Developmental and biophysical leaf characteristics that influence post-harvest shelf life in lettuce, an important leafy crop, have been examined. The traits were studied using 60 informative F-9 recombinant inbed lines (RILs) derived from a cross between cultivated lettuce (Lactuca sativa cv. Salinas) and wild lettuce (L. serriola acc. UC96US23). Quantitative trait loci (QTLs) for shelf life co-located most closely with those for leaf biophysical properties such as plasticity, elasticity, and breakstrength, suggesting that these are appropriate targets for molecular breeding for improved shelf life. Significant correlations were found between shelf life and leaf size, leaf weight, leaf chlorophyll content, leaf stomatal index, and epidermal cell number per leaf, indicating that these pre-harvest leaf development traits confer post-harvest properties. By studying the population in two contrasting environments in northern and southern Europe, the genotype by environment interaction effects of the QTLs relevant to leaf development and shelf life were assessed. In total, 107 QTLs, distributed on all nine linkage groups, were detected from the 29 traits. Only five QTLs were common in both environments. Several areas where many QTLs co-located (hotspots) on the genome were identified, with relatively little overlap between developmental hotspots and those relating to shelf life. However, QTLs for leaf biophysical properties (breakstrength, plasticity, and elasticity) and cell area correlated well with shelf life, confirming that the ideal ideotype lettuce should have small cells with strong cell walls. The identification of QTLs for leaf development, strength, and longevity will lead to a better understanding of processability at a genetic and cellular level, and allow the improvement of salad leaf quality through marker-assisted breeding.

Lettuce is an important leafy vegetable, consumed across the world, containing bitter sesquiterpenoid lactone (SL) compounds that may negatively affect consumer acceptance and consumption. We assessed liking of samples with differing absolute abundance and different ratios of bitter:sweet compounds by analysing recombinant inbred lines (RILs) from an interspecific lettuce mapping population derived from a cross between a wild (L. serriola acc. UC96US23) and domesticated lettuce, (L. sativa, cv. Salinas). We found that the ratio of bitter:sweet compounds was a key determinant of bitterness perception and liking. We were able to demonstrate that SLs such as 8-deoxylactucin-15-sulphate contribute most strongly to bitterness perception, whilst 15-p-hydroxylphenylacetyllactucin-8-sulphate does not contribute to bitter taste. Glucose was the sugar most highly correlated with sweetness perception. There is a genetic basis to the biochemical composition of lettuce. This information will be useful in lettuce breeding programmes in order to produce leaves with more favourable taste profiles.

Enterohaemorrhagic Escherichia coli (EHEC) are a group of food and contact-borne pathogens responsible for haemorrhagic colitis. The bacteria can be transmitted by contaminated meat, but importantly, also by plants. The bacteria can use plants as an alternative host, where they associate with both the leaves and the roots. Colonisation in the rhizosphere of plants is thought to be the main habitat for colonisation. Four different plant species, commonly associated with EHEC outbreaks, were infected with EHEC O157:H7 isolates Sakai and TUV 93-0 over ten days to assess the colonisation potential of the bacteria in both the phyllosphere and rhizosphere of plants. The rhizosphere was found to sustain a higher population level of bacteria over time in comparison to the phyllosphere, yet both strains were unable to utilize root exudates for growth. Global gene expression changes of EHEC O157:H7 strain Sakai were measured in response to plant extracts such as leaf lysates, root exudates and leaf cell wall polysaccharides from spinach cultivar Amazon and lettuce cultivar Salinas. Microarrays analysis showed a significant change in expression of 17 % of genes on exposure to leaf lysates of spinach. A more specific response was seen to spinach leaf cell wall polysaccharides with only a 1.5 % change. In contrast, when exposed to lettuce leaf cell wall polysaccharides a higher change of 4.8 % was seen. Genes that were differentially expressed belonged to multiple functional groups, including metabolism, indicating the utilization of plant-specific polysaccharides. Several areas of further investigation have been determined from this project, including the importance of culturing bacterial strains at a relevant temperature, the proposed lack of the type III secretion system in plant colonization by EHEC O157:H7 and the utilization of plant components for growth and persistence in the plant environment.

We introduce a Sherrington-Kirkpatrick spin-glass model with the addition of elastic degrees of freedom. The problem is formulated in terms of an effective four-spin Hamiltonian in the pressure ensemble, which can be treated by the replica method. In the replica-symmetric approximation, we analyze the pressure-temperature phase diagram, and obtain expressions for the critical boundaries between the disordered and the ordered (spin-glass and ferromagnetic) phases. The second-order para-ferromagnetic border ends at a tricritical point, beyond which the transition becomes discontinuous. We use these results to make contact with the temperature-concentration phase diagrams of mixtures of hydrogen-bonded crystals.

We performed Monte Carlo simulations to investigate the steady-state critical behavior of a one-dimensional contact process with an aperiodic distribution of rates of transition. As in the presence of randomness, spatial fluctuations can lead to changes of critical behavior. For sufficiently weak fluctuations, we give numerical evidence to show that there is no departure from the universal critical behavior of the underlying uniform model. For strong spatial fluctuations, the analysis of the data indicates a change of critical universality class.

This paper considers an extension to the skew-normal model through the inclusion of an additional parameter which can lead to both uni- and bi-modal distributions. The paper presents various basic properties of this family of distributions and provides a stochastic representation which is useful for obtaining theoretical properties and to simulate from the distribution. Moreover, the singularity of the Fisher information matrix is investigated and maximum likelihood estimation for a random sample with no covariates is considered. The main motivation is thus to avoid using mixtures in fitting bimodal data as these are well known to be complicated to deal with, particularly because of identifiability problems. This work presents a Bayesian semiparametric approach for dealing with regression models where the covariate is measured with error. Given that (1) the error normality assumption is very restrictive, and (2) assuming a specific elliptical distribution for errors (Student-t for example), may be somewhat presumptuous; there is need for more flexible methods, in terms of assuming only symmetry of errors (admitting unknown kurtosis). In this sense, the main advantage of this extended Bayesian approach is the possibility of considering generalizations of the elliptical family of models by using Dirichlet process priors in dependent and independent situations. Conditional posterior distributions are implemented, allowing the use of Markov Chain Monte Carlo (MCMC), to generate the posterior distributions. An interesting result shown is that the Dirichlet process prior is not updated in the case of the dependent elliptical model. Furthermore, an analysis of a real data set is reported to illustrate the usefulness of our approach, in dealing with outliers. Finally, semiparametric proposed models and parametric normal model are compared, graphically with the posterior distribution density of the coefficients. (C) 2009 Elsevier Inc. All rights reserved.

In this article, we study some results related to a specific class of distributions, called skew-curved-symmetric family of distributions that depends on a parameter controlling the skewness and kurtosis at the same time. Special elements of this family which are studied include symmetric and well-known asymmetric distributions. General results are given for the score function and the observed information matrix. It is shown that the observed information matrix is always singular for some special cases. We illustrate the flexibility of this class of distributions with an application to a real dataset on characteristics of Australian athletes.

Tropomyosin (Tm) is a dimeric coiled-coil protein that polymerizes through head-to-tail interactions. These polymers bind along actin filaments and play an important role in the regulation of muscle contraction. Analysis of its primary structure shows that Tm is rich in acidic residues, which are clustered along the molecule and may from sites for divalent cation binding. In a previous study, we showed that the Mg(2+)-induced increase in stability of the C-terminal half of Tin is sensitive to imitations near the C-terminus. In the present report, we study the interaction between Mg(2+) and full-length Tin and smaller fragments corresponding to the last 65 and 26 Tin residues. Although the smaller Tin peptide (Tm(259-284(W269))) is flexible and to large extent unstructured, the larger Tm(220-284(W269)) fragments forms a coiled coil in solution whose stability increases significantly in the presence of Mg(2+). NMR analysis shows thin Mg(2+) induces chemical shift perturbations in both Tm(220-284(W269)) and Tm(259-284(W269)) in the vicinity of His276, in which are located several negatively charged residues. Skeletal alpha-tropomyosin (Tm) is a dimeric coiled-coil protein that forms linear assemblies under low ionic strength conditions in vitro through head-to-tail interactions. A previously published NMR structure of the Tin head-to-tail complex revealed that it is formed by the insertion of the N-terminal coiled-coil of one molecule into a cleft formed by the separation of the helices at the C-terminus of a second molecule. To evaluate the contribution of charged residues to complex stability, we employed single and double-mutant Tm fragments in which specific charged residues were changed to alanine in head-to-tail binding assays, and the effects of the mutations were analyzed by thermodynamic double-mutant cycles and protein-protein docking. The results show that residues K5, K7, and D280 are essential to the stability of the complex. Though D2, K6, D275, and H276 are exposed to the solvent and do not participate in intermolecular contacts in the NMR structure, they may contribute to head-to-tail complex stability by modulating the stability of the helices at the Tm termini.

The PilZ protein was originally identified as necessary for type IV pilus (T4P) biogenesis. Since then, a large and diverse family of bacterial PilZ homology domains have been identified, some of which have been implicated in signaling pathways that control important processes, including motility, virulence and biofilm formation. Furthermore, many PilZ homology domains, though not PilZ itself, have been shown to bind the important bacterial second messenger bis(3`-> 5`)cyclic diGMP (c-diGMP). The crystal structures of the PilZ orthologs from Xanthomonas axonopodis pv Citri (PilZ(XAC1133), this work) and from Xanthomonas campestris pv campestris (XC1028) present significant structural differences to other PilZ homologs that explain its failure to bind c-diGMP. NMR analysis of PilZ(XAC1133) shows that these structural differences are maintained in solution. In spite of their emerging importance in bacterial signaling, the means by which NZ proteins regulate specific processes is not clear. In this study, we show that PilZ(XAC1133) binds to PilB, an ATPase required for TV polymerization, and to the EAL domain of FiMX(XAC2398), which regulates TV biogenesis and localization in other bacterial species. These interactions were confirmed in NMR, two-hybrid and far-Western blot assays and are the first interactions observed between any PilZ domain and a target protein. While we were unable to detect phosphodiesterase activity for FimXX(AC2398) in vitro, we show that it binds c-diGMP both in the presence and in the absence of PilZ(XAC1133). Site-directed mutagenesis studies for conserved and exposed residues suggest that PilZ(XAC1133) interactions with FimX(XAC2398) and PilB(XAC3239) are mediated through a hydrophobic surface and an unstructured C-terminal extension conserved only in PilZ orthologs. The FimX-PilZ-PilB interactions involve a full set of ""degenerate"" GGDEF, EAL and PilZ domains and provide the first evidence of the means by which PilZ orthologs and FimX interact directly with the TP4 machinery. A Fonnação Alagamar corresponde ao estágio transicional na evolução tectono-sedimentar da bacia Potiguar, representada inicialmente por um ambiente tlúvio-deltaico passando a lagunar restrito com indícios de intluência marinha. Os resultados obtidos nas análises por DRX, a partir das frações> 2 11me < 211m, demonstram variações na composição mineralógica ao longo das perfurações analisadas. A principal característica é a forte contribuição de minerais detríticos na fração >2 I-lme a predominância de argilominerais na fração <2 I-lmrepresentados por esmectita, ilita, caulinita, clorita, sepiolita e interestratificados de ilitalesmectita (IIE) e cloritalesmectita (CIE). As carapaças de ostracodes identificadas correspondem a espécies nãomarinhas de três famílias mais comuns no Cretáceo: Cyprididae, Limnocytheridae e Darwinulidae. A avaliação estatística da composição ontogenética das espécies fósseis, tomou-se útil neste trabalho para estimar os níveis de energia do paleoambiente deposicional. As associações de argilominerais e ostracodes, caracterizadas ao longo dos cilindros de sondagens estudados na Fonnação Alagamar, sugerem a variação dos ambientes deposicionais a partir de um ambiente lacustre e de clima árido. Ainda a passagem por uma fase transgressiva com o aumento da salinidade, condições de fundo redutoras e margens subaquosas, ocasionam exposições subaéreas intennitentes, culminandono topo com um período mais úmido e condições de águas salinas O rico conteúdo em esmectita assemelha os depósitos da Fonnação Alagamar aos depósitos neoaptianos, os quais estão associados a condições climáticas com tendência à aridez. Sugere, ainda, morfologia pouco acidentada e drenagem reduzida ao redor de um paleolago restrito, o que favorece o desenvolvimento de solos do tipo vertissolo, fonte principal da esmectita da área. Os teores baixos de ilita e caulinita são, em grande parte, do resultado de erosão reduzida, atribuído ao relevo pouco acidentado da área. A associação dos argilominerais ilita, esmectita e interestratificados não ordenados indica condições rasas de soterramento e baixas temperaturas de diagênese nas amostras da base da perfuração RN6, alto de Macau. Ainda, a coloração apresentada pelas carapaças de ostracodes, entre o amarelo-laranja muito claro (2,5 Y 8/4) e amarelo amarronado(10 YR 6/6), sugerem níveis imaturos quanto à maturação da matéria orgânica. Na perfuração RN9, próxima a falha de Ubarana, condições de maturação da matéria orgânica são indicadas pelas cores cinza muito preto (5 Y 3/1) a preto (5 Y 2,5/1) apresentadas pelas carapaças de Candona sp.1, originalmente de cor branca (5 YR S/1 aiO YR 8/1). As intensidades e a fonna dos picos da ilita e da caulinita, a ausência de interestratificados e de argilominerais expansivos indicam condições de diagênese mais intensas que na perfuraçãoRN6. Associações de argilominerais identificadas em análises de difração de raios-X e mudanças na coloração das carapaças de ostracodes sugerem a utilização destes constituintes como indicadores ténnicos (geotennômetros) da maturação da matéria orgânica presente em rochas geradoras de hidrocarbonetos. Diante de mercados globais, interligados em tempo real, as empresas se vêem diante do desafio de se tornarem mais flexíveis, centrarem-se em suas principais competências e, sobretudo, terem a capacidade de dar respostas rápidas e adequadas às mudanças, em um ambiente onde a tempestividade da resposta pode ditar o sucesso ou o fracasso de uma empresa. Nesse contexto, a auditoria interna, em razão do seu posicionamento estratégico e permeabilidade por toda a organização, pode contribuir mais efetivamente para a melhoria dos processos produtivos e de gestão das organizações, através de uma atuação diferenciada de seus auditores. Por sua vez, a aprendizagem organizacional, entendida como um processo de apropriação e geração de novos conhecimentos, voltado para a construção ou desenvolvimento de novas competências, pode potencializar o valor agregado dos trabalhos de auditoria. Dentro dessa perspectiva, a resolução coletiva de problemas, baseada nas constatações de auditoria, pode tornar-se um veículo eficaz para alavancar o processo de aprendizagem por toda a organização. O presente trabalho procura avaliar os impactos da difusão dos princípios e conceitos de aprendizagem organizacional nas práticas de trabalho dos auditores internos, através de uma estratégia focada na resolução coletiva de problemas, tomando como objeto de pesquisa o processo desenvolvido na área de auditoria interna do Banco do Brasil S.A. A pesquisa, qualitativa na sua essência e orientada pela Grounded Theory (Glaser & Strauss, 1967; Strauss & Corbin, 1990), abrangeu o acompanhamento de 30 eventos desenvolvidos no âmbito da unidade de pesquisa, desembocando em 34 fontes primárias, derivadas de relatos, relatórios, transcrição de vídeos e entrevistas, memorandos, etc. Como resultante, o trabalho apresenta e avalia os principais impactos verificados nas práticas de auditoria interna, bem como nas ações dos indivíduos participantes do processo. O estudo abrange, também, a identificação das principais barreiras para aprendizagem e das competências básicas requeridas para condução de um processo de aprendizagem organizacional dessa natureza.
