Virus invasion of minor veins in inoculated leaves of a host is the likely prelude to systemic movement of the pathogen and to subsequent yield reduction and quality loss. In this study we have analyzed the cell number and arrangement in minor veins within mature leaves of various members of the Solanaceae and Fabaceae families. We then monitored the accumulation pattern of several tobamoviruses and potyviruses in these veins at the time of rapid, phloem-mediated movement of viruses. Vascular parenchyma cells were the predominant and sometimes only cells to become visibly infected among the cells surrounding the sieve elements in minor veins containing 9 to 12 cells. In no instance did we observe a companion cell infected without a vascular parenchyma cell also being infected in the same vein. This suggests that the viruses used in this study first enter the vascular parenchyma cells and then the companion cells during invasion. The lack of detectable infection of smooth-walled companion or transfer cells, respectively, from inoculated leaves of bean (Phaseolus vulgaris) and pea (Pisum sativum) during a period of known rapid, phloem-mediated movement suggests that some viruses may be able to circumvent these cells in establishing phloem-mediated infection. The cause of the barrier to virus accumulation in the companion or transfer cells, the relationship of this barrier to previously identified barriers for virus or photoassimilate transport, and the relevance of these findings to photoassimilate transport models are discussed.

Petunia hybrida is a popular bedding plant that has a long history as a genetic model system. We report the whole-genome sequencing and assembly of inbred derivatives of its two wild parents, P. axillaris N and P. inflata S6. The assemblies include 91.3% and 90.2% coverage of their diploid genomes (1.4 Gb; 2n = 14) containing 32,928 and 36,697 protein-coding genes, respectively. The genomes reveal that the Petunia lineage has experienced at least two rounds of hexaploidization: the older gamma event, which is shared with most Eudicots, and a more recent Solanaceae event that is shared with tomato and other solanaceous species. Transcription factors involved in the shift from bee to moth pollination reside in particularly dynamic regions of the genome, which may have been key to the remarkable diversity of floral colour patterns and pollination systems. The high-quality genome sequences will enhance the value of Petunia as a model system for research on unique biological phenomena such as small RNAs, symbiosis, self-incompatibility and circadian rhythms.

In addition to producing medicinally important tropane alkaloids, some species in the mainly Australian Solanaceous tribe Anthocercideae, sister to genus Nicotiana, are known to also contain substantial levels of the pyridine alkaloids nicotine and nornicotine. Here, we demonstrate that axenic hairy root cultures of two tribe Anthocercideae species, Cyphanthera tasmanica Miers and Anthocercis ilicifolia ssp. ilicifolia Hook, contain considerable amounts of both nicotine and nornicotine (∼0.5-1% DW), together with lower levels of the tropane alkaloid hyoscyamine (<0.2% DW). Treatment of growing hairy roots of both species with micromolar levels of the wound stress hormone methyl-jasmonate (MeJa) led to significant increases (P<0.05) in pyridine alkaloid concentrations but not of hyoscyamine. Consistent with previous studies involving Nicotiana species, we also observed that transcript levels of key genes required for pyridine alkaloid synthesis increased in hairy roots of both Anthocercideae species following MeJa treatment. We hypothesise that wound-associated induction of pyridine alkaloid synthesis in extant species of tribe Anthocercideae and genus Nicotiana was a feature of common ancestral stock that existed before the separation of both lineages ∼15million years ago.

A Physalis angulata, conhecida popularmente como camapú, pertence à família Solanaceae. Possui propriedades medicinais dentre elas ações imunomoduladoras, antimicrobianas, antitumorais, moluscicidas, antiparasitárias, antivirais e antineoplásicas. Os meios de cultura se baseiam nas exigências nutricionais das plantas, sendo adequados às necessidades de cada espécie/genótipo e cultivo in vitro. Desta forma, este estudo teve como objetivo avaliar o efeito do meio de cultura MS no cultivo in vitro de P. angulata na ausência de regulador de crescimento. O trabalho foi realizado no Laboratório de Recursos Genéticos e Biotecnologia Vegetal na Embrapa Amazônia Oriental, Belém-PA. Para o experimento foram utilizadas três concentrações de meios de cultura sólidos: MS, ½MS e ¼MS, cada concentração de meio possuía sete repetições com dois frascos, e, cada frasco continha três explantes. As variáveis avaliadas foram número de raízes, número de brotações, comprimento da maior raiz e comprimento do maior broto. Para analise estatística foi utilizado o programa Sisvar. Houve brotação e enraizamento em todas as concentrações de meio MS. Entretanto, não ocorreu diferença estatisticamente significativa para todas as variáveis brotos e raízes avaliadas sob diferentes concentrações do meio de cultura MS. Plântulas foram regeneradas na ausência de regulador de crescimento.

Heliothine moths (Lepidoptera: Heliothinae) include some of the world's most devastating pest species. Whereas the majority of nonpest heliothinae specialize on a single plant family, genus, or species, pest species are highly polyphagous, with populations often escalating in size as they move from one crop species to another. Here, we examine the current literature on heliothine host-selection behavior with the aim of providing a knowledge base for research scientists and pest managers. We review the host relations of pest heliothines, with a particular focus on Helicoverpa armigera (Hubner), the most economically damaging of all heliothine species. We then consider the important question of what constitutes a host plant in these moths, and some of the problems that arise when trying to determine host plant status from empirical studies on host use. The top six host plant families in the two main Australian pest species (H. armigera and Helicoverpa punctigera Wallengren) are the same and the top three (Asteraceae, Fabaceae, and Malvaceae) are ranked the same (in terms of the number of host species on which eggs or larvae have been identified), suggesting that these species may use similar cues to identify their hosts. In contrast, for the two key pest heliothines in the Americas, the Fabaceae contains approximate to 1/3 of hosts for both. For Helicoverpa zea (Boddie), the remaining hosts are more evenly distributed, with Solanaceae next, followed by Poaceae, Asteraceae, Malvaceae, and Rosaceae. For Heliothis virescens (F.), the next highest five families are Malvaceae, Asteraceae, Solanaceae, Convolvulaceae, and Scrophulariaceae. Again there is considerable overlap in host use at generic and even species level. H. armigera is the most widely distributed and recorded from 68 plant families worldwide, but only 14 families are recorded as a containing a host in all geographic areas. A few crop hosts are used throughout the range as expected, but in some cases there are anomalies, perhaps because host plant relation studies are not comparable. Studies on the attraction of heliothines to plant odors are examined in the context of our current understanding of insect olfaction, with the aim of better understanding the connection between odor perception and host choice. Finally, we discuss research into sustainable management of pest heliothines using knowledge of heliothine behavior and ecology. A coordinated international research effort is needed to advance our knowledge on host relations in widely distributed polyphagous species instead of the localized, piecemeal approaches to understanding these insects that has been the norm to date.

We have characterised six Australian Cucumber mosaic virus (CMV) strains belonging to different subgroups, determined by the sequence of their complete RNA 3 and by their host range and the symptoms they cause on species in the Solanaceae, Cucurbitaceae and on sweet corn. These data allowed classification of strains into the known three CMV subgroups and identification of plant species able to differentiate the Australian strains by symptoms and host range. Western Australian strains 237 and Twa and Queensland strains 207 and 242 are closely related members of CMV subgroup IA, which cause similar severe symptoms on Nicotiana species. Strains 207 and 237 (subgroup IA) were the only strains tested which systemically infected sweet corn. Strain 243 caused the most severe symptoms of all strains on Nicotiana species, tomato and capsicum and appears to be the first confirmed subgroup IB strain reported in Australia. Based on pair-wise distance analysis and phylogeny of RNA 3, as well as mild disease symptoms on Nicotiana species, CMV 241 was assigned to subgroup II, as the previously described Q-CMV and LY-CMV.

Although only recently described, Colletotrichum boninense is well established in literature as an anthracnose pathogen or endophyte of a diverse range of host plants worldwide. It is especially prominent on members of Amaryllidaceae, Orchidaceae, Proteaceae and Solanaceae. Reports from literature and preliminary studies using ITS sequence data indicated that C. boninense represents a species complex. A multilocus molecular phylogenetic analysis (ITS, ACT, TUB2, CHS-1, GAPDH, HIS3, CAL) of 86 strains previously identified as C. boninense and other related strains revealed 18 clades. These clades are recognised here as separate species, including C. boninense s. str., C. hippeastri, C. karstii and 12 previously undescribed species, C. annellatum, C. beeveri, C. brassicicola, C. brasiliense, C. colombiense, C. constrictum, C. cymbidiicola, C. dacrycarpi, C. novae-zelandiae, C. oncidii, C. parsonsiae and C. torulosum. Seven of the new species are only known from New Zealand, perhaps reflecting a sampling bias. The new combination C. phyllanthi was made, and C. dracaenae Petch was epitypified and the name replaced with C. petchii. Typical for species of the C. boninense species complex are the conidiogenous cells with rather prominent periclinal thickening that also sometimes extend to form a new conidiogenous locus or annellations as well as conidia that have a prominent basal scar. Angiosperms represent a huge diversity in floral structures. Thus, they provide an attractive target for comparative developmental genetics studies. Research on flower development has focused on few main model plants, and studies on these species have revealed the importance of transcription factors, such as MADS-box and TCP genes, for regulating the floral form. The MADS-box genes determine floral organ identities, whereas the TCP genes are known to regulate flower shape and the number of floral organs. In this study, I have concentrated on these two gene families and their role in regulating flower development in Gerbera hybrida, a species belonging to the large sunflower family (Asteraceae). The Gerbera inflorescence is comprised of hundreds of tightly clustered flowers that differ in their size, shape and function according to their position in the inflorescence. The presence of distinct flower types tells Gerbera apart from the common model species that bear only single kinds of flowers in their inflorescences. The marginally located ray flowers have large bilaterally symmetrical petals and non-functional stamens. The centrally located disc flowers are smaller, have less pronounced bilateral symmetry and carry functional stamens. Early stages of flower development were studied in Gerbera to understand the differentiation of flower types better. After morphological analysis, we compared gene expression between ray and disc flowers to reveal transcriptional differences in flower types. Interestingly, MADS-box genes showed differential expression, suggesting that they might take part in defining flower types by forming flower-type-specific regulatory complexes. Functional analysis of a CYCLOIDEA-like TCP gene GhCYC2 provided evidence that TCP transcription factors are involved in flower type differentiation in Gerbera. The expression of GhCYC2 is ray-flower-specific at early stages of development and activated only later in disc flowers. Overexpression of GhCYC2 in transgenic Gerbera-lines causes disc flowers to obtain ray-flower-like characters, such as elongated petals and disrupted stamen development. The expression pattern and transgenic phenotypes further suggest that GhCYC2 may shape ray flowers by promoting organ fusion. Cooperation of GhCYC2 with other Gerbera CYC-like TCP genes is most likely needed for proper flower type specification, and by this means for shaping the elaborate inflorescence structure. Gerbera flower development was also approached by characterizing B class MADS-box genes, which in the main model plants are known regulators of petal and stamen identity. The four Gerbera B class genes were phylogenetically grouped into three clades; GGLO1 into the PI/GLO clade, GDEF2 and GDEF3 into the euAP3 clade and GDEF1 into the TM6 clade. Putative orthologs for GDEF2 and GDEF3 were identified in other Asteraceae species, which suggests that they appeared through an Asteraceae-specific duplication. Functional analyses indicated that GGLO1 and GDEF2 perform conventional B-function as they determine petal and stamen identities. Our studies on GDEF1 represent the first functional analysis of a TM6-like gene outside the Solanaceae lineage and provide further evidence for the role of TM6 clade members in specifying stamen development. Overall, the Gerbera B class genes showed both commonalities and diversifications with the conventional B-function described in the main model plants.

Groundnut bud necrosis virus (GBNV), a member of genus Tospovirus in the family Bunyaviridae, infects a large number of leguminosae and solanaceae plants in India. With a view to elucidate the function of nonstructural protein, NSs encoded by the small RNA genome (S RNA), the NSs protein of GBNV-tomato (Karnataka) [1] was over-expressed in E.coli and purified by Ni-NTA chromatography. The purified rNSs protein exhibited an RNA stimulated NTPase activity. Further, this activity was metal ion dependent and was inhibited by adenosine 5' (beta, gamma imido) triphosphate, an ATP analog. The rNSs could also hydrolyze dATP.Interestingly, in addition to the NTPase and dATPase activities, the rNSs exhibited ATP independent 5' RNA/DNA phosphatase activity that was completely inhibited by AMP. The 5' alpha phosphate could be removed from ssDNA, ssRNA, dsDNA and dsRNA thus confirming that rNSs has a novel 5' alpha phosphatase activity. K189A mutation in the Walker motif A (GxxxxGKT) resulted in complete loss of ATPase activity, but the 5'phosphatase activity was unaffected. On the other hand, D159A mutation in the Walker motif B (DExx) resulted in partial loss of both the activities. These results demonstrate for the first time that NSs is a bifunctional enzyme, which could participate in viral movement, replication or in suppression of the host defense mechanism.

El ensayo fue realizado en la unidad experimental “El Plantel” de la Universidad Nacional Agraria, ubicada en el kilómetro 42.5 en la carretera Tipitapa - Masaya, mu nicipio de Masaya, departamento de Masaya, durante los meses de Noviembre 2007 a Marzo de 2008, con el objetivo de: Evaluar el comportamiento de las arvenses en el cultivo del maíz, variedad NB - 6, bajo dos sistemas de producción (convencional y orgánico). Éste consistió en el establecimiento de dos sistemas, convencional y orgánico con dos tratamientos, cuatro sub - muestras y cuatro parcelas útiles por cada tratamiento. El tamaño de la parcela experimental fue de 432 m 2 (16 m x 27 m). La distancia entre las parcelas fue de 1 m. Las variables evaluadas en las arvenses fueron: diversidad, abundancia, biomasa , cobertura y en el cultivo fue rendimiento. Las especies de arvenses que predominaron en ambos sistemas fueron similares encontrándose siete familias repr esentadas por siete especies, la mayoría de ellas pertenecientes a la clase dicotiledóneas con cinco especies. Entre las dicotiledóneas se encontraron Sida acuta Burn. F, Physalis angulata (L), Amaranthus spinosus , Portulaca oleracea (L) y Ricinus comunis (L) . L as monocotiledoneas estuvieron representadas por las familias Cyperaceae y Poaceae, predominando las especies Ixophorus unicetus y Cyperus rotundus. En el sistema orgánico hubo mayor cantidad de individuos. En el comportamiento de la biomasa hubo var iación en los diferentes muestreos, encontrándose mayor cantidad de biomasa en el sistema orgánico con 2,261.17 g/m 2 y en el sistema convencional con 1368.90 g/m 2 . En el sistema orgánico la familia que presentó mayor acumulación de biomasa fueron las Poace ae con 915.18 g/m 2 y la que obtuvo menor biomasa fueron las Solanaceae con 46.30 g/m 2 . Las arvenses en e l sistema orgánico presentaron mayor porcentaje de cobertura en los cuatro muestreos realizados; en los primeros 15 días después de l a siembra se observ ó un 24% y 19% en el sistema convencional. En el rendimiento h ubo diferenci as estadísticas significativas en ambos sistemas de manejo. El mayor rendimiento fue obtenido por el sistema orgánico con 3,327.50 kgha - 1 , el sistema convencional con 1,963.80 kgh a - 1 esto representó para el sistema orgánico un 25.77% de rendimiento mayor que el sistema convencional

El presente trabajo se realizó en la comunidad Amak, zona de régimen especial Mayangna Sauni Bu Reserva Biósfera BOSAWAS, Nicaragua; con el fin de recopilar el uso popular sobre plantas medicinales en el tratamiento de enfermedades en animales domésticos. Fueron entrevistados 15 informantes clave entre curanderos, parteras y veterinarios empíricos, que utilizan las plantas medicinales en la práctica habitual. Después del análisis de las entrevistas se encontró una amplia gama de indicaciones terapéuticas sobre plantas medicinales utilizadas para el tratamiento de enfermedades en los animales. Se identificaron 42 plantas pertenecientes a las familias: Fabácea, Tiliácea, Malvácea, Solanaceae, Begoniaceae, Rubiaceae y Cucurbitácea. Las partes utilizadas son hojas, flores, corteza, frutos, raíces además de la manera de preparación que acompañan a los diversos tratamientos. La introducción de fármacos de síntesis química a la comunidad ha ido desplazando la práctica etnobotánica, no obstante las personas con mayor riqueza de conocimientos van transmitiendo de manera oral todos estos saberes. Esto permitió obtener una riqueza del manejo etnobotánico como saber popular en la práctica de la salud animal.

本文研究了茄科天仙子族7属以及有亲缘关系的颠茄属、茄参属和枸杞属的形态学、解剖学、孢粉学、种子形态学、胚胎学、花器官发生、分子系统学和分支系统学;论述了它们的结构、分化和系统关系。主要内容包括: 1.解剖学 在光学显微镜和扫描电镜下,观察了茄科天仙子族(Hyoscyameae)7属21种及邻近4属4种共25种植物的叶表皮特征,结果表明:气孔器在各种植物的上、下的表皮均有分布,多为无规则型,也有不等细胞型;叶表皮细胞形状有不规则形、十字形、近长圆形和不规则多边形,垂周壁较平直或各种波状。叶表皮气孔器外拱盖内缘近平滑、浅波状或波状;角质膜多为具条纹,也有同时具条纹和颗粒或鳞片,偶尔仅具颗粒和鳞片或光滑。气孔器类型可以作为区分部分属的依据之一。颠茄叶表皮细胞形状和垂周壁式样在一些属和种中有一定的规律性,并与外部形态、孢粉学性状或地理分布相关;叶片表面角质膜的特征对属和种的划分也有一定意义。 2.种子形态学 在扫描电子显微镜下首次全面研究了天仙子族及颠茄属和茄参属等共10属25种植物的种子形态学特征,结果表明: 该族植物种子表面纹饰有脑纹状或近脑纹状和网状两大类,除了山莨菪属Anisodus和Hyoscyamus.pusillus 外,其余类群均为网状。外种皮细胞形状和周壁式样有两大类,一类为多边形至近圆形,等径,周壁近平直,另一类为形状不规则,常不等径,周壁为各式波状;外种皮细胞周壁内侧有皱波状、瘤状、小颗粒、片状或小刺等各式附属物。各属植物的种子大小、形状及种脐着生位置的系统学意义不大。但是,种脐突出与否、种子外种皮细胞形状和周壁式样,尤其是周壁内侧附属物等性状具有较大的分类学意义。 3.孢粉学 在扫描电镜下研究了该族7属及其它3属共10属23种植物的花粉形态学特征。结果表明:天仙子族的花粉粒形状为球形、近球形、扁球形和长球形;萌发孔类型有无萌发孔、不规则的拟孔、3沟、4 沟、3- 4沟、散沟和3孔沟;外壁纹饰为小刺状、瘤状、各式条纹状、条纹-穴状、条纹-网状、皱波状、细网状和网状。沟膜近光滑、具小颗粒、具小刺状突起或具瘤状突起。各属植物的花粉形态在萌发孔的有无、萌发孔的类型和外壁纹饰等方面有较大的差异,可以作为探讨属间分类和系统关系的重要依据。天仙子族植物花粉萌发孔的演化趋势为:无萌发孔→ 3、4沟→3孔沟。无萌发孔的山莨菪属是原始类群,三孔沟的马尿泡和天仙子等属是进化类群。 4.胚胎学 本论文首次研究了天仙子族山莨菪Anisodus tanguticus的胚胎学特征,结果表明:山莨菪的小孢子形成为同时型,腺质绒毡层;胚珠半倒生,单珠被,薄珠心;蓼型胚囊;其胚胎发生类型可能为茄型;胚乳形成为细胞型。山莨菪的胚胎学性状与马尿泡十分相似。胚胎学证据支持山莨菪属与马尿泡属关系密切的观点。 5.花器官发生 首次全面研究了天仙子族的山莨菪属(山莨菪)、马尿泡属(马尿泡)、天仙子属(天仙子)及其邻近2属共5种植物的花器官发生特征,结果表明:山莨菪和天仙子的5个花萼裂片原基突起最早发生,且几乎是近同时环状发生,其排列方式从花发育早期直至花成熟后始终为轮状排列。而马尿泡和颠茄首先是5个花萼裂片原基突起依次螺旋状发生,但在其5个花萼裂片原基都出现后,其排列方式仍为轮状排列。不同之处是:马尿泡的花萼裂片原基在其花冠裂片原基刚刚不明显地发生时,就已经在基部联合并形成了一个环,而颠茄的花萼裂片原基直到雄蕊原基突起开始发生并较明显时,花萼裂片原基仍然是分离的,当花顶的中央开始出现很不明显的雌蕊原基的凹陷时,花萼裂片原基才开始联合。青海茄参与其它4种植物的不同之处在于柱头2裂或3裂,而后4者的柱头均为2裂。花器官发生的特征表明马尿泡与天仙子和山莨菪的关系较近。 6.基于叶绿体DNA trnL-F序列和核糖体DNA ITS序列的初步分析 以宁夏枸杞作为外类群,对代表天仙子族及其邻近3属的10种植物的ITS区和trnL-F序列进行了测定,将这两个序列结合构建系统树。将gaps作为fifth base处理时,马尿泡属和茄参属构成最基部的一支,赛莨菪属和天仙子属构成一支然后与泡囊草属构成姐妹群,另外一支由天蓬子属、颠茄属和山莨菪属组成,其中天蓬子属与颠茄属构成一支后与山莨菪属构成姐妹群。分子证据只是一个尝试,还不足以说明天仙子族各属的系统关系。 7.分支分析 以枸杞属为外类群,基于形态学、叶表皮、种子形态学、孢粉学等特征对天仙子族7属及颠茄属和茄参属进行了分支分析。结果表明:目前公认的天仙子族7属构成一个单系类群,而颠茄属和茄参属并没有包括在内,它们两者构成一支,并与天仙子族形成姐妹群的关系。这与该族传统的分类结果相一致。 8.天仙子族的分化和系统关系 基于上述研究结果,结合天仙子族各属的现代地理分布,讨论了各属的分化和系统关系。认为根据传统分类包括有7个属的天仙子族是一个自然的单系类群,目前还没有足够的证据说明是否应该将颠茄属和茄参属放在天仙子族中。
