The pathogenic members of the picornavirus superfamily have adverse effects on humans, their crops and their livestock. As structure is related to function, detailed structural studies on these viruses are important not only for fundamental understanding of the viral life cycle, but also for the rational design of vaccines and inhibitors for disease control. These viruses have positive sense, single-stranded RNA genomes enclosed in a protein capsid. X-ray crystallography and cryo-electron microscopy studies have revealed that the isometric members of this group have icosahedrally-symmetric capsids made up of 60 copies of each of the structural proteins. The members that infect animal cells often employ one or more cellular receptors to facilitate cell entry which in some cases is known to initiate the uncoating sequence of the genome. The nature of the interactions between individual viruses and alternative cellular receptors has rarely been probed. The capsid assembly of the members of the picornavirus superfamily is considered to be cooperative and the interactions of RNA and capsid proteins are thought to play an important role in orchestrating virus assembly. The major aims of this thesis were to solve the structures of blackcurrant reversion virus (BRV), human parechovirus 1 (HPEV1) and coxsackievirus A7 (CAV7), as well as the structure of HPEV1 complexed with two of its cellular receptors using cryo-electron microscopy, three-dimensional image reconstruction and homology modeling. Each of the selected viruses represents a taxonomic group where little or no structural data was previously available. The results enabled the detailed comparison of the new structures to those of known picornaviruses, the identification of surface-exposed epitopes potentially important for host interaction, the mapping of RNA-capsid protein interactions and the elucidation of the basis for the specificity of two different receptor molecules for the same capsid. This work will form the basis for further studies on the influence of RNA on parechovirus assembly as a potential target for drug design.

Neste estudo investigamos as consequências da restrição protéica materna durante a lactação sobre a resposta de timócitos da prole jovem de ratos Wistar (grupo D), identificando o papel da leptina nas alterações encontradas. Observamos que, quando comparados ao grupo controle, os animais do grupo D apresentaram, aos 30 dias de vida, uma diminuição significativa tanto do peso corporal quanto do timo. Contudo, não observamos alterações no número de timócitos, no perfil de células CD4/CD8 ou na resposta proliferativa destas células. Sistemicamente, o grupo D não apresentou alterações nos níveis séricos de corticosterona ou no conteúdo nuclear do seu receptor (GR) em timócitos. Apesar dos animais D não apresentarem alterações nos níveis circulantes de leptina, a expressão do seu receptor, ObRb, estava aumentada nos timócitos. Esta alteração foi acompanhada pela amplificação da resposta de sinalização da leptina nestas células, observada por um aumento na ativação de JAK2 e STAT3 após a incubação com leptina. Os timócitos isolados do grupo D apresentaram uma diminuição significativa na taxa de apoptose espontânea quando comparados ao grupo controle. Corroborando estes resultados, demonstramos que os timócitos dos animais D apresentam um aumento na expressão da proteína antiapoptótica Bcl-2 e uma redução da expressão da proteína próapoptótica Bax, além de um maior conteúdo de Pró-caspase-3, entretanto, não encontramos alterações no conteúdo de Bad. Além disso, timócitos do grupo D apresentaram um maior conteúdo da subunidade p65 do NFĸB no núcleo, associado a uma menor expressão de IĸBα no citoplasma. Finalmente, observamos um aumento na expressão do RNAm para o gene ob (leptina) mas não para o gene db (receptor) no microambiente tímico dos animais D. Em conjunto, nossos dados mostram que a restrição protéica durante a lactação afeta a homeostase tímica, induzindo uma maior atividade de leptina, que protege os timócitos da apoptose na prole jovem, sugerindo que esses animais poderiam ser mais suscetíveis a alterações na resposta imune na vida aduta.

Estreptococos do grupo B (EGB) é a principal causa de sepse e meningite neonatal e tem sido recentemente reconhecido como patógeno responsável por infecções invasivas em adultos imunocomprometidos (idosos ou portadores de doenças crônicas). Os EGB produzem inúmeras enzimas extracelulares, várias das quais interagem com o sistema imune do hospedeiro e são importantes durante a interação EGB-hospedeiro, bem como para o desenvolvimento da doença. Estudos anteriores mostraram que metaloproteases estão envolvidas em várias vias metabólicas em diferentes tipos celulares. Por esta razão, nós decidimos investigar o possível envolvimento de metaloproteases de EGB durante a interação celular e apoptose/necrose induzida pelo micro-organismo em células endoteliais da veia umbilical humana (HUVEC) e da linhagem de epitélio respiratório (A549). Tratamento de EGB com inibidores de metaloproteases (EDTA, EGTA e FEN) não induziu alterações no crescimento bacteriano, mas promoveu alterações na expressão de proteínas de superfície, capacidade adesiva e perfil de sobrevivência intracelular do patógeno. O EGB e o sobrenadante do crescimento bacteriano (meio condicionado; MC) promoveram a morte das células HUVEC e A549. Contudo, o tratamento com inibidores de metaloproteases restauraram a viabilidade celular induzida pelos EGB e o MC, sugerindo que metaloproteases bacteriana estão envolvidas no rompimento da barreira celular, promovendo a disseminação bacteriana. Este trabalho descreve pela primeira vez apoptose e necrose induzidas pelo EGB e MC em HUVEC e células A549 após 24h de incubação, respectivamente. Nós também observamos redução da pró-caspase-3 após infecção das HUVEC com EGB e MC, sugerindo ativação da caspase-3. Além disso, o aumento da expressão da proteína pró-apoptótica Bax e diminuição dos níveis da proteína anti-apoptótica Bcl-2 em HUVEC, demonstram o envolvimento do mecanismo apoptótico mitocondrial (via intrínseca). A melhor compreensão das bases moleculares da patogênese do EGB contribui para identificar novas moléculas bacterianas e hospedeiras que podem representar novos alvos terapêuticos ou imunoprofiláticos contra a doença causada por esse patógeno neonatal.

A hiperplasia prostática benigna (HPB) é a doença na qual a próstata demonstra um crescimento anormal e sua prevalência aumenta com o envelhecimento. Leptina, a adipocina mais notável, tem um importante papel na regulação do sistema reprodutivo. Esse trabalho tem por fim avaliar o papel da leptina no tecido prostático humano, utilizando a cultura de tecido in vitro, avaliando a proliferação celular e a expressão dos genes do fator de crescimento do fibroblasto 2 (FGF2), da enzima aromatase e dos genes apoptóticos. De 2009 a 2011, amostras de tecido hiperplásico humano foram obtidas pela prostatectomia transvesical em quinze pacientes com próstatas de volume aumentadas. Cada amostra foi dividida em quatro partes simétricas, mantidas no meio RPMI suplementado com soro fetal bovino a 10%, e 1ng/mL de gentamicina, adicionados a 16 ng/mL de leptina (Leptina) ou não (Controle). Após três horas a expressão dos genes de FGF2, aromatase, Bax, Bcl-x e Bcl-2 foram avaliados por RT-PCR em tempo real. A proliferação celular foi avaliada por imunohistoquímica para PCNA. O tratamento com leptina levou a um aumento na expressão de Bax (C=0.40.1; L=0.90.2; p<0.05), enquanto as expressões de Bcl-2 (C=19.95.6; L=5.61.8; p< 0.05) e Bcl-x (C=0.20.06; L=0.070.02; p<0.05) foram significativamente reduzidas. Não houve alteração significativa na expressão de FGF2, enquanto a expressão da aromatase foi significativamente (C=1.90.6; L=0.40.1; p<0.04) reduzida. A leptina também levou a um aumento na proliferação celular (C=21.80.5; L=64.80.9; p<0.0001). Dessa forma, concluímos que a leptina tem um importante papel na manutenção do crescimento fisiológico da próstata, desde que estimula tanto a proliferação celular como a apoptose, com diminuição na expressão do gene da aromatase.

Cytokine-driven signalling shapes immune homeostasis and guides inflammatory responses mainly through induction of specific gene expression programmes both within and outside the immune cell compartment. These transcriptional outputs are often amplified via cytokine synergy, which sets a stimulatory threshold that safeguards from exacerbated inflammation and immunopathology. In this study, we investigated the molecular mechanisms underpinning synergy between two pivotal Th1 cytokines, IFN-γ and TNF-α, in human intestinal epithelial cells. These two proinflammatory mediators induce a unique state of signalling and transcriptional synergy implicated in processes such as antiviral and antitumour immunity, intestinal barrier and pancreatic β-cell dysfunction. Since its discovery more than 30 years ago, this biological phenomenon remains, however, only partially defined. Here, using a functional genomics approach including RNAi perturbation screens and small-molecule inhibitors, we identified two new regulators of IFN-γ/TNF-α-induced chemokine and antiviral gene and protein expression, a Bcl-2 protein BCL-G and a histone demethylase UTX. We also discovered that IFN-γ/TNF-α synergise to trigger a coordinated shutdown of major receptor tyrosine kinases expression in colon cancer cells. Together, these findings extend our current understanding of how IFN-γ/TNF-α synergy elicits qualitatively and quantitatively distinct outputs in the intestinal epithelium. Given the well-documented role of this synergistic state in immunopathology of various disorders, our results may help to inform the identification of high quality and biologically relevant druggable targets for diseases characterised by an IFN-γ/TNF-α high immune signature

PURPOSE: There is now no doubt that bystander signalling from irradiated cells occurs and causes a variety of responses in cells not targeted by the ionizing track. However, the mechanisms underlying these processes are unknown and the relevance to radiotherapy and risk assessment remains controversial. Previous research by our laboratory has shown bystander effects in a human keratinocyte cell line, HPV-G cells, exposed to medium from gamma irradiated HPV-G cells. The aim of this work was to investigate if similar mechanisms to those identified in medium transfer experiments occurred in these HPV-G cells when they are in the vicinity of microbeam irradiated cells. Demonstration of a commonality of mechanisms would support the idea that the process is not artifactual. MATERIALS AND METHODS: HPV-G cells were plated as two separate populations on mylar dishes. One population was directly irradiated using a charged particle microbeam (1 - 10 protons). The other population was not irradiated. Bystander factor-induced apoptosis was investigated in both populations following treatment by monitoring the levels of reactive oxygen species and mitochondrial membrane potential using fluorescent probes. Expression of the anti-apoptotic protein, bcl-2, and cytochrome c were determined, as well as apoptosis levels. RESULTS: Microbeam irradiation induced increases in reactive oxygen species and decreases in mitochondrial membrane potential at 6 h post-exposure, increased expression of bcl-2 and cytochrome c release at 6.5 h and increased apoptosis at 24 h. CONCLUSION: This study shows that similar bystander signalling pathways leading to apoptosis are induced following microbeam irradiation and following medium transfer. This demonstrates that the mechanisms involved are common across different radiation qualities and conditions and indicates that they may be relevant in vivo.

Le virus de l’immunodéficience humaine ou VIH est l’agent qui cause le SIDA. Le VIH donne lieu à une dérégulation dans la production de certaines cytokines qui ont un rôle immunologique très important chez les patients infectés. L’IL-18, autrement nommé facteur inducteur d’IFN-γ, est une cytokine pro-inflammatoire qui affecte le système immunitaire de façon importante. Son activité est régulée par l’"IL-18 Binding Protein" (IL-18BP), une autre cytokine qui se lie avec l’IL-18 et inhibe son activité biologique. Des études ultérieures ont montré des niveaux élevés d’Il-18 chez les patients infectés par le VIH par rapport aux personnes saines. Cependant, aucune étude n’a été réalisée concernant la production d’IL-18BP chez ces patients. Due à sa relevance dans la régulation de l’IL-18, nous avons étudié l’effet de l’infection par le VIH sur l’équilibre entre ces deux facteurs et l’impact de cet équilibre sur l’homéostasie des cellules NK. Nous avons mesuré les taux de l’IL-18 et de l’IL-18BP circulantes dans les sérums des patients infectés par le VIH en les comparants avec le même nombre de personnes saines et séronégatives. Nous avons aussi déterminé le nombre total des différents sous-types de cellules NK et analysé l’activité des cellules NK (Natural Killer). Finalement nous avons cherché à déterminer si l’IL-18 pouvait induire l’apoptose des cellules NK en activant l’expression de Fas ligand. Nos résultats nous démontrent que les patients infectés par le VIH ont trois fois plus d’IL-18 que les donneurs sains. Cependant les niveaux d’IL-18BP sont plus bas chez les patients infectés comparés aux donneurs sains. Alors, le ratio IL-18/IL-18BP est augmenté chez les patients infectés, ce qui entraîne une grande quantité d’IL-18 libre et biologiquement active circulante dans leur organisme. Nos études démontrent que chez ces patients, les concentrations d’IL-18 sont en corrélation négative avec l’activité cytotoxique de leurs cellules NK. Nos études in vitro démontrent que le traitement des cellules NK par l’IL-18 induit de façon fratricide leur apoptose en augmentant l’expression de Fas ligand. Finalement, cette production non coordonnée de ces deux facteurs pourrait contribuer à une immunopathologie induite par l’IL-18 en entraînant une apoptose fratricide des cellules NK qui possèdent un rôle important dans la réponse antivirale. Le dérèglement de l’homéostasie des cellules NK pourrait donc contribuer à la pathogenèse induite par le VIH.

Le mécanisme biologique responsable pour l’augmentation de l’expression de la protéine nestin dans les cellules souches neurales (CSN) du cœur après un infarctus du myocarde (IM) demeure inconnu. Des études antérieures ont démontré que le traitement au dexamethasone, un glucocorticoïde aux propriétés anti-inflammatoires, abolit la régulation positive de nestin après un IM. Ceci suggère un lien avec la réponse inflammatoire. Nous avons vérifié dans cette étude l’hypothèse que la cytokine inflammatoire interleukin-1beta (IL-1beta) peut modifier le phénotype de cellules souches neurales. Le deuxième objectif de l’étude fut d’établir l’impact, suivant un IM, de l’inhibition de la signalisation de IL-1beta sur la fonction et la guérison cardiaque. Suite à une ligature complète de l’artère coronaire du rat mâle, le dysfonctionnement contractile du ventricule gauche fut associé à une régulation positive de la protéine nestin dans le myocarde non-infarci. Le traitement avec Xoma 052 (1 mg/kg), un anticorps anti-IL-1beta, 24h, 7 et 14 jours après un évènement ischémique, eu aucun effet sur la taille de l’infarctus ou la contractilité du ventricule gauche. De plus, le traitement avec Xoma 052 après un IM n’a pu supprimer l’augmentation de l’expression de nestin et Bcl-2 malgré une réduction modeste du niveau de la protéine Bax. Pour déterminer directement le rôle de la réponse inflammatoire en l’absence d’ischémie, nous avons injecté des rats mâles avec du LPS (10mg/kg, 18hrs). Dans le coeur du rat-LPS, nous avons noté une augmentation significative du niveau d’ARNm de IL-1beta et de l’expression de la protéine nestin. Le prétraitement avec 10mg/kg de Xoma 052 a aboli l’augmentation de l’expression de nestin dans le coeur des rats-LPS. Ces observations indiquent que les cellules souches neurales pourraient représenter une cible potentielle de l’IL-1beta.

ZUSAMMENFASSUNG: Proteinkinasen übernehmen zentrale Aufgaben in der Signaltransduktion höherer Zellen. Dabei ist die cAMP-abhängige Proteinkinase (PKA) bezüglich ihrer Struktur und Funktion eine der am besten charakterisierten Proteinkinasen. Trotzdem ist wenig über direkte Interaktionspartner der katalytischen Untereinheiten (PKA-C) bekannt. In einem Split-Ubiquitin basiertem Yeast Two Hybrid- (Y2H-)System wurden potenzielle Interaktionspartner der PKA-C identifiziert. Als Bait wurden sowohl die humane Hauptisoform Cα (hCα) als auch die Proteinkinase X (PrKX) eingesetzt. Nach der Bestätigung der Funktionalität der PKA-C-Baitproteine, dem Nachweis der Expression und der Interaktion mit dem bekannten Interaktionspartner PKI wurde ein Y2H-Screen gegen eine Mausembryo-cDNA-Expressionsbibliothek durchgeführt. Von 2*10^6 Klonen wurden 76 Kolonien isoliert, die ein mit PrKX interagierendes Preyprotein exprimierten. Über die Sequenzierung der enthaltenen Prey-Vektoren wurden 25 unterschiedliche, potenzielle Interaktionspartner identifiziert. Für hCα wurden über 2*10^6 S. cerevisiae-Kolonien untersucht, von denen 1.959 positiv waren (1.663 unter erhöhter Stringenz). Über die Sequenzierung von ca. 10% der Klone (168) konnten Sequenzen für 67 verschiedene, potenzielle Interaktionspartner der hCα identifiziert werden. 15 der Preyproteine wurden in beiden Screens identifiziert. Die PKA-C-spezifische Wechselwirkung der insgesamt 77 Preyproteine wurde im Bait Dependency Test gegen largeT, ein Protein ohne Bezug zum PKA-System, untersucht. Aus den PKA-C-spezifischen Bindern wurden die löslichen Preyproteine AMY-1, Bax72-192, Fabp3, Gng11, MiF, Nm23-M1, Nm23-M2, Sssca1 und VASP256-375 für die weitere in vitro-Validierung ausgewählt. Die Interaktion von FLAG-Strep-Strep-hCα (FSS-hCα) mit den über Strep-Tactin aus der rekombinanten Expression in E. coli gereinigten One-STrEP-HA-Proteinen (SSHA-Proteine) wurde über Koimmunpräzipitation für SSHA-Fabp3, -Nm23-M1, -Nm23-M2, -Sssca1 und -VASP256-375 bestätigt. In SPR-Untersuchungen, für die hCα kovalent an die Oberfläche eines CM5-Sensorchips gekoppelt wurde, wurden die ATP/Mg2+-Abhängigkeit der Bindungen sowie differentielle Effekte der ATP-kompetitiven Inhibitoren H89 und HA-1077 untersucht. Freie hCα, die vor der Injektion zu den SSHA-Proteinen gegeben wurde, kompetierte im Gegensatz zu FSS-PrKX die Bindung an die hCα-Oberfläche. Erste kinetische Analysen lieferten Gleichgewichtsdissoziationskonstanten im µM- (SSHA-Fabp3, -Sssca1), nM- (SSHA-Nm23-M1, –M2) bzw. pM- (SSHA-VASP256-375) Bereich. In funktionellen Analysen konnte eine Phosphorylierung von SSHA-Sssca1 und VASP256-375 durch hCα und FSS-PrKX im Autoradiogramm nachgewiesen werden. SSHA-VASP256-375 zeigte zudem eine starke Inhibition von hCα im Mobility Shift-Assay. Dieser inhibitorische Effekt sowie die hohe Affinität konnten jedoch auf eine Kombination aus der Linkersequenz des Vektors und dem N-Terminus von VASP256-375 zurückgeführt werden. Über die Wechselwirkungen der hier identifizierten Interaktionspartner Fabp3, Nm23-M1 und Nm23-M2 mit hCα können in Folgeuntersuchungen neue PKA-Funktionen insbesondere im Herzen sowie während der Zellmigration aufgedeckt werden. Sssca1 stellt dagegen ein neues, näher zu charakterisierendes PKA-Substrat dar.

Recently we have described an HPMA copolymer conjugate carrying both the aromatase inhibitor aminoglutethimide (AGM) and doxorubicin (Dox) as combination therapy. This showed markedly enhanced in vitro cytotoxicity compared to the HPMA copolymer-Dox (FCE28068), a conjugate that demonstrated activity in chemotherapy refractory breast cancer patients during early clinical trials. To better understand the superior activity of HPMA copolymer-Dox-AGM, here experiments were undertaken using MCF-7 and MCF-7ca (aromatase-transfected) breast cancer cell lines to: further probe the synergistic cytotoxic effects of AGM and Dox in free and conjugated form; to compare the endocytic properties of HPMA copolymer-Dox-AGM and HPMA copolymer-Dox (binding, rate and mechanism of cellular uptake); the rate of drug liberation by lysosomal thiol-dependant proteases (i.e. conjugate activation), and also, using immunocytochemistry, to compare their molecular mechanism of action. It was clearly shown that attachment of both drugs to the same polymer backbone was a requirement for enhanced cytotoxicity. FACS studies indicated both conjugates have a similar pattern of cell binding and endocytic uptake (at least partially via a cholesterol-dependent pathway), however, the pattern of enzyme-mediated drug liberation was distinctly different. Dox release from PK1 was linear with time, whereas the release of both Dox and AGM from HPMA copolymer-Dox-AGM was not, and the initial rate of AGM release was much faster than that seen for the anthracycline. Immunocytochemistry showed that both conjugates decreased the expression of ki67. However, this effect was more marked for HPMA copolymer-Dox-AGM and, moreover, only this conjugate decreased the expression of the anti-apoptotic protein bcl-2. In conclusion, the superior in vitro activity of HPMA copolymer-Dox-AGM cannot be attributed to differences in endocytic uptake, and it seems likely that the synergistic effect of Dox and AGM is due to the kinetics of intracellular drug liberation which leads to enhanced activity. The molecular mechanisms underlying the initiation and control of the release of cytochrome c during mitochondrion-dependent apoptosis are thought to involve the phosphorylation of mitochondrial Bcl-2 and Bcl-x(L). Although the c-Jun N-terminal kinase (JNK) has been proposed to mediate the phosphorylation of Bcl-2/Bcl-x(L) the mechanisms linking the modification of these proteins and the release of cytochrome c remain to be elucidated. This study was aimed at establishing interdependency between JNK signalling and mitochondrial apoptosis. Using an experimental model consisting of isolated, bioenergetically competent rat brain mitochondria, these studies show that (i) JNK catalysed the phosphorylation of Bcl-2 and Bcl-x(L) as well as other mitochondrial proteins, as shown by two-dimensional isoelectric focusing/SDS/PAGE; (ii) JNK-induced cytochrome c release, in a process independent of the permeability transition of the inner mitochondrial membrane (imPT) and insensitive to cyclosporin A; (iii) JNK mediated a partial collapse of the mitochondrial inner-membrane potential (Deltapsim) in an imPT- and cyclosporin A-independent manner; and (iv) JNK was unable to induce imPT/swelling and did not act as a co-inducer, but as an inhibitor of Ca-induced imPT. The results are discussed with regard to the functional link between the Deltapsim and factors influencing the permeability transition of the inner and outer mitochondrial membranes. Taken together, JNK-dependent phosphorylation of mitochondrial proteins including, but not limited to, Bcl-2/Bcl-x(L) may represent a potential of the modulation of mitochondrial function during apoptosis.

Nonsteroidal antiinflammatory drugs (NSAIDs) have been shown to reduce cell growth in several tumors. Among these possible antineoplastic drugs are cyclooxygenase-2 (COX-2)-selective drugs, such as celecoxib, in which antitumoral mechanisms were evaluated in rats bearing Walker-256 (W256) tumor. W256 carcinosarcoma cells were inoculated subcutaneously (10(7) cells/rat) in rats submitted to treatment with celecoxib (25 mg kg(-1)) or vehicle for 14 days. Tumor growth, body-weight gain, and survival data were evaluated. The mechanisms, such as COX-2 expression and activity, oxidative stress, by means of enzymes and lipoperoxidation levels, and apoptosis mediators were also investigated. A reduction in tumor growth and an increased weight gain were observed. Celecoxib provided a higher incidence of survival compared with the control group. Cellular effects are probably COX-2 independent, because neither enzyme expression nor its activity, measured by tumoral PGE(2), showed significant difference between groups. It is probable that this antitumor action is dependent on an apoptotic way, which has been evaluated by the expression of the antiapoptotic protein Bcl-xL, in addition to the cellular changes observed by electronic microscopy. Celecoxib has also a possible involvement with redox homeostasis, because its administration caused significant changes in the activity of oxidative enzymes, such as catalase and superoxide dismutase. These results confirm the antitumor effects of celecoxib in W256 cancer model, contributing to elucidating its antitumoral mechanism and corroborating scientific literature about its effect on other types of cancer.

O objetivo nesta pesquisa foi avaliar o efeito de dois ingredientes protéicos da dieta (farelo de soja e de algodão) e de dois processamentos físicos do concentrado (farelado e extrusado) na terminação de 16 bovinos machos não-castrados da raça Canchim. Avaliaram-se ainda a excreção de nutrientes nos dejetos e o potencial de produção de biogás. Os animais tinham 12 meses de idade e 315 kg PC, em média, e foram confinados em baias individuais durante 147 dias (os primeiros 35 dias foram de adaptação). Como volumoso utilizou-se silagem de milho, em uma relação volumoso:concentrado de 50:50, com base na MS. Os resultados foram analisados em delineamento inteiramente casualizado, em esquema fatorial 2 × 2 (fonte protéica × processamento físico). A fonte protéica influenciou o ganho de peso corporal (1,50 e 1,35 kg/dia para o farelo de soja e de algodão, respectivamente), a conversão alimentar (4,73 e 5,31 kg MS ingerida/kg de ganho de peso, respectivamente) e a eficiência protéica (1,78 e 1,59 kg de ganho de peso/kg PB ingerida, respectivamente). O tratamento físico do concentrado e a fonte protéica não influenciaram a ingestão de MS, a área de olho-de-lombo e a espessura de gordura, estimadas por ultra-som. O balanço de nutrientes foi semelhante entre tratamentos para MS, FDN e FDA, mas a fonte protéica determinou diferenças no balanço de PB. Entre os macro e microminerais quantificados nas fezes, os teores de P e Mg diferiram entre as fontes protéicas e a extrusão aumentou o conteúdo de Ca, com médias de 0,39 e 0,43 g/100 g de MS de dejetos, respectivamente, para os concentrados farelado e extrusado. Os dejetos produziram biogás de maneira efetiva entre o 70º e o 200º dia.

Foi conduzido um experimento para avaliar a utilização de subprodutos de origem animal em dietas para frangos de corte, formuladas com base na proteína bruta ou proteína ideal. O delineamento foi inteiramente casualizado em esquema fatorial 2x2+1, com duas fontes de proteína de origem animal (farinha de vísceras de aves e farinha de sangue bovino), dois conceitos de formulação (proteína bruta e proteína ideal) e uma dieta testemunha à base de milho e farelo de soja, com quatro repetições. As características avaliadas foram ganho de peso, consumo de ração e conversão alimentar. Houve efeito significativo das interações entre fontes de proteína de origem animal e conceitos de formulação para consumo de ração e ganho de peso. Quanto à conversão alimentar, não houve diferença significativa na interação e os melhores valores de conversão alimentar foram encontrados quando se incorporou a farinha de vísceras às dietas. Os melhores ganhos de peso foram obtidos com as dietas com farinha de sangue formuladas com base na proteína bruta e com farinha de vísceras de aves com base na proteína ideal.

Vascular anomalies constitute a distinct group of lesions, but they may present similar clinical and histopatological characteristics, which can lead to diagnostic mistakes. This study aimed by histopathology and immunohistochemical expression of human glucose transporter protein (GLUT-1), correctly identify and classify oral vascular anomalies, besides analyzing the immunoexpression of markers proliferation and apoptosis (Ki-67 and Bcl-2). All cases diagnosed as "oral hemangiomas" belonging to the archives of the Service of Pathological Anatomy from the subject of Oral Pathology of the Department of Dentistry (DOD), of the Federal University of Rio Grande do Norte (UFRN) were reviewed, totalizing 77 cases. Immunohistochemical analysis for GLUT-1 showed that only 26 (33.8%) of the specimens were true infantile hemangiomas (IHs). The 51 (66.2%%) GLUT-1 negative specimens were then reclassified as pyogenic granulomas (PGs) and vascular malformations (VMs) from their histopathologic characteristics,totalizing 26 (33.8%) cases of IHs, 20 (26.0%) of PGs and 31 (40.2) cases of oral VMs. The cases analyzed by the marker Ki-67 showed different median IH (13,85), PG (33,70) and VM (4.55) with statistically significant differences between them (p <0.001). In relation to the protein Bcl-2, the groups also showed different median of the established scores IH (1.00), PG (1.50), VMs (0.0) demonstrating statistically significant differences between them (p<0,001). No statistically significant correlation between the indexes of positivity for Ki-67 and the scores of immunoexpression of Bcl-2 were observed in any group. Thus, we can conclude that it is necessary a careful and parameterized review of cases of vascular anomalies making use of auxiliary tools such as GLUT-1, since the histopathological findings alone, sometimes, are not sufficient to differentiate some anomalies. Furthermore, analysis of the expressions of markers involved in the levels of proliferation of lesions is important for a better understanding of its biological behavior aspect
