A method was developed using the multi-element graphite furnace atomic absorption spectrometry technique for the direct and simultaneous determination of As, Cu, and Pb in Brazilian sugar cane spirit (cachaqa) samples. Also employed was the end-capped transversely heated graphite atomizer (THGA) with platforms pre-treated with W permanent modifier and co-injection of Pd/Mg(NO3)(2). Pyrolysis and atomization temperature curves were established in a cachaqa medium (1+1; v/v) containing 0.2% (v/v) HNO3 and spiked with 20 mu g L-1 As and Pb and 200 mu g L-1 Cu. The effect of the concentration of major elements usually present in cachaqa matrices (Ca, Mg, Na, and K) and ethanol on the absorbance of As, Cu, and Pb was investigated. Analytical working solutions of As, Cu, and Pb were prepared in 10% (v/v) ethanol plus 5.0 mg L-1 Ca, Mg, Na, and K. Acidified to 0.2% (v/v) HNO3, these solutions were suitable to build calibration curves by matrix matching. The proposed method was applied to the simultaneous determination of As, Cu, and Pb in commercial sugar cane spirits. The characteristic mass for the simultaneous determination was 16 pg As, 119 pg Cu, and 28 pg Pb. The pretreated tube lifetime was about 450 firings. The limit of detection (LOD) was 0.6 mu g L-1 As, 9.2 mu g L-1 Cu, and 0.3 pig L-1 Pb. The found concentrations varied from 0.81 to 4.28 mu g L-1 As, 0.28 to 3.82 mg L-1 Cu and 0.82 to 518 mu g L-1 Pb. The recoveries of the spiked samples varied from 94-112% (As), 97-111% (Cu), and 95-101% (Pb). The relative standard deviation (n=12) was 6.9%, 7.4%, and 7.7% for As, Cu, and Pb, respectively, present in a sample at 0.87 mu g L-1, 0.81 mg L-1, and 38.9 mu g L-1 concentrations.

Objective To report the severe metabolic acidosis identified in a group of 11 healthy mules anaesthetized with halothane for castration.Study design Data generated from a prospective study.Animals Eleven mules aged 2.5-8 years, weighing 230-315 kg and 11 horses aged 1.5-3.5 years, weighing 315-480 kg.Methods Animals were anaesthetized for castration as part of an electroencephalographic study. Preanaesthetic medication was acepromazine (0.03 mg kg(-1)) administered through a preplaced jugular venous catheter. Anaesthesia was induced 30-90 minutes later with intravenous thiopental (10 mg kg(-1)). After orotracheal intubation, anaesthesia was maintained with halothane vaporised in oxygen. The animals' lungs were ventilated to maintain the end-tidal CO(2) concentration between 3.9 and 4.5 kPa (29-34 mmHg). Anaesthetic monitoring included invasive blood pressure measurement via the auricular artery (mules) and submandibular branch of the facial artery (horses). Arterial blood gas samples were drawn from these catheters at three time points during surgery and pH, PaCO(2), base excess (ecf) and HCO(3)(-) were measured. Values were compared between groups using a Mann-Whitney test. p was taken as <0.05. Results are reported as median (range).Results PaCO(2) did not differ between groups but pH was significantly lower in mules [7.178 (7.00-7.29)] compared to horses [7.367 (7.24-7.43)] (p = 0.0002). HCO(3)(-) values were significantly lower in the mules [16.6 (13.0-22.3) mM] compared to horses [23.7 (20.9-23.7) mM] (p = 0.0001), whilst base excess (ecf) was significantly more negative in the mules [-11.4 (-1.27 to -16) mM] compared to horses [-1.3 (-5.8 to +2.4) mM] (p = 0.0004).Conclusion and clinical relevance This study demonstrated severe metabolic acidosis in healthy mules, which may have prompted intervention with drug therapies in a clinical arena. It is probable that the acidosis existed prior to anaesthesia and caused by diet, but other possible causes are considered.

Aim of the study was to evaluate the influence of an extra corporal perfusion (cardiopulmonary bypass operation - cpb) on activation and biodistribution of Tc-99m labelled granulocytes in pigs with and without inhibition of the granulocytes by a leukocyte inhibition module (LIM). The cpb is often related to an activation of granulocytes resulting in an inflammatory answer. The biological mechanisms are unsolved yet. First trials of our group showed that LIM may inhibit the activation of neutrophils and therefore antagonize a cpb-caused impairment of cardiac function. This study is the continuation of these experiments with a higher number of animals and the focus on scintigraphic imaging. Animals, material, methods: 39 German landrace pigs were subdivided into three groups: group A (control) median sternotomy without cpb, group B with cpb, group C with LIM in addition to cpb. After labelling with Tc-99m-HMPAO autologues granulocytes were reinjected. Subsequently to cpb, the animals underwent scintigraphic imaging. Quantification was performed with ROI evaluation and with tissue samples (section analysis) examined in a well counter. Results:A high uptake of Tc-99m-HMPAO was found in the liver. The count rates in brain, heart, lung, spleen and kidneys were far below. The amount of Tc-99m-activity in the organ related to the half life corrected administered activity [%] was for the tissue samples (group A/B/C): brain 0.01/0.02/0.03; lung 12.1/8.3/11.5; heart 0.35/0.54/0.42; kidney 1.24/0.87/1.02; spleen 4.0/4.0/4.5, liver 16.8/20.9/19.6. The count rates determined by ROI-evaluation of the scintigraphic images related to the total count rate in the image [%] were (group A/B/C): brain 1.1/0.9/1.0; lung 15.6/10.4/12.2; heart 4.0/3.5/3.4; kidney 4.0/2.9/3.2; spleen 7.6/7.7/9.5, liver 23.1/36.7/31.4. A significant difference in the tracer uptake between the groups could neither be detected by scintigraphic imaging nor evaluation of tissue samples. Conclusion: Scintigraphic imaging as well as section analysis showed a comparable biodistribution of the tracer. Therefore, the initial results of our group were not confirmed with a considerably higher number of animals. Neither cpb nor the use of the LIM influenced distribution of Tc-99m-labelled granulocytes in pigs significantly.

Morphological features of the mid-palatal suture were studied in human foetuses from 4 to 9 months of intra-uterine life. The foetuses were divided into three age groups, GI (16-23 weeks), GII (24-31 weeks) and GIII (32-39 weeks). The mid-palatal suture in GI foetuses is rectilineal in form with a wide space between the palatal processes of the maxilla. The suture has a sinuous nature in GII and GIII foetuses due to growth of the bone processes crossing the mid-line. A wide zone of cellular proliferation observed in GI narrows in GII and GIII foetuses. The imbricating nature of the suture in GII and GIII is caused by bone growth adjacent to the mid-palatal suture. Sharpey's fibres, emerging from the bone processes, run to the median region of the mid-palatal suture and are observed from GI foetuses onwards. The collagen fibres of the mid-palatal suture are orientated transversely under the oral epithelium and exhibit a regular meshwork with a predominance of sagittal fibres in the median region of the suture. These fibres are orientated transversely and obliquely at the junction with the nasal septum.

Background. An interaction between lectins from marine algae and PLA 2 from rattlesnake was suggested some years ago. We, herein, studied the effects elicited by a small isolectin (BTL-2), isolated from Bryothamnion triquetrum, on the pharmacological and biological activities of a PLA 2 isolated from rattlesnake venom (Crotalus durissus cascavella), to better understand the enzymatic and pharmacological mechanisms of the PLA 2 and its complex. Results. This PLA2 consisted of 122 amino acids (approximate molecular mass of 14 kDa), its pI was estimated to be 8.3, and its amino acid sequence shared a high degree of similarity with that of other neurotoxic and enzymatically-active PLA2s. BTL-2 had a molecular mass estimated in approximately 9 kDa and was characterized as a basic protein. In addition, BTL-2 did not exhibit any enzymatic activity. The PLA2 and BTL-2 formed a stable heterodimer with a molecular mass of approximately 24-26 kDa, estimated by molecular exclusion HPLC. In the presence of BTL-2, we observed a significant increase in PLA2 activity, 23% higher than that of PLA2 alone. BTL-2 demonstrated an inhibition of 98% in the growth of the Gram-positive bacterial strain, Clavibacter michiganensis michiganensis (Cmm), but only 9.8% inhibition of the Gram-negative bacterial strain, Xanthomonas axonopodis pv passiflorae (Xap). PLA2 decreased bacterial growth by 27.3% and 98.5% for Xap and Cmm, respectively, while incubating these two proteins with PLA2-BTL-2 inhibited their growths by 36.2% for Xap and 98.5% for Cmm. PLA2 significantly induced platelet aggregation in washed platelets, whereas BTL-2 did not induce significant platelet aggregation in any assay. However, BTL-2 significantly inhibited platelet aggregation induced by PLA2. In addition, PLA 2 exhibited strong oedematogenic activity, which was decreased in the presence of BTL-2. BTL-2 alone did not induce oedema and did not decrease or abolish the oedema induced by the 48/80 compound. Conclusion. The unexpected results observed for the PLA2-BTL-2 complex strongly suggest that the pharmacological activity of this PLA2 is not solely dependent on the presence of enzymatic activity, and that other pharmacological regions may also be involved. In addition, we describe for the first time an interaction between two different molecules, which form a stable complex with significant changes in their original biological action. This opens new possibilities for understanding the function and action of crude venom, an extremely complex mixture of different molecules. © 2008 Oliveira et al; licensee BioMed Central Ltd.

A leishmaniose tegumentar (LT) encontra-se em expansão no Estado do Pará, Brasil. Juruti é um dos 143 municípios desse Estado e atualmente cenário de grandes transformações ambientais devido à mineração de bauxita, o que poderá influenciar o padrão de transmissão. Objetivo: Este estudo buscou elucidar aspectos epidemiológicos relevantes para o controle da LT em Juruti. Materiais e Métodos: A frequência de LT e o perfil dos pacientes no hospital municipal "Francisco Barros" foram determinados de janeiro a dezembro/2007. Espécies de flebotomíneos silvestres existentes no entorno de uma área de prospecção da bauxita foram também descritas, durante levantamento entomológico em janeiro/2008 (armadilha Shannon/18h às 20h/2 noites). Em 21 indivíduos, portadores de lesão cutânea suspeita de LT, biópsias de pele foram realizadas entre fevereiro e junho de 2007. Neste grupo procedeu-se ao diagnóstico parasitológico (esfregaço corado e cultura), molecular e teste intradérmico de Montenegro. Utilizaram-se sondas de DNA ribossomal (PCR-SSUrDNA) gênero específicas (S4, S12; S17, S18) e de G6PD, para distinguir o subgênero Viannia (ISVC, ISVA: ISVC, ISVG) e a espécie L. (V.) braziliensis (ISVC, ISVA; ISVC, ISVB). Resultados: No ano de 2007 foram confirmados 42 casos novos de LT, com média mensal inferior a quatro (3,5 ± 0,8), maior frequência em julho (11) e menor em junho e novembro (0). A maioria dos pacientes foi de homens (41/42, 98%) com menos de 20 anos (<10 anos: 30%; 10-20: 57%; 20-40: 12%). A maioria também residia em localidades rurais (33/42, 79%), incluindo áreas impactadas pela mineração (19/42, 45%), e exercia atividades de risco (28/42, 67%). Doze eram funcionários de empresas (29%). A análise molecular das 21 amostras identificou 12 resultados positivos para o gênero Leishmania (57%), sendo 11 (52%) parasitologicamente confirmados. A PCRG6PD identificou 75% das amostras como sendo L. (V.) braziliensis. As demais (3/12, 25%) não hibridizaram com os oligonucleotídeos da PCR-G6PD e, por isso, os produtos da reação de nested-PCR SSUrDNA foram clonados e sequenciados, confirmando que se tratavam de Leishmania (Viannia) sp. Apenas 9/12 (75%) casos confirmados pelos métodos parasitológico e/ou olecular tiveram reações de hipersensibilidade tardia em resposta ao antígeno de Montenegro, cujos diâmetros variaram de 7 a 40mm (16,3 ± 3,2). Capturaram-se 105 flebotomíneos de 13 espécies nas seguintes frequências: 1- Lutzomyia (Ps.) geniculata (23, 22%), 2- Lutzomyia (Ps.) paraensis (21, 20%), 3- Lutzomyia (Ps.) complexa (18, 17%), 4-Lutzomyia (Ps.) davisi (10, 10%), 5- Lutzomyia (N.) flaviscutellata (13, 13%) e outras oito espécies (20, 18%). Discussão: Espécies de Leishmania do subgênero Viannia, sobretudo L. (V.) braziliensis predominam em Juruti, o que é compatível com o extenso diâmetro das reações cutâneas observadas ao antígeno de Montenegro e com os relatos comuns de persistência e recidiva, apesar do tratamento específico. Entre os flebotomíneos antropófilos destacam-se L. (Ps.) complexa (17%) e L. (Ps.) flaviscutellata (13%) por serem vetores de L. (V.) braziliensis e L. (L.) amazonensis respectivamente, associadas às formas severas da LT humana. Conclusão: Medidas de controle em Juruti devem priorizar a redução da morbidade, diagnóstico precoce, busca ativa de LT humana, vigilância entomológica e de microambientes no entorno da área de impacto de mineração.

O objetivo deste estudo foi avaliar o comportamento de dois cimentos de ionômero de vidro: um de alta viscosidade (Ketac Molar - ESPE) e um modificado por resina (Fuji VIII – GC) em restaurações envolvendo duas ou mais superfícies dentárias, confeccionadas pela técnica do Tratamento Restaurador Atraumático. Sessenta restaurações (30 com cada material) foram inseridas em estudantes (9-16 anos) por dois operadores. Após 6 meses, dois examinadores independentes avaliaram as restaurações de acordo com os critérios utilizados em trabalhos prévios semelhantes. Os dados foram analisados pelos testes de McNemar e Fischer. A porcentagem de sucesso do tratamento foi de 98,3%. Uma restauração (Ketac Molar) foi substituída por outro material e classificada como falha. As porcentagens de sucesso das restaurações foram de 100% e 96,6% para o Fuji VIII e Ketac Molar, respectivamente. Não houve diferença estatisticamente significante no sucesso das restaurações entre o baseline e 6 meses (p>0,05). Da mesma forma, não houve diferença estatística entre os materiais, tipos de cavidade ou entre operadores.(p>0,05). A técnica ART foi altamente apropriada e efetiva em restaurações envolvendo duas ou mais superfícies, após 6 meses. Os resultados mostraram um comportamento promissor com ambos os materiais.
