In amphibia, steroidogenesis remains quiescent in distinct seasonal periods, but the mechanism by which spermatogenesis is maintained under low steroidogenic conditions is not clear. In the present study, testosterone location in the testes of Rana catesbeiana was investigated immunohistochemically during breeding (summer) and nonbreeding (winter) periods. In winter, the scarce interstitial tissue exhibited occasional testosterone immunopositivity in the interstitial cells but the cytoplasm of primordial germ cells (PG cells) was clearly immunopositive. By contrast, in summer, PG cells contained little or no immunoreactivity whereas strong immunolabelling was present in the well-developed interstitial tissue. These results suggest that PG cells could retain testosterone during winter. This androgen reservoir could be involved in the control of early spermatogenesis in winter and/or to guarantee spermiogenesis and spermiation in the next spring/summer. The weak or negative immunoreaction in the summer PG cells might reflect consumption of androgen reservoir by the intense spermatogenic activity from spring to summer. Thus, besides acting as stem cells, PG cells of R. catesbeiana could exert an androgen regulatory role during seasonal spermatogenesis.

The aim of this study was to investigate the ultrastructural characteristics of primordial follicles after culturing of sheep ovarian cortical slices in the presence of indol acetic acid (IAA), Epidermal Growth Factor (EGF), and FSH. To evaluate ultrastructure of primordial follicles cultured in MEM (control) or in MEM containing IAA, EGF, and FSH, fragments of cultured tissue were processes for transmission electron microscopy. Except in the control, primordial follicles cultured in supplemented media for 6d were ultrastructurally normal. They had oocyte with intact nucleus and the cytoplasm contained heterogeneous-sized lipid droplets and numerous round or elongated mitochondria with intact parallel cristae were observed. Rough endoplasmic reticulum (RER) was rarely found. The granulosa cells cytoplasm contained a great number of mitochondria and abundant RER. In conclusion, the presence of IAA, EGF, and FSH helped to maintain ultrastructural integrity of sheep primordial follicles cultured in vitro. © 2011 Evelyn Rabelo Andrade et al.

In vertebrate species, testosterone seems to inhibit spermatogonial differentiation and proliferation. However, this androgen can also be converted, via aromatase, into estrogen which stimulates spermatogonial differentiation and mitotic activity. During seasonal spermatogenesis of adult bullfrogs Lithobates catesbeianus, primordial germ cells (PGCs) show enhanced testosterone cytoplasm immunoexpression in winter; however, in summer, weak or no testosterone immunolabelling was observed. The aim of this study was to confirm if PGCs express stem cell markers-alkaline phosphatase (AP) activity and GFRα1 (glial-cell-line-derived neurotrophic factor)-and verify whether testosterone is maintained in these cells by androgen receptors (ARs) and/or sex hormone-binding globulin (SHBG) in winter. Furthermore, regarding the possibility that testosterone is converted into estrogen by PGCs in summer, the immunoexpression of estrogen receptor (ER)β was investigated. Bullfrog testes were collected in winter and in summer and were embedded in glycol methacrylate for morphological analyses or in paraffin for the histochemical detection of AP activity. GFRα1, AR, SHBG and ERβ expression were detected by Western blot and immunohistochemical analyses. The expression of AP activity and GFRα1 in the PGCs suggest that these cells are spermatogonial stem cells. In winter, the cytoplasmic immunoexpression of ARs and SHBG in the PGCs indicates that testosterone is maintained by these proteins in these cells. The cytoplasmic immunoexpression of ERβ, in summer, also points to an ER-mediated action of estrogen in PGCs. The results indicate a participation of testosterone and estrogen in the control of the primordial spermatogonia during the seasonal spermatogenesis of L. catesbeianus. © 2012 S. Karger AG, Basel.

The scope of this study was to investigate the grieving experiences of women who terminated pregnancies under judicial authorization, due to life-incompatible fetal malformation. Ten women attended in the Fetal Medicine Department of Botucatu Clinical Hospital participated in the study. Data collection was conducted by means of semi-structured interviews forty days after termination. The interviews were recorded and transcribed in full, with the data analyzed from the thematic content analysis perspective. The results revealed that the mothers sought explanations and meanings for the loss, with religious responses and self-blame being very frequent. The reports were marked by feelings of sadness, longing and sensations of emptiness due to the loss of the child, revealing the need of the mothers to dwell on the issue. The mothers were and continued to be linked to their children; the termination of the pregnancy, although being a choice to minimize the pain of an inevitable loss, did not spare the women from experiences of great suffering. The study includes input for the discussion and planning of health approaches and care for women who terminate their pregnancy due to lethal fetal malformation, by means of judicial authorization.

O luto pode ser compreendido como um processo esperado de adaptação quando de uma perda significativa. Configura-se em uma experiência inevitável e contínua, dada a diversidade de eventos que impõe um ciclo de rompimento e reconstrução ao longo da existência. Contudo, o luto por morte de um ente querido tem sido considerado um potencial agente complicador das condições de saúde e da qualidade do viver. O estudo teve o objetivo de compreender as demandas de pessoas que vivenciam esta modalidade de perda, sendo que o recorte consistiu no desvelamento do "pedido de ajuda", no momento em que buscou o atendimento. Para tanto, a pesquisa foi desenvolvida em um hospital público da cidade de Belém do Pará, onde funcionava um serviço de Pronto Atendimento Psicológico a essa população. Tratou-se de um estudo qualitativo sob o método de análise de conteúdo. Os colaboradores foram cinco mulheres que procuraram atendimento psicológico, no período de maio a agosto de 2008. Os dados foram coletados por meio de entrevistas semi-estruturadas com base em três questões norteadoras: O que aconteceu para que você procurasse esse atendimento psicológico? O que você veio buscar nesse atendimento psicológico? Como foi a sua experiência de atendimento psicológico? Foram destacadas quatro categorias: 1) Quem chegou? Apresentação e perfil socioeconômico das colaboradoras; 2) sobre o ente querido falecido e o processo de luto; 3) Demanda: desvelando o pedido de ajuda; 4) A experiência de atendimento psicológico. Os achados sugerem que a demanda inicial por atendimento psicológico pode condensar "pedidos de ajuda" a permear o movimento pessoal das colaboradoras na atenção e cuidado ao seu sofrimento decorrente da vivência do luto, sendo que corresponderam a pedidos de acolhimento e expressão do pesar; de certificação de morte; de aceitação desse evento e de redução do sentimento de responsabilidade pela morte do ente querido, entre outros. Ressaltou-se ainda, a relevância da oferta de atendimento psicológico às pessoas que atravessam o processo de luto como uma possibilidade de acolhimento para a expressão da dor, do medo da morte, de chorar a saudade, e como um possível caminho de elaboração frente à perda de um ente querido.

Este estudo tem por objetivo compreender como as mães da criança com Fibrose Cística (FC) vivenciam o luto pela perda da saúde do seu filho, considerando que esta ocorrência representa uma ameaça de morte continua à vida da criança, quando da ausência da adesão do tratamento. A Fibrose Cística é uma doença crônica, genética, sem cura e potencialmente letal, com prognóstico reservado, que demanda tratamento de alto impacto e intenso cuidado. A estratégia metodológica fundamentou-se na abordagem clínico qualitativa, com ênfase na análise de conteúdo. Participaram deste estudo onze mães com filho diagnosticado com FC e que se encontrava em acompanhamento ambulatorial no Programa de Assistência de FC, do Hospital Universitário João de Barros Barreto. A coleta de dados foi realizada a partir de um encontro com a mãe para uma entrevista semiestruturada e a realização de dois desenhos com objetivo de compreender o luto destas mães em relação à doença FC de seu filho, suas perdas e significados em relação ao adoecimento da criança. Os resultados mostram que as mães vivenciam o luto pela perda da saúde da criança, desvelando os significados atribuídos a morte e o morrer, confirmado pela hipótese de que a mãe da criança com FC sabe sobre a doença, tem consciência da ameaça de morte e compreende que o tratamento pode proporcionar ao seu filho melhor qualidade de vida. Para elas a proximidade da existência de uma morte iminente traz uma reorganização e mudanças internas e externas, pessoais e familiares que favorecem a ressignificação suas vidas a partir do enfrentamento da doença.

This paper presents the basic trends by which psychoanalytical thought understands the subject of death and mourning: the melancholic identification and the death instinct. The implications of these concepts to the comprehension of death and mourning are discussed, specially showing that the death instinct, differently than a wish to die, must be understood as a traumatic potentiality to the psychic apparatus, which needs to be bound and represented. In this sense, it is argued that the narcissist personality configurations in place on post-modernity lead to violence explosion on the field of culture. One of the results of this process can be traced in the lack of psychic elaboration work of mourning and death, which demonstrate the relevance of the definition of "wide-open death" as a social representation of death nowadays.

In gene-banking, primordial germ cells (PGCs), which are embryonic precursor cells of germ cells, are useful for cryopreservation because PGCs have a potential to differentiate into both eggs and sperm via germ-line chimera. Here, we have established vitrification methods for PGCs cryopreservation using 12- to 17-somite stage embryos in loach, Misgurnus anguillicaudatus, which were dechorionated, removed their yolk and injected with green fluorescent protein (GFP) -nos1 3'UTR mRNA to visualize their PGCs. In order to optimize cryopreservation medium for vitrification, the toxicity of cryoprotectants was analyzed. Different concentrations (2, 3, 4, 5 m) of dimethyl sulfoxide (DMSO), methanol (MeOH), ethylene glycol (EG) and propylene glycol (PG) as cryoprotectants were tested. Then, 5 m DMSO showed significantly-high toxicity. Based on this information, combinations called DMP (2 m (14.2% [v/v]) DMSO, 2 m (8.1% [v/v]) MeOH and 2 m (14.4% [v/v]) PG), DP (2 m (14.2% [v/v]) DMSO and 4 m (28.7% [v/v]) PG) and DE (2.1 m (15% [v/v]) DMSO and 2.7 m (15% [v/v]) EG) were evaluated for their toxicities and efficacy of PGCs cryopreservation using two types of equilibration step: direct immersion of cryopreservation media (one-step) and serial exposure to half and full concentration of cryopreservation media (two-step). Viable PGCs were obtained from post-thaw embryos which were cryopreserved by DP and DE with both 1- and 2-step equilibrations. Despite DP showing the highest toxicity, it gave the highest survival rate of embryonic cells after cryopreservation. When PGCs recovered from vitrified embryos were transplanted into host embryos at the blastula stage, the transplanted PGCs were able to migrate to a host genital ridge similarly as endogenous PGCs. It suggests that our methods could be useful to create a germ-line chimera for the production of gametes from PGCs of cryopreserved embryos.
