Nowadays generation ethanol second, that t is obtained from fermentation of sugars of hydrolyses of cellulose, is gaining attention worldwide as a viable alternative to petroleum mainly for being a renewable resource. The increase of first generation ethanol production i.e. that obtained from sugar-cane molasses could lead to a reduction of lands sustainable for crops and food production. However, second generation ethanol needs technologic pathway for reduce the bottlenecks as production of enzymes to hydrolysis the cellulose to glucose i.e. the cellulases as well as the development of efficient biomass pretreatment and of low-cost. In this work Trichoderma reesei ATCC 2768 was cultivated under submerged fermentation to produce cellulases using as substrates waste of lignocellulosic material such as cashew apple bagasse as well as coconut bagasse with and without pretreatment. For pretreatment the bagasses were treated with 1 M NaOH and by explosion at high pressure. Enzyme production was carried out in shaker (temperature of 27ºC, 150 rpm and initial medium pH of 4.8). Results showed that T.reesei ATCC 2768 showed the higher cellulase production when the cashew apple bagasse was treated with 1M NaOH (2.160 UI/mL of CMCase and 0.215 UI/mL of FPase), in which the conversion of cellulose, in terms of total reducing sugars, was of 98.38%, when compared to pretreatment by explosion at high pressure (0.853 UI/mL of CMCase and 0.172 UI/mL of Fpase) showing a conversion of 47.39% of total reducing sugars. Cellulase production is lower for the medium containing coconut bagasse treated with 1M NaOH (0.480 UI/mL of CMcase and 0.073 UI/mL of FPase), giving a conversion of 49.5% in terms of total reducing sugars. Cashew apple bagasse without pretreatment showed cellulase activities lower (0.535 UI/mL of CMCase and 0,152 UI/mL of FPase) then pretreated bagasse while the coconut bagasse without pretreatment did not show any enzymatic activity. Maximum cell concentration was obtained using cashew nut bagasse as well as coconut shell bagasse treated with 1M NaOH, with 2.92 g/L and 1.97 g/L, respectively. These were higher than for the experiments in which the substrates were treated by explosion at high pressure, 1.93 g/L and 1.17 g/L. Cashew apple is a potential inducer for cellulolytic enzymes synthysis showing better results than coconut bagasse. Pretreatment improves the process for the cellulolytic enzyme production

Biosurfactants are amphiphilic molecules synthesized by microorganisms such as bacteria, yeast or filamented fungi cultivated in various carbon sources among sucrose and hydrocarbons. These molecules are composed by a hydrophilic and hydrophobic part. They operate mostly at interfaces of fluids of different polarities. Because of this characteristic, they are potentially employed in numerous industries, such as the textile, medical, cosmetics, food and mainly in the petrochemical ones. Therefore industry has interest in developing new biosurfactant production processes in high scale, in order to become them economically competitive when compared to synthetic biosurfactants. This work aims to evaluate the biosurfactant production applying a non-conventional substrate sugar cane molasses proceeding from the sugar industry thus reducing the production costs. The strain identified as AP029/GLIIA, isolated from oil wells in Rio Grande do Norte state and used in these experiments belongs to the culture collection of Antibiotics Department of UFPE. The fermentation were carried out using different conditions according to a factorial planning 24 with duplicate at center point, in which the studied factors were molasse concentration, nitrate concentration, agitation and aeration ratio. The experiments were performed in a shaker at 38ºC of temperature. Samples were withdrawn in regular periods of time of up to 72 hours of fermentation in order to analyze substrate consumption, cellular concentration, superficial tension, critical micelle dilution (CMD-1 e CMD-2) as well as extracelullar protein production. The results showed a production of 3,480 g/L of biomass, a reduction of 41% on superficial tension, 67% of substrate consumption and 0,2805 g/L of extracellular protein

Considering the increasing demand for seedlings of plants for aforestation and the scarcity of raw materials for the substrates used for plant growth, it is necessary to evaluate new components and formulations that ensure the quality of seedlings. In this experiment the development and quality of production of Eucalyptus urophylla x E. grandis seedlings was studied through vegetative propagation, as a function of nine compositions of substrate, produced from fine-grained vermiculite, carbonized rice chaff and coconut fiber in hard plastic tubes of 50 mL as containers. The physical properties of substrates and morphological development and the quality of the root system of seedlings were evaluated at 90 days after staking. The substrates with higher total porosity promotes better quality of the root system, which consequently results in cuttings with larger diameter, shoot and root dry weight and Dickson Quality Index. Other properties for the physical characteristics of substrates other than mentioned in literature can also be considered adequate.

Pectinolytic enzymes, or simply pectinases, are complex enzymes that degrade pectic polymers. They have many uses, such as fruit juice extraction and purification, textile fiber treatment and vegetal oil extraction. The aim of this work was to study the kinetics of pectinases production by solid-state fermentation, using dry cashew apple residue as substrate and the microorganism Aspergillus niger CCT 0916. The influence of the initial medium moisture and medium supplementation with a source of nitrogen and phosphorus was evaluated using the factorial experimental planning and response surface methodology. Ammonia sulphate and potassium phosphate were used as nitrogen and phosphorus source, respectively. The variables time of contact (T) and ratio volume solvent/fermented medium (RZ), in systems with and without agitation, were evaluated in order to study the best extraction condition of the produced enzyme. Washed and unwashed cashew apple residues were tested as the growth medium. The unwashed residue was obtained by drying the residue after the extraction of the juice, while the washed residue was obtained by water washing 5 times using the proportion of 1 kg pulp/2 liters of water. Samples were taken every 12 hours for moisture content, pH, protein, reducing sugars, polygalacturonase activity (PG) and viscosity reduction. The physical-chemical composition of the residues had different sugar and pectin levels. For the unwashed residue, the peak activity was reached with 40% of initial moisture content, 1% of nitrogen supplementation without phosphorus addition after 30 hours of process. These conditions led to 16 U/g of PG activity and 82% of viscosity reduction. The calculated models reached similar values to the experimental ones in the same process conditions: 15.55 U/g of PG and 79.57% of viscosity eduction. Similarly, the greatest enzyme production for washed residue was reached with 40% initial moisture content, 1% nitrogen supplementation without phosphorus addition after 22 hours of cultivation. In this condition it was obtained polygalacturonase activity of 9.84 U/g and viscosity reduction of 81.36%. These values are close to experimental values that were of 10.1 U/g and 81%, respectively. The conditions that led to the best PG activity results was the agitated one and the best extraction condition was obtained with 100 minutes of solvent/medium contact and RZ of 5 (mL/g)

Biosurfactants are molecules produced by microorganisms mainly bacteria as Pseudomonas and Bacillus. Among the biosurfactants, rhamnolipids play an important role due to their tensoactive as well as emulsifying properties. Besides can be produced in a well consolidated way the production costs of biosurfactants are quite expansive mainly if downstream processing is goning to be considered. Actually, attention has been given to identification of biosurfactants as well as optimization of its fermentative processes including downstream ones. This work deals with the development of strategies to recovery and purification of rhamnolipids produced by Pseudomonas aeruginosa P029-GVIIA using sugar-cane molasses as substrate. Broth free of cells was used in order to investigate the best strategies to recovery and purification produced by this system. Between the studied acids (HCl and H2SO4) for the acid precipitation step, HCl was the best one as has been showed by the experimental design 24. Extraction has been carried out using petroleum ether and quantification has been done using the thioglycolic acid method. Adsorption studies were carried out with activated carbon in a batch mode using a 24 experimental design as well as combined with an hydrophobic resin Streamline Phenyl aiming to separate the produced biosurfactant. Biosurfactant partial identification was carried out using High Performance Liquid Chromatography (HPLC). Experiments in batch mode showed that adsorption has been controlled mainly by pH and temperature. It was observed a reduction of 41.4% for the liquid phase and the solid phase it was possible to adsorb up to 15 mg of rhamnolipd/g of activated carbon. The kinetics of adsorption has been well fitted to a pseudo-first order reaction with velocity constant (k1) of 1.93 x 10-2 min-1. Experiments in packed bed ranging concentration on eluent (acetone) has been shown the highest recovery factor of 98% when pure acetone has been used. The combined effect if using activated carbon with an hydrophobic resin Streamline Phenyl has been shown successful for the rhamnolipids purification. It has been possible to purify a fraction of the crude broth with 98% of purity when the eluted of activated carbon packed bed was used with pure acetone

Carciniculture in Brazil occupies world-wide prominence due to shrimp culture, and the state of Rio Grande do Norte has presented the best results in the culture of the Litopenaeus vannamei in the last decade. This species has been shown to adapt easily to different environments and is between the five most cultivated penaeids of the world. The ponds are usually constructed in areas close to water courses and estuaries. Stock density and substrate ponds can pollute environment, causing losses in the growth and survival of the shrimps, being considered stress factors. Shrimps in inadequate densities and substrates can result reduced productivity of the farm; and favor diseases. So, it is important to verify how these variables influence the development of the animals in the culture farms. Our objective was to study the influence of the type of substrate and the stock density on the behavior and haemocyte count of the L. vannamei. Individually marked juvenile shrimps were kept in aquaria with 30 L of seawater and continuous aeration, in 12L-12D photoperiod. They were observed through Ad libitum and focal sampling instantaneous methods during thirty days, five times per week, six times per day (8:00 to 18:00) in windows of 15 minutes every two hours. The marking of carapace permitted quantifying molting and the feeding was supplied three times a day. Two experiments were carried out: the first one tested animals in the three different substrates (fine sand, smaller rocks-SPP and biggest rocks-SGR) with 33 shrimp/m2. In the second one, the animals were tested in three stock densities (26, 52 and 66 shrimp/m2) in fine sand substrate. At the end of experiment, biometry (first and second ones) and haemocyte count (second one) were made. The behavior of the L. vannamei seems to have been influenced by substrate and stocking density. In low granulometry of the substrate; the exploratory behavior became more frequent and inactivity of the shrimps was reduced. Burrowing was registered in sand substrate, specially in the initial period of the day. Cleaning was gradually higher along the day, presenting the biggest levels as the dark phase approached. The ingestion of feeding was more frequent in low density, and the animals were bigger and heavier at the end of the experiment. In the fine sand condition, the animals presented better growth, probably associated with the burrowing. The molting was equivalent in all types of substrate, but it was more frequent in high densities. Mortality of the shrimps was more frequent in high densities, and cannibalism and diseases were also registered in that condition. The clinical signals were similar to the ones of infectious mionecrosis (IMNV), generally associated with environment and physical stress. The haemocyte count was low for the hematologic standards of the penaeid, which we attributed for greater dilution of haemolymph in the postmolting phase. Smaller shrimps presented lower levels of haemocytes in relation to the bigger animals, count was also low in 26 shrimp/m2 density. The study demonstrates that stocking density and the granulometry of the substrate can affect the welfare, the health and the behavior of the L. vannamei. The sand substrate and low stocking density can be important tools in the management systems of shrimp production

Com o objetivo de avaliar a influência da composição química do substrato no cultivo do cogumelo comestível Pleurotus florida, seis resíduos agrícolas foram testados: palha de arroz, palha de feijão, palha de trigo, folha de bananeira, palha de sorgo e sabugo de milho. O substrato palha de feijão apresentou resultados semelhantes para a produção (189,8 g kg-1), eficiência biológica (89,2%) e número de cogumelos (12) à palha de arroz, substrato utilizado tradicionalmente no cultivo de Pleurotus em escala comercial. Não foi possível atribuir apenas a um fator químico as altas produções e eficiências biológicas observadas em palha de arroz e palha de feijão e muito menos para a baixa produção em palha de sorgo (77,8 g kg-1) e sabugo de milho (53,2 g g-1). No geral, substratos com relação C/N em torno de 45 (N = 1,0%), maior conteúdo de cinzas, acrescido de altos teores de P, K, Ca, Mg and Mn foram os melhores para o cultivo de P. florida.

Foram estudadas as respostas plásticas em duas espécies nativas. em G. hymenifolia, não foram verificadas modificações morfológicas significativas, exceto quanto ao conteúdo de amido de reserva, que foram menores nos lotes capacidade de campo desnutrido (CCD) e alagamento desnutrido (ALD). em Genipa americana os indivíduos do lote (CCN - capacidade de campo nutrido) exibiram raízes adventícias com aerênquima distinto, mais freqüentes nas plantas do lote controle submetidas ao alagamento (ALN - alagado nutrido). Nessa última espécie, nos lotes ALN, verificou-se o surgimento de aerênquima lisógeno no caule, além da hipertrofia lenticelar. A diversidade de respostas às condições de alagamento, aliada à uma larga ocorrência de populações de Genipa americana em áreas sazonalmente inundadas, como por exemplo no Pantanal de Miranda-MS, sugere um complexo de interações entre caractes adaptativos morfológicos e fisiológicos, sendo o potencial gênico fundamental neste tipo de resposta.

Foram estudadas as respostas plásticas em plântulas e plantas jovens de duas espécies arbóreas nativas, Adelia membranifolia (Müll. Arg.) Pax & K. Hoffm. (Euphorbiaceae) e Peltophorum dubium (Spreng.) Taub. (Leguminosae-Caesalpinoidae), submetidas ao alagamento do substrado associado ao déficit nutricional. Avaliou-se o acréscimo em superfície no módulo de expressão, definido como a região do eixo da plântula que, sob condições de estresse, sofre rediferenciação sendo considerada a hipertrofia lenticelar como a expressão morfogenética mais comum. em A. membranifolia a associação do estresse nutricional e alagamento do substrato foi a condição mais restritiva tanto para o crescimento em altura e diâmetro do colo quanto para as expressões morfogenéticas. Já em P. dubium o estado nutricional foi mais restritivo que o alagamento para o crescimento do colo. em geral, para ambas as espécies, plantas nutridas mantidas alagadas restabeleceram seu ritmo de crescimento a partir do terceiro mês de tratamento, além de expressarem respostas plásticas como a hipertrofia lenticelar e a formação de raízes adventícias. Quanto ao aumento em superfície, nos lotes desnutridos, verificou-se que em A. membranifolia de um total de 38% de superfície expressa, 12% corresponderam à hipertrofia lenticelar. em P. dubium verificou-se as maiores variações de aumento em superfície expressa por hipertrofia lenticelar, elevando de 5% para 13%. em geral a superfície modular nos lotes tratados foi o dobro em relação ao controle. É possível que o aumento em superfície, para algumas espécies, seja significativo como tendência ao aumento da estabilidade no desenvolvimento a partir de interações fisiológicas.

Foi estudado o efeito do cobre do substrato no crescimento, sintomatologia e composição mineral do cafeeiro novo. Verificou-se ser a var. Mundo Novo mais sensível à toxidez do cobre por translocar maior proporção do elemento que a var. Catuaí. O uso de calcário e de matéria orgânica no solo neutraliza o sobre tóxico.

Caesalpinia ferrea Mart. ex Tul. é uma leguminosa arbórea tropical que ocorre na região amazônica, sendo muito utilizada como planta medicinal e na arborização e paisagismo urbanos. Para viabilizar a produção de mudas, determinaram-se a melhor temperatura e o melhor substrato para a germinação das sementes. Sementes recém-colhidas apresentaram teor médio de água de 7,46%, porcentagem de germinação de 3,33% e baixo ganho de água durante a embebição, mostrando dormência tegumentar. A escarificação mecânica com lixa nº 40 foi um método eficiente para superação da dormência, comprovado pela alta porcentagem de germinação e embebição de água em sementes escarificadas. A porcentagem de germinação dessas sementes foi influenciada pela temperatura, mas não pelo substrato. Com base no tempo médio de germinação, recomenda-se a temperatura de 30 ºC e areia como substrato para germinação mais rápida de sementes escarificadas.
