Placing a gene of interest under the control of an inducible promoter greatly aids the purification, localization and functional analysis of proteins but usually requires the sub-cloning of the gene of interest into an appropriate expression vector. Here, we describe an alternative approach employing in vitro transposition of Tn Omega P(BAD) to place the highly regulable, arabinose inducible P(BAD) promoter upstream of the gene to be expressed. The method is rapid, simple and facilitates the optimization of expression by producing constructs with variable distances between the P(BAD) promoter and the gene. To illustrate the use of this approach, we describe the construction of a strain of Escherichia coli in which growth at low temperatures on solid media is dependent on threshold levels of arabinose. Other uses of the transposable promoter are also discussed.

During the process of lysozyme protein crystallization with batch method, the macroscopic flow field of solid/liquid system was observed by particle image velocimetry (PIV). Furthermore, a normal growth rate of (110) face and local flow field around a single protein crystal were obtained by a long work distance microscope. The experimental results showed that the average velocity, the maximal velocity of macroscopic solid/liquid system and the velocity of local flow field around single protein crystal were fluctuant. The effective boundary layer thickness delta(eff), the concentration at the interface Q and the characteristic velocity V were calculated using a convection-diffusion model. The results showed that the growth of lysozyme crystal in this experiment was dominated by interfacial kinetics rather than bulk transport, and the function of buoyancy-driven flow in bulk transport was small, however, the effect of bulk transport in crystal growth had a tendency to increase with the increase of lysozyme concentration. The calculated results, also showed that the order of magnitude of shear force was about 10(-21) N, which was much less than the bond force between the lysozyme molecules. Therefore the shear force induced by buoyancy-driven flows cannot remove the protein molecules from the interface of crystal.

The growth behaviour of zero-mean-shear turbulent-mixed layer containing suspended solid particles has been studied experimentally and analysed theoretically in a two-layer fluid system. The potential model for estimating the turbulent entrainment rate of the mixed layer has also been suggested, including the results of the turbulent entrainment for pure two-layer fluid. The experimental results show that the entrainment behaviour of a mixed layer with the suspended particles is well described by the model. The relationship between the entrainment distance and the time, and the variation of the dimensionless entrainment rate E with the local Richardson number Ri1 for the suspended particles differ from that for the pure two-layer fluid by the factors-eta-1/5 and eta-1, respectively, where eta = 1 + sigma-0-DELTA-rho/DELTA-rho-0.

[EN] Neurodegeneration together with a reduction in neurogenesis are cardinal features of Alzheimer’s disease (AD) induced by a combination of toxic amyloid-β peptide (Aβ) and a loss of trophic factor support. Amelioration of these was assessed with diverse neurotrophins in experimental therapeutic approaches. The aim of this study was to investigate whether intranasal delivery of plasma rich in growth factors (PRGF-Endoret), an autologous pool of morphogens and proteins, could enhance hippocampal neurogenesis and reduce neurodegeneration in an amyloid precursor protein/presenilin-1 (APP/PS1) mouse model. Neurotrophic and neuroprotective actions were firstly evident in primary neuronal cultures, where cell proliferation and survival were augmented by Endoret treatment. Translation of these effects in vivo was assessed in wild type and APP/PS1 mice, where neurogenesis was evaluated using 5-bromodeoxyuridine (BdrU), doublecortin (DCX), and NeuN immunostaining 5 weeks after Endoret administration. The number of BrdU, DCX, and NeuN positive cell was increased after chronic treatment. The number of degenerating neurons, detected with fluoro Jade-B staining was reduced in Endoret-treated APP/PS1 mice at 5 week after intranasal administration. In conclusion, Endoret was able to activate neuronal progenitor cells, enhancing hippocampal neurogenesis, and to reduce Aβ-induced neurodegeneration in a mouse model of AD.

The aim of the investigation is to know the percentage of fish meal required to support the best growth of Heterotis niloticus in a semi intensive pond culture system. To achieve this, feed was formulated with equal percentages of blood meal, and corn meal and varying levels of fish meal. The experiment was in four treatments. Results showed that the mean weight gained was directly proportional to the quantity of fish meal made available to the fish fence 31.58g, 33.79g, 45.15g and 51.24g were recorded for treatments I, II, III and IV respectively. Result from this study when compared with previous works, shows that size of the water body to a greater extent affects the growth. The availability of fish meal in the feed made it more acceptable to the fish and hence a commensurate conversion in to flesh. The analysis of variance showed that there is significant difference in the growth performance in the treatments

Hybrids of Clariid catfishes; C. gariepinus (Netherlands), C. anguillaris, H. bidorsalis and their parental species were monitored for 8 weeks in 2 x 2 x 1m outdoor concrete tanks. The fry were fed NIFFR diet (40% crude protein) twice daily, 7 days of the week. Growth and survival records were taken weekly. The male HEB X female CLG hybrid showed an overall highest performance in growth rate while the lowest was recorded in male CLA X female CLG hybrid. The male HEB X female CLG hybrid grew at a faster rate than its reciprocal hybrid. In view or their growth rate, it is possible that the growth and survival rates or H. bidorsalis especially at the fry to fingerling stage could be improved through hybridization. The hybrid have potential as commercial food fish

Local, exotic and hybrid tilapia fingerlings were fed 45% crude protein diet containing 18% fish meal in a flow through system in triplicate and their growth and food utilization observed for 14 weeks. At the end of the study, the hybrid (Exotic Oreochromis niloticus male x Exotic Oreochromis aureus female) fingerlings had higher growth rate and food conversion ratio (FCR) than the other treatments. This was followed by Exotic Oreochromis niloticus fingerlings. The exotic Oreochromis niloticus fingerlings came next while the local Oreochromis niloticus fingerlings were the least in growth performance. The survival rate of the local O. niloticus was however higher than the other treatments

Pig dung was used as manure at 18 and 36 t/ha/year in carp polyculture (without supplementary feeding) for 270 days. It was observed that pig dung at both the levels did not degrade the physico-chemical properties (pH, dissolved oxygen and alkalinity) of water. The nutrient (phosphates and nitrates) level of water was higher in manured ponds than control ponds (no manuring, only supplementary feeding). Further, plankton levels (phyto- and zooplankton) were also significantly higher in manured ponds. The growth of Catla catla and Labeo rohita was significantly more in manured ponds than in control ponds. Growth of Cirrhinus cirrhosus and Cyprinus carpio was significantly more in ponds manured with pig dung at 18 t/ha/year than in control ponds and the growth of Ctenopharyngodon idellus was significantly more in control than in manured ponds.

Comparative production potential of red tilapia (a mutant hybrid of Oreochromis mossambicus) and Nile tilapia (Oreochromis niloticus) under low-input aquaculture was studied in six ponds of 360 m² each with an average water depth of 90 cm. Three ponds were stocked with fingerlings of O. niloticus (average weight 11.4±3.48 g) while three other ponds were stocked with red tilapia (average weight 10.72±2.5 g) at a density of 20,000 fingerlings/ha. Supplementary feed consisting of rice bran was given daily at 4-6% of standing biomass. Ponds were fertilized at fortnightly intervals with cattle manure 750 kg/ ha. After six months of rearing, gross fish productions of 3,218 and 3,017 kg/ha were obtained from O. niloticus and red tilapia ponds, respectively. Of this, table size fish (>80 g in size) production amounted to 2,366 and 2,823 kg/ha from O. niloticus and red tilapia culture, respectively. Analysis of cost and benefits showed higher benefit from red tilapia culture.

Samples were collected to study the age and growth of Labeo calbasu (Hamilton) from the river Ghaghra (Guptarghat centre, Faizabad). The scales of L. calbasu have been used for age and growth studies in present paper. Study of the marginal rings on the scales of L. calbasu indicates their annual nature. The fish attained growth in 1st 18.7 cm, 2nd 27.8 cm, 3rd 35.7 cm, 4th 41.8 cm, 5th 46.9 cm, 6th 54.9 cm and 7th 57.4 cm years of the life. The growth rate was observed 18.7, 9.1, 7.9, 6.7, 5.1, 8.0 and 2.5 cm for 1st to 7th age classes respectively. The age groups 1+ to 4+ constituted 91.17% of the total exploited population and 8.83% of remaining age groups (5+ to 7+). The maximum exploited population was observed in 2+ age group with 33.68%. Overall exploitation pattern was systematic and a good indicator for heavy recruitment.

We study graphene growth on hafnia (HfO2) nanoparticles by chemical vapour deposition using optical microscopy, high resolution transmission electron microscopy and Raman spectroscopy. We find that monoclinic HfO2 nanoparticles neither reduce to a metal nor form a carbide while nucleating nanometer domain-sized few layer graphene. Hence we regard this as an interesting non-metallic catalyst model system with the potential to explore graphene growth directly on a (high-k) dielectric. HfO2 nanoparticles coated with few layer graphene by atmospheric pressure CVD with methane and hydrogen at 950 °C. (© 2011 WILEY-VCH Verlag GmbH & Co. A nerve growth factor (NGF) was isolated from the venom of Chinese cobra (Naja naja ntr a) by ion exchange chromatography, gel filtration and fast protein liquid chromatography (FPLC). The N-terminal sequence of 22 amino acid residues was identical with other NGFs previously purified from the venom of the same genus. The NGF monomer molecular weight was estimated to be 13 500 by reducing SDS-PAGE and the isoelectric point was determined to be 7.2 by isoelectric focusing electrophoresis. NGF improved the epididymal sperm motility of male rats and increased the pregnancy rate and fetus number of mated female rats. The serum levels of luteinizing hormone (LH) and follicle stimulating hormone (FSH) of male rats administrated NGF + gossypol was lower than that of male rats administrated gossypol. Histological sections of testes and epididymides showed that NGF reduced the destructive effects of gossypol on rat testes. (C) 1999 Elsevier Science Inc. All rights reserved.
