Calcium (Ca2+) is a critical regulator of many aspects of the Plasmodium reproductive cycle. In particular, intra-erythrocyte Plasmodium parasites respond to circulating levels of the melatonin in a process mediated partly by intracellular Ca2+. Melatonin promotes the development and synchronicity of parasites, thereby enhancing their spread and worsening the clinical implications. The signalling mechanisms underlying the effects of melatonin are not fully established, although both Ca2+ and cyclic AMP (cAMP) have been implicated. Furthermore, it is not clear whether different strains of Plasmodium use the same, or divergent, signals to control their development. The aim of this study was to explore the signalling mechanisms engaged by melatonin in P. chabaudi, a virulent rodent parasite. Using parasites at the throphozoite stage acutely isolated from mice erythrocytes, we demonstrate that melatonin triggers cAMP production and protein kinase A (PKA) activation. Interestingly, the stimulation of cAMP/PKA signalling by melatonin was dependent on elevation of Ca2+ within the parasite, because buffering Ca2+ changes using the chelator BAPTA prevented cAMP production in response to melatonin. Incubation with melatonin evoked robust Ca2+ signals within the parasite, as did the application of a membrane-permeant analogue of cAMP. Our data suggest that P. chabaudi engages both Ca2+ and cAMP signalling systems when stimulated by melatonin. Furthermore, there is positive feedback between these messengers, because Ca2+ evokes cAMP elevation and vice versa. Melatonin more than doubled the observed extent of parasitemia, and the increase in cAMP concentration and PKA activation was essential for this effect. These data support the possibility to use melatonin antagonists or derivates in therapeutic approach.

Chitosan, a biopolymer obtained from chitin, and its derivates, such as chitosan hydrochloride, has been reported as wound healing accelerators and as possible bone substitutes for tissue engineering, and therefore these Substances could be relevant in dentistry and periodontology. The purpose of this investigation was to make a histological evaluation of chitosan and chitosan hydrochloride biomaterials (gels) used in the correction of critical size bone defects made in rat`s calvaria. Bone defects of 8 mm in diameter were surgically created in the calviria of 50 Holtzman (Rattus norvegicus) rats and filled with blood clot (control), low molecular weight chitosan, high molecular weight chitosan, low molecular weight chitosan hydrochloride, and high molecular weight chitosan hydrochloride, numbering 10 animals, divided into two experimental periods (15 and 60 days), for each biomaterial. The histological evaluation was made based on the morphology of the new-formed tissues in defect`s region, and the results indicated that there was no statistical difference between the groups when the new bone formation in the entire defect`s area were compared (p > 0.05) and, except in the control groups, assorted degrees of inflammation Could be Seen. In Conclusion, chitosan and chitosan hydrochloride biomaterials used in this study were not able to promote new bone formation in critical size defects made in rat`s calvaria. The increasing in the consumption of plant medicine by parts of the population generated a bigger need for studies. Drug substitutions, changes and adulterations at the production techniques are common places at plant-originated drugs trade, leading governmental departments of drug control round the world to adopt many analytical practices to medicinal plants. However, agronomic and technological issues cause characteristics and chemical composition variation at the drug, problem to be solved by the subject researchers. The present work aims to obtain a spray dried extract from a extractive solution obtained from Psidium guajava L. leaves based in book references that stress the intermediate dosage forms advantages. It also tries to validate useful methodologies for the quality control for both raw material and its derivates. Using eight sets of the spray dried extract (with Eudragit®, Aerosil ® e Avicel PH101 ® as drying adjuvants), the study proposes analytical methods using techniques commonly performed to plant medicines and its intermediate forms. As results, a viable spray-dried extract was obtained from a standartized extract solution. Among the studied adjuvants, the combination Aerosil ® with Eudragit ® showed the drying outcome, rheology, humidity and tannin content values that best fitted the demands of the Brazilian Pharmacopaea

Architecture and relevance of several strongly adhered biofilms over a polyester imide (PEI) surface

Um estudo microscópico foi considerado para analisar a eventual adesão de fungos sobre uma superfície de poliéster-imida presente em fios de cobre esmaltados. A microscopia eletrônica de varredura, permitiu observar nestes biofilmes aderidos, uma alta quantidade de pigmentos, hifas e um arsenal enzimático possivelmente atuando na superfície desta macromolécula. Devido a natureza altamente aromática deste material e traços de derivados fenólicos usados como solventes - que se fazem ainda presentes no polímero já reticulado, uma certa atividade anti-fúngica poderia ser esperada, todavia não foram observadas alterações no crescimento dos microrganismos, bem como no processo de adesão dos fungos. Adicionalmente a este fato, os fios esmaltados revelaram total descaracterização de suas propriedades isolantes. Os estudos visam compreender e avaliar o grande potencial demonstrado pelos fungos que poderia em caráter vindouro, explorado em processos de biodeterioração e biodegradação

Comparou-se a sensibilidade microbiana in vitro de isolados de Rhodococcus equi pelo teste padrão de difusão com discos, com o modificado, pela adição de 5% de dimetilsulfóxido-DMSO. Observou-se aumento da sensibilidade do R. equi no teste com DMSO, frente a aminoglicosídeos (canamicina, amicacina, estreptomicina) e ao cloranfenicol, enquanto para a eritromicina e derivados ß-lactâmicos (penicilina G, cefalosporinas, amoxicilina, oxacilina), constatou-se redução da sensibilidade do agente.

Este estudo discute a relação entre as condições e a organização do trabalho como elementos que contribuem para a ocorrência de acidentes do trabalho. Os dados foram coletados em uma indústria produtora de açúcar, álcool e derivados, situada no Estado de São Paulo. Para a coleta dos dados utilizamos a observação direta do trabalho e realizamos entrevistas semidirigidas individuais como 22 trabalhadores do setor de produção de açúcar. A produção de açúcar foi indicada pela Equipe de Segurança e Higiene no Trabalho como o setor em que havia a maior ocorrência de acidentes. Por destacar o papel que a relação homem-trabalho desempenha na saúde física e psíquica dos trabalhadores, utilizamos a Psicodinâmica do Trabalho (Dejours, 1994) como referencial teórico para a análise dos dados obtidos nas entrevistas. A análise das entrevistas envolveu três aspectos: condições e organização do trabalho e insatisfação. Os resultados revelaram que o ambiente estudado apresenta fatores físicos, químicos e biológicos desfavoráveis à saúde dos trabalhadores. Quanto à organização do trabalho, os dados revelaram que a divisão do trabalho bem como o conteúdo das tarefas determinavam sobrecarga aos trabalhadores. O relato sobre a insatisfação envolveu: ausência de perspectiva para progressão profissional, falta de treinamento técnico, dificuldade em manejar equipamentos e inadequação dos equipamentos de proteção. Destaca-se também no discurso dos trabalhadores a ineficiência das ações organizacionais para a eliminação ou a neutralização dos riscos de acidentes do trabalho e a predominância da teoria do Ato inseguro na apuração da causalidade dos acidentes do trabalho.

This work presents the application of a scalar finite element formulation for Ex (TE-like) modes in anisotropic planar and channel waveguides with diagonal permittivity tensor, diffused in both transversal directions. This extended formulation considers explicitly both the variations of the refractive index and their spatial derivates inside of each finite element. Dispersion curves for Ex modes in planar and channel waveguides are shown, and the results compared with solutions obtained by other formulations.
