The therapeutic use of medicinal plants has contributed since antiquity in a beneficial way for health. However, many species lacks of scientific evidence which provide basis for their use in therapeutic practice. In this context is the Genipa americana L. species (Rubiaceae), popularly known as jenipapo and used to treat syfilis, ulcer and hemorrhagic disturbs. It's also used against bruising, as tonic and as aphrodisiac. Due this species lacks toxicological studies, the aim of this study was to evaluate the toxicity in vivo (acute and sub-chronic toxicity) and in vitro (cytotoxicity) of the hydroethanolic extract from G. americana fruits. The hydroethanolic extract of G. americana fruits was prepared by maceration. A preliminary phytochemical analysis was performed to assess the presence of secondary metabolites in the extract. The cytotoxicity study of the extract (0.1, 1.0, 10, 100 and 1000 mg / 100 ul) were performed against normal cells (3T3) and tumor (786-0, HepG2 and B16), analyzed by the MTT assay. To evaluate the acute (single dose of 2000 mg / Kg) and subchronic (100, 500 and 1000 mg / kg for 30 days) toxicity Swiss mice of both sexes were used. At the end of the experiment, blood samples and organs were collected for analysis. Data between groups were compared by t test or ANOVA with Dunnett's post-test with 5% significance level. The phytochemical study of the extracts mainly indicated the presence of iridoids. Results for cytotoxicity tests showed up to 70% inhibition of B16 cell line at a dose of 1000 mg / 100 ul, and up to 29% inhibition of 786-0 at a dose of 10 ug / 100 ul. The extract did not cause death in 3T3 and HepG2 cells. During the in vivo assays, there were no animal deaths. Analysis of blood samples revealed that the animals submitted to the evaluation of acute toxicity had changes in AST and ALT, and that the animals evaluated for subchronic toxicity showed changes in the relative wet weight of the kidney and plasma urea concentration. No differences were observed between groups on histopathological evaluation of the collected organs. Despite the changes found in the in vivo toxicity tests, using the criteria described by the OECD Guidelines, it is suggested that the hydroethanolic extract of the fruits of the G. americana is classified as low toxicity. The cytotoxicity of the extract suggests that they have potential against melanoma cell lines (B16).

The Banisteriopsis genus is widespread in traditional medicine. This work aims to contribute with information about the chemical composition and on the evaluation of the biological activity of the essential oil, the ethanol extract of the leaves and partitions of the Banisteriopsis laevifolia. The phytochemical screeningtest of ethanol extract and partitions of leaves indicated the presence of flavonoids, terpenoids, saponins, phenols and steroids compounds. Nitrogenous compounds, characteristic of some species of this family, were not detected. Flavonoids were the predominant metabolite, with the highest concentrations on the partitions ethyl acetate and n-butanol. The antibacterial activity, antifungal and cytotoxicity of the essetial oil, ethanol extract and partitions were assyed by microdilution broth method (MBM), where the minimum inhibitory concentrations (MIC) were calculated. The ethanol extract and partitions did not inhibit growth against to Gram positive bacteria tested, with MIC less than 400 mg L-1. For the Gram negative bacteria tested, the hexane and hydroethanol partitios were more effective against F. nucleatum bacteria (MIC 100 ug mL-1). The ethanol extract showed antifungal activity with MIC of 31.2 mg L-1. Ethyl acetate and n-butanol partitions showed MIC 187.5 mg L-1 and 93.7 mg L-1, respectively, arousing interest for isolation studies. The antioxidant activity was evaluated by the DPPH free radical method. The ethanolic extract, ethyl acetate and n-butanol partitions were active, since they showed EC50 values (4.53 ug mL-1, 4.07 and 8.39 ug mL-1, respectively), values equivalent to the BHT (7.3 mg L-1). The analysis by HPLC-MS/MS of the most active fractions (ethyl acetate and n-butanol) identified phenolic compounds (flavonols and phenolic acids) which exert recognized biological activity. The GC-MS analysis of the essential oils from leaves collected in two periods studied (dry and wet), showed a small variation in the number of compounds. The major classes identified for the oil collected in the dry period were aliphatic alcohols (23,4%), terpenoids (18.7%), sterols (10.4%) and long-chain alkanes (9.2%) compounds. Terpenoids (26.8%) were the major class for the rain season. The major compounds (3Z) -hexenol, phytol and untriacontano are present in the two seasons but in different amounts (19.4%, 9.8% and 7.5% during the dry season, and 17.0 %, 14.9% and 15.3% in the rainy season, respectively). The essential oil from rainy season was not effective against to the oral bacteria Gram positive and Gram negative tested. However, showed significant antifungal activity with MIC 1000 mg L-1 against Candidas. Thus, the promising results with respect to biological assays of ethanolic extract and partitions from B. laevifolia contributed to the chemical and biological knowledge of the species B. laevifolia.

The objective of this study was to evaluate the productive performance, metabolic and feeding behavior of sheep after ninety days deferred pasture at different heights. The experiment was conducted at Capim Branco experimental farm of the Universidade Federal de Uberlândia. During the period of 90 days, from June to September 2013, forty-eight crossbred lambs Santa Inês x Dorper, divided into groups of four animals, occupied twelve pickets deferred pasture with four initial heights (15 cm, 25 cm, 35 cm and 45 cm). During this period we evaluated the structural characteristics of pasture. For confinement, from September to December 2013, 32 of these animals were used (16 males and 16 females), divided into four bays, separated as pasture were using. The consumption was assessed daily, while biometric measurements were made every 21 days. In relation to gender, there were differences in average daily gain weight. Reviews of feeding behavior occurred at the beginning, middle and end of the experiment for 24 hours. The rumination and leisure activities do not present statistical differences, both initial height of pasture and by period. The time spent on intake was higher during the daytime both treatments starting height as the experimental periods, however, rumination activity was more intense at night. Blood glucose was achieved in five periods of the day, while other metabolites have been obtained with a collection made fortnightly. The different initial heights not promoted effects on blood glucose. The harvesting times were not affected. However, there was a reduction of basal blood glucose of animals throughout the experimental period. There was a significant interaction between the initial pasture heights and periods of evaluation of basal glucose. Cholesterol and triglyceride levels were below recommended levels, however the final phase of confinement cholesterol level increased significantly. The values of VLDL and GGT were above the reference range. FA and AST showed average values within the recommended values. Total protein was influenced by different initial heights of pasture. The creatinine and albumin had values below the recommended range. Moreover, the albumin decreased during the confinement time. Uric acid showed close to the recommended maximum. There was stabilization of compensatory growth, with modification of consumption and weight gain at 45 days of experiment.

In captive common marmoset groups, the reproductive inhibition observed in subordinate female seems to be a result of olfactory, visual and behavioral cues from the dominant female. However, few studies have examined the relationship among adult males living in the same social group. These studies have shown that reproductive failure among peer males seems to be based on hormonal and behavioral mechanisms. New insights on sexual strategies in primates have been shown using fecal steroids, but so far no information is available for common marmoset males. In the present study, we evaluated the influence of light-dark cycle, age and reproductive condition on the profile of fecal androgens in males living in the same family group. Feces were collected from six fathers and six sons for androgen determination during the light phase of the 24-h cycle for eight days randomly distributed over a 4-week period. Androgen levels were determined by enzyme immunoassay technique. Adult sons showed higher androgen levels (166.97 ± 22.95 ng/g) than fathers (80.69 ± 44.38 ng/g) and juveniles (49.06 ± 23.15 ng/g; P < 0.05). No diurnal variation (P > 0.05) in fecal androgen profile was observed in adults or juveniles. No indication of androgen-mediated social competition between fathers and adult sons was demonstrable. These results provide basic information on fecal androgen profile useful to investigate the socioendocrinology of free-ranging common marmoset males and verify that, in contrast to daughters, the reproductive suppression of sons is not based on physiological inhibition of their gonads

The red deer (Cervus elaphus) is currently one of the most widespread and abundant wild ungulates in the Iberian Peninsula and is extremely important both ecologically, as a key species for the functioning of the ecosystems, and economically, as a major game species. In Iberia, red deer populations are subjected to different management systems that may affect the physical condition of the individuals, with further consequences for population dynamics. Studies investigating the effects of management practices and environmental conditions on the performance of red deer are still rare regarding Mediterranean ecosystems. Much of the knowledge concerning the ecology of red deer and the impact of management on its physical condition is based on studies conducted in northern and central regions of Europe, where climatological features and management practices differ from those observed in the Mediterranean areas of Iberia. Studies on a biogeographical scale can provide important insights into the relationships between species and a particular environment and contribute to the development of more targeted and appropriate management practices. The optimisation of sampling procedures and the fine-tuning of pre-existing analytical techniques are also fundamental to a more cost-effective monitoring and, therefore, are of enormous value to wildlife managers. In this context, the main aims of this thesis were: 1) to optimise the procedures used to assess the physical condition of red deer; and 2) to identify relevant management and environmental factors affecting the nutritional condition and stress physiology of red deer in the Mediterranean ecosystems of Iberia, as well as any potential interactions between those factors. Two studies with a methodological focus, presented in the first part of the thesis, demonstrated that the physical condition of red deer can be evaluated more simply, using more cost- and time-effective procedures than those traditionally used: i) it was shown that only one kidney and its associated fat is enough to assess nutritional condition in red deer; and ii) the feasibility of using near infrared spectroscopy to predict the concentrations of stress hormone metabolites was demonstrated using faeces of red deer for the first time. Subsequently, two large-scale observational studies, conducted in representative red deer populations found in Mediterranean Iberia, highlighted the importance of considering seasonal environmental variations and variables related to hunting management practices to better understand the nutritional and physiological ecology of red deer. High population densities had adverse effects on the nutritional condition of the deer and were associated with increased stress levels in natural populations without supplementary feeding. Massive hunting events involving the use of hounds were also identified as a potential source of chronic stress in red deer. The research presented in this thesis has clear implications regarding the management and monitoring of red deer populations in Mediterranean environments and is intended to help wildlife managers to implement more effective monitoring programmes and sustainable management practices.

The jabuticaba fruit tree from classified in the Myrtaceae family and Plinia genre. There are about nine species of this fruit tree, that include as most important, Plinia trunciflora (jabuticaba de cabinho), naturally occurring in southwestern Paraná State, Brazil, P. cauliflora (jabuticaba Paulista or Jabuticaba Açu) and P. jaboticaba (Vell) (jabuticaba sabará), with all the over species producing fruit for the industry or fresh consumption. Nevertheless, there aren‟t commercial orchards with this culture, with highest yield part from extractive. This fact can be combined with lack of technical knowledge for the plants produce in the field. As these species are found in the forest, the first point is whether they can adapt to other light intensity conditions. The aim of this work was to identify the adaptive behavior of jabuticaba fruit seedling and tree when they were put in different light intensities and what this can be considered ideal for the growth, as well as, its influence in the leaves secondary compounds production. Two experiments were conducted, with the first involved with the study of the seedlings and the second with plants in the field. The work was carried out at Universidade Tecnológica Federal do Paraná – Câmpus Dois Vizinhos, Paraná State - Brazil. The experimental design was a completely randomized and a block design with four treatments and four replications of 10 seedlings or two plants per plot, according to nursery or orchard conditions, respectively. The treatments were base according to the light intensity. The treatments used were, 1 - full sun, similar the orchard condition, with 0% shading; 2 - side cover with shade cloth and top with transparent plastic, representing a gap forest condition; 3 - side and top cover with shade cloth, representing stage where the forest canopy is closing, focusing only indirect sunlight; 4 - side and top cover with shade cloth, simulating a closed canopy condition, with PPD (photon flux density) of 10% (90% shading); 5 - side and top cover with shade cloth, simulating a more open canopy condition with PPD 65% (35% shading). The growth and development seedling and plant characteristics were evaluated once by month, as also, during time part in the plants the secondary metabolites leaves, soil activity microbiological and the fresh and dry matter root and shoot and, root length from seedlings. For the growth and development of jabuticaba Açú Paulista seedling recommend to use of side cover with shade cloth and top with transparent plastic, representing a gap forest condition. In orchard, for the growth and development of plants jabuticaba Híbrida tree it was recommended the use of side and top cover with shade cloth of some type. For production of secondary metabolites of leaves, the plant must to be full sunlight condition orchard.

A ocratoxina A é um composto formado a partir do metabolismo secundário de fungos dos gêneros Aspergillus e Penicillium. Uma vez que a presença dessa micotoxina nos alimentos causa sérios danos à saúde humana e animal, surge o interesse pelo desenvolvimento de métodos que visem a redução dos seus níveis em diferentes matrizes. Diversos processos de descontaminação têm sido propostos, sendo que os métodos de redução biológica tem recebido destaque. Esses métodos consistem na aplicação de micro-organismos ou de suas enzimas, o que gera a biotransformação ou degradação da toxina produzindo metabólitos com menor ou nenhuma toxicidade. Diante disso, o objetivo geral do trabalho foi avaliar o efeito da peroxidase na redução dos níveis de ocratoxina A. As enzimas peroxidases testadas foram a comercial e a obtida do farelo de arroz. Para a extração enzimática foram utilizadas as frações granulométricas do farelo de arroz de 48 a 100 mesh, sendo estas frações caracterizadas quimicamente. A peroxidase foi extraída do farelo de arroz em tampão 10 mM pH 5,0 e purificada por partição trifásica, obtendo 77,1% de recuperação e 9,2 para o fator de purificação. O método utilizado para a extração da ocratoxina A do sistema aquoso foi por partição líquido-líquido utilizando como solvente o clorofórmio, sendo esse método validado segundo os parâmetros de linearidade (0,1 a 20 ng mL-1), coeficientes de correlação (0,9997) e de determinação (0,9994), e limites de detecção (0,02) e quantificação (0,03). A afinidade entre as peroxidases e a ocratoxina A foi verificada segundo os parâmetros de KM e Vmáx, resultando em 0,00027 mM e 0,000015 mM min-1, respectivamente, para a peroxidase comercial, e 0,0065 mM e 0,000031 mM min-1 para a obtida do farelo de arroz. Com relação aos percentuais de redução de ocratoxina A, foram avaliadas 3 proporções enzima:substrato (1:10, 1:5 e 8:1 para a comercial e 1:10, 1:5 e a com atividade de 0,063 U mL-1 para a do farelo), sendo que as proporções que forneceram maior redução foi a de 8:1 para a enzima comercial (0,063 U mL-1) e a correspondente a 0,063 U mL-1 para a enzima obtida do farelo. Os percentuais de redução de ocratoxina A foram de 59% para a peroxidase comercial em 300 min e 41% para a peroxidase do farelo de arroz em 1440 min. O efeito de adsorção da ocratoxina A pela enzima peroxidase foi descartado uma vez que foi realizada a sua hidrólise com a enzima pepsina e verificado um percentual de 2,7% de adsorção, demonstrando que a redução foi por ação enzimática. A enzima obtida de farelo de arroz com atividade de 0,063 U mL-1 foi aplicada em suco de uva tinto e branco. Observou-se que para o primeiro não houve redução significativa, enquanto que para o segundo a redução foi de 17%. Neste trabalho, então, foi possível verificar a capacidade de redução dos níveis da ocratoxina A pela enzima peroxidase, tanto em sistema aquoso como no suco de uva integral branco.

Há uma crescente procura por alimentos mais saudáveis e seguros para atender uma população cada vez maior e mais exigente. Nos últimos anos o interesse por surfactantes de origem microbiana tem aumentado significativamente em decorrência de serem naturalmente biodegradáveis diminuindo assim o impacto ambiental. Uma grande variedade de microorganismos produz biossurfactantes, sendo que o tipo, a quantidade e a qualidade do biossurfactante são influenciados pelos constituintes do meio, tais como, fontes de carbono, nitrogênio e sais inorgânicos, além das condições de cultivo, como pH, temperatura, agitação e disponibilidade de oxigênio. Os biossurfactantes são metabólitos microbianos de superfície ativa que apresentam uma vasta aplicação no setor industrial. Os objetivos deste trabalho foram selecionar microalgas com potencial para produzir biossurfactantes e estudar a produção por microalgas em diferentes fotobiorreatores e condições nutricionais. O trabalho foi dividido em quatro etapas: 1) cultivo autotrófico e mixotrófico de microalgas para produção de biossurfactantes; 2) Seleção de microalgas para produção de biossurfactantes; 3) Produção de biossurfactantes por microalgas em diferentes fotobiorreatores e 4) Cultivo outdoor da microalga marinha Tetraselmis suecica OR para produção de biossurfactantes. Na primeira etapa Spirulina sp. LEB-18, Synechococcus nidulans LEB-25, Chlorella vulgaris LEB-106, Chlorella minutissima LEB-108 e Chlorella homosphaera foram cultivadas com glicose (cultivo mixotrófico). Spirulina sp. LEB-18 apresentou concentrações máximas de biomassa (2,55 g.L-1 ) quando foi utilizada 5 g.L-1 de glicose no meio de cultivo. A tensão superficial dos meios das microalgas foi reduzida de 70 para 43 mN.m-1 para as microalgas estudadas utilizando glicose como fonte de carbono. Resultados da segunda etapa mostraram que a microalga Scenedesmus sp. 3PAV3 apresentou valor de atividade emulsificante óleo em água (AE o/a) superior (339,8 UE.g-1 ) ao encontrado para as demais microalgas. Os menores valores de tensões superficiais variaram de 27,4 a 31,2 mN.m-1 . Na terceira etapa verificou-se que a microalga Chlorella sp. PROD1 apresentou valor de AE o/a semelhante (258,2 UE g -1 ) ao encontrado para o emulsificante comercial lecitina de soja (257,0 UE g -1 ) e ambas as microalgas estudadas alcançaram valores de tensões superficiais abaixo de 30 mN.m -1 . Na última etapa, Tetraselmis suecica OR cultivada em fotobiorreator do tipo Green Wall Panel apresentou menores valores de tensões superficiais para cultura com limitação de nitrogênio. Os resultados demonstraram a potencialidade das microalgas estudadas na produção de biossurfactantes, tanto pela redução da tensão superficial e interfacial, como pelo aumento da atividade emulsificante, confirmando uma possível aplicação como emulsificante, detergente, lubrificante, estabilizante, entre outras.

As Microcistinas são heptapeptídios cíclicos produzidos como metabólitos secundários por diferentes espécies de cianobactérias, sendo relevantes pelo seu potencial hepatotóxico. Peixes apresentam estratégias bioquímicas para detoxificar contaminantes ambientais, incluindo a ativação de enzimas de fase II de biotransformação, que incluem as isoformas de glutationa S-transferase (GST). As GST catalizam a conjugação de glutationa reduzida (GSH) com uma variedade de xenobióticos, incluindo as microcistinas. O presente estudo avaliou os níveis transcricionais de quinze isoformas de GST a fim de identificar isoformas possivelmente envolvidas na detoxificação de contaminantes ambientais como a microcistina-LR (MC-LR) em Danio rerio. A técnica de PCR em tempo real (RT-qPCR) foi utilizada para avaliação dos níveis transcricionais, permitindo análise das GST em diferentes órgãos, abundância e a ativação/repressão das isoformas de GST pela exposição à MC-LR. Foram avaliados os possíveis efeitos causados em brânquia e fígado após exposição por 24 hs às concentrações de 5 µg.L-1 e 50 µg.L-1 de MC-LR. Baseado nos scores de estabilidade para oito genes normalizadores, foram selecionados glicose-6-fosfato desidrogenase (g6pdh), β-actina1 e beta-2-microglobulina (b2m); b2m, alfa-tubulina 1 (tuba) e β- actin1; e tuba, b2m e g6pdh, para normalização dos níveis trancricionais de GST para distribuição órgão-específica, abundância e efeito da MC-LR em brânquia e fígado, respectivamente. A avaliação transcricional da distribuição órgão-específica revelou níveis significativos de gstal e gstk1.1 no fígado; gstp1 e gstp2 em brânquia; mgst3a, gstr1, gstm2, gstm33, gstp1, gstp2 e gstk1.1 no intestino; gstm2, gstm3 e gstal no olho e gstt1a e gsta2.1 no cérebro. Considerando os níveis de transcritos para um dado órgão, gstk1.1, gstal, gstp1 e gstt2 foram mais abundantes nos órgãos de detoxificação, tais como o fígado, brânquias e intestino, enquanto gstt1a e gsta2.1 foram mais abundantes no rim. Em brânquia, gsta2.1 e gstt1b foram reprimidas por 5 µg.L-1 de MC-LR e mgst1.1 foi reprimida em 50 µg.L-1 de MC-LR. No fígado, as isoformas gst2.2 e gstp2 foram reprimidas em ambas as concentrações, gstal foi reprimida em 5 µg.L-1, e gstt1a e gstk1.1 foram reprimidas em 50 µg.L-1 de MC-LR. As isoformas gstal, gstr1, gstp1, mgst3a, gstm1, gstm2 e gstm3 não foram alteradas pela exposição a MC-LR. Os resultados obtidos fornecem informações para a escolha de isoformas específicas de GST possivelmente envolvidas na detoxificação/toxicidade de MC-LR, a serem melhores caracterizadas ao nível protéico e também contribui para a escolha de genes normalizadores a serem utilizados em outros estudos da mesma natureza

A avaliação do aporte de matéria orgânica no ambiente aquático por atividades antrópicas pode ser realizada através da identificação e quantificação de marcadores moleculares. Diversos estudos apontam à aplicação dos marcadores moleculares com esta finalidade, no entanto, poucos avaliam a variação das concentrações desses compostos ao longo do tempo, registrada nas camadas sedimentares. O presente trabalho realiza um estudo a partir de três classes de marcadores moleculares presentes em perfis sedimentares da região do Complexo Estuarino de Paranaguá (CEP) no Paraná (PR), que nos últimos anos vêm sofrendo com o crescente desenvolvimento de atividades antrópicas. Como objetivo, tem-se identificar as principais fontes de matéria orgânica e estudar o histórico destes aportes em colunas sedimentares do CEP, relacionando as taxas de sedimentação com a deposição de origem natural e antrópica. A legislação vigente para o monitoramento ambiental, no que diz respeito à contaminação por esgoto fecal, sugere a avaliação por indicadores microbiológicos, porém, indicadores químicos como os esteróides fecais são uma alternativa bastante promissora, pois estes são persistentes, sendo menos sensíveis a variações ambientais. Outros dois marcadores moleculares de aportes antrópicos ao ambiente que foram determinados neste estudo são os alquilbenzenos lineares (LABs), presente em detergentes, que indicam aportes antrópicos oriundos de esgoto doméstico e a determinação de cafeína, tendo em vista que os esteróides fecais podem ser originários de fezes de animais de sangue quente, podendo indicar outras fontes. Para o presente trabalho foram coletados 12 testemunhos de até 1 m de profundidade em maio de 2006, totalizando 12 pontos de coleta e um montante de 121 amostras. As análises foram realizadas por cromatografia em fase gasosa com detecção por espectrometria de massas (CG-EM). Os esteróides encontrados em maior concentração foram o β- sitosterol (71,4 µg g-1), estigmasterol (8,7 µg g-1), colestanol (3,6 µg g-1) e o estigmastanol (2,8 µg g-1), todos oriundos de fonte natural, indicando que a maior contribuição para o CEP é por aporte biogênico. O coprostanol, que é um esterol fecal, foi encontrado entre as concentrações de 0,001 e 4,10 µg g-1, outros dois esteróides de origem fecal também foram detectados, coprostanona e epicoprostanol, onde as maiores concentrações foram 3,6 e 0,2 µg g-1, respectivamente, sendo encontrados em regiões próximas a centros urbanos, indicando origem antrópica. As maiores concentrações para o ∑LABs também foram encontradas em regiões próximas às cidades de Antonina e Paranaguá, sendo a maior encontrada no testemunho #3 Gererês (208 ng g-1). Para o último marcador molecular analisado, a cafeína, foi encontrada a maior concentração de 18,41 ng g-1, sendo este ponto localizado longe dos centros urbanos, porém este contaminante é bastante solúvel em água podendo ser transportado na coluna d’água e percorrer grandes distâncias. Através dos compostos analisados, pode-se perceber que a intervenção antropogênica foi mais marcante nos testemunhos coletados no eixo leste-oeste do CEP, ficando registrado nas camadas sedimentares.

O Empreendimento de Fins Múltiplos de Alqueva (EFMA) é um projecto estruturante centrado na barragem do Alqueva, que pretende garantir o recurso Água na região e com isso viabilizar uma série de infra-estruturas que contribuirão para o desenvolvimento, em termos económicos e sociais, de uma das regiões mais desfavorecidas da Europa. O EFMA é constituído pelo "Reservatório Mãe" e por um conjunto de outras albufeiras de menor dimensão, interligados por uma rede com cerca de dois mil quilómetros de canais e condutas e mais de meia centena de estações de bombagem, contemplando os seguintes usos da água: - Abastecimento público. - Fornecimento de água para o desenvolvimento de actividades agrícolas e agro-industriais; - Produção de energia; Este trabalho tem por objectivo avaliar a adequabilidade da qualidade da água para o uso rega na origem e produzir informação adequada de suporte à decisão. Os resultados obtidos permitem afirmar que de um modo geral a qualidade da água captada no sistema Alqueva- Pedrógão é adequada para o uso rega, apesar de se verificar uma persistente contaminação de coliformes fecais. ABSTRACT; The Multipurpose Alqueva Project (EFMA- Empreendimento de Fins Múltiplos de Alqueva) is a groundwork project centred in Alqueva dam and its reservoir, which intends to guarantee a strategic water reserve in the region, and therefore making possible a series of associated infrastructures, contributing for the potential development of the region, in economic and social terms, in one of the most poorest regions of Europe. The EFMA includes a "Main Reservoir" and set of other dams with lesser dimension, connected by a several canals and pipelines about two thousand kilometers long (including Primary and Secondary Network), and about 50 pumping stations, with the goal of attaining the following main objectives: - Water supply for the development of agricultural and agro-industrial activities; - Production of energy; - Public Water Supply. This study intends to evaluate the suitability of water quality in the origin for irrigation use and to achieve adequate information for support decision. The results have revealed that the quality of water abstracted in the Alqueva - Pedrógão System is suitable for irrigation, although the persistent contamination of fecal coliforms.

Nutritional status is an important determinant to the response against Leishmania infection, although few studies have characterized the molecular basis for the association found between malnutrition and the disease. Vitamin A supplementation has long been used in developing countries to prevent mortality by diarrheal and respiratory diseases, but there are no studies on the role of vitamin A in Leishmania infection, although we and others have found vitamin A deficiency in visceral Leishmaniasis (VL). Regulatory T cells are induced in vitro by vitamin A metabolites and are considered important cells implicated T CD4+ cell suppression in human VL. This work aimed to examine the correlation of nutritional status and the effect of vitamin A in the response against Leishmania infantum infection. A total of 179 children were studied: 31 had active VL, 33 VL history, 44 were DTH+ and 71 were DTH- and had negative antibody to Leishmania (DTH-/Ac-). Peripheral blood monuclear cells were isolated in a subgroup of 10 active VL and 16 DTH-/Ac- children and cultivated for 20h under 5 different conditions: 1) Medium, 2) Soluble promastigote L. infantum antigens (SLA), 3) All-trans retinoic acid (ATRA), 4) SLA + ATRA and 5) Concanavalin A. T CD4+CD25highFoxp3+, T CD4+CD25-Foxp3- and CD14+ monocytes were stained and studied by flow cytometry for IL-10, TGF-β and IL-17 production. Nutritional status was compromised in VL children, which presented lower BMI/Age and retinol concentrations when compared to healthy controls. We found a negative correlation between nutritional status (measured by BMI/Age and serum retinol) and anti-Leishmania antibodies and acute phase proteins. There was no correlation between nutritional status and parasite load. ATRA presented a dual effect in Treg cells and monocytes: In healthy children (DTH-/Ac-), it induced a regulatory response, increasing IL-10 and TGF-β production; in VL children it modulated the immune response, preventing increased IL-10 production after SLA stimulation. Furthermore, we found a positive correlation between BMI/Age and IL-17 production and negative correlation between serum retinol and IL-10 and TGF-β production in T CD4+CD25highFoxp3+ cells after SLA stimulus. Our results show a potential dual role of vitamin A in the immune system: improvement of regulatory profile during homeostasis and down modulation of IL-10 in Treg cells and monocytes during symptomatic VL. Therefore, the use of vitamin A concomitant to VL therapy might improve recovery from disease status in Leishmania infantum infection

The mobilization of food reserves in storage tissues and allocation of their hydrolysis products in the growing axis are critical processes for the establishment of seedlings after germination. Therefore, it is crucial for mobilization of reserves to be synchronized with the growing axis, so that photosynthetic activity can be started before depletion of reserves. For this, integrative approaches involving different reserves, different hydrolysis products and interaction between storage and growing axis tissues, either through hormones or metabolites with signaling role, can contribute greatly to the elucidation of the regulation mechanisms for reserve mobilization. In this study, was hypothesized that hormones and metabolites have different actions on reserve mobilization, and there must be a crossed effect of sugars on the mobilization of proteins and amino acids on lipids and starch mobilization in sunflower seedlings. This study was conducted with seeds of sunflower (Helianthus annuus L.) hybrid Helio 253 using in vitro culture system. Seeds were germinated on Germitest® paper and grown on agar-water 4 g/L without addition of nutrients during 9 days after imbibition (DAI) for growth curve. To verify the effect of metabolites and hormones, seedlings were transferred in the 2nd DAI to agar-water 4 g/L supplemented with increasing concentrations of sucrose or L-glutamine, abscisic acid, gibberellic acid or indolebutyric acid. The results of this study confirm that the mobilization of lipids and storage proteins occurs in a coordinated manner during post-germination growth in sunflower, corroborating the hypothesis that the application of external carbon (sucrose) and nitrogen (L-glutamine) sources can delay the mobilization of these reserves in a crossed way. Moreover, considering the changes in the patterns of reserve mobilization and partition of their products in seedlings treated with different growth regulators, it is evident that the effects of metabolites and hormones must involve, at least in part, distinct mechanisms of action
