To better understand the dynamics of bee populations in crops, we assessed the effect of landscape context and habitat type on bee communities in annual entomophilous crops in Europe. We quantified bee communities in five pairs of crop-country: buckwheat in Poland, cantaloupe in France, field beans in the UK, spring oilseed rape in Sweden, and strawberries in Germany. For each country, 7-10 study fields were sampled over a gradient of increasing proportion of semi-natural habitats in the surrounding landscape. The CORINE land cover classification was used to characterize the landscape over a 3 km radius around each study field and we used multivariate and regression analyses to quantify the impact of landscape features on bee abundance and diversity at the sub-generic taxonomic level. Neither overall wild bee abundance nor diversity, taken as the number of sub-genera. was significantly affected by the proportion of semi-natural habitat. Therefore, we used the most precise level of the CORINE classification to examine the possible links between specific landscape features and wild bee communities. Bee community composition fell into three distinct groups across Europe: group I included Poland, Germany, and Sweden, group 2 the UK, and group 3 France. Among all three groups, wild bee abundance and sub-generic diversity were affected by 17 landscape elements including some semi-natural habitats (e.g., transitional wood land-shrub), some urban habitats (e.g., sport and leisure facilities) and some crop habitats (e.g., non-irrigated arable land). Some bee taxa were positively affected by urban habitats only, others by semi-natural habitats only, and others by a combination of semi-natural, urban and crop habitats. Bee sub-genera favoured by urban and crop habitats were more resistant to landscape change than those favoured only by semi-natural habitats. In agroecosystems, the agricultural intensification defined as the loss of semi-natural habitats does not necessarily cause a decline in evenness at the local level, but can change community composition towards a bee fauna dominated by common taxa. The aim of this study was to evaluate the survivability of Bifidobacterium breve NCIMB 702257 in a three malt-based media supplemented with cysteine and yeast extract, and to determine the protective effect of these growth factors. A number of parameterised mathematical models were used to predict of kinetics of viability and total acidity during storage at different temperatures. Results demonstrated a good fit to the experimental mathematical model. The Arrhenius equations showed only reasonable fits and the polynomial plots contained a large area without data between 4 and 25 degrees C. In addition, it was shown that cysteine promotes growth and acid production by bifidobacteria, but does not extend survivability. On the other hand, increasing the yeast extract content of the fermentation media enhances the survivability of B. breve. To our knowledge, this is the first study to address the modelling of the survivability of probiotic bacteria in a cereal based fermentation media at different temperatures, introducing a more quantitative approach to the study of the shelf-life of a probiotic product. The Cannabis sativa herb contains over 100 phytocannabinoid (pCB) compounds and has been used for thousands of years for both recreational and medicinal purposes. In the past two decades, characterisation of the body's endogenous cannabinoid (CB) (endocannabinoid, eCB) system (ECS) has highlighted activation of central CB1 receptors by the major pCB, Δ9-tetrahydrocannabinol (Δ9-THC) as the primary mediator of the psychoactive, hyperphagic and some of the potentially therapeutic properties of ingested cannabis. Whilst Δ9-THC is the most prevalent and widely studied pCB, it is also the predominant psychotropic component of cannabis, a property that likely limits its widespread therapeutic use as an isolated agent. In this regard, research focus has recently widened to include other pCBs including cannabidiol (CBD), cannabigerol (CBG), Δ9tetrahydrocannabivarin (Δ9-THCV) and cannabidivarin (CBDV), some of which show potential as therapeutic agents in preclinical models of CNS disease. Moreover, it is becoming evident that these non-Δ9-THC pCBs act at a wide range of pharmacological targets, not solely limited to CB receptors. Disorders that could be targeted include epilepsy, neurodegenerative diseases, affective disorders and the central modulation of feeding behaviour. Here, we review pCB effects in preclinical models of CNS disease and, where available, clinical trial data that support therapeutic effects. Such developments may soon yield the first non-Δ9-THC pCB-based medicines.

Pharmacological levels of zinc oxide (ZnO) incorporated into the post-weaning piglet diet reduce the incidence of diarrhoea caused by enterotoxigenic Escherichia coli (ETEC) K88. The mechanism for this is not understood. Here, Intestinal Porcine Epithelial Cells (IPEC) J2 were used as an in vitro model of the porcine intestine. ZnO reduced IPEC J2 viability at concentrations >= 200 mu M, and ETEC adhesion to the host cell was unaffected by ZnO. Characterisation of the metabolism of IPEC J2 cells and ETEC established the effects of ZnO treatment on the metabolic profile of both. Although 100 mu M ZnO did not inhibit growth of either host or pathogen in fully supplemented media, metabolic profiles were significantly altered. Glucose and mannose were essential energy sources for IPEC J2 cells in the presence of ZnO, as the ability to utilise other sources was compromised. The increase in specificity of requirements to support respiration in ETEC was more pronounced, in particular the need for cysteine as a nitrogen source. These findings indicate that ZnO impacts on both host cell and pathogen metabolism and may provide insight into the mechanism for diarrhoea reduction. (C) 2010 Elsevier B.V. All rights reserved.

Prostaglandins (PG) are bioactive lipids derived from the metabolism of membrane polyunsaturated fatty acids (PUFA), and play important roles in a number of biological processes including cell division, immune responses and wound healing. Cyclooxygenase (COX) is the key enzyme in PG synthesis from arachidonic acid. The hypothesis of the present study was that expression of COX-2 in porcine intestine was dependent on the microbial load and the age of piglets. Piglets were obtained from sows raised either on outdoor free-range farms or on indoor commercial farms, and littermates were divided into three treatments: One group of piglets suckled the sow, a second group was put into an isolator and fed a milk formula, and a third group was put into the isolator fed milk formula and injected with broad spectrum antibiotics. Samples were collected from the 75% level of the small intestine at day 5, 28 and 56 of age. Tissue section from four piglets from each of these six treatment groups was analysed by immunofluorescence for COX-2 and type-IV collagen (basement membrane, defining lamina propria (LP)). Image analysis was used to determine the number of positive pixels expressing LP and epithelial COX-2. COX-2 expressing cells were observed in LP and epithelium in all porcine intestinal samples. When analysing images obtained on day 28, injection of antibiotics seemed to reduce the COX-2 expression in intestinal samples of piglets when compared to other treatments (P=0.053). No significant effect of farm, treatments or age of piglets was observed on COX-2 expressing data when analysing all data of images obtained at day 28 and 56. By double-labelling experiments, COX-2 was found not to be expressed on cell co-expressing CD45, CD16, CD163 or CD2, thus indicating that mucosal leukocytes, including dendritic cells, macrophages and NK cells did not express COX-2. Future research should investigate the role of COX-2 expression in the digestive tract in relation to pig health.

The protein encoded by the PPARGC1A gene is expressed at high levels in metabolically active tissues and is involved in the control of oxidative stress via reactive oxygen species detoxification. Several recent reports suggest that the PPARGC1A Gly482Ser (rs8192678) missense polymorphism may relate inversely with blood pressure. We used conventional meta-analysis methods to assess the association between Gly482Ser and systolic (SBP) or diastolic blood pressures (DBP) or hypertension in 13,949 individuals from 17 studies, of which 6,042 were previously unpublished observations. The studies comprised cohorts of white European, Asian, and American Indian adults, and adolescents from South America. Stratified analyses were conducted to control for population stratification. Pooled genotype frequencies were 0.47 (Gly482Gly), 0.42 (Gly482Ser), and 0.11 (Ser482Ser). We found no evidence of association between Gly482Ser and SBP [Gly482Gly: mean = 131.0 mmHg, 95% confidence interval (CI) = 130.5-131.5 mmHg; Gly482Ser mean = 133.1 mmHg, 95% CI = 132.6-133.6 mmHg; Ser482Ser: mean = 133.5 mmHg, 95% CI = 132.5-134.5 mmHg; P = 0.409] or DBP (Gly482Gly: mean = 80.3 mmHg, 95% CI = 80.0-80.6 mmHg; Gly482Ser mean = 81.5 mmHg, 95% CI = 81.2-81.8 mmHg; Ser482Ser: mean = 82.1 mmHg, 95% CI = 81.5-82.7 mmHg; P = 0.651). Contrary to previous reports, we did not observe significant effect modification by sex (SBP, P = 0.966; DBP, P = 0.715). We were also unable to confirm the previously reported association between the Ser482 allele and hypertension [odds ratio: 0.97, 95% CI = 0.87-1.08, P = 0.585]. These results were materially unchanged when analyses were focused on whites only. However, statistical evidence of gene-age interaction was apparent for DBP [Gly482Gly: 73.5 (72.8, 74.2), Gly482Ser: 77.0 (76.2, 77.8), Ser482Ser: 79.1 (77.4, 80.9), P = 4.20 x 10(-12)] and SBP [Gly482Gly: 121.4 (120.4, 122.5), Gly482Ser: 125.9 (124.6, 127.1), Ser482Ser: 129.2 (126.5, 131.9), P = 7.20 x 10(-12)] in individuals <50 yr (n = 2,511); these genetic effects were absent in those older than 50 yr (n = 5,088) (SBP, P = 0.41; DBP, P = 0.51). Our findings suggest that the PPARGC1A Ser482 allele may be associated with higher blood pressure, but this is only apparent in younger adults.

We know that from mid-childhood onwards most new words are learned implicitly via reading; however, most word learning studies have taught novel items explicitly. We examined incidental word learning during reading by focusing on the well-documented finding that words which are acquired early in life are processed more quickly than those acquired later. Novel words were embedded in meaningful sentences and were presented to adult readers early (day 1) or later (day 2) during a five-day exposure phase. At test adults read the novel words in semantically neutral sentences. Participants’ eye movements were monitored throughout exposure and test. Adults also completed a surprise memory test in which they had to match each novel word with its definition. Results showed a decrease in reading times for all novel words over exposure, and significantly longer total reading times at test for early than late novel words. Early-presented novel words were also remembered better in the offline test. Our results show that order of presentation influences processing time early in the course of acquiring a new word, consistent with partial and incremental growth in knowledge occurring as a function of an individual’s experience with each word.

The timing and nature of the penultimate deglaciation, also known as Termination II (T-II), is subject of controversial discussions due to the scarcity of precisely-dated palaeoclimate records. Here we present a new precisely-dated and highly-resolved multi-proxy stalagmite record covering T-II from the high alpine Schafsloch Cave in Switzerland, an area where climate is governed by the North Atlantic. The inception of stalagmite growth at 137.4 ± 1.4 kyr before present (BP) indicates the presence of drip water and cave air temperatures of above 0°C, and is related to a climate-induced change in the thermal state (from cold- to warm-based) of the glacier above the cave. The cessation of stalagmite growth between 133.1 ± 0.7 and 131.9 ± 0.6 kyr BP is most likely related to distinct drop in temperature associated with Heinrich stadial 11. The resumption of stalagmite growth at 131.9 ± 0.6 kyr BP is accompanied by an abrupt increase in temperature and precipitation as indicated by distinct shifts in the oxygen and carbon isotopic composition as well as in trace element concentrations. The mid-point of T-II is around 131.8 ± 0.6 kyr BP in the Schafsloch Cave record is significantly earlier compared to the age of 129.1 ± 0.1 kyr BP in the Sanbao Cave record from China. The different ages between both records can be best explained by the competing effects of insolation and glacial boundary forcing on seasonality and snow cover extent in Eurasia.

Carbon dioxide electroreduction on copper electrode was studied by surface enhanced Raman scattering (SERS) in K(2)SO(4) aqueous solutions with different pH values. CO(2) was bubbled into the solution at 0 V vs. Ag/AgCl, i.e., on an oxidized copper surface. In acidic solutions (pH around 2.5), at -0.2 V, bands indicative of the presence of ethylene on the electrode surface were detected. Although ethylene is knowledgably a product of CO(2) electroreduction on copper, it was not experimentally identified on the electrode`s surface at such a low cathodic potential in prior works. In solutions with pH around 2.5, CO bands were not observed, suggesting that hydrocarbons could be formed by a pathway that does not occur via adsorbed CO. In solutions with higher pHs, a complex spectral pattern, between 800 and 1700 cm(-1), was observed at approximately -0.4 V. The observed spectrum closely resembles those reported in the literature for adsorption of monocarboxylic acids with small chains. The spectral features indicate the presence of a structure containing a double C=C bond. a carboxyl group, and C-H bonds on the electrode`s surface. SERS spectra obtained in CO-saturated solution are also presented. However, in this case, no SERS bands were observed in the region between 800 and 1700 cm(-1) at low cathodic potentials. Foi desenvolvido um experimento, numa criação comercial localizada na região subtropical do Estado do Paraná, para identificar os efeitos que influenciam a taxa de prenhez de 117 vacas de corte Nelore x Hereford, com idades entre 2 e 20 anos e com diferentes frações gênicas de Nelore (25,0%; 37,5%; 50,0% e 100,0%), paridas no período de 11/08/03 a 23/12/03 e acasaladas, por monta natural, entre 10/12/03 a 12/03/04. Foram avaliados os efeitos do peso ao início do acasalamento (PI), ganho diário médio de peso durante o acasalamento (GDA), escore de condição corporal ao início do acasalamento (ECCI), data juliana de parto (DJ), altura de garupa (H), fração gênica de Nelore (FGN), ordem de parto (OP), sexo do terneiro (S), peso ao nascer (PN) e peso ao desmame ajustado (PAJ205) sobre a probabilidade de prenhez (Pi). Os dados foram submetidos à análise de regressão logística, por meio do procedimento Logistic do pacote computacional SAS, para identificar os efeitos de cada variável na Pi. A média da taxa de prenhez foi de 43,2%. A Pi foi explicada pelas variações de ECCI (P=0,25), PI (P=0,02), GDA (P=0,09) e DJ (P=0,07). A mudança na chance de prenhez para cada acréscimo de unidade na variável regressora foi estimada com base na estatística da razão entre chances (odds ratio) dada por OR = exp(k), considerando que chance é a razão entre a probabilidade do evento ocorrer e a probabilidade do evento não ocorrer A razão entre chances foi estimada em 1,341, 1,133, 1,197 e 0,886 para ECCI, PI, GDA e DJ, respectivamente. Não foram observados efeitos significativos (P>0,30) da FGN, OP, S, PN e PAJ205 sobre a Pi. A chance das vacas conceberem esteve associada ao escore de condição corporal no início do acasalamento, sendo de 34,1% para cada 0,5 unidade de ECCI e de 19,7% para cada 0,100 kg/d na variação de peso durante o acasalamento. Além disto, vacas com partos mais cedo dentro do ano e com maiores pesos no início do acasalamento também apresentaram maiores chances de conceber, 12,9% e 13,3%, respectivamente. Portanto, para aumentar as taxas de prenhez de vacas de corte são necessárias melhorias no escore de condição corporal no início do acasalamento e no ganho de peso durante a estação de monta, ambos decorrentes de uma adequada nutrição pré e pós-parto.

This study evaluated stress indicators of juvenile piau (Leporinus friderici) during and after a 4-hour transportation in order to establish an appropriate transportation protocol for this type of fish. Fish were transported in plastic bags (133.1 g/L) and sampled before loading, during 1, 2, 3 and 4 h and after transportation (2, 6, 12 and 24 h). Blood samples were analyzed for cortisol and glucose levels, hematocrit, hemoglobin level, number and mean corpuscular volume of erythrocytes. Water pH, dissolved oxygen, temperature and ammonia were monitored before, during and after transportation. No mortality was observed through the experiment. Ammonia levels increased throughout transportation, but the low pH values kept NH3 in safe levels for fish. Cortisol levels increased within 4 h of transportation, and returned to control condition 2 h after arrival. Plasma glucose increased within one hour of transportation, reaching peak value within 4 h and returning to initial condition 2 h after arrival. Erythrocyte number and hemoglobin levels showed the lowest levels 2 h after arrival, and mean corpuscular volume increased during transportation, decreasing at 12 and 24 h after arrival. Transporting piau is stressful, but fish recover the initial condition in short time, showing tolerance to the changes in the water quality parameters.

We re-analyse the non-standard interaction (NSI) solutions to the solar neutrino problem in the light of the latest solar as well as atmospheric neutrino data. The latter require oscillations (OSC), while the former do not. Within such a three-neutrino framework the solar and atmospheric neutrino sectors are connected not only by the neutrino mixing angle theta(13) constrained by reactor and atmospheric data, but also by the flavour-changing (FC) and non-universal (NU) parameters accounting for the solar data. Since the NSI solution is energy-independent the spectrum is undistorted, so that the global analysis observables are the solar neutrino rates in all experiments as well as the Super-Kamiokande day-night measurements. We find that the NSI description of solar data is slightly better than that of the OSC solution and that the allowed NSI regions are determined mainly by the rate analysis. By using a few simplified ansatzes for the NSI interactions we explicitly demonstrate that the NSI values indicated by the solar data analysis are fully acceptable also for the atmospheric data. Objective: This study was conducted to evaluate the relationship between fistulae of the lower lip and cleft lip and/or palate in patients with Van der Woude syndrome.Methods: the medical records of 11,000 patients with cleft lip and/or palate registered at the Cleft Lip-Palate Research and Rehabilitation Hospital, University of São Paulo, Bauru were reviewed. of these patients, 133 (1.2%) presented with Van der Woude syndrome.Results: of the 133 patients, 88 (66.2%) exhibited full clefts, 22 (16.5%) only cleft lip, and 23 (17.3%) only cleft palate. The lower-lip fistulae observed in these 133 patients were bilateral symmetric in 66 (49.7%), bilateral asymmetric in 42 (31.6%), microform in 19 (14.3%), median in 5 (3.8%), and unilateral in 1 (0.7%).Conclusion: This population sample appears to exhibit the previously published tendency for bilateral, unilateral, or mixed-type congenital fistulae to be associated with cleft lip with or without cleft palate, while so-called microforms or conic elevations are almost exclusively associated with cleft palate.
