We report a method for estimating the positions of charge transfer (CT) bands in Eu3+-doped complex crystals. The environmental factor ( he) influencing the CT energy is presented. he consists of four chemical bond parameters: the covalency, the bond volume polarization, the presented charge of the ligand in the chemical bond, and the coordination number of the central ion. These parameters are calculated with the dielectric theory of complex crystals. The relationship between the experimental CT energies and calculated environmental factors was established by an empirical formula. The calculated values are in good agreement with the experimental results. Such a relationship was confirmed by detailed analysis. In addition, our method is also useful to predict the charge-transfer position of any other rare earth ion.

The energy difference DeltaE between the spin-allowed and spin-forbidden states of Tb3+ in crystals is studied. The environmental factor he representing the character of the host is redefined by using the chemical band of complex crystals. The relationship between h(e) and DeltaE is found to be a linear relation. The results show that the energy difference between the spin-forbidden and spin-allowed states for Tb3+ ions in crystals can be predicted from the environmental factor.

By using the chemical bond theory of complex crystals, the chemical bond properties of REBa2Cu3O7 (RE = Eu, Y) were calculated. The calculated covalencies for Cu(1)-O and Cu(2)-O bond in REBa2Cu3O7 compounds are 0.41 and 0.28 respectively. Mossbauer isomer shifts of Fe-57 doped, and Sn-119 doped in REBa2Cu3O7-x were calculated by using the chemical environmental factor, h(e), defined by covalency and electronic polarizability. Four valence state tin ion and iron ion sites were identified in Fe-57 and Sn-119 doped REBa2Cu3O7-x superconductors.

By using the clinical bond theory of dielectric description, the chemical bond parameters of (Tl.Pb) - 1223 was calculated. The results show that the Sr-O, Tl-O, and Ca-O types of bond have higher ionic character and the Cu-O types of bond have more covalent, character. Mossbauer isomer shifts of Fe-57 and Sn-119 doped in (Tl, Pb) -1223 were calculated by using the chemical environmental factor, h, defined by covalency and electronic polarizability. Four valence state tin and three valence iron sites were identified ill Fe-57, and Sn-119 doped (Tl, Pb) -1223 superconductor. We conclude that all of' the Fe atoms substitute the Cu at square planar Cu (H site, whereas Sn prefers to Substitute the square pyramidal Cu (2) site.

用平板画线法从患病栉孔扇贝(Chlamys farreri)体内分离到了一种原核生物(简称QDP)。QDP可以在改进的液体培养基MEM(含2.2%NaCl,5%小牛血清)和脑心浸液(含2.2% NaCl)中生长;菌落在显微镜下(150×)为无色、透明的小点状;革兰氏染色阴性;菌体为圆形或近似圆形。QDP在发育过程中有两种状态,一种为未成熟阶段,直径小于100nm;另一种为成熟阶段,直径变化很大,最小约60nm,最大可达4µm以上。较小的个体有拟核、核糖体和新月状的空泡,未见细胞壁;较大的个体有细胞壁,胞内大部分被空泡充满,未见拟核和核糖体。栉孔扇贝组织超簿切片电镜观查证实QDP的存在。QDP的密度随着生长发育时间的不同而有所变化,繁殖高峰期密度较大。 建立了密度梯度离心结合滤膜过滤分离技术,优化人工培养条件。最适生长温度为23℃,最适生长pH值为7.4,最适生长盐度相当于细胞培养液所需的盐浓度(0.85%NaCl)。 提取的QDP核酸能被RNase A 降解,且没有检测到DNA。以PCR、RT-PCR扩增其16SrRNA基因序列片段,PCR反应没有扩增出扩增子,而RT-PCR则扩增出了16S rRNA基因序列片段,经测定其序列全长度为1430bp,经与GENEBANK中的16S rRNA片段比较分析,与6种不同科的微生物的同源率最高的为95%-95.47%。 采用温度梯度和病原浓度梯度回归感染实验方法,较为系统地研究了QDP的致病性。研究结果表明:QDP对栉孔扇贝有强烈的致病作用,高温(23℃以上)是其致病的必要条件,证实DQP是栉孔扇贝大规模死亡的病原体之一。

A cruise was undertaken from 3rd to 8th November 2004 in Changjiang (Yangtze) River Estuary and its adjacent waters to investigate the spatial biomass distribution and size composition of phytoplankton. Chlorophyll-a (Chl-a) concentration ranged 0.42-1.17 mu g L-1 and 0.41-10.43 mu g L-1 inside and outside the river mouth, with the mean value 0.73 mu g L-1 and 1.86 mu g L-1, respectively. Compared with the Chl-a concentration in summer of 2004, the mean value was much lower inside, and a little higher outside the river mouth. The maximal Chl-a was 10.43 mu g L-1 at station 18 (122.67 degrees E, 31.25 degrees N), and the region of high Chl-a concentration was observed in the central survey area between 122.5 degrees E and 123.0 degrees E. In the stations located east of 122.5 degrees E, Chl-a concentration was generally high in the upper layers above 5 m due to water stratification. In the survey area, the average Chl-a in sizes of > 20 mu m and < 20 mu m was 0.28 mu g L-1 and 1.40 mu g L-1, respectively. High Chl-a concentration of < 20 mu m size-fraction indicated that the nanophytoplankton and picophytoplankton contributed the most to the biomass of phytoplankton. Skeletonema costatum, Prorocentrum micans and Scrippsiella trochoidea were the dominant species in surface water. The spatial distribution of cell abundance of phytoplankton was patchy and did not agree well with that of Chl-a, as the cell abundance could not distinguish the differences in shape and size of phytoplankton cells. Nitrate and silicate behaved conservatively, but the former could probably be the limitation factor to algal biomass at offshore stations. The distribution of phosphate scattered considerably, and its relation to the phytoplankton biomass was complicated.

Grazing intensity may alter the soil respiration rate in grassland ecosystems. The objectives of our study were to (1) determine the influence of grazing intensity on temporal variations in soil respiration of an alpine meadow on the northeastern Tibetan Plateau; and (2) characterise, the temperature response of soil respiration under different grazing intensities. Diurnal and seasonal soil respiration rates were measured for two alpine meadow sites with different grazing intensities. The light grazing (LG) meadow site had a grazing intensity of 2.55 sheep ha(-1), while the grazing intensity of the heavy grazing (HG) meadow site, 5.35 sheep ha(-1), was approximately twice that of the LG site. Soil respiration measurements - showed that CO2 efflux was almost twice as great at the LG site as at the HG site during the growing season, but the diurnal and seasonal patterns of soil respiration rate were similar for the two sites. Both exhibited the highest annual soil respiration rate in mid-August and the lowest in January. Soil respiration rate was highly dependent on soil temperature. The Q(10) value for annual soil respiration was lower for the HG site (2.75) than for the LG site (3.22). Estimates of net ecosystem CO2 exchange from monthly measurements of biomass and soil respiration revealed that during the period from May 1998 to April 1999, the LG site released 2040 g CO2 m(-2) y(-1) to the atmosphere, which was about one third more than the 1530g CO2 m(-2) y(-1) released at the HG site. The results suggest that (1) grazing intensity alters not only soil respiration rate, but also the temperature dependence of soil CO2 efflux; and (2) soil temperature is the major environmental factor controlling the temporal variation of soil respiration rate in the alpine meadow ecosystem. Toppling is a major failure model in anti-dip layered rock slopes. Because of the limited by testing means and experimental apparatus, present research on the deformation mechanism and stability analysis are mainly focus on the 2-Dimensional deformation, and the research really based on 3-Dimension is still limited. Therefore, based on the present research station, the article rely on the important hydroelectric project of typical anti-dip layered rock slopes -- The left bank slope of Long-tan hydropower-station in Guang Xi, China, and focused on the influencing factors, deformation mechanism and stability analysis of anti-dip layered rock slopes, three problems as follows are researched in this paper. (1) Deformation influencing factor analysis on ant-dip layered rock slopes Three influencing factors are included: geological factor, engineering factor and environmental factor. It is concluded that the toppling deformation of anti-dip layered rock slopes are more sensitive to geological and engineering factors, but less sensitive to environmental factor. In addition, the sensitivity of various factors to the rock toppling deformation is also arranged sequentially as follows: construction, gravitation, rainfall (underground water) and rock structure intensity, etc. (2) 3D deformation study on the anti-dip layered toppling rock slopes Used 3D Distinct Element Method (3DEC) analyzed the 3D deformation characteristic of anti-dip layered rock slops. It can be seen that the toppling characteristics are obvious when the inter-angle between slope direction and layer striking direction is under 20o, when the inter-angle is over 20o and equal or less than 40o,the toppling deformation characteristics decrease sharply with increase of inter-angle, when the inter-angle is over 40o , the slope deformation is not controlled by joints but influenced by other failure mode. Therefore, in order to quantify the toppling characteristics, a differential value of displacement vector angle between layered rock slope and block rock slope is proposed as a key index to distinguish failure model for anti-dip layered rock slopes, and it was applied to study the toppling of the rock slopes at Guangxi Long-tan hydropower station, China. The results indicated that the index was effective and instructive for analyzing the anti-dip layered rock slopes. (3) Stability analysis methods Because of the imperfection of some present slope analysis methods, based on slope failure mode and those three influencing factors, “slope stability entropy” method is defined in this paper, which makes good use of the sensitivity of relational matrix to influencing factors on slope stability and the qualification characteristics for information entropy to the irregularity of slope deformation. By this method, not only the randomness of geologic body on the base of dynamic analysis of slope failure mode is fully concerned, but also it makes the analysis time-saving and simple. Finally, the research findings were used to the engineering example successfully, and rational conclusion has been obtained.

The composition and distribution of phytoplankton assemblages around the tip of the Antarctic Peninsula were studied during two summer cruises (February/March 2008 and 2009). Water samples were collected for HPLC/CHEMTAX pigment and microscopic analysis. A great spatial variability in chlorophyll a (Chl a) was observed in the study area: highest levels in the vicinity of the James Ross Island (exceeding 7 mg m−3 in 2009), intermediate values (0.5 to 2 mg m−3) in the Bransfield Strait, and low concentrations in the Weddell Sea and Drake Passage (below 0.5 mg m−3). Phytoplankton assemblages were generally dominated by diatoms, especially at coastal stations with high Chl a concentration, where diatom contribution was above 90% of total Chl a. Nanoflagellates, such as cryptophytes and/or Phaeocystis antarctica, replaced diatoms in open-ocean areas (e.g., Weddell Sea). Many species of peridinin-lacking autotrophic dinoflagellates (e.g., Gymnodinium spp.) were also important to total Chl a biomass at well-stratified stations of Bransfield Strait. Generally, water column structure was the most important environmental factor determining phytoplankton communities’ biomass and distribution. The HPLC pigment data also allowed the assessment of different physiological responses of phytoplankton to ambient light variation. The present study provides new insights about the dynamics of phytoplankton in an undersampled region of the Southern Ocean highly susceptible to global climate change.

Identifying adaptive genetic variation is a challenging task, in particular in non-model species for which genomic information is still limited or absent. Here, we studied distribution patterns of amplified fragment length polymorphisms (AFLPs) in response to environmental variation, in 13 alpine plant species consistently sampled across the entire European Alps. Multiple linear regressions were performed between AFLP allele frequencies per site as dependent variables and two categories of independent variables, namely Moran's eigenvector map MEM variables (to account for spatial and unaccounted environmental variation, and historical demographic processes) and environmental variables. These associations allowed the identification of 153 loci of ecological relevance. Univariate regressions between allele frequency and each environmental factor further showed that loci of ecological relevance were mainly correlated with MEM variables. We found that precipitation and temperature were the best environmental predictors, whereas topographic factors were rarely involved in environmental associations. Climatic factors, subject to rapid variation as a result of the current global warming, are known to strongly influence the fate of alpine plants. Our study shows, for the first time for a large number of species, that the same environmental variables are drivers of plant adaptation at the scale of a whole biome, here the European Alps.

Le diabète de type 2 (DT2) est caractérisé par une résistance des tissus périphériques à l’action de l’insuline et par une insuffisance de la sécrétion d’insuline par les cellules β du pancréas. Différents facteurs tels que le stress du réticulum endoplasmique (RE) et l’immunité innée affectent la fonction de la cellule β-pancréatique. Toutefois, leur implication dans la régulation de la transcription du gène de l’insuline demeure imprécise. Le but de cette thèse était d’identifier et de caractériser le rôle du stress du RE et de l’immunité innée dans la régulation de la transcription du gène de l’insuline. Les cellules β-pancréatiques ont un RE très développé, conséquence de leur fonction spécialisée de biosynthèse et de sécrétion d’insuline. Cette particularité les rend très susceptible au stress du RE qui se met en place lors de l’accumulation de protéines mal repliées dans la lumière du RE. Nous avons montré qu’ATF6 (de l’anglais, activating transcription factor 6), un facteur de transcription impliqué dans la réponse au stress du RE, lie directement la boîte A5 de la région promotrice du gène de l’insuline dans les îlots de Langerhans isolés de rat. Nous avons également montré que la surexpression de la forme active d’ATF6α, mais pas ATF6β, réprime l’activité du promoteur de l’insuline. Toutefois, la mutation ou l’absence de la boîte A5 ne préviennent pas l’inhibition de l’activité promotrice du gène de l’insuline par ATF6. Ces résultats montrent qu’ATF6 se lie directement au promoteur du gène de l’insuline, mais que cette liaison ne semble pas contribuer à son activité répressive. Il a été suggéré que le microbiome intestinal joue un rôle dans le développement du DT2. Les patients diabétiques présentent des concentrations plasmatiques élevées de lipopolysaccharides (LPS) qui affectent la fonction de la cellule β-pancréatique. Nous avons montré que l’exposition aux LPS entraîne une réduction de la transcription du gène de l’insuline dans les îlots de Langerhans de rats, de souris et humains. Cette répression du gène de l’insuline par les LPS est associée à une diminution des niveaux d’ARNms de gènes clés de la cellule β-pancréatique, soit PDX-1 (de l’anglais, pancreatic duodenal homeobox 1) et MafA (de l’anglais, mammalian homologue of avian MafA/L-Maf). En utilisant un modèle de souris déficientes pour le récepteur TLR4 (de l’anglais, Toll-like receptor), nous avons montré que les effets délétères des LPS sur l’expression du gène de l’insuline sollicitent le récepteur de TLR4. Nous avons également montré que l’inhibition de la voie NF-kB entraîne une restauration des niveaux messagers de l’insuline en réponse à une exposition aux LPS dans les îlots de Langerhans de rat. Ainsi, nos résultats montrent que les LPS inhibent le gène de l’insuline dans les cellules β-pancréatiques via un mécanisme moléculaire dépendant du récepteur TLR4 et de la voie NF-kB. Ces observations suggèrent ainsi un rôle pour le microbiome intestinal dans la fonction de la cellule β du pancréas. Collectivement, ces résultats nous permettent de mieux comprendre les mécanismes moléculaires impliqués dans la répression du gène de l'insuline en réponse aux divers changements survenant de façon précoce dans l’évolution du diabète de type 2 et d'identifier des cibles thérapeutiques potentielles qui permettraient de prévenir ou ralentir la détérioration de l'homéostasie glycémique au cours de cette maladie, qui affecte plus de deux millions de Canadiens.

Ce mémoire visait à comprendre la dynamique temporelle et les patrons floristiques actuels de deux tourbières du sud-ouest du Québec (Small et Large Tea Field) et à identifier les facteurs anthropiques, environnementaux et spatiaux sous-jacents. Pour répondre aux objectifs, des inventaires floristiques anciens (1985) ont d’abord été comparés à des inventaires récents (2012) puis les patrons actuels et les facteurs sous-jacents ont été identifiés à l’aide d’analyses multi-variables. Mes résultats montrent d’abord qu’un boisement important s’est produit au cours des 30 dernières années dans les tourbières à l’étude, probablement en lien avec le drainage des terres agricoles avoisinantes, diminuant la hauteur de la nappe phréatique. Simultanément, les sphaignes ont proliférées dans le centre des sites s’expliquant par une recolonisation des secteurs ayant brûlés en 1983. D’autre part, mes analyses ont montré que les patrons floristiques actuels étaient surtout liés aux variables environnementales (pH et conductivité de l’eau, épaisseur des dépôts), bien que la variance associée aux activités humaines était aussi significative, notamment dans la tourbière Large (18.6%). Les patrons floristiques ainsi que les variables environnementales et anthropiques explicatives étaient aussi fortement structurés dans l’espace, notamment selon un gradient bordure-centre. Enfin, la diversité béta actuelle était surtout liée à la présence d’espèces non-tourbicoles ou exotiques. Globalement, cette étude a montré que les perturbations humaines passées et actuelles avaient un impact important sur la dynamique et la distribution de la végétation des tourbières Small et Large Tea Field.

S'han estudiat els efectes dels factors ambientals sobre el perífiton dels sistemes lenític fluctuants del aiguamolls de l'Empordà. L'estudi s'ha realitzat als tres nivells d'integració: nivell d'ecosistema considerant el rol del perífiton envers els altres productors primaris; a nivell de comunitat, estudiant la composició específica de les diatomees i a nivell de població estudiant la plasticitat fenotípica d'una espècie de diatomea (Nitzschia frustulum). A nivell d'ecosistema s'observa que els factors que afavoreixen el predomini dels diferents tipus de productors primaris (perífiton, fitoplàncton i macròfits) són la renovació i el grau d'eutròfia de l'aigua. A nivell de comunitat els factors determinants en la composició i distribució de les espècies de diatomees són els gradients confinament-inundació així com la productivitat del sistema. En funció d'aquest factors s'han establert 5 associacions de diatomees. A nivell de població es demostra que tant la salinitat, com la relació N : P a l'aigua com el moviment de l'aigua afecten la morfologia i ultraestructura de la valva de N. frustulum. De forma interessant s'observa que la salinitat, considerada com a factor individual, afecta N. frustulum a nivell poblacional provocant-li modificacions en la morfologia de la valva, per en canvi, no afecta a nivell de comunitat, ja que totes les espècies de diatomees presents en ambients de salinitat fluctuant són eurihalines.
