Die vorliegende Arbeit liefert erstmals einen umfassenden Überblick über die molekulare Epidemiologie von Methicillin resistenten Staphylococcus aureus (MRSA) eines nordhessischen Krankenhauses inklusive seines Umfeldes und deren Entwicklung in einem Zeitraum von fünf Jahren. Von besonderer Bedeutung ist, dass die MRSA-Stämme hierfür nicht nur anhand ihrer SCCmec-Region (staphylococcal cassette chromosome) typisiert wurden, sondern eine weitergehende Charakterisierung auf Grund der Bestimmung des Vorkommens von Antibiotikaresistenz- und Toxingenen, sowie Plasmiden erfolgte. Dabei wurde ein neuer SCCmec-Typ entdeckt und charakterisiert und weitere noch unbekannte SCCmec-Elemente beschrieben. Bei der Charakterisierung der MRSA-Kollektive konnten bzgl. aller untersuchten Eigenschaften im Laufe der Zeit signifikante Veränderungen beobachtet werden. Am deutlichsten waren diese Unterschiede zwischen dem ältesten Kollektiv aus 1999 und allen nachfolgenden Kollektiven. Die Kollektive aus 2001, 2002, 2003 und 2004 zeigten untereinander größere Ähnlichkeiten, aber dennoch gleichzeitig eine tendenziell divergente Entwicklung einzelner Eigenschaften. Besonders auffallend war das dominante Auftreten von SCCmecIV mit 63-87% der Isolate eines Kollektivs ab 2001, gegenüber 16% in 1999. Weiterhin erfolgte eine markante Veränderung im Vorkommen einzelner Antibiotikaresistenzgene von 1999 bis 2004. So waren aacA-aphD und ermA bei MRSA aus 1999 mit 84% bzw. 90% deutlich häufiger als in allen Kollektiven der folgenden Jahre (aacA-aphD: max. 32%, ermA: max. 40%). Wohingegen ermC ein stets zunehmendes Vorkommen von 3% auf 67% über den Untersuchungszeitraum zeigte. Unkontinuierliches aber statistisch relevant vermehrtes Auftreten von tetM konnte bei Isolaten aus 1999 (40%) und 2004 (74%) nachgewiesen werden. Auch bei Toxingenen zeigten sich deutliche Unterschiede in der zeitlichen Verteilung. Ab 2001 zeigten alle Isolate wesentlich höhere Anteile an sec, seg und sei verglichen mit den MRSA aus 1999. So konnte sec im Kollektiv aus 1999 gar nicht nachgewiesen werden, in denen der Folgejahre mit 54-77%. Die Werte für seg und sei stiegen von 48% bzw. 41% in 1999 kontinuierlich auf über 90% in 2004. Die Häufigkeit von MRSA sowohl mit mehreren Resistenzgenen als auch die mit mehreren Toxingenen nahm im Laufe der Zeit zu und korrelierte mit dem Vorkommen von Plasmiden. Bezüglich seiner Korrelation mit den vorkommenden Plasmiden zeigte SCCmecIV im Erhebungszeitraum besonders deutlich eine Veränderung. So nahm über den Zeitraum der Beobachtung die Anzahl der Stämme die zusätzlich zu einem großen Plasmid ein weiteres kleines Plasmid besaßen signifikant zu. Auch beim Vergleich der SCCmec-Typen der MRSA-Isolate konnten Unterschiede bzgl. aller weiteren untersuchten Eigenschaften dargestellt werden. So zeigten z.B. alle SCCmecIIIA das sea-Gen, während dies bei allen anderen in der vorliegenden Arbeit untersuchten SCCmec-Typen nur vereinzelt vorkam. SCCmecII-Stämme wiesen sowohl die meisten Antibiotikaresistenz- als auch Toxingene auf. Es wurde ferner gezeigt, dass Stämme mit vielen Resistenzgenen auch eine hohe Anzahl Toxingene besaßen und dies im Zusammenhang mit einem erhöhten Plasmidgehalt stehen könnte. Aus den MRSA-Kollektiven isolierte Plasmide konnten aufgrund von Restriktionsanalysen als verwandt zu β-Laktamase-Plasmiden des Grundtyps pI524 und pI258 beschrieben werden. Der in vorliegender Arbeit gezeigte Zusammenhang zwischen der Anzahl von direct repeat units (dru) in der Hypervariablen Region (HVR) und dem SCCmec-Typ half den Unterschied zwischen SCCmecIV und SCCmecIVA, sowie die Sonderstellung des in vorliegender Arbeit erstmalig beschriebenen SCCmecIA/II darzustellen. Nicht alle Isolate konnten einem bekannten SCCmec-Typ zugeordnet werden, es handelt sich bei diesen Ausnahmen um weitere noch unbekannte und hier erstmalig beschriebene SCCmec-Typen. Aufgrund der vorliegenden Arbeit konnte ein neuer SCCmec-Typ definiert werden, namentlich der Typ SCCmecIA/II, der seit 1999 in der Region gehäuft vorkommt Die vorliegenden Untersuchungen zeigten somit, dass die Epidemiologie von MRSA der Region Nordhessen trotz bestehender Gemeinsamkeiten zur MRSA-Situation in ganz Deutschland auch Besonderheiten aufweist. Diese nun zu kennen kann einen Beitrag zur gezielten Verbesserung bisheriger Maßnahmen zur Ausbreitungskontrolle von MRSA in der nordhessischen Region leisten.

Virulence in Staphylococcus aureus is regulated via agr-dependent quorum sensing in which an autoinducing peptide (AIP) activates AgrC, a histidine protein kinase. AIPs are usually thiolactones containing seven to nine amino acid residues in which the thiol of the central cysteine is linked to the alpha-carboxyl of the C-terminal amino acid residue. The staphylococcal agr locus has diverged such that the AIPs of the four different S. aureus agr groups self-activate but cross-inhibit. Consequently, although the agr system is conserved among the staphylococci, it has undergone significant evolutionary divergence whereby to retain functionality, any changes in the AIP-encoding gene (agrD) that modifies AIP structure must be accompanied by corresponding changes in the AgrC receptor. Since AIP-1 and AIP-4 only differ by a single amino acid, we compared the transmembrane topology of AgrC1 and AgrC4 to identify amino acid residues involved in AIP recognition. As only two of the three predicted extracellular loops exhibited amino acid differences, site-specific mutagenesis was used to exchange the key AgrC1 and AgrC4 amino acid residues in each loop either singly or in combination. A novel lux-based agrP3 reporter gene fusion was constructed to evaluate the response of the mutated AgrC receptors. The data obtained revealed that while differential recognition of AIP-1 and AIP-4 depends primarily on three amino acid residues in loop 2, loop 1 is essential for receptor activation by the cognate AIP. Furthermore, a single mutation in the AgrC1 loop 2 resulted in conversion of (Ala5)AIP-1 from a potent antagonist to an activator, essentially resulting in the forced evolution of a new AIP group. Taken together, our data indicate that loop 2 constitutes the predicted hydrophobic pocket that binds the AIP thiolactone ring while the exocyclic amino acid tail interacts with loop 1 to facilitate receptor activation.

PURPOSE. Interleukin (IL)-17, which is responsible for the initial influx of leukocytes into the target tissue, was recently described as the main cytokine involved in autoimmune diseases. Vogt-Koyanagi-Harada (VKH) syndrome is a significant cause of noninfectious blindness in the world. Herein the authors aimed at unraveling the involvement of IL-17 in VKH and in experimental autoimmune uveitis, focusing on the signaling pathways involved in IL-17 synthesis. METHODS. Mice were immunized with 161-180 peptide and pertussis toxin. Draining lymph node cells, harvested 21 days after immunization, were cultured in the presence or absence of p38 alpha mitogen-activated protein kinase (MAPK) inhibitor (SB203580) and assayed for cytokine production and quantification of CD4(+)IL-17(+) cells. Mice received intraocular injections of SB203580, and disease severity was evaluated by histologic examination of the enucleated eyes at day 21. CD4(+) lymphocytes from MSK-1/2-deficient mice, human CD4(+) cells silenced with MSK1 siRNA, or peripheral blood mononuclear cells (PBMCs) from VKH patients were cultured in the presence or absence of p38 alpha MAPK inhibitor and then assayed for IL-17, IFN-gamma, and IL-4 production. RESULTS. The inhibition of p38 alpha MAPK fully blocked the synthesis of IL-17 by PBMCs from VKH patients and lymphocytes from EAU mice. The absence of the msk1/2 gene resulted in failure to produce IL-17 by murine and human lymphocytes. Interestingly, intraocular injections of SB203580 in EAU mice did not suppress development of the disease. CONCLUSIONS. These data show that p38 alpha MAPK-MSK1/2 is involved in the control of IL-17 synthesis by CD4(+) T cells and that inhibition of p38 alpha MAPK in vitro suppresses IL-17 synthesis but that inhibition of this kinase in vivo did not protect from EAU. A total of 72 strains of Staphylococcus aureus were examined for the production of staphylococcal enterotoxins (SE) A, B, C, D and toxic shock syndrome toxin (TSST-1). The strains were isolated from milk samples from cows with mastitis in dairy herds of São Paulo State, Brazil. Off 72 isolates, 38 (52.8%) produced SEA, 38 (52.8%) SEB, 32 (44.4%) SED, 28 (38.9%) SEC and 27 (37.5%) TSST-1. From the 72 strains, 66 (91.7%) produced, at least, one or more toxin, including TSST-1.

O Staphylococcus aureus é um dos principais agentes das mastites consideradas contagiosas, apresentando elevada incidência na maioria dos rebanhos leiteiros em vários países. Além de perdas econômicas é importante salientar o aspecto de saúde pública para cepas produtoras de enterotoxinas e da toxina do choque tóxico. A enterotoxina A, relacionada com maior ênfase nos casos de toxinfecções alimentares, pode ser veiculada pelo leite cru, pasteurizado e subprodutos lácteos. A síndrome do choque tóxico é determinada mais freqüentemente pela toxina do choque tóxico, porém as enterotoxinas do tipo B e C também podem ser implicadas. O objetivo deste estudo foi verificar a ocorrência de S.aureus produtores de enteroxinas e da toxina do choque tóxico em amostras de leite de animais com mastite subclínica, e correlacionar estes resultados com a contagem de células somáticas; utilizando a técnica de celofane over agar para detecção da TNAase, kit comercial para identificação das enterotxinas e contagem eletrônica de células somáticas. Avaliod]MORENO, B.[u-se 209 amostras de leite oriundas de vacas com mastite subclínica por S.aureus, e dentre estas, 209 (98,86%) produziram TNAse, nove amostras (4,39%) foram produtoras de enterotoxinas, sendo que uma (0,49%) dentre elas foi produtora de EED, três (1,46%) de EEC, e três (1,46%) de EEB. em uma amostra (0,49%), detectou-se concomitantemente EEA e EEB e em outra EEB e EEC. A toxina do choque tóxico não foi encontrada nas cepas avaliadas neste estudo, assim como não houve aumento estatisticamente significativo, na contagem de células somáticas, das amostras de cepas produtoras de enteroxinas.

The structural specificity of alpha-PMTX, a novel peptide toxin derived from wasp venom has been studied on the neuromuscular synapse in the walking leg of the lobster. alpha-PMTX is known to induce repetitive action potentials in the presynaptic axon due to sodium channel inactivation. We synthesized 29 analogs of alpha-PMTX by substituting one or two amino acids and compared threshold concentrations of these mutant toxins for inducing repetitive action potentials. In 13 amino acid residues of alpha-PMTX, Arg-1, Lys-3 and Lys-12 regulate the toxic activity because substitution of these basic amino acid residues with other amino acid residues greatly changed the potency. Determining the structure-activity relationships of PMTXs will help clarifying the molecular mechanism of sodium channel inactivation. (C) 2000 Elsevier B.V. Ireland Ltd. All rights reserved.

Pompilidotoxins (PMTXs), derived from the venom of solitary wasp has been known to facilitate synaptic transmission in the lobster neuromuscular junction, and a recent further study from rat trigeminal neurons revealed that the toxin slows Na+ channel inactivation without modifying activation process. Here we report that beta -PMTX modifies rat brain type II Na+ channel alpha -subunit (rBII) expressed in human embryonic kidney cells but fails to act on the rat heart alpha -subunit (rH1) at similar concentrations. We constructed a series of chimeric mutants of rBII and rH1 Na+ channels and compared modification of the steady-state Na+ currents by beta -PMTX. We found that a difference in a single amino acid between Glu-1616 in rBII and Gln-1615 in rH1 at the extracellular loop of D4S3-S4 is crucial for the action of beta -PMTX. PMTXs, which are small peptides with 13 amino acids, would be a potential tool for exploring a new functional moiety of Na+ channels.

To investigate the role of the N-terminal region in the lytic mechanism of the pore-forming toxin sticholysin II (St II), we studied the conformational and functional properties of peptides encompassing the first 30 residues of the protein. Peptides containing residues 1-30 (P1-30) and 11-30 (P11-30) were synthesized and their conformational properties were examined in aqueous solution as a function of peptide concentration, pH, ionic strength, and addition of the secondary structure-inducing solvent trifluoroethanol (TFE). CD spectra showed that increasing concentration, pH, and ionic strength led to aggregation of P1-30; as a consequence, the peptide acquired beta-sheet conformation. In contrast, P11-30 exhibited practically no conformational changes under the same conditions, remaining essentially structureless. Moreover, this peptide did not undergo aggregation. These differences clearly point to the modulating effect of the first 10 hydrophobic residues on the peptides aggregation and conformational properties. In TFE both the first ten hydrophobic peptides acquired alpha-helical conformation, albeit to a different extent, P11-30 displayed lower alpha-helical content. P1-30 presented a larger-fraction of residues in alpha-helical conformation in TFE than that found in St II's crystal structure for that portion of the protein. Since TFE mimics the membrane em,, such increase in helical content could also occur upon toxin binding to membranes and represent a step in the mechanism of pore formation. The peptides conformational properties correlated well with their functional behaviour. Thus, P1-30 exhibited much higher hemolytic activity than P11-30. In addition, P11-30 was able to block the toxin's hemolytic activity. The size of pores formed in red blood cells by P 1-30 was estimated by measuring the permeability PEGs of different molecular mass. The pore radius (0.95 +/- 0.01 nm) was very similar to that of the PEGs of different pore formed by the toxin. The results demonstrate that the synthetic peptide P1-30 is a good model of St 11 conformation and function and emphasize the contribution of the toxin's N-terminal region, and, in particular, the hydrophobic residues 1-10 to pore formation. A study of 215 women from different socioeconomic backgrounds in Botucatu, Brazil, was conducted to reveal possible clues why toxic shock syndrome (TSS) is seldom diagnosed in Brazil. Of the 215 women, 79 were colonized with Staphylococcus aureus either in the nasal passages and/or in the vaginal area, which is comparable to the colonization of individuals in the developed countries Thirteen of the women were colonized with S. aureus that produced toxic shock syndrome toxin-1 (TSST-1), the toxin responsible for the majority of cases of TSS. Eleven strains produced enterotoxin B, the only enterotoxin implicated in TSS, primarily in non-menstrual TSS. Enterotoxin A was produced by 15 strains and is commonly associated with the production of TSST-1, but has not been implicated in TSS. Seven strains produced enterotoxin D and one strain produced enterotoxin C, but these have not been implicated in TSS. Only 9 women used tampons which may be a major reason for the lack of menstrual TSS in Brazil, Only two of the 49 women whose sera were examined for the presence of antibodies to TSST-1 had no or very low antibody titers, the major protection against the development of TSS, both menstrual and non-menstrual TSS. This is a lower percentage than has been observed in the developed countries. Although another possibility for the lack of TSS in Brazil is the failure to recognize the disease, however, the results of this limited study indicate the importance of low usage of tampons and the high percentage of individuals with antibodies to TSST-1. The socioeconomic backgrounds of the participants were of little significance.

The detection of staphylococcal enterotoxins is decisive for the confirmation of an outbreak and for the determination of the enterotoxigenicity of strains. Since the recognition of their antigenicity, a large number of serological methods for the detection of enterotoxins in food and culture media have been proposed. Since immunological methods require detectable amounts of toxin, molecular biology techniques represent important tools in the microbiology laboratory. In the present study, polymerase chain reaction (PCR) was used to identify genes responsible for the production of enterotoxins and toxic shock syndrome toxin 1 (TSST-1) in S. aureus and coagulase-negative staphylococci (CNS) isolated from patients and the results were compared with those obtained by the reverse passive latex agglutination (RPLA) assay. PCR detection of toxin genes revealed a higher percentage of toxigenic S. aureus strains (46.7%) than the RPLA method (38.3%). Analysis of the toxigenic profile of CNS strains showed that 26.7% of the isolates produced some type of toxin, and one or more toxin-specific genes were detected in 40% of the isolates. These results suggests the need for further studies in order to better characterize the pathogenic potential of CNS and indicate that attention should be paid to the toxigenic capacity of this group of microorganisms.
