It is well known that melatonin participates in the regulation of many important physiological functions such as sleep-wakefulness cycle, motor coordination and neural plasticity, and cognition. However, as there are contradictory results regarding the melatonin production diurnal profile under alcohol consumption, the aim of this paper was to study the phenomenology and mechanisms of the putative modifications on the daily profile of melatonin production in rats submitted to chronic alcohol intake. The present results show that rats receiving 10% ethanol in drinking water for 35 days display an altered daily profile of melatonin production, with a phase delay and a reduction in the nocturnal peak. This can be partially explained by a loss of the daily rhythm and the 25% reduction in tryptophan hydroxylase activity and, mainly, by a phase delay in arylalkylamine N-acetyltransferase gene expression and a 70% reduction in its peak activity. Upstream in the melatonin synthesis pathway, the results showed that noradrenergic signaling is impaired as well, with a decrease in beta 1 and alpha 1 adrenergic receptors` mRNA contents and in vitro sustained loss of noradrenergic-stimulated melatonin production by glands from alcohol-treated rats. Together, these results confirm the alterations in the daily melatonin profile of alcoholic rats and suggest the possible mechanisms for the observed melatonin synthesis modification.

The pst operon of Escherichia coli is composed of five genes pstS, pstC, pstA, pstB and phoU, that encode a high-affinity phosphate transport system and a negative regulator of the PHO regulon. Transcription of pst is induced under phosphate shortage and is initiated at the promoter located upstream of the first gene of the operon, pstS. Here, we show by four different technical approaches the existence of additional internal promoters upstream of pstC, pstB and phoU. These promoters are not induced by Pi-limitation and do not possess PHO-box sequences. Plasmids carrying the pst internal genes partially complement chromosomal mutations in their corresponding genes, indicating that they are translated into functional proteins.

This work presents a numerical method suitable for the study of the development of internal boundary layers (IBL) and their characteristics for flows over various types of coastal cliffs. The IBL is an important meteorological occurrence for flows with surface roughness and topographical step changes. A two-dimensional flow program was used for this study. The governing equations were written using the vorticity-velocity formulation. The spatial derivatives were discretized by high-order compact finite differences schemes. The time integration was performed with a low storage fourth-order Runge-Kutta scheme. The coastal cliff (step) was specified through an immersed boundary method. The validation of the code was done by comparison of the results with experimental and observational data. The numerical simulations were carried out for different coastal cliff heights and inclinations. The results show that the predominant factors for the height of the IBL and its characteristics are the upstream velocity, and the height and form (inclination) of the coastal cliff. Caulobacter crescentus sigma(E) belongs to the ECF (extracytoplasmic function) subfamily of RNA polymerase sigma factors, whose members regulate gene expression in response to distinct environmental stresses. During physiological growth conditions, data indicate that sigma(E) is maintained in reduced levels due to the action of ChrR, a negative regulator of rpoE gene expression and function. However, once bacterial cells are exposed to cadmium, organic hydroperoxide, singlet oxygen or UV-A irradiation, transcription of rpoE is induced in a sigma(E)-dependent manner. Site-directed mutagenesis indicated that residue C188 in ChrR is critical for the cadmium response while residues H140 and H142 are required for the bacterial response to organic hydroperoxide, singlet oxygen and UV-A. Global transcriptional analysis showed that sigma(E) regulates genes involved in protecting cells against oxidative damages. A combination of transcriptional start site identification and promoter prediction revealed that some of these genes contain a putative sigma(E)-dependent motif in their upstream regions. Furthermore, deletion of rpoE and two sigma(E)-dependent genes (cfaS and hsp20) impairs Caulobacter survival when singlet oxygen is constantly generated in the cells.

The ability to appropriately interact with the environment is crucial to an organism’s survival. The establishment of functional sensory systems, such as the bristles and eyes in Drosophila, is a critical event during the development of the organism. The transcription factor D Pax2 is involved in the differentiation of the shaft and glial cells in the developing bristle (Kavaler et al., Dev, 126:2261-2272, 1999) and of the cone and primary pigment cells in the developing eye (Fu and Noll, Genes Dev, 11:389-405, 1997). How D-Pax2 contributes to distinct differentiative pathways in different cell types is not known. Recent work by Anna Czechowski and Katherine Harmon (personal communication) identified a mutation in the D-Pax2 gene that introduced a stop codon at the end of exon 9, effectively truncating the protein. This mutation affects bristle, but not eye, development. We thus suspected regions after exon 9 are required for D-Pax2 function only in the bristles and may also be associated with alternative splicing of the D Pax2 transcript. We plan to assess the role of the carboxy terminal region of the protein by establishing transgenic lines bearing rescue constructs of D-Pax2 with either the complete coding sequence or with deletions of specific exons. To date, we have generated the first rescue construct bearing the complete coding region of the gene driven by a 3 KB upstream regulatory region of D-Pax2 and are currently generating transgenic fly lines with this construct.

The phytohormones gibberellin (GA) and abscisic acid (ABA) regulate important developments events in germinating seeds. Specifically, GA induces the expression of hyrolase genes, like the α-amylase gene Amy32b, which mobilizes starch reserves to be used by the embryo, and ABA suppresses this induction. Recent advancements identified ABA and GA receptors and key components in the signaling pathways, however, the mechanism of crosstalk between the hormones remains largely unknown. To further elucidate the mechanism of ABA suppression of GA-induced genes, we focused on the transcription factor TaABF1, a member of the ABA response element binding factor family. TaABF1 has been shown to physically interact with the SnRK2 kinase PKABA1 and overexpression of TaABF1 or PKABA1 can suppress Amy32b. We carried out particle bombardment experiments to investigate how TaABF1 suppresses Amy32b and how TaABF1 is activated by ABA. The role of TaABF1 in ABA-mediated suppression of Amy32b is more complicated than hypothesized. Unlike PKABA1, overexpression of TaABF1 did not cause a decrease of GAMyb expression and in fact resulted in an increase of GAMyb expression. When TaABF1 and GAMyb were simultaneously overexpressed in aleurone, the GAMyb induction of Amy32b was unaffected, indicating that the target of TaABF1 action must be upstream of GAMyb. Furthermore, TaABF1 and ABA demonstrated an additive effect on the suppression of Amy32b. Based on our findings, we propose a model in which PKABA1 activates two separate targets, one being TaABF1 which then modifies an unknown target upstream of GAMyb and the other being an unknown transcription factor that suppresses GAMyb transcription.

In the UK, urban river basins are particularly vulnerable to flash floods due to short and intense rainfall. This paper presents potential flood resilience approaches for the highly urbanised Wortley Beck river basin, south west of the Leeds city centre. The reach of Wortley Beck is approximately 6km long with contributing catchment area of 30km2 that drain into the River Aire. Lower Wortley has experienced regular flooding over the last few years from a range of sources, including Wortley Beck and surface and ground water, that affects properties both upstream and downstream of Farnley Lake as well as Wortley Ring Road. This has serious implications for society, the environment and economy activity in the City of Leeds. The first stage of the study involves systematically incorporating Wortley Beck’s land scape features on an Arc-GIS platform to identify existing green features in the region. This process also enables the exploration of potential blue green features: green spaces, green roofs, water retention ponds and swales at appropriate locations and connect them with existing green corridors to maximize their productivity. The next stage is involved in developing a detailed 2D urban flood inundation model for the Wortley Beck region using the CityCat model. CityCat is capable to model the effects of permeable/impermeable ground surfaces and buildings/roofs to generate flood depth and velocity maps at 1m caused by design storm events. The final stage of the study is involved in simulation of range of rainfall and flood event scenarios through CityCat model with different blue green features. Installation of other hard engineering individual property protection measures through water butts and flood walls are also incorporated in the CityCat model. This enables an integrated sustainable flood resilience strategy for this region.

Scour around hydraulic structures is a critical problem in hydraulic engineering. Under prediction of scour depth may lead to costly failures of the structure, while over prediction might result in unnecessary costs. Unfortunately, up-to-date empirical scour prediction formulas are based on laboratory experiments that are not always able to reproduce field conditions due to complicated geometry of rivers and temporal and spatial scales of a physical model. However, computational fluid dynamics (CFD) tools can perform using real field dimensions and operating conditions to predict sediment scour around hydraulic structures. In Korea, after completing the Four Major Rivers Restoration Project, several new weirs have been built across Han, Nakdong, Geum and Yeongsan Rivers. Consequently, sediment deposition and bed erosion around such structures have became a major issue in these four rivers. In this study, an application of an open source CFD software package, the TELEMAC-MASCARET, to simulate sediment transport and bed morphology around Gangjeong weir, which is the largest multipurpose weir built on Nakdong River. A real bathymetry of the river and a geometry of the weir have been implemented into the numerical model. The numerical simulation is carried out with a real hydrograph at the upstream boundary. The bedmorphology obtained from the numerical results has been validated against field observation data, and a maximum of simulated scour depth is compared with the results obtained by empirical formulas of Hoffmans. Agreement between numerical computations, observed data and empirical formulas is judged to be satisfactory on all major comparisons. The outcome of this study does not only point out the locations where deposition and erosion might take place depending on the weir gate operation, but also analyzes the mechanism of formation and evolution of scour holes after the weir gates.

Due to the increase in water demand and hydropower energy, it is getting more important to operate hydraulic structures in an efficient manner while sustaining multiple demands. Especially, companies, governmental agencies, consultant offices require effective, practical integrated tools and decision support frameworks to operate reservoirs, cascades of run-of-river plants and related elements such as canals by merging hydrological and reservoir simulation/optimization models with various numerical weather predictions, radar and satellite data. The model performance is highly related with the streamflow forecast, related uncertainty and its consideration in the decision making. While deterministic weather predictions and its corresponding streamflow forecasts directly restrict the manager to single deterministic trajectories, probabilistic forecasts can be a key solution by including uncertainty in flow forecast scenarios for dam operation. The objective of this study is to compare deterministic and probabilistic streamflow forecasts on an earlier developed basin/reservoir model for short term reservoir management. The study is applied to the Yuvacık Reservoir and its upstream basin which is the main water supply of Kocaeli City located in the northwestern part of Turkey. The reservoir represents a typical example by its limited capacity, downstream channel restrictions and high snowmelt potential. Mesoscale Model 5 and Ensemble Prediction System data are used as a main input and the flow forecasts are done for 2012 year using HEC-HMS. Hydrometeorological rule-based reservoir simulation model is accomplished with HEC-ResSim and integrated with forecasts. Since EPS based hydrological model produce a large number of equal probable scenarios, it will indicate how uncertainty spreads in the future. Thus, it will provide risk ranges in terms of spillway discharges and reservoir level for operator when it is compared with deterministic approach. The framework is fully data driven, applicable, useful to the profession and the knowledge can be transferred to other similar reservoir systems.

This paper describes the formulation of a Multi-objective Pipe Smoothing Genetic Algorithm (MOPSGA) and its application to the least cost water distribution network design problem. Evolutionary Algorithms have been widely utilised for the optimisation of both theoretical and real-world non-linear optimisation problems, including water system design and maintenance problems. In this work we present a pipe smoothing based approach to the creation and mutation of chromosomes which utilises engineering expertise with the view to increasing the performance of the algorithm whilst promoting engineering feasibility within the population of solutions. MOPSGA is based upon the standard Non-dominated Sorting Genetic Algorithm-II (NSGA-II) and incorporates a modified population initialiser and mutation operator which directly targets elements of a network with the aim to increase network smoothness (in terms of progression from one diameter to the next) using network element awareness and an elementary heuristic. The pipe smoothing heuristic used in this algorithm is based upon a fundamental principle employed by water system engineers when designing water distribution pipe networks where the diameter of any pipe is never greater than the sum of the diameters of the pipes directly upstream resulting in the transition from large to small diameters from source to the extremities of the network. MOPSGA is assessed on a number of water distribution network benchmarks from the literature including some real-world based, large scale systems. The performance of MOPSGA is directly compared to that of NSGA-II with regard to solution quality, engineering feasibility (network smoothness) and computational efficiency. MOPSGA is shown to promote both engineering and hydraulic feasibility whilst attaining good infrastructure costs compared to NSGA-II.

Throughout the industrial era, the introduction of innovations by companies ¿ new technologies, new products ¿ has followed a downstream diffusion model, that is, from the owners of the conception and production means and governmental agents to the market. These innovations frequently brought great benefits to a part of the population, but ended up creating serious social and environmental unbalances. As this Diffusion of Innovations model became more and more successful outside the developed nations, the greater the reaction of social scientists towards the need of calculating the consequences and implementing measures that could provide an equal distribution of the benefits as of the losses generated by this model. What this work aims to show, illustrated by the controversial episode of the introduction of the genetically modified foods in Brazil, is that nowadays the diffusion model should incorporate the existence of an upstream movement, that is, from the organized and informed civil society onto the corporations and governments, which questions the companies¿ goals and production processes and the efficiency of the regulating institutions that are under the pressure of the global capital flow¿s dynamic. It¿s the consumer-citizen putting himself in as an active participant of strategic importance at the center of the conception and diffusion of innovations¿ processes.

As empresas brasileiras com atividades internacionais (MNC) possuem uma estrutura de capital diferente das empresas domésticas (DC)? Se sim, é válida a hipótese upstream-downstream, com empresas internacionalizadas utilizando mais dívida do que as empresas domésticas? Encontramos que as MNCs brasileiras utilizam mais dívida devido à atividade internacional, com 9,6% mais alavancagem, dos quais 5,8% são oriundos de fontes de longo prazo. Nós ainda lançamos uma luz sobre uma explicação alternativa para o maior uso de dívida pelas empresas internacionalizadas. Esta dissertação testa se existe um vínculo entre a atividade internacional e o financiamento com dívida estrangeira. O acesso a dívida estrangeira ajuda a explicar porque MNCs utilizam mais dívida do que DCs? Nossos resultados revelam que a atividade internacional está positivamente relacionada ao uso de dívida estrangeira, sendo que MNCs médias carregam 12,7% mais dívida estrangeira em sua estrutura de capital. Nossa amostra consiste em 131 companhias no período 2004-2008, resultando em 538 observações.

Esta tese analisa três lacunas que ainda não foram discutidas na literatura. As lacunas dizem respeito aos efeitos da internacionalização na estrutura de capital e de propriedade de empresas multinacionais. Para tanto, três ensaios sobre os efeitos da internacionalização foram elaborados. O primeiro analisou os efeitos dos modos de entrada sobre o endividamento; o segundo os efeitos da internacionalização nas formas de endividamento e o terceiro os efeitos da internacionalização na estrutura de propriedade. A partir de dados de empresas multinacionais e domésticas de capital aberto latino-americanas de 2007 a 2011, foram montados painéis de dados com variáveis de teste e controle para cada ensaio. Os resultados do primeiro ensaio apontam que os modos de entrada são relevantes para determinar o nível de endividamento das multinacionais e complementam os resultados de trabalhos baseados na hipótese upstream-downstream. Evidenciou-se também que empresas com modos de entrada patrimoniais tendem a ser mais endividadas no longo prazo e total que empresas com modos de entrada não-patrimoniais, reforçando explicações dadas pela teoria do static trade-off; e menos endividadas no curto prazo, reforçando as explicações das teorias da agência e pecking order. Os resultados do segundo ensaio apontam que o grau de internacionalização: (i) aumenta o nível de endividamento da quase totalidade das dívidas providas por agentes financeiros (bancos), conforme a hipótese upstream-downstream e diminuem as dívidas providas por agentes não-financeiros (trade-credit), conforme teoria da restrição financeira; (ii) não produz efeitos sobre a maturidade das dívidas (iii) diferente do que esperava, não aumenta endividamento via banco nacional de desenvolvimento (empréstimos incentivados), (iv) foi relevante para alterar a composição do endividamento das multinacionais se comparada às empresas domésticas e (v) a forma de entrada na internacionalização não afeta a composição de endividamento. No terceiro ensaio o principal resultado encontrado foi que as empresas mais internacionalizadas e com modo de entrada patrimonial têm menores níveis de concentração de propriedade. Outro importante resultado do terceiro ensaio foi de que há uma simultaneidade ainda não explorada em estudos anteriores na determinação do nível de internacionalização e da concentração de propriedade. Ambos os resultados do terceiro ensaio estão amparados pela Visão Baseada em Recursos contrapondo a visão tradicional da teoria da agência.

This work analyzes the entry problem in the hydroelectric generation industry. The operation of a generator upstream regularizes the river flow for generators located downstream on the same river, increasing the production capacity of the latter. This positive externality increases the attractiveness of the locations downstream whenever a generator decides to enter upstream. Therefore, the entry decision of a generator in a given location may affect all entry decisions in potential locations for plants downstream. I first model the problem of generators located in cascade on the same river to show the positive effect of the externality. Next, I develop a method to estimate an entry model specific to the hydro generation industry which takes into account the externality of the entry decisions. Finally, I use a data set on investment decisions of Brazilian hydro-generators to estimate the model. The results show a positive incentive to locate downstream from existing plants and from locations where entry is likely to occur. An interesting by-product of the analysis is that the year effects’ estimates show an increase one year before the energy crisis of 2001, providing evidence that the market anticipated the crisis. It contradicts the governmental version that the crisis was due to an unexpected drought.

O objetivo deste trabalho é propor a utilização do arcabouço teórico das opções reais e a posterior aplicação do modelo binomial na avaliação de projetos relacionados à exploração e produção de petróleo, tendo em vista a flexibilidade gerencial, os riscos e as incertezas técnicas e de mercado que norteiam o setor petrolífero upstream. Ademais, a aplicação do modelo proposto capta o papel crucial da volatilidade do preço do petróleo na avaliação da decisão de investimento e revela a existência dos custos irrecuperáveis extremos decorrentes do ativo real, neste caso, a unidade marítima de petróleo. Assim, com o intuito de prolongar o ciclo de produção de unidade marítima de petróleo com características preestabelecidas, propõe-se a avaliação econômica de duas alternativas tecnológicas para a extensão de vida útil da plataforma marítima objeto de estudo, sendo estas alternativas tratadas como opções de expansão. As alternativas propostas são duas: o afretamento da UMS (Unidade de Manutenção e Segurança) acoplada à plataforma e a docagem da plataforma a partir da desmobilização, isto é, o descomissionamento, e envio da plataforma ao estaleiro. Na aplicação da primeira opção, a UMS se configura em uma embarcação equipada com toda a estrutura necessária para a realização de serviço de manutenção e revitalização, sem que ocorra interrupção da produção de petróleo. Por outro lado, a opção de descomissionamento é desprovida de receita até o retorno da plataforma do estaleiro. No que tange à metodologia do presente trabalho, o modelo binomial com probabilidades de risco neutro é aplicado considerando a receita proveniente da produção de petróleo de uma plataforma marítima com sistema de produção flutuante com 14 poços, sendo 10 produtores e 4 injetores e sustentada por 8 linhas de ancoragem. Também é definida a volatilidade do projeto como sendo a volatilidade do preço do petróleo. Por fim, as opções de expansão podem ser exercidas a qualquer momento antes da data de expiração das opções, data esta coincidente para ambas as opções e referente ao término de contrato de afretamento da UMS, que corresponde ao período de cinco anos. Neste período de cinco anos, as duas alternativas são exercidas a partir do primeiro ano, com receitas e custos distintos em virtude das especificidades decorrentes das alternativas tecnológicas propostas. A partir da aplicação do modelo binomial com probabilidades de risco neutro sob o enfoque das opções reais, as duas alternativas tecnológicas são tratadas como opções americanas na avaliação econômica da revitalização e manutenção da plataforma marítima. Também realiza-se a análise tradicional do VPL para as duas alternativas. As duas análises apontam para a escolha da UMS como alternativa ótima de expansão da vida útil da plataforma. Ademais, a análise sob o enfoque das opções reais capta um valor adicional em ambas as alternativas tecnológicas, fruto das características inerentes à indústria petrolífera. Quanto à estrutura do trabalho em questão se divide em cinco capítulos: introdução, referencial teórico, metodologia, apresentação dos resultados e as considerações finais.
