We recently showed that oxadiazoles have anti-Trypanosoma cruzi activity at micromolar concentrations. These compounds are easy to synthesize and show a number of clear and interpretable structure-activity relationships (SAR), features that make them attractive to pursue potency enhancement. We present here the structural design, synthesis, and anti-T. cruzi evaluation of new oxadiazoles denoted 5a-h and 6a-h. The design of these compounds was based on a previous model of computational docking of oxadiazoles on the T. cruzi protease cruzain. We tested the ability of these compounds to inhibit catalytic activity of cruzain, but we found no correlation between the enzyme inhibition and the antiparasitic activity of the compounds. However, we found reliable SAR data when we tested these compounds against the whole parasite. While none of these oxadiazoles showed toxicity for mammalian cells, oxadiazoles 6c (fluorine), 6d (chlorine), and 6e (bromine) reduced epimastigote proliferation and were cidal for trypomastigotes of T. cruzi Y strain. Oxadiazoles 6c and 6d have IC50 of 9.5 +/- 2.8 and 3.5 +/- 1.8 mu M for trypomastigotes, while Benznidazole, which is the currently used drug for Chagas disease treatment, showed an IC50 of 11.3 +/- 2.8 mu M. Compounds 6c and 6d impair trypomastigote development and invasion in macrophages, and also induce ultrastructural alterations in trypomastigotes. Finally, compound 6d given orally at 50 mg/kg substantially reduces the parasitemia in T. cruzi-infected BALB/c mice. Our drug design resulted in potency enhancement of oxadiazoles as anti-Chagas disease agents, and culminated with the identification of oxadiazole 6d, a trypanosomicidal compound in an animal model of infection. Als Phenanthro-Alkaloide wird eine kleine Gruppe von pentacyclischen, auf dem Phenanthren-Strukturmotiv basierenden Indolizidinen sowie Chinolizidinen bezeichnet. Von Letztgenannten sind bisher fünf, von den homologen Phenanthroindolizidinen mehr als sechzig natürliche Vertreter gefunden worden. Das wohl bekannteste Alkaloid in dieser Gruppe ist das Indolizidin-Alkaloid Tylophorin, das beispielsweise aus Tylophora indica (Apocynaceae, "Hundsgiftgewächse") gewonnen werden kann. Tylophorin und verwandte Derivate besitzen potente biologische und physiologische Wirkungen. So entfalten sie sowohl antiinflammatorische als auch antineoplastische Effekte (wirksam auch bei MDR-Tumorzelllinien, MDR = "multi drug resistant").rnrnDas Ziel der vorliegenden Arbeit war es, neue Methoden zur Herstellung von Phenanthro-Alkaloiden und deren Derivaten zu entwickeln. Ausgehend von (S)-Prolin konnten sowohl (S)-(+)-Tylophorin (>99% ee) als auch ein bisher noch nicht beschriebenes Derivat, das sich durch eine deutlich geringere Lichtempfindlichkeit im Vergleich zu Tylophorin aus¬zeichnet, in 33%-iger Gesamtausbeute über neun lineare Stufen, hergestellt werden. Die Synthese von (R)-(-)-Tylophorin gelang in analoger Weise aus (R)-Prolin in 21%-iger Ausbeute (93% ee). Der Einsatz von Schutzgruppen war nicht notwendig.rnDer Schlüsselschritt ist in beiden Fällen eine Cyclisierung an eine C=N-Doppelbindung über freie Radikale, die bei der Synthese des neuen Derivats hochstereoselektiv zur Bildung des (13aS,14S) Diastereomers führt. Die Synthese von 7-Methoxycryptopleurin gelang durch eine ähnliche Synthesestrategie. rnrnZur Herstellung von Cryptopleurin ist zunächst ein neuer Syntheseweg für 9-(Hydroxymethyl)-2,3,6-trimethoxyphenanthren entwickelt worden. Dieser führt über den Aufbau eines Biphenylsystems durch palladiumkatalysierte Kreuzkupplung, eine anschließende COREY-FUCHS-Transformation und als Schlüsselschritt über eine Gold-NHC-Komplex katalysierte 6-endo-dig-Cyclisierung zum entsprechenden Phenanthren. Diese Ringschlussreaktion verläuft in gewünschter Weise regioselektiv unter Bildung des 2,3,6-trimethoxysubstituierten Phenanthrens. Die Bil¬dung des regioisomeren 2,3,8-Trimethoxyphenanthrens wird nicht beobachtet. Der Alkohol wird dann in fünf linearen Stufen (34%) in das Xanthogenat überführt, aus dem sich durch eine zweistufige Reaktionssequenz, bestehend aus einer Radikal¬cyclisierung nach ZARD und einer Reduktion mit LiAlH4 das extrem lichtempfindliche und hochtoxische Alkaloid (R)-(-)-Cryptopleurin gewinnen ließ (50%).rnNachdem beide Enantiomere und das Racemat von Tylophorin synthetisiert worden waren und zum Vergleich bereit standen, wurde Tylophorin aus Tylophora indica extrahiert. rnDie Motivation rührte unter anderem daher, dass in der bisherigen Literatur Unstimmigkeiten über das in der Natur vorkommende Enantiomer des Tylophorins herrschten. Vor Beginn dieser Arbeit ging man davon aus, dass in T. indica nur (R)-(-)-Tylophorin vorkommt und für die Diskrepanzen zwischen den berichteten Drehwerten von, aus Pflanzenmaterial isolierten und des synthetisierten Naturstoffs, dessen Zersetzung vor oder während der Messung verantwortlich ist. Dieser Effekt kann zwar auch beobachtet werden, jedoch trägt er nur in geringem Maße zur Erniederigung des Drehwertes bei. Schließlich sind Proben von synthetisiertem Tylophorin in gleichem Maße von der schnell eintretenden Oxidation des Alkaloids betroffen. Aus dem Rohextrakt ist Tylophorin durch RP-HPLC isoliert worden. Anschließend wurde die Probe mittels chiraler HPLC/MS analysiert. Durch den Vergleich mit den bereit stehenden synthetischen Proben von (R)- und (S)-Tylophorin konnte in dieser Arbeit erstmals experimentell belegt werden, dass es sich bei (−)-Tylophorin aus T. indica um ein scalemisches Gemisch im Verhältnis von 56:44 (R:S) handelt.rnrnDas Ziel dieses Teilprojektes war die Entwicklung einer Synthese für den bisher noch nicht synthetisch hergestellten phytotoxischen Sekundärmetabolit (+)-Phenguignardiasäure. Isoliert wurde diese Verbindung aus Guignardia bidwellii, dem Erreger der Schwarzfäule der Weinrebe. Die absolute Konfiguration des quartären Stereozentrums war zu Beginn dieser Arbeit nicht bekannt. Ausgehend von (R)-Phenylmilchsäure und 3-Phenylprop-2-in-1-ol gelang die Synthese beider Enantiomere des Dioxolanons in acht linearen Stufen. Sie liefert den experimentellen Beweis (ECD, Polarimetrie) für die (S)-Konfiguration von natürlicher (+)-Phenguignardiasäure.rnrn

Hydrazine $\rm (N\sb2H\sb4),$ an important liquid propellant and derivative chemical for pharmaceuticals and pesticides, produces coma and convulsions sometimes resulting in death. Hyperammonia was found in rabbits exposed to 18 mg/Kg of hydrazine. Results of Part One of this study of rabbits emphasize the importance of acute ammonia toxicity during the first three hours following exposure to hydrazine. At no time during this post exposure period did a significant reduction of hydrazine to ammonia occur. Therefore, the elevated blood ammonia was apparently secondary to the effects of hydrazine on metabolic pathways. Further, the results support the theory of competitive inhibition of ammonia by hydrazine and emphasize the need to monitor plasma ammonia following toxic exposure to hydrazine.^ In Part Two, urea, ammonia, CO$\sb2,$ pH, glucose, sodium, potassium, chloride and creatinine were measured for up to 4 hours following injection of 18 mg/Kg of hydrazine in each of two groups of five rabbits. One group received normal saline and the other group received 5% dextrose and water/normal saline. Hyperammonemia, minimal metabolic acidosis and hyperglycemia without increased urea were found in the rabbits receiving normal saline intravenous infusion and hydrazine injection. Hence, hypoglycemia does not appear to play a role in the development of hyperammonemia. A significant difference in the elevated ammonia levels between the two groups receiving dextrose and water/normal saline and normal saline at 1 hour occurred. There was no significant difference in the elevated ammonia levels seen between the two groups receiving dextrose and water/normal saline and normal saline at 2.5 and 4 hours. Thus at 1 hour the group receiving dextrose was able to utilize excess glucose to detoxify ammonia, while at 2.5 and 4 hours there was no significant difference in the two groups' ability to detoxify ammonia.^ Findings support the theory that hydrazine inhibits the formation of urea resulting in hyperammonemia. Results suggest that hydrazine at 18 mg/Kg, a known hypoglycemic agent, causes serious hyperammonemia without increasing urea production during hyperglycemia. These experiments support a unified theory for the toxic mechanism of action of hydrazine, i.e., the intermediary metabolic effects of hydrazine are brought about by the formation of hydrazones which encumber ATP synthesis and vitamin B$\sb6$ enzymatic reactions. ^

Dynamic combinatorial libraries are mixtures of compounds that exist in a dynamic equilibrium and can be driven to compositional self adaptation via selective binding of a specific assembly of certain components to a molecular target. We present here an extension of this initial concept to dynamic libraries that consists of two levels, the first formed by the coordination of terpyridine-based ligands to the transition metal template, and the second, by the imine formation with the aldehyde substituents on the terpyridine moieties. Dialdehyde 7 has been synthesized, converted into a variety of ligands, oxime ethers L11–L33 and acyl hydrazones L44–L77, and subsequently into corresponding cobalt complexes. A typical complex, Co(L22)22+ is shown to engage in rapid exchange with a competing ligand L11 and with another complex, Co(L22)22+ in 30% acetonitrile/water at pH 7.0 and 25°C. The exchange in the corresponding Co(III) complexes is shown to be much slower. Imine exchange in the acyl hydrazone complexes (L44–L77) is strongly controlled by pH and temperature. The two types of exchange, ligand and imine, can thus be used as independent equilibrium processes controlled by different types of external intervention, i.e., via oxidation/reduction of the metal template and/or change in the pH/temperature of the medium. The resulting double-level dynamic libraries are therefore named orthogonal, in similarity with the orthogonal protecting groups in organic synthesis. Sample libraries of this type have been synthesized and showed the complete expected set of components in electrospray ionization MS.

A doença de Chagas é uma parasitose extremamente negligenciada, cujo agente etiológico é o protozoário Trypanosoma cruzi. Atualmente, 21 países da América Latina são considerados regiões endêmicas, onde 75-90 milhões de pessoas estão expostas à infecção, 6-7 milhões estão infectadas e mais de 41 mil novos casos surgem por ano. Entretanto, apenas os fármacos nifurtimox e benznidazol estão disponíveis no mercado. Estes, além da baixa eficácia na fase crônica da parasitose, apresentam diversos efeitos adversos, sendo que no Brasil apenas o benznidazol é utilizado. Este fato mostra a importância de se ampliar o número de fármacos disponíveis e propor quimioterapia mais eficaz para o tratamento da doença de Chagas. Como forma de contribuir para essa busca, este trabalho objetiva a síntese de compostos híbridos bioisostéricos N-acilidrazônicos e sulfonilidrazônicos, contendo grupo liberador de óxido nítrico, com potencial de interação com cisteíno-proteases parasitárias, tais como a cruzaína. Nestes derivados, os grupos liberadores de óxido nítrico utilizados foram os grupos furoxano (contendo substituinte metílico e fenílico) e éster nitrato. Propôs-se a variação de anéis aromáticos substituídos e não-substituídos, com o intuito de avaliar a possível relação estrutura-atividade (REA) desses análogos. Até o momento, somente os compostos da série N-acilidrazônica tiveram avaliação biológica realizada. Os valores de IC50 dos compostos na forma amastigota do parasita variaram entre >100 a 2,88 µM, sendo este último valor comparável ao fármaco de referência. A atividade inibitória frente à cruzaína foi de 25,2 µM a 2,2 µM. Já a liberação de óxido nítrico foi avaliada pelo método indireto de detecção de nitrato e os valores variaram entre 52,0 µM e 4.232,0 µM. Estes são bem inferiores ao composto padrão, além de não se identificar correlação direta entre a atividade biológica e a liberação de NO. Na sequência, os dois compostos mais ativos (6 e 14) foram submetidos a estudos de permeabilidade e de citotoxicidade. O composto 6 foi considerado o de maior permeabilidade segundo o Sistema de Classificação Biofarmacêutica (SCB) e todos os compostos apresentaram a taxa de fluxo menor que 2, indicando a ausência de mecanismo de efluxo. Na avaliação do potencial citotóxico desses compostos em células humanas, o derivado 6 apresentou índice de seletividade superior ao do benznidazol. Em estudos de modelagem molecular usando análise exploratória de dados (HCA e PCA), propriedades estéricas/geométricas e eletrônicas foram consideradas as mais relevantes para a atividade biológica. Além disso, estudos de docking mostraram que a posição do grupo nitro no anel aromático é importante para a interação com a cruzaína. Ademais o composto 6 não provocou mudanças significativas no ciclo celular e na fragmentação de DNA em células humanas, mostrando-se como líder promissor para futuros estudos in vivo. Atividade tripanomicida, citotoxicidade, potencial de liberação de NO e estudos de permeabilidade dos 23 derivados sulfonilidrazônicos e ésteres nitrato estão sendo avaliados.

Azomethine imines are considered 1,3-dipoles of the aza-allyl type which are transient intermediates and should be generated in situ but can also be stable and isolable compounds. They react with electron-rich and electron-poor olefins as well as with acetylenic compounds and allenoates mainly by a [3 + 2] cycloaddition but they can also take part in [3 + 3], [4 + 3], [3 + 2 + 2] and [5 + 3] with different dipolarophiles. These 1,3-dipolar cycloadditions (1,3-DC) can be performed not only under thermal or microwave conditions but also using metallo- and organocatalytic systems. In recent years enantiocatalyzed 1,3-dipolar cycloadditions have been extensively considered and applied to the synthesis of a great variety of dinitrogenated heterocycles with biological activity. Acyclic azomethine imines derived from mono and disubstituted hydrazones could be generated by prototropy under heating or by using Lewis or Brønsted acids to give, after [3 + 2] cycloadditions, pyrazolidines and pyrazolines. Cyclic azomethine imines, incorporating a C–N bond in a ring, such as isoquinolinium imides are the most widely used dipoles in normal and inverse-electron demand 1,3-DC allowing the synthesis of tetrahydro-, dihydro- and unsaturated pyrazolo[1,5-a]isoquinolines in racemic and enantioenriched forms with interesting biological activity. Pyridinium and quinolinium imides give the corresponding pyrazolopyridines and indazolo[3,2-a]isoquinolines, respectively. In the case of cyclic azomethine imines with an N–N bond incorporated into a ring, N-alkylidene-3-oxo-pyrazolidinium ylides are the most popular stable and isolated dipoles able to form dinitrogen-fused saturated and unsaturated pyrazolopyrazolones as racemic or enantiomerically enriched compounds present in many pharmaceuticals, agrochemicals and other useful chemicals.
