Enterococci are increasingly responsible for nosocomial infections worldwide. This study was undertaken to compare the identification and susceptibility profile using an automated MicrosScan system, PCR-based assay and disk diffusion assay of Enterococcus spp. We evaluated 30 clinical isolates of Enterococcus spp. Isolates were identified by MicrosScan system and PCR-based assay. The detection of antibiotic resistance genes (vancomycin, gentamicin, tetracycline and erythromycin) was also determined by PCR. Antimicrobial susceptibilities to vancomycin (30 µg), gentamicin (120 µg), tetracycline (30 µg) and erythromycin (15 µg) were tested by the automated system and disk diffusion method, and were interpreted according to the criteria recommended in CLSI guidelines. Concerning Enterococcus identification the general agreement between data obtained by the PCR method and by the automatic system was 90.0% (27/30). For all isolates of E. faecium and E. faecalis we observed 100% agreement. Resistance frequencies were higher in E. faecium than E. faecalis. The resistance rates obtained were higher for erythromycin (86.7%), vancomycin (80.0%), tetracycline (43.35) and gentamicin (33.3%). The correlation between disk diffusion and automation revealed an agreement for the majority of the antibiotics with category agreement rates of > 80%. The PCR-based assay, the van(A) gene was detected in 100% of vancomycin resistant enterococci. This assay is simple to conduct and reliable in the identification of clinically relevant enterococci. The data obtained reinforced the need for an improvement of the automated system to identify some enterococci.

Intestinal parasites are a problem for public health all over the world. The infection with Blastocystis, a protozoan of controversial pathogenicity, is one of the most common among them all. In this study, the occurrence of intestinal parasites, with emphasis on Blastocystis, in patients at the Universidade Federal do Triângulo Mineiro was investigated in Uberaba (MG) through microscopy of direct smears and fecal concentrates using Ritchie’s method. Feces of 1,323 patients were examined from April 2011 to May 2012. In 28.7% of them at least one intestinal parasite was identified, and the most frequent organisms were Blastocystis spp. (17.8%) and Giardia intestinalis (7.4%). The occurrence of parasitism was higher in children aged 6 -10 years old, and the infection with Blastocystis spp. was higher above the age of six (p < 0.001). The exclusive presence of G. intestinalis and of Blastocystis spp. was observed in 5.4% and 12.2% of the patients, respectively. Regarding patients with diarrheic feces, 8% revealed unique parasitism of Blastocystis spp. Other intestinal parasites observed in children were Ascaris lumbricoides(0.3%) and Entamoeba histolytica/dispar/moshkovskii (1.4%). The Ritchie’s method was more sensitive (92.8%) when compared to direct microscopy (89.8%), with high agreement between them (97.7%, kappa = 0.92). In conclusion, the occurrence of Blastocystis spp. in Uberaba is high and the presence of diarrheic feces with exclusive presence of the parasite of Blastocystis spp. was observed.

Entamoeba histolytica e Giardia lamblia são protozoários com distribuição mundial que frequentemente infectam o Homem, sendo causadores de elevada morbilidade associada com quadros de diarreia. Este estudo consistiu na utilização de métodos moleculares na detecção e identificação destes parasitas em amostras de fezes recebidas no Laboratório de Patologia Tropical do Instituto de Higiene e Medicina Tropical (Lisboa, Portugal), no período decorrente entre Setembro de 2007 e Agosto de 2009. . Foi igualmente avaliada a eficácia e custo-benefício de um método alternativo de conservação para material biológico fecal – o papel de filtro, para subsequente extracção de DNA. Foram analisadas microscopicamente 80 amostras, 23,8 % (19/80) das quais positivas para Giardia e 27,5% (22/80) positivas para Entamoeba spp.. Através do método molecular da PCR, amplificou-se com sucesso DNA de Giardia para o gene ssurRNA em 94,7% (18/19) das amostras microscopicamente positivas. No que se refere às amostras positivas por exame microscópico para Entamoeba spp foi detectada E. dispar em 50,0% (11/22) das amostras após amplificação de parte do gene 16S rRNA. Adicionalmente foi também detectado DNA de E. histolytica em 20,0% (4/20) das amostras analisadas, microscopicamente negativas para Entamoeba spp.. Neste estudo realizou-se a genotipagem de G. lamblia utilizando parte dos genes bg (beta-giardina), tpi (triose-fosfato isomerase) e gdh (glutamato desidrogenase), que revelou haver 61,5% (8/13) dos isolados pertencentes ao ge³tipo B e 38,5% (5/13) do ge³tipo A. A determinação de subge³tipos através da a¡lise de SNP’s para os 3 genes só foi possível para o gene bg do ge³tipo A, revelando 3 amostras correspondentes ao subge³tipo A2 e uma para o subge³tipo A3. Para os restantes genes £o foi possível a determinação de subge³tipos devido à prese§a de polimorfismos ge©ticos para ambos os ge³tipos A e B. Realizou-se também a a¡lise filoge©tica concatenada que apenas permitiu a integração de três amostras identificadas com o ge³tipo A no subge³tipo AII. Os resultados obtidos neste trabalho demonstram que a microscopia associada às técnicas moleculares possibilita a diferenciação das espécies do complexo Entamoeba favorecendo o correcto diagnóstico desta patologia e consequente tratamento. Para além disso, o uso dos métodos moleculares contribuiu para o esclarecimento e compreensão dos ge³tipos de Giardia em humanos. Neste estudo a utilização do método de conservação, papel de filtro apresentou um menor custo e elevada eficácia em relação aos métodos normalmente utilizados, conservação a -20ºC. Sugerindo a sua utilização com sucesso em estudos epidemiológicos em especial em zonas endémicas de condições precárias.

The leaf-feeding species Syntermes molestus and S. spinosus are two dominant termite species in Reserva Ducke in Central Amazonia; two other species (S. aculeosus, S. longiceps) exist in the area. All species except S. aculeosus were also found in urban areas. The workers of S. molestus and S. spinosus have average body dry weights of 4.5±0.2 and 13.1±1.4 mg, and the weight of their soldiers is 8.2±0.2 and 51.0±1.7 mg, respectively. Therefore, S. spinosus is among the largest termites of the world. In both species, fresh weight is about 4.7 higher than dry weight (a wider relation than in other termite species). The biomass of the populations of both species amounted to about 1 g m-2 (dry weight; indirect estimate), which rises previous assessments of the total termite biomass by about 36-45%, to a value of 3.0-3.5 g m-2.

A banana (Musa spp.) é uma das frutas mais consumidas no mundo, e amplamente cultivada no Brasil, porém doe§as como as sigatokas, negra e amarela, vêm reduzindo a sua produção. A disponibilização imediata de novas cultivares resistentes às principais doe§as é limitada pela propagação convencional. A micropropagação é uma alternativa para a produção de mudas com qualidade fitossanitária e vegetativa, mas apresenta fatores que dificultam sua aplicação como a contaminação por fungos e bactérias, associada à oxidação dos explantes. O objetivo desse trabalho foi adaptar e/ou otimizar as etapas do processo de micropropagação para diferentes cultivares de bananeira, por meio do controle de oxidação, contaminação, e multiplicação de brotos, sendo utilizadas as cultivares Caipira (AAA), BRS Caprichosa (AAAB), Pacovan Ken (AAAB), Preciosa (AAAB), PV 03-76 (AAAB), Thap Maeo (AAB). No estudo foram utilizados o antibiótico sulfato de estreptomicina e o fungicida Opera® (BASF) visando reduzir a contaminação in vitro provocada por bactérias e fungos, além do anti-oxidante PVP (polivinilpirrolidona) para controlar a oxidação. Houve redução da contaminação com uso do sulfato de estreptomicina à concentração de 100 mg L-1 e da oxidação com PVP a 4 g L-1. Na fase de multiplicação de brotos, as cultivares apresentaram médias que variaram de 1,90 a 4,75 brotos/explante. A cultivar caipira (AAA) destacou-se das demais com a maior taxa de multiplicação de brotos após três subcultivos, média de 41,50 brotos por rizoma.

Com a finalidade de determinar as doses de nitroªnio, fósforo e potássio mais adequadas para obte§Ã£o de plantas aptas para enxertia em viveiro de seringueira, instalou-se um experimento em Latossolo Amarelo textura média na Ilha do Mosqueiro-PA. O delineamento experimental foi de blocos ao acaso com duas repetições obedecendo ao arranjo fatorial 33 Foram utilizadas as doses de 0-2,1-4,2 g/planta de N; 0-3,5-7,0 g/planta de P2O5; 0-1,4-2,8 g/planta de K2O e dose constante de 0,8 g/planta de MgO, empregando-se como fontes, respectivamente, sulfato de amônio, superfosfato triplo, cloreto de potássio e sulfato de magnésio. Os resultados foram obtidos duzentos e vinte dias após a instalação do experimento, sendo realizadas as seguintes avaliações: a¡lises químicas do solo e folhas, altura das plantas, diâmetro do caule, peso da matéria seca da parte aérea e plantas aptas para a enxertia. Pelos resultados obtidos chega-se à conclusão que as doses mais adequadas foram 330 kg/ha (4,6 g/p) de N, 340 kg/ha (4,8 g/p) de P2O5 e 190 kg/ha (2,7 g/p) de K2O, aliadas a dose constante de 60 kg/ha (0,8 g/p) de MgO, propiciando um índice de aproveitamento de oitenta e sete porcento de plantas aptas para enxertia.

A interação Ficus (Moraceae) - vespas de figo é considerada um dos exemplos mais extremos de mutualismo entre planta e inseto. Neste trabalho, descreve-se a fauna de vespas de figo associada a cinco espécies de Ficus na Amazônia Central, considerando alguns aspectos do modo de polinização nas espécies Ficus (Urostigma) cremersii, Ficus (Urostigma) greiffiana, Ficus (Urostigma) mathewsii, Ficus (Urostigma) pertusa e Ficus (Pharmacosycea) maxima. O estudo foi desenvolvido durante o período de abril a julho de 2004 em Manaus e Presidente Figueiredo, Estado do Amazonas. O ºmero de espécies de vespas de figo por hospedeiro variou de uma a 13. Vespas do ªnero Pegoscapus Cameron, 1906, polinizadoras de Ficus (Urostigma) spp., apresentam pentes coxais e bolsos torácicos adaptados à coleta e ao transporte de pólen, indicando modo ativo de polinização. No subªnero Pharmacosycea, a polinizadora do ªnero Tetrapus Mayr, 1885, £o apresenta estrutura morfológica adaptada ao transporte de pólen, condizente com o modo passivo de polinização. Além das vespas de figo, F. (Pharmacosyceae) maxima e F. (Urostigma) pertusa apresentaram associação com ácaros, formigas (Solenopsis sp., Formicidae), besouros (Staphylinidae) e larvas de Diptera e Lepidoptera.

Invasive candidiasis (IC) is a relatively common syndrome in neonates and children and is associated with significant morbidity and mortality. These guidelines provide recommendations for the prevention and treatment of IC in neonates and children. Appropriate agents for the prevention of IC in neonates at high risk include fluconazole (A-I), nystatin (B-II) or lactoferrin ± Lactobacillus (B-II). The treatment of IC in neonates is complicated by the high likelihood of disseminated disease, including the possibility of infection within the central nervous system. Amphotericin B deoxycholate (B-II), liposomal amphotericin B (B-II), amphotericin B lipid complex (ABLC) (C-II), fluconazole (B-II), micafungin (B-II) and caspofungin (C-II) can all be potentially used. Recommendations for the prevention of IC in children are largely extrapolated from studies performed in adults with concomitant pharmacokinetic data and models in children. For allogeneic HSCT recipients, fluconazole (A-I), voriconazole (A-I), micafungin (A-I), itraconazole (B-II) and posaconazole (B-II) can all be used. Similar recommendations are made for the prevention of IC in children in other risk groups. With several exceptions, recommendations for the treatment of IC in children are extrapolated from adult studies, with concomitant pharmacokinetic studies. Amphotericin B deoxycholate (C-I), liposomal amphotericin B (A-I), ABLC (B-II), micafungin (A-I), caspofungin (A-I), anidulafungin (B-II), fluconazole (B-I) and voriconazole (B-I) can all be used.

Specimens collected from the large intestine of the sidenecked turtle Podocnemis unifilis Troschel, 1848 in the region of Cumi¡ and Trombetas rivers near Pará, Brazil are assigned to a new genus and new species of the nematode superfamily Cosmocercoidea and family Atractidae and named Paraorientatractis semiannulata. The new genus is separated from the nearest genus Orientatractis by the funnelshaped mouth opening, the presence of 4 distinct lips, 4 papillae in the internal cycle, one on each lip margin, 2 lateral amphids with large amphidial pores and absence of submedian papillae. It is also separated from Orientatractis and Proatractis by the presence of striated lateral alae which curve dorsally extending from mid oesophagus to mid tail, the difference in size of the vulvar opening and the presence of large transverse ridges or semiannules on the dorsal surface. The new species can be separated from the species of the genera Orientatractis and Proatractis by the characters that distinguish the genera and the arrangement of the caudal papillae on the male. A host/parasite list for Podocnemis spp. is included

The infection rates of Babesia sporokinetes in engorged Boophilus microplus were evaluated during a 2-year period in a dairy farm located in an area of enzootic stability. Every 14 days engorged females were collected from calves and from adult animals. Ticks were incubated at 27 ± 0.5ºC and 80-90% relative humidity and Babesia infection rates were determined by microscopic examination of Giemsa-stained hemolymph smears. After 52 collections, 2105 ticks were obtained, from which 982 were collected from calves and 1123 from cows. The total Babesia infection rate was 10%, however the incidence was higher (p < 0.05) in ticks collected from calves (17.5%) than in those collected from cows (3.6%). Females collected from cows showed the highest infection rates in January, March, and August, and absence of infection in April and May. Ticks feeding on calves were infected throughout the experimental period. The infection rates of engorged females collected from naturally infected calves that were artificially infested with Babesia-free-larvae of B. microplus gradually decreased until the calves were four months old. No differences were observed among infection rates of ticks collected from calves maintained under natural conditions.

Infections by Cryptococcus strains other than C. neoformans have been detected in immunocompromised patients. Of these strains, three are considered human pathogens: C. albidus, C. laurenttii, and C. uniguttulatus. This study deals with the in vitro susceptibility of Cryptococcus to drugs such as amphotericin B, itraconazole, fluconazole, and 5-fluorocytosine. Environmental Cryptococcus isolates (50) distributed as follows: C. neoformans var. neoformans (16), C. albidus (17), C. laurentii (14), and C. uniguttulatus (3) were evaluated by the micro and macrodilution techniques, according to EUCAST and NCCLS recommendations, respectively. Considering both methodologies the respective minimal inhibitory concentrations (MIC) were 0.125 and 2 µg/ml for amphotericin B, 0.06 and 8 µg/ml for itraconazole, and 0.5 and more than 64 µg/ml for fluconazole and 5-fluorocytosine. Agreement percentages for the two methodologies were 100% for amphotericin B and fluconazole for all the strains tested. For itraconazole, the agreement percentage was 81.3% in the C. neoformans strain and 100% for all the others. All species had a agreement percentage of 94.1 to 100% when susceptibility to 5-fluorocytosine was tested. It is concluded that environmental isolates of C. neoformans var. neoformans, C. albidus, C. laurentii, and C. uniguttulatus may show high MICs against certain drugs, suggesting in vitro primary resistance to the antifungals tested.

Antibacterial and antifungal properties of wax and hexane extracts of Citrus spp. peels were tested using bioautographic and microdilution techniques against three plant pathogenic fungi (Penicillium digitatum, Curvularia sp., and Colletotrichum sp.), two human pathogens (Trichophyton mentagrophytes and Microsporum canis), and two opportunistic bacteria (Escherichia coli and Staphylococcus aureus). Two polymethoxylated flavonoids and a coumarin derivative, were isolated and identified from peel extracts, which presented antimicrobial activity especially against M. canis and T. mentagrophytes: 4',5,6,7,8-pentamethoxyflavone (tangeritin) and 3',4',5,6,7,8-hexamethoxyflavone (nobiletin) from C. reticulata; and 6,7-dimethoxycoumarin (also known as escoparone, scoparone or scoparin) from C. limon.

Three Plumbago spp have been tested for mosquito larvicidal activity. The crude extracts exhibiting the highest larvicidal activity against Anopheles gambiae were hexane (LC50 = 6.4 μg/mL) and chloroform (LC50 = 6.7 μg/mL) extracts from Plumbago zeylanica Linn, chloroform (LC50 = 6.7 ug/mL) extract from Plumbago stenophylla Bull and ethyl acetate (LC50 = 4.1 μg/mL) extract from Plumbago dawei Rolfe. These LC50 values were within 95% confidence limits. 5-hydroxy-2-methyl-1,4-naphthoquinone (plumbagin) 1 (LC50 = 1.9 μg/mL) and β-sitosterol 2 were characterised from ethyl acetate extract of root bark of P. dawei, a native medicinal plant growing in Kenya, based on spectral analysis and comparisons with data in literature.

The chemical composition and biological activities of 19 essential oils and seven of their major components were tested against free and intracellular forms of Leishmania chagasi and Trypanosoma cruzi parasites as well as Vero and THP-1 mammalian cell lines. The essential oils were obtained from different species of Lippia, a widely distributed genus of Colombian plants. They were extracted by microwave radiation-assisted hydro-distillation and characterised by GC-FID and GC-MS. The major components were geranial, neral, limonene, nerol, carvacrol, p-cymene, γ-terpinene, carvone and thymol. The essential oil of Lippia alba exhibited the highest activity against T. cruzi epimastigotes and intracellular amastigotes with an IC50 of 5.5 μg/mL and 12.2 μg/mL, respectively. The essential oil of Lippia origanoides had an IC50 of 4.4 μg/mL in L. chagasi promastigotes and exhibited no toxicity in mammalian cells. Thymol (IC50 3.2 ± 0.4 μg/mL) and S-carvone (IC50 6.1 ± 2.2 μg/mL), two of the major components of the active essential oils, were active on intracellular amastigotes of T. cruziinfected Vero cells, with a selective index greater than 10. None of the essential oils or major components tested in this study was active on amastigotes of L. chagasi infected THP-1 cells.

The effects of Crypthecodinium cohnii (Cryp.), Chlorela spp. (Chlo.) and Isochrysis galbana (Iso.) addition to milk replacer on goat kids and lambs growth were evaluated. About 80 Majorera goat kids (males and females) and 80 Canarian sheep lambs were randomly assigned into four different groups (by specie) according to diet. Control groups were fed with a commercial milk replacer at 16% (w/w); Cryp. groups received a commercial milk replacer (15.1% w/w) supplemented with 9 g of a paste of C. cohnii; Chlo. groups received a commercial milk replacer (15.1% w/w) supplemented with 9 g of a paste of Chlorela spp.; Iso. groups received a commercial milk replacer (15.1% w/w) supplemented with 9 g of a paste of I. galbana. After colostrum period, animals were individually bottle-fed twice daily (8 am and 8 pm) ad libitum with the corresponding diet until day 60 of life. Animals were weighted every week at 8 am and liquid diet intake was recorded weekly. No effects of microseaweed addition were observed, neither growth nor milk replacer intake.
