RESUMO: Os Staphylococcus aureus resistentes à meticilina (MRSA, do inglês “methicillin-resistant Staphylococcus aureus”) são um dos principais agentes responsáveis por infeções hospitalares. Os MRSA são resistentes a praticamente todos os antibióticos β-lactâmicos devido a dois mecanismos principais: produção de β-lactamase (bla), codificada pelo gene blaZ, e produção de uma proteína de ligação à penicilina (PBP2a, do inglês “penicillin binding protein 2”), codificada pelo gene mecA. Estes dois genes são regulados por sistemas homólogos, constituídos por um sensor-transdutor (BlaR1 e MecR1) e um repressor (BlaI e MecI), de tal modo que ambos os sistemas são capazes de co-regular os genes mecA e blaZ, embora com eficiências de indução muito diferentes. De facto, a indução mediada pelo sistema mecI-mecR1 é tão lenta que se acredita que este sistema não está funcional na maioria das estirpes MRSA. No entanto, dados recentes do nosso laboratório, demonstram a ausência de relação entre a presença do gene mecI e o nível de resistência à meticilina em estirpes MRSA epidémicas, e também que, o fenótipo de resistência da grande maioria das estirpes não é perturbado pela sobre-expressão em trans do repressor mecI. Curiosamente, as duas estirpes em que a expressão da resistência foi afetada pela sobre-expressão do mecI são negativas para o locus da β-lactamase, o que sugere que este locus pode interferir diretamente com a repressão do gene mecA mediada pelo MecI. Nesta tese de mestrado esta hipótese foi explorada usando estratégias de biologia molecular e ensaios fenotípicos da resistência aos -lactâmicos. Os resultados obtidos demonstram que a presença do plasmídeo nativo da β-lactamase não só anula a repressão mediada pelo MecI, como também aumenta o nível de resistência das estirpes parentais. Várias hipóteses foram então formuladas para explicar estas observações. Dados preliminares, em conjunto com evidências experimentais publicadas, sugerem que o BlaI forma hetero-dímeros com o MecI que, após a indução, são inativados eficientemente pelo BlaR1. Em conclusão, estes resultados apresentam novas perspetivas para o mecanismo de regulação do mecA e para uma nova importante função do operão da β-lactamase para o fenótipo das estirpes MRSA.-------------------ABSTRACT: Methicillin-resistant Staphylococcus aureus (MRSA) is an important nosocomial pathogen and is also emerging in the community. MRSA is cross-resistant to virtually all β-lactam antibiotics and has acquired two main resistance mechanisms: production of β-lactamase (bla), coded by blaZ, and production of penicillin binding protein 2a (PBP2a), coded by mecA. Both genes are regulated by homologous sensor-transducers (BlaR1 and MecR1) and repressors (BlaI and MecI), and coregulation of mecA and blaZ by both systems has been demonstrated, although with remarkable different efficiencies. In fact, induction of mecA by mecI-mecR1 is so slow that it is believed it is not functional in most MRSA strains. However, recent data from our laboratory has unexpectedly demonstrated that not only there is no correlation between the presence of mecI gene and the resistance level in epidemic MRSA strains, but also that for most strains there were no significant changes on the resistance phenotype upon the mecI overexpression in trans. Interestingly, the two strains in which mecI overexpression affected the resistance expression were negative for the bla locus, suggesting that this locus may interfere directly with the MecI-mediated repression of mecA and account for those puzzling observations. In this master thesis we have explored this hypothesis using molecular biology strategies and phenotypic analysis of -lactam resistance. The data obtained demonstrate that the presence of a wild-type plasmid containing the bla locus not only disrupts the MecImediated repression, but also significantly enhances the expression of resistance. Several preliminary hypotheses were formulated to explain these observations and preliminary data, together with published evidence, support the working model that BlaI forms functional hetero-dimers with MecI, which upon induction are readily inactivated by BlaR1. These results provide new insights into the regulatory mechanism(s) of mecA and open new perspectives for the role of β-lactamase operon in the MRSA phenotype.

Microalgae are promising microorganisms for the production of food and fine chemicals. Several species of microalgae are used in aquaculture with the purpose of transfer bioactive compounds up to the aquatic food chain. The main objective of this project was to develop a stress–inducement strategy in order to enhance the biochemical productivity of Nannochloropsis gaditana, Rhodomonas marina and Isochrysis sp. for aquaculture purposes having in account their growth and organizational differences. In this regard, two experiments were design: the first one consisted on the alteration of overall nutrient availabilities in growth medium; and the second one comprised changes in nitrogen and sulfur concentrations maintaining the concentrations of the other nutrients present in a commercial growth medium (Nutribloom plus), which is frequently used in aquaculture. Microalgae dried biomass was characterized biochemically and elemental analysis was also performed for all samples. In first experimental design: linear trends between nutrient availability in growth media and microalgae protein content were obtained; optimum productivities of eicosapentaenoic (EPA) and docosahexaenoic acids (DHA) were attained for both R. marina and N. gaditana in growth media enriched with 1000 L L-1 of nutrient solution whereas for Isochrysis sp. the double of Nutribloom plus was needed; the decrease of glucans and total monosaccharides with nutrient availability for R. marina and Isochrysis sp. showed the occurrence of a possible depletion of carbohydrates towards lipids and proteins biosynthesis. Second experimental desing: N. gaditana exhibited the highest variation in their biochemical composition against the applied perturbation; variations observed for microalgae in their biochemical composition were reflected in their elemental stoichiometry; in N. gaditana the highest nitrogen concentrations lead to overall maximum productivities of the biochemical parameters. The results of the present work show two stress-inducement strategies for microalgae that may constitute a base for further investigations on their biochemical enhancement.

O presente trabalho tem como objetivo a otimização da etapa de fermentação dos açúcares obtidos a partir da drêche cervejeira para produção do bioetanol através da utilização das leveduras Pichia stipitis NCYC 1541 e Kluyveromyces marxianus NCYC 2791 como agentes fermentativos. O meio de cultura usado para manter as culturas destas leveduras foi Yeast Extract Peptone Dextrose (YEPD). O principal propósito deste trabalho foi o de encontrar alternativas aos combustíveis fósseis, pautando-se por soluções inofensivas para o meio ambiente e sustentáveis. Assim, o trabalho está dividido em quatro etapas: 1) caraterização química e biológica da drêche; 2) pré-tratamento ácido e hidrólise enzimática para primeiramente quebrar as moléculas de lenhina que envolvem os polímeros de celulose e hemicelulose e em seguida romper as ligações poliméricas destas macromoléculas por ação enzimática e transforma-las em açúcares simples, respetivamente, obtendo-se então a glucose, a maltose, a xilose e a arabinose; e, por último, 3) otimização da etapa de fermentação da glucose, maltose e das pentoses que constitui a condição essencial para se chegar à síntese do bioetanol de um modo eficiente e sustentável e 4) a recuperação do bioetanol produzido por destilação fracionada. A quantificação dos açúcares libertados no processo foi feita recorrendo a análises por cromatografia líquida de alta eficiência (HPLC). Neste estudo foram identificados e quantificados cinco açúcares: Arabinose, Glucose, Maltose, Ribose e Xilose. Na etapa de pré-tratamento e hidrólise enzimática foram usados os ácidos clorídrico (HCl) e nítrico (HNO3) com a concentração de 1% (m/m), e as enzimas Glucanex 100g e Ultraflo L. Foram testadas seis condições de pré-tratamento e hidrólise enzimática, alterando os parâmetros tempo de contacto e razão enzimas/massa de drêche, respetivamente, e mantendo a temperatura (50 ºC), velocidade de agitação (75 rpm) e concentração dos ácidos (1% (m/m)). No processamento de 25 g de drêche seca com 0,5 g de Glucanex, 0,5 mL de Ultraflo e um tempo de reação de 60 minutos para as enzimas foi obtida uma eficiência de 15%, em hidrolisado com 6% da celulose. Realizou-se a fermentação do hidrolisado resultante do pré-tratamento ácido e hidrólise enzimática de drêche cervejeira e de meios sintéticos preparados com os açúcares puros, usando as duas estirpes selecionadas para este estudo: Pichia stipitis NCYC 1541 e Kluyveromyces marxianus NYCY 2791. As eficiências de fermentação dos açúcares nos meios sintéticos foram superiores a 80% para ambas as leveduras. No entanto, as eficiências de fermentação do hidrolisado da drêche foram de 45,10% pela Pichia stipitis e de 36,58 para Kluyveromyces marxianus, para um tempo de fermentação de 72 horas e à temperatura de 30 °C. O rendimento teórico em álcool no hidrolisado da drêche é de 0,27 g/g, três vezes maior do que o real (0,0856 g/g), para Pichia stipitis e de 0,19 g/g seis vezes maior do que o real (0,0308 g/g), para a Kluyveromyces marxianus.

O decréscimo das reservas de petróleo e as consequências ambientais resultantes do recurso a combustíveis fósseis nos motores a diesel têm levado à procura de combustíveis alternativos. Esta pesquisa alicerçada nas fontes de energia renovável tornou-se essencial, face à crescente procura de energia e ao limitado fornecimento de combustíveis fósseis . Resíduos de óleo de cozinha, gordura animal, entre outros resíduos de origem biológica, tais como a borra de café, são exemplos de matérias-primas para a produção de biodiesel. A sua valorização tem interesse quer pela perspetiva ambiental, quer pela económica, pois aumenta não só a flexibilidade e diversificação das matérias-primas, mas também contribui para uma estabilidade de custos e alteração nas políticas agrícolas e de uso do solo. É neste contexto que se enquadra o biodiesel e a borra de café, pretendendo-se aqui efetuar o estudo da produção, à escala laboratorial, de biodiesel a partir da borra de café, por transesterificação enzimática, visando a procura das melhores condições reacionais. Iniciando-se com a caracterização da borra de café, foram avaliados antes e após a extração do óleo da borra de café, diversos parâmetros, de entre os quais se destacam: o teor de humidade (16,97% e 6,79%), teor de cinzas (1,91 e 1,57%), teor de azoto (1,71 e 2,30%), teor de proteínas (10,7 e 14,4%), teor de carbono (70,2 e 71,7%), teor de celulose bruta (14,77 e 18,48%), teor de lenhina (31,03% e 30,97%) e poder calorifico superior (19,5 MJ/kg e 19,9 MJ/kg). Sumariamente, constatou-se que os valores da maioria dos parâmetros não difere substancialmente dos valores encontrados na literatura, tendo sido evidenciado o potencial da utilização desta biomassa, como fonte calorifica para queima e geração de energia. Sendo a caracterização do óleo extraído da borra de café um dos objetivos antecedentes à produção do biodiesel, pretendeu-se avaliar os diferentes parâmetros mais significativos. No que diz respeito à caracterização do óleo extraído, distingue-se a sua viscosidade cinemática (38,04 mm2/s), densidade 0,9032 g/cm3, poder calorífico de 37,9 kcal/kg, índice de iodo igual a 63,0 gI2/ 100 g óleo, o teor de água do óleo foi de 0,15 %, o índice de acidez igual a 44,8 mg KOH/g óleo, ponto de inflamação superior a 120 ºC e teor em ácidos gordos de 82,8%. Inicialmente foram efetuados ensaios preliminares, a fim de selecionar a lipase (Lipase RMIM, TL 100L e CALB L) e álcool (metanol ou etanol puros) mais adequados à produção de biodiesel, pelo que o rendimento de 83,5% foi obtido através da transesterificação mediada pela lipase RMIM, utilizando como álcool o etanol. Sendo outro dos objetivos a otimização do processo de transesterificação enzimática, através de um desenho composto central a três variáveis (razão molar etanol: óleo, concentração de enzima e temperatura), recorrendo ao software JMP 8.0, determinou-se como melhores condições, uma razão molar etanol: óleo 5:1, adição de 4,5% (m/m) de enzima e uma temperatura de 45 ºC, que conduziram a um rendimento experimental equivalente a 96,7 % e teor de ésteres 87,6%. Nestas condições, o rendimento teórico foi de 99,98%. Procurou-se ainda estudar o efeito da adição de água ao etanol, isto é, o efeito da variação da concentração do etanol pela adição de água, para teores de etanol de 92%, 85% e 75%. Verificou-se que até 92% decorreu um aumento da transesterificação (97,2%) para um teor de ésteres de (92,2%), pelo que para teores superiores de água adicionada (75% e 85%) ocorreu um decréscimo no teor final em ésteres (77,2% e 89,9%) e no rendimento da reação (84,3% e 91,9%). Isto indica a ocorrência da reação de hidrólise em maior extensão, que leva ao desvio do equilíbrio no sentido contrário à reação de formação dos produtos, isto é, dos ésteres. Finalmente, relativamente aos custos associados ao processo de produção de biodiesel, foram estimados para o conjunto de 27 ensaios realizados neste trabalho, e que corresponderam a 767,4 g de biodiesel produzido, sendo o custo dos reagentes superior ao custo energético, de 156,16 € e 126,02 €, respetivamente. Naturalmente que não esperamos que, a nível industrial os custos sejam desta ordem de grandeza, tanto mais que há economia de escala e que as enzimas utilizadas no processo deveriam ser reutilizadas diversas vezes.

OBJECTIVE: Statins are among the most prescribed drugs worldwide and their recently discovered anti-inflammatory effect seems to have an important role in inhibiting proinflammatory cytokine production, chemokines expression and counteracting the harmful effects of sepsis on the coagulation system. We decided to perform a meta-analysis of all randomized controlled trials ever published on statin therapy in septic patients to evaluate their effect on survival and length of hospital stay. DATA SOURCES AND STUDY SELECTION: Articles were assessed by four trained investigators, with divergences resolved by consensus. BioMedCentral, PubMed, Embase and the Cochrane Central Register of clinical trials were searched for pertinent studies. Inclusion criteria were random allocation to treatment and comparison of statins versus any comparator in septic patients. DATA EXTRACTION AND SYNTHESIS: Data from 650 patients in 5 randomized controlled studies were analyzed. No difference in mortality between patients receiving statins versus control (44/322 [14%] in the statins group vs 50/328 [15%] in the control arm, RR = 0.90 [95% CI 0.65 to 1.26], p = 0.6) was observed. No differences in hospital stay (p = 0.7) were found. CONCLUSIONS: Published data show that statin therapy has no effect on mortality in the overall population of adult septic patients. Scientific evidence on statins role in septic patients is still limited and larger randomized trials should be performed on this topic.

Tyrosine hydroxylase (TH) deficiency is an inborn error of dopamine biosynthesis and a cause of early parkinsonism. Two clinical phenotypes have been described. Type “B”: early onset severe encephalopathy; type “A”: later onset, less severe and better response to L-dopa. We aimed to study the expression of several key dopaminergic and gabaergic synaptic proteins in the cerebrospinal fluid (CSF) of a series of patients with TH deficiency and their possible relation with the clinical phenotype and response to L-DOPA. Dopamine transporter (DAT), D2-receptor and vesicularmonoamine transporter (VMAT2)weremeasured in the CSF of 10 subjectswith THdeficiency byWestern blot analysis. In 3 patients, data of pre- and post-treatmentwith L-DOPA were available, and in one of them, GABA vesicular transporter was determined. Results were compared to an age-matched control population. The concentration of D2-receptors in CSFwas significantly higher in patients with TH deficiency than in controls. Similarly, DAT and vesicular monoamine transporter type 2 were up-regulated. Studies performed before LDOPA, and on L-DOPA therapy showed a paradoxical response with D2 receptor expression increase as L-Dopa doses and homovanillic concentration gradually raised in a B phenotype patient. The opposite results were found in two patients with A phenotype. However, this is a very small sample, and further studies are needed to conclude robust differences between phenotypes. Synaptic proteins are detectable in the CSF and their quantification can be useful for understanding the pathophysiology of neurotransmitter defects and potentially to adjust and personalize treatments in the future.

Characterized native and recombinant Hevea brasiliensis (rHev b) natural rubber latex (NRL) allergens are available to assess patient allergen sensitization profiles. OBJECTIVE: Quantification of individual IgE responses to the spectrum of documented NRL allergens and evaluation of cross-reactive carbohydrate determinants (CCDs) for more definitive diagnosis. METHODS: Sera of 104 healthcare workers (HCW; 51 German, 21 Portuguese, 32 American), 31 spina bifida patients (SB; 11 German, 20 Portuguese) and 10 Portuguese with multiple surgeries (MS) were analysed for allergen-specific IgE antibody (sIgE) to NRL, single Hev b allergens and CCDs with ImmunoCAP technology. RESULTS: In all patient groups rHev b 5-sIgE concentrations were the most pronounced. Hev b 2, 5, 6.01 and 13 were identified as the major allergens in HCW and combined with Hev b 1 and Hev b 3 in SB. In MS Hev b 1 displayed an intermediate relevance. Different sIgE antibody levels to native Hevea brasiliensis (nHev b) 2 and rHev b 6.01 allowed discrimination of SB with clinical relevant latex allergy vs. those with latex sensitization. Sensitization profiles of German, Portuguese and American patients were equivalent. rHev b 5, 6.01 and nHev b 13 combined detected 100% of the latex-allergic HCW and 80.1% of the SB. Only 8.3% of the sera showed sIgE response to CCDs. CONCLUSIONS: Hev b 1, 2, 5, 6.01 and 13 were identified as the major Hev b allergens and they should be present in standardized latex extracts and in vitro allergosorbents. CCDs are only of minor relevance in patients with clinical relevant latex allergy. Component-resolved diagnostic analyses for latex allergy set the stage for an allergen-directed immunotherapy strategy

Several risk factors for asthma have been identified in infants and young children with recurrent wheeze. However, published literature has reported contradictory findings regarding the underlying immunological mechanisms. OBJECTIVES: This study was designed to assess and compare the immunological status during the first 2 years in steroid-naive young children with >or= three episodes of physician-confirmed wheeze (n=50), with and without clinical risk factors for developing subsequent asthma (i.e. parental asthma or a personal history of eczema and/or two of the following: wheezing without colds, a personal history of allergic rhinitis and peripheral blood eosinophilia >4%), with age-matched healthy controls (n=30). METHODS: Peripheral blood CD4(+)CD25(+) and CD4(+)CD25(high) T cells and their cytotoxic T-lymphocyte-associated antigen-4 (CTLA-4), GITR and Foxp3 expression were analysed by flow cytometry. Cytokine (IFN-gamma, TGF-beta and IL-10), CTLA-4 and Foxp3 mRNA expression were evaluated (real-time PCR) after peripheral blood mononuclear cell stimulation with phorbol 12-myristate 13-acetate (PMA) (24 h) and house dust mite (HDM) extracts (7th day). RESULTS: Flow cytometry results showed a significant reduction in the absolute number of CD4(+)CD25(high) and the absolute and percentage numbers of CD4(+)CD25(+)CTLA-4(+) in wheezy children compared with healthy controls. Wheezy children at a high risk of developing asthma had a significantly lower absolute number of CD4(+)CD25(+) (P=0.01) and CD4(+)CD25(high) (P=0.04), compared with those at a low risk. After PMA stimulation, CTLA-4 (P=0.03) and Foxp3 (P=0.02) expression was diminished in wheezy children compared with the healthy children. After HDM stimulation, CTLA-4 (P=0.03) and IFN-gamma (P=0.04) expression was diminished in wheezy children compared with healthy children. High-risk children had lower expression of IFN-gamma (P=0.03) compared with low-risk and healthy children and lower expression of CTLA-4 (P=0.01) compared with healthy children. CONCLUSIONS: Although our findings suggest that some immunological parameters are impaired in children with recurrent wheeze, particularly with a high risk for asthma, further studies are needed in order to assess their potential as surrogate predictor factors for asthma in early life.

The present work is devoted to study the pre-treatment of lignocellulosic biomass, especially wheat straw, by the application of the acidic ionic liquid (IL) such as 1-butyl-3-methylimidazolium hydrogen sulphate. The ability of this IL to hydrolysis and conversion of biomass was scrutinised. The pre-treatment with hydrogen sulphate-based IL allowed to obtain a liquor rich in hemicellulosic sugars, furans and organic acids, and a solid fraction mainly constituted by cellulose and lignin. Quantitative and qualitative analyses of the produced liquors were made by capillary electrophoresis and high-performance liquid chromatography. Pre-treatment conditions were set to produce xylose or furfural. Specific range of temperatures from 70 to 175 °C and residence times from 20.0 to 163.3 min were studied by fixing parameters such as biomass/IL ratio (10 % (w/w)) and water content (1.25 % (w/w)) in the pre-treatment process. Statistical modelling was applied to maximise the xylose and furfural concentrations. For the purpose of reaction condition comparison the severity factor for studied ionic liquid was proposed and applied in this work. Optimum conditions for xylose production were identified to be at 125 °C and 82.1 min, at which 16.7 % (w/w) xylose yield was attained. Furfural was preferably formed at higher pre-treatment temperatures and longer reaction time (161 °C and 104.5 min) reaching 30.7 % (w/w) maximum yield. The influence of water content on the optimum xylose formation was also studied. Pre-treatments with 5 and 10 % (w/w) water content were performed and an increase of 100 % and 140 % of xylose yield was observed, respectively, while the conversion into furfural maintained unchanged.

In this study, the larvicidal activity of dried leaf extracts from Pinus caribaea Morelet against Aedes aegypti was evaluated for the first time. Pinus caribaea extracts were obtained by macerating dried leaves in alkaline hydroethanol, ethanol and acetone solutions followed by evaporation under reduced pressure. The lignin content was quantified using the thioglycolic acid complexation method. Lethality bioassays (LC50 and LC90) were carried out in accordance with the recommendations of the World Health Organization. The results showed that the acetone extract from Pinus caribaea was more active, and that larvicidal activity was associated with lignin concentration.
