In the framework of the European Project for Ice Coring in Antarctica (EPICA), a comprehensive glaciological pre-site survey has been carried out on Amundsenisen, Dronning Maud Land, East Antarctica, in the past decade. Within this survey, four intermediate-depth ice cores and 13 snow pits were analyzed for their ionic composition and interpreted with respect to the spatial and temporal variability of volcanic sulphate deposition. The comparison of the non-sea-salt (nss)-sulphate peaks that are related to the well-known eruptions of Pinatubo and Cerro Hudson in AD 1991 revealed sulphate depositions of comparable size (15.8 ± 3.4 kg/km**2) in 11 snow pits. There is a tendency to higher annual concentrations for smaller snow-accumulation rates. The combination of seasonal sodium and annually resolved nss-sulphate records allowed the establishment of a time-scale derived by annual-layer counting over the last 2000 years and thus a detailed chronology of annual volcanic sulphate deposition. Using a robust outlier detection algorithm, 49 volcanic eruptions were identified between AD 165 and 1997. The dating uncertainty is ±3 years between AD 1997 and 1601, around ±5 years between AD 1601 and 1257, and increasing to ±24 years at AD 165, improving the accuracy of the volcanic chronology during the penultimate millennium considerably.

A Cd2+-selective vibrating microelectrode was constructed using a neutral carrier-based Cd ionophore to investigate ion-transport processes along the roots of wheat (Triticum aestivum L.) and two species of Thlaspi, one a Zn/Cd hyperaccumulator and the other a related nonaccumulator. In simple Cd(NO3)2 solutions, the electrode exhibited a Nernstian response in solutions with Cd2+ activities as low as 50 nm. Addition of Ca2+ to the calibration solutions did not influence the slope of the calibration curve but reduced the detection limit to a solution activity of 1 μm Cd2+. Addition of high concentrations of K+ and Mg2+ to the calibration solution to mimic the ionic composition of the cytoplasm affected neither the slope nor the sensitivity of the electrode, demonstrating the pH-insensitive electrode's potential for intracellular investigations. The electrode was assayed for selectivity and was shown to be at least 1000 times more selective for Cd2+ than for any of those potentially interfering ions tested. Flux measurements along the roots of the two Thlaspi species showed no differences in the pattern or the magnitude of Cd2+ uptake within the time frame considered. The Cd2+-selective microelectrode will permit detailed investigations of heavy-metal ion transport in plant roots, especially in the area of phytoremediation.

Fluid and macromolecule secretion by submucosal glands in mammalian airways is believed to be important in normal airway physiology and in the pathophysiology of cystic fibrosis (CF). An in situ fluorescence method was applied to measure the ionic composition and viscosity of freshly secreted fluid from airway glands. Fragments of human large airways obtained at the time of lung transplantation were mounted in a humidified perfusion chamber and the mucosal surface was covered by a thin layer of oil. Individual droplets of secreted fluid were microinjected with fluorescent indicators for measurement of [Na+], [Cl−], and pH by ratio imaging fluorescence microscopy and viscosity by fluorescence recovery after photobleaching. After carbachol stimulation, 0.1–0.5 μl of fluid accumulated in spherical droplets at gland orifices in ≈3–5 min. In gland fluid from normal human airways, [Na+] was 94 ± 8 mM, [Cl−] was 92 ± 12 mM, and pH was 6.97 ± 0.06 (SE, n = 7 humans, more than five glands studied per sample). Apparent fluid viscosity was 2.7 ± 0.3-fold greater than that of saline. Neither [Na+] nor pH differed in gland fluid from CF airways, but viscosity was significantly elevated by ≈2-fold compared to normal airways. These results represent the first direct measurements of ionic composition and viscosity in uncontaminated human gland secretions and indicate similar [Na+], [Cl−], and pH to that in the airway surface liquid. The elevated gland fluid viscosity in CF may be an important factor promoting bacterial colonization and airway disease.

The microbiota of multi-pond solar salterns around the world has been analyzed using a variety of culture-dependent and molecular techniques. However, studies addressing the dynamic nature of these systems are very scarce. Here we have characterized the temporal variation during 1 year of the microbiota of five ponds with increasing salinity (from 18% to >40%), by means of CARD-FISH and DGGE. Microbial community structure was statistically correlated with several environmental parameters, including ionic composition and meteorological factors, indicating that the microbial community was dynamic as specific phylotypes appeared only at certain times of the year. In addition to total salinity, microbial composition was strongly influenced by temperature and specific ionic composition. Remarkably, DGGE analyses unveiled the presence of most phylotypes previously detected in hypersaline systems using metagenomics and other molecular techniques, such as the very abundant Haloquadratum and Salinibacter representatives or the recently described low GC Actinobacteria and Nanohaloarchaeota. In addition, an uncultured group of Bacteroidetes was present along the whole range of salinity. Database searches indicated a previously unrecognized widespread distribution of this phylotype. Single-cell genome analysis of five members of this group suggested a set of metabolic characteristics that could provide competitive advantages in hypersaline environments, such as polymer degradation capabilities, the presence of retinal-binding light-activated proton pumps and arsenate reduction potential. In addition, the fairly high metagenomic fragment recruitment obtained for these single cells in both the intermediate and hypersaline ponds further confirm the DGGE data and point to the generalist lifestyle of this new Bacteroidetes group.

Em invertebrados eurialinos, a exposição a metais pode induzir distúrbios respiratórios, iônicos e osmóticos, bem como estresse oxidativo. Diversos estudos sobre o efeito combinado da salinidade da água e a exposição a metais em invertebrados estuarinos estão relatados na literatura, porém a maioria destes estudos estão focados em apenas alguns metais como Cd, Cu, Pb e Zn. Entretanto, poucos estudos avaliaram as respostas bioquímicas e fisiológicas de invertebrados eurialinos à exposição ao Ni em diferentes salinidades. No presente estudo, o caranguejo estuarino Neohelice granulata foi mantido sob condições controle (sem adição de Ni na água) ou exposto (96 h) a concentrações subletais de Ni (100 e 1000 µg/L) em duas salinidades (2 e 30). Após exposição, o consumo de oxigênio corporal foi medido e amostras de tecidos (hemolinfa, hepatopâncreas, músculo, e brânquias anteriores e posteriores) foram coletadas para análises posteriores. A concentração osmótica e a composição iônica (Na+ , Cl- , Ca2+, Mg2+ e K+ ) foram determinadas nas amostras de hemolinfa. A atividade da lactato desidrogenase (LDH) foi medida na hemolinfa, hepatopâncreas e músculo, enquanto a peroxidação lipídica (LPO) foi analisada no hepatopâncreas, músculo e brânquias (anteriores e posteriores). Os caranguejos controle não apresentaram diferença na concentração osmótica em função da salinidade, porém aqueles aclimatados à salinidade 2 apresentaram menores concentrações hemolinfáticas de Na+ , K + e Mg2+, bem como maiores níveis de LPO nas brânquias (anteriores e posteriores) e hepatopâncreas do que aqueles aclimatados à salinidade 30. O consumo de oxigênio corporal e a atividade tecidual da LDH foram semelhantes nos caranguejos controles aclimatados a 2 e 30. Estes resultados indicam que, após duas semanas de manutenção em laboratório, N. granulata apresenta ajustes fisiológicos da concentração osmótica (2‰: hiper-regulação; 30‰: hipo-regulação), composição iônica hemolinfática e taxas 4 metabólicas (aeróbica e anaeróbica) em função da salinidade, com conseqüente maior dano oxidativo em lipídios durante a hiper-regulação em baixa salinidade. Quanto à exposição ao Ni, houve aumento do consumo de oxigênio corporal, da atividade da LDH hemolinfática e da concentração hemolinfática de K+ na salinidade 2. Na salinidade 30 foi observado um aumento da atividade da LDH hemolinfática, da concentração osmótica e de Cl- hemolinfática, bem como uma diminuição das concentrações hemolinfáticas de K+ e Mg2+. Nos caranguejos aclimatados à salinidade 2, os efeitos do Ni parecem estar associados a distúrbios metabólicos (aeróbico e anaeróbico), enquanto distúrbios osmóticos e ionoregulatórios foram mais evidentes nos caranguejos aclimatados e expostos ao Ni na salinidade 30.

Os sistemas intensivos associados ao sistema de bioflocos (BFT), não requerem troca de água, permitem manutenção da qualidade da água do cultivo e proporciona a utilização de elevadas densidades de estocagem. Esta prática envolvendo a formação de floco microbiano e utilização de elevadas densidades de estocagem ainda é pouco explorada comercialmente no Brasil, igualmente ao uso de água de subsolo que é uma alternativa para realizar cultivos em regiões interiores. Os estudos existentes indicam que L. vannamei em meio heterotrófico apresenta sobrevivência elevada e produtividade pelo menos cinco vezes maior que nos sistemas tradicionais. O objetivo deste trabalho foi determinar as densidades de estocagem (100 e 150 camarões m-2 ) apropriadas para a região sul do Brasil na engorda do camarão Litopenaeus vannamei no sistema de bioflocos (BFT), avaliar os parâmetros de desempenho zootécnico e os parâmetros de qualidade da água. No presente estudo, a composição iônica da água do subsolo e os parâmetros de qualidade de água do cultivo estiveram dentro dos limites aceitáveis para o crescimento e sobrevivência dos camarões. A comunidade microbiana presente foi importante como suplemento na dieta dos animais cultivados, melhorando a taxa de conversão alimentar. As elevadas densidades de estocagem testadas apresentaram diferença estatística na produtividade: 9.900 e 13.700 kg ha-1 para o tratamento 100 e 150 camarões m-2 , respectivamente. Quando utilizado o sistema BFT, estas densidades de estocagem apresentam-se viáveis tanto no desempenho zootécnico dos camarões quanto nos parâmetros de qualidade da água. Entretanto a densidade mais elevada (150 camarões m-2 ) foi mais rentável devido sua maior produtividade.

We have prepared, characterized and investigated a new PEG-2000 based solid polymer electrolyte (PEG) x NH4I. Ionic conductivity measurements have been made as a function of salt concentration as well as temperature in the range 265–330 K. Selected compositions of the electrolyte were exposed to a beam of 8 MeV electrons to an accumulated dose of 10 kGy to study the effect on ionic conductivity. The electrolyte samples were also quenched at liquid nitrogen temperature and conductivity measurements were made. The ionic conductivity at room temperature exhibits a characteristic double peak for the composition x = 20 and 70. Both electron beam irradiation and quenching at low temperature have resulted in an increase in conductivity by 1–2 orders of magnitude. The enhancement of conductivity upon irradiation and quenching is interpreted as due to an increase in amorphous region and decrease in crystallinity of the electrolyte. DSC and proton NMR measurements also support this conclusion.

In the present study, a method based on transmission-line mode for a porous electrode was used to measure the ionic resistance of the anode catalyst layer under in situ fuel cell operation condition. The influence of Nafion content and catalyst loading in the anode catalyst layer on the methanol electro-oxidation and direct methanol fuel cell (DMFC) performance based on unsupported Pt-Ru black was investigated by using the AC impedance method. The optimal Nafion content was found to be 15 wt% at 75 degrees C. The optimal Pt-Ru loading is related to the operating temperature, for example, about 2.0 mg/cm(2) for 75-90 degrees C, 3.0 mg/cm2 for 50 degrees C. Over these values, the cell performance decreased due to the increases in ohmic and mass transfer resistances. It was found that the peak power density obtained was 217 mW/cm(2) with optimal catalyst and Nafion loading at 75 degrees C using oxygen. (c) 2005 International Association for Hydrogen Energy. The ultimate goal of periodontal tissue engineering is to produce predictable regeneration of alveolar bone, root cementum, and periodontal ligament, which are lost as a result of periodontal diseases. To achieve this goal, it is of great importance to develop novel bioactive materials which could stimulate the proliferation, differentiation and osteogenic/cementogenic gene expression of periodontal ligament cells (PDLCs) for periodontal regeneration. In this study, we synthesized novel Ca7Si2P2O16 ceramic powders for the first time by the sol–gel method and investigated the biological performance of PDLCs after exposure to different concentrations of Ca7Si2P2O16 extracts. The original extracts were prepared at 200 mg ml-1 and further diluted with serum-free cell culture medium to obtain a series of diluted extracts (100, 50, 25, 12.5 and 6.25 mg ml–1). Proliferation, alkaline phosphatase(ALP) activity, Ca deposition, and osteogenesis/cementogenesis-related gene expression (ALP, Col I, Runx2 and CEMP1) were assayed for PDLCs on days 7 and 14. The results showed that the ionic products from Ca7Si2P2O16 powders significantly stimulated the proliferation, ALP activity, Ca deposition and osteogenesis/cementogenesisrelated gene expression of PDLCs. In addition, it was found that Ca7Si2P2O16 powders had excellent apatite-mineralization ability in simulated body fluids. This study demonstrated that Ca7Si2P2O16 powders with such a specific composition possess the ability to stimulate the PDLC proliferation and osteoblast/cemenoblast-like cell differentiation, indicating that they are a promising bioactive material for periodontal tissue regeneration application.

Periodontitis results from the destructive inflammatory reaction of the host elicited by a bacterial biofilm adhering to the tooth surface and if left untreated, may lead to the loss of the teeth and the surrounding tissues, including the alveolar bone. Cementum is a specialized calcified tissue covering the tooth root and an essential part of the periodontium which enables the attachment of the periodontal ligament to the root and the surrounding alveolar bone. Periodontal ligament cells (PDLCs) represent a promising cell source for periodontal tissue engineering. Since cementogenesis is the critical event for the regeneration of periodontal tissues, this study examined whether inorganic stimuli derived from bioactive bredigite (Ca7MgSi4O16) bioceramics could stimulate the proliferation and cementogenic differentiation of PDLCs, and further investigated the involvement of the Wnt/β-catenin signalling pathway during this process via analysing gene/protein expression of PDLCs which interacted with bredigite extracts. Our results showed that the ionic products from bredigite powder extracts led to significantly enhanced proliferation and cementogenic differentiation, including mineralization–nodule formation, ALP activity and a series of bone/cementum-related gene/protein expression (ALP, OPN, OCN, BSP, CAP and CEMP1) of PDLCs in a concentration dependent manner. Furthermore, the addition of cardamonin, a Wnt/β-catenin signalling inhibitor, reduced the pro-cementogenesis effect of the bredigite extracts, indicating the involvement of the Wnt/β-catenin signalling pathway in the cementogenesis of PDLCs induced by bredigite extracts. The present study suggests that an entirely inorganic stimulus with a specific composition of bredigite bioceramics possesses the capacity to trigger the activation of the Wnt/β-catenin signalling pathway, leading to stimulated differentiation of PDLCs toward a cementogenic lineage. The results indicate the therapeutic potential of bredigite ceramics in periodontal tissue engineering application.

A generic approach towards tailoring of ion species composition in reactive plasmas used for nanofabrication of various functional nanofilms and nanoassemblies, based on a simplified model of a parallel-plate rf discharge, is proposed. The model includes an idealized reactive plasma containing two neutral and two ionic species interacting via charge exchange collisions in the presence of a microdispersed solid component. It is shown that the number densities of the desired ionic species can be efficiently managed by adjusting the dilution of the working gas in a buffer gas, rates of electron impact ionization, losses of plasma species on the discharge walls, and surfaces of fine particles, charge exchange rates, and efficiency of three-body recombination processes in the plasma bulk. The results are relevant to the plasma-aided nanomanufacturing of ordered patterns of carbon nanotip and nanopyramid microemitters.
