1. 1. Routine oxygen consumption and blood glucose were determined from freshwater fishes, Prochilodus scrofa and Cyprinus carpio, exposed at high temperatures for 1 hr. 2. 2. Prochilodus scrofa had a significantly higher rate of oxygen consumption at 30°C than at 25°C, and carp higher at 25°C than at 30°C. 3. 3. Blood glucose was significantly higher for Cyprinus carpio than for Prochilodus scrofa at 25 and 30°C; however, after exposure to these temperatures for 1 hr blood glucose did not change significantly for both species. 4. 4. The results suggest that these interspecific variations may be linked to the differences between native and foreign fishes and their way of life. © 1985.

In order to access the effects of cultivation in soil, sand or a commercial substrate on the productivity of Bonus #2 cultivars, 'Don Carlos' and 'Hy Mark', two experiments were conducted in the greenhouse at FCAV-UNESP, Jaboticabal- SP, Brazil, 21° 15' 22 S, 48° 18'58 W, and altitude of 595 m, from November 1999 to April 2000 (summer), and from July to November 2000 (Winter). For cultivation in soil, chemical nutrients were added, and plants were irrigated with drip irrigation. Fertigation with recirculation of the nutrient solution was used on sand; slabs were used on commercial substrate with the fertigation with non circulating nutrient solution. Bonus #2 cultivar yielded the highest production of marketable fruit, but were later in production, while Hy Mark cultivar had early production but a lower number of fruit per plant. The winter planting yielded higher production of marketable fruit while summer plantings yielded lower number of fruit per plant, but with higher average weigh. The combination between cultivation systems and cultivars yielded higher fruit production in winter.

Background: Hematophagous insects digest large amounts of host hemoglobin and release heme inside their guts. In Rhodnius prolixus, hemoglobin-derived heme is detoxified by biomineralization, forming hemozoin (Hz). Recently, the involvement of the R. prolixus perimicrovillar membranes in Hz formation was demonstrated. Methodology/Principal Findings: Hz formation activity of an α-glucosidase was investigated. Hz formation was inhibited by specific α-glucosidase inhibitors. Moreover, Hz formation was sensitive to inhibition by Diethypyrocarbonate, suggesting a critical role of histidine residues in enzyme activity. Additionally, a polyclonal antibody raised against a phytophagous insect α-glucosidase was able to inhibit Hz formation. The α-glucosidase inhibitors have had no effects when used 10 h after the start of reaction, suggesting that α-glucosidase should act in the nucleation step of Hz formation. Hz formation was seen to be dependent on the substrate-binding site of enzyme, in a way that maltose, an enzyme substrate, blocks such activity. dsRNA, constructed using the sequence of α-glucosidase gene, was injected into R. prolixus females' hemocoel. Gene silencing was accomplished by reduction of both α-glucosidase and Hz formation activities. Insects were fed on plasma or hemin-enriched plasma and gene expression and activity of α-glucosidase were higher in the plasma plus hemin-fed insects. The deduced amino acid sequence of α-glucosidase shows a high similarity to the insect α-glucosidases, with critical histidine and aspartic residues conserved among the enzymes. Conclusions/Significance: Herein the Hz formation is shown to be associated to an a-glucosidase, the biochemical marker from Hemipteran perimicrovillar membranes. Usually, these enzymes catalyze the hydrolysis of glycosidic bond. The results strongly suggest that α-glucosidase is responsible for Hz nucleation in the R. prolixus midgut, indicating that the plasticity of this enzyme may play an important role in conferring fitness to hemipteran hematophagy, for instance. © 2009 Mury et al.

The climates of the central and southern regions of São Paulo state in Brazil favor pathogens such as Puccinia psidii Winter, which causes a common and severe disease in Eucalyptus plantations under 2 years old. We studied genetic parameters including genotype by environment interaction (G × E) of resistance to P. psidii rust in Eucalyptus grandis at nine sites in São Paulo State. Open-pollinated progeny from ten 'provenances' were established in a randomized complete block design; at individual sites there were from 134 to 160 progenies, from four to eight blocks, and five to six trees per plot. Significant provenance and progeny(provenance) differences were detected, as was G × E involving progeny(provenance). However, the G × E involved little if any rank changes, indicating that selection can be done efficiently at a single site, if the disease level is sufficient. The estimated coefficient of genetic variation among the progeny within provenances CVg was high and variable among the sites (ranging from 11 % to 36. 7 %), demonstrating different expression of genetic variability among the sites. The estimated heritability at the individual-tree level h2 and within a plot hw 2 ranged from low to intermediate (ranging from 0. 04 to 0. 46) and was high at the progeny-mean level hf 2 (ranging from 0. 30 to 0. 86). Our study shows good prospects of controlling this disease by selection among and within progenies in a single site. © 2012 Springer-Verlag Berlin Heidelberg.

Tillering dynamics during regrowth in the spring (September to December 2009) on signal grass (Brachiaria decumbens cv. Basilisk) pastures deferred in the beginning of April 2009 is evaluated. Four pasture heights were evaluated at the beginning of deferment (10, 20, 30 and 40 cm). The experimental design consisted of completely randomized blocks with two replications. Rise in pasture height at the beginning of deferment decreased the appearance rate, increased the mortality rate and reduced the survival rate, the balance between appearance and mortality and the stability index of the tiller population at the beginning of the spring. Pastures deferred at initial height of 30 and 40 cm presented relatively stable tiller appearance rate between the beginning and end of spring. However, these pastures presented greater mortality rate, lower survival rate, lower balance between appearance and mortality and lower stability index of the tiller population at the beginning of spring when compared to rates at the end of spring. Pastures managed with shorter initial heights (10 and 20 cm) provided more tissue renewal in the subsequent growth season after their utilization in the winter.

Buffaloes and bovines are polyestrous and seasonal or annual livestock, respectively, that show reduced fertility during heat stress. To investigate whether reduced fertility is related to oocyte competence in both species, immature oocytes from buffalo and bovine heifers were collected during winter and summer and subjected to molecular analyses. In each season, heifers of both species had their follicular wave emergence synchronized with a standard protocol (Ferreira et al., 2011). Before being subjected to ovum pick up (OPU), cutaneous (CT; degrees C) and rectal (RT; degrees C) temperatures and respiratory rate (RR; breaths/min) were measured. Oocytes' RNA was extracted to evaluate the expression of target genes related to mtDNA replication/transcription (PPARGC1A, TFAM and MT-CO1), apoptosis (BAX and BCL2) and HS (HSP90AA1 and HSPA1AB). ACTB, HIST1H2AG and GAPDH were initially chosen as housekeeping genes. In buffaloes, CT (35.0 +/- 0.4 vs 23.8 +/- 0.5), RT (38.7 +/- 0.1 vs 38.0 +/- 0) and RR (21.3 +/- 1.2 vs 15.4 +/- 1.1) were higher during summer than winter. However, in bovine heifers, RT (38.7 +/- 0.1 vs 38.6 +/- 0.1) and RR (44.8 +/- 1.5 vs 40.6 +/- 1.5) were similar in both seasons, while CT (31.6 +/- 0.3 vs 30.2 +/- 0.3) was increased during summer. Reduced expression of ACTB, HIST1H2AG and GAPDH was evidenced during summer, disqualifying them as housekeeping genes. Similarly, the expression of all target genes was reduced during summer in oocytes of both species. In summary, physiological responses to heat stress seem to be more intense in buffalo than bovine heifers. However, in both species, negative effects of heat stress upon oocyte quality occur at the molecular level and affects genes related to several biological functions.

Stink bugs are among the major pests of soybean [Glycine max (L.) Merrill] worldwide. Piezodorus guildinii [Westwood] (Hemiptera: Pentatomidae) is one of the predominant pest species, causing more severe damage in many regions than other stink bugs. Its attack reduces yield and quality of the beans. Plant resistance is a valuable strategy in integrated pest management that can reduce insect populations below economic injury level. Here, we report the resistance of 17 soybean entries to P. guildinii. PI 229358, PI 274454, L1-1-01, IAC 19, PI 171451, PI 227687, IAC 100, IAC 78-2318, PI 274453, and IAC 74-2832 caused high nymphal mortality (greater than 90 %), indicating the expression of antibiosis. IAC 100, IAC 74-2832, PI 274453, and IAC 24 also increased the length of the nymphal stage of P. guildinii, showing the same mechanism of resistance. Our findings may be useful for breeding programs that focus on the resistance of soybeans to insects.

The aim of the present study was to evaluate the effects of type of norgestomet auricular implant (new - N or previously used during 5 days - U), season of the year (summer - S and winter - W), and parity (12 heifers - H and 23 cows - C) on synchronization of follicular wave emergence in buffaloes. For this purpose, 35 buffaloes were examined daily by ultrasonography until follicular wave emergence was detected. Data were analysed by ANOVA, using PROC GLIMMIX. No interactions were observed in none variables. Time of follicular wave emergence and number of follicles at emergence were not affected by type of implant or season of the year. Parity also did not influence the number of follicles at emergence. However, follicular wave emergence occurred later in heifers than in cows. In conclusion, the previous use of a norgestomet auricular implant independent of the season of the year does not affect the time or the number of follicles at follicular wave emergence in buffaloes. Nevertheless, although heifers and cows had a similar number of follicles at emergence, the time of follicular wave emergence occurs earlier in cows than in heifers.
