mechanisms inside the cell, thereby resulting in improved defense against ROS. Sulforaphane (SF) (1-isothiocyanato-(4R)-(methylsulfinyl)butane) is a naturally occurring cancer chemopreventive agent found as a precursor glucosinolate in Cruciferous vegetables like broccoli. Although SF is not a direct-acting antioxidant, there is substantial evidence that SF acts indirectly to increase the antioxidant capacity of animal cells and their abilities to cope with oxidative stress. Induction of phase 2 enzymes is one means by which SF enhances the cellular antioxidant capacity. Enzymes induced by SF include Glutathione S-transferases (GST) and NAD[P]H:quinone oxidoreductase (NQO1) which can function as protectors against oxidative stress. To protect themselves from oxidative stress, cells are equipped with reducing buffer systems including the GSH and thioredoxin (Trx) reductase. GSH is an important tripeptide thiol which in addition to being the substrate for GSTs maintains the cellular oxidation– reduction balance and protects cells against free radical species. Aim of the first part of this thesis was to investigate the ability of SF to induce the expression and the activity of different phase 2 and antioxidant enzymes (such as GST, GR, GPx, NQO1, TR, SOD, CAT) in an in vitro model of rat cardiomyocytes, and also to define if SF treatment supprts cells in counteracting oxidative stress induced by H2O2 It is well known that acute exhaustive exercise causes significant reactive oxygen species generation that results in oxidative stress, which can induce negative effects on health and well being. In fact, increased oxidative stress and biomarkers (e.g., protein carbonyls, MDA, and 8- hydroxyguanosine) as well as muscle damage biomarkers (e.g. plasmatic Creatine cinase and Lactate dehydrogenase) have been observed after supramaximal sprint exercises, exhaustive longdistance cycling or running as well as resistance-type exercises, both in trained and untrained humans. Markers of oxidative stress also increase in rodents following exhaustive exercise. Moreover, antioxidant enzyme activities and expressions of antioxidant enzymes are known to increase in response to exhaustive exercise in both animal and human tissues. Aim of this project was to evaluate the effect of SF supplementation in counteracting oxidative stress induced by physical activity through its ability to induce phase 2, and antioxidant enzymes in rat muscle. The results show that SF is a nutraceutical compound able to induce the activity of different phase 2 and antioxidant enzymes in both cardiac muscle and skeletal muscle. Thanks to its actions SF is becoming a promising molecule able to prevent cardiovascular damages induced by oxidative stress and muscle damages induced by acute exhaustive exercise.

Im Rahmen dieser Dissertation wurden die Synthese und die Charakterisierung verschiedener, zum Teil neuartiger Blockcopolymere beschrieben, wobei die Einbeziehung radikalischer Polymeri-sationsmechanismen den konzeptionellen Kern ausmachte. Mit einer auf die jeweilige Kombination von Monomeren zugeschnittenen Syntheseroute gelang die Verknüpfung von Segmenten, die allein mittels der herkömmlich zur Synthese von Blockcopolymeren genutzten, ionischen Mechanismen nur mit hohem Aufwand oder gar nicht zu verbinden sind. Auf materieller Seite stand die Herstellung amphiphiler Strukturen im Vordergrund. Diese wurden entweder direkt beim Aufbau der Blockcopo-lymere oder nach anschließender polymeranaloger Umsetzung eines ihrer Segmente erhalten. Solche amphiphilen Substanzen besitzen aufgrund ihrer Grenzflächenaktivität Anwendungspotential z. B. als Stabilisatoren in der Dispersionspolymerisation oder als Flokkulantien. Es wurden drei Verfahren zum Aufbau von Blockcopolymeren untersucht:1. Die Transformation von anionischer zu freier radikalischer Polymerisation für die Synthese von Polystyrol-b-poly(N-vinylformamid) (PS-b-P(VFA)).2. Die Transformation von anionischer zu kontrollierter radikalischer Polymerisation (ATRP) für den Aufbau von Blockcopolymeren aus Poly(dimethylsiloxan) PDMS und Segmenten von t-Butylacrylat (t-BuA) bzw. (2-(Trimethylsiloxy)ethyl)methacrylat (TMS-HEMA).3. Die kontrollierte radikalische Polymerisation unter Einsatz von Triazolinyl als Gegenradikal zur Synthese von Poly[(2-(trimethylsiloxy)ethyl)methacrylat]-b-polystyrol (P(TMS-HEMA)-b-PS) als alternative Route zur anionischen Polymerisation.

In den letzten Jahren ist die Nachfrage nach Datenspeichern und ihrer Speicherkapazität immer größer geworden. Dazu sind neue Speichermaterialien notwendig sind, wofür sich in vielen Fällen polymere schaltbare Materialien eignen.Aus diesem Grunde wurden in der vorliegenden Arbeit polymergebundene Nitrone hergestellt, um diese als photoreaktives Material zu untersuchen. Die direkte freie radikalische Polymerisation war nicht möglich. Um zu den polymergebundenen Nitronen zu gelangen, mussten zuerst die Doppelbindungen enthaltenden Aldehyde polymerisiert und dann polymeranalog, meist quantitativ mit den N?Alkylhydroxylaminen zum Nitron kondensiert werden. Des weiteren wurden nitronhaltige Polymere auf direktem Weg durch enzymatisch-oxidative Polymerisation von nitronhaltigen Phenolen erhalten.Auch wurden Aldehydmonomere mit dem sperrigen dimethylierten b-Cyclodextrin als 'Schutzgruppe' komplexiert und im wässrigen Medium radikalisch polymerisiert, was zu höheren Molgewichten führte. Hierbei trat auch ein interessanter Verzögerungseffekt bei der Polymerisation von 4-Vinylbenzaldehyd auf, was sich in längeren Polymerisationszeiten und geringeren Polymerisationsausbeuten bemerkbar machte.Die UV-Bestrahlung der Nitronfunktion führt zum entsprechenden dreigliedrigen Oxaziridinring, der eine geringere Polarität als das Nitron besitzt. Die Wellenleitermodenspektroskopie der nitronhaltigen Polymerfilme zeigte, dass die Schichtdicke weitgehend konstant bleibt, während die Brechungsindices stark abnehmen. Eine thermische Ringöffnung der polymergebundenen Oxaziridine zu den entsprechenden Nitronen scheint bei dieser Art von Polymeren nicht möglich zu sein.

In der vorliegenden Arbeit wurden verschiedene Topologien von Polymakromonomeren auf unterschiedlichen chemischen Routen synthetisiert und mit verschiedenen Methoden charakterisiert. Es wurden Polyalkylmakromonomere, Kern-Schale zylindrische Bürsten (Poly[styrol-block-alkylmakromonomere]), Polystyrolmakromonomere und Blockcopolymere aus zylindrischer Polystyrolbürste und linearem t--BuMA--Knäuel synthetisiert. Die Synthese der Polyalkylmakromonomere und der Kern--Schale zylindrischen Bürsten wurde durch die freie radikalische Polymerisation von Makromonomeren erreicht. Die unterschiedlichen Eigenschaften der Polymakromonomere wurden mit verschiedenen Methoden (Lichtstreuung, Neutronenstreuung, DSC, AFM und NMR) untersucht. Die metalloceninitiierte Polymerisation von Polystyrolmakromonomeren führte zum ersten Mal zu Polymerisationsgraden der Hauptkette von mehr als 1000, so dass eine neue chemische Route zur Synthese von zylindrischen Bürsten entwickelt werden konnte. Die partiell lebende metalloceninitiierte Polymerisation erlaubt weiterhin zum ersten Mal die Synthese von Blockstrukturen, die einen zylindrischen Bürstenteil und einen linearen Knäuelteil (t--BuMA) aufweisen. Diese Blockcopolymere bilden nach Abspaltung der tert.--Butylgruppe und Neutralisation der freien Polymethacrylsäure mit Cäsiumhydroxid ein sehr großes Amphiphil, das in einem selektiven Lösungsmittel (z.~B. THF) eine mizellare überstruktur ausbildet. Der mizellare Charakter dieser überstrukturen wurde mit der hochauflösenden Transmissionselektronenmikroskopie/EDX bewiesen. Der mit der TEM beobachtete Durchmesser einer solchen Riesenmizelle erreicht Werte von bis zu 300 nm.
