Research with soybean seeds has revealed that the results of the electrical conductivity test may be influenced by storage temperature, particularly low temperature, such as 10ºC, suggesting that seed deterioration at low storage temperatures does not seem to be directly related to the loss of the cell membrane integrity. This study was conducted with seeds of two soybean cultivars with the objective of: a) studying the effect of different storage temperatures (10ºC; 20ºC; 25ºC; 20/10ºC and 25/10ºC) on the results of the electrical conductivity test; b) observing the behavior of fatty acids and carbohydrates during storage and studying its relation with the electrical conductivity results. Every three months, from a total of 18 months of storage, the physiological quality of seeds was evaluated using the germination, accelerated aging and electrical conductivity tests. Based on the obtained results, it can be concluded that the electrical conductivity test was not shown to be a good indicative of the deterioration process of seeds stored at low temperatures, and no direct relationship between changes in the fatty acids and carbohydrates and the behavior of the mentioned test for seeds stored at 10ºC was found.

This experiment viewed to evaluate the physiological quality of grain sorghum seeds as well as to determine the respective drying curve of each of three drying methods. The seeds harvested at 18.9%, 18.1%, and 18.2% of moisture content were submitted to the following drying methods : a) under natural conditions, b) an intermittent dryer in which the combustion of firewood was the source of caloric energy, and c) a stationary dryer in which the source of caloric energy was the burning of liquefied petroleum gas. The experimental design was a completely randomized one with 25 repetitions of one hundred seeds each. The water contents and weight of one thousand seeds were evaluated. Seeds physiological quality was evaluated by germination and vigor tests. Seed drying rates were of 0.11, 1.25, and 0.55 percent points per hour (pph -1) for the natural, intermittent and stationary drying methods, respectively. The intermittent treatment permits the highest loss of water in the shortest period of time, and germination and vigor remaining unchanged.

Hot and dry weather conditions during soybean [Glycine max (L.) Merrill] seed maturation can cause forced maturation of the seed, resulting in the production of high levels of green seed, which may be detrimental to seed germination. These stressful conditions were imposed on soybean plants during seed maturation to investigate the production of green seeds and seed quality. Plants of the CD 206 cultivar were grown in a greenhouse until the R5.5 growing stage and then transferred to phytotrons at R6 and R7.2 for stress induction. Plants were subjected to two temperature regimes, high (28ºC to 36ºC) and normal (19ºC to 26ºC), and four soil water availability conditions, control (adequate water supply), 30% gravimetric moisture (GM), 20% GM and no water supply. Seed were harvested at R9. Green seed percentages and 100-seed weights from the lower, middle and upper thirds of each plant were determined. Seed quality was assessed by germination, tetrazolium (viability and vigor) and electrical conductivity tests. Occurrence of green seed varied from 9% to 86%, depending on the severity of the stresses imposed. High temperature, coupled with no water supply at R6, resulted in a pronounced occurrence of green seeds. There was no difference in the percentage of green seeds among the plant segments. Seed quality was negatively affected by the incidence of green seeds. A procedure for screening soybean genotypes in a phytotron for their tolerance and/or susceptibility to the production of green seeds was developed.

The impermeability of seed coat to water is common mechanism in Fabaceae seeds. Treatments to overcome hardseededness include scarification with sulphuric acid, scarification on abrasive surface and soaking in water among others. The objective of this study was to identify an effective method to overcome dormancy in Dinizia excelsa seeds. A pre-test (untreated seed) and three experiments were carried out: immersion of seeds in acid sulphuric for 10, 20, 30, 40, 50 and 60min (experiment 1); scarification on abrasive surface at the positions distal end, near of the mycrophyle and on the lateral tissue and tegument clipping at 1mm of the distal end, near of the mycrophyle and on the lateral tissue (experiment 2); scarification on abrasive surface and immersion in water for 0, 12, 24 and 48h (experiment 3). The experimental design was completely with four replications of 50 seeds for each treatment. The statistical analysis was carried out by ANOVA and regression analysis. Seedlings emergence on untreated seeds started on the 8th day after sowing and reached 52.5% on the 1,709th day. In general, the treatments to overcome dormancy increase emergence. Emergence was higher for seeds treated with sulphuric acid for 20 and 30min with emergence of 93.6% and 86.6%, respectively. For seeds scarified on abrasive surface higher emergences were recorded for scarification on distal end, near of the mycrophyle and on the lateral, 82.7%, 74.3% and 75.7%, respectively. Seeds scarified manually showed higher emergence when not immersed in water (75%), or when immersed for 12 and 24h (75%, 73.6% and 65.6%, respectively). Immersion seeds in sulphuric acid for 20 and 30min and scarification on abrasive surface of distal end are effective to overcome dormancy in D. excelsa.

The objective in this research was to evaluate the isoflavone content and the physiological quality of seed from conventional and transgenic soybean cultivars before and after 180 days of storage. Twenty one soybean cultivars: CD 202, CD 206, CD 208, CD 213RR, CD 214RR, CD 215, CD 216, CD 217, CD 218, CD 221, BRS 184, BRS 185, BRS 214, BRS 244RR, BRS 245RR, BRS 246RR, BRS 255, BRS 257, BRS 258, BRS 261 and BRS 262, grown in the 2005/2006 crop season, were assayed. The seeds were packed in Kraft paper bags and stored at room temperature under laboratory conditions. Seeds were evaluated with respect to their germination and vigor (first germination count, accelerated aging and tetrazolium test) and their total isoflavone contents and respective aglycon forms (daidzein, genistein and glycitein),glycosides (daidzine, genistine and glycitine) and malonyl conjugates. A completely randomized block design with six replications with the treatments set out within a subplot scheme (21 cultivars x 2 storage periods) was used. The F-test was used to compare means between storage periods and the Scott-Knott test to compare cultivars for each storage period, both with a 95% probability. It was concluded that isoflavone contents differ between cultivars and show a distinct behavior throughout storage.

Seed vigor is one of the factors that determines the speed and uniformity of seedling emergence and initial plant growth, which are crucial aspects in the competition against weeds. The objective of this study was to verify the direct effects of soybean seed vigor on weed competition and grain yield. A field experiment was conducted with three seed lots of the "Conquista" cultivar, previously characterized by physiological tests as having high, intermediate and low vigor. The experiment was divided into weeded and unweeded plots with six treatments. Crop plant height, weed dry mass accumulation and crop grain yield data were recorded. Seed vigor did not affect plant height and grain yield. Plants which developed from seeds with high and intermediate vigor showed the best results for competition against weeds, reducing weed dry mass accumulation. Plants which developed from high vigor seeds give the best results for grain yield for both weeded and unweeded treatments.

Schinus terebinthifolius is a native forest species which may be used in recuperating degraded areas and in urban afforestation. In order to evaluate the efficiency of methods for separating lots of S. terebinthifolius seeds into vigor levels, five lots of seeds were qualitatively evaluated for their moisture content, germination, first count, accelerated aging (the traditional and the saturated salt methods), germination speed index, average time of germination and seedling length and dry weight. The results showed that the tests of first germination count, accelerated aging at 41 ºC for 72 hours, average time of germination, seedling length and dry matter permitted the separation of S. terebinthifolius seeds into different vigor levels.

The effect of chemical and biological treatments on castor bean emergence, seedling vigor, dry matter production, and also the control of microorganisms associated with seeds of the AL Guarany 2002 and Lyra cultivars, was evaluated. The products tested were carbendazim + thiram, carboxin + thiram and a product based on Trichoderma. Total seed and seedling emergence were evaluated at 27 days after sowing whereas dry matter production was verified for plants removed 45 days after sowing. The Guarany 2002 AL cultivar had a higher incidence of microorganisms than the Lyra cultivar. The chemical treatment was 100% effective in controlling fungi but the biological treatment did not reduce microorganism incidence on the seeds. Chemical treatment resulted in plants with more dry matter and the best results were for carbendazim + thiram and carboxin + thiram at doses of 60 g + 140 g and 50 g + 50 g/100 kg of seeds, respectively. The carbendazim + thiram mixture was the only treatment which was statistically higher for total emergence whereas the biological treatment increased emergence only for the Lyra cultivar, thus demonstrating its lower efficiency. The importance of fungicides to control pathogens associated with seeds was discussed.

The development of new procedures for quickly obtaining accurate information on the physiological potential of seed lots is essential for developing quality control programs for the seed industry. In this study, the effectiveness of an automated system of seedling image analysis (Seed Vigor Imaging System - SVIS) in determining the physiological potential of sun hemp seeds and its relationship with electrical conductivity tests, were evaluated. SVIS evaluations were performed three and four days after sowing and data on the vigor index and the length and uniformity of seedling growth were collected. The electrical conductivity test was made on 50 seed replicates placed in containers with 75 mL of deionised water at 25 ºC and readings were taken after 1, 2, 4, 8 and 16 hours of imbibition. Electrical conductivity measurements at 4 or 8 hours and the use of the SVIS on 3-day old seedlings can effectively detect differences in vigor between different sun hemp seed lots.

The X-ray test is a precise, fast and non-destructive method to detect mechanical damage in seeds. In the present study, the efficiency of X-ray analysis in identifying the extent of mechanical damage in sweet corn seeds and its relationship with germination and vigor was evaluated. Hybrid 'SWB 551' (sh2) seeds with round (R) and flat (F) shapes were classified as large (L), medium (M1, M2 and M3) and small (S), using sieves with round and oblong screens. After artificial exposure to different levels of damage (0, 1, 3, 5 and 7 impacts), seeds were X-rayed (15 kV, 5 min) and submitted to germination (25 °C/5 days) and cold (10 °C/7 days) tests. Digital images of normal and abnormal seedlings and ungerminated seeds from germination and cold tests were jointly analyzed with the seed X-ray images. Results showed that damage affecting the embryonic axis resulted in abnormal seedlings or dead seeds in the germination and cold tests. The X-ray analysis is efficient for identifying mechanical damage in sweet corn seeds, allowing damage severity to be associated with losses in germination and vigor.

Coffee seeds have slow and irregular germination, losing fast their viability during storage, and the standard germination test of these seeds requires at least 30 days. Besides, the results may not reflect the actual physiological quality of these seeds. The objective of this work was to develop a fast and practical test for evaluating the viability of coffee seeds, which is based on the interpretation of different color hues of exudates from seeds. Coffee seeds of the cultivar Catuai 44 from six lots were submitted to germination, accelerated aging, and electrical conductivity tests. In the exudates color hue test, coffee seeds without the parchment and the silvery pellicle (four replications of 10 seeds each) were distributed on top of paper towels moistened and then maintained into a germinator, at 25 ºC for 24, 48, 72, 96, and 120 h. Three classes of color hues were established: colorless, light color hue, and dark color hue, assigning the values of 0, 1, and 3, for each class, respectively. The proposed exudates color hue test can be recommended for the fast assessment of viability for coffee seeds. The most promising results were obtained for seeds with 12% moisture content, after imbibition periods of 72, 96, and 120 h; and with 30% moisture content, after imbibition periods of 72 and 120 h.

The Leguminosae family is one of the most representative botanical families of the Caatinga, with 80 endemic species, highlighting the catingueira (Poincianella pyramidalis). The objective of this research work was to study the maturation process of P. pyramidalis seeds based on the physiological maturity. Five harvest of fruits and seeds were carried out, with 15 days interval each, in a period from July to September 2010. The harvests began 75 days after anthesis (d.a.a.) and lasted until 135 d.a.a. Fruits and seeds were subjected to the following assessments: size, moisture content, and dry mass of fruits and seeds; and germination and vigor of seeds (first count of germination, germination speed index, length and dry mass of seedling). Under the environmental conditions of municipality of Soledade, State of Paraiba, Northeast Brazil, the point of physiological maturity of P. pyramidalis seeds occurs at 125 d.a.a., when the maximum accumulation of dry mass is 1.993 g and moisture content is 21%. The ideal point of harvest is between 130 d.a.a and 135 d.a.a., before natural dehiscence, when the moisture content of seeds is between 13.0% and 5.0%.

The objective of this study was to evaluate physiological quality, content, and activity of antioxidants, in soybean seeds subjected to accelerated aging during different periods. Seeds of cultivars BRS 258, BRS 262 and BRS 268, subjected to accelerated aging during 12, 24, 36 and 48 hours and non-aged seeds were used. After each aging period, the seed were evaluated by tests of: germination; first count and tetrazolium. The total of phenolic compounds, total flavonoides, total of isoflavones, and activity for eliminating ABTS●+ radicals were quantified. There were differences among cultivars according to vigor and viability only after seeds were aged. Cultivars BRS 158 and BRS 268 have shown better seed physiological quality in each aging period; however, not presenting higher amounts of isoflavones and efficiency in removing free radicals. For all cultivars, the values for total of phenolic compounds, as well as total of flavonoids have shown quadratic positive behavior; the values for isoflavones remained constant and the vigor and viability showed contrary trend to activity of antioxidant agents.

In Brazil, although the coffee plantations are predominantly grown under full sunlight, the use of agroforestry systems can lead to socioeconomic advantages, thus providing a favorable environment to the crop by promoting its sustainability as well as environmental preservation. However, there is a lack of information on physiological quality of the coffee seeds produced under different levels of solar radiation. Within this context, the objective of this study was to evaluate the influence of different levels of solar radiation and maturation stages on the physiological quality of coffee (Coffea arabica L.) seeds, cv. Acaiá Cerrado MG-1474. Three levels of solar radiation (plants grown under full sunlight; under plastic screens of 35% shading; and under plastic screens of 50% shading) and three maturation stages (cherry, greenish-yellow and green) were assessed. Physiological quality of seeds was assessed by using germination test, first count of germination, abnormal seedlings, dead seeds, and seedlings with open cotyledonary leaves. Electrophoretic analysis of isoenzymes catalase, esterase, superoxide dismutase and peroxidase was also performed. With the evolution of development the coffee seeds presents increases on physiological quality, and at its beginning the seeds show improvements on quality with the reduction of solar radiation.
