The use of in situ techniques to detect DNA and RNA sequences has proven to be an invaluable technique with paraffin-embedded tissue. Advances in non-radioactive detection systems have further made these procedures shorter and safer. We report the detection of Trypanosoma cruzi, the causative agent of Chagas disease, via indirect and direct in situ polymerace chain reaction within paraffin-embedded murine cardiac tissue sections. The presence of three T. cruzi specific DNA sequences were evaluated: a 122 base pair (bp) sequence localized within the minicircle network, a 188 bp satellite nuclear repetitive sequence and a 177 bp sequence that codes for a flagellar protein. In situ hybridization alone was sensitive enough to detect all three T. cruzi specific DNA sequences.

Afin de saisir la portée du débat nord-américain sur les communautarismes pour l'éthique théologique chrétienne actuelle, il paraît nécessaire de faire le point sur la différence entre le communautarisme des traditions intellectuelles, morales et spirituelles partagées, ou communautarisme éthico-religieux et éthico-politique, et le communautarisme politique. Le second, pour autant qu'il postule l'existence de communautés séparées au sein du politique, en est selon nous réduit à échouer comme modèle politique, aussi bien en ce qui concerne la démocratie à l'intérieur des États-nations (il est impossible de circonscrire des communautés distinctes stables en leur sein) qu'en ce qui touche les relations entre États-nations (qui ne constituent pas des communautés). D. Mûller prête ici attention à la valeur herméneutique constructive des traditions intellectuelles, morales et ou spirituelles, et plaide pour la fécondité partielle d'une telle approche lorsqu'il est question de construire une éthique théologique à visée vraiment universelle.

Research has demonstrated that landscape or watershed scale processes can influence instream aquatic ecosystems, in terms of the impacts of delivery of fine sediment, solutes and organic matter. Testing such impacts upon populations of organisms (i.e. at the catchment scale) has not proven straightforward and differences have emerged in the conclusions reached. This is: (1) partly because different studies have focused upon different scales of enquiry; but also (2) because the emphasis upon upstream land cover has rarely addressed the extent to which such land covers are hydrologically connected, and hence able to deliver diffuse pollution, to the drainage network However, there is a third issue. In order to develop suitable hydrological models, we need to conceptualise the process cascade. To do this, we need to know what matters to the organism being impacted by the hydrological system, such that we can identify which processes need to be modelled. Acquiring such knowledge is not easy, especially for organisms like fish that might occupy very different locations in the river over relatively short periods of time. However, and inevitably, hydrological modellers have started by building up piecemeal the aspects of the problem that we think matter to fish. Herein, we report two developments: (a) for the case of sediment associated diffuse pollution from agriculture, a risk-based modelling framework, SCIMAP, has been developed, which is distinct because it has an explicit focus upon hydrological connectivity; and (b) we use spatially distributed ecological data to infer the processes and the associated process parameters that matter to salmonid fry. We apply the model to spatially distributed salmon and fry data from the River Eden, Cumbria, England. The analysis shows, quite surprisingly, that arable land covers are relatively unimportant as drivers of fry abundance. What matters most is intensive pasture, a land cover that could be associated with a number of stressors on salmonid fry (e.g. pesticides, fine sediment) and which allows us to identify a series of risky field locations, where this land cover is readily connected to the river system by overland flow. In most initiatives to publish Open Educational Resources (OER), the production of OER is the activity with the highest costs. Based on literature and personal experiences a list of relevant characteristics of production processes for OER are determined. Three cases are compared with each other on these characteristics. Most influence on costs are human costs and the type of OER created.

Aims: 1) to create a new and reproducible animal model to produce heterotopic ossification (HO) 2) to be able to exactly quantify the amount of HO using a microCT scan and 3) to prove the hypothesis that COX-2 inhibitors are efficacious in the prevention of HO. Methods: We developed a IACUC-approved Lewis rat model, in which the ventral side of the right femur was scraped to mechanically disrupt the periosteum. By clamping the vastus intermedius ischemic injury to the muscle was produced to enhance HO. Finally homologous bone marrow from a donor rat was placed on the anterior surface of the femur. Half of the study group (8 rats) received chow mixed with a COX-2 inhibitor, while the other half received normal chow. After 6 weeks the animals were sacrificed, the femurs removed and imaged by microCT. Grading of HO was based on the thickness of ectopic bone as evaluated in a blinded fashion by 3 independent observers. Results: All animals developed bilateral HO. Rats treated with COX-2 inhibitors developed significantly less ectopic bone than the control group rats. Conclusions: The results suggest that we have created a very reliable, reproducible model to form ectopic bone in rats. Using the microCT we can precisely quantify the amount of HO. We have been able to show that COX-2 inhibitors significantly decrease the amount of HO formation and are thus a good alternative to non-specific NSAIDs with their potential serious side effects on the gastrointestinal tract and on hemo-stastis.

This retrospective study (April-September 2003) was designed to investigate the roles of the main viruses responsible for cases of acute infantile gastroenteritis in hospitalised children up to two years of age. The viruses were identified in 64.7% (88/136) of the cases and the detection rates of rotavirus A (RVA), norovirus (NoV) and astrovirus were 41.9% (57/136), 30.3% (24/79) and 12.7% (7/55), respectively. RVA and NoV were detected in 20 of the 24 reported nosocomial infection cases. This study identified the first circulation of the genotype NoV GII.21 in Brazil and highlights the need to establish differential diagnoses through active laboratorial surveillance.

In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

Estudi realitzat a partir d’una estada al Laboratoire d’études sur les monothéismes (UMR 8584, Centre national de la recherche scientifique / École pratique des hautes études / Université Paris IV-Sorbonne), França, entre 2010 i 2011. Anàlisi de la crisi estructural que afectà a l’església gal•la entre el darrer quart del segle IV i el primer del segle VI, crisi causada per la cristianització a gran escala de les elits aristocràtiques gal•loromanes i per la reivindicació per part d’aquest estament de la translació a l’esfera de la jerarquia institucional de l’Església de la seva preeminència econòmica i social. Aquest procés implicà l’aparició d’algunes interpretacions del “fet existencial cristià” que tractaven de legitimar en el plànol teòric la presa del control de les comunitats cristianes per part de la noblesa senatorial. En relació a aquest últim punt, s’ha donat particular rellevància a l’anomenada “controvèrsia semipelagiana” a Provença, amb especial èmfasi en dos punts: a) la relació entre l’oposició a la teologia agustiniana de la gràcia en alguns cercles monàstics provençals –Marsella, Lérins– i l’emergència en aquests ambients d’una literatura autobiogràfica en la que la reflexió sobre els conceptes de uocatio divina i conuersio a l’ascetisme cristià està estretament vinculada a un esforç teòric de redefinició i reorientació de l’ethos aristocràtic; i b) la relació entre els punts teològics debatuts en aquesta controvèrsia i les concepcions eclesiològiques dels pensadors que hi prengueren part –entengui’s aquí per eclesiologia la definició teòrica dels límits i dels fonaments de la “comunitat cristiana”, amb especial incidència en aquest cas en els plantejaments sobre el rol que l’aristòcrata havia d’exercir en aquestes noves comunitats “transversals”–. Aquest projecte bianual ha posat de manifest la inexistència d’una “teologia semipelagiana”, ateses les antagòniques concepcions eclesiològiques dels autors tradicionalment associats a aquesta corrent de pensament: Cassià entén la comunitat cristiana com una elit ascètica en la que els criteris “laics” d’estratificació social queden suspesos, i rebutja –en la teoria i en la pràctica– que aquesta elit hagi d’assumir el lideratge de la comunitat de fidels seglars; en els autors del cercle de Lérins, en canvi, l’oposició a la teologia agustiniana de la gràcia és inspirada per l’esforç d’importar a tota la comunitat cristiana els ideals monàstics, quelcom que fou també una via de legitimació de l’autoritat dels monjos-bisbes d’origen aristocràtic sorgits del cenobi de Lérins.

This Commentary draws together recently published work relating to the relationship between climate change and geomorphology to address the surprising observation that geomorphic work seems to have had little impact upon the work of the Intergovernmental Panel for Climate Change. However, recent papers show that methodological innovation has allowed geomorphological reconstruction over timescales highly relevant to late 20th century and 21st century climate change. In turn, these and other developments are allowing links to be made between climatic variability and geomorphology, to begin to predict geomorphic futures and also to appreciate the role that geomorphic processes play in the flux of carbon and the carbon cycle.

In this article we compare the images used by Dante Alighieri with the ones used by her“heretical” contemporary Marguerite Porete (†1310) for expressing the final contact-vision of God. We will analyze the use of the images of light, the gradual ascent and the knot in bothauthors, putting it into the context of the theological doctrines about the visio Dei. This will allow us, in the first place, to claim a shared cultural background based in the conception of God as a visible being and, in the second, to discuss the historical implications ofheteredoxy/ortodoxy in the Commedia and the Mirouer.
