Reports about acquired resistance to colistin in different bacteria species are increasing, including E. coli of animal origin, but reports of resistance in wild S. enterica of different serotypes from swine are not found in the literature. Results obtained with one hundred and twenty-six E. coli strains from diseased swine and one hundred and twenty-four S. enterica strains from diseased and carrier swine showed a frequency of 6.3% and 21% of colistin-resistant strains, respectively. When comparing the disk diffusion test with the agar dilution test to evaluate the strains, it was confirmed that the disk diffusion test is not recommended to evaluate colistin resistance as described previously. The colistin MIC 90 and MIC 50 values obtained to E. coli were 0.25 mu g/mL and 0.5 mu g/mL, the MIC 90 and MIC 50 to S. enterica were 1 mu g/mL and 8 mu g/mL. Considering the importance of colistin in control of nosocomial human infections with Gram-negative multiresistant bacteria, and the large use of this drug in animal production, the colistin resistance prevalence in enterobacteriaceae of animal origin must be monitored more closely.

Los objetivos principales de esta Tesis Doctoral fueron estudiar en 4 ensayos los efectos a) del procesado del maíz y la inclusión en los piensos de ingredientes de alta calidad como harina de pescado o fuentes de lactosa en lechones blancos b) inclusión en el pienso de diferentes productos derivados del haba de soja, con diferente contenido de proteína bruta (PB), tamaño de partícula y origen en lechones blancos e ibéricos y c) inclusión en el pienso de lechones ibéricos de ingredientes de alta calidad; forma de presentación del pienso y la duración del suministro del pienso prestárter sobre los parámetros productivos, la digestibilidad de los nutrientes, y las características morfológicas de la mucosa digestiva en lechones blancos e ibéricos recién destetados. En el experimento 1, los efectos de la complejidad del pienso prestárter sobre los parámetros productivos y la digestibilidad total aparente (TTAD) de los nutrientes fueron estudiados en lechones blancos recién destetados. Se utilizaron 10 tratamientos experimentales como resultado de 5 piensos prestárter (21 a 41 d de edad) y 2 piensos estárter (42 a 62 d de edad). Los piensos prestárter consistieron en un control negativo que incluía 40% de maíz crudo, 4% de harina de pescado y 7% de lactosa, un control positivo que incluía 40% de maíz cocido, 10% de harina de pescado, y 14% de lactosa, y 3 piensos adicionales con similares ingredientes que el pienso control positivo pero en los que a) 40% de maíz cocido fue sustituido por el mismo porcentaje de maíz crudo, b) se redujo el nivel de harina de pescado del 10 al 4%, y c) se redujo el nivel de lactosa del 14 al 7%. Cada tratamiento se replicó 6 veces (6 lechones/departamento). De 42 a 62 d de edad, la mitad de cada uno de los 5 piensos prestárter recibió un pienso estándar compuesto por harina de soja- maíz crudo y manteca y la otra mitad un pienso con similar perfil nutricional pero incluyendo un 20% de maíz cocido, 5% de harina de pescado, 1.3% de lactosa, 2% de concentrado de proteína de soja obtenido por fermentación y 1% de aceite de soja en lugar de harina de soja, maíz sin procesar y manteca. La complejidad del pienso no afectó a los parámetros productivos en ninguno de los periodos estudiados, pero el índice de diarreas durante la fase prestárter fue mayor en los lechones que recibieron el pienso control negativo que en los alimentados con cualquiera de los otros piensos (P<0.05). A los 30 días de edad (piensos prestárter), la digestibilidad de la materia orgánica (MO) y de la energía bruta (EB) fue menor (P<0.001) en los lechones que consumieron el pienso control negativo que en los lechones que consumieron cualquiera de los otros piensos. Sin embrago, la digestibilidad fecal de la PB no fue afectada. A los 50 días de edad (piensos estárter), la digestibilidad de los nutrientes fue similar en ambos piesnsos. Se concluye que la utilización de niveles elevados de ingredientes de alta calidad en los piensos no mejora los parámetros productivos de los lechones blancos en ninguno de los períodos estudiados. De 21 a 41 días de edad, el índice de diarreas se redujo y la digestibilidad de los nutrientes aumentó con la utilización de piensos de mayor calidad. Por lo tanto, la utilización de piensos con niveles elevados de ingredientes de calidad para reducir problemas digestivos y por lo tanto, mejorar los parámetros productivos podría estar justificada en algunos casos. En el experimento 2, se estudiaron los efectos de la inclusión en el pienso de harina de soja con diferente contenido de PB (44 vs. 49 % PB), la micronización de la harina de soja de alta proteína (AP-HS; 49% PB) y la utilización de concentrado de proteína de soja (CPS; 65% PB) sobre los parámetros productivos y la TTAD de los nutrientes en lechones blancos recién destetados de 28 a 63 días de edad. De 28 a 49 días de edad (fase I), hubo un pienso control positivo con un 10% de CPS, un pienso control negativo con 14.8% de harina de soja estándar (R-HS; 44% de PB) y otros 4 piensos que incluían 13.3% de AP-HS de origen Americano (USA) o Argentino (ARG) y molidas groseramente (980 μm) o micronizadas (80 μm). Cada tratamiento se replicó 8 veces (6 lechones/departamento). De 49 a 63 días de edad (fase II), todos los lechones recibieron un pienso comercial común en forma de harina. En el global de la fase I, el tratamiento experimental no afectó a ninguno de los parámetros productivos estudiados. Sin embargo, de 28 a 35 días de edad, los lechones alimentados con AP-HS micronizadas tuvieron un mejor índice de conversión (IC; 1.11 vs. 0.98; P<0.05) que los alimentados con AP-HS molidas groseramente. También, de 35 a 42 días de edad, los lechones que recibieron el pienso con AP-HS micronizada tendieron (P=0.08) a consumir más pienso que los lechones que consumieron el pienso con AP-HS molida. Durante la fase II (49 a 63 días de edad), cuando todos los lechones recibieron un pienso común, no se observaron diferencias en productividad de los lechones debido al tratamiento previo. En general, la digestibilidad de los nutrientes a los 35 días de edad fue mayor para los lechones que consumieron CPS que para los lechones que consumieron R-HS con los lechones que consumieron AP-HS en una posición intermedia. La digestibilidad de la PB fue mayor (P≤0.01) para el pienso que contenía CPS que para el promedio de los 5 tratamientos en base a HS. También, la digestibilidad de la MO y de la materia seca (MS) fue mayor para el pienso que contenía AP-HS micronizada o molida groseramente que para el pienso que contenía R-HS. La micronización de la AP-HS no tuvo efecto alguno sobre la digestibilidad de los nutrientes. Se concluye que cuando el CPS sustituye en el pienso a R-HS, la digestibilidad de la PB aumenta pero no tiene efecto alguno sobre los parámetros productivos. La utilización de AP-HS en sustitución de R-HS en el pienso mejora la digestibilidad de los nutrientes pero no afecta a los parámetros productivos. La utilización de harina de soja micronizada en los piensos mejora la eficiencia alimenticia durante la primera semana post-destete pero no tiene efecto alguno sobre la digestibilidad de los nutrientes. En general, la inclusión de productos derivados del haba de soja con un alto valor añadido (CPS o AP-HS) en el pienso presenta pocas ventajas en términos productivos al uso de AP-HS en lechones blancos recién destetados. En el experimento 3, se estudiaron los mismos productos de soja y piensos similares al experimento 2 en lechones ibéricos recién destetados. Además de los parámetros productivos y la TTAD de los nutrientes, en este ensayo se estudió también la digestibilidad ileal aparente (AID) de los nutrientes, así como las características histológicas y morfometría de la mucosa ileal. Cada uno de los 6 tratamientos fue replicado 6 veces (6 lechones/departamento). De 30 a 51 días de edad la fuente de harina de soja no afectó a los parámetros productivos, pero el índice de diarreas fue mayor (P<0.001) y la TTAD y AID de los nutrientes menor en los lechones alimentados con R-HS que en los alimentados con CPS o AP-HS. Sin embargo, no se encontró ninguna diferencia para éstos parámetros entre los piensos que contenían AP-HS y CPS. La TTAD de la MO (P=0.07) y de la EB (P=0.05) tendieron a ser mayores en los piensos basados en AP-HS micronizada que en los basados en AP-HS molida. La TTAD de la EB tendió (P<0.05) a ser mayor para la AP-HS de origen USA que para la AP-HS de origen ARG. Los lechones que consumieron R-HS presentaron villi de menor longitud (P<0.01) que los lechones que consumieron AP-HS o CPS, pero no se observaron diferencias en el caso de los lechones que recibieron los piensos que contenían AP-HS o CPS. Se concluye que la inclusión de AP-HS o CPS en el pienso en sustitución de R-HS reduce el índice de diarreas y mejora la digestibilidad de los nutrientes y las características morfológicas del íleon sin afectar a los parámetros productivos. La utilización de piensos basados en productos derivados del haba de soja con mayor valor añadido (CPS o AP-HS) en sustitución de la R-HS, mejora la TTAD de todos los nutrientes y reduce el índice de diarreas si llegar afectar a los parámetros productivos. En el experimento 4 se estudiaron los efectos del contenido de PB y la complejidad del pienso, la presentación física y la duración del suministro del pienso prestárter sobre los parámetros productivos y la TTAD de los nutrientes en lechones ibéricos recién destetados de 28 a 63 días de edad. Hubo 12 tratamientos experimentales con 2 tipos de pienso (AC; calidad alta y BC: calidad media), 2 presentaciones del pienso (gránulo y harina) y 3 duraciones de suministro del pienso prestárter (7, 14 y 21 días). Desde los 7, 14 y 21 días de experimento (dependiendo del tratamiento), hasta los 35 días, todos los lechones recibieron un pienso comercial en forma de harina. Cada uno de los tratamientos fue replicado 3 veces (6 lechones/departamento). En el global del experimento, la ganancia media diaria (GMD; P<0.05) y el consumo medio diario (CMD; P<0.01) fue menor en los lechones que recibieron el pienso AC que para los que recibieron el pienso de BC, si bien el IC no se vio afectado. La granulación del pienso prestárter no afectó a los crecimientos pero mejoró la eficiencia alimenticia. La utilización del pienso prestárter de 0 a 21 días de prueba mejoró el IC (P<0.05), pero redujo la GMD (P<0.01) en comparación con la utilización de éste pienso solo durante 7 o 14 días. El índice de diarreas tendió a ser mayor (P=0.06) en los lechones alimentados con los piensos AC que en los alimentados con los piensos BC. Asimismo, el índice de diarreas fue superior en los lechones que recibieron el pienso en gránulo que los que los recibieron en harina (P<0.001). Además, el índice de diarreas fue superior en los lechones que recibieron el pienso prestárter durante 14 o 21 días que en los que lo recibieron solo durante 7 días (P<0.01). De 28 a 49 días de edad, la GMD y el IC no se vieron afectados por la complejidad del pienso, pero la presentación en gránulo o el aumento en la duración de suministro del pienso prestárter mejoró el IC (P<0.01). También, en este periodo el índice de diarreas fue mayor en lechones alimentados con piensos granulados que aquellos alimentados con piensos en harina. Asimismo, fue superior para los lechones alimentados con el pienso prestárter durante 14 o 21 días que para los que recibieron éste pienso solo durante 7 días (P<0.01). De 49 a 63 días de edad, los lechones que previamente habían recibido piensos BC crecieron más que los que recibieron piensos AC (P<0.001). Asimismo, los lechones que recibieron el pienso prestárter durante 21 días comieron (P< 0.001) y crecieron menos (P<0.05) presentando una peor eficacia alimenticia (P<0.05) que los lechones que lo recibieron solo durante 7 14 días. La digestibilidad de la MO fue mayor en los lechones alimentados con los piensos AC que en los alimentados con piensos BC (P<0.05). La granulación del pienso mejoró la digestibilidad de los principales nutrientes. Los piensos prestárter AC mejoraron la digestibilidad de los nutrientes pero no la eficiencia alimenticia en lechones ibéricos de 28 a 63 días de edad. La granulación del pienso mejoró la eficiencia alimenticia. El aumento del suministro del pienso prestárter de 7 a 21 días mejoró la eficiencia alimenticia pero redujo la GMD. Por lo tanto, la utilización de piensos granulados de alta calidad durante el periodo prestárter es recomendable en lechones ibéricos, pero solo durante la primera semana post-destete. ABSTRACT The main objectives of this PhD Thesis were to study the effects of a) heat processing (HP) of corn and inclusion of high quality ingredients of animal origin such as fish meal (FM) and dried milk products in the diet, b) inclusion of different soy products varying in crude protein (CP) content, particle size, and origin of the beans in diets for conventional white and Iberian weanling pigs, and c) effects of ingredient quality, feed form, and duration of supply of the phase I diets on growth performance, nutrient digestibility, and intestinal morphology of weanling pigs. In experiment 1, the effect of diet complexity on total tract apparent digestibility (TTAD) and growth performance was studied in piglets from 21 to 62 d of age. There were 10 experimental treatments which resulted from the combination of 5 phase I (21 to 41 d of age) and 2 phase II (42 to 62 d of age) diets. The 5 phase I diets consisted of a negative control diet that contained 40 % raw corn, 4% FM, and 7% lactose (LAC); a positive control diet that contained 40 % HP corn, 10% FM, and 14% LAC, and 3 extra diets that used similar ingredients to those of the positive control diet but in which a) 40% of HP corn was substituted by raw corn, b) 4% FM rather than 10% FM, and c) 7% LAC instead of 14% LAC were included in the diet. Each treatment was replicated 6 times (6 pigs per pen). From 42 to 62 d of age, half of the pens of each of the 5 phase I treatments received a standard soybean meal (SBM)–native corn–lard diet wheras the other half received a diet with similar nutrient profile but that included 20% HP corn, 5% FM, 1.3% lactosa, 2% fermented soy protein concentrate, and 1% soybean oil in substitution of variables amounts of non-processed corn, SBM, and lard. Dietary treatment did not affect piglet performance at any age, but the incidence of post-weaning diarrhea (PWD) was higher during phase I in piglets fed the negative control diet than in piglets fed any of the other diets (P<0.05). At 30 d of age (phase I diets), the TTAD of organic matter (OM) and gross energy (GE) was lower (P<0.001) in pigs fed the negative control diet than in pigs fed the other diets but CP digestibility was not affected. At 50 d of age (phase II diets), dietary treatment did not affect TTAD of any dietary component. It is concluded that the use of high quality ingredients at high levels in the diet did not improve growth performance of piglets at any age. From 21 to 41 d of age, PWD was reduced and nutrient digestibility was increased in pigs fed the more complex diets. Consequently, the inclusion of high levels of high quality ingredients in piglet diets to maximize growth performance might not be justified under all circumstances In experiment 2, the effect of CP content (44 vs. 49 % CP) of SBM, micronization (fine grinding) of the high CP SBM (HP-SBM; 49% CP), and soy protein concentrate (SPC; 65% CP) on TTAD and growth performance was studied in conventional white piglets from 28 to 63 d of age. From 28 to 49 d of age (phase I), there was a positive control diet that included 6.5% CP from SPC and a negative control diet that supplied the same amount of CP as regular SBM (R-SBM; 44% CP) of Argentina (ARG) origin. The other 4 diets included the same amount of dietary CP from 2 sources of HP-SBM (USA or ARG origin), either ground (990 μm) or micronized (60 μm). Each treatment was replicated 8 times (6 pigs per pen). From 49 to 63 d of age (phase II), all pigs were fed a common commercial starter diet. For the entire phase I, type of soy product included in the diet did not affect growth performance of the pigs. However, from 28 to 35 d of age pigs fed the micronized HP-SBM had better feed conversion ratio (FCR; 0.90 vs. 1.01; P<0.05) than pigs fed the ground HP-SBM. Also, from 35 to 42 d of age, average daily feed intake (ADFI) tended to be higher (P=0.08) for pigs fed the micronized HP-SBM than for pigs fed the ground HP-SBM. During phase II, when all the pigs received the same diet, no differences among treatments were observed. In general, the TTAD of nutrients at 35 d of age was higher for the SPC than for the R-SBM diet with the HP-SBM diets being intermediate. The TTAD of CP was higher (83.8% vs. 81.9%; P≤0.01) for the SPC diet than for the average of 5 SBM containing diets. Also, the digestibility of OM and dry matter (DM) was higher (P<0.01) for the HP-SBM, either ground or micronized, than for the R-SBM diet. Micronization of the HP-SBM did not affect nutrient digestibility. It is concluded that when R-SBM was substituted by SPC, CP digestibility was improved but no effects on growth performance were observed. The use of HP-SBM in substitution of R-SBM in the diet improved nutrient digestibility but did not affect piglet performance. The inclusion of micronized HP-SBM in the diet improved FCR during the first week post-weaning but did not affect TTAD of nutrients. Therefore, the inclusion of added value soy products (SPC or micronized SBM) in the diet presents little advantage in terms of growth performance over the use of HP-SBM in pigs weaned at 28 d of age. In experiment 3, the effects of the same sources of soy protein used in experiment 2 on TTAD and growth performance of crossbreed Iberian pigs from 30 to 61 d of age were studied. In addition, the apparent ileal digestibility (AID) of nutrients and mucosa ileum morphology were also determined. Dietary treatment did not affect growth performance of the pigs at any age but from 30 to 51 d of age (phase I diets), PWD was higher (P<0.001) and the TTAD and AID of all nutrients were lower for pigs fed the R-SBM diet than for pigs fed the HP-SBM or the SPC diets. However, no differences between the HP-SBM and the SPC containing diets were detected for digestibility of any dietary component. The TTAD of OM (P=0.07) and GE (P=0.05) tended to be higher for the micronized HP-SBM than for the ground HP-SBM and that of GE was higher (P<0.05) for the USA meal than for the ARG meal. Pigs fed R-SBM had lower villus height (P<0.01) than pigs fed HP-SBM or SPC but no differences in ileal mucosal morphology were detected between SPC and HP-SBM containing diets. It is concluded that feeding the HP-SBM or SPC in substitution of R-SBM reduced PWD and improved nutrient digestibility and ileal morphology in piglets as compared with feeding the R-SBM, but had no effect on growth performance. The inclusion in the diet of added value soy products (micronized SBM or SPC) in substitution of the R-SBM increased the TTAD of all nutrients and reduced PWD but had no advantage in terms of growth performance over the use of ground HP-SBM. In experiment 4, the effect of CP content and ingredient complexity, feed form, and duration of feeding of the phase I diets on growth performance and TTAD of nutrients were studied in Iberian pigs from 28 to 63 d of age. There were 12 dietary treatments with 2 type of feeds (HQ; higher quality and LQ; medium quality), 2 feed forms (pellets vs. mash), and 3 durations of supply (7, 14, and 21 d) of the phase I diets. From d 7, 14, or 21 (depending on treatment) to d 35 of experiment, all pigs received a common diet in mash form. Each treatment was replicated 3 times (6 pigs/pen). For the entire experiment, average daily gain (ADG; P<0.05) and ADFI (P<0.01) were lower with the HQ than with the LQ phase I diets but FCR was not affected. Pelleting of the phase I diets did not affect ADG but improved FCR (P<0.01). Feeding the phase I diets from d 0 to 21 improved FCR (P<0.05) but decreased ADG (P<0.01) as compared with 7 or 14 d of feeding. Post-weaning diarrhea tended to be higher (P=0.06) for pigs fed the HQ diets than for pigs fed the LQ diets and for pigs fed pellets than for pigs fed mash (P<0.001). Also, PWD was higher for pigs fed the phase I diet for 14 or 21 d than for pigs fed this diet for 7 d (P<0.01). From d 0 to 21, ADG and FCR were not affected by feed quality but feeding pellets or increasing the duration of feeding the phase I diets improved FCR (P<0.01). Also, in this period PWD was higher with pellets than with mash and for pigs fed the phase I diets for 14 or 21 d than for pigs fed this diet for only 7 d (P<0.01). From d 21 to 35, pigs previously fed the LQ diet had higher ADG than pigs fed the HQ phase I diets (P<0.001). Also, pigs that were fed the phase I diets for 21 d had lower ADG (P<0.05) and ADFI (P< 0.001) and poor FCR (P<0.05) than pigs fed these diets for 7 or 14 d. Organic matter digestibility was higher for pigs fed the HQ phase I diets than for pigs fed the LQ phase I diets (P<0.05). Pelleting improved TTAD of all nutrients (P<0.01). It is concluded that HQ phase I diets increased TTAD of nutrients but not feed efficiency of Iberian pigs from d 28 to 63 d of age. Also, pelleting improved nutrient digestibility and feed efficiency. Increasing the duration of supply of the phase I diets from 7 to 21 d improved feed efficiency but reduced ADG. Therefore, the use of LQ phase I diets in pellet form for no more than 7 d after weaning is recommended in Iberian pigs.

O presente trabalho foi desenvolvido para compor um modelo de educação ambiental no Jardim Botânico do Estado de São Paulo, localizado no Parque Estadual Fontes do Ipiranga (PEFI). Devido ao seu propósito educacional, o projeto educativo foi desenvolvido com o apoio de dois professores do ensino básico e de pesquisadores do Núcleo de Educação Ambiental do Jardim Botânico do Estado de São Paulo e das Seções de Ecologia e Ficologia do Instituto de Botânica. O modelo considerou quatro sistemas hídricos do PEFI, a Nascente do Riacho do Ipiranga e os Lagos do Jardim Botânico \"Nascentes, Ninféias e Bugios\", como base para a construção de instrumentos ordenadores de visitas monitoradas de professores do ensino básico e seus alunos ao Jardim Botânico. Como base do projeto, abordou-se o tema meio ambiente como transversal às disciplinas clássicas do sistema educacional brasileiro e empreenderam-se as ações do projeto através do conhecimento de diferentes profissionais sobre o meio ambiente que compuseram o grupo de trabalho e, ao mesmo, buscou-se integrar as ações às reais necessidades dos docentes do Ensino Básico. Nesse sentido, foi essencial a participação dos dois professores de escolas do entorno do Jardim Botânico nas etapas organizacionais do projeto. A caracterização da área de estudo baseou-se em seus aspectos históricos e físicos, bem como na qualidade sanitária dos corpos aquáticos considerados. As determinações físicas-químicas e microbiológicas revelaram uma condição sanitária satisfatória das águas, contudo os valores de coliformes fecais e totais encontrados nos lagos sugerem contaminação de origem animal, comum à região. A produtividade primária também foi determinada para cada corpo aquático, encontrando-se baixos valores de clorofila a nos três lagos e na nascente. Os tipos de algas microscópicas observadas foram os gêneros Chrysochromulina, Ankistrodesmus, Scenedesmus, Cosmarium, Navicula, Eunotia, Cryptomonas, Closterium, Gomphonema, Pinnularia, Pleurotaenium, Arthrospira, Oedogonium, Euastrum, Monoraphidium, Staurastrum, Pleurotaenium, Mallomonas e Chlamydomonas. De acordo com a tabela Carlson (Índice de Estado Trófico) a Nascente e o Lago das Nascentes são considerados oligotróficos, enquanto que o Lago dos Bugios e o das Ninféias, mesotróficos. A primeira ação do projeto, um curso teórico-prático, buscou-se integrar os professores do ensino básico em temas relacionados ao meio ambiente, particularmente naqueles de maior relevância aos profissionais atuantes no Jardim Botânico. Posteriormente, os professores participaram de ações para formatação de instrumentos educativos para visitação monitorada ao jardim, que resultaram na elaboração de um manual de visitação do professor, contendo um mapa da área de visitação e explicações de placas sinalizadoras. Para o núcleo de educação ambiental do Jardim Botânico foi disponibilizada a estrutura básica de um curso de capacitação de professores do ensino básico em temas sobre o meio ambiente, bem como a documentação fotográfica da área estudada, os critérios para a admissão de visita monitorada e os procedimentos para adequação do local da Nascente do Riacho do Ipiranga à visitação pública. Finalmente, deve-se ressaltar que o tema escolhido para a realização dessa dissertação de mestrado foi originado da percepção dos pesquisadores do Instituto de Botânica em destacar a importância do Jardim Botânico através de seus sistemas hídricos, e como poderiam ser utilizados como instrumento educativo nas escolas de São Paulo. Nesse sentido, optou-se por enriquecer o clássico foco dado aos jardins botânicos em todo o mundo, no qual a maioria dos visitantes apenas observa a vegetação local e exótica. Assim, pode-se afirmar que o trabalho empreendido se constituirá em uma contribuição ao professor com a missão de inserir a dimensão ambiental nas mais variadas disciplinas do ensino básico.

A utilização de pesticidas organoclorados é motivo de preocupação das entidades ligadas à área de saúde em todo o mundo. Apesar de as formas de contaminação serem bem conhecidas, não há um controle eficaz na fiscalização do seu uso no Brasil. Sabe-se que altos níveis séricos destes compostos nos organismos de seres humanos e animais acarretam sérios problemas de saúde. Tendo em vista essa realidade, foi realizado, em 2009, o Projeto Piloto do I Inquérito Nacional de Populações Expostas a Substâncias Químicas, cujo subprojeto \"doadores de sangue\" teve como objetivo mensurar as concentrações de substâncias químicas no sangue de 547 residentes da região metropolitana de São Paulo, dentre elas os pesticidas organoclorados. Este trabalho teve como objetivos avaliar as concentrações dos pesticidas hexaclorobenzeno (HCB), alfa-HCH, ?-HCH, beta-HCH, beta-HCH, heptacloro, heptacloro epóxido, dieldrin, mirex, o,p\'-DDT, p,p\'-DDT, o,p\'-DDE, p,p\'-DDE, o,p\'-DDD e p,p\'-DDD nesta população e compará-las com as encontradas em outros países e determinar fatores associados aos níveis mais elevados destas substâncias. O método analítico utilizado foi de cromatografia a gás. Os resultados deste estudo indicam que a população adulta de São Paulo não está exposta a níveis preocupantes de pesticidas organoclorados, pois dentre os compostos analisados, apenas o beta-HCH e o p,p\'-DDE tiveram um número significante de amostras acima do limite de quantificação, 10,7% e 31,2% das amostras respectivamente. Quando utilizada a metade do limite de quantificação para substituir os valores abaixo do limite de quantificação do método, o valor médio encontrado para o beta-HCH foi de 0,028 ug/dL e para o p,p\'-DDE foi de 0,045 ug/dL. Este estudo propôs dois modelos multivariados para explicar os fatores associados aos compostos beta-HCH e p,p\'-DDE no sangue dos doadores. Segundo o modelo de Regressão Logística Ordinal Multivariado, os fatores associados a níveis mais altos de beta-HCH foram ter idade entre 26 e 45 anos e ser do sexo feminino. Para o p,p\'-DDE os fatores associados a níveis mais altos foram ter idade entre 26 e 45 anos, ser do sexo feminino e ter trabalhado com pesticidas, enquanto receber renda mensal de 3 a 5 salários mínimos e consumir derivados de origem animal uma ou mais vezes por semana foram associados a níveis mais baixos de p,p\'-DDE. Segundo o modelo de Regressão Linear Múltipla, os fatores associados a níveis mais altos de beta-HCH foram o sexo feminino, ter contato prévio com pesticidas na região agrícola, ter trabalhado com pesticidas em campanhas de saúde pública, ter trabalhado em empresas de capacitores ou transformadores, ter trabalhado em indústrias de solventes clorados, ter renda mensal de 3 a 5 salários mínimos, consumo de carnes uma ou duas vezes por semana e consumo de frutos do mar uma ou duas vezes por semana, enquanto consumo frequente de cerveja e ter renda mensal de 1 a 3 salários mínimos foram associado a níveis menores de beta-HCH. Já para o p,p\'-DDE, os fatores associados a níveis mais elevados foram ser do sexo feminino, ser não branco, ter trabalhado com pesticidas e consumir água de fontes que não sejam minerais ou de rede, enquanto o consumo frequente de bebidas alcoólicas foi associado a níveis mais baixos de p,p\'-DDE

Lactic acid bacteria (LAB) play a key role in the biopreservation of a wide range of fermented food products, such as yogurt, cheese, fermented milks, meat, fish, vegetables (sauerkraut, olives and pickles), certain beer brands, wines and silage, allowing their safe consumption, which gave to these bacteria a GRAS (Generally Recognised as Safe) status. Besides that, the use of LAB in food and feed is a promising strategy to reduce the exposure to dietary mycotoxins, improving their shelf life and reducing health risks, given the unique mycotoxin decontaminating characteristic of some LAB. Mycotoxins present carcinogenic, mutagenic, teratogenic, neurotoxic and immunosuppressive effects over animals and Humans, being the most important ochratoxin A (OTA), aflatoxins (AFB1), trichothecenes, zearalenone (ZEA), fumonisin (FUM) and patulin. In a previous work of our group it was observed OTA biodegradation by some strains of Pediococcus parvulus isolated from Douro wines. So, the aim of this study was to enlarge the screening of the biodetoxification over more mycotoxins besides OTA, including AFB1, and ZEA. This ability was checked in a collection of LAB isolated from vegetable (wine, olives, fruits and silage) and animal (milk and dairy products, sausages) sources. All LAB strains were characterized phenotypically (Gram, catalase) and genotypically. Molecular characterisation of all LAB strains was performed using genomic fingerprinting by MSP- PCR with (GTG)5 and csM13 primers. The identification of the isolates was confirmed by 16S rDNA sequencing. To study the ability of LAB strains to degrade OTA, AFB1 and ZEA, a MRS broth medium was supplemented with 2.0 g/mL of each mycotoxin. For each strain, 2 mL of MRS supplemented with the mycotoxins was inoculated in triplicate with 109 CFU/mL. The culture media and bacterial cells were extracted by the addition of an equal volume of acetonitrile/methanol/acetic acid (78:20:2 v/v/v) to the culture tubes. A 2 mL sample was then collected and filtered into a clean 2 mL vial using PP filters with 0.45 m pores. The samples were preserved at 4 °C until HPLC analysis. Among LAB tested, 10 strains isolated from milk were able to eliminate AFB1, belonging to Lactobacillus casei (7), Lb. paracasei (1), Lb. plantarum (1) and 1 to Leuconostoc mesenteroides. Two strains of Enterococcus faecium and one of Ec. faecalis from sausage eliminated ZEA. Concerning to strains of vegetal origin, one Lb. plantarum isolated from elderberry fruit, one Lb. buchnerii and one Lb. parafarraginis both isolated from silage eliminated ZEA. Other 2 strains of Lb. plantarum from silage were able to degrade both ZEA and OTA, and 1 Lb. buchnerii showed activity over AFB1. These enzymatic activities were also verified genotypically through specific gene PCR and posteriorly confirmed by sequencing analysis. In conclusion, due the ability of some strains of LAB isolated from different sources to eliminate OTA, AFB1 and ZEA one can recognize their potential biotechnological application to reduce the health hazards associated with these mycotoxins. They may be suitable as silage inoculants or as feed additives or even in food industry.

The regional population growth in West Africa, and especially its urban centers, will bring about new and critical challenges for urban development policy, especially in terms of ensuring food security and providing employment for the growing population. (Peri-) urban livestock and vegetable production systems, which can contribute significantly to these endeavours, are limited by various constraints, amongst them limited access to expensive production factors and their (in)efficient use. To achieve sustainable production systems with low consumer health risks, that can meet the urban increased demand, this doctoral thesis determined nutrient use efficiencies in representative (peri-) urban livestock production systems in three West African cities, and investigated potential health risks for consumers ensuing from there. The field study, which was conducted during July 2007 to December 2009, undertook a comparative analysis of (peri-) urban livestock production strategies across 210 livestock keeping households (HH) in the three West African cities of Kano/Nigeria (84 HH), Bobo Dioulasso/Burkina Faso (63 HH) and Sikasso/Mali (63 HH). These livestock enterprises were belonging to the following three farm types: commercial gardening plus field crops and livestock (cGCL; 88 HH), commercial livestock plus subsistence field cropping (cLsC; 109 HH) and commercial gardening plus semi-commercial livestock (cGscL; 13 HH) which had been classified in a preceding study; they represented the diversity of (peri-) urban livestock production systems in West Africa. In the study on the efficiency of ruminant livestock production, lactating cowsand sheep herd units were differentiated based on whether feed supplements were offered to the animals at the homestead (Go: grazing only; Gsf: mainly grazing plus some supplement feeding). Inflows and outflows of nutrients were quantified in these herds during 18 months, and the effects of seasonal variations in nutrient availability on animals’ productivity and reproductive performance was determined in Sikasso. To assess the safety of animal products and vegetables, contamination sources of irrigated lettuce and milk with microbiological contaminants, and of tomato and cabbage with pesticide residues in (peri-) urban agriculture systems of Bobo Dioulasso and Sikasso were characterized at three occasions in 2009. Samples of irrigation water, organic fertilizer and ix lettuce were collected in 6 gardens, and samples of cabbage and tomato in 12 gardens; raw and curdled milk were sampled in 6 dairy herds. Information on health risks for consumers of such foodstuffs was obtained from 11 health centers in Sikasso. In (peri-) urban livestock production systems, sheep and goats dominated (P<0.001) in Kano compared to Bobo Dioulasso and Sikasso, while cattle and poultry were more frequent (P<0.001) in Bobo Dioulasso and Sikasso than in Kano. Across cities, ruminant feeding relied on grazing and homestead supplementation with fresh grasses, crop residues, cereal brans and cotton seed cake; cereal grains and brans were the major ingredients of poultry feeds. There was little association of gardens and livestock; likewise field cropping and livestock were rarely integrated. No relation existed between the education of the HH head and the adoption of improved management practices (P>0.05), but the proportion of HH heads with a long-term experience in (peri-) urban agriculture was higher in Kano and in Bobo Dioulasso than in Sikasso (P<0.001). Cattle and sheep fetched highest market prices in Kano; unit prices for goats and chicken were highest in Sikasso. Animal inflow, outflow and dairy herd growth rates were significantly higher (P<0.05) in the Gsf than in the Go cattle herds. Maize bran and cottonseed expeller were the main feeds offered to Gsf cows as dry-season supplement, while Gsf sheep received maize bran, fresh grasses and cowpea pods. The short periodic transhumance of Go dairy cows help them maintaining their live weight, whereas Gsf cows lost weight during the dry season despite supplement feeding at a rate of 1506 g dry matter per cow and day, resulting in low productivity and reproductive performance. The daily live weight gains of calves and lambs, respectively, were low and not significantly different between the Go and the Gsf system. However, the average live weight gains of lambs were significantly higher in the dry season (P<0.05) than in the rainy season because of the high pressure of gastrointestinal parasites and of Trypanosoma sp. In consequence, 47% of the sheep leaving the Go and Gsf herds died due to diseases during the study period. Thermo-tolerant coliforms and Escherichia coli contamination levels of irrigation water significantly exceeded WHO recommendations for the unrestricted irrigation of vegetables consumed raw. Microbial contamination levels of lettuce at the farm gate and the market place in Bobo Dioulasso and at the farm gate in Sikasso were higher than at the market place in Sikasso (P<0.05). Pesticide residues were detected in only one cabbage and one tomato sample and were below the maximum residue limit for consumption. Counts of thermo-tolerant coliforms and Escherichia coli were higher in curdled than in raw milk (P<0.05). From 2006 to x 2009, cases of diarrhea/vomiting and typhoid fever had increased by 11% and 48%, respectively, in Sikasso. For ensuring economically successful and ecologically viable (peri-) urban livestock husbandry and food safety of (peri-) urban foodstuffs of animal and plant origin, the dissemination and adoption of improved feeding practices, livestock healthcare and dung management are key. In addition, measures fostering the safety of animal products and vegetables including the appropriate use of wastewater in (peri-) urban agriculture, restriction to approve vegetable pesticides and the respect of their latency periods, and passing and enforcement of safety laws is required. Finally, the incorporation of environmentally sound (peri-) urban agriculture in urban planning by policy makers, public and private extension agencies and the urban farmers themselves is of utmost importance. To enable an efficient (peri-) urban livestock production in the future, research should concentrate on cost-effective feeding systems that allow meeting the animals’ requirement for production and reproduction. Thereby focus should be laid on the use of crop-residues and leguminous forages. The improvement of the milk production potential through crossbreeding of local cattle breeds with exotic breeds known for their high milk yield might be an accompanying option, but it needs careful supervision to prevent the loss of the local trypanotolerant purebreds.

The interest in animal welfare and welfare-friendly food products has been increasing in Europe over the last 10 years. The media, highlighting traditional farming methods and food scares such as those related to salmonella, bovine spongiform encephalopathy/variant Creutzfeldt-Jakob disease (BSE) and avian influenza, have brought the methods of animal farming to public attention. Concerns about farm animal welfare are reflected in the increase in the number of vegetarians and vegans and an increase in consumers wishing to purchase food which is more animal welfare-friendly. This paper considers consumers’ attitudes to animal welfare and to marketing practices, such as product labelling, welfare grading systems and food assurance marks using comparative data collected in a survey of around 1500 consumers in each of Great Britain, Italy and Sweden as part of the EU-funded Welfare Quality research project. The findings suggest a need for the provision of improved consumer information on the welfare provenance of food using appropriate product labelling and other methods.

Methicillin-resistant Staphylococcus aureus (MRSA) is one of the most important bacterial pathogens based on its incidence and the severity of its associated infections. In addition, severe MRSA infections can occur in hospitalised patients or healthy individuals from the community. Studies have shown the infiltration of MRSA isolates of community origin into hospitals and variants of hospital-associated MRSA have caused infections in the community. These rapid epidemiological changes represent a challenge for the molecular characterisation of such bacteria as a hospital or community-acquired pathogen. To efficiently control the spread of MRSA, it is important to promptly detect the mecA gene, which is the determinant of methicillin resistance, using a polymerase chain reaction-based test or other rapidly and accurate methods that detect the mecA product penicillin-binding protein (PBP)2a or PBP2’. The recent emergence of MRSA isolates that harbour a mecA allotype, i.e., the mecC gene, infecting animals and humans has raised an additional and significant issue regarding MRSA laboratory detection. Antimicrobial drugs for MRSA therapy are becoming depleted and vancomycin is still the main choice in many cases. In this review, we present an overview of MRSA infections in community and healthcare settings with focus on recent changes in the global epidemiology, with special reference to the MRSA picture in Brazil.

Genetic background, prenatal and post-natal early-life conditions influence the development of interconnected physiological systems and thereby shape the phenotype. Certain combinations of genotypes and pre- and post-natal conditions may provide higher fitness in a specific environmental context. Here, we investigated how grey partridges Perdix perdix of two strains (wild and domesticated) cope physiologically with pre- and post-natal predictable vs. unpredictable food supply. Food unpredictability occurs frequently in wild environments and requires physiological and behavioural adjustments. Well-orchestrated and efficient physiological systems are presumably more vital in a wild environment as compared to captivity. We thus predicted that wild-strain grey partridges have a stronger immunity, glucocorticoid (GC) stress response and oxidative stress resistance (OSR) than domesticated birds, which have undergone adaptations to captivity. We also predicted that wild-strain birds react more strongly to environmental stimuli and, when faced with harsh prenatal conditions, are better able to prepare their offspring for similarly poor post-natal conditions than birds of domesticated origin. We found that wild-strain offspring were physiologically better prepared for stressful situations as compared to the domesticated strain. They had a high GC stress response and a high OSR when kept under predictable food supply. Wild-strain parents reacted to prenatal unpredictable food supply by lowering their offspring's GC stress response, which potentially lowered GC-induced oxidative pressure. No such pattern was evident in the domesticated birds. Irrespective of strain and prenatal feeding scheme, post-natal unpredictable food supply boosted immune indices, and GC stress response was negatively related to antibody response in females and to mitochondrial superoxide production. Wild-strain grey partridge showed fitness-relevant physiological advantages and appeared to prepare their offspring for the prospective environment. Negative relationships between GC stress response, immunity and oxidative indices imply a pivotal role of an organism's oxidative balance and support the importance of considering multiple physiological systems simultaneously.

From 2003-2006, an EU network project ‘Sustaining Animal Health and Food Safety in Organic Farming' (SAFO), was carried out with 26 partners from 20 EU-countries and 4 related partners from 4 candidate or new member states. The focus was the integration of animal health and welfare issues in organic farming with food safety aspects. Four very consistent conclusions became apparent: 1) The climatic, physical and socio-economic conditions vary considerably throughout Europe, leading to different livestock farming systems. This limits the possibility for technology transfer between regions, and creates several challenges for a harmonised regulation, 2) Implementing organic standards at farm level does not always ensure that animal health and welfare reach the high ideals of the organic principles, 3) To overcome these deficiencies, organic farmers and farmer organisations need to take ownership of organic values and, 4) In all participating countries, a strong need for training of farmers and in particular veterinarians in animal health promotion and organic principles was identified. The article presents a summary of papers presented at the five SAFO workshops.

Over the past decade genomic approaches have begun to revolutionise the study of animal diversity. In particular, genome sequencing programmes have spread beyond the traditional model species to encompass an increasing diversity of animals from many different phyla, as well as unicellular eukaryotes that are closely related to the animals. Whole genome sequences allow researchers to establish, with reasonable confidence, the full complement of any particular family of genes in a genome. Comparison of gene complements from appropriate genomes can reveal the evolutionary history of gene families, indicating when both gene diversification and gene loss have occurred. More than that, however, assembled genomes allow the genomic environment in which individual genes are found to be analysed and compared between species. This can reveal how gene diversification occurred. Here, we focus on the Fox genes, drawing from multiple animal genomes to develop an evolutionary framework explaining the timing and mechanism of origin of the diversity of animal Fox genes. Ancient linkages between genes are a prominent feature of the Fox genes, depicting a history of gene clusters, some of which may be relevant to understanding Fox gene function.

O objetivo do trabalho foi efetuar a comparação das principais espécies de interesse agropecuário, em relação à eficiência de conversão das dietas em produtos de origem animal (carne ou ovo), produção de resíduo e potencial de emissão de metano, a partir da fermentação dos resíduos. Para isso, foram selecionadas cinco espécies de animais durante a fase de produção: 1) suínos, do nascimento ao abate (peso vivo (PV) final de 90 kg) ; 2) bovinos, do desmame ao abate (PV: 520 kg); 3) caprinos, do desmame ao abate (PV: 30 kg); 4) aves, durante toda a fase de postura (14,7 kg de ovos); 5) frango de corte, do nascimento ao abate (PV: 3,1 kg). Para a estimativa dos parâmetros propostos, foram medidos os dados do desempenho e efetuou-se a biodigestão anaeróbia dos dejetos produzidos pelos animais. de maneira geral, os dejetos dos ruminantes apresentaram altas concentrações de fibra e baixos potenciais de produção de biogás; no entanto, o menor desempenho destes animais na conversão do alimento em produto e a maior produção de dejetos fizeram com que eles apresentassem maiores produções de metano por kg de alimento produzido.

The habitat of the mycelial saprobic form of Paracoccidio ides brasiliensis, which produces the infectious propagula, has not been determined and has proven difficult for mycologists to describe. The fungus has been rarely isolated from the environment, the disease has a prolonged latency period and no outbreaks have been reported. These facts have precluded the adoption of preventive measures to avoid infection. The confirmation of natural infections in nine-banded armadillos (Dasypus novemcinctus) with P. brasiliensis, in high frequency and wide geographic distribution, has opened new avenues for the study and understanding of its ecology. Armadillos belong to the order Xenarthra, which has existed in South America ever since the Paleocene Era (65 million years ago), when the South American subcontinent was still a detached land, before the consolidation of what is now known as the American continent. on the other hand, strong molecular evidence suggests that P. brasiliensis and other dimorphic pathogenic fungi - such as Blastomyces dermatitidis, Coccidioides immitis and Histoplasma capsulatum - belong to the family Onygenaceae sensu Into (order Onygenales, Ascomycota), which appeared around 150 million years ago.P. brasiliensis ecology and relation to its human host are probably linked to the fungal evolutionary past, especially its long coexistence with and adaptation to animal hosts other than Homo sapiens, of earlier origin. Instead of being a blind alley, the meaning of parasitism for dimorphic pathogenic fungi should be considered as an open two-way avenue, in which the fungus may return to the environment, therefore contributing to preserve its teleomorphic (sexual) and anamorphic (asexual) forms in a defined and protected natural habitat.
