Oxidative stress is considered to be of major relevance for a variety of pathological processes. Thus, it is valuable to identify compounds, which might act as antioxidants, i.e. compounds that antagonize the deleterious action of reactive oxygen species (ROS) on biomolecules. The mode of action of these compounds could be either to scavenge ROS directly or to trigger protective mechanisms inside the cell, thereby resulting in improved defense against ROS. Sulforaphane (SF) (1-isothiocyanato-(4R)-(methylsulfinyl)butane) is a naturally occurring cancer chemopreventive agent found as a precursor glucosinolate in Cruciferous vegetables like broccoli. Although SF is not a direct-acting antioxidant, there is substantial evidence that SF acts indirectly to increase the antioxidant capacity of animal cells and their abilities to cope with oxidative stress. Induction of phase 2 enzymes is one means by which SF enhances the cellular antioxidant capacity. Enzymes induced by SF include Glutathione S-transferases (GST) and NAD[P]H:quinone oxidoreductase (NQO1) which can function as protectors against oxidative stress. To protect themselves from oxidative stress, cells are equipped with reducing buffer systems including the GSH and thioredoxin (Trx) reductase. GSH is an important tripeptide thiol which in addition to being the substrate for GSTs maintains the cellular oxidation– reduction balance and protects cells against free radical species. Aim of the first part of this thesis was to investigate the ability of SF to induce the expression and the activity of different phase 2 and antioxidant enzymes (such as GST, GR, GPx, NQO1, TR, SOD, CAT) in an in vitro model of rat cardiomyocytes, and also to define if SF treatment supprts cells in counteracting oxidative stress induced by H2O2 It is well known that acute exhaustive exercise causes significant reactive oxygen species generation that results in oxidative stress, which can induce negative effects on health and well being. In fact, increased oxidative stress and biomarkers (e.g., protein carbonyls, MDA, and 8- hydroxyguanosine) as well as muscle damage biomarkers (e.g. plasmatic Creatine cinase and Lactate dehydrogenase) have been observed after supramaximal sprint exercises, exhaustive longdistance cycling or running as well as resistance-type exercises, both in trained and untrained humans. Markers of oxidative stress also increase in rodents following exhaustive exercise. Moreover, antioxidant enzyme activities and expressions of antioxidant enzymes are known to increase in response to exhaustive exercise in both animal and human tissues. Aim of this project was to evaluate the effect of SF supplementation in counteracting oxidative stress induced by physical activity through its ability to induce phase 2, and antioxidant enzymes in rat muscle. The results show that SF is a nutraceutical compound able to induce the activity of different phase 2 and antioxidant enzymes in both cardiac muscle and skeletal muscle. Thanks to its actions SF is becoming a promising molecule able to prevent cardiovascular damages induced by oxidative stress and muscle damages induced by acute exhaustive exercise.

We observed 82 healthy subjects, from both sexes, aged between 19 and 77 years. All subjects performed two different tests: for being scientifically acknowledged, the first one was used as a reference and it was a stress test (CPX). During the entire test, heart rate and gas exchange were recorded continuously; the second, the actual object of this study, was a submaximal test (TOP). Only heart rate was recorded continuously. The main purpose was to determinate an index of physical fitness as result of TOP. CPX test allowed us to individuate anaerobic threshold. We used an incremental protocol of 10/20 Watt/min, different by age. For our TOP test we used an RHC400 UPRIGHT BIKE, by Air Machine. Each subject was monitored for heart frequency. After 2 minutes of resting period there was a first step: 3 minutes of pedalling at a constant rate of 60 RPM, (40 watts for elder subjects and 60 watts for the younger ones). Then, the subject was allowed to rest for a recovery phase of 5 minutes. Third and last step consisted of 3 minutes of pedalling again at 60 RPM but now set to 60 watts for elder subjects and 80 watts for the young subjects. Finally another five minutes of recovery. A good correlation was found between TOP and CPX results especially between punctua l heart rate reserve (HRR’) and anaerobic threshold parameters such as Watt, VO2, VCO2 . HRR’ was obtained by subtracting maximal heart rate during TOP from maximal theoretic heart rate (206,9-(0,67*age)). Data were analyzed through cluster analysis in order to obtain 3 homogeneous groups. The first group contains the least fit subjects (inactive, women, elderly). The other groups contain the “average fit” and the fittest subjects (active, men, younger). Concordance between test resulted in 83,23%. Afterwards, a linear combinations of the most relevant variables gave us a formula to classify people in the correct group. The most relevant result is that this submaximal test is able to discriminate subjects with different physical condition and to provide information (index) about physical fitness through HRR’. Compared to a traditional incremental stress test, the very low load of TOP, short duration and extended resting period, make this new method suitable to very different people. To better define the TOP index, it is necessary to enlarge our subject sample especially by diversifying the age range.

Quello che viene presentato costituisce un estratto del lavoro di tesi svolto presso il Dipartimento di Architettura e Pianificazione Territoriale della Facoltà di Ingegneria. La Convenzione Europea del Paesaggio (Firenze, 2000), sottoscritta da vari paesi dell'U.E. (tra cui l'Italia), obbliga ad una rivalutazione del concetto di paesaggio, che si svincola dalla classica definizione estetico-percettiva per abbracciare un significato ben più vasto: ogni paesaggio, inteso come mosaico di valori storici, culturali, sociali, naturali, diventa condizione fondamentale non solo per il benessere delle popolazioni che lo abitano, ma anche per uno sviluppo economico durevole nel tempo. In quest'ottica la pianificazione è tenuta oggi ad occuparsi in maniera integrata di tutti i paesaggi, da quelli eccellenti, fino a quelli ritenuti privi di valore e "degradati". Assumendo come area oggetto di studio la Regione Emilia-Romagna, la presente trattazione intende dapprima ripercorrere l'evoluzione del concetto di paesaggio e della sua gestione all'interno del quadro normativo nazionale e regionale; successivamente si approfondisce lo studio di aree compromesse e degradate, fornendo una base metodologica che consenta di "interpretare" un paesaggio, di individuarne le componenti fondamentali e le possibili alterazioni che queste subiscono, quindi definire indirizzi progettuali che consentano di gestire le trasformazioni antropiche sul territorio in maniera compatibile dal punto di vista paesistico ed ambientale.

The recent introduction of free form NC machining in the ophthalmic field involved a full review of the former product development process both from the design and the manufacturing viewpoint. Aim of the present work is to investigate and to set up innovative methods and tools supporting the product development, particularly for lenses characterized by free form geometry, as e.g. progressive lenses. In the design stage, the research addressed geometric modeling of complex lens shapes and relevant analysis tools for the optical-geometrical characterization of the produced models. In the manufacturing stage, the main interest was focused on the set-up of the fabrication process, particularly on the NC machining process for which an integration CADCAM software was developed for the generation and the simulation of the machining cycle. The methodologies and tools made available by the present work are currently used in the development of new complex geometry product typologies as, e.g. progressive lenses.

Recent statistics have demonstrated that two of the most important causes of failures of the UAVs (Uninhabited Aerial Vehicle) missions are related to the low level of decisional autonomy of vehicles and to the man machine interface. Therefore, a relevant issue is to design a display/controls architecture which allows the efficient interaction between the operator and the remote vehicle and to develop a level of automation which allows the vehicle the decision about change in mission. The research presented in this paper focuses on a modular man-machine interface simulator for the UAV control, which simulates UAV missions, developed to experiment solution to this problem. The main components of the simulator are an advanced interface and a block defined automation, which comprehend an algorithm that implements the level of automation of the system. The simulator has been designed and developed following a user-centred design approach in order to take into account the operator’s needs in the communication with the vehicle. The level of automation has been developed following the supervisory control theory which says that the human became a supervisor who sends high level commands, such as part of mission, target, constraints, in then-rule, while the vehicle receives, comprehends and translates such commands into detailed action such as routes or action on the control system. In order to allow the vehicle to calculate and recalculate the safe and efficient route, in term of distance, time and fuel a 3D planning algorithm has been developed. It is based on considering UASs representative of real world systems as objects moving in a virtual environment (terrain, obstacles, and no fly zones) which replicates the airspace. Original obstacle avoidance strategies have been conceived in order to generate mission planes which are consistent with flight rules and with the vehicle performance constraints. The interface is based on a touch screen, used to send high level commands to the vehicle, and a 3D Virtual Display which provides a stereoscopic and augmented visualization of the complex scenario in which the vehicle operates. Furthermore, it is provided with an audio feedback message generator. Simulation tests have been conducted with pilot trainers to evaluate the reliability of the algorithm and the effectiveness and efficiency of the interface in supporting the operator in the supervision of an UAV mission. Results have revealed that the planning algorithm calculate very efficient routes in few seconds, an adequate level of workload is required to command the vehicle and that the 3D based interface provides the operator with a good sense of presence and enhances his awareness of the mission scenario and of the vehicle under his control.

Questo lavoro di tesi nasce da un progetto di ricerca promosso nel 2001 dal Prof. Leonardo Seccia (Seconda Facoltà di Ingegneria, sede di Forlì, e C.I.R.A.M., Università di Bologna), dal Prof. Nicola Santopuoli (Facoltà di Architettura Valle Giulia, Sapienza Università di Roma), dal Prof. Ingo Muller e dal Dott. André Musolff (Technical University Berlin, Facultat III, Thermodynamics). Tale progetto ha avuto come obiettivo lo studio, la progettazione e la realizzazione di un dispositivo di ancoraggio in lega a memoria di forma per il restauro di affreschi e mosaici parietali, che presentino distacchi più o meno evidenti fra gli strati di intonaco di supporto, proponendosi come mezzo efficace per la salvaguardia strutturale di tali zone variamente ammalorate. In particolare, è stata programmata una serie di prove di laboratorio per caratterizzare in modo preciso il comportamento del materiale prescelto, al fine di realizzare un prototipo rispondente alle caratteristiche di progetto ed anche per implementare un modello numerico sufficientemente realistico. A questo proposito, è stato anche approfondito il problema della scelta del modello costitutivo più adeguato. Successivamente, i risultati ottenuti sono stati impiegati nella progettazione e realizzazione di nuovi dispositivi in lega a memoria di forma da impiegare nel campo dei beni culturali, fra cui sistemi reversibili per il ricongiungimento di parti fratturate e sistemi di movimentazione intelligenti sia per lastre di protezione di superfici affrescate, sia per finestre da inserire in contesti museali per il controllo del microclima.
