The study's objective was to realize comparisons among different acepromazine, chlorpromazine and methotrimeprazine doses, evaluate parametric changes, test analgesia using press and term algimetry, and evaluate bispectral condition. 90 mongrel dogs were used, male and female, adult, weighting 10 to 15 Kg as a rule, distributed in 9 groups with 10 animals each. At first, second and third groups acepromazine was used at 0,1; 0,05 e 0,025 mg/Kg, respectively. At forth, fifth and sixth groups, chlorpromazine was used at 1,0; 0,5 and 0,25 mg/Kg, respectively. At seventh, eighth and ninth groups, methotrimeprazine at 1,0; 0,5 and 0,25 mg/Kg was used, respectively. All drugs were administered intravenously. objects of study: heart rate (HR), non invasive blood pressure (SAP, MAP, DAP), respiratory rate (f), capnography (ETCO(2)), pulse oxymetry (SatO(2)), mouth and rectal temperature, bispectral index (BIS), electromyography (EMG%), press and term algimetry. Somatic analgesia was evaluated by animal's response to nociceptives stimulus. We concluded that chlorpromazine had more bypotension. Dogs showed higher hypnosis level at: chlorpromazine group, with evident myorelaxation. All groups showed analgesia to thermic and mechanic stimulus. Acepromazine group showed high duration to both pain stimuli. Bispectral index was shorten at chlorpromazine group at 1,0 mg/kg doses, showing higher hypnosis index, and acepromazine was the less depressing considering the bispectral index.

Several findings suggest that catecholaminergic neurones in the caudal ventrolateral medulla (CVLM) contribute to body fluid homeostasis and cardiovascular regulation. The present study sought to determine the effects of lesions of these neurones on the cardiovascular responses induced by changes in circulating volume. All experiments were performed in male Wistar rats (320-360 g). Medullary catecholaminergic neurones were lesioned by microinjection of anti-dopamine beta-hydroxylase-saporin (6.3 ng in 60 nl; SAP rats, n = 14) into the CVLM, whereas sham rats received microinjections of free saporin (1.3 ng in 60 nl, n = 15). Two weeks later, rats were anaesthetized (urethane, 1.2 g kg(-1), I.V..), instrumented for measurement of mean arterial pressure (MAP), renal blood flow (RBF) and renal vascular conductance (RVC), and infused with hypertonic saline (HS; 3 M NaCl, 0.18 ml (100 g body weight)(-1), I.V.) or an isotonic solution (volume expansion, VE; 4% Ficoll, 1% of body weight, I.V.). In sham rats, HS induced sustained increases in RBF and RVC (155 +/- 7 and 145 +/- 6% of baseline, at 20 min after HS). In SAP rats, RBF responses to HS were blunted (125 +/- 6%) and RVC increases were abolished (108 +/- 5%) 20 min after HS. Isotonic solution increased RBF and RVC in sham rats (149 +/- 10 and 145 +/- 12% of baseline, respectively, at 20 min). These responses were reduced in SAP rats (131 +/- 6 and 126 +/- 5%, respectively, at 20 min). Pressor responses to HS were larger in SAP rats than in sham rats (17 +/- 5 versus 9 +/- 2 mmHg, at 20 min), whereas during VE these responses were similar in both groups (6 +/- 3 versus 4 +/- 6 mmHg, at 20 min). Immunohistochemical analysis indicates that microinjections of anti-D beta H-saporin produced extensive destruction within the A1/C1 cell groups in the CVLM. These results suggest that catecholaminergic neurones mediate the cardiovascular responses to VE or increases in plasma sodium levels.

The nucleus of the solitary tract (NTS) receives primary afferents involved in cardiovascular regulation. We investigated the role of NK1-receptor bearing neurons in the NTS on cardiovascular reflexes in awake rats fitted with chronic venous and arterial cannulae. These neurons were lesioned selectively with saporin conjugated with substance P (SP-SAP, 2 mu M, bilateral injections of 20 nL in the subpostremal NTS, or 200 nL in both the subpostremal and the commissural NTS). Before, and 7 and 14 days after injection of SP-SAP, we measured changes in blood pressure and heart rate induced by i.v. injection of phenylephrine and nitroprusside (baroreceptor reflex), cyanide (arterial chemoreceptor reflex), and phenylbiguanide (Bezold-Jarisch reflex). The smaller injections with SP-SAP completely abolished NK1 receptor staining in the subpostremal NTS. The larger injections abolished NK1 receptor immunoreactivity in an area that extended from the commissural NTS to the rostral end of the subpostremal NTS. The lesions seemed to affect only a limited number of neurons, since neutral red stained sections did not show any obvious reduction in cell number. The smaller lesions reduced the gain of baroreflex bradycardia and the hypotension induced by phenylbiguanide. The larger lesions completely abolished the response to phenylbiguanide, blocked the baroreflex bradycardia induced by phenylephrine, severely blunted the baroreflex tachycardia, and blocked the bradycardia and reduced the hypertension induced by cyanide. Thus, these responses depend critically on NK1-receptor bearing neurons in the NTS. (c) 2006 Elsevier B.V. All rights reserved.

The ultrastructural features and the plastid changes caused by sample preparation were studied in sieve elements of Panicum maximum leaves. Samples of expanded leaves, taken near the ligule region, were fixed and processed by common light and transmission electron microscopy methods. In mature sieve-tube elements, the protoplast is electron-translucent and plastids are the most frequent organelles. Mitochondria and smooth endoplasmic reticulum segments are also visible and occupy a parietal position within the cell. The plastids are globular and show electron-dense proteinaceous inclusions in the stroma. The protein crystals are predominantly cuneate, but thin crystalloids and amorphous and/or filamentous proteins also occur. The presence of intact plastids plus others in different phases of plastid envelope rupture were interpreted as evidence that this rupture is a normal event in response to injury. This plastid envelope rupture is possibly activated by the release of pressure in the sieve-tube element. After plastid membrane vesiculation, the stroma and the protein crystals are dispersed within the sieve-element ground cytoplasm. The vesicles originating from the plastid envelope move to one cell pole, while protein crystalloids move to the opposite pole and agglomerate in the sieve-plate region. Our findings indicate that these protein crystalloids, which deposit in the sieve plate, may act in sieve-plate pores occlusion, preventing the release of phloem sap, similar to the role of P-protein in dicotyledons. Leaf-cutting ants belonging to the tribe Attini are major herbivores and important agriculture pests in the neotropics, these ants being thought to feed on the sap which exudes from the plant material which they cut and also on the mycelium of a symbiotic fungus that grows on plant material inside their nests in what is called 'the fungus garden'. However, we have found that the survival of Atta sexdens worker ants on leaves, on mycelium. of the ants' symbiotic fungus, Leucoagaricus gongylophorus, or on plant polysaccharides was the same as that of starved A. sexdens, while, conversely, significantly longer survival was achieved by ants fed on the fungus garden material or on some of the products (especially glucose) of the hydrolysis of plant polysaccharides. We found that the fungus garden contained glucose at a higher concentration than that found in leaves or fungal mycelium, and that this glucose was consumed by the ant to the extent that it was probably responsible for up to 50% of the nutritional needs of the workers. The fungus garden contained polysaccharide degrading enzymes (pectinase, amylase, xylanase and cellulase) in proportions similar to that observed in laboratory cultures of L. gongylophorus. It thus appears that A. sexdens workers obtain a significant part of their nutrients from plant polysaccharide hydrolysis products produced by the action of extracellular enzymes released by L. gongylophorus. In this paper we discuss the symbiotic nutrition strategy of A. sexdens workers and brood and the role played by plant polysaccharides in the nutrition of attine ants. (C) 2003 Elsevier B.V. Ltd. Spittlebugs are insects that suck sap from plants and regurgitate saliva containing toxic enzymes into the leaves. As a consequence, the conductive channels are blocked resulting in dry leaves, thus giving a burned aspect to the plantation. This work performed ultrastructural analyses of the salivary glands of the sugarcane spittlebug Mahanarva fimbriolata, since these organs produce the enzymes that are injected into the plants, thus being responsible for the economic losses in the production of sugarcane. Three kinds of secretory cells are found in principal gland, forming the lobules I-IV. The main differences among these cells relate to size, morphology and electron density of the secretory vesicles. The accessory glands contain different secretory vesicles to those in the principal gland. Muscular cells are found around the entire gland. The different secretory vesicles found in both principal and accessory glands indicate that the gland produces different substances or that the secretion in the interior of cells passes through a maturation process. Em lavouras de feijoeiro (Phaseolus vulgaris) da cultivar Carioca Comum, no município de Londrina, Estado do Paraná, foram encontradas plantas com sintomas de necrose da haste, mosaico clorótico leve e porte reduzido, semelhantes aos sintomas causados por infecção viral. Exames de microscopia eletrônica revelaram a presença de partículas isométricas. em testes de imunodifusão dupla em gel de ágar os extratos foliares de plantas infetadas reagiram positivamente com anti-soro específico para o Southern bean mosaic virus (SBMV). O vírus foi purificado e a massa molecular de sua proteína capsidial foi estimada em 30 kDa, valor esperado para proteínas do capsídeo de vírus do gênero Sobemovirus. A gama de hospedeiras do SBMV isolado no Paraná foi restrita ao feijoeiro e a algumas cultivares de soja (Glycine max). A separação de dois vírus isométricos comuns em infecções mistas no feijoeiro foi possível através da reação de imunidade ao SBMV apresentada por Crotalaria sp, Chenopodium quinoa e Mucuna deeringiana, e da reação de susceptibilidade dessas mesmas hospedeiras ao Bean rugose mosaic virus (BRMV).

Within a wide class of models, the CERN LEP2 lower limit of 95 GeV on the chargino mass implies gluinos are heavier than similar to 300 GeV. In this case electroweak (W) over tilde(1)(W) over tilde(1) production and (W) over tilde(1)(Z) over tilde(2) production are the dominant supersymmerry (SUSY) processes at the Fermilab Tevatron, and the extensively examined isolated trilepton signal From (W) over tilde(1)(Z) over tilde(2) production assumes an even greater importance. We update our previous calculations of the SUSY reach of luminosity upgrades of the Fermilab Tevatron in this channel incorporating (i) decay matrix elements in the computation of the momenta of leptons from chargino and neutralino decays, (ii) the trilepton background from W*Z* and W*gamma* production which, though neglected in previous analyses, turns out to be the dominant background, and finally, (iii) modified sets of cuts designed to reduce these new backgrounds and increase the range of model parameters for which the signal is observable. We show our improved projections for the reach for SUSY of both the Fermilab Main Injector and the proposed TeV33 upgrade. We also present opposite sign same flavor dilepton invariant mass distributions as well as the p(T) distributions of leptons in SUSY trilepton events, and comment upon how the inclusion of decay matrix elements impacts upon the Tevatron reach, as well as upon the extraction of neutralino masses.

Levantamentos realizados no estado de São Paulo indicaram a ocorrência isolada e em infecções mistas do Lettuce mosaic virus (LMV) e do Lettuce mottle virus (LeMoV) em plantas de alface (Lactuca sativa). O presente trabalho teve como objetivo estudar os efeitos da infecção isolada e mista entre o LMV (patótipos II e IV) e o LeMoV, em cultivares de alface suscetível (White Boston) e tolerante (Elisa - gene mol¹) ao LMV patótipo II. As plantas foram inoculadas via extrato vegetal tamponado com isolados de LMV-II, LMV-IV e LeMoV separadamente e em diferentes combinações, com intervalo de 24 h ou simultaneamente com os dois vírus. As plantas infetadas foram analisadas utilizando-se hospedeiras diferenciais para o LMV e o LeMoV, e no caso do LMV pelo teste sorológico de PTA-ELISA. Nas avaliações de peso fresco e seco, área foliar e teor de clorofila, observou-se que a cultivar White Boston foi a mais afetada por ambos os vírus. As infecções mistas e isoladas na cultivar Elisa causaram efeitos semelhantes, provavelmente devido a presença do gene mo1¹ de tolerância ao LMV-II. O isolado LMV-IV foi considerado o mais agressivo nestas cultivares quando comparado ao LMV-II e o LeMoV.

Estudaram-se possíveis alterações hemodinâmicas e intracranianas em cães submetidos à hemorragia aguda e anestesiados pelo isofluorano. Verificou-se também a influência do anestésico no mecanismo de auto-regulação cerebral. Utilizaram-se 20 cães adultos que foram induzidos à anestesia geral com isofluorano por máscara naso-oral a 3,5V% (volume %). Após a intubação orotraqueal, reajustou-se o vaporizador para 2,1V%. Induziu-se a hipovolemia retirando-se volume total de 35ml/kg de sangue. Avaliaram-se pressão intracraniana (PIC), temperaturas intracraniana (TIC) e corpórea (T), pressão de perfusão cerebral (PPC), pressões arteriais sistólica (PAS), diastólica (PAD) e média (PAM), freqüências cardíaca (FC) e respiratória (FR), índices cardíaco (IC) e sistólico (IS), pressão venosa central (PVC), pressão da artéria pulmonar (PAP), concentração de dióxido de carbono ao final da expiração (ETCO2) e saturação de oxihemoglobina (SpO2). Imediatamente após a hipovolemia, houve redução significativa da PIC, PPC, PAS, PAD, PAM, IC, IS e PAP. Após 10 minutos, houve aumento gradativo das médias, permanecendo neste patamar até o final do período experimental. Concluiu-se que a hemorragia aguda promoveu redução das variáveis hemodinâmicas, sendo possível verificar a ativação de mecanismos compensatórios. Além disso, houve redução da perfusão sangüínea e ativação do mecanismo de auto-regulação cerebral, conseqüentes à hipovolemia associada à anestesia com isofluorano.

Estudaram-se os efeitos do óxido nitroso (N2O) sobre a concentração alveolar mínima (CAM) do desfluorano. Trinta cães foram distribuídos em dois grupos: desfluorano (GD) e N2O e desfluorano (GDN). Os do GD receberam propofol (8,9±1,65mg/kg) para intubação orotraqueal e após, 11,5V% de desfluorano em 100% de O2. Após 30 minutos, os animais receberam estímulo elétrico e não havendo reação do animal, reduziu-se a concentração em 1,5V%. Repetiu-se o protocolo a cada 15 minutos, cessando-se os estímulos quando observada reação voluntária. Os GDN foram submetidos ao mesmo protocolo, substituindo-se o fluxo diluente por 30% O2 e 70% N2O. Mensuraram-se freqüências cardíaca (FC) e respiratória (FR), pressões arteriais sistólica, diastólica e média (PAS, PAD e PAM), concentração de dióxido de carbono ao final da expiração (ETCO2), saturação de oxihemoglobina (SpO2), temperatura corpórea (TC) e a CAM do desfluorano. Observou-se aumento da FC, ETCO2 e SpO2, e redução da FR e da TC concomitantemente à administração da maior dose de desfluorano, além de redução da CAM do desfluorano. As pressões arteriais diminuíram em M30 aumentando posteriormente. Concluiu-se que o N2O associado ao desfluorano reduz em 16% a CAM do anestésico volátil. Além disso, essa associação promoveu aumento da FC e depressão respiratória.

Seis cães adultos, de raças e sexos variados, com peso de 13,3±3,4kg (média±DP), foram utilizados no estudo. Os animais foram tranqüilizados com acepromazina (0,1mg/kg, IV) e, após 30 minutos, foram aleatoriamente submetidos à anestesia epidural com um dos seguintes tratamentos: lidocaína 2% 0,25ml/kg (controle); neostigmine 0,01mg/kg+lidocaína (NEO); metadona 0,3mg/kg+lidocaína (MET). Todos os animais foram submetidos aos três tratamentos com intervalo mínimo de uma semana. Foram mensuradas as freqüências cardíaca (FC) e respiratória (FR), a pressão arterial sistólica (PAS), o tempo para a perda do reflexo interdigital, a duração e a altura do bloqueio sensitivo, durante um período de 90 minutos. Não houve diferença significativa entre os tratamentos nos valores de FC, PAS e FR, bem como na duração do bloqueio sensitivo e no tempo para a perda do reflexo interdigital. No grupo MET, houve diminuição de FC dos 30 aos 90 minutos em relação ao valor basal. Bloqueio sensitivo mais cranial também foi observado em MET. A associação de neostigmine ou metadona não prolongou o período hábil de anestesia epidural produzido pela lidocaína em cães. A metadona, mas não o neostigmine, parece estender mais cranialmente o bloqueio epidural pela lidocaína.
