La cuestión del asentamiento socialista en la URSS durante la década de 1920 estuvo caracterizada por el objetivo de definir y establecer un estado socialista en términos políticos, sociales y económicos. En este contexto de inestabilidad y cambio, un grupo de arquitectos pertenecientes a la Asociación de Arquitectos Contemporáneos, OSA, y liderado por Moisey Ginzburg, abordó el tema de la vivienda obrera asumiendo la responsabilidad y el compromiso por alcanzar un nuevo orden social. Su misión no consistió únicamente en solucionar el problema del alojamiento para los trabajadores en las grandes ciudades soviéticas, sino en redefinirlo como el marco adecuado para una sociedad sometida a un cambio sin precedentes que, al mismo tiempo y en un proceso dialéctico, debía contribuir a la construcción de esa nueva sociedad. La respuesta dada por la OSA trascendió el diseño inmediato bajo los estándares modernos establecidos en Occidente y tomó forma en un proceso de investigación que habría de prolongarse durante cinco años. Este trabajo, que culminó con la construcción y puesta en crisis de la Casa Narkomfin, se desarrolló en tres aproximaciones sucesivas. La primera, de carácter conceptual, consideró la participación ciudadana, así como de especialistas independientes, formalizándose en el Concurso entre Camaradas convocado por la OSA en 1926. La segunda aproximación al problema de la vivienda obrera se articuló a través de la investigación llevada a cabo por la Sección de Tipificación del Stroykom, esta vez desde premisas científicas y metodológicas. Finalmente, las conclusiones alcanzadas fueron transferidas a la práctica arquitectónica por medio de la construcción de seis Casas Experimentales de Transición, entre las que destacó la Casa Narkomfin. Este último acercamiento, de carácter empírico, ha sido tradicionalmente examinado por los expertos como un hecho aislado. Sin embargo, su estudio debe trascender necesariamente el genio del autor-creador en favor del proceso de investigación al que pertenece. En esta tesis, la Casa Narkomfin no se presenta sólo como el paradigma de vivienda soviética de vanguardia al que estamos acostumbrados, sino como un prototipo que recoge los principios y conclusiones alcanzados en las aproximaciones conceptuales y científicas precedentes. Únicamente desde este punto de vista cobra sentido la consideración de Ginzburg sobre su propio edificio como un medio propositivo y no impositivo: un proyecto concebido como una herramienta de transición hacia una sociedad más avanzada. ABSTRACT The question of mass housing in the USSR during the Twenties was marked by the drive to define and establish a socialist state in political, social and economic terms. In this context of instability and change, a group of architects gathered together under the Association of Contemporary Architects, OSA, led by Moisey Ginzburg, to address the issue of mass housing, thus taking on the responsibility and being committed to creating a new social order. Their quest not only involved solving the problem of housing for workers in large Soviet cities, but also redefining this solution as an appropriate framework for a society undergoing dramatic changes which, at the same time and in a dialectical process, would contribute to the creation of this new society. The solution provided by OSA transcended Modern standards of immediate design set by the West and was the result of a research process that would last five years. This work culminated in the construction of Narkomfin House and its self-criticism, developed in three successive approaches. The first was conceptual, being formalized in the Comradely Competition held by the OSA in 1926 and taking into account the participation of citizens and independent experts. The second approach to the problem of mass housing involved research developed by the Typification Section of the Stroykom, this time under scientific and methodological premises. Finally, the conclusions reached were put in practice with the construction of six Experimental Transitional Houses of which the most notable is Narkomfin House. This third empirical approach has traditionally been examined by scholars in isolation. However, its study must necessarily transcend the genius of the author-creator and involve the research process of which it is part. In this thesis, Narkomfin House is presented not only as the paradigm in Soviet housing avant-garde we are used to, but also as a prototype reflecting the principles and conclusions reached in the preceding conceptual and scientific approaches. Only from this point of view does Ginzburg’s understanding of his own building as a proactive and non-imposed environment make sense: a project conceived as a transition tool towards a more advanced society.

Esta dissertação de mestrado analisará a expressão grega ta. stoicei/a tou/ ko,smou, “os elementos do mundo”, que ocorre na carta de Colossenses nos versículos 8 e 20 do segundo capítulo. Será feito um estudo exegético na perícope bíblica 2.8-3.4 da referida carta, bem como uma análise histórica especificamente do termo stoicei/a. O estudo desta expressão é importante para poder se compreender a filosofia colossense mencionada em Cl 2.8. A igreja cristã na cidade de Colossos estava inserida em um contexto social religioso sincrético. Esse sincretismo é percebido claramente em textos de magia como os Papiros Mágicos Gregos, muito comuns na região da Ásia Menor, a mesma onde a igreja colossense estava situada. O sincretismo religioso, envolvendo crenças judaicas e pagãs, reflete as bases dessa filosofia. O autor da carta aos Colossenses refuta a crença nos “elementos do mundo”, bem como a subserviência aos mesmos. Dentre outras crenças, acreditava-se que esses “elementos” poderiam influenciar os acontecimentos sobre a terra e o destino das pessoas. Questões que envolvem práticas acéticas, adoração a anjos e observância de calendário litúrgico, dão os contornos dessa filosofia. O autor da carta enfatiza o senhorio de Cristo, bem como as obras dele em favor dos cristãos colossenses, que proporcionavam a eles, segurança quanto a terem um bom destino. E, além disso, é assegurada uma liberdade aos cristãos colossenses que não podia lhes ser cerceada por quaisquer outras crenças religiosas. Então, as obras de Cristo, bem como o seu senhorio, são os principais argumentos utilizados pelo autor da carta, a fim de afirmar aos cristãos em Colossos que eles não precisam mais temer o destino e nem se submeter aos “elementos do mundo”.

Human serum albumin (HSA) derivatized with cis-aconitic anhydride was covalently coupled to liposomes with a size of approximately 100 nm [polyaconitylated HSA (Aco-HSA) liposomes]. Within 30 min after injection into a rat, Aco-HSA liposomes were completely cleared from the blood and almost exclusively taken up by the liver, whereas in control liposomes 80% was still present in the blood at that time. Endothelial cells were shown to account for almost two-thirds of the hepatic uptake of the Aco-HSA liposomes, the remainder being recovered mainly in the liver macrophages (Kupffer cells). With fluorescently labeled liposomes it was shown that the Aco-HSA liposomes target a vast majority (>85%) of the cells in the endothelial cell population. Control liposomes were not taken up to a significant extent by the endothelial cells. Uptake of Aco-HSA liposomes by both endothelial and Kupffer cells was inhibited by preinjection with polyinosinic acid, indicating the involvement of scavenger receptors in the uptake process. The uptake of Aco-HSA liposomes by liver endothelial cells was dependent on liposome size; with increasing liposome diameter endothelial cell uptake decreased in favor of Kupffer cell uptake. We have demonstrated that massive in vivo targeting of liposomes to a defined cell population other than macrophages is possible. Aco-HSA liposomes thus may represent an attractive drug carrier system for treatment of various liver or liver endothelium-associated disorders.

The inhibition of β-galactosidase expression in a medium containing both glucose and lactose is a typical example of the glucose effect in Escherichia coli. We studied the glucose effect in the lacL8UV5 promoter mutant, which is independent of cAMP and cAMP receptor protein (CRP). A strong inhibition of β-galactosidase expression by glucose and a diauxic growth were observed when the lacL8UV5 cells were grown on a glucose–lactose medium. The addition of isopropyl β-d-thiogalactoside to the culture medium eliminated the glucose effect. Disruption of the crr gene or overproduction of LacY also eliminated the glucose effect. These results are fully consistent with our previous finding that the glucose effect in wild-type cells growing in a glucose–lactose medium is not due to the reduction of CRP–cAMP levels but is due to the inducer exclusion. We found that the glucose effect in the lacL8UV5 cells was no longer observed when either the crp or the cya gene was disrupted. Evidence suggested that CRP–cAMP may not enhance directly the lac repressor action in vivo. Northern blot analysis revealed that the mRNA for ptsG, a major glucose transporter gene, was markedly reduced in a Δcrp or Δcya background. The constitutive expression of the ptsG gene by the introduction of a multicopy plasmid restored the glucose effect in Δcya or Δcrp cells. We conclude that CRP–cAMP plays a crucial role in inducer exclusion, which is responsible for the glucose–lactose diauxie, by activating the expression of the ptsG gene.

In subjects suffering from early onset strabismus, signals conveyed by the two eyes are not perceived simultaneously but in alternation. We exploited this phenomenon of interocular suppression to investigate the neuronal correlate of binocular rivalry in primary visual cortex of awake strabismic cats. Monocularly presented stimuli that were readily perceived by the animal evoked synchronized discharges with an oscillatory patterning in the γ-frequency range. Upon dichoptic stimulation, neurons responding to the stimulus that continued to be perceived increased the synchronicity and the regularity of their oscillatory patterning while the reverse was true for neurons responding to the stimulus that was no longer perceived. These differential changes were not associated with modifications of discharge rate, suggesting that at early stages of visual processing the degree of synchronicity rather than the amplitude of responses determines which signals are perceived and control behavioral responses.

Antithrombin, a plasma serpin, is relatively inactive as an inhibitor of the coagulation proteases until it binds to the heparan side chains that line the microvasculature. The binding specifically occurs to a core pentasaccharide present both in the heparans and in their therapeutic derivative heparin. The accompanying conformational change of antithrombin is revealed in a 2.9-Å structure of a dimer of latent and active antithrombins, each in complex with the high-affinity pentasaccharide. Inhibitory activation results from a shift in the main sheet of the molecule from a partially six-stranded to a five-stranded form, with extrusion of the reactive center loop to give a more exposed orientation. There is a tilting and elongation of helix D with the formation of a 2-turn helix P between the C and D helices. Concomitant conformational changes at the heparin binding site explain both the initial tight binding of antithrombin to the heparans and the subsequent release of the antithrombin–protease complex into the circulation. The pentasaccharide binds by hydrogen bonding of its sulfates and carboxylates to Arg-129 and Lys-125 in the D-helix, to Arg-46 and Arg-47 in the A-helix, to Lys-114 and Glu-113 in the P-helix, and to Lys-11 and Arg-13 in a cleft formed by the amino terminus. This clear definition of the binding site will provide a structural basis for developing heparin analogues that are more specific toward their intended target antithrombin and therefore less likely to exhibit side effects.

The effect of different total enzyme concentrations on the flux through the bacterial phosphoenolpyruvate:carbohydrate phosphotransferase system (PTS) in vitro was determined by measuring PTS-mediated carbohydrate phosphorylation at different dilutions of cell-free extract of Escherichia coli. The dependence of the flux on the protein concentration was more than linear but less than quadratic. The combined flux–response coefficient of the four enzymes constituting the glucose PTS decreased slightly from values of ≈1.8 with increasing protein concentrations in the assay. Addition of the macromolecular crowding agents polyethylene glycol (PEG) 6000 and PEG 35000 led to a sharper decrease in the combined flux–response coefficient, in one case to values of ≈1. PEG 6000 stimulated the PTS flux at lower protein concentrations and inhibited the flux at higher protein concentrations, with the transition depending on the PEG 6000 concentration. This suggests that macromolecular crowding decreases the dissociation rate constants of enzyme complexes. High concentrations of the microsolute glycerol did not affect the combined flux–response coefficient. The data could be explained with a kinetic model of macromolecular crowding in a two-enzyme group-transfer pathway. Our results suggest that, because of the crowded environment in the cell, the different PTS enzymes form complexes that live long on the time-scale of their turnover. The implications for the metabolic behavior and control properties of the PTS, and for the effect of macromolecular crowding on nonequilibrium processes, are discussed.
