We analyzed the codon usage bias of eight open reading frames (ORFs) across up to 79 human papillomavirus (HPV) genotypes from three distinct phylogenetic groups. All eight ORFs across HPV genotypes show a strong codon usage bias, amongst degenerately encoded amino acids, toward 18 codons mainly with T at the 3rd position. For all 18 degenerately encoded amino acids, codon preferences amongst human and animal PV ORFs are significantly different from those averaged across mammalian genes. Across the HPV types, the L2 ORFs show the highest codon usage bias (73.2 +/- 1.6% and the E4 ORFs the lowest (51.1 +/- 0.5%), reflecting as similar bias in codon 3rd position A + T content (L2: 76.1 +/- 4.2%; E4: 58.6 +/- 4.5%). The E4 ORF, uniquely amongst the HPV ORFs, is G + C rich, while the other ORFs are A + T rich. Codon usage bias correlates positively with A + T content at the codon 3rd position in the E2, E6, L1 and L2 ORFs, but negatively in the E4 ORFs. A general conservation of preferred codon usage across human and non-human PV genotypes whether they originate from a same supergroup or not, together with observed difference between the preferred codon usage for HPV ORFs and for genes of the cells they infect, suggests that specific codon usage bias and A + T content variation may somehow increase the replicational fitness of HPVs in mammalian epithelial cells, and have practical implications for gene therapy of HPV infection. In seeking to explain why oral estrogen inhibits the GH-IGF-I axis, a recent study has unearthed a new way that steroid hormones can influence growth hormone action. This involves suppressors of cytokine signalling (SOCS), which offer a new level of understanding in signal control and crosstalk.

Glucocorticoids are pivotal for adipose tissue development. Rodent studies suggest that corticosteroid-binding globulin (CBG) modulates glucocorticoid action in adipose tissue. In humans, both genetic CBG deficiency and suppressed CBG concentrations in hyperinsulinemic states are associated with obesity. We hypothesized that CBG deficiency in humans modulates the response of human preadipocytes to glucocorticoids, predisposing them to obesity. We compared normal preadipocytes with subcultured preadipocytes from an individual with the first ever described complete deficiency of CBG due to a homozygous null mutation. CBG-negative preadipocytes proliferated more rapidly and showed greater peroxisome proliferator-activated receptor-gamma-mediated differentiation than normal preadipocytes. CBG was not expressed in normal human preadipocytes. Glucocorticoid receptor number and binding characteristics and 11beta-hydroxysteroid dehydrogenase activity were similar for CBG-negative and normal preadipocytes. We propose that the increased proliferation and enhanced differentiation of CBG-negative preadipocytes may promote adipose tissue deposition and explain the obesity seen in individuals with genetic CBG deficiency. Furthermore, these observations may be relevant to obesity occurring with suppressed CBG concentrations associated with hyperinsulinemia.

A procura por madeiras oriundas de reflorestamentos destinadas à serraria é uma realidade já há muitos anos, principalmente aquelas das espécies do gênero Eucalyptus. Visando buscar novas informações importantes para esse mercado, este trabalho objetivou determinar algumas propriedades mecânicas da madeira de um híbrido clonal de Eucalyptus urophylla x Eucalyptus grandis de duas idades e provenientes de talhadia simples e de reforma. Os resultados indicaram que a madeira desse híbrido apresenta boas características tecnológicas, destacando-se a segunda tora (a partir de 3 m) com as melhores propriedades de flexão estática (Módulo de Elasticidade - MOE e Módulo de Ruptura - MOR) e Compressão Axial das fibras. As árvores de maior idade (166 meses) e que sofreram dois desbastes apresentaram as melhores propriedades de flexão estática e compressão axial.

Smart grids are envisaged as infrastructures able to accommodate all centralized and distributed energy resources (DER), including intensive use of renewable and distributed generation (DG), storage, demand response (DR), and also electric vehicles (EV), from which plug-in vehicles, i.e. gridable vehicles, are especially relevant. Moreover, smart grids must accommodate a large number of diverse types or players in the context of a competitive business environment. Smart grids should also provide the required means to efficiently manage all these resources what is especially important in order to make the better possible use of renewable based power generation, namely to minimize wind curtailment. An integrated approach, considering all the available energy resources, including demand response and storage, is crucial to attain these goals. This paper proposes a methodology for energy resource management that considers several Virtual Power Players (VPPs) managing a network with high penetration of distributed generation, demand response, storage units and network reconfiguration. The resources are controlled through a flexible SCADA (Supervisory Control And Data Acquisition) system that can be accessed by the evolved entities (VPPs) under contracted use conditions. A case study evidences the advantages of the proposed methodology to support a Virtual Power Player (VPP) managing the energy resources that it can access in an incident situation.

Serious games are games where the entertainment aspect is not the most relevant motivation or objective. TimeMesh is an online, multi-language, multiplayer, collaborative and social game platform for sharing and acquiring knowledge of the history of European regions. As such it is a serious game with educational characteristics. This article evaluates the use of TimeMesh with students of 13 and 14 years-old. It shows that this game is already a significant learning tool about European citizenship.

This study focused on the occurrence of several EDCs including bisphenol A, estrone (E1), the 17b-estradiol (E2) and 17a- thinylestradiol (EE2) in fourteen rivers of Portugal. Samples analysis revealed a widespread contamination of BPA especially in Ave, Cávado, Douro, Ferro, Sousa and Vizela Rivers. Achieving 98.4 ng/L for the highest concentration. The estrogens achieved above the method quantification limit (MQL) were E1 in Águeda River and E2 in Ave, Lima and Tâmega Rivers. The maximum concentration detected for E1 was 26.9 ng/L. EE2 was detected only below MQL.

Bisphenol A (BPA) is an endocrine disrupting chemical (EDC) whose migration from food packaging is recognized worldwide. However, the real overall food contamination and related consequences are yet largely unknown. Among humans, children’s exposure to BPA has been emphasized because of the immaturity of their biological systems. The main aim of this study was to assess the reproductive impact of BPA leached from commercially available plastic containers used or related to child nutrition, performing ecotoxicological tests using the biomonitoring species Daphnia magna. Acute and chronic tests, as well as single and multigenerational tests were done. Migration of BPA from several baby bottles and other plastic containers evaluated by GC-MS indicated that a broader range of foodstuff may be contaminated when packed in plastics. Ecotoxicological test results performed using defined concentrations of BPA were in agreement with literature, although a precocious maturity of daphnids was detected at 3.0 mg/L. Curiously, an increased reproductive output (neonates per female) was observed when daphnids were bred in the polycarbonate (PC) containers (145.1±4.3 % to 264.7±3.8 %), both in single as in multigenerational tests, in comparison with the negative control group (100.3±1.6 %). A strong correlated dose-dependent ecotoxicological effect was observed, providing evidence that BPA leached from plastic food packaging materials act as functional estrogen in vivo at very low concentrations. In contrast, neonate production by daphnids cultured in polypropylene and non-PC bottles was slightly but not significantly enhanced (92.5±2.0 % to 118.8±1.8 %). Multigenerational tests also revealed magnification of the adverse effects, not only on fecundity but also on mortality, which represents a worrying trend for organisms that are chronically exposed to xenoestrogens for many generations. Two plausible explanations for the observed results could be given: a non-monotonic dose–response relationship or a mixture toxicity effect.

Tamoxifen is a selective estrogen receptor modulator that is used as an adjuvant and/or chemotherapeutic agent for the treatment of all stages of hormone-dependent breast cancer. Currently there is a deep interest in the study of tamoxifen biotransformation and identification of metabolites since they can significantly contribute to the overall pharmacological or adverse effects of the drug. Accordingly, the study of the electrochemical behavior of tamoxifen in aqueous solution is reported. To clarify the occurring oxidative process and to assess the influence of the functional groups on the oxidation mechanism, the voltammetric assessment was extended to the study of tamoxifen’s analogues (E)-tamoxifen and dihydrotamoxifen, and to its main phase I oxidative metabolite, N-desmethyl tamoxifen. The data found shows that the oxidative processes occurring in tamoxifen are essentially related with the two chemical moieties present in the molecule: the substituted aromatic nucleus and the tertiary amine group. Moreover, the results obtained suggest that the ethylenic linkage is not critical for tamoxifen’s oxidation although it could play an important role in the course of the oxidation process. These results could contribute to highlight some remaining questions regarding tamoxifen’s metabolic behavior and to the development of new analytical strategies, based on electrochemical approaches.

Desde os anos 90 que se suspeita que um vasto leque de compostos, que estão presentes no ambiente aquático é susceptível de causar disrupção endócrina, podendo provocar efeitos adversos na nível do sistema reprodutor de vários organismos, de entre os quais os surfactantes não-iónicos alquifenois polietoxilatos(APEO), estrogénios, compostos organoclorados, entre outros. Estes, têm sido amplamente utilizados nos últimos 50 anos numa vasta diversidade de aplicações domésticas e comerciais. Durante o tratamento de águas residuais urbanas e industriais, os APEO são degradados sucessivamente até formas menos biodegradáveis, como por exemplo o NP (nonilfenol) e o OP (octilfenol), acabando por ser descarregados no ambiente aquático. A informação disponível, relativamente ao efeito das elevadas descargas nos meios receptores e da sua potencial toxicidade é ainda muito limitada, nomeadamente em sistemas aquáticos. Por outro lado, a informação sobre remoção de EDC em estações de tratamento de águas residuais (ETAR) é igualmente muito escassa, circunstância que impede a realização de estimativas de balanços materiais, indispensáveis à previsão dos correspondentes impactes nos meios hídricos, pelo que, este estudo teve como principal objectivo, a avaliação do potencial estrogénico de um efluente de uma ETAR com tratamento terciário. O presente estudo foi realizado na ETAR de Chelas, em Lisboa (Portugal). O efluente da ETAR é caracterizado por não ser só, efluente doméstico mas também ser constituído por quantidades significativas de efluente industrial. A escolha desta ETAR para realizar este estudo recaiu também na sua linha de tratamento, isto é, a ETAR possui tratamento terciário com remoção de azoto e desinfecção final do efluente através de U.V. De forma a detectar e quantificar o potencial estrogénico de uma água seleccionaramse compostos específicos, isto é alguns EDC “alvo”, nomeadamente: (i) nonilfenol e octilfenol (NP e OP); (ii) bisfenol A (BPA) e (iii) 17 β-estradiol (E2). Foram utilizadas três técnicas diferentes para identificar e quantificar os EDC seleccionados ELISA, LC-MS-MS e HPLC. As técnicas foram igualmente comparadas. Em todos os casos, isto é, independentemente da técnica analítica usada ficou demonstrado que os EDC seleccionados estavam presentes no efluente da ETAR,variando as suas concentrações de composto para composto e também ao longo da linha de tratamento da ETAR, e que ainda são descarregadas, no Estuário do Tejo,níveis de concentrações que poderão causar efeitos fisiológicos na vida animal. Contudo, devido ao elevado nível de diluição existente no Estuário do Tejo (caudais muito grandes) os efeitos nos organismos poderão não ser relevantes e,ou imediatos. Os níveis mais elevados de APE (NP e OP) foram detectados no afluente à ETAR e os menores no efluente da ETAR. O valor mais elevado de E2 foi registado após a decantação primária. A variação de E2 ao longo da linha de tratamento foi pouco significativa, facto este que poderá estar relacionado também com a “cross-reactivity”, bem como a própria actividade microbiológica existente numa água residual. Os maiores valores para o BPA foram obtidos nas lamas primárias (acima dos limites de detecção), tendo sido registada uma redução significativa deste composto ao longo da linha de tratamento, o que poderá indicar que a ETAR de Chelas remove eficientemente este composto.

The development of scaffolds that combine the delivery of drugs with the physical support provided by electrospun fibres holds great potential in the field of nerve regeneration. Here it is proposed the incorporation of ibuprofen, a well-known non-steroidal anti-inflammatory drug, in electrospun fibres of the statistical copolymer poly(trimethylene carbonate-co-ε-caprolactone) [P(TMC-CL)] to serve as a drug delivery system to enhance axonal regeneration in the context of a spinal cord lesion, by limiting the inflammatory response. P(TMC-CL) fibres were electrospun from mixtures of dichloromethane (DCM) and dimethylformamide (DMF). The solvent mixture applied influenced fibre morphology, as well as mean fibre diameter, which decreased as the DMF content in solution increased. Ibuprofen-loaded fibres were prepared from P(TMC-CL) solutions containing 5% ibuprofen (w/w of polymer). Increasing drug content to 10% led to jet instability, resulting in the formation of a less homogeneous fibrous mesh. Under the optimized conditions, drug-loading efficiency was above 80%. Confocal Raman mapping showed no preferential distribution of ibuprofen in P(TMC-CL) fibres. Under physiological conditions ibuprofen was released in 24h. The release process being diffusion-dependent for fibres prepared from DCM solutions, in contrast to fibres prepared from DCM-DMF mixtures where burst release occurred. The biological activity of the drug released was demonstrated using human-derived macrophages. The release of prostaglandin E2 to the cell culture medium was reduced when cells were incubated with ibuprofen-loaded P(TMC-CL) fibres, confirming the biological significance of the drug delivery strategy presented. Overall, this study constitutes an important contribution to the design of a P(TMC-CL)-based nerve conduit with anti-inflammatory properties.
