Awareness of antibiotics in wastewaters and aquatic ecosystems is growing as investigations into alternate pollutants increase and analytical techniques for detecting these chemicals improve. The presence of three antibiotics (ciproffoxacin, norfloxacin and cephalexin) was evaluated in both sewage effluent and environmental waters downstream from a sewage discharge. Bacteria cultured from the sewage bioreactor and receiving waters were tested for resistance against six antibiotics (ciprofloxacin, tetracycline, ampicillin, trimethoprim, erythromycin and trimethoprim/sulphamethoxazole) and effects of short term exposure (24h) to antibiotics on bacterial denitrification rates were examined. Antibiotics were detected entering the sewage treatment plant with varying levels of removal during the treatment process. Antibiotics were also detected in effluent entering receiving waters and detectable 500m from the source. Among the bacteria cultured from the sewage bioreactor, resistance was displayed against all six antibiotics tested and bacteria cultured from receiving waters were resistant against two of the antibiotics tested. Rates of denitrification were observed to decrease in response to some antibiotics and not to others, though this was only observed at concentrations exceeding those likely to be found in the environment. Findings from this preliminary research have indicated that antibiotics are entering our aquatic systems and pose a potential threat to ecosystem function and potentially human health. Fungi are ubiquitous organisms in nature and can be found in association with healthy eyes. The incidence of actual fungal infection of the eye, however, is relatively low compared with that attributable to viruses and bacteria. Nevertheless, fungal infection of the eye is increasing especially in immuno-compromised patients and a wide variety of fungal infections have now been described worldwide with species of Fusarium, Aspergillus, Candida, and dematiaceous fungi predominating. At present there are a limited number of compounds available to control ocular mycoses while resistance to anti-fungal agents has been growing in recent years, especially to azoles. Several mechanisms of resistance have been identified including modification of sterol synthesis pathways by the fungus, modification of enzymes to reduce the binding of azoles to fungal components and increased efficiency of removal of the azole within fungal cells. Although resistance to amphotericin-B has been reported, it continues to be the most important treatment for life-threatening conditions and more severe ophthalmic infections. Natamycin is often first choice for filamentous fungal keratitis and topical amphotericin-B for Candida keratitis. Continued monitoring of the behaviour of ocular fungi will be essential in future together with the development of new anti-fungal agents.

The advancement of nanotechnology in the synthesis and characterisation of nanoparticles (NP's) has played an important role in the development of new technologies for various applications of nano-scale materials that have unique properties. The scientific development in the last decades in the field of nanotechnology has sought ceaselessly, the discovery of new materials for the most diverse applications, such as biomedical areas, chemical, optical, mechanical and textiles. The high bactericidal efficiency of metallic nanoparticles (Au and Ag), among other metals is well known, due to its ability to act in the DNA of fungi, viruses and bacteria, interrupting the process of cellular respiration, making them important means of study, in addition to its ability to protect UVA and UVB. The present work has as its main objective the implementation of an innovative method in the impregnation of nanoparticles of gold in textile substrate, functionalized with chitosan, by a dyeing process by exhaustion, with the control of temperature, time and velocity, thus obtaining microbial characteristics and UV protection. The exhausted substrates with colloidal solutions of NPAu's presented the colours, lilac and red (soybean knits) due to their surface plasmon peak around 520-540 nm. The NPAu's were synthesized chemically, using sodium citrate as a reducing agent and stabilizer. The material was previously cationised with chitosan, a natural polyelectrolyte, with the purpose of functionalising it to enhance the adsorption of colloid, at concentrations of 5, 7, 10 and 20 % of the bonding agent on the weight of the material (OWM). It was also observed, through an experimental design 23 , with 3 central points, which was the best process of exhaustion of the substrates, using the following factors: Time (min.), temperature (OC) and concentration of the colloid (%), having as a response to variable K/S (ABSORBÂNCIA/ Kubelka-Munk) of the fibres. Furthermore, it was evidenced as the best response, the following parameters: concentration 100%, temperature 70 ºC and time 30 minutes. The substrate with NPAu was characterised by XRD; thermal analysis using TGA; microstructural study using SEM/EDS and STEM, thus showing the NP on the surface of the substrate confirming the presence of the metal. The substrates showed higher washing fastness, antibacterial properties and UV radiation protection.

Antarctic meiofauna is still strongly understud- ied, and so is its trophic position in the food web. Primary producers, such as phytoplankton, and bacteria may repre- sent important food sources for shallow water metazoans, and the role of meiobenthos in the benthic-pelagic coupling represents an important brick for food web understanding. In a laboratory, feeding experiment 13C-labeled freeze- dried diatoms (Thalassiosira weissflogii) and bacteria were added to retrieved cores from Potter Cove (15-m depth, November 2007) in order to investigate the uptake of 3 main meiofauna taxa: nematodes, copepods and cumaceans. In the surface sediment layers, nematodes showed no real difference in uptake of both food sources. This outcome was supported by the natural delta 13C values and the community genus composition. In the first centimeter layer, the dominant genus was Daptonema which is known to be opportunistic, feeding on both bacteria and diatoms. Copepods and cumaceans on the other hand appeared to feed more on diatoms than on bacteria. This may point at a better adaptation to input of primary production from the water column. On the other hand, the overall carbon uptake of the given food sources was quite low for all taxa, indicating that likely other food sources might be of relevance for these meiobenthic organisms. Further studies are needed in order to better quantify the carbon requirements of these organisms.

Molecular biological methods were used to investigate the microbial diversity and community structure in intertidal sandy sediments near the island of Sylt (Wadden Sea) at a site which was characterized for transport and mineralization rates in de Beer et al., (2005, hdl:10013/epic.21375). The sampling was performed during low tide in the middle of the flat, approximately 40 m in the offshore direction from the high water line on October 6, 1999, March 7, 2000, and July 5, 2000. Two parallel cores were collected from each season for molecular analyses. Within 2 h after sampling the sediment cores were sub-sampled and fixed in formaldehyde for FISH analysis. The cells were hybridized, stained with 4',6'-diamidino-2-phenylindole (DAPI) and microscopically counted as described previously [55]. Details of probes and formamide concentrations which were used are shown in further details. Counts are reported as means calculated from 10-15 randomly chosen microscopic fields corresponding to 700-1000 DAPI-stained cells. Values were corrected for the signals counted with the probe NON338. Fluorescence in situ hybridization (FISH)with group-specific rRNA-targeted oligonucleotide probes were used to characterize the microbial community structure over depth (0-12 cm) and seasons (March, July, October). We found high abundances of bacteria with total cell numbers up to 3×109 cells ml-1 and a clear seasonal variation, with higher values in July and October versus March. The microbial community was dominated by members of the Planctomycetes, the Cytophaga/Flavobacterium group, Gammaproteobacteria, and bacteria of the Desulfosarcina/Desulfococcus group. The high abundance (1.5×10**7 - 1.8×10**8 cells/ml accounting for 3-19% of all cells) of presumably aerobic heterotrophic polymer-degrading planctomycetes is in line with the high permeability, deep oxygen penetration, and the high rates of aerobic mineralization of algal biomass measured in the sandy sediments by de Beer et al., (2005, hdl:10013/epic.21375). The high and stable abundance of members of the Desulfosarcina/Desulfococcus group, both over depth and season, suggests that these bacteria may play a more important role than previously assumed based on low sulfate reduction rates in parallel cores de Beer et al., (2005).

1. ATP in deep-sea sediments can be determined after it is adsorbed on a mixture of the sediment and calcium carbonate by measuring the luminescence of the reaction of the mixture and luciferin-luciferase. 2. ATP contents of the toplayer of northeastern Atlantic sediments (Josephine Bank and northern Canary Basin) decrease with increasing depths of 252, 408, 1445, 1769, 2149, 4897, 5510m: 0.96, 0.61, 0.13, 0.10, 0.21, 0.05, 0.07 µg ATP/ml wet sediment. The decreasing values are in accordance with the decrease of macrobenthos and meiobenthos biomass in the deep-sea. 3. The ATP content of deep-sea nematodes is about 1 ? of their wet weight. 4. At the two deepest stations, less than 50% of the ATP measured in the sediment is represented by nematodes, copepods, other "hard" meiofauna groups and bacteria.

An increasing number of empirical studies are challenging the central fundamentals on which the classical soil food web model is built. This model assumes that bacteria consume labile substrates twice as fast as fungi, and that mycorrhizal fungi do not decompose organic matter. Here, we build on emerging evidence that points to significant consumption of labile C by fungi, and to the ability of ectomycorrhizal fungi to decompose organic matter, to show that labile C constitutes a major and presently underrated source of C for the soil food web. We use a simple model describing the dynamics of a recalcitrant and a labile C pool and their consumption by fungi and bacteria to show that fungal and bacterial populations can coexist in a stable state with large inputs into the labile C pool and a high fungal use of labile C. We propose a new conceptual model for the bottom trophic level of the soil food web, with organic C consisting of a continuous pool rather than two or three distinct pools, and saprotrophic fungi using substantial amounts of labile C. Incorporation of these concepts will increase our understanding of soil food web dynamics and functioning under changing conditions.

When most people think of food safety they think of food poisoning and bacteria. They also, one hopes, generally follow the well-understood public advice on bacterial risks and store their food properly and cook it thoroughly. But what about chemical risks in food? Do many consumers ask the question “if drug residues are in my food, does cooking make it safe?” Or do they assume that following the good advice on bacterial risks also affords some protection against the health risks of chemical contaminants? In this short report we highlight some difficulties in assessing the stability of veterinary drug residues during cooking and summarise our cooking studies on anthelmintics, nitroimidazoles and nitrofuran residues in various foods. safefood Knowledge Networks http://safefood.ning.com/

[EN] Different types of fungi and bacteria have been isolated from hatched and non-hatched as well as failed and non-failed eggs in natural sea turtles nests (Marco et al. 2006, Phillott and Parmenter, 2001, Phillott et al. 2001). Microbiota infections are common in artificial incubation activities and they seem to have an important negative impact on embryo development (Phillott, 2002). However, no clear evidences of their pathogenic effects have been described. The aim of this study was to investigate whether fungi and bacteria represent pathogenic agents to sea turtle eggs, and to assess whether there exists a specific period during incubation in which eggs are more susceptible to microorganisms.

The presence of microorganisms in ophthalmic instruments and surfaces can lead to the exposure of patients to several infections. However, there is no information regarding fungal and bacteria contamination in optical shops. This study aims to characterize fungi and bacteria contamination in air and surfaces from 10 optical shops covering also ophthalmic instruments. Air samples were collected through an impaction method onto malt extract agar (MEA) supplemented with chloramphenicol (0.05%) used for fungi and Tryptic Soy Agar (TSA) supplemented with nystatin (0.2%) used for bacteria. Outdoor samples were also performed to be used as reference. Surface and equipment’s swab samples were also collected side-by-side. All the collected samples were incubated at 27ºC for 5 to 7 days (fungi) or at 30º for 7 days (bacteria). Regarding fungal distribution, thirteen different species/genera were found in the air, being the most common Alternaria sp. (62.0%). Eight different species/genera were identified in the surfaces, ranging from 2 to 5x104 CFU/m2, being the most common A. versicolor complex and Penicillium sp. (40.0%). The trial frames were the most contaminated equipment, since 50.0% of the collected samples were with countless colonies. The airborne bacterial population indicated higher concentrations in the contactology office (average: 133 CFU/m3) than in the client’s waiting rooms (average: 126 CFU/m3). The surface samples indicated bacterial concentrations ranging from 2x104 to 1x106 CFU/m2, pointing out the automatic refractometer as the surface with higher bacterial load.

Sampling the total air concentration of particulate matter (PM) only provides a basic estimate of exposure that normally not allows correlating with the observed health effects. Therefore is of great importance to recognize the particles size distribution and, particularly, the exposure to fine particles (≤ 2.5 μm). This particles dimension corresponds to the respirable fraction, the one that can implicate local and systemic effects due to particle deposition and clearance from the lungs and transport within the organism. This study intended to describe occupational exposure to PM2.5 in three units related with swine production and consumption, namely: feed production, swine production and swine slaughterhouse. A size-selective particle measuring in five to six workplaces of each unit was performed. Measurements of PM were done using a portable direct-reading hand-held equipment (Lighthouse, model 3016 IAQ). Data showed slaughterhouse unit with higher values, with values ranging from 0.030 to 0.142 mg/m3 (0.073 + 0.043), being the cutting room the workplace with higher values. In feed production unit, values were between 0.026 and 0.033 mg/m3 (0.028 + 0.003) with the warehouse of pharmacy products as the workplace with higher values. Finally, in swine unit values ranged from 0.006 to 0.048 mg/m3 (0.023 + 0.017) with the batteries area presenting the higher values. PM can be rich in fungi and bacteria and their metabolites, such as endotoxins and mycotoxins. Previous publications already showed high contamination in these occupational settings and particles can have an important role in exposure since can easily act as carrier of these agents. Data acquired allow not only a better prediction of particle penetration into respiratory regions of the respiratory tract, but also a better estimation of PM health effects. Moreover, data permit to identify the workplaces where investment should be made to prevent and reduce exposure.

Com a realização deste trabalho, pretendeu-se efetuar uma seleção de culturas mistas em reatores semi-descontínuos (SBR) com capacidade de acumulação de polihidroxialcanoatos (PHA). Para a seleção de culturas foram utilizados inóculos provenientes de diferentes Estações de Tratamento de Águas Residuais (ETAR) e ácidos orgânicos voláteis (AOV) como fonte de carbono. Foram testadas diferentes condições como a proveniência do inóculo, as cargas orgânicas aplicadas e a seleção de culturas utilizando soro de queijo. Verificaram-se elevadas remoções da CQO (acima de 90%) em grande parte dos ensaios realizados, apresentando uma acumulação de PHA por parte de algumas espécies de bactérias presentes. Ocorreu o aparecimento de microrganismos filamentosos com capacidade de acumulação de PHA em alguns ensaios, levando a serem testadas como culturas acumuladoras de PHA. A estabilidade das culturas mistas não foi atingida, mesmo havendo ensaios com 80 dias de operação. Efetuaram-se ensaios de acumulação de PHA em reatores descontínuos, utilizando as culturas selecionadas anteriormente em reatores SBR, com AOV provenientes da acidificação anaeróbia de diferentes resíduos (Fração Orgânica dos Resíduos Sólidos Urbanos - FORSU e Soro de Queijo). Verificou-se uma melhor acumulação por parte das culturas selecionadas com soro de queijo, na qual a quantidade de polímero acumulado triplicou.

Com o aumento na captura de pescado e da poluição do meio ambiente, esta-se à margem de exceder a estimativa do limite da sustentabilidade, e obviamente isto faz com se utilize os recursos marítimos com mais inteligência e precaução. Aplicando tecnologia enzimática ou química é possível recuperar as proteínas do processamento do pescado, produzindo hidrolisados e isolados protéicos. Uma grande quantidade de proteínas insolúveis está disponível em escamas, peles e ossos, subprodutos do processamento do pescado, que podem ser solubilizadas através de fungos e bactérias. Utilizando isolados protéicos é possível obter biopolímeros, estes têm chamado a atenção nos últimos anos, pois são biodegradáveis, não-tóxicos e geralmente biocompatíveis. Os hidrogéis protéicos são polímeros que podem absorver uma quantidade de água a partir de 10 até centenas de vezes o seu peso seco. O objetivo deste trabalho foi desenvolver um hidrogel protéico, com propriedades superabsorventes, a partir das proteínas solúveis e insolúveis da corvina (Micropogonias furnieri). Para a produção dos hidrolisados a partir das proteínas solúveis foi utilizado processo enzimático (Alcalase e Flavourzyme) e químico (solubilização ácida e alcalina). Nos processos de solubilização das proteínas insolúveis foram utilizados microrganismos (bactérias e fungos). Tanto as bactérias como os fungos avaliados apresentaram capacidade de solubilizar as proteínas insolúveis presentes nos resíduos (escamas, ossos, cartilagens e outros). A bactéria que atingiu a maior atividade proteolítica foi a Bacillus velesensis (47,56 U mL-1) e o fungo foi o Penicillium sp. (E20) (31,20 U mL-1). Para a produção dos hidrogéis, foram utilizados isolados protéicos provenientes de solubilização ácida ou alcalina, produzidos a partir de resíduos da industrialização de pescado, modificados quimicamente com dianidrido etilenodiamino tetraacético (EDTAD) e adicionados de agente de ligação cruzada (glutaraldeído). Algumas proteínas modificadas ainda foram submetidas a tratamento com etanol. Foram realizadas análise estrutural das proteínas modificadas e estudo da capacidade de retenção de água dos hidrogéis assim obtidos. Os hidrogéis produzidos apresentaram alta capacidade de retenção de água. A máxima absorção de água foi alcançada pelo hidrogel ácido sem o tratamento com etanol foi de 103,25 gágua/ggel seco, enquanto que a mesma amostra tratada com etanol alcançou 216,05 gágua/ggel seco. Os hidrogéis produzidos podem ser utilizados em diversas indústrias, tais como, farmacêutica, alimentícia, médica, agroindústria, entre outras, que necessitem de hidrogéis com alta capacidade de retenção de água.
