Cells from rat bone marrow exhibit the proliferation-differentiation sequence of osteoblasts, form mineralized extracellular matrix in vitro and release alkaline phosphatase into the medium. Membrane-bound alkaline phosphatase was obtained by method that is easy to reproduce, simpler and fast when compared with the method used to obtain the enzyme from rat osseous plate. The membrane-bound alkaline phosphatase from cultures of rat bone marrow cells has a MWr of about 120 kDa and specific PNPP activity of 1200 U/tng. The ecto-enzyme is anchored to the plasma membrane by the GPI anchor and can be released by PIPLC (selective treatment) or polidocanol (0.2 mg/mL protein and 1% (w/v) detergent). The apparent optimum pH for PNPP hydrolysis by the enzyme was pH 10. This fraction hydrolyzes ATP (240 U/mg), ADP (350 U/ mg), glucose 1-phosphate (1100 U/mg), glucose 6-phosphate (340 Wing), fructose 6-phosphate (460 U/mg), pyrophosphate (330 U/mg) and (3glycerophosphate (600 U/mg). Cooperative effects were observed for the hydrolysis of PPi and beta-glycerophosphate. PNPPase activity was inhibited by 0.1 mM vanadate (46%), 0.1 mM ZnCl2 (68%), 1 mM levamisole (66%), 1 mM arsenate (44%), 10 mM phosphate (21%) and 1 mM theophylline (72%). We report the biochemical characterization of membrane-bound alkaline phosphatase obtained from rat bone marrow cells cultures, using a method that is simple, rapid and easy to reproduce. Its properties are compared with those of rat osseous plate enzyme and revealed that the alkaline phosphatase obtained has some kinetics and structural behaviors with higher levels of enzymatic activity, facilitating the comprehension of the mineralization process and its function. Success of tooth replantation is limited because part of the replanted tooth is lost because of progressive root resorption. This study used histomorphometry and immunohistochemistry to evaluate the effect of low-level laser therapy (LLLT) on the healing process of rat teeth replanted after different extra-oral periods, simulating immediate and delayed replantation. Sixty Wistar rats (Rattus norvegicus albinus) had their maxillary right incisors extracted and randomly assigned to six groups (n = 10): C4, C30 and C45, in which the teeth were replanted 4 min (immediate), 30 min (delayed) and 45 min (delayed) after extraction, respectively, and L4, L30 and L45, in which the teeth were replanted after the same extra-alveolar times, but the root surfaces and the alveolar wounds were irradiated with a gallium-aluminum-arsenate (GaAlAs) diode laser before replantation. The animals were sacrificed after 60 days. The anatomic pieces containing the replanted teeth were obtained and processed for either histomorphometrical analysis under optical microscopy or immunohistochemical expression of receptor activator of nuclear factor Kappa-B (RANK), and its ligand (RANKL), osteoprotegerin (OPG) and tartrate-resistant acid phosphatase (TRAP) proteins. Areas of external replacement and inflammatory root resorption were observed in all groups, without statistically significant differences (P > 0.05). Ankylosis was more frequent in L30 than in C30 (P < 0.05). RANKL immunostaining predominated over RANK and OPG immunostaining in both groups with immediate tooth replantation (P < 0.05). For the 45-min extra-alveolar time, however, there was greater evidence of RANK immunostaining compared to RANKL for both control and laser-treated groups (P < 0.05). Positive TRAP immunostaining predominated in L4 and L30 (P < 0.05). In conclusion, under the tested conditions, the treatment of the root surface and the alveolar wound with LLLT did not improve the healing process after immediate and delayed tooth replantation in rats.

Arsenic is an environmental pollutant that induces congenital malformations in experimental models and can contribute to human birth defects. The environmental exposure to arsenic is relatively small when compared with the doses required to cause teratogenicity in mice and other laboratory animals. In order to study the action of zinc in the arsenic-induced teratogenicity, in the present work mice were either pretreated with zinc and later with arsenic or were treated simultaneously with zinc and arsenic in vivo and in vitro. Following administration of arsenate on gestation day 8, pregnant females were killed on the 17th day of gestation; maternal and fetal data were collected by laparotomy and used to calculate reproductive parameters. Fetuses were analyzed for the presence of external malformation and, after the appropriate processing, visceral and skeletal analyses were accomplished. Conceptuses were exposed in whole embryo culture to arsenicals on gestation day 8 (3-6 somite stage). After a 26 h culture period, morphological development was assessed. Neither pretreatment with zinc nor simultaneous administration of zinc prevented arsenic teratogenicity in these experimental models. (C) 2002 Wiley-Liss, Inc.

During the growth of Kluyveromyces marxianus var. marxianus ATCC 10022 on lactose, peaks of glucose, but not β-galactosidase activity, were detected in culture medium. Harvested and washed whole cells produced glucose and galactose from lactose, or ortho-nitro-phenol from the chromogenic substrate ortho-nitro-phenyl-β-D-galactopyranoside (ONPG), indicating that β-galactosidase is physically associated with cells. ONPG hydrolysis by whole cells presented a monophasic kinetics (Km 36.6 mM) in lactose exponential growth phase cells, but a biphasic kinetics (Km 0.2 and 36.6 mM) in stationary growth phase cells. Permeabilization with digitonin or disruption of cells from both growth phases led to monosite ONPG hydrolysis (Km 2.2 to 2.5 mM), indicating that β-galactosidase is not located in the periplasm. In addition, the energy inhibitors fluoride or arsenate, as well as the uncouplercarbonyl cyanide m-chlorophenylhydrazone (CCCP) prevented ONPG hydrolysis by whole cells. These findings indicate that energy coupled transmembrane transport is the rate-limiting step for intracellular ONPG cleavage. The taxonomic and physiologic implications of the exclusive intracellular location of β-galactosidase of K. marxianus var. marxianus ATCC 10022 are discussed. © 1996 Kluwer Academic Publishers.

Arsenic is an environmental pollutant that influences the male reproductive function, affecting the spermatogenesis process in the testis and causing alterations in the sperm. The objective of this work was evaluate the reproductive efficiency in adult male rats exposed to arsenic and the possible male-mediated structural malformations. The animal were treated with sodium arsenato (3,6 mg/Kg, i.p.), twice a weak, for 15, 30, 45, 60 or 75 days whereas control received only distilled water. Results have showed a reduced pregnancy rate associated to a decrease in the fertility potencial and to a increase in the preimplantation loss. There was a decrease in the fetal weights, as showed by its average. Malformations rate, postimplantation loss and resorptions were unaffect by treatment. These results suggest that chronic exposure to arsenic provokes reduction on fertility of male rats. Further studies are required to clarify the arsenic effects on male reproductive system and male-mediated effects on progeny.

A contaminação ambiental por arsênio vem aumentando nos últimos anos, seja através de fontes naturais ou antropogênicas, o que pode ocasionar uma maior exposição do homem a este composto tóxico. Vários estudos vêm analisando o potencial de espécies cianobacterianas como possíveis biorremediadoras na busca de soluções para diminuir os impactos causados por este metaloide. A cianobactéria filamentosa Phormidium sp. se destaca por apresentar mecanismos bem desenvolvidos de adaptação às diversas condições ambientais. O presente estudo objetivou analisar o perfil de resistência da Phormidium sp. em diferentes concentrações de arsenato de sódio. A cianobactéria foi inoculada em tubos contendo 4 mL de meio BG-11 líquido e diferentes concentrações de arsenato (5, 10, 30, 50, 100, 130, 150, 200 e 250 mM) e sem a presença do metaloide (controle) e cultivadas durante 20 dias a 25ºC, sem agitação e com fotoperíodo de 12/12 horas de luz/escuro. A toxicidade do arsenato para Phormidium sp. foi caracterizada pela inibição do crescimento, sendo este determinado pela concentração de clorofila a. Todas as condições foram realizadas em triplicatas. A determinação do arsênio total presente nas amostras foi obtida através da técnica de espectrometria de emissão óptica com plasma induzido, do Instituto Evandro Chagas. A resistência de Phormidium sp. ao arsenato foi observada em até 50 mM do composto (p>0,05). À partir de 100 mM de arsenato foi observada inibição do crescimento cianobacteriano (p<0,05). As análises da dosagem de arsênio total no meio de cultura mostram que, durante os primeiros dias de experimento, a concentração de arsênio no meio de cultura foi reduzido, seguido de um aumento gradativo na concentração deste metaloide. Provavelmente, esta cianobacteria pode acumular o arsênio e posteriormente excretar este metaloide para o meio extracelular. Os resultados obtidos indicam a capacidade que a cianobactéria Phormidium sp. possui de crescer em meio contendo altas concentrações de arsênio. No entanto, outras análises se tornam fundamental para elucidar as vias metabólicas envolvidas durante o processo de resistência a este metaloide.

O arsênio é um metalóide tóxico que se tornou um problema de saúde pública em todo o mundo. Como forma de reduzir a contaminação ambiental por este metalóide, a qual é proveniente de atividades antropogênicas e naturais, a utilização de microorganismos em processos de biorremediação se tem mostrado uma estratégia promissora. A cianobactéria filamentosa homocitada, Geitlerinema unigranulatum UFV-E01, pertencente à ordem Pseudanabaenales, foi isolada de um ambiente contaminado por arsênio, sugerindo uma habilidade em lidar com o efeito tóxico deste metalóide. Com vista nisso, o presente trabalho objetivou caracterizar a resistência ao arsenato de sódio e quantificar o arsênio total extracelular da cianobactéria G. unigranulatum UFV-E01. As análises de resistência ao arsenato de sódio revelaram que a cianobactéria foi capaz de crescer em até 50 mM por 20 dias. Além disso, a cianobactéria G. unigranulatum UFV-E01 acumulou arsenato de sódio por 10 dias, reduzindo em até 67% o arsênio extracelular. Pelos dados obtidos neste estudo, a cianobactéria G. unigranulatum UFV-E01 foi capaz de resistir a altas concentrações de arsenato de sódio, no entanto outras análises, como a caracterização das vias metabólicas envolvidas no processo de resistência, devem ser realizadas para considerar sua aplicação em ambientes impactados por arsênio.

Cyclic voltammograms and capacitance measurements are presented to characterize the mineral response at relatively moderate environmental conditions, pH 4.5 and T = 25degreesC. The experiments involve examining the rates of oxidation and the surface morphology of arsenopyrite, which is oxidized abiotically. The semiconducting properties of the mineral have been investigated in attempt to gain additional information of FeAsS dissolution behavior in acidic solutions at potentials close to the open circuit potential of the mineral. A mechanistic pathway for the anodic dissolution of arsenopyrite in open circuit conditions is also suggested. At high overpotentials, anodic reactions produce mainly sulfate and arsenate ions and may be described as hole limited. The reduction of orpiment-like compounds at potentials more negative than the open circuit potential is discussed.
