The marine environment is constantly affected by anthropic actions, with causes consequent degradation of the waters and marine biota by various discharges of xenobiotics. In the present study, the focus was the study of a region of the marine coast of the State of So Paulo (city of Santos), which is one of the most industrialized parts of Brazil and suffers also from a strong impact of domestic effluents. The mussel Perna perna, very abundant in the coast of the State of So Paulo, Brazil, was selected as the biomonitoring organism for the determination of inorganic elements and a passive biomonitoring was performed. The organisms were collected at two sites in So Paulo State coast: Cocanha beach in Caraguatatuba (mussel farm) and Santos Bay (Itaipu and Palmas). Seasonally, the Perna perna were collected between September/08 and July/09 in each study sites. After removal and sample preparation, the elements As, Co, Cr, Fe, Se and Zn were determined by instrumental neutron activation analysis (INAA) and Cd, Pb and Hg were determined by atomic absorption spectroscopy (AAS) in this organism.

Intranasal inoculation of equid herpesvirus type-1 (EHV-1) Brazilian strains A4/72 and A9/92 induced an acute and lethal infection in four different inbred mouse strains. Clinical and neurological signs appeared between the 2nd and 3rd day post inoculation (dpi) and included weight loss, ruffled fur, a hunched posture, crouching in corners, nasal and ocular discharges, dyspnoea, dehydration and increased salivation. These signs were followed by increased reactivity to external stimulation, seizures, recumbency and death. The virus was recovered consistently from the brain and viscera of all mice with neurological signs. Histopathological changes consisted of leptomeningitis, focal haemorrhage, ventriculitis, neuronal degeneration and necrosis, neuronophagia, non-suppurative inflammation, multifocal gliosis and perivascular infiltration of polymorphonuclear and mononuclear cells. Immunohistochemical examination demonstrated that EHV-1 strains A4/72 and A9/92 replicated in neurons of the olfactory bulb, the cortex and the hippocampus. In contrast, mice inoculated with the EHV-1 Brazilian strain A3/97 showed neither weight loss nor apparent clinical or neurological signs; however, the virus was recovered consistently from their lungs at 3 dpi. These three EHV-1 strains showed distinct degrees of virulence and tissue tropism in mice. EHV-1 strains A4/72 and A9/92 exhibited a high degree of central nervous system tropism with neuroinvasion and neurovirulence. EHV-1 strain A3/97 was not neurovirulent despite being detected in the brains of infected BALB/c nude mice. These findings indicate that several inbred mouse strains are susceptible to neuropathogenic EHV-1 strains and should be useful models for studying the pathogenesis and mechanisms contributing to EHV-induced myeloencephalopathy in horses. We review symplectic nontwist maps that we have introduced to describe Lagrangian transport properties in magnetically confined plasmas in tokamaks. These nontwist maps are suitable to describe the formation and destruction of transport barriers in the shearless region (i.e., near the curve where the twist condition does not hold). The maps can be used to investigate two kinds of problems in plasmas with non-monotonic field profiles: the first is the chaotic magnetic field line transport in plasmas with external resonant perturbations. The second problem is the chaotic particle drift motion caused by electrostatic drift waves. The presented analytical maps, derived from plasma models with equilibrium field profiles and control parameters that are commonly measured in plasma discharges, can be used to investigate long-term transport properties. The study was conducted at the Research Laboratory of Hydraulic and Irrigation Group in the Rural Engineering Department, Technical University of Madrid (Universidad Politecnica de Madrid), Madrid, Spain. Water temperatures of 20, 30, 40 degrees C and system pressures often encountered in irrigation practices of 80, 90, 100, 110, 120, 130, 140, 150, 160, 170, 180, 190 and 200 k Pa were applied to determine the effects of different water temperatures and pressures on emitter discharge. Non-pressure compensating in-line emitter which has turbulent flow regime with a long-path (labyrinth), emitter discharge was 4 L h(-1) at system pressure of 100 kPa according to the manufacturer recommended, was used. Emitters were spaced 20 cm along the drip laterals with 16 mm diameter. Discharge equations and coefficients of variation related to temperatures of 20, 30 and 40 degrees C were obtained as q = 0.375H(0.51), q = 0.358H(0.52), q = 0.346H(0.53) and 2.68, 2.09, 3.65, respectively. Discharge of the emitter was affected by different system pressures and increased as potentially (R = 0993-0996). In general. the emitter discharge increased with increasing temperature. However, especially in the common system pressures of 90-120 k Pa, differences of obtained emitter discharges between the different water temperatures were not significant (1%).

BECTS represents the vast majority of childhood focal epilepsy. Owing to the age peculiarity of children who suffer from this disease, i.e., school-going age of between 6 and 9 years, the condition is often referred to as a school disorder by parents and teachers. Objective: The aim of this study was to evaluate the academic performance of children with BED'S, according to the clinical and electroencephalographic ILAE criteria, and compare the results of neuropsychological tests of language and attention to the frequency of epileptic discharges. Methods: The performances of 40 school children with BED'S were evaluated by applying a school performance test (SBT), neuropsychological tests (WISC and Trail-Making), and language tests (Illinois Test Psycholinguistic Abilities - ITPA - and Staggered Spondaic Word - SSW). The same tests were applied in the control group. Results: Children with BED'S, when compared to those in the control group, showed lower scores in academic performance (SPT), digits and similarities subtests of WISC, auditory processing subtest of SSW, and ITPA - representational and automatic level. The study showed that epileptic discharges did not influence the results. Conclusion: Children with BED'S scored significantly lower scores in tests on academic performance, when compared with those in the control group probably due to executive dysfunction. Recent reports have shown an increase in potentially harmful phytoplankton in Santos bay (Southeastern Brazilian Coast), located in a highly urbanised estuarine complex. Prediction of blooms is, thus, essential but the phytoplankton community structure in very dynamic regions is difficult to determine. In the present work, we discriminate bloom forming microphytoplankton dominance and their relationship to physical and meteorological variables to look for patterns observed in different tides and seasons. Comparing 8 distinct situations, we found five scenarios of dominance that could be related to winds, tides and rainfall: i) Surfers, diatoms occurring during high surf zone energies; ii) Sinkers, represented by larger celled diatoms during spring tide, after periods of high precipitation rates; iii) Opportunistic mixers, composed of chain forming diatoms with small or elongate cells occurring during neap tides; iv) Local mixers, microplanktonic diatoms and dinoflagellates which occurred throughout the 298 sampling stations; and v) Mixotrophic dinoflagellates, after intense estuarine discharges. Results suggest alterations in the temporal patterns for some bloom-forming species, while others appeared in abundances above safe limits for public health. This approach can also illustrate possible impacts of changes in freshwater discharge in highly urbanised estuaries.

Estuaries are extremely dynamic environments that are vulnerable to anthropogenic alterations. Thus, monitoring phytoplankton abundances and composition is an essential tool for the prediction of eutrophication and its effects on coastal ecosystems. Phytoplankton biomass, as chlorophyll-a, in the São Vicente estuary (Brazil) varies in response to tidal cycles and seasonal rainfall. Objectives. To present two datasets designed to assess the relationship between chlorophyll-a and changes in water turbidity driven by tide and rain. Methods. Weekly observations were made in the shallow embayment (February to September 2008; site 1) and observations recorded on alternate days (summer 2010, site 2). Results. At site 1, turbidity differed between high and low tides, but on most days was over 3000 RU, maintaining moderate chlorophyll-a levels (4 mg.m-3) and only two blooms developed during low turbidity. Site 2 mean turbidity was 1500 RU, nutrient level was higher during neap tides and phytoplankton blooms were mainly observed at the end of neap tides at 15-day intervals, dominated by chain-forming diatoms and occasionally flagellates and pennate diatoms. Conclusions. Taxonomic composition of the blooms was different and their frequency altered by events characterized by intense freshwater discharges from the Henry Borden Hydroelectric Dam (> 9*106.m³), inhibiting phytoplankton accumulation during neap tide periods.

The spectral reflectance of the sea surface recorded using ocean colour satellite sensors has been used to estimate chlorophyll-a concentrations for decades. However, in bio-optically complex coastal waters, these estimates are compromised by the presence of several other coloured components besides chlorophyll, especially in regions affected by low-salinity waters. The present work aims to (a) describe the influence of the freshwater plume from the La Plata River on the variability of in situ remote sensing reflectance and (b) evaluate the performance of operational ocean colour chlorophyll algorithms applied to Southwestern Atlantic waters, which receive a remarkable seasonal contribution from La Plata River discharges. Data from three oceanographic cruises are used, in addition to a historical regional bio-optical dataset. Deviations found between measured and estimated concentrations of chlorophyll-a are examined in relation to surface water salinity and turbidity gradients to investigate the source of errors in satellite estimates of pigment concentrations. We observed significant seasonal variability in surface reflectance properties that are strongly driven by La Plata River plume dynamics and arise from the presence of high levels of inorganic suspended solids and coloured dissolved materials. As expected, existing operational algorithms overestimate the concentration of chlorophyll-a, especially in waters of low salinity (S<33.5) and high turbidity (Rrs(670)>0.0012 sr−1). Additionally, an updated version of the regional algorithm is presented, which clearly improves the chlorophyll estimation in those types of coastal environment. In general, the techniques presented here allow us to directly distinguish the bio-optical types of waters to be considered in algorithm studies by the ocean colour community.

In order to protect river water quality, highly affected in urban areas by continuos as intermittent immissions, it is necessary to adopt measures to intercept and treat these polluted flows. In particular during rain events, river water quality is affected by CSOs activation. Built in order to protect the sewer system and the WWTP by increased flows due to heavy rains, CSOs divert excess flows to the receiving water body. On the basis of several scientific papers, and of direct evidences as well, that demonstrate the detrimental effect of CSOs discharges, also the legislative framework moved towards a stream standard point of view. The WFD (EU/69/2000) sets new goals for receiving water quality, and groundwater as well, through an integrated immission/emissions phylosophy, in which emission limits are associated with effluent standards, based on the receiving water characteristics and their specific use. For surface waters the objective is that of a “good” ecological and chemical quality status. A surface water is defined as of good ecological quality if there is only slight departure from the biological community that would be expected in conditions of minimal anthropogenic impact. Each Member State authority is responsible for preparing and implementing a River Basin Management Plan to achieve the good ecological quality, and comply with WFD requirements. In order to cope with WFD targets, and thus to improve urban receiving water quality, a CSOs control strategy need to be implemented. Temporarily storing the overflow (or at least part of it) into tanks and treating it in the WWTP, after the end of the storm, showed good results in reducing total pollutant mass spilled into the receiving river. Italian State Authority, in order to comply with WFD statements, sets general framework, and each Region has to adopt a Water Remediation Plan (PTA, Piano Tutela Acque), setting goals, methods, and terms, to improve river water quality. Emilia Romagna PTA sets 25% reduction up to 2008, and 50% reduction up to 2015 fo total pollutants masses delivered by CSOs spills. In order to plan remediation actions, a deep insight into spills dynamics is thus of great importance. The present thesis tries to understand spills dynamics through a numerical and an experimental approach. A four months monitoring and sampling campaign was set on the Bologna sewer network, and on the Navile Channel, that is the WWTP receiving water , and that receives flows from up to 28 CSOs during rain events. On the other hand, the full model of the sewer network, was build with the commercial software InfoWorks CS. The model was either calibrated with the data from the monitoring and sampling campaign. Through further model simulations interdependencies among masses spilled, rain characteristics and basin characteristics are looked for. The thesis can be seen as a basis for further insighs and for planning remediation actions.

Many efforts have been devoting since last years to reduce uncertainty in hydrological modeling predictions. The principal sources of uncertainty are provided by input errors, for inaccurate rainfall prediction, and model errors, given by the approximation with which the water flow processes in the soil and river discharges are described. The aim of the present work is to develop a bayesian model in order to reduce the uncertainty in the discharge predictions for the Reno river. The ’a priori’ distribution function is given by an autoregressive model, while the likelihood function is provided by a linear equation which relates observed values of discharge in the past and hydrological TOPKAPI model predictions obtained by the rainfall predictions of the limited-area model COSMO-LAMI. The ’a posteriori’ estimations are provided throw a H∞ filter, because the statistical properties of estimation errors are not known. In this work a stationary and a dual adaptive filter are implemented and compared. Statistical analysis of estimation errors and the description of three case studies of flood events occurred during the fall seasons from 2003 to 2005 are reported. Results have also revealed that errors can be described as a markovian process only at a first approximation. For the same period, an ensemble of ’a posteriori’ estimations is obtained throw the COSMO-LEPS rainfall predictions, but the spread of this ’a posteriori’ ensemble is not enable to encompass observation variability. This fact is related to the building of the meteorological ensemble, whose spread reaches its maximum after 5 days. In the future the use of a new ensemble, COSMO–SREPS, focused on the first 3 days, could be helpful to enlarge the meteorogical and, consequently, the hydrological variability.

Die Ziele der vorliegenden Arbeit waren 1) die Entwicklung und Validierung von sensitiven und substanz-spezifischen Methoden für die quantitative Bestimmung von anionischen, nichtionischen und amphoteren Tensiden und deren Metaboliten in wässrigen Umweltproben unter Einsatz leistungsfähiger, massenspektrometrischer Analysengeräte,2) die Gewinnung von aeroben, polaren Abbauprodukten aus Tensiden in einem die realen Umweltbedingungen simulierenden Labor-Festbettbioreaktor (FBBR), dessen Biozönose oberflächenwasserbürtig war,3) zur Aufklärung des Abbaumechanismus von Tensiden neue, in 2) gewonnene Metabolite zu identifizieren und massenspektrometrisch zu charakterisieren ebenso wie den Primärabbau und den weiteren Abbau zu verfolgen,4) durch quantitative Untersuchungen von Tensiden und deren Abbauprodukten in Abwasser und Oberflächenwasser Informationen zu ihrem Eintrag und Verhalten bei unterschiedlichen hydrologischen und klimatischen Bedingungen zu erhalten,5) das Verhalten von persistenten Tensidmetaboliten in Wasserwerken, die belastetes Oberflächenwasser aufbereiten, zu untersuchen und deren Vorkommen im Trinkwasser zu bestimmen,6) mögliche Schadwirkungen von neu entdeckten Metabolite mittels ökotoxikologischer Biotests abzuschätzen,7) durch Vergleich der Felddaten mit den Ergebnissen der Laborversuche die Umweltrelevanz der Abbaustudien zu belegen. Die Auswahl der untersuchten Verbindungen erfolgte unter Berücksichtigung ihres Produktionsvolumens und der Neuheit auf dem Tensidmarkt. Sie umfasste die Waschmittelinhaltsstoffe lineare Alkylbenzol-sulfonate (LAS), welches das Tensid mit der höchsten Produktionsmenge darstellte, die beiden nichtionischen Tenside Alkylglucamide (AG) und Alkylpolyglucoside (APG), ebenso wie das amphotere Tensid Cocamidopropylbetain (CAPB). Außerdem wurde der polymere Farbübertragungsinhibitor Polyvinylpyrrolidon (PVP) untersucht.

Reaching and grasping an object is an action that can be performed in light, under visual guidance, as well as in darkness, under proprioceptive control only. Area V6A is a visuomotor area involved in the control of reaching movements. V6A, besides neurons activated by the execution of reaching movements, shows passive somatosensory and visual responses. This suggests fro V6A a multimodal capability of integrating sensory and motor-related information, We wanted to know whether this integration occurrs in reaching movements and in the present study we tested whether the visual feedback influenced the reaching activity of V6A neurons. In order to better address this question, we wanted to interpret the neural data in the light of the kinematic of reaching performance. We used an experimental paradigm that could examine V6A responses in two different visual backgrounds, light and dark. In these conditions, the monkey performed an istructed-delay reaching task moving the hand towards different target positions located in the peripersonal space. During the execution of reaching task, the visual feedback is processed in a variety of patterns of modulation, sometimes not expected. In fact, having already demonstrated in V6A reach-related discharges in absence of visual feedback, we expected two types of neural modulation: 1) the addition of light in the environment enhanced reach-related discharges recorded in the dark; 2) the light left the neural response unmodified. Unexpectedly, the results show a complex pattern of modulation that argues against a simple additive interaction between visual and motor-related signals.

Satellite remote sensing has proved to be an effective support in timely detection and monitoring of marine oil pollution, mainly due to illegal ship discharges. In this context, we have developed a new methodology and technique for optical oil spill detection, which make use of MODIS L2 and MERIS L1B satellite top of atmosphere (TOA) reflectance imagery, for the first time in a highly automated way. The main idea was combining wide swaths and short revisit times of optical sensors with SAR observations, generally used in oil spill monitoring. This arises from the necessity to overcome the SAR reduced coverage and long revisit time of the monitoring area. This can be done now, given the MODIS and MERIS higher spatial resolution with respect to older sensors (250-300 m vs. 1 km), which consents the identification of smaller spills deriving from illicit discharge at sea. The procedure to obtain identifiable spills in optical reflectance images involves removal of oceanic and atmospheric natural variability, in order to enhance oil-water contrast; image clustering, which purpose is to segment the oil spill eventually presents in the image; finally, the application of a set of criteria for the elimination of those features which look like spills (look-alikes). The final result is a classification of oil spill candidate regions by means of a score based on the above criteria.

Nozizeptive Spinalganglienneurone detektieren mit einer Vielzahl liganden- und spannungsgesteuerter Ionenkanäle noxische Reize, d.h. Reize, die eine Gewebeschädigung bewirken können, wandeln sie in Aktionspotenzialentladungen um und leiten sie über das Rückenmark zum Gehirn weiter, wo eine Schmerzempfindung ausgelöst wird. Die pronozizeptiven transienten Rezeptor-Potenzial-Kanäle der Vanilloidrezeptorfamilie, TRPV1 und TRPV2, sind die klassischen Transduktionsmoleküle für noxische Hitzereize in den Spinalganglien und werden von Reiztemperaturen über 43°C bzw. 52°C aktiviert. Daneben finden sich auch antinozizeptive Membranproteine, wie z.B. der metabotrope Cannabinoidrezeptor CB1. Er koppelt an spannungsgesteuerte Kaliumkanäle, die neben Natrium- und Kalziumkanälen ebenfalls an der neuronalen Erregbarkeit beteiligt sind. Von den spannungsgesteuerten Kaliumkanälen könnte der Kv1.4, der einen schnell inaktivierenden A-Strom vermittelt, an antinozizeptiven Signalwegen beteiligt sein. Um die molekulare Physiologie der Regulation von Nozizeption und Antinozizeption zu charakterisieren, wurde die Expression bzw. Ko-Expression dieser Membranproteine auf der einen als auch die funktionelle Charakterisierung von TRPV1 auf der anderen Seite im Soma der Spinalganglienneurone und im heterologen Expressionssystem untersucht. TRPV1 wurde in je einem Drittel und TRPV2 in je einem Zehntel aller Spinalganglienneurone nachgewiesen. Das Expressionsmuster veränderte sich nicht zwischen verschiedenen Präparationsmethoden, die zur Aufarbeitung der Zellen für unterschiedliche experimentelle Ansätze notwendig sind. Somit können die aus Expressionsanalysen und funktionellen Untersuchungen gewonnenen Ergebnisse miteinander verglichen werden. Obwohl TRPV1 und TRPV2 in unterschiedlich großen Zellen exprimiert werden, überlappen dennoch ihre Größenverteilungen. Durch Ko-Expressionsanalysen konnten hier erstmalig TRPV1-TRPV2-ko-exprimierende Neurone detektiert werden. Mit dem neu entwickelten N-terminalen Antikörper gegen TRPV1 (3C11) konnte gezeigt werden, dass für TRPV1 verschiedene Splice-Varianten existieren. Neben den bereits bekannten Splice-Varianten wurde hier die neue Variante Vr.3’sv isoliert. Diese besitzt zwischen Exon 15 und 16 eine Insertion aus 104 Basen und exprimiert daher einen veränderten C-Terminus. Trotz dieser Veränderung bildeten sich im heterologen Expressionssystem funktionelle Kanäle aus, die im Gegensatz zu den anderen Varianten immer noch durch Capsaicin aktivierbar waren. Vr.3’sv könnte als Homo- oder Heterotetramer die Eigenschaften TRPV1-positiver Neurone beeinflussen. Bei der Bestimmung der Häufigkeit von TRPV1 in einem Gewebe ist somit die Wahl des Antikörpers von entscheidender Bedeutung. Für TRPV2 dagegen gibt es hier keine Hinweise auf Splice-Varianten. TRPV1 wird durch das Vanilloid Capsaicin aktiviert, wobei diese Substanz neurotoxisch ist und eine Degeneration von Neuronen und epidermalen Nervenfasern bewirkt. Hier wurde nun gezeigt, dass unabhängig von den Splice-Varianten nicht alle TRPV1-positiven Neurone bei langer Inkubationszeit absterben. Funktionelle Untersuchungen belegten, dass auch Capsaicin-sensitive Zellen unter dem Einfluss des Agonisten überleben können. Dieser Schutzmechanismus wird möglicherweise von den verschiedenen Splice-Varianten vermittelt. Ko-Expressionsanalysen zeigten, dass der spannungsgesteuerte Kaliumkanal Kv1.4 in nahezu allen TRPV1- aber nicht TRPV2-positiven Neuronen exprimiert wird. Desweiteren ko-exprimierten nahezu alle TRPV1-positiven Neurone auch den Cannabinoidrezeptor CB1. Diese fast vollständige Ko-Lokalisation von CB1 und Kv1.4 in nozizeptiven Spinalganglienneuronen spricht für eine funktionell synergistische Aktivität. Der Kaliumkanal kann unter der regulativen Kontrolle von CB1 als Vermittler von A-Typ-Kaliumströmen an der Kontrolle der repetitiven Entladungen in der Peripherie und der Transmitterausschüttung zentral beteiligt sein. Es ergeben sich daraus Ansatzpunkte für die Entwicklung neuer Medikamente. Mit Kv1.4-Aktivatoren und/oder peripher wirkenden Cannabinoiden könnten die Nebenwirkungen der Cannabinoide im zentralen Nervensystem umgangen werden.

L’invarianza spaziale dei parametri di un modello afflussi-deflussi può rivelarsi una soluzione pratica e valida nel caso si voglia stimare la disponibilità di risorsa idrica di un’area. La simulazione idrologica è infatti uno strumento molto adottato ma presenta alcune criticità legate soprattutto alla necessità di calibrare i parametri del modello. Se si opta per l’applicazione di modelli spazialmente distribuiti, utili perché in grado di rendere conto della variabilità spaziale dei fenomeni che concorrono alla formazione di deflusso, il problema è solitamente legato all’alto numero di parametri in gioco. Assumendo che alcuni di questi siano omogenei nello spazio, dunque presentino lo stesso valore sui diversi bacini, è possibile ridurre il numero complessivo dei parametri che necessitano della calibrazione. Si verifica su base statistica questa assunzione, ricorrendo alla stima dell’incertezza parametrica valutata per mezzo di un algoritmo MCMC. Si nota che le distribuzioni dei parametri risultano in diversa misura compatibili sui bacini considerati. Quando poi l’obiettivo è la stima della disponibilità di risorsa idrica di bacini non strumentati, l’ipotesi di invarianza dei parametri assume ancora più importanza; solitamente infatti si affronta questo problema ricorrendo a lunghe analisi di regionalizzazione dei parametri. In questa sede invece si propone una procedura di cross-calibrazione che viene realizzata adottando le informazioni provenienti dai bacini strumentati più simili al sito di interesse. Si vuole raggiungere cioè un giusto compromesso tra lo svantaggio derivante dall’assumere i parametri del modello costanti sui bacini strumentati e il beneficio legato all’introduzione, passo dopo passo, di nuove e importanti informazioni derivanti dai bacini strumentati coinvolti nell’analisi. I risultati dimostrano l’utilità della metodologia proposta; si vede infatti che, in fase di validazione sul bacino considerato non strumentato, è possibile raggiungere un buona concordanza tra le serie di portata simulate e osservate.
