Facing growth in demand, dairy production in peri-urban areas of developing countries is changing rapidly. To characterise this development around Bamako (Mali), this study establishes a typology of dairy production systems with a special focus on animal genetic resources. The survey included 52 dairy cattle farms from six peri-urban sites. It was conducted in 2011 through two visits, in the dry and harvest seasons. The median cattle number per farm was 17 (range 5-118) and 42% of farmers owned cropland (8.3 +/- 7.3 ha, minimum 1 ha, maximum 25 ha). Feeding strategy was a crucial variable in farm characterisation, accounting for about 85% of total expenses. The use of artificial insemination and a regular veterinary follow-up were other important parameters. According to breeders’ answers, thirty genetic profiles were identified, from local purebreds to different levels of crossbreds. Purebred animals raised were Fulani Zebu (45.8%), Maure Zebu (9.2%), Holstein (3.0%), Azawak Zebu (1.3%), Mere Zebu (0.5%) and Kuri taurine (0.1%). Holstein crossbred represented 30.5% of the total number of animals (19.0% Fulani-Holstein, 11.2% Maure-Holstein and 0.3% Kuri-Holstein). Montbéliarde, Normande and Limousin crossbreds were also found (6.6%, 0.7% and 0.3%, respectively). A multivariate analysis helped disaggregate the diversity of management practices. The high diversity of situations shows the need for consideration of typological characteristics for an appropriate intervention. Although strongly anchored on local breeds, the peri-urban dairy systems included a diversity of exotic cattle, showing an uncoordinated quest of breeders for innovation. Without a public intervention, this dynamic will result in an irremediable erosion of indigenous animal genetic resources.

Das Mahafaly Plateau im südwestlichen Madagaskar ist gekennzeichnet durch raue klimatische Bedingungen, vor allem regelmäßige Dürren und Trockenperioden, geringe Infrastruktur, steigende Unsicherheit, hohe Analphabetenrate und regelmäßige Zerstörung der Ernte durch Heuschreckenplagen. Da 97% der Bevölkerung von der Landwirtschaft abhängen, ist eine Steigerung der Produktivität von Anbausystemen die Grundlage für eine Verbesserung der Lebensbedingungen und Ernährungssicherheit in der Mahafaly Region. Da wenig über die Produktivität von traditionellen extensiven und neu eingeführten Anbaumethoden in diesem Gebiet bekannt ist, waren die Zielsetzungen der vorliegenden Arbeit, die limitierenden Faktoren und vielversprechende alternative Anbaumethoden zu identifizieren und diese unter Feldbedingungen zu testen. Wir untersuchten die Auswirkungen von lokalem Viehmist und Holzkohle auf die Erträge von Maniok, der Hauptanbaufrucht der Region, sowie die Beiträge von weiteren Faktoren, die im Untersuchungsgebiet ertragslimitierend sind. Darüber hinaus wurde in der Küstenregion das Potenzial für bewässerten Gemüseanbau mit Mist und Holzkohle untersucht, um zu einer Diversifizierung von Einkommen und Ernährung beizutragen. Ein weiterer Schwerpunkt dieser Arbeit war die Schätzung von Taubildung und deren Beitrag in der Jahreswasserbilanz durch Testen eines neu entworfenen Taumessgerätes. Maniok wurde über drei Jahre und in drei Versuchsfeldern in zwei Dörfern auf dem Plateau angebaut, mit applizierten Zeburindermistraten von 5 und 10 t ha-1, Holzkohleraten von 0,5 und 2 t ha-1 und Maniokpflanzdichten von 4500 Pflanzen ha-1. Maniokknollenerträge auf Kontrollflächen erreichten 1 bis 1,8 t Trockenmasse (TM) ha-1. Mist führte zu einer Knollenertragssteigerung um 30 - 40% nach drei Jahren in einem kontinuierlich bewirtschafteten Feld mit geringer Bodenfruchtbarkeit, hatte aber keinen Effekt auf den anderen Versuchsfeldern. Holzkohle hatte keinen Einfluss auf Erträge über den gesamten Testzeitraum, während die Infektion mit Cassava-Mosaikvirus zu Ertragseinbußen um bis zu 30% führte. Pflanzenbestände wurden felder-und jahresübergreifend um 4-54% des vollen Bestandes reduziert, was vermutlich auf das Auftreten von Trockenperioden und geringe Vitalität von Pflanzmaterial zurückzuführen ist. Karotten (Daucus carota L. var. Nantaise) und Zwiebeln (Allium cepa L. var. Red Créole) wurden über zwei Trockenzeiten mit lokal erhältlichem Saatgut angebaut. Wir testeten die Auswirkungen von lokalem Rindermist mit einer Rate von 40 t ha-1, Holzkohle mit einer Rate von 10 t ha-1, sowie Beschattung auf die Gemüseernteerträge. Lokale Bewässerungswasser hatte einen Salzgehalt von 7,65 mS cm-1. Karotten- und Zwiebelerträge über Behandlungen und Jahre erreichten 0,24 bis 2,56 t TM ha-1 beziehungsweise 0,30 bis 4,07 DM t ha-1. Mist und Holzkohle hatten keinen Einfluss auf die Erträge beider Kulturen. Beschattung verringerte Karottenerträge um 33% im ersten Jahr, während sich die Erträge im zweiten Jahr um 65% erhöhten. Zwiebelerträge wurden unter Beschattung um 148% und 208% im ersten und zweiten Jahr erhöht. Salines Bewässerungswasser sowie Qualität des lokal verfügbaren Saatgutes reduzierten die Keimungsraten deutlich. Taubildung im Küstendorf Efoetsy betrug 58,4 mm und repräsentierte damit 19% der Niederschlagsmenge innerhalb des gesamten Beobachtungszeitraum von 18 Monaten. Dies weist darauf hin, dass Tau in der Tat einen wichtigen Beitrag zur jährlichen Wasserbilanz darstellt. Tageshöchstwerte erreichten 0,48 mm. Die getestete Tauwaage-Vorrichtung war in der Lage, die nächtliche Taubildung auf der metallischen Kondensationsplatte zuverlässig zu bestimmen. Im abschließenden Kapitel werden die limitierenden Faktoren für eine nachhaltige Intensivierung der Landwirtschaft in der Untersuchungsregion diskutiert.
Correlating Bayesian date estimates with climatic events and domestication using a bovine case study

The tribe Bovini contains a number of commercially and culturally important species, such as cattle. Understanding their evolutionary time scale is important for distinguishing between post-glacial and domestication-associated population expansions, but estimates of bovine divergence times have been hindered by a lack of reliable calibration points. We present a Bayesian phylogenetic analysis of 481 mitochondrial D-loop sequences, including 228 radiocarbon-dated ancient DNA sequences, using a multi-demographic coalescent model. By employing the radiocarbon dates as internal calibrations, we co-estimate the bovine phylogeny and divergence times in a relaxed-clock framework. The analysis yields evidence for significant population expansions in both taurine and zebu cattle, European aurochs and yak clades. The divergence age estimates support domestication-associated expansion times (less than 12 kyr) for the major haplogroups of cattle. We compare the molecular and palaeontological estimates for the Bison-Bos divergence.

An experiment on restricted suckling of crossbred dairy cows was conducted at the Livestock Research Centre, Tanga in northeast Tanzania. Thirty-six Bos taurus (Holstein Friesian and Jersey) x Bos indicus (East African Zebu) cows were allocated alternately as they calved to suckling their calves for either 12 or 24 weeks after calving. Cows grazed improved pastures and were offered 4 kg concentrate daily. Milking occurred twice daily by hand; calves were allowed to suck residual milk for 30 min following each milking. Calves were also allowed access to grazing and were offered a maximum of I kg concentrate daily to 24 weeks of age. Weaning age had no significant effect on lactation milk yield for human consumption, the mean (SE) yield being 1806 (102.0) L and 1705 (129. 1) L for 12- and 24-week weaning, respectively. Cows from the two treatments suffered similar losses of live weight and body condition score during lactation and neither group had returned to the original body condition score 40 weeks following calving. Post-partum anoestrous intervals were prolonged. Although not significant, cows suckling calves to 24 weeks had a mean interval to first oestrus extended by 38 days compared with cows suckling calves to 12 weeks. The mean (SE) daily live weight gains of the calves to 52 weeks were 263 (14.1) g/day and 230 (18.1) g/day for calves weaned at 12 and 24 weeks, respectively, such that 12-month weights were 119 (5.6) kg and 110 (7.3) kg, respectively. Twelve-week-weaned calves consumed more concentrate (p < 0.05) from 13 to 24 weeks than did 24-week weaned calves. Calculation of residual milk consumption removed by calves from birth to 12 weeks indicated that it accounted for 28% of total yield. No benefits in cow and calf performance and welfare were found to justify prolonging the suckling period to 24 weeks.

The ability of PCR to detect infections of Theileria parva, the cause of East Coast Fever, in field-collected tick and bovine samples from Tanzania was evaluated. PCR-detected infection prevalence was high (15/20, 75%) in unfed adult Rhipicephalus appendiculatus ticks that fed as nymphs on an acutely-infected calf, but low (22/836, 2.6%) in unfed adult R. appendiculatus collected from field sites in Tanzania. Tick infection prevalence was comparable to that in previous studies that used salivary gland staining to detect T parva infection in field-collected host-seeking ticks. Of 282 naturally-exposed zebu calves, seven had PCR-positive buffy coat samples prior to detection of Theileria spp. parasites in stained huffy coat cells or lymph node biopsies. Evidence of Theileria spp. infections was detected in stained smears of lymph node biopsies from 109 calves (38.6%) and huffy coat samples from 81 (28.7%), while huffy coat samples from 66 (23.4%) were PCR-positive for T parva. Implications of these findings for the sensitivity and specificity of the PCR are discussed. (C) 2003 Elsevier Science B.V. All rights reserved.

A retrospective cross-sectional study was conducted on 200 randomly selected smallholder farms from a mixed dairy farming system in Tanga, Tanzania, between January and April 1999. We estimated the frequency and determinants of long calving interval (LCI), retention of fetal membrane (RFM), dystocia, and abortion in smallholder crossbred cattle and explored birth trends. The mean calving interval was 500 days and birth rate was 65 per 100 cow-years. Dystocia was reported to affect 58% of calvings, and 17.2% of animals suffered RFM. Using mixed effect models, the variables associated with LCI, RFM and dystocia were breed, level of exotic blood and condition score. Zebu breeding was associated with LCI (odds ratio (OR) = 2.3, p = 0.041) and Friesian breeding with lower odds for RF (OR = 0.26, p = 0.020). Animals with higher levels of exotic blood had lower odds for evidence of dystocia (OR = 0.45, p = 0.021). Evidence of dystocia was significantly associated with poor condition score (beta = -1.10, p = 0.001). Our observations suggest that LCIs are common in smallholder dairy farms in this region and a likely source of economic loss. Dystocia, RFM, poor condition score and mineral deficiency were common problems and were possibly linked to LCI.

Thiocyanate content and lactoperoxidase activity of individual cow's milk of different breeds were determined, and the effects of different lactoperoxidase system (LP-s) activation strategies were compared. Lactoperoxidase activity varied significantly between Friesian and both Ayrshire and Tanzania Short Horn Zebu (TSHZ), but differences between Ayrshire and TSHZ were not significant. There was no significant variation in SCN- content between breeds. The LP-s was activated using three strategies based on SCN-: namely; equal concentrations of SCN- and H2O2 (7:7, 10:10, 15 :15 mg/l), excess SCN- concentrations (15:10, 20:10, 25:10 mg SCN-: H2O2/I), and excess H2O2 concentrations (10:15, 10:20, 10:25 mg SCN-: H2O2/I), plus a fourth strategy based on I- (15 : 15 mg I- : H2O2/I). The keeping quality (KQ) was assessed using pH, titratable acidity, clot on boiling and alcohol stability tests. All activation strategies enhanced the shelf life of milk (typically increasing KQ from around 10 to around 20 h), but it was clear that the effectiveness of the LP-s depends on the type and concentrations of the activators of the system. The LP-s activated using I- as an electron donor was more effective than the LP-s activated using SCN- as an electron donor, increasing the KQ by a further 6-8 h compared with SCN-.

Influences of inbreeding on daily milk yield (DMY), age at first calving (AFC), and calving intervals (CI) were determined on a highly inbred zebu dairy subpopulation of the Guzerat breed. Variance components were estimated using animal models in single-trait analyses. Two approaches were employed to estimate inbreeding depression: using individual increase in inbreeding coefficients or using inbreeding coefficients as possible covariates included in the statistical models. The pedigree file included 9,915 animals, of which 9,055 were inbred, with an average inbreeding coefficient of 15.2%. The maximum inbreeding coefficient observed was 49.45%, and the average inbreeding for the females still in the herd during the analysis was 26.42%. Heritability estimates were 0.27 for DMY and 0.38 for AFC. The genetic variance ratio estimated with the random regression model for CI ranged around 0.10. Increased inbreeding caused poorer performance in DMY, AFC, and CI. However, some of the cows with the highest milk yield were among the highly inbred animals in this subpopulation. Individual increase in inbreeding used as a covariate in the statistical models accounted for inbreeding depression while avoiding overestimation that may result when fitting inbreeding coefficients.

This study investigated the effect of human-animal interaction (HAI) and the stress response on the quality of embryo production in superovulated Nelore (Bos indicus) cattle, under tropical conditions. Thirty-two females underwent a superovulation protocol for 5 days. Cortisol concentrations were determined in blood plasma collected on days 0, 4, and 5. Artificial insemination was performed on days 4 and 5, and nonsurgical embryo flushing on day 11. Embryo production and viability were determined. Human stimulation, animal behaviors, accidents, and handling time were recorded to assess HAI. Cattle age was negatively correlated with accidents, frequency of aversive behaviors, and negative stimuli by stockperson during transit through corral compartments to receive superovulation treatments. The factor analysis revealed two distinct groups. The first group was called stressed and had higher cortisol concentration than the nonstressed group, 16.0 +/- 2.1 and 12.5 +/- 1.0 ng/mL, respectively. Comparisons between these groups showed that the frequency of voice emissions by the stockperson and the number of accidents were higher in the stressed group, and also, the mean handling time was longer in the stressed group than for the nonstressed. As a result, viability rate of the embryos was 19% lower in the stressed group (P < 0.05). This indicates that intensive negative HAI is likely related to stress, which affects embryo production in a superovulation program.

O gado zebu (Bos indicus) é predominante em todo o mundo. Atualmente, no Brasil, o rebanho de bovinos e zebuínos é composto por aproximadamente 170 milhões de indivíduos, no qual, 80% são zebuínos ou animais de raças cruzadas com zebu. O principal objetivo deste estudo foi determinar a possibilidade da utilização de marcadores moleculares de bovinos na identificação genética de zebuínos. A confirmação desta possibilidade seria de extrema valia, já que a quantidade de informações sobre o genoma zebuíno é limitada e metodologias específicas de identificação de marcadores genéticos para raças zebuínas são raras. Assim, o DNA de 65 zebus, de 4 raças diferentes (Gir, Tabapuã, Nelore e Brahman), foi extraído a partir de sangue aplicado em papel filtro, utilizando o “kit” DNA IQTM SYSTEM (Promega®). Após a extração de DNA, foi feito um PCR Multiplex utilizando o “kit” StockMarks® (Applied Biosystems®), o qual amplifica um total de 11 loci (BM1824, BM2113, ETH10, ETH225, ETH3, INRA023, SPS115, TGLA122, TGLA126, TGLA227 e TGLA53). Os fragmentos gerados pela PCR foram analisados por eletroforese capilar utilizando o analisador genético ABI Prism 3100 (Applied Biosystems®) e o programa GeneScan® v.3.7 (Applied Biosystems®). A freqüência dos marcadores e os índices de variabilidade genética foram calculados utilizando o programa Microsatellite Toolkit v.3.1 e o equilíbrio de Hardy-Weinberg foi determinado através do programa GENEPOP v.3.4. Através destas técnicas foi possível demonstrar que os marcadores moleculares utilizados para identificação genética de bovinos podem também ser utilizados para a realização da técnica de identificação genética de zebuínos Além disso, foi possível demonstrar a freqüência alélica na população de zebuínos analisada como um todo, assim como foi determinada a freqüência alélica para os diferentes marcadores moleculares dentro das 4 raças estudas. Considerando o total de 100 diferentes alelos identificados entre os 11 STR (Short Tandem Repeats) analisados, foi possível observar que a heterozigosidade esperada (He) foi relativamente alta na população analisada, assim como na análise por raça. Estes dados indicam alto grau de diversidade genética nas diferentes raças. Neste sentido, a raça Brahman apresentou a menor diversidade (0,60) e a raça Tabapuã, a maior (0,69). Contudo, estudos complementares são necessários, a fim de confirmar as freqüências alélicas estabelecidas, uma vez que os resultados encontrados no presente trabalho referem-se a um número relativamente pequeno de zebuínos, quando comparados com o total de zebus encontrados no rebanho brasileiro.

Este experimento foi realizado para avaliar o efeito de dois níveis de casca de soja (CS) e de três fontes de nitrogênio sobre a digestão total e parcial aparentes no rúmen e nos intestinos. Seis novilhos Holandês x Zebu com idade média de 12 meses e 229 kg de PV foram fistulados no rúmen e no abomaso e distribuídos em quadrado latino 6x6 e alimentados individualmente ad libitum. O consumo e as digestibilidades totais e parciais dos nutrientes foram avaliados. Não houve efeito da fonte de proteína sobre o consumo, quantidades de matéria seca (MS), matéria orgânica (MO), proteína bruta (PB), fibra em detergente neutro (FDN) e da fibra em detergente ácido (FDA) no rúmen, no abomaso e nas fezes e sobre os coeficientes de digestibilidades destes componentes. O aumento, porém, do nível de casca de soja elevou os coeficientes de digestibilidade de todos os componentes estudados nos locais do trato digestivo, com exceção nos intestinos, onde o nível de casca de soja não afetou a digestão da PB, FDN e da FDA. Os resultados indicam que a casca de soja pode substituir tanto o milho como, parcialmente, a fração volumosa nas rações de ruminantes. A farinha de subproduto avícola e a amiréia podem substituir o farelo de soja nas rações de bovinos sem reflexos negativos sobre a digestão de seus componentes nutritivos.

Os efeitos de ano e época de nascimento, propriedade, sexo do bezerro, regime alimentar, idade do bezerro e idade da vaca ao parto, os efeitos aditivos direto e materno da raça Charolesa e o efeito de heterose materna entre as raças Charolesa e zebu foram avaliados pelo método dos quadrados míninos sobre características de importância econômica, como peso, perímetro escrotal e escores visuais de conformação frigorífica, de umbigo e de pelame à desmama em bovinos da raça Canchim. Utilizaram-se dados de 12.334 animais Canchim nascidos de 1999 a 2005. O ano e a época de nascimento, a propriedade, o regime alimentar e a idade da vaca ao parto (linear e quadrático) tiveram efeitos significativos sobre todas as características estudadas. Houve efeitos do sexo sobre o peso do bezerro à desmama e o escore de umbigo; da idade do bezerro sobre peso, perímetro escrotal e escores de conformação frigorífica e de pelame; e das proporções de Charolês no bezerro e na mãe sobre o peso, a conformação frigorífica e a qualidade de pelagem; e da heterozigose materna sobre peso, perímetro escrotal e escores de conformação frigorífica e de umbigo. Os efeitos aditivos diretos da raça Charolesa como desvio do zebu foram negativos sobre peso, conformação frigorífica e qualidade da pelagem à demama, enquanto os efeitos aditivos maternos foram positivos. Os efeitos heteróticos maternos foram positivos sobre peso, perímetro escrotal e escores de conformação frigorífica e de umbigo. em geral, essas fontes de variação devem ser consideradas nas estimativas de parâmetros genéticos e nas avaliações genéticas dos animais Canchim para as características estudadas.

O objetivo deste trabalho foi avaliar a influência da ingestão alimentar nas concentrações séricas de hormônios reprodutivos e metabólicos em vacas azebuadas. Dezoito vacas foram divididas em dois grupos: 170% (alta ingestão = A) e 66% (baixa ingestão = B) da dieta de manutenção. Com 21 dias nas dietas experimentais, as vacas tiveram o estro sincronizado. Posteriormente, os ovários foram avaliados por ultra-sonografia transretal e sangue foi coletado diariamente até o dia 7 do ciclo (ovulação = dia 1). Na análise estatística, utilizou-se o teste t. As vacas ganharam 1,1 kg por dia no grupo A e perderam 1,5 kg por dia de PV no grupo B. Apesar de não ter havido diferença entre os grupos no diâmetro máximo do folículo ovulatório, o grupo A apresentou pico pré-ovulatório de estradiol sérico menor. Não foi observada diferença entre os grupos quanto ao volume luteal e concentração sérica de progesterona no dia 7 do ciclo e de FSH, IGF-I e insulina séricos no período peri-ovulatório. As dietas experimentais não alteraram a função ovariana e as concentrações séricas de hormônios reprodutivos e metabólicos, com exceção do estradiol, sugerindo que, no grupo de alta ingestão, ocorreu maior metabolismo desse hormônio.

The aim of this work was to assess the morphometry and chromatin integrity of bovine sperm head after a three layers discontinuous Percoll (TM) density gradient centrifugation. Frozen semen doses were obtained from six bulls of different breeds, including three taurine and three Zebu animals. Three ejaculates per bull were evaluated. The semen doses were thawed and two smears were made from each sample before (control) and after the Percoll (TM) centrifugation (Percoll (TM) group). The smears were stained with toluidine blue and grayscale digital images were captured and processed in Scilab environment software. It was observed that chromatin heterogeneity was reduced (P<0.05) and chromatin decondensation was increased (P<0.05) after the Percollm treatment utilized. In addition, it was observed that sperm head length was higher (P<0.05) and the side symmetry was lower (P<0.05) in centrifuged sperm cells. When analyzed separately by subspecies, it was observed that the decrease (P<0.05) in chromatin heterogeneity and the increase (P<0.05) in chromatin decondensation occurred in Zebu sperm heads. In addition, the length and the width:length ratio of sperm heads was affected by Percoll (TM) centrifugation in Zebu semen. In conclusion, the three layers discontinuous Percoll (TM) centrifugation increased the chromatin decondensation and the morphometric alterations of frozen-thawed bovine semen. However, the real implication of these findings in fertility rates of centrifuged sperm must be investigated. (C) 2011 Elsevier B.V. All rights reserved.

The objective of this study was to evaluate the quality of bovine frozen-thawed sperm cells after Percoll gradient centrifugation. Frozen semen doses were obtained from six bulls of different breeds, including three taurine and three Zebu animals. Four ejaculates per bull were evaluated before and after discontinuous Percoll gradient centrifugation. Sperm motility was assessed by computer-assisted semen analysis and the integrity of the plasma and acrosomal membranes, as well as mitochondrial function, were evaluated using a combination of fluorescent probes propidium iodide, fluorescein isothiocyanate-conjugated Pisum sativum agglutinin and 5,5',6,6'-tetrachloro-1,1',3,3'-tetraethylbenzimidazolcarbocyanine iodide. The procedure of Percoll gradient centrifugation increased the percentage of total and progressive sperm motility, beat frequency, rectilinear motility, linearity and rapidly moving cells. In addition, the percentage of cells with intact plasma membrane and mitochondrial membrane potential was increased in post-centrifugation samples. However, the percentage of sperm cells with intact acrosomal membrane was markedly reduced. The method used selected the motile cells with intact plasma membrane and higher mitochondrial functionality in frozen-thawed bull semen, but processing, centrifugation and/or the Percoll medium caused damage to the acrosomal membrane.
