The occurrence of Biomphalaria intermedia (Paraense & Deslandes, 1962) is recorded at Três Lagoas and Dourados (state of Mato Grosso do Sul, Brazil), and Parque Nacional Iguazú (province of Misiones, Argentina), west and southwest, respectively, of its known western limit, in the state of São Paulo. The species was also found at Parapuã, a new locality to São Paulo state.

Objetivando ampliar o conhecimento da biologia de flebótomos em cativeiro, que propicie condições para mantê-los regularmente, estabelecemos colônias autônomas de Lutzomyia intermedia e Lutzomyia longipalpis, apresentando aqui dados referentes às observações sobre a alimentaçãodas larvas e adultos. A ração comercializada para peixes é bem aceita pelas larvas das duas espécies, em todos os estádios; é de fácil aquisição e de baixo custo, não favorecendo a proliferação de fungos. As larvas de L. intermedia e de L. longipalpis, em todos os estádios, aceitam rações alimentares de origem vegetal e de origem mista; porém as de 1º e 2º estádios de L. intermedia têm certa preferência pela ração de base vegetal, enquanto que as de 3º e 4º estádios de L. longipalpis ainda que discretamente, preferem ração de origem mista. A prévia alimentação com solução açucarada não é fator indispensável ao hematofagismo nas duas espécies. Ambas se alimentam bem em homem, cão, pinto ou hamster, mas a fonte de alimento sanguíneo mais adequada é o hamster, analisando-se aceitação da isca, desova, duração do ciclo e produtividade a partir do número de ovos postos. As fêmeas de L. longipalpis mostraram maior resistência ao jejum de sangue que as l. intermedia, embora ambas possam resistir, em mais de 70% até o 7º dia, apenas com alimentação de solução açucarada.

Com o objetivo de ampliar os conheciemntos sobre a biologia de Lutzomyia intermedia e Lutzomya longipalpis, mantidos em colônias autônomas no laboratório, apresentamos dados referentes a alterações em seu comportamento determinadas por influência de fatores ambientais. L. longipalpis foi mais fácil de criar, mais produtiva e mais resitente ás variações das condições ambientais; suga a qualquer hora do dia, enquanto que L. intermedia prefere fazê-lo ao crepúsculo e à noite, quando também ocorrem masi freqüentemente as desovas e as ecdises dos adultos das duas espécies. As fases imaturas de ambas as espécies resistem à imersão na água por até 1 hora e a baixa temperatura de 5ºC por até 6 horas.

Three Yersinia pestis strains isolated from humans and one laboratory strain (EV76) were grown in rich media at 28§C and 37§C and their outer membrane protein composition compared by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-Page). Several proteins with molecular weights ranging from 34 kDa to 7 kDa were observed to change in relative abundance in samples grown at different temperatures. At least seven Y. pestis outer membrane proteins showed a temperature-dependent and strain-specific behaviour. Some differences between the outer membrane proteins of full-pathogenic wild isolates and the EV76 strain could aldso be detected and the relevance of this finding on the use of laboratory strains as a reference to the study of Y. pestis biological properties is discuted.

The development of Leishmania (Viannia) braziliensis in experimentally infected Lutzomyia intermedia, showed colonization of the hindgut from 48h after the infective blood-meal, and the migration flagellates to the foregut, with a massive infection of the cardia at the 5th day post infection. Up to 10 days following the infective blood-meal, very few parasites were seen in the pharynx and cibarium. The role of L. intermedia as a vector of cutaneous leishmaniasis is discussed according to the estabilished criteria.

The development of Colombian Leishmania species of the subgenus Viannia in Lutzomyia intermedia was similar to that observed with Brazilian Le. (V.) braziliensis: colonization of the pylorus by paramastigotes; promastigotes in the midgut and massive infection of stomodeal valve. Difference was observed in the number of paramastigotes colonizing the pylorus, which was smaller in Colombian Leishmania species than Brazilian Le. braziliensis.

The irp2 gene codes for a 190 kDa protein (HMWP2) synthesidez when highly pathogenic Yersinia are grown under conditions of iron starvation. In this work, the presence of irp2 in strains of Y. pestis isolated from different hosts during several plague outbreaks in the foci of Northeast Brazil wasstudied. For this purpose, 53 strains were spotted onto nylon filters and their DNA was hybridized with the A13 probe which is a 1 kb fragment of the irp2 coding sequence. All strains except two hybridized with the probe. However, when the initial stock culture of these two strains were analyzed, they both proved to bepositive with the A13 probe, indicating that the locus was lost after subculturein vitro but was always present in vivo. To examine the degree of conservation of the chromosomal fragment carrying irp2 among Brazilian strains, the hybridization profiles of 15 strains from different outbreaks, different hosts and different foci were compared. The hybridization profiles of these strains were all identical when their DNA was digested with either EcoRI, EcorRV or AvaII, indicatingthat the restriction sites surrounding the irp2 locus are very well conserved among Northeast Brazilian strains of Y. pestis. Altogether, these results suggest that the irp2 chromosomal region should be of prime importance for the bacteria during their multiplication in the host.

Lutzomyia (Nyssomyia) intermedia (Lutz & Neiva) is redescribed, a neotype is designated, and a very similar species, L. (N.) neivai (Pinto, 1926), is resurrected; the male and the female are redescribed. The two species can be differentiated by differences in the spermathecae, common ducts and number of cibarial horizontal teeth of the females and in the size and proportions of several structures of both sexes. The known geographical distribution of both species is given.

The influence of altitude and latitude on some structure sizes of Lutzomyia intermedia was noted; several structures of insects collected in higher localities were greater, according to Bergmann's rule. This influence was more remarkable in two localities of the State of Espírito Santo, probably due to greater differences in altitude. Comparing insects from different latitudes, more differences were noted in comparisons of insects from low altitude localities than in those of material from higher altitudes. The small number of differences between insects collected in July and in December does not indicate a defined influence of season and temperature on the size of adults. The possible epidemiological implications of these variations are discussed.

The females of the two species of the Lutzomyia intermedia complex can be easily distinguished, but the males of each species are quite similar. The ratios between the extra-genital and the genital structures of L. neivai are larger than those of L. intermedia s. s., according to ANOVA. An artificial neural network was trained with a set of 300 examples, randomly taken from a sample of 358 individuals. The input vectors consisted of several ratios between some structures of each insect. The model was tested on the remaining 58 insects, 56 of which (96.6%) were correctly identified. This ratio of success can be considered remarkable if one takes into account the difficulty of attaining comparable results using traditional statistical techniques.
