Short and long-term thyroidectomy and Methimazole treatment reduced food intake in young growing pigs. The thermic effect of feeding assessed by the increment in rectal temperature after the beginning of food ingestion was reduced in thyroidectomized animals, but no effect could be observed in Methimazole-treated pigs. Propranolol injection after short-term treatment decreased food intake in sham-operated and treated animals, but reduced the thermic effect of feeding only in the thyroidectomized and Methimazole-treated pigs. Long-term treatment inhibited the effect of propranolol in reducing food intake and the thermic effect of feeding. On the basis of these data, it was suggested that the interaction between thyroid hormones and catecholamines (noradrenaline) plays an important role in the regulation of food intake and in the thermic effect of feeding in thyroid-deficient pigs.

Food intake and plasma thyroid hormone levels (T4 and T3) were higher in pigs acclimated to cold (12°) than hot (32°) environments. The exposure of cold pigs to hot ambient temperature decreased food intake and plasma T4 and T3, whereas for hot acclimated animals the change in ambient temperature (from 32 to 12° C) increased food intake and plasma thyroid hormone levels, but the new steady state level of food intake was reached only after 96 hr of temperature transfer despite the rapid change in plasma levels of thyroid hormones. Cold-acclimated pigs, when transferred to a hot environment after thyroidectomy, also reduced food intake, but hot pigs shifted to cold ambient temperature after thyroidectomy did not significantly increase food ingestion. The results of this experiment suggest that food intake adjustment depends on the previous living temperature and that thyroid hormones seem to play an important role in increasing the metabolically active mass that probably sustains the new steady state level of food intake, particularly in a cold environment.

The development of the febrile response to E. coli lipopolysaccharide (1.5 μg/kg, i.v.) in thyroid-deficient rabbits has been studied. Twenty-eight New Zealand White rabbits weighing 2.1-2.3 kg were used. Hypothyroidism was induced by treatment with propylthiouracil (100 or 200 mg/kg body wt./15 days). Thyroid-deficient animals showed a reduction in the febrile response to lipopolysaccharide, but the effect was significantly different (p<0.01) from the control only for rabbits treated with 200 mg/kg of propylthiouracil. Propranolol (2 mg/kg, i.p.) given 30 min before lipopolysaccharide also reduced (p<0.01) the fever response in control rabbits. The results of this experiment are consistent with the hypothesis that the reduction in the febrile response of thyroid-deficient rabbits is due to the reduced number of β-adrenergic receptors, or to a change in the availability of neurotransmitter in thermogenically active tissues, such as brown fat.

The development of a fever in response to intravenous (IV, 1.5 μg/kg body mass) and intracerebroventricular (ICV, 1.5 μg/animal) injections of Escherichia coli lipopolysaccharide (LPS) was studied in control, thyroidectomised and protein-calorie malnourished rabbits (New Zealand Whites, n = 55). ICV injection of LPS is control rabbits produced a fever response, the characteristics of which differed from those obtained after IV pyrogen injection. Thyroid deficiency caused an attenuated fever response, irrespective of whether LPS had been administered by IV or ICV injection. Protein-calorie malnourished rabbits showed a smaller fever response after IV or ICV pyrogen injections. Malnourished rabbits, refed over a period of 15 days, showed a typical biphasic fever response, but with lower magnitude than controls. The results of these experiments suggest that ICV injection of LPS is not an appropriate model for the study of fever mechanisms in disease states, and that the attenuated fever response observed in protein-calorie malnourished rabbits may be related, at least in part, to a decreased ability to produce the endogenous pyrogen interleukin-1.

Short-term cultures of a collagenase disaggregated multinodular goiter was shown by cytogenetic analysis to have the mosaic karyotype 47,XX,+7/48,XX,+7,+17/49,XX,+7,+10,+17. No cytogenetic data on goiter are available for comparison with the present case.

A tireoide sintetiza a tiroxina (T4) e a 3,5,3’-triiodotironina (T3), ambos hormônios apresentam uma função crucial no desenvolvimento do sistema nervoso central, incluindo o sistema visual e a retina. A diminuição dos níveis sanguíneos do T3 e T4 ocasionam uma síndrome denominada de hipotireoidismo, o que pode levar à prejuízos visuais. Os déficits visuais gerados pelo hipotireoidismo estão diretamente relacionados ao período de desenvolvimento do indivíduo. Foi demonstrado em modelos murinos que o hipotireoidismo congênito diminui a espessura da retina, o número de células, e interfere na diferenciação da subpopulação de cones M. Desta forma buscaremos investigar possíveis alterações funcionais na retina de ratos wistar jovens após a tireoidectomia bilateral, utilizando respostas eletrofisiológicas não invasivas. Para tanto, dividimos os ratos em três grupos (controle, sham e tireoidectomizado) cada um contendo ≥ 8 animais. As cirurgias foram realizadas 30 dias pós-natal. Os eletrorretinogramas de campo total foram realizados 10, 15, 20, 25 e 30 dias após a cirurgia, utilizando protocolos para avaliar a resposta escotópica máxima, resposta fotópica (com e sem o uso de filtros de luz) e a resposta ao flicker (12, 15, 18 e 30 Hz). Os parâmetros analisados foram o tempo implícito e a amplitude das ondas a e b. Além disso, realizamos o monitoramento dos parâmetros clínicos dos animais, visando identificar características que indiquem um quadro de hipotireoidismo, bem como a dosagem dos hormônios tireoidianos. Os resultados obtidos demonstraram que em todos dos protocolos de estimulação utilizados no ERG houve diminuição nas amplitudes das ondas a e b nos animais tireoidectomizados em todos os dias avaliados após a cirurgia, quando comparados com animais do grupo controle e sham. Os resultados da avaliação do tempo implícito para ambas as ondas não demonstraram diferença estatística quando comparamos os diversos grupos ao controle. Também podemos constatar uma redução do ganho de peso e tamanho nos animais que sofreram tireoidectomia, associados à redução dos níveis de hormônio tireoidiano (T3). Concluímos dessa forma que os hormônios tireoidianos estão diretamente ligados a alterações funcionais na retina dos animais que sofreram tireoidectomia, bem com, na redução da aquisição de peso e aumento de tamanho.

O diabetes mellitus constitui um problema de saúde pública mundial devido ao seu potencial de morbidade e mortalidade. Esta patologia é caracterizada como uma desordem metabólica com hiperglicemia crônica, resultado de uma deficiência absoluta ou relativa na secreção e/ou ação da insulina. Associado a esse quadro, problemas com a tireoide são frequentemente encontrados em pacientes acometidos por diabetes, principalmente com o avançar da idade. Além disso, há um corpo de conhecimento que descreve que tanto o diabetes quanto o hipotiroidismo provocam isoladamente perdas visuais, no entanto, não há muitas publicações acerca da fisiologia envolvendo em conjunto ambas patologias com as complicações visuais. O presente trabalho objetivou investigar questões relevantes para uma compreensão mais detalhada do quadro de evolução do comprometimento retiniano em modelos animais acometidos concomitantemente por duas doenças: diabetes e hipotireoidismo. Para isso, foram utilizados 50 ratos Wistar machos (Rattus norvegicus), com dois meses de idade, pesando entre 80 a 120 gramas, divididos em quatro grupos: um grupo controle sem procedimentos; um grupo com hipotireoidismo optando por um modelo experimental de tireoidectomia bilateral; um grupo com diabetes, com aplicação de 200 mg/Kg a 2% de Aloxana; e dois grupos com as duas patologias, adotando ambos os procedimentos experimentais, alterando a sequência das patologias em questão. Nos grupos estudados foi utilizado para avaliar as alterações visuais, um método não invasivo amplamente utilizado no meio acadêmico que é o eletrorretinograma (ERG) em diferentes períodos de 30, 45 e 60 dias. Assim, encontramos uma diminuição da amplitude média da onda-a dos animais nos grupos com diabetes e com as duas patologias em todos os registros realizados, os que apresentaram maiores diferenças estatísticas foram em relação ao registro de estocópica máxima, como também diferença no potencial oscilatório de ambos os grupos com as duas patologias. Os resultados obtidos dão suporte à hipótese que as duas patologias concomitantes (diabetes e hipotireoidismo) diminuem significativamente as respostas do ERG.

The 131I (radioactive iodine) is one of the most used radionuclides in nuclear medicine for diagnosis and treatment. The present study evaluates the dosimetric aspects related to radioiodine therapy after thyroidectomy in patients with thyroid cancer. The samples were studied with 50 patients undergoing treatment, by assessing the exposures of workers (nursing assistants, staff hygiene, medical and physical), the general public (accompanying and family members) and on the environment. To evaluate the workers, was made a survey of the environmental conditions of the room radioiodine and routines adopted by them during the treatment period. Estimating the dose equivalent per month for each employee, we observed that the maximum levels obtained for nursing assistants, the team of hygiene, medical and physicians were considered low in relation to the extent permitted by law. In order to assess the public, some situations have been suggested for the calculation of equivalent doses in which it was possible to verify the fundamental importance of isolating the patient in the 2 days first. Regarding the environment, the radioactive waste generated by patient had volume of 1.0 m3 and activity estimated at 0.91 mCi, taking a decay time for eliminating them about 75 days to reach the allowable value of 2 μCi / kg system of collecting garbage. Therefore, all radioactive waste removed from the patient's room should be sent to the warehouse for temporary storage of radioactive waste, located away from normal work areas

A case of primary squamous-cell carcinoma (SCC) of the thyroid which had been initially diagnosed as an anaplastic carcinoma (ATC) is described: female, 73 years old, with a fast-growing cervical nodule on the left side and hoarseness for 3 months. Ultrasonography showed a 4.5 cm solid nodule. FNA was compatible with poorly differentiated carcinoma with immunoreactivity for AE1/AE3, EMA. Thyroidectomy was performed. Histopathological examination showed a nonencapsulated tumor. Immunohistochemistry disclosed positivity for AE1/AE3, p53,p63, and Ki67. The diagnosis was ATC. A second opinion reported tumor consisting of squamous cells, with intense inflammatory infiltrate both in tumor and in the adjacent thyroid, with final diagnosis of SCC, associated with Hashimoto thyroiditis. No other primary focus of SCC was found. Patient has shown a 48-month survival period. Clinically, primary SCCs of the thyroid and ATCs are similar. The distinction is often difficult particularly when based on the cytological analysis of FNA material.

To evaluate the impact of iodine-131 therapy received during childhood and adolescence and correlate it with the quality of life in these patients. Methods: We studied 19 patients diagnosed with cancer in childhood or adolescence who underwent thyroidectomy and supplemental therapy with I-131. We also recruited a control group of healthy subjects with the same demographic parameters. All patients were subjected to a scintigraphy examination of the salivary glands, and were also asked to complete a questionnaire in order to assess their overall quality of life. In addition, a more specific questionnaire for patients with head and neck cancer was also given to all study participants. Results: The quantitative and qualitative analyses of the salivary glands showed functional deficits with greater involvement of the parotid gland for volume, concentration and excretion. The right submandibular gland showed significant changes for volume in the patient group. The questionnaires made it possible to observe significant differences between the patient and control groups for symptoms such as thick saliva, dry mouth and speech problems. Conclusion: In spite of being very effective and widely used, iodine radionuclide therapy is correlated with a lower quality of life in young people.
